1. PP2A modulation overcomes multidrug resistance in chronic lymphocytic leukemia via mPTP-dependent apoptosis

Abstract

Targeted therapies such as venetoclax (VEN) (Bcl-2 inhibitor) have revolutionized the treatment of chronic lymphocytic leukemia (CLL). We previously reported that persister CLL cells in treated patients overexpress multiple antiapoptotic proteins and display resistance to proapoptotic agents. Here, we demonstrated that multidrug-resistant CLL cells in vivo exhibited apoptosis restriction at a pre-mitochondrial level due to insufficient activation of the Bax and Bak (Bax/Bak) proteins. Co-immunoprecipitation analyses with selective BH domain antagonists revealed that the pleiotropic proapoptotic protein (Bim) was prevented from activating Bax/Bak by “switching” interactions to other upregulated antiapoptotic proteins (Mcl-1, Bcl-xL, Bcl-2). Hence, treatments that bypass Bax/Bak restriction are required to deplete these resistant cells in patients. Protein phosphatase 2A (PP2A) contributes to oncogenesis and treatment resistance. We observed that small-molecule activator of PP2A (SMAP) induced cytotoxicity in multiple cancer cell lines and CLL samples, including multidrug-resistant leukemia and lymphoma cells. The SMAP (DT-061) activated apoptosis in multidrug-resistant CLL cells through induction of mitochondrial permeability transition pores, independent of Bax/Bak. DT-061 inhibited the growth of wild-type and Bax/Bak double-knockout, multidrug-resistant CLL cells in a xenograft mouse model. Collectively, we discovered multidrug-resistant CLL cells in patients and validated a pharmacologically tractable pathway to deplete this reservoir.

Abstract

We determined the fundamental properties of HD 140283 by obtaining new interferometric and spectroscopic measurements and combining them with photometry from the literature. The interferometric measurements were obtained using the visible interferometer VEGA on the CHARA array and we determined a 1D limb-darkened angular diameter of 0.353 +/- 0.013 milliarcseconds. Using photometry from the literature we derived the bolometric flux with two solutions: a zero-reddening one of Fbol = 3.890 +/- 0.066 1E-8 erg/s/cm2 and a solution with a maximum of Av = 0.1 mag, Fbol= 4.220 +/- 0.067 1E-8 erg/s/cm2. The interferometric Teff is thus 5534 +/- 103 K or 5647 +/- 105 K and its radius is R = 2.21 +/- 0.08 Rsol. Spectroscopic measurements of HD140283 were obtained using HARPS, NARVAL, and UVES and a 1D LTE analysis of H-alpha line wings yields Teff(Halpha) = 5626 +/- 75 K. Using fine-tuned stellar models including diffusion of elements we then determined the mass M and age t of HD140283. Once the metallicity has been fixed, the age of the star depends on M, initial helium abundance Yi and mixing-length parameter alpha, only two of which are independent. We need to adjust alpha to much lower values than the solar one (~2) in order to fit the observations, and if Av = 0.0 mag then 0.5 < alpha < 1. We give an equation to estimate t from M, Yi (alpha) and Av. Establishing a reference alpha = 1.00 and adopting Yi = 0.245 we derive a mass and age of HD140283: M = 0.780 +/- 0.010 Msol and t = 13.7 +/- 0.7 Gyr (Av = 0.0) or M = 0.805 +/- 0.010 Msol and t = 12.2 +/- 0.6 Gyr (Av=0.1 mag). Our stellar models yield an initial metallicity of [Z/X]i = -1.70 and logg = 3.65 +/- 0.03. Abstract

Venetoclax combination therapies are becoming the standard of care in acute myeloid leukemia (AML). However, the therapeutic benefit of these drugs in older/unfit patients is limited to only a few months, highlighting the need for more effective therapies. Protein phosphatase 2A (PP2A) is a tumor suppressor phosphatase with pleiotropic functions that becomes inactivated in ∼70% of AML cases. PP2A promotes cancer cell death by modulating the phosphorylation state in a variety of proteins along the mitochondrial apoptotic pathway. We therefore hypothesized that pharmacological PP2A reactivation could increase BCL2 dependency in AML cells and, thus, potentiate venetoclax–induced cell death. Here, by using 3 structurally distinct PP2A-activating drugs, we show that PP2A reactivation synergistically enhances venetoclax activity in AML cell lines, primary cells, and xenograft models. Through the use of gene editing tools and pharmacological approaches, we demonstrate that the observed therapeutic synergy relies on PP2A complexes containing the B56α regulatory subunit, of which expression dictates response to the combination therapy. Mechanistically, PP2A reactivation enhances venetoclax-driven apoptosis through simultaneous inhibition of antiapoptotic BCL2 and extracellular signal-regulated kinase signaling, with the latter decreasing MCL1 protein stability. Finally, PP2A targeting increases the efficacy of the clinically approved venetoclax and azacitidine combination in vitro, in primary cells, and in an AML patient-derived xenograft model. These preclinical results provide a scientific rationale for testing PP2A-activating drugs with venetoclax combinations in AML.

Abstract

Posterior cortical atrophy (PCA) describes a neurodegenerative syndrome characterized by progressive difficulties in cortical visual and other posterior cortical functions consistent with parieto-occipital and occipito-temporal involvement. It is increasingly recognized that many patients develop difficulties with other aspects of daily living, in particular, with language and communication. We present a case emphasizing how language difficulties may emerge in PCA. Difficulties are interpreted as arising from interacting effects of linguistic deficits and impaired detection of nonverbal (particularly, visual) turns that normally facilitate, schedule, and disambiguate the exchange of verbal messages between speakers. We propose that relatively simple speech and language therapy interventions may hold promise in addressing language and communication difficulties as secondary features of PCA by targeting the behaviors of both the person with PCA and their communication partners.

