Flacourtia montana J. Graham wild edible fruit tree, endemic to the Western Ghats, India was found infected with leaf spot disease. Based on morphological characteristics, molecular analyses (ITS and LSU) and pathogenicity, the pathogen was identified as Lasiodiplodia theobromae (Pat.) Griffon & Maubl. (Botryosphaeriaceae).This is the first report of L. theobromae causing leaf spots on F. montana from Western Ghats, India.

We study the feasibility of volume imaging from a few angular views/scans in a light sheet fluorescence microscopy. Two-dimensional (2D) images (angular views) were acquired at an angular separation of 108 and volume images were constructed with merely 18, 9, and 6 views. We study the structural changes in a 5-day old Zebrafish embryo labeled with Phalloidin TRITC that binds to F-Actin of embryo cell. To collect the data, the specimen is rotated (for varying sampling angles Du) with respect to a fixed vertical axis passing through the volume-of-interest (yolk sac). In the proposed realization of selective plane illumination microscopy (SPIM) technique, the translation is completely avoided. Analysis shows rich structural information with marginal reduction in contrast. Comparison with the state-of-the-art SPIM shows appreciable volume reconstruction (from an order less 2D scans) that may be good enough for rapid monitoring of macroscopic specimens. Here we present a light sheet based imaging flow cytometry system to image 2D cross sections of the sample (HeLa cells) and these 2D sections in turn can be stitched together to form 3D image. The images are corrected for motion blur using Maximum a Posteriori algorithm(MAP).

We propose and demonstrate a light-sheet-based 3D interrogation system on a microfluidic platform for screening biological specimens during flow. To achieve this, a diffraction-limited light-sheet (with a large field-of-view) is employed to optically section the specimens flowing through the microfluidic channel. This necessitates optimization of the parameters for the illumination sub-system (illumination intensity, light-sheet width, and thickness), microfluidic specimen platform (channel-width and flow-rate), and detection sub-system (camera exposure time and frame rate). Once optimized, these parameters facilitate cross-scctional imaging and 3D reconstruction of biological specimens. The proposed integrated light-sheet imaging and flow-based enquiry (iLIFE) imaging technique enables single-shot sectional imaging of a range of specimens of varying dimensions, ranging from a single cell (HeLa cell) to a multicellular organism (C. elegans). 3D reconstruction of the entire C. elegans is achieved in real-time and with an exposure time of few hundred micro-seconds. A maximum likelihood technique is developed and optimized for the iLIFE imaging system. We observed an intracellular resolution for mitochondria-labeled HeLa cells, which demonstrates the dynamic resolution of the iLIFE system. The proposed technique is a step towards achieving flow-based 3D imaging. We expect potential applications in diverse fields such as structural biology and biophysics. Published by AIP Publishing.

We propose and demonstrate a limited-view light sheet microscopy (LV-LSM) for three dimensional (3D) volume imaging. Realizing that longer and frequent image acquisition results in significant photo-bleaching, we have taken limited angular views (18 views) of the macroscopic specimen and integrated with maximum likelihood (ML) technique for reconstructing high quality 3D volume images. Existing variants of light-sheet microscopy require both rotation and translation with a total of approximately 10-fold more views to render a 3D volume image. Comparatively, LV-LSM technique reduces data acquisition time and consequently minimizes light-exposure by many-folds. Since ML is a post-processing technique and highly parallelizable, this does not cost precious imaging time. Results show noise-free and high contrast volume images when compared to the state-of-the-art selective plane illumination microscopy. (C) 2015 AIP Publishing LLC.
