In a sample of 229 individuals who recently undertook a large-scale sacrifice by relocating for their romantic partner's job or schooling (i.e., accompanying partners), we tested preregistered predictions linking accompanying partners' attachment insecurities (i.e., attachment anxiety and attachment avoidance) and relationship functioning (i.e., relationship quality, relationship conflict, and move-related relationship benefits). We also examined whether any negative associations found among accompanying partners' attachment insecurities and relationship functioning could be buffered by partner prosociality in the form of perceived partner gratitude (i.e., partners' expressions of move-related and general gratitude) and perceived partner sacrifice (i.e., partners' daily sacrifice behaviors and general willingness to sacrifice). Results showed that more insecurely attached accompanying partners reported worse relationship functioning after moving than their secure counterparts. Although gratitude and sacrifice did not buffer insecurely attached individuals' relationship conflict, both perceived partner general gratitude and willingness to sacrifice partially buffered avoidantly attached individuals from experiencing lower relationship quality, while move-related gratitude helped avoidantly attached individuals to feel that the move benefitted their relationship. Meanwhile, perceived partner sacrifice behaviors buffered anxiously attached individuals from experiencing lower relationship quality. This is the first study to demonstrate, in an ecologically valid sample, the implications of a large-scale sacrifice for insecurely attached accompanying partners' relationship functioning, as well as the protective effects of perceiving a partner's prosociality following the major life transition of job relocation. Agri-food industries such as chicken meat production face increasing pressure to quantify and improve their environmental performance over time, while simultaneously increasing production to meet global demand. Using life cycle assessment, this study aimed to quantify resource use, environmental impacts and hotspots for Australian chicken meat production using updated inventories and new methods. Two contrasting states; Queensland, and South Australia, and two housing systems; conventional and free range were analysed to indicate the variation expected between regions and systems. Lower impacts were observed per kilogram of chicken meat produced in South Australia compared to Queensland for fossil fuel energy, greenhouse gas (including land use and direct land use change) and fresh water consumption (18.1 and 21.4 MJ, 2.8 and 3.4 kg CO 2 -e, 38 and 111 L respectively), but not arable land or stress weighted water use (22.5 and 14 m 2 , 36 and 26 L H 2 O-e respectively). Feed production was the largest contributor to all impact categories, and also showed the largest variation between regions, highlighting the importance of spatially specific feed grain datasets to determine resource use and greenhouse gas from chicken meat production. While the feed conversion ratio was lower in South Australia, this was found to be less significant than differences related to crop yield, irrigation water use and the use of imported feed ingredients, suggesting that incremental improvements in feed conversion ratio will result in lower impacts only when feed inputs and production systems do not change. Fresh water consumption was lower in South Australia, but did not correlate with stress weighed water use (lower in Queensland), highlighting that volumetric water use is not a reliable indicator of the impact of water use. We did not observe substantial differences between conventional and free range production when feed related differences were removed, because key productivity factors such as feed conversion ratio were similar between the two housing types in Australia. While results were found to vary between regions, total greenhouse gas emissions were low from these Australian supply chains, and resource use was moderate. Expansion of the study to include additional regions and impact categories is recommended in future benchmarking studies.

Over the past three decades major changes have occurred in Australia's beef industry, affecting productivity and potentially the amount of resources used and environmental impacts from production. Using a life cycle assessment (LCA) approach with a ‘cradle-to-farm gate’ boundary the changes in greenhouse gas (GHG) emission intensity and key resource use efficiency factors (water use, fossil fuel energy demand and land occupation) are reported for the 30 years from 1981 to 2010, for the Australian beef industry. The analysis showed that over the three decades since 1981 there has been a decrease in GHG emission intensity (excluding land use change emissions) of 14% from 15.3 to 13.1 kg CO2-e/kg liveweight (LW). The improvement was largely due to efficiency gains through heavier slaughter weights, increases in growth rates in grass-fed cattle, improved survival rates and greater numbers of cattle being finished on grain. However, the increase in supplement and grain use on farms, and the increase in feedlot finishing, resulted in a twofold increase in fossil fuel energy demand for beef production over the same time. Fresh water consumption for beef production dropped to almost a third from 1465 L/kg LW in 1981 to 515 L/kg LW in 2010. Three contributing factors for this dramatic reduction in water use were: (i)an increase in the competitive demand for irrigation water, resulting in a transfer away from pasture for cattle to higher value industries such as horticulture, (ii) an initiative to cap free flowing artesian bores in the rangelands, and (iii) an overall decline in water available for agriculture compared to industrial and domestic uses. While there was higher uncertainty relating to estimates of land occupation and emissions from land use (LU) and direct land use change (dLUC), an inventory of land occupation indicated a decline in non-arable land occupation of about 19%, but a sevenfold increase in land occupation for feed production, albeit from a low base in 1981. GHG emissions associated with LU and dLUC for grazing were estimated to have declined by around 42% since 1981, due largely to legislated restrictions on broad-scale deforestation which were introduced progressively between 1996 and 2006. This paper discusses the prospects and challenges for further gains in resource use efficiency and reductions in greenhouse gas intensity for Australian beef production.