Abstract

The 6-min walk test (6MWT) is a common means of functional assessment. Its relationship to disability-free survival (DFS) is uncertain.This sub-study of the Measurement of Exercise Tolerance for Surgery study had co-primary outcome measures: correlation of the preoperative 6MWT distance with 30 day quality of recovery (15-item quality of recovery) and 12 month WHO Disability Assessment Schedule scores. The prognostic utility of the 6MWT and other risk assessment tools for 12 month DFS was assessed with logistic regression and receiver-operating-characteristic-curve analysis.Of 574 patients recruited, 567 (99%) completed the 6MWT. Twelve months after surgery, 16 (2.9%) patients had died and 444 (77%) had DFS. The 6MWT correlated weakly with 30 day 15-item quality of recovery (ρ=0.14; P=0.001) and 12 month WHO Disability Assessment Schedule (ρ=-0.23; P0.0005) scores. When the cohort was split into 6MWT distance tertiles, the adjusted odds ratio of low vs high tertiles for DFS was 3.13 [95% confidence interval (CI): 1.54-6.35]. The only independent variable predictive of DFS was the Duke Activity Status Index (DASI) score (adjusted odds ratio: 1.06; P0.0005). The area under the receiver-operating-characteristic curve for DFS was 0.63 (95% CI: 0.57-0.70) for the 6MWT, 0.60 (95% CI: 0.53-0.67) for cardiopulmonary-exercise-testing-derived peak oxygen consumption, and 0.70 (95% CI: 0.64-0.76) for the DASI score.Of the risk assessment tools analysed, the DASI was the most predictive of DFS. The 6MWT was safe and comparable with cardiopulmonary exercise testing for all predictive assessments. Future research should aim to determine the optimal 6MWT distance thresholds for risk prediction.

Abstract

Targeted cancer therapies, which act on specific cancer-associated molecular targets, are predominantly inhibitors of oncogenic kinases. While these drugs have achieved some clinical success, the inactivation of kinase signaling via stimulation of endogenous phosphatases has received minimal attention as an alternative targeted approach. Here, we have demonstrated that activation of the tumor suppressor protein phosphatase 2A (PP2A), a negative regulator of multiple oncogenic signaling proteins, is a promising therapeutic approach for the treatment of cancers. Our group previously developed a series of orally bioavailable small molecule activators of PP2A, termed SMAPs. We now report that SMAP treatment inhibited the growth of KRAS-mutant lung cancers in mouse xenografts and transgenic models. Mechanistically, we found that SMAPs act by binding to the PP2A Aα scaffold subunit to drive conformational changes in PP2A. These results show that PP2A can be activated in cancer cells to inhibit proliferation. Our strategy of reactivating endogenous PP2A may be applicable to the treatment of other diseases and represents an advancement toward the development of small molecule activators of tumor suppressor proteins.

Abstract

The tumor suppressor protein phosphatase 2A (PP2A) is a serine/threonine phosphatase whose activity is inhibited in most human cancers. One of the best-characterized PP2A substrates is MYC proto-oncogene basic helix–loop–helix transcription factor (MYC), whose overexpression is commonly associated with aggressive forms of this disease. PP2A directly dephosphorylates MYC, resulting in its degradation. To explore the therapeutic potential of direct PP2A activation in a diverse set of MYC-driven cancers, here we used biochemical assays, recombinant cell lines, gene expression analyses, and immunohistochemistry to evaluate a series of first-in-class small-molecule activators of PP2A (SMAPs) in Burkitt lymphoma, KRAS-driven non–small cell lung cancer, and triple-negative breast cancer. In all tested models of MYC-driven cancer, the SMAP treatment rapidly and persistently inhibited MYC expression through proteasome-mediated degradation, inhibition of MYC transcriptional activity, decreased cancer cell proliferation, and tumor growth inhibition. Importantly, we generated a series of cell lines expressing PP2A-dependent phosphodegron variants of MYC and demonstrated that the antitumorigenic activity of SMAPs depends on MYC degradation. Collectively, the findings presented here indicate a pharmacologically tractable approach to drive MYC degradation by using SMAPs for the management of a broad range of MYC-driven cancers.

Abstract

Background The Duke Activity Status Index (DASI) questionnaire might help incorporate self-reported functional capacity into preoperative risk assessment. Nonetheless, prognostically important thresholds in DASI scores remain unclear. We conducted a nested cohort analysis of the Measurement of Exercise Tolerance before Surgery (METS) study to characterise the association of preoperative DASI scores with postoperative death or complications. Methods The analysis included 1546 participants (≥40 yr of age) at an elevated cardiac risk who had inpatient noncardiac surgery. The primary outcome was 30-day death or myocardial injury. The secondary outcomes were 30-day death or myocardial infarction, in-hospital moderate-to-severe complications, and 1 yr death or new disability. Multivariable logistic regression modelling was used to characterise the adjusted association of preoperative DASI scores with outcomes. Results The DASI score had non-linear associations with outcomes. Self-reported functional capacity better than a DASI score of 34 was associated with reduced odds of 30-day death or myocardial injury (odds ratio: 0.97 per 1 point increase above 34; 95% confidence interval [CI]: 0.96–0.99) and 1 yr death or new disability (odds ratio: 0.96 per 1 point increase above 34; 95% CI: 0.92–0.99). Self-reported functional capacity worse than a DASI score of 34 was associated with increased odds of 30-day death or myocardial infarction (odds ratio: 1.05 per 1 point decrease below 34; 95% CI: 1.00–1.09), and moderate-to-severe complications (odds ratio: 1.03 per 1 point decrease below 34; 95% CI: 1.01–1.05). Conclusions A DASI score of 34 represents a threshold for identifying patients at risk for myocardial injury, myocardial infarction, moderate-to-severe complications, and new disability.

