1. Distinguishing the combined vegetation and soil component of δ13C variation in speleothem records from subsequent degassing and prior calcite precipitation effects

Abstract: The carbon isotopic signature inherited from soil and epikarst processes may be modified by degassing and prior calcite precipitation (PCP) before its imprint on speleothem calcite. Despite laboratory demonstration of PCP effects on carbon isotopes and increasingly sophisticated models of the governing processes, to date, there has been limited effort to deconvolve the dual PCP and soil–epikarst components in measured speleothem isotopic time series. In this contribution, we explore the feasibility, advantages, and disadvantages of using trace element ratios and δ44Ca to remove the overprinting effect of PCP on measured δ13C to infer the temporal variations in the initial δ13C of drip water prior to degassing and PCP. In nine examined stalagmites, the most widely utilized PCP indicators Mg/Ca and δ44Ca covary as expected. However, Sr / Ca does not show consistent relationships with δ44Ca so PCP is not the dominant control on Sr / Ca. From δ44Ca and Mg/Ca, our calculation of PCP as fCa, the fraction of initial Ca remaining in solution at the time the stalagmite layer is deposited, yields multiple viable solutions depending on the assumed δ44Ca fractionation factor and inferred variation in DMg. Uncertainty in the effective fractionation of δ13C during degassing and precipitation contributes to uncertainty in the absolute value of estimated initial δ13C. Nonetheless, the trends in initial δ13C are less sensitive to these uncertainties. In coeval stalagmites from the same cave spanning the 94 to 82 ka interval, trends in calculated initial δ13C are more similar than those in measured δ13C and reveal a common positive-anomaly initial δ13C during a stadial cooling event. During deglaciations, calculated initial δ13C implies a trend of greater respiration rates and higher soil CO2, although the higher interglacial drip water saturation favors more extensive PCP. Initial δ13C can be estimated for active and fossil speleothems from a range of settings, wherever there is confidence that Mg/Ca and/or δ44Ca provides a quantitative indication of past changes in PCP. Further study of Mg partitioning in speleothems will improve the robustness of Mg/Ca as a PCP proxy.

Abstract: Experiments on ASDEX Upgrade (AUG) in 2021 and 2022 have addressed a number of critical issues for ITER and EU DEMO. A major objective of the AUG programme is to shed light on the underlying physics of confinement, stability, and plasma exhaust in order to allow reliable extrapolation of results obtained on present day machines to these reactor-grade devices. Concerning pedestal physics, the mitigation of edge localised modes (ELMs) using resonant magnetic perturbations (RMPs) was found to be consistent with a reduction of the linear peeling-ballooning stability threshold due to the helical deformation of the plasma. Conversely, ELM suppression by RMPs is ascribed to an increased pedestal transport that keeps the plasma away from this boundary. Candidates for this increased transport are locally enhanced turbulence and a locked magnetic island in the pedestal. The enhanced D-alpha (EDA) and quasi-continuous exhaust (QCE) regimes have been established as promising ELM-free scenarios. Here, the pressure gradient at the foot of the H-mode pedestal is reduced by a quasi-coherent mode, consistent with violation of the high-n ballooning mode stability limit there. This is suggestive that the EDA and QCE regimes have a common underlying physics origin. In the area of transport physics, full radius models for both L- and H-modes have been developed. These models predict energy confinement in AUG better than the commonly used global scaling laws, representing a large step towards the goal of predictive capability. A new momentum transport analysis framework has been developed that provides access to the intrinsic torque in the plasma core. In the field of exhaust, the X-Point Radiator (XPR), a cold and dense plasma region on closed flux surfaces close to the X-point, was described by an analytical model that provides an understanding of its formation as well as its stability, i.e., the conditions under which it transitions into a deleterious MARFE with the potential to result in a disruptive termination. With the XPR close to the divertor target, a new detached divertor concept, the compact radiative divertor, was developed. Here, the exhaust power is radiated before reaching the target, allowing close proximity of the X-point to the target. No limitations by the shallow field line angle due to the large flux expansion were observed, and sufficient compression of neutral density was demonstrated. With respect to the pumping of non-recycling impurities, the divertor enrichment was found to mainly depend on the ionisation energy of the impurity under consideration. In the area of MHD physics, analysis of the hot plasma core motion in sawtooth crashes showed good agreement with nonlinear 2-fluid simulations. This indicates that the fast reconnection observed in these events is adequately described including the pressure gradient and the electron inertia in the parallel Ohm’s law. Concerning disruption physics, a shattered pellet injection system was installed in collaboration with the ITER International Organisation. Thanks to the ability to vary the shard size distribution independently of the injection velocity, as well as its impurity admixture, it was possible to tailor the current quench rate, which is an important requirement for future large devices such as ITER. Progress was also made modelling the force reduction of VDEs induced by massive gas injection on AUG. The H-mode density limit was characterised in terms of safe operational space with a newly developed active feedback control method that allowed the stability boundary to be probed several times within a single discharge without inducing a disruptive termination. Regarding integrated operation scenarios, the role of density peaking in the confinement of the ITER baseline scenario (high plasma current) was clarified. The usual energy confinement scaling ITER98( p,y ) does not capture this effect, but the more recent H20 scaling does, highlighting again the importance of developing adequate physics based models. Advanced tokamak scenarios, aiming at large non-inductive current fraction due to non-standard profiles of the safety factor in combination with high normalised plasma pressure were studied with a focus on their access conditions. A method to guide the approach of the targeted safety factor profiles was developed, and the conditions for achieving good confinement were clarified. Based on this, two types of advanced scenarios (‘hybrid’ and ‘elevated’ q -profile) were established on AUG and characterised concerning their plasma performance.