Sixty-four market-weight (130.0 +/- 0.65 kg) barrows (n = 16) and gilts (n = 48) were used in a split-plot design with a 2 x 2 x 2 factorial arrangement of treatments: 1) handling intensity (gentle vs. aggressive), 2) transport floor space (0.39 vs. 0.49 m(2)/pig), and 3) distance moved during handling (25 vs. 125 m) to determine the effects of multiple concurrent stressors on metabolic responses. For the handling intensity treatment, pigs were moved individually approximately 50 m through a handling course with either 0 (gentle) or 8 (aggressive) shocks from an electric goad. Pigs were loaded onto a trailer and transported for approximately 1 h at floor spaces of either 0.39 or 0.49 m(2)/pig. After transport, pigs were unloaded, and the distance moved treatment was applied; pigs were moved 25 or 125 m through a handling course using livestock paddles. Rectal temperature was measured, and blood samples (to measure blood acid-base status) were collected 2 h before the handling intensity treatment was applied and immediately after the distance moved treatment was applied. A LM sample to measure glycolytic potential was collected after the distance moved treatments on a subset of 32 pigs. There were handling intensity x distance moved interactions (P < 0.05) for several blood acid-base measurements. In general, there was no effect of distance moved on these traits when pigs were previously handled gently. However, when pigs were previously handled aggressively, pigs moved 125 compared with 25 m had greater (P < 0.05) blood lactate and less (P < 0.05) blood pH, bicarbonate, and base-excess. Pigs transported at 0.39 compared with 0.49 m(2)/pig had a greater (P < 0.01) increase in creatine kinase values; however, transport floor space did not affect any other measurements. Effects of distance moved during loading and floor space on the trailer during transport on the incidence of transport losses (dead and nonambulatory pigs) on arrival at the packing plant were evaluated in a study involving 42 loads of pigs (average BW = 131.2 kg, SD 5.05). A split-plot design was used with a 2 x 6 factorial arrangement of the following treatments: 1) distance moved from the pen to the exit of the building [short (0 to 30.5 m) vs. long (61.0 to 91.4 m)] and 2) transport floor space (0.396, 0.415, 0.437, 0.462, 0.489, or 0.520 m(2)/pig). Loading distance treatments (sub-plots) were compared within transport floor space treatments (main plot). Pigs were loaded at the farm using sorting boards and, if necessary, electric goads, transported approximately 3 h to a commercial packing plant and unloaded using livestock paddles. The number of nonambulatory pigs during loading and the number of dead and nonambulatory pigs at the plant were recorded. Nonambulatory pigs were classified as fatigued, injured, or injured and fatigued. In addition, the incidence of pigs exhibiting signs of stress (open-mouth breathing, skin discoloration, and muscle tremors) during loading and unloading was recorded. There were no interactions (P > 0.05) between distance moved and transport floor space treatments. Moving pigs long compared with short distances during loading increased (P < 0.001) the incidence of open-mouth breathing after loading (24.9 vs. 11.0 +/- 1.03%, respectively) and tended to increase the incidence of nonambulatory pigs during loading (0.32 vs. 0.08 +/- 0.09%, respectively; P = 0.09) and of nonambulatory, injured pigs at the plant (0.24 vs. 0.04 +/- 0.07%, respectively; P = 0.06). However, distance moved did not affect other losses at the plant. Total losses at the plant were greater (P < 0.05) for the 3 lowest floor spaces compared with the 2 highest floor spaces, and pigs provided 0.462 m(2)/pig during transport had similar transport losses to those provided 0.489 and 0.520 m(2)/pig (total losses at the plant = 2.84, 1.88, 1.87, 0.98, 0.13, and 0.98 +/- 0.43% of pigs transported, for 0.396, 0.415, 0.437, 0.462, 0.489, and 0.520 m(2)/pig, respectively). These data confirm previous findings that transport floor space has a major effect on transport losses and suggest that these losses are minimized at a floor space of 0.462 m(2)/pig or greater.

Utilisation of water, energy and land resources is under pressure globally because of increased demand for food, fibre and fuel production. Australian pork production utilises these resources both directly to grow and process pigs, and indirectly via the consumption of feed and other inputs. With increasing demand and higher costs associated with these resources, supply chain efficiency is a growing priority for the industry. This study aimed to quantify fresh water consumption, stress-weighted water use, fossil fuel energy use and land occupation from six case study supply chains and the national herd using a life cycle assessment approach. Two functional units were used: 1 kg of pork liveweight (LW) at the farm-gate, and 1 kg of wholesale pork (chilled, bone-in). At the farm-gate, fresh water consumption from the case study supply chains ranged from 22.2 to 156.7 L/kg LW, with a national average value of 107.5 L/kg LW. Stress-weighted water use ranged from 6.6 to 167.5 L H2O-e /kg LW, with a national average value of 103.2 L H2O-e /kg LW. Fossil fuel energy demand ranged from 12.9 to 17.4 MJ/kg LW, with a national average value of 14.5 MJ/kg LW, and land occupation ranged from 10.9 to 16.1 m2/kg LW, with a national average value of 16.1 m2/kg LW and with arable land representing 97% to 99% of total land occupation. National average impacts associated with production of wholesale pork, including impacts from meat processing, were 184 ± 43 L fresh water consumption, 172 ± 53 L H2O-e stress-weighted water, 27 ± 2.6 MJ fossil fuel energy demand and 25.9 ± 5.5 m2 land/kg wholesale pork. Across all categories through to the wholesale product, resource use was highest from the production of feed inputs, indicating that improving feed conversion ratio is the most important production metric for reducing the resource use. Housing type and energy generation from manure management also influence resource use requirements and may offer improvement opportunities.

Background: Arthrodesis of the hip remains a viable treatment for severe unilateral arthritis after traumatic injury or infection in a young but otherwise healthy individual. The goal of the present study was to review the long-term clinical and radiographic results after conversion of a fused hip to a total hip arthroplasty and to identify the risk factors that would lead to a higher rate of failure. Methods: We performed a retrospective review of the charts and radiographs of 187 patients (208 hips) who had conversion of a fused hip to a total hip arthroplasty. The mean duration of follow-up after the conversion to total hip arthroplasty was 9.2 years (range, two to twenty-six years). Results: The mean age at time of the arthroplasty was fifty-one years. The mean time-interval between the arthrodesis and the conversion to a total hip arthroplasty was twenty-seven years. According to the information in the charts, at a mean duration of follow-up of 9.2 years after the total hip arthroplasty, 79% of hips were either pain-free or had minimal pain, 83% had good-to-excellent function, and 79% had good-to-excellent range of motion. Complications, which included fifteen nerve palsies, occurred in twenty-four hips. Twenty-eight hips had heterotopic ossification, but it was not associated with a recurrence of ankylosis or a marked reduction of motion. Revision arthroplasty was performed in twelve hips. The probability of survival of the implant was 96.1% (95% confidence interval, 91.5% to 98.2%) at ten years, 89.9% (95% confidence interval, 85.3% to 96.1%) at fifteen years, and 72.8% (95% confidence interval, 36% to 90.6%) at twenty-six years. Conclusions: Conversion of a fused hip to a total hip arthroplasty has a favorable outcome. However, the technically demanding nature of the procedure should not be underestimated. Patients should be cautioned with regard to the possibility of a higher rate of complications than that seen with primary total hip arthroplasty.

Volatile anaesthetic agents potentiate neuromuscular blocking agents and retard their rate of reversal. We hypothesised that there was a difference in the rate of reversal of rocuronium-induced neuromuscular blockade based on the selection of inhalation agent. Thirty-eight patients undergoing elective surgical procedures received either sevoflurane or isoflurane, by random allocation. Neuromuscular blockade was induced using rocuronium 0.6 mg.kg-1 followed by continuous intravenous infusion to maintain 90% suppression of the single twitch response. Upon completion of surgery, the rocuronium infusion was discontinued, neostigmine 50 microg.kg-1 and glycopyrrolate 10 microg.kg-1 were administered. Times from reversal to T1 = 25, 50 and 60% and train-of-four ratio = 0.6 were recorded. The mean (SD) times to train-of-four ratio = 0.6 in the isoflurane and sevoflurane groups were 327 (132) and 351 (127) s, respectively. The mean (SD) times to single twitch response T1 = 25, 50 and 60% in the isoflurane group were 81 (33), 161 (59) and 245 (84) s, respectively, and in the sevoflurane group were 95 (35), 203 (88) and 252 (127) s, respectively. It is concluded that reversal of rocuronium-induced neuromuscular blockade is similar during isoflurane and sevoflurane anaesthesia.