Abstract

BACKGROUND: Preoperative anaemia is associated with elevated risks of postoperative complications. This association may be explained by confounding related to poor cardiopulmonary fitness. We conducted a pre-specified substudy of the Measurement of Exercise Tolerance before Surgery (METS) study to examine the associations of preoperative haemoglobin concentration with preoperative cardiopulmonary exercise testing performance (peak oxygen consumption, anaerobic threshold) and postoperative complications. METHODS: The substudy included a nested cross-sectional analysis and nested cohort analysis. In the cross-sectional study (1279 participants), multivariate linear regression modelling was used to determine the adjusted association of haemoglobin concentration with peak oxygen consumption and anaerobic threshold. In the nested cohort study (1256 participants), multivariable logistic regression modelling was used to determine the adjusted association of haemoglobin concentration, peak oxygen consumption, and anaerobic threshold with the primary endpoint (composite outcome of death, cardiovascular complications, acute kidney injury, or surgical site infection) and secondary endpoint (moderate or severe complications). Abstract

Increasing ethical attention and debate is focusing on whether individuals who take part in clinical trials should be given access to post-trial care. However, the main focus of this debate has been upon drug trials undertaken in low-income settings. To broaden this debate, we report findings from interviews with individuals (n = 24) who participated in a clinical trial of a closed-loop system, which is a medical device under development for people with type 1 diabetes that automatically adjusts blood glucose to help keep it within clinically recommended ranges. Individuals were recruited from UK sites and interviewed following trial close-out, at which point the closed-loop had been withdrawn. While individuals were stoical and accepting of the requirement to return the closed-loop, they also conveyed varying degrees of distress. Many described having relaxed diabetes management practices while using the closed-loop and having become deskilled as a consequence, which made reverting back to pre-trial regimens challenging. Participants also described unanticipated consequences arising from using a closed-loop. As well as deskilling, these included experiencing psychological and emotional benefits that could not be sustained after the closed-loop had been withdrawn and participants reevaluating their pre- and post-trial life in light of having used a closed-loop and now perceiving this life much more negatively. Participants also voiced frustrations about experiencing better blood glucose control using a closed-loop and then having to revert to using what they now saw as antiquated and imprecise self-management tools. We use these findings to argue that ethical debates about post-trial provisioning need to be broadened to consider potential psychological and emotional harms, and not just clinical harms, that may result from withdrawal of investigated treatments. We also suggest that individuals may benefit from information about potential nonclinical harms to help make informed decisions about trial participation.

Abstract

The ability of breast cancer cells to transiently transition between epithelial and mesenchymal states contributes to their metastatic potential. Therefore, driving tumor cells into a stable mesenchymal state, as opposed to complete tumor cell eradication, presents an opportunity to pharmacologically limit disease progression by promoting an asymptomatic state of dormancy. Here, we compare a reversible model of epithelial-mesenchymal transition (EMT) induced by TGFβ to a stable mesenchymal phenotype induced by chronic exposure to the ErbB kinase inhibitor lapatinib. Only cells capable of returning to an epithelial phenotype resulted in skeletal metastasis. Gene expression analyses of the two mesenchymal states indicated similar transition expression profiles. A potently downregulated gene in both datasets was spleen tyrosine kinase (SYK). In contrast to this similar diminution in mRNA, kinome analyses using a peptide array and DNA-conjugated peptide substrates showed a robust increase in SYK activity upon TGFβ-induced EMT only. SYK was present in cytoplasmic RNA processing depots known as P-bodies formed during the onset of EMT, and SYK activity was required for autophagy-mediated clearance of P-bodies during mesenchymal-epithelial transition (MET). Genetic knockout of autophagy-related 7 (ATG7) or pharmacologic inhibition of SYK activity with fostamatinib, a clinically approved inhibitor of SYK, prevented P-body clearance and MET, inhibiting metastatic tumor outgrowth. Overall, this study suggests assessment of SYK activity as a biomarker for metastatic disease and the use of fostamatinib as a means to stabilize the latency of disseminated tumor cells. SIGNIFICANCE: These findings present inhibition of spleen tyrosine kinase as a therapeutic option to limit breast cancer metastasis by promoting systemic tumor dormancy.

Abstract

EGFR activation is both a key molecular driver of disease progression and the target of a broad class of molecular agents designed to treat advanced cancer. Nevertheless, resistance develops through several mechanisms, including activation of AKT signaling. Though much is known about the specific molecular lesions conferring resistance to anti-EGFR–based therapies, additional molecular characterization of the downstream mediators of EGFR signaling may lead to the development of new classes of targeted molecular therapies to treat resistant disease. We identified a transcriptional network involving the tumor suppressors Krüppel-like factor 6 (KLF6) and forkhead box O1 (FOXO1) that negatively regulates activated EGFR signaling in both cell culture and in vivo models. Furthermore, the use of the FDA-approved drug trifluoperazine hydrochloride (TFP), which has been shown to inhibit FOXO1 nuclear export, restored sensitivity to AKT-driven erlotinib resistance through modulation of the KLF6/FOXO1 signaling cascade in both cell culture and xenograft models of lung adenocarcinoma. Combined, these findings define a novel transcriptional network regulating oncogenic EGFR signaling and identify a class of FDA-approved drugs as capable of restoring chemosensitivity to anti-EGFR–based therapy for the treatment of metastatic lung adenocarcinoma.