Abstract: After a long device enhancement phase, scientific operation resumed in 2022. The main new device components are the water cooling of all plasma facing components and the new water-cooled high heat flux divertor units. Water cooling allowed for the first long-pulse operation campaign. A maximum discharge length of 8 min was achieved with a total heating energy of 1.3 GJ. Safe divertor operation was demonstrated in attached and detached mode. Stable detachment is readily achieved in some magnetic configurations but requires impurity seeding in configurations with small magnetic pitch angle within the edge islands. Progress was made in the characterization of transport mechanisms across edge magnetic islands: Measurement of the potential distribution and flow pattern reveals that the islands are associated with a strong poloidal drift, which leads to rapid convection of energy and particles from the last closed flux surface into the scrape-off layer. Using the upgraded plasma heating systems, advanced heating scenarios were developed, which provide improved energy confinement comparable to the scenario, in which the record triple product for stellarators was achieved in the previous operation campaign. However, a magnetic configuration-dependent critical heating power limit of the electron cyclotron resonance heating was observed. Exceeding the respective power limit leads to a degradation of the confinement.

Abstract: Particle balance calculations are done for the seeding species N _2 , Ne, Ar, Kr as well as He with the aim of obtaining a realistic description of the divertor and core plasma impurity content. Experimental time traces of main plasma impurity densities are fitted by a single, time-independent parameter v $_{z,\textrm{in}}^\textrm{eff}$ . This parameter represents the product of the impurity inward pinch in the pedestal, used for the description of gross fueling here, and the enrichment factor between the sub-divertor gas reservoir and the upstream separatrix. v $_{z,\textrm{in}}^\textrm{eff}$ depends strongly on the first ionization energy as well as on the charge Z or mass of the impurity. The prevailing dependence of the enrichment values on the ionization energy suggests the importance of the relative impurity and deuterium ionization lengths in the divertor. Regression analysis of v $_{z,\textrm{in}}^\textrm{eff}$ yields an expression for the impurity concentrations in the core and the divertor of ASDEX Upgrade which allows the prediction of the corresponding impurity densities and their divertor enrichment within a factor of 2 with only engineering parameters as input. A simple wall model has been introduced to take into account wall storage and release of impurities, e.g. for conditions of pre-loaded walls due to seeding in previous discharges. Wall effects are observed for all species considered, but wall storage turns out to be more important for N and He compared to Ne, Ar, Kr. Similar enrichment values are obtained for ELMy H-modes and EDA/QCE no-ELM regimes. A factor of approximately 1.4 reduction in enrichment is observed for divertor conditions for pronounced detachment with Ar and N _2 . The obtained analytical model for the core and sub-divertor impurity densities is well suited for integration into a discharge flight simulator or a real time state observer.