The glycosylphosphatidylinositol-linked platelet protein CD109 carries the biallelic alloantigen system Gov. There is limited information on the incidence of Gov alloantibodies in neonatal alloimmune thrombocytopenia (NAITP), post-transfusion purpura (PTP) and platelet refractoriness. We adapted the monoclonal antibody-specific immobilization of platelet antigens (MAIPA) assay to the detection of Gov antibodies and determined their incidence in 605 archived samples (112 with HPA antibodies) referred for the aforementioned conditions. Here, we show that CD109 expression was reduced upon platelet storage in saline or by cryopreservation, but was stable when stored as whole blood or therapeutic platelet concentrate. Fourteen of the 605 samples contained Gov alloantibodies (anti-Gova, n = 10; anti-Govb, n = 4), with the majority in platelet refractoriness (n = 9) and, of the remaining five, four in NAITP and one in PTP. In seven cases, no other HPA antibodies were detected, three being NAITP cases. The incidence of Gov antibodies was significantly lower than HPA-1 system antibodies (n = 87), but equalled the number of HPA-5 system antibodies (n = 14) and outnumbered HPA-2 and -3 system antibodies (10 altogether).

Precise studies of sea-level rise require accurate calibrations to avoid misinterpretation of any spurious drift. Calibrations of altimeters can be undertaken in the relative sense for concurrent missions or independently against in situ data. In this study tide gauge enhanced repeat pass data is used to investigate non-oceanic signatures within TOPEX/Poseidon and ERS-1 with the individual time series cross-calibrated by inclusion of tide gauge enhanced dual satellite crossovers. The procedure is general in the sense that non-concurrent altimetric missions may be inter-calibrated, the only requirement being well calibrated tide gauge time series spanning the operational lifetimes of the two satellites.

Studies of sea level rise or ice sheet deformation from altimeter missions require careful calibration if systematic errors are not to be aliased into the result. For satellite missions that have distinct phases or when the repeat period is too long for repeat pass studies, altimetric crossover differences provide the mechanism for examination of residual systematic effects. In this study, altimetry, augmented with sea-surface variability time series as provided by the global network of tide gauges, is used to examine the stability of the ERS 1 range bias from April 1992 to August 1995. This 3-year period covered the first multidisciplinary phase, the geodetic phase, and the first four cycles of the second multidisciplinary phase. A correlation analysis using TOPEX/Poseidon and ERS 1 was first employed to identify a subset of island tide gauges where the time series is representative of that observed by the altimeter in the open ocean. Tide gauge augmented crossovers and all available repeat pass data were analyzed in the vicinity of these gauges. In particular, crossovers linking the ERS 1 multidisciplinary and geodetic phases permitted the range bias to be monitored continuously. Dual-crossover residuals between ERS 1 and TOPEX/Poseidon were utilized to gain insight into the characteristics of the temporal variation in the long-wavelength, geographically correlated error for the 3-year period. Failure to incorporate this geographically correlated error over the 3-year period led to a spurious drift of some 2–3 mm/yr. However, its inclusion reduces the drift to the extent that the observed rate is not statistically different from zero.

Two hundred and thirty-two total hip replacements with cement were performed between 1965 and 1987 in 190 patients who had osteoarthrosis secondary to acetabular dysplasia resulting from congenital dysplasia or dislocation of the hip. Forty-nine patients (fifty hips) who died or who did not have at least three years of clinical and radiographic follow-up were excluded; therefore, the study comprised 141 patients (182 hips), 125 of whom were women and sixteen of whom were men. The mean age at the time of the operation was 42.5 years (range, 19.5 to 76.5 years), and the mean duration of follow-up was 9.9 years (range, 3.1 to 22.8 years). A Charnley low-friction prosthesis with a 22.25-millimeter femoral head was used in all hips. A trochanteric osteotomy was performed in 164 hips. A direct lateral approach was used only when there was a limb-length discrepancy of less than two centimeters. Bulk autogenous graft was not used to augment the fixation of the acetabular component. The hips were divided into two groups on the basis of the degree of subluxation or dislocation according to the classification of Crowe et al. One hundred and thirty-six hips were grade I, II, or III (indicating subluxation) (Group S), and forty-six hips were grade IV (indicating dislocation) (Group D). At the most recent follow-up evaluation, 128 (94 per cent) of the 136 hips in Group S and forty-four (96 per cent) of the forty-six hips in Group D caused no or only slight pain (a score of 6 or 5 points, respectively, according to the system of Merle d'Aubigné and Postel). Nineteen cups (10 per cent) and five stems (3 per cent) failed and were revised. The rate of revision for loosening of the femoral component in Group D (2 per cent [one stem]) was similar to that in Group S (3 per cent [four stems]). In comparison, the rate of revision of the acetabular component in Group D (15 per cent [seven cups]) was almost twice that in Group S (9 per cent [twelve cups]). This study demonstrated satisfactory clinical results at a mean of nearly ten years. The fixation of the femoral components appeared to be satisfactory; however, the failure of the fixation of the acetabular components in Group D (the dislocated hips) is a cause for concern.

Derivatized bovine adenosine deaminase is used in enzyme replacement therapy and as an adjunct to gene therapy against severe combined immunodeficiency syndrome. Although a gene sequence is known for human adenosine deaminase, the structure of the bovine enzyme has not been characterized. Structure studies using mass spectrometry are reported here that evaluate sequence, processing, post-translational modifications and the extent of homology between the human protein and its therapeutic surrogate.

A calibration of the TOPEX/POSEIDON and ERS 1 altimeters has been undertaken using altimetry across the English Channel. Data from the tide gauge installed at Newhaven and the satellite laser ranger at Herstmonceux were used in conjunction with local models for the geoid, ocean tide, and storm surge to generate bias values at each subsatellite altimetry point. A range bias value of −17.3 cm was generated for the NASA TOPEX altimeter relative to the four-parameter sea state bias model calculated by Gaspar et al. [1994]. The corresponding value for the Centre National d'Etudes Spatiales (CNES) POSEIDON solid state altimeter was −0.3 cm. The impact of other parameterizations for the sea state bias on the altimeter bias is studied. The derived results are in close agreement with results from the NASA calibration campaign at Harvest oil platform near California and the CNES experiment at the Italian Lampedusa island. The bias derived for ERS 1 was −39.3 cm relative to an Aston-derived single-parameter sea state bias model.