Abstract

Background Tranexamic acid reduces blood loss and transfusion requirements in cardiac surgery but may increase the risk of coronary graft thrombosis. We previously reported the 30-day results of a trial evaluating tranexamic acid for coronary artery surgery. Here we report the 1-year clinical outcomes. Methods Using a factorial design, we randomly assigned patients undergoing coronary artery surgery to receive aspirin or placebo and tranexamic acid or placebo. The results of the tranexamic acid comparison are reported here. The primary 1-year outcome was death or severe disability, the latter defined as living with a modified Katz activities of daily living score of less than 8. Secondary outcomes included a composite of myocardial infarction, stroke, and death from any cause through to 1 year after surgery. Results The rate of death or disability at 1 year was 3.8% in the tranexamic acid group and 4.4% in the placebo group (relative risk, 0.85; 95% confidence interval, 0.64-1.13; P = .27), and this did not significantly differ according to aspirin exposure at the time of surgery (interaction P = .073). The composite rate of myocardial infarction, stroke, and death up to 1 year after surgery was 14.3% in the tranexamic acid group and 16.4% in the placebo group (relative risk, 0.87; 95% CI, 0.76-1.00; P = .053). Conclusions In this trial of patients having coronary artery surgery, tranexamic acid did not affect death or severe disability through to 1 year after surgery. Further work should be done to explore possible beneficial effects on late cardiovascular events.

Abstract

Ionomeric networks (IN) with lithium sulfate and salt-solvating oligo(oxyethylene) chain were synthesized on the purpose of improving the conductivity of single-Li+−ionic conductors. Li+−ionic conductivity depends considerably on the salt content of the INs although the apparent degree of cross-linking is fixed in a constant of 10 mol%. As salt content (Li/O value) equals 0.0467, conductivity of the IN containing neither small-molecular salt nor low molecular weight plasticizer reaches a maximum of 7×10−6 S/cm at 25°C. Temperature-conductivity relationship of the INs shows curved Arrhenius plots, suggesting that the ionic conduction is primarily influenced by segmental motion of the polymer host. In addition, WLF (Williams-Landel-Ferry) equation is used to analyze the conductivity data, from which the related WLF parameters are determined.

Abstract

Protein phosphatase 2A (PP2A) is a serine/threonine phosphatase involved in the regulation of many cellular processes. A confirmed tumor suppressor protein, PP2A is genetically altered or functionally inactivated in many cancers highlighting a need for its therapeutic reactivation. In this review we discuss recent literature on PP2A: the elucidation of its structure and the functions of its subunits, and the identification of molecular lesions and post-translational modifications leading to its dysregulation in cancer. A final section will discuss the proteins and small molecules that modulate PP2A and how these might be used to target dysregulated forms of PP2A to treat cancers and other diseases.

Abstract

Background— Previous studies have shown that pericytes can differentiate into osteoblasts and form bone. This study investigated whether pericytes can also differentiate into chondrocytes and adipocytes. Methods and Results— Reverse transcription–polymerase chain reaction demonstrated that pericytes express mRNA for the chondrocyte markers Sox9, aggrecan, and type II collagen. Furthermore, when cultured at high density in the presence of a defined chondrogenic medium, pericytes formed well-defined pellets comprising cells embedded in an extracellular matrix rich in sulfated proteoglycans and type II collagen. In contrast, when endothelial cells were cultured under the same conditions, the pellets disintegrated after 48 hours. In the presence of adipogenic medium, pericytes but not endothelial cells expressed mRNA for peroxisome proliferator–activated receptor-γ2 (an adipocyte-specific transcription factor) and incorporated lipid droplets that stained with oil red O. To confirm that pericytes can differentiate along the chondrocytic and adipocytic lineages in vivo, these cells were inoculated into diffusion chambers and implanted into athymic mice for 56 days. Accordingly, mineralized cartilage, fibrocartilage, and a nonmineralized cartilaginous matrix with lacunae containing chondrocytes were observed within these chambers. Small clusters of cells that morphologically resembled adipocytes were also identified. Conclusions— These data demonstrate that pericytes are multipotent cells that may contribute to growth, wound healing, repair, and/or the development and progression of various pathological states.

Abstract

Calcification and fibrointimal proliferation are associated with advanced complicated atherosclerosis in large arteries but may also occur in smaller vessels, resulting in ischaemic tissue necrosis. This study investigates whether the mechanisms of calcification and intimal fibrosis are similar in vessels of different sizes. The localization of osteopontin (OPN), matrix Gla protein (MGP), thrombospondin-1 (TSP-1), and cartilage oligomeric matrix protein (COMP) was investigated in three types of human vascular lesions: atherosclerosis, chronic vascular rejection (CVR) in renal allografts, and calcific uraemic arteriolopathy (calciphylaxis). These lesions were chosen as they affect different sized blood vessels and they exhibit a fibroproliferative intimal reaction, with or without calcification, resulting in luminal obliteration and ischaemic complications. OPN, MGP, TSP-1, and COMP were not detected in normal blood vessels. However, OPN and MGP were expressed at sites of calcification within atherosclerotic lesions and in microvessels in calciphylaxis, suggesting that calcification in different sized vessels may occur by a common mechanism. These proteins were not detected in areas of fibrointimal proliferation. In contrast, TSP-1 was localized primarily within the fibrous tissue of atherosclerotic lesions and was also expressed in the expanded fibrous intima of arteries showing CVR. COMP was localized primarily within the fibrous tissue under the lipid core of the majority of advanced atherosclerotic lesions. TSP-1 and COMP were also detected in areas of microcalcification in atherosclerotic lesions and TSP-1 was detected adjacent to areas of calcification in calciphylaxis. However, neither TSP-1 nor COMP was localized to calcific foci within these lesions. The localization of OPN, MGP, TSP-1, and COMP to pathological, but not normal arterial intima supports a pathogenetic role for these proteins in the development of vascular fibrosis and calcification. Modulation of their production and activity may offer a novel approach to the therapy of a number of vascular diseases.