Grain finishing of cattle has become increasingly common in Australia over the past 30 years. However, interest in the associated environmental impacts and resource use is increasing and requires detailed analysis. In this study we conducted a life cycle assessment (LCA) to investigate impacts of the grain-finishing stage for cattle in seven feedlots in eastern Australia, with a particular focus on the feedlot stage, including the impacts from producing the ration, feedlot operations, transport, and livestock emissions while cattle are in the feedlot (gate-to-gate). The functional unit was 1 kg of liveweight gain (LWG) for the feedlot stage and results are included for the full supply chain (cradle-to-gate), reported per kilogram of liveweight (LW) at the point of slaughter. Three classes of cattle produced for different markets were studied: short-fed domestic market (55–80 days on feed), mid-fed export (108–164 days on feed) and long-fed export (>300 days on feed). In the feedlot stage, mean fresh water consumption was found to vary from 171.9 to 672.6 L/kg LWG and mean stress-weighted water use ranged from 100.9 to 193.2 water stress index eq. L/kg LWG. Irrigation contributed 57–91% of total fresh water consumption with differences mainly related to the availability of irrigation water near the feedlot and the use of irrigated feed inputs in rations. Mean fossil energy demand ranged from 16.5 to 34.2 MJ lower heating values/kg LWG and arable land occupation from 18.7 to 40.5 m2/kg LWG in the feedlot stage. Mean greenhouse gas (GHG) emissions in the feedlot stage ranged from 4.6 to 9.5 kg CO2-e/kg LWG (excluding land use and direct land-use change emissions). Emissions were dominated by enteric methane and contributions from the production, transport and milling of feed inputs. Linear regression analysis showed that the feed conversion ratio was able to explain >86% of the variation in GHG intensity and energy demand. The feedlot stage contributed between 26% and 44% of total slaughter weight for the classes of cattle fed, whereas the contribution of this phase to resource use varied from 4% to 96% showing impacts from the finishing phase varied considerably, compared with the breeding and backgrounding. GHG emissions and total land occupation per kilogram of LWG during the grain finishing phase were lower than emissions from breeding and backgrounding, resulting in lower life-time emissions for grain-finished cattle compared with grass finishing.

We compared the outcome of total knee arthroplasty in 19 patients who had had previous patellectomy with the results in a matched series of arthroplasties performed on knees in which the patella was intact. The mean follow-up was 63 months (21 to 114). In the study group, the outcome was poor in five patients. There was instability in the coronal plane in three patients and persistent pain in four. Three supracondylar fractures occurred. The overall complication rate was 36%. In the control group, pain was relieved in every case and there were no complications. Total knee arthroplasty has a higher complication rate and inferior results if the knee has undergone prior patellectomy.

The sarcoplasmic reticulum of rabbit skeletal muscle contains a small “proteolipid,” i.e., a protein which is soluble in acidic CHCl 3 CH 3 OH . We propose the name sarcolipin for this small protein, to signify its lipid-like solubility and association with the sarcoplasmic reticulum. We have determined the following amino acid sequence for sarcolipin, using protein chemistry methods: M E R S T R E L C L N F T V V L I T V I L I W L L V R S Y Q Y. This 31-residue sequence includes a 19-residue hydrophobic segment which probably spans the sarcoplasmic reticulum membrane. The molecular weight calculated from the sequence, 3733, agrees with that measured by fast atom bombardment mass spectrometry, showing that sarcolipin contains no attached fatty acyl or other prosthetic groups.

Cadmium-induced metallothionein in a mollusc, the oyster Crassostrea virginica, occurs in both blocked and unblocked forms (Roesijadi, G., Kielland, S.L. and Klerks, P. (1989) Arch. Biochem. Biophys. 273, 403-413). The block, which is the sole difference in the structure of the two proteins, was identified as an acetyl group with use of tandem mass spectrometry. The blocked and unblocked proteins carried N-acetylserine and serine, respectively, at the N-terminus and were designated CvNAcMT and CvMT. Only CvNAcMT was detected under basal conditions. Both forms were induced by Cd. Pulse-labeling with [35S]cyteine at specified times during exposure showed that the rate of CvNAcMT synthesis in gills increased rapidly, initially exceeding that of CvMT, then declined to the rate attained by CvMT. Turnover rates for Cd-induced CvMT and CvNAcMT were similar to each other. They appeared to be faster when measured in the absence of Cd in the external medium (k = 0.18 and 0.16/day, respectively), than in its presence (k = 0.03 and 0.06/day, respectively).

This study evaluated effects of trailer design and season on physical indicators of stress during loading and unloading and transport losses (dead and nonambulatory pigs) in market-weight pigs (BW = 129.6 +/- 0.40 kg). A total of 109 trailer loads of pigs (n = 17,256 pigs) from 1 farm were used in a randomized complete block design with a 2 x 4 factorial arrangement of treatments: 1) trailer design (potbelly vs. straight-deck) and 2) season (spring vs. summer vs. fall vs. winter). A subset of loads (n = 42) was used to examine effect of distance pigs were moved during loading [short (24 m) vs. long (47 to 67 m)] on physical indicators of stress and transport losses. This study was conducted on 7 d per season at 1 farm with 4 loads (2 on potbelly and 2 on straight-deck trailers) being transported each day to 1 commercial packing plant. Pigs from different farm groups were mixed on the trailer and provided with 0.45 m(2)/pig floor space during an approximately 4-h journey to the plant. The percentage of pigs exhibiting open-mouth breathing, skin discoloration, and muscle tremors was recorded during loading and unloading. Additionally, dead pigs on arrival at the plant and nonambulatory pigs at the farm and at the plant were recorded. Effects of trailer design on open-mouth breathing and skin discoloration during unloading were dependent on season (trailer design x season interaction; P0.05). Pigs unloaded from potbelly trailers had a greater (Por = 0.05) incidence of open-mouth breathing in the spring and summer and a greater (P0.05) incidence of skin discoloration in the spring, summer, and winter than pigs unloaded from straight-deck trailers. The incidence of total nonambulatory pigs at the plant was greater (P0.05) in the winter than in the spring and summer. The long compared with short distance moved treatment resulted in a greater (P = 0.001) incidence of open-mouth breathing and skin discoloration during loading and tended (P = 0.06) to increase the incidence of nonambulatory pigs at the farm. However, there was no effect of trailer design, season, or loading distance on total losses at the plant. In summary, physical indicators of stress (open-mouth breathing and skin discoloration) were increased with the long distance moved during loading treatment and were greater during unloading for potbelly than straight-deck trailers; however, trailer design, season, and loading distance had minimal effects on total transport losses.

Transportation has been recognized as an important aspect that affects meat quality and profit in pork production. The objective of this study was to measure the microenvironment surrounding the animals in the transport trailer through seasonal testing. Fourteen tests were performed over three seasons (hot, mild and cold) to determine seasonal trends in the microenvironment of the animals during transport, with exposure to temperature extremes. A total of 156 market-weight pigs were loaded onto a straight, dual-deck semi-trailer for each trip. Results showed that the average compartment temperatures were often higher than external temperature before traveling, but generally cooled down to be consistent with external temperature during transport, because of relative air velocity. Average compartment air velocity was highly correlated to truck travel speed. Skin surface temperatures of pigs were highest while waiting at the farm after loading and decreased during transport. Carbon dioxide levels were higher in compartments towards the front of the trailer, indicating that ventilating air entered the rear of the trailer and exited out the front. The data collected in this study is important to evaluate the animal microenvironment during transport. These data can also be used to help develop a simulation model to improve trailer designs, reduce animal stress level and reduce economic loss from transport losses and optimize the practice of animal loading and transporting.

Greenhouse gas (GHG) and ammonia emissions are important environmental impacts from meat chicken houses. This study measured ammonia (NH3), nitrous oxide (N2O) and methane (CH4) in two trials from paired, commercial meat chicken houses using standard (control) and mitigation strategies. In Trial 1, emissions from houses with standard litter depth of 47 mm (LD47) or increased litter depth of 67 mm (LD67) were compared. When standardised to a 42-day-old bird, emissions were 11.9 g NH3/bird, 0.30 g N2O/bird and 0.16 g CH4/bird from the LD47 and 11.7 g NH3/bird, 0.69 g N2O/bird and 0.12 g CH4/bird from the LD67. Emissions per kilogram of manure N were 0.14 and 0.11 for NH3-N, 0.003 and 0.005 N2O-N and CH4 conversion factors were 0.08% and 0.05%. Total direct and indirect GHG emissions reported in carbon dioxide equivalents were found to be higher in LD67 in response to the elevated direct N2O emissions. Trial 2 compared the impact of reduced crude protein (CP19.8) and a standard diet (CP21.3) developed using least-cost ration formulation, on emissions. Emissions per bird for the CP19.8 diet were 7.7 g NH3/bird, 0.39 g N2O/bird and 0.14 g CH4/bird, while emissions from birds fed the CP21.3 diet were 10.6 g NH3/bird, 0.42 g N2O/bird and 0.19 g CH4/bird. Significant differences were observed only in the NH3 results, where emissions were reduced by 27% for the low-CP diet. Because of the low emission levels, total mitigation potential from indirect GHG emissions was relatively small in Trial 2, corresponding to 11 t carbon dioxide equivalents/year per million birds.

This study conducted a life cycle assessment (LCA) investigating energy, land occupation, greenhouse gas (GHG) emissions, fresh water consumption and stress-weighted water use from production of export lamb in the major production regions of New South Wales, Victoria and South Australia. The study used data from regional datasets and case study farms, and applied new methods for assessing water use using detailed farm water balances and water stress weighting. Land occupation was assessed with reference to the proportion of arable and non-arable land and allocation of liveweight (LW) and greasy wool was handled using a protein mass method. Fossil fuel energy demand ranged from 2.5 to 7.0 MJ/kg LW, fresh water consumption from 58.1 to 238.9 L/kg LW, stress-weighted water use from 2.9 to 137.8 L H2O-e/kg LW and crop land occupation from 0.2 to 2.0 m2/kg LW. Fossil fuel energy demand was dominated by on-farm energy demand, and differed between regions and datasets in response to production intensity and the use of purchased inputs such as fertiliser. Regional fresh water consumption was dominated by irrigation water use and losses from farm water supply, with smaller contributions from livestock drinking water. GHG emissions ranged from 6.1 to 7.3 kg CO2-e/kg LW and additional removals or emissions from land use (due to cultivation and fertilisation) and direct land-use change (due to deforestation over previous 20 years) were found to be modest, contributing between –1.6 and 0.3 kg CO2-e/kg LW for different scenarios assessing soil carbon flux. Excluding land use and direct land-use change, enteric CH4 contributed 83–89% of emissions, suggesting that emissions intensity can be reduced by focussing on flock production efficiency. Resource use and emissions were similar for export lamb production in the major production states of Australia, and GHG emissions were similar to other major global lamb producers. The results show impacts from lamb production on competitive resources to be low, as lamb production systems predominantly utilised non-arable land unsuited to alternative food production systems that rely on crop production, and water from regions with low water stress.

Mitigation of agricultural greenhouse gas emissions is a target area for the Australian Government and the pork industry. The present study measured methane (CH4), nitrous oxide (N2O) and ammonia (NH3) from a deep-litter piggery and litter stockpile over two trials in southern New South Wales, to compare emissions from housing pigs on deep litter with those of pigs from conventional housing with uncovered anaerobic effluent-treatment ponds. Emissions were measured using open-path Fourier transform infrared spectrometry, in conjunction with a backward Lagrangian stochastic model. Manure excretion was determined by mass balance and emission factors (EFs) were developed to report emissions relative to volatile solids and nitrogen (N) input. Nitrous oxide emissions per animal unit (1 AU = 500 kg liveweight) from deep-litter sheds were negligible in winter, and 8.4 g/AU.day in summer. Ammonia emissions were 39.1 in winter and 52.2 g/AU.day in summer, while CH4 emissions were 16.1 and 21.6 g/AU.day in winter and summer respectively. Emission factors averaged from summer and winter emissions showed a CH4 conversion factor of 3.6%, an NH3-N EF of 10% and a N2O-N EF of 0.01 kg N2O-N/kg N excreted. For the litter stockpile, the simple average of summer and winter showed an EF for NH3-N of 14%, and a N2O-N EF of 0.02 kg N2O-N/kg-N of spent litter added to the stockpile. We observed a 66% and 80% decrease in emissions from the manure excreted in litter-based housing with litter stockpiling or without litter stockpiling, compared with conventional housing with an uncovered anaerobic effluent-treatment pond. This provides a sound basis for mitigation strategies that utilise litter-based housing as an alternative to conventional housing with uncovered anaerobic effluent-treatment ponds.