This paper examines knowledge exchange dynamics in a specialist integrated intellectual (learning) disability service, comprising specialist healthcare provision with social care commissioning and management, and considers their significance in terms of integrated service delivery.A qualitative study focusing on knowledge exchange and integrated services. Semi-structured interviews (n = 25) were conducted with members of an integrated intellectual disability service in England regarding their perceptions of knowledge exchange within the service and the way in which knowledge exchange impinges on the operation of the integrated service.Exchange of 'explicit' (codifiable) knowledge between health and care management components of the service is problematic because of a lack of integrated clinical governance and related factors such as IT and care record systems and office arrangements. Team meetings and workplace interactions allowed for informal exchange of explicit and 'tacit' (non-codifiable) knowledge, but presented challenges in terms of knowledge exchange completeness and sustainability.Knowledge exchange processes play an important role in the functioning of integrated services incorporating health and care management components. Managers need to ensure that knowledge exchange processes facilitate both explicit and tacit knowledge exchange and do not rely excessively on informal, 'ad hoc' interactions. Research on integrated services should take account of micro-scale knowledge exchange dynamics and relationships between social dynamics and physical factors.

The beta-alumina family of materials has played a central role in the field of solid-state ionics. β- and β-alumina have been a principal theme in the field ever since Yao and Kummer reported the extraordinarily high conductivity for sodium β-alumina. The material “came of age” at a time when there was great interest in the science and technology of highconductivity solid electrolytes. A previous MRS Bulletin review introduced this broad family of materials, examining the range of compositions and the two major structures. The beta-alumina family emerged as almost an ideal system in which to explore structure-property relations, as its unusual structure is responsible for its remarkable ion-transport properties. Although the initial interest in this material, and the one which endures to this day, is its rapid sodium-ion transport, the rich ion-exchange chemistry added a dimension to this material that few inorganic systems can match, let alone exceed. β-alumina, in particular, is an almost universal solid electrolyte for cations. It constitutes a broad family of solid electrolytes whose properties are dependent upon the nature of the ion inserted into the conduction plane. As a result, the β-aluminas have served as model systems for a wide range of studies: proton transport and mixed-ion diffusion, order-disorder reactions, and superlattice phenomena. Moreover, these β-alumina compositions demonstrated that fast-ion transport was not limited to a few monovalentions, but could be extended to divalent and even trivalent cations. With the latter materials, the interest was not necessarily ion transport, but optical properties instead. The presence of trivalent cations in this unusual structure, coupled with the ability to control the chemistry of the local environment, enabled the β-aluminas to exhibit a number of novel optical properties.

Abstract

Materials which display ionic conductivity were produced by the sol-gel and microemulsion-gel methods. The charging and discharging characteristics were investigated and it was found that a single ionic species was responsible for the conduction, the ionic species being residual protons from the initial component mixture. The complex impedence of various samples was measured and the ionic conductivities ranged in value from 9 × 10−8 to 1 × 10−5 ohm−1 cm−1. An Arrhemius plot for a microemulsion-gel revealed that the energy of activation was 0.15 eV.

LiMn{sub 2}O{sub 4}-based spinels have been extensively studied as positive electrode materials for lithium-ion batteries. These investigations have shown that, by using the Pechini process, the performance of these materials can be improved significantly through adjustments to the synthesis conditions and composition by means of selective doping. This paper reports the results of neutron and ex situ X-ray diffraction studies performed to examine the structural changes that occur during lithium ion insertion into various Li{sub x}Mn{sub 2}O{sub 4} compositions. It appears that an ordering intercalation of lithium ions occurs in the lithium concentration range of 0.15--0.35, followed by a second-order phase transformation when the lithium population is close to 0.5, leading to a random lithium insertion into a single-spinel phase from x = 0.5 to 1.0.

Abstract

The reaction rate was determined for copper nitrate hydrate with methoxydimethyloctylsilane (MDOS) in methanol. The rate constants of hydrolysis and condensation were established by quantitative measurement of the product and Karl Fischer water determination. The reaction with the hydrated copper salt resulted in the phase separation of an insoluble product from the reaction mixture. The structure of the product was determined, by Fourier Transform Infrared Spectrometry (FTIR) and Nuclear Magnetic Resonance (NMR) to be a dimer of the MDOS. The results showed the alcohol, producing condensation reaction was negligible in the formation of the dimer. contrary to the case for the well-known reaction by trialkoxysilanes and tetraalkoxysilanes.

Fe(NO3))-SiO2 glasses were prepared by hydrorlyzing silicon alkoxides with various amount of ferric nitrate hydrate following the sol-gel process. The gel glasses were allowed to dry completely at room temperature then several thermally treatments were done. The thermal transition was studied with differential scanning calorimetry (DSC) and thermal gravimetric analysis (TGA). The structural features of the glassy materials were analyzed by means of infrared (IR) spectroscopy and X-ray diffraction (XRD). The samples with a higher concentration of 30 % mole of iron existed in the crystalline structure; the thermal transitions did not affect the crystalline structure of these mixed samples as indicated by the X-ray diffraction. The IR spectra indicated the existence of Si-O-Fe bond. The thermal analysis indicated similar heat transitions for the glass as the pure iron nitrate with some modification due to the interactions with the silica network.