In the Australian pork industry, manure is the main source of greenhouse gases (GHG). In conventional production systems, effluent from sheds is transferred into open anaerobic ponds where the effluent is typically stored for many months, with the potential of generating large quantities of GHG. The present study measured methane (CH4), nitrous oxide (N2O) and ammonia (NH3) emissions from a conventional anaerobic effluent pond (control), a short hydraulic retention-time tank (short HRT, mitigation) and from the animal housing for a flushing piggery in south-eastern Queensland, over two 30-day trials during summer and winter. Emissions were compared to determine the potential for a short HRT to reduce emissions. Average CH4 emissions from the pond were 452 ± 37 g per animal unit (AU; 1 AU = 500 kg liveweight) per day, during the winter trial and 789 ± 29 g/AU.day during the summer trial. Average NH3 emissions were 73 ± 8 g/AU.day during the winter trial and 313 ± 18 g/AU.day during the summer trial. High emission factors during summer will be temperature driven and influenced by the residual volatile solids and nitrogen (N) deposited in the pond during winter. Average NH3 emissions from the piggery shed were 0.707 ± 0.050 g/AU.day and CH4 emissions were 0.344 ± 0.116 g/AU.day. The N2O concentrations from both the pond and shed were close to, or below, the detection limits. Total emissions from the short HRT during the winter and summer trials, respectively, were as follows: CH4 10.65 ± 0.616 mg/AU.day and 4108 ± 473 mg/AU.day; NH3-N 1.15 ± 0.07 mg/AU.day and 29.8 ± 2.57 mg/AU.day; N2O-N 0.001 ± 0.00052 mg/AU.day and 5.9 ± 0.321 mg/AU.day. On the basis of a conservative analysis of CH4 emissions relative to the inflow of volatile solids, and NH3 and N2O emissions as a fraction of the excreted N, GHG emissions were found to be 79% lower from the short-HRT system. This system provides a potential mitigation option to reduce GHG emissions from conventional pork production in Australia.

Agricultural industries are under increasing pressure to measure and reduce greenhouse gas emissions from the supply chain. The Australian pork industry has established proactive goals to improve greenhouse-gas (GHG) performance across the industry, but while productivity indicators are benchmarked by industry, similar data have not previously been collected to determine supply chain GHG emissions. To assess total GHG emissions from Australian pork production, the present study conducted a life-cycle assessment of six case study supply chains and the national herd for the year 2010. The study aimed to determine total GHG emissions and hotspots, and to determine the mitigation potential from alternative manure treatment systems. Two functional units were used: 1 kg of pork liveweight (LW) at the farm gate, and 1 kg of wholesale pork (chilled, bone-in) ready for packaging and distribution. Mean GHG emissions from the case study supply chains ranged from 2.1 to 4.5 kg CO2-e/kg LW (excluding land-use (LU) and direct land use-change (dLUC) emissions). Emissions were lowest from the piggeries that housed grower-finisher pigs on deep litter and highest from pigs housed in conventional systems with uncovered anaerobic effluent ponds. Mean contribution from methane from effluent treatment was 64% of total GHG at the conventional piggeries. Nitrous oxide arose from both grain production and manure management, comprising 7–33% of the total emissions. The GHG emissions for the national herd were 3.6 kg CO2-e/kg LW, with the largest determining factor on total emissions being the relative proportion of pigs managed with high or low emission manure management systems. Emissions from LU and dLUC sources ranged from 0.08 to 0.7 kg CO2-e/kg LW for the case study farms, with differences associated with the inclusion rate of imported soybean meal in the ration and feed-conversion ratio. GHG intensity (excluding LU, dLUC) from the national herd was 6.36 ± 1.03 kg CO2-e/kg wholesale pork, with the emission profile dominated by methane from manure management (50%), followed by feed production (27%) and then meat processing (8%). Inclusion of LU and dLUC emissions had a minor effect on the emission profile. Scenarios testing showed that biogas capture from anaerobic digestion with combined heat and power generation resulted in a 31–64% reduction in GHG emissions. Finishing pigs on deep litter as preferred to conventional housing resulted in 38% lower GHG emissions than conventional finishing.

Greenhouse gas and ammonia emissions are important environmental impacts from manure management in the layer-hen industry. The present study aimed to quantify emissions of nitrous oxide (N2O), methane (CH4) and ammonia (NH3) from layer-hen manure stockpiles, and assess the use of an impermeable cover as an option to mitigate emissions. Gaseous emissions of N2O, CH4 and NH3 were measured using open-path FTIR spectroscopy and the emission strengths were inferred using a backward Lagrangian stochastic model. Emission factors were calculated from the relationship between gaseous emissions and stockpile inputs over a 32-day measurement period. Total NH3 emissions were 5.97 ± 0.399 kg/t (control) and 0.732 ± 0.116 kg/t (mitigation), representing an 88% reduction due to mitigation. Total CH4 emissions from the mitigation stockpile were 0.0832 ± 0.0198 kg/t. Methane emissions from the control and N2O emissions (control and mitigation) were below detection. The mass of each stockpile was 27 820 kg (control) and 25 120 kg (mitigation), with a surface area of ~68 m2 and a volume of ~19 m3. Total manure nitrogen (N) and volatile solids (VS) were 25.2 and 25.8 kg/t N, and 139 and 106 kg/t VS for the control and mitigation stockpiles respectively. Emission factors for NH3 were 24% and 3% of total N for the control and mitigation respectively. Methane from the mitigation stockpile had a CH4 conversion factor of 0.3%. The stockpile cover was found to reduce greenhouse gas emissions by 74% compared with the control treatment, primarily via reduced NH3 and associated indirect N2O emissions.

Spectra were recorded of all fragment ions formed by elimination of 28-u neutral fragments in fast atom bombardment spectra of peptides in the mass range 1000–1800 u. This approach can provide less complex spectra than either conventionally scanned spectra or product ion scans from collisionally activated four-sector experiments, and spectra that contain information that is both overlapping and complementary to those from the other techniques. Constant neutral loss spectra may provide a reading frame for distinguishing sequence ion series in tandem or single analyzer spectra.

From these studies we conclude that endothermic ion-molecule reactions may be effective for the fragmentation of large peptides and/or as the first step in a neutralization/chemical reionization scheme in which the reverse (exothermic) reaction is used for reprotonation

Effects of distance moved during loading and floor space on the trailer during transport on the incidence of transport losses (dead and nonambulatory pigs) on arrival at the packing plant were evaluated in a study involving 42 loads of pigs (average BW = 131.2 kg, SD 5.05). A split-plot design was used with a 2 x 6 factorial arrangement of the following treatments: 1) distance moved from the pen to the exit of the building [short (0 to 30.5 m) vs. long (61.0 to 91.4 m)] and 2) transport floor space (0.396, 0.415, 0.437, 0.462, 0.489, or 0.520 m(2)/pig). Loading distance treatments (sub-plots) were compared within transport floor space treatments (main plot). Pigs were loaded at the farm using sorting boards and, if necessary, electric goads, transported approximately 3 h to a commercial packing plant and unloaded using livestock paddles. The number of nonambulatory pigs during loading and the number of dead and nonambulatory pigs at the plant were recorded. Nonambulatory pigs were classified as fatigued, injured, or injured and fatigued. In addition, the incidence of pigs exhibiting signs of stress (open-mouth breathing, skin discoloration, and muscle tremors) during loading and unloading was recorded. There were no interactions (P0.05) between distance moved and transport floor space treatments. Moving pigs long compared with short distances during loading increased (P0.001) the incidence of open-mouth breathing after loading (24.9 vs. 11.0 +/- 1.03%, respectively) and tended to increase the incidence of nonambulatory pigs during loading (0.32 vs. 0.08 +/- 0.09%, respectively; P = 0.09) and of nonambulatory, injured pigs at the plant (0.24 vs. 0.04 +/- 0.07%, respectively; P = 0.06). However, distance moved did not affect other losses at the plant. Total losses at the plant were greater (P0.05) for the 3 lowest floor spaces compared with the 2 highest floor spaces, and pigs provided 0.462 m(2)/pig during transport had similar transport losses to those provided 0.489 and 0.520 m(2)/pig (total losses at the plant = 2.84, 1.88, 1.87, 0.98, 0.13, and 0.98 +/- 0.43% of pigs transported, for 0.396, 0.415, 0.437, 0.462, 0.489, and 0.520 m(2)/pig, respectively). These data confirm previous findings that transport floor space has a major effect on transport losses and suggest that these losses are minimized at a floor space of 0.462 m(2)/pig or greater.

Arthrodesis of the hip remains a viable treatment for severe unilateral arthritis after traumatic injury or infection in a young but otherwise healthy individual. The goal of the present study was to review the long-term clinical and radiographic results after conversion of a fused hip to a total hip arthroplasty and to identify the risk factors that would lead to a higher rate of failure.We performed a retrospective review of the charts and radiographs of 187 patients (208 hips) who had conversion of a fused hip to a total hip arthroplasty. The mean duration of follow-up after the conversion to total hip arthroplasty was 9.2 years (range, two to twenty-six years).The mean age at time of the arthroplasty was fifty-one years. The mean time-interval between the arthrodesis and the conversion to a total hip arthroplasty was twenty-seven years. According to the information in the charts, at a mean duration of follow-up of 9.2 years after the total hip arthroplasty, 79% of hips were either pain-free or had minimal pain, 83% had good-to-excellent function, and 79% had good-to-excellent range of motion. Complications, which included fifteen nerve palsies, occurred in twenty-four hips. Twenty-eight hips had heterotopic ossification, but it was not associated with a recurrence of ankylosis or a marked reduction of motion. Revision arthroplasty was performed in twelve hips. The probability of survival of the implant was 96.1% (95% confidence interval, 91.5% to 98.2%) at ten years, 89.9% (95% confidence interval, 85.3% to 96.1%) at fifteen years, and 72.8% (95% confidence interval, 36% to 90.6%) at twenty-six years.Conversion of a fused hip to a total hip arthroplasty has a favorable outcome. However, the technically demanding nature of the procedure should not be underestimated. Patients should be cautioned with regard to the possibility of a higher rate of complications than that seen with primary total hip arthroplasty.

The results of cemented total hip arthroplasty (THA) in patients with ankylosing spondylitis were studied to determine the utility of THA for these patients. A total of 103 patients with ankylosing spondylitis underwent 181 THAs; 72 patients (69.9%) had bilateral surgery. The mean follow-up was 10.3 years. The mean age of patients at surgery was 47 years. Before surgery, 42 hips (23.2%) were ankylosed. Revision surgery was carried out in 25 hips (13.8%). Heterotopic ossification was present in 21 hips (11.6%); however, no patients had functional impairment or reankylosis. At final follow-up examination, 173 hips (96%) had an excellent (low) pain score, and 53 hips had a normal or near-normal function score (29.2%). The probability of survival of the implant was 71% at 27 years. THA provides long-term improvement in hip function for patients with ankylosing spondylitis. Copyright 2002, Elsevier Science (USA). Volatile anaesthetic agents potentiate neuromuscular blocking agents and retard their rate of reversal. We hypothesised that there was a difference in the rate of reversal of rocuronium-induced neuromuscular blockade based on the selection of inhalation agent. Thirty-eight patients undergoing elective surgical procedures received either sevoflurane or isoflurane, by random allocation. Neuromuscular blockade was induced using rocuronium 0.6 mg.kg-1 followed by continuous intravenous infusion to maintain 90% suppression of the single twitch response. Upon completion of surgery, the rocuronium infusion was discontinued, neostigmine 50 microg.kg-1 and glycopyrrolate 10 microg.kg-1 were administered. Times from reversal to T1 = 25, 50 and 60% and train-of-four ratio = 0.6 were recorded. The mean (SD) times to train-of-four ratio = 0.6 in the isoflurane and sevoflurane groups were 327 (132) and 351 (127) s, respectively. The mean (SD) times to single twitch response T1 = 25, 50 and 60% in the isoflurane group were 81 (33), 161 (59) and 245 (84) s, respectively, and in the sevoflurane group were 95 (35), 203 (88) and 252 (127) s, respectively. It is concluded that reversal of rocuronium-induced neuromuscular blockade is similar during isoflurane and sevoflurane anaesthesia.

The glycosylphosphatidylinositol-linked platelet protein CD109 carries the biallelic alloantigen system Gov. There is limited information on the incidence of Gov alloantibodies in neonatal alloimmune thrombocytopenia (NAITP), post-transfusion purpura (PTP) and platelet refractoriness. We adapted the monoclonal antibody-specific immobilization of platelet antigens (MAIPA) assay to the detection of Gov antibodies and determined their incidence in 605 archived samples (112 with HPA antibodies) referred for the aforementioned conditions. Here, we show that CD109 expression was reduced upon platelet storage in saline or by cryopreservation, but was stable when stored as whole blood or therapeutic platelet concentrate. Fourteen of the 605 samples contained Gov alloantibodies (anti-Gova, n = 10; anti-Govb, n = 4), with the majority in platelet refractoriness (n = 9) and, of the remaining five, four in NAITP and one in PTP. In seven cases, no other HPA antibodies were detected, three being NAITP cases. The incidence of Gov antibodies was significantly lower than HPA-1 system antibodies (n = 87), but equalled the number of HPA-5 system antibodies (n = 14) and outnumbered HPA-2 and -3 system antibodies (10 altogether).