In situ electrochemical atomic force microscopy (ECAFM) was used to study the surface films that form on highly ordered pyrolytic graphite (HOPG) electrodes during cathodic polarization in 1 M EC:DMC (1:1) and 1 M EC‐DMC (1.1) electrolytes. Cyclic voltammetry experiments confirmed that distinct reduction reactions occur when the potential of a fresh HOPG electrode is swept from 2.4 to 0.01 V vs. in a Li/HOPG cell. The reactions appear to be irreversible since no evidence of corresponding oxidation reactions was observed. ECAFM results, when combined with slow‐scan cyclic voltammetry data, suggest that different electrochemical reactions are occurring in these two electrolytes. The deposits were found to be several hundreds of nanometers thick. Auger analysis confirmed the presence of elements consistent with electrolyte reduction products. These observations have implications for understanding the formation of a solid electrolyte interphase on HOPG electrodes in lithium‐ion batteries.

The crystal structure and ionic distribution in the conduction plane of the partially exchanged Na+ \beta-alumina system Li0.75Na0.47Al11O17.11 has been determined from single-crystal X-ray diffraction at 30 and 298 K, in combination with a single-crystal neutron diffraction study at 295 K. At 30 K the Li+ ions occupy two sites: one in (2/3, 1/3, 1/4) and one ∼1.0 Å out of the z = 1/4 conduction plane. The Na+ ions are also found in two sites, both in the conduction plane. At 298 K the Li+ ions are found in three sites; the same two sites as at 30 K, as well as at a third site between these two. The Na+ ions occupy the same sites as at 30 K. The combination of X-ray and neutron diffraction is used to resolve the distribution of Na+ and Li+ ions. The column O5 atom in the Al3—O5—Al3 bridge between the spinel blocks is found to be disordered. Charge compensation is achieved through a Frenkel defect of two interstitial Al atoms, one on either side of the conduction plane, together with an extra oxygen in the conduction plane. One such defect occurs, on average, in every 4.5 unit cells.

It also has been shown that different firing temperatures and the selective doping of LiMn{sub 2}O{sub 4} both produce materials with different electrochemical characteristics that merit further exploration.

Abstract

Atomic force microscopy, a powerful, high-resolution imaging technique for determining the structure of surfaces in gaseous and liquid environments, was used to examine the reactivity of an electropolished surface of a naturally aged aluminum-copper-magnesium alloy (2024-T3) in aqueous hydrochloric acid (0.01, 0.1, and 1 M). When first exposed to acid, the matrix dissolved uniformly. Dissolution then accelerated and pits formed predominantly in the vicinity of the second-phase precipitates. The pits developed into characteristic intergranular damage: i.e., elongated pits (incipient corrosion cracks) along grain boundaries. Postexperimental ex situ energy dispersive X-ray analysis and Auger electron spectroscopy were employed to characterize the composition of the various surface features of corroded samples.

LiMn{sub 2}O{sub 4}-based spinels are of great interest as positive electrode materials for lithium-ion batteries. The authors describe here what is believed to be the first synthesis of these materials using the Pechini process, a low temperature synthetic method that often yields inorganic oxides of excellent phase purity and well-controlled stoichiometry. Using this process, it has been possible to synthesize phase-pure crystalline spinel LiMn{sub 2}O{sub 4} by calcining the appropriate polymeric precursors in air at 250 C for several hours. The influence of different firing temperatures and the effect of substituting a small amount of Mn with Ni have also been explored. Electrochemical studies show that the Pechini-synthesized materials appear to offer not only high quality performance but also significant analytical advantages which allows one to understand the structural mechanism of Li intercalation.

We have found it possible to determine salt diffusion coefficients and ion transference numbers by obtaining, respectively, the steady-state ion-transport-limited current at a microdisk electrode in spherical diffusion geometry and the transient ion-transport-limited current at a macrodisk electrode in planar diffusion geometry in ionic solutions containing no supporting electrolyte. The method is demonstrated in aqueous solutions of AgNO 3 and in propylene carbonate solutions of LiClO 4 . The ion transport parameters obtained agree well with those reported previously. It would appear that the method also could be used in the study of ion transport in solid electrolytes

A systematic characterization of the electrokinetics of the Li/Li + couple was carried out in poly(ethylene glycol dimethyl ether) (PEGM) (MW = 400) and in its small molecule analog, 1,2-dimethoxyethane (DME), using LiAsF 6 as the electrolyte salt. Microelectrodes (nickel disks of 25 or 50 μm diam) were used for the investigations because of their many advantages over conventional macroelectrodes. In the kinetic studies, lithium was freshly deposited and then immediately stripped to minimize interference from lithium surface films. The exchange current densities, i 0 , for the Li/Li + couple are an order of magnitude smaller in PEGM than in DME between 296 and 338 K. A typical value of i 0 is 0.72 mA/cm 2 in PEGM and 9.8 mA/cm in DME, for salt concentrations of 0.3 M at 313 K. The transfer coefficient was close to 0.5 in both solvents at all temperatures. The exchange current density in PEGM is strongly temperature dependent, with a high activation energy of 66 kJ/mol. The concentration dependence of i 0 in PEGM is unusual, with a linear relationship between i 0 and C 1/2 salt from 0.02 to 0.6 M after which the i 0 drops off sharply. The results can be interpreted microscopically in light of recent advances concerning the influence of solvent dynamics on electron transfer. Abstract

PP2A is a phosphatase that is functionally dysregulated and inactivated in over 50% of all breast cancers independent of histological type. While much attention has focused on oncogenic kinases as targets for cancer treatment, therapeutic targeting of phosphatases, the key negative regulators of these same signaling pathways, has remained largely unexplored. Starting with the observation that tricyclic neuroleptic drugs exert anticancer effects, our laboratory reverse engineered these drugs to generate a novel series of compounds that retain the anti-proliferative effects and favorable pharmacokinetic properties but are devoid of the undesirable central nervous system pharmacology of the parent drugs. It was subsequently demonstrated that these first generation novel derivatives exert anticancer effects in cell culture and xenograft models of cancer (without overt toxicity) by modulation of critical oncogenic signaling pathways. Recently, it was determined that the molecular basis of the actions of these novel derivatives is the activation of PP2A. Comprehensive profiling for activity in a panel of 240 cancer cell lines has shown that these first-in-class small molecule activators of PP2A (SMAPs) have significant anti-proliferative activity in breast cancer cell lines, and studies have been extended to disease relevant in vivo models (patient derived xenograft, GEMM, and traditional xenograft models). PP2A's ability to negatively regulate a diverse set of oncogenic drivers in breast cancer means SMAPs are able to exert their effects through multiple biological mechanisms, swiftly disrupting cellular energetics and metabolism, inducing apoptosis and inhibiting proliferation. This will be exhibited through a variety of biological assays evaluating cell growth, apoptosis and changes in cellular metabolism. Target engagement assessed via western blotting demonstrates inhibition of PP2A targets critical for mediating SMAP activity, notably c-MYC and AKT. As a result, this research demonstrates SMAPs have potent activity in a wide variety of breast cancer contexts where response to current therapies is modest and limited by the development of treatment resistance. Thus, SMAPs may be a viable treatment option for high-risk populations of patients who do not respond to the current standard of care. This research strives to demonstrate that therapeutic reactivation of PP2A may represent a novel approach for breast cancer treatment and that these molecules may favorably impact the lives of women suffering from breast cancer. Abstract

We report an analysis of the Lambda(0)(b) -> Lambda(+)(c)pi(-)pi(+)pi(-) decay in a data sample collected by the CDF II detector at the Fermilab Tevatron corresponding to 2.4 fb(-1) of integrated luminosity. We reconstruct the currently largest samples of the decay modes Lambda(0)(b) -> Lambda(c)(2595)(+)pi(-) (with Lambda(c)(2595)(+) -> Lambda(+)(c)pi(+)pi(-)), Lambda(0)(b) -> Lambda(c)(2625)(+)pi(-) (with Lambda(c)(2625)(+) -> Lambda(+)(c)pi(+)pi(-)), Lambda(0)(b) -> Sigma(c)(2455)(++)pi(-)pi(-) (with Sigma(c)(2455)(++) -> Lambda(+)(c)pi(+)), and Lambda(0)(b) ->Sigma(c)(2455)(0)pi(+)pi(-) (with Sigma(c)(2455)(0) -> Lambda(+)(c)pi(-)) and measure the branching fractions to the Lambda(0)(b) -> Lambda(+)(c)pi(-) branching fraction. We measure the ration B(Lambda(0)(b) -> Lambda(+)(c)pi(-)pi(+)pi(-))/B(Lambda(0)(b) -> Lambda(+)(c)pi(-)) = 3.04 +/- 0.33(stat)(-0.55)(+0.70)(syst) which is used to derive B(Lambda(0)(b) -> Lambda(+)(c)pi(-)pi(+)pi(-)) = (26.8(-11.2)(+11.9)) x 10(-3).

Abstract

A search for a narrow Higgs boson resonance in the diphoton mass spectrum is presented based on data corresponding to 7.0 fb(-1) of integrated luminosity from p (p) over bar collisions at root s = 1: 96 TeV collected by the CDF experiment. No evidence of such a resonance is observed, and upper limits are set on the cross section times branching ratio of the resonant state as a function of Higgs boson mass. The limits are interpreted in the context of the standard model and one fermiophobic benchmark model where the data exclude fermiophobic Higgs bosons with masses below 114 GeV/c(2) at a 95% Bayesian credibility level.

Abstract

We present a search for neutral Higgs bosons phi decaying into b (b) over bar, produced in association with b quarks in p (p) over bar collisions. This process could be observable in supersymmetric models with high values of tan beta. The event sample corresponds to 2.6 fb(-1) of integrated luminosity collected with the CDF II detector at the Fermilab Tevatron collider. We search for an enhancement in the mass of the two leading jets in events with three jets identified as coming from b quarks using a displaced vertex algorithm. A data-driven procedure is used to estimate the dijet mass spectrum of the nonresonant multijet background. The contributions of backgrounds and a possible Higgs boson signal are determined by a two-dimensional fit of the data, using the dijet mass together with an additional variable which is sensitive to the flavor composition of the three tagged jets. We set mass-dependent limits on sigma(p (p) over bar -> phi b) x B(phi -> b (b) over bar) which are applicable for a narrow scalar particle phi produced in association with b quarks. We also set limits on tan beta in supersymmetric Higgs models including the effects of the Higgs boson width.

Abstract

We report a set of measurements of inclusive invariant p(T) differential cross sections of Lambda(0), (Lambda) over bar (0), Xi(+/-), and Omega(+/-) hyperons reconstructed in the central region with pseudorapidity vertical bar eta vertical bar < 1 and p(T) up to 10 GeV/c. Events are collected with a minimum-bias trigger in p collisions at a center-of-mass energy of 1.96 TeV using the CDF II detector at the Tevatron Collider. As p(T) increases, the slopes of the differential cross sections of the three particles are similar, which could indicate a universality of the particle production in p(T). The invariant differential cross sections are also presented for different charged-particle multiplicity intervals.