We have determined the amino acid sequences of the three major isoforms of lobster troponin C, using protein chemistry methods. Each isoform contains 150 amino acid residues, with a calculated M r of 17,000 ± 100. Sequence differences occur in all regions of the polypeptides, indicating that the isoforms are products of three different genes, rather than alternative transcripts of a single gene. Analysis of the sequences predicts that functional Ca 2+ -binding sites are present only in regions II and IV of four ancestral regions.

Initial investigations of a multifactorial approach to the measurement of emotional abuse in dating relationships are presented. A set of 54 items was generated to assess 4 rationally derived subscales measuring Restrictive Engulfment, Hostile Withdrawal, Denigration, and Dominance/Intimidation. An exploratory factor analysis on reports of partner behavior by 157 female undergraduate students in dating relationships provided support for the hypothesized subscales. Denigration and Dominance/Intimidation had consistently higher correlations with physical aggression than did the other two forms of emotional abuse. Further evidence for discriminant and convergent validity was apparent in correlations with the circumplex scales of the Inventory of Interpersonal Problems, and with self-reported attachment variables. The results support the assessment of emotional abuse in dating relationships as a multifactorial construct.

To explore opinions about the OCN credential, the ways in which it was obtained and retained, and the extent to which it is valued by employers.A descriptive comparison study using a cross-sectional survey design.Questionnaires were mailed to a nationwide sample of 2,429 RN members of the Oncology Nursing Society; 1,217 (50%) surveys were returned. The majority of respondents were female, 30-49 years of age. Caucasian, and had practiced nursing for more than 11 years.Certification status, work role characteristics, preparation strategies for the certification examination, and motivation for obtaining certification.Oncology nurses recognize the importance and value of OCN certification. The primary reasons oncology nurses obtain and retain certification include the desire for personal achievement, professional growth, and development. OCNs were more likely to work in a setting where the employer supports professional development through continuing nursing education.Because health care is increasingly delivered in ambulatory/home settings and the population is aging, oncology certification needs to be encouraged among nurses who work in these settings or with geriatric populations. Certified nurses tended to experience more job satisfaction than noncertified nurses.

To conduct a role delineation study of advanced oncology nursing practice as a basis for the blueprint for the Advanced Oncology Certified Nurse (AOCN) Examination.Descriptive and comparative.A group of 802 Oncology Nursing Society members comprised of 258 oncology nurse practitioners (NPs), 235 oncology clinical nurse specialists (CNSs), and 309 baccalaureate-prepared oncology nurses.A pilot survey was mailed to a small group to allow refinement of the survey instrument. The survey then was mailed to a total sample of 2,400.Frequency and importance to practice of 121 survey items as well as whether each item reflected an entry-level skill. Survey items were divided into five major domains: (a) Direct Caregiver, (b) Consultant, (c) Administrator/Coordinator, (d) Researcher and (e) Educator.Direct Caregiver domain was weighted most heavily (63%), followed by Educator (18%), Consultant (10%), Administrator/Coordinator (5%), and Researcher (4%). Most of the items were identified as entry-level skills, and fewer than 10% of the items differentiated between CNS and NP respondents.The blueprint for the AOCN Examination reflects entry-level advanced practice oncology nursing and is appropriate for both CNSs and NPs in oncology.Examinees, consumers, and employers can have confidence that the AOCN Examination will be based on a blueprint that was revised to reflect current oncology nursing practice and, therefore, is valid for its purpose.

This study investigated relations between lifetime exposure to interparental aggression and (1) anxiety, depression, and anger; (2) interpersonal problems; and (3) symptoms of trauma, in a sample of young adults. Participants were 326 (219 female, 107 male) undergraduates from an eastern university. Participants reported on their demographic backgrounds, memories of interparental verbal and physical aggression, past experiences of severe stressors (including child physical and sexual abuse and community violence), and recent symptoms. Results demonstrate that exposure to interparental violence is associated with elevations in depression, anxiety, interpersonal problems, and trauma symptoms. Further analyses indicated that interparental verbal aggression predicted all symptom areas and was a stronger predictor than interparental violence. The impact of both forms of aggression was attenuated and in some cases negated by controlling for the effects of other severe stressors.

We analysed in-vivo migration and wear over a long period of all-polyethylene acetabular cups which had not been affected by mechanical loosening. The selection criteria of regular radiological follow-up, good clinical outcome (Charnley score of 5 or 6), continued walking without crutches and no radiological signs of loosening of the acetabular cups were fulfilled by 25 Charnley total hip arthroplasties. Mean migration, measured by the Nunn method, was 0.6 mm in the medial and 0.2 mm in the cranial direction. The mean yearly rate of wear was 0.05 mm and 0.04 mm, with six and two cups having no detectable wear, as measured by the Livermore and Charnley-Cupic methods, respectively. The maximal detected wear was 3.7 mm. There were no changes in the rate of wear with time. Computerised Ein Bild Röntgen Analyse (single-image radiological analysis) measurements of 20 hips indicated plastic deformation of the cups. We conclude that long-term successful cups do not migrate and have a very low rate of wear which was not affected by ageing of the polyethylene. There was no evidence that polyethylene wear alone caused mechanical loosening of the cup but high rates of wear seem to have an adverse prognostic value in terms of the long-term survival of the prosthesis.

One hundred sixty-one dislocations after cemented total hip arthroplasty, with a mean follow-up period of 8 years after dislocation, were reviewed with the aim of establishing the prognosis. There were 84% single and 16% recurrent dislocations. Closed reduction was successful in 81% of cases. Thirty-seven percent of dislocations were early (within 5 weeks), 36% occurred in patients who had had previous surgery, and in 47% there was nonunion of the trochanter. There was a two-way interaction between these factors, and all factors were significant for recurrent dislocation. Twenty-six (16%) recurrent dislocations required surgery. The most common causes of recurrent dislocation demonstrated at operation were component malposition (58%) and failure of the abductor mechanism (42%). In total, 96% of cases were successfully treated.

The cell nucleus plays an essential role in all aspects of cell function, including DNA replication, gene transcription, RNA processing and cell division. Within the cell nucleus there are many proteins and peptides that have regulatory roles. Although several higher molecular weight protein components (30 kDa) have been identified and characterized, limited information is available for the lower molecular weight components. Nuclei from human peripheral blood lymphocytes were isolated and rapidly characterized by employing a strategy using reversed-phase high-performance liquid chromatography, tryptic digestion, and post-source decay analysis of matrix-assisted laser desorption/ionization (MALDI) ions in combination with database searches. Database searches utilizing molecular weight, proteolytic digest fragments, and peptide sequence results identified known proteins. The results illustrate the usefulness of MALDI as a tool in the characterization of low abundance proteins in the cell nucleus.