Abstract

We report on a search for the standard model Higgs boson decaying into pairs of tau leptons in p (p) over bar collisions produced by the Tevatron at root s = 1.96 TeV. The analyzed data sample was recorded by the CDFII detector and corresponds to an integrated luminosity of 6.0 fb(-1). The search is performed in the final state with one tau decaying leptonically and the second one identified through its semihadronic decay. Since no significant excess is observed, a 95% credibility level upper limit on the production cross section times branching ratio to the tau tau final state is set for hypothetical Higgs boson masses between 100 and 150 GeV/c(2). For a Higgs boson of 120 GeV/c(2) the observed (expected) limit is 14.6 (15.3) the predicted value.

Abstract

We present a search for the standard model Higgs boson production in association with a W boson in proton-antiproton collisions (p (p) over bar -> W+ H -> lvb (b) over bar) at a center of mass energy of 1.96 TeV. The search employs data collected with the CDF II detector which correspond to an integrated luminosity of approximately 2.7 fb(-1). We recorded this data with two kinds of triggers. The first kind required high-p(T) charged leptons and the second required both missing transverse energy and jets. The search selects events consistent with a signature of a single lepton (e(+/-)/mu(+/-)),missing transverse energy, and two jets. Jets corresponding to bottom quarks are identified with a secondary vertex tagging method and a jet probability tagging method. Kinematic information is fed in an artificial neural network to improve discrimination between signal and background. The search finds that both the observed number of events and the neural network output distributions are consistent with the standard model background expectations, and sets 95% confidence level upper limits on the production cross section times branching ratio. The limits are expressed as a ratio to the standard model production rate. The limits range from 3.6 (4.3 expected) to 61.1 (43.2 expected) for Higgs masses from 100 to 150 GeV/c(2), respectively.

Abstract

Diboson production (WW + WZ + ZZ) has been observed at the Tevatron in hadronic decay modes dominated by the WW process. This paper describes the measurement of the cross section of WZ and ZZ events in final states with large (sic)(T) and using b-jet identification as a tool to suppress WW contributions. Because of the limited energy resolution, we cannot distinguish between partially hadronic decays of WZ and ZZ, and we measure the sum of these processes. The number of signal events is extracted using a simultaneous fit to the invariant mass distribution of the two jets for events with two b-jet candidates and events with fewer than two b-jet candidates. We measure a cross section sigma(p (p) over bar -> WZ, ZZ) = 5.8(3.0)(+3.6) pb, in agreement with the standard model.

Abstract

We report an indirect search for nonstandard model physics using the flavor-changing neutral current decays B -> K-(*)mu(+)mu(-). We reconstruct the decays and measure their angular distributions, as a function of q(2) = M(mu mu)(2)c(2), where M-mu mu is the dimuon mass, in p (P) over bar collisions at root s = 1.96 Tev using a data sample corresponding to an integrated luminosity of 6.8 fb(-1). The transverse polarization asymmetry A(T)((2)) and the time-reversal-odd charge-and-parity asymmetry A(im) are measured for the first time, together with the K* longitudinal polarization fraction F-L and the muon forward-backward asymmetry A(FB) for the decays B-0 -> K*(0)mu(+)mu(-) and B -> K*(+)mu(+)mu(-). Abstract

Sol-gel chemistry has a great many potential applications in the preparation of unique electrochemical materials, from non-equilibrium transition metal oxides which may be of use as high energy density electrodes, to high surface area mixed oxides which may possess high proton conductivity, to novel composite structures consisting of inorganic gels in combination with organic electronic and ionic conductors. This papar reviews prior work on proton conduction in gel systems and presents recent work regarding electrode materials prepared by sol-gel methods and inorganic-organic materials.

Abstract

The morphology of silver deposited on a platinum electrode from aqueous solutions of H 2 SO 4 , Na 2 SO 4 , and Ag 2 SO 4 was investigated for different concentrations of silver ions and different potential sweep rates (1, 4, and 10 mV)s) using in situ electrochemical atomic force microscopy (ECAFM). At a sweep rate of 1 mV/s two kinds of silver crystals formed: smaller ones (distributed uniformly over the electrode surface) and larger ones (nucleated on scratches). At faster sweep rates the deposit becomes more uniform. The roughness of the silver deposit decreased and the deposit uniformity increased during extended cycling at a constant sweep rate. Some artifacts due to the presence of the AFM tip were observed when the force between the tip and the electrode surface was large

Abstract

We report the first observation of the baryonic flavor-changing neutral current decay Lambda(0)(b) -> Lambda mu(+)mu(-) with 24 signal events and a statistical significance of 5.8 Gaussian standard deviations. This measurement uses a p (p) over bar collisions data sample corresponding to 6.8 fb(-1) at root s = 1.96 TeV collected by the CDF II detector at the Tevatron collider. The total and differential branching ratios for Lambda(0)(b) -> Lambda mu(+)mu(-) are measured. We find B(Lambda(0)(b) -> Lambda mu(+)mu(-)) = [1.73 +/- 0.42(stat) +/- (syst)] x 10(-6). We also report the first measurement of the differential branching ratio of B-s(0) -> phi mu(+)mu(-), using 49 signal events. In addition, we report branching ratios for B+ -> K+mu(+)mu(-), B-0 -> K-0 mu(+)mu(-), and B -> K*(892)mu(+)mu(-) decays.