Abstract: Superconducting (SC) tokamak JT-60SA plays an essential role in fusion research and development by supporting and complementing the ITER project, providing directions to the DEMO design activity and fostering next generation scientists and engineers. Since the short circuit incident at the terminal joints of equilibrium field coil #1 during the integrated commissioning (IC) in March 2021, both EU and JA implementing agencies (IAs) have examined how to ensure safe operation of JT-60SA by mitigating the risk of possible discharge occurrence inside the cryostat. Based on the experience of the global Paschen tests, the IAs have established a strategy of risk mitigation measures, which is a combination of (i) reinforcement of insulation, (ii) avoiding unnecessary voltage application to the coil systems and (iii) immediate de-energization of the coils when deteriorated vacuum conditions are detected. Thanks to the considerable efforts of the Integrated Project Team members, the IC restarted in May 2023. After confirmation of the SC state of the coil systems (TF, EF and CS), the coil energization test and the plasma operation phase 1 (OP-1) started. The first plasma was successfully achieved on 23 October 2023 with a limited value of voltage and current applied to the coils. The plasma configuration control was also confirmed with low plasma current and low auxiliary heating power conditions. Based on the IO–F4E–QST collaboration, activities of JT-60SA have been shared with the IO and provided an important lesson for ITER assembly and commissioning, and will provide an outstanding contribution to fusion research at large. After OP-1, maintenance & enhancement phase 1 (M/E-1) starts from January 2024, in which in-vessel components are installed, and heating and diagnostic systems are extensively upgraded to allow a high power heating experiment planned in OP-2. In order to make the best use of JT-60SA, a newly organized JT-60SA experiment team will refine the research plan for the future high heating power operation phase.

6. Numerical simulation of neutral gas dynamics in the W7-X sub-divertor

Abstract: The present work presents a 2D and 3D modeling of the neutral gas flow in the sub-divertor region of the W7-X. The investigations have been done using the DIVGAS code. The complex 2D and 3D geometries of the divertor components in the sub-divertor region have been considered and the Standard and High-Iota magnetic configurations have been numerically simulated. The main objective of this study is to investigate the dynamics of neutral particles in the sub-divertor region including the effects due to geometry and toroidal and poloidal leakages located at the divertor targets and baffles on the achieved pumping efficiency. A sensitivity analysis has been performed for the effect of various geometrical and flow parameters on the pumping performance, under different plasma scenarios. The considered incoming fluxes in the sub-divertor range between 10 ^20 to 10 ^22 (H _2 s ^−1 ). The main conclusions, which can be extracted from the present numerical analysis could be summarized as follows; a large fraction of incoming neutral particle flux i.e. >70% on the low iota side and >40% for the high iota side is leaked back to the main divertor region, while higher incoming neutral fluxes facilitate the increase of the pumped flux as well as the decrease of the outflux. It has been estimated that a small fraction ∼3%–4% of the incoming neutral flux is being pumped via the turbo-molecular pumps. The closure of the toroidal leakages as well as the inclination of the pumping gap panel by 9 ^o facilitate the increase of the pumped flux, but considering the all the engineering constraints, the latter option seems to be more easy to be implemented. For low incoming neutral fluxes (∼10 ^20 H _2 s ^−1 ) and for the case of AEH section, free molecular flow conditions are estimated and therefore neutral-neutral collisions could be neglected. For higher incoming neutral fluxes and for both AEH and AEP sections neutral-neutral collisions play a significant role in the flow establishment. A comparison with available experimental measurements of the neutral pressure in the sub-divertor has been performed for Standard and High-Iota plasma discharges. The 3D DIVGAS simulations predict qualitatively the experimental data with relative deviation between 25 and 63%. All the above numerical findings will actively support the optimization of the W7-X particle exhaust, in view of the experimental campaign OP2.

The construction of the toroidal field magnet system is progressing. The prototype of the 170 GHz gyrotron has been produced and it is now under test on the FALCON facility. The design of the vacuum vessel, the poloidal field coils and the civil infrastructures has been completed. The shape of the first DTT divertor has been agreed with EUROfusion to test different plasma and exhaust scenarios: single null, double null, X-divertor and negative triangularity plasmas. A detailed research plan is being elaborated with the involvement of the EUROfusion laboratories.

Abstract: This paper is dealing with the physics basis used for the design of the Divertor Tokamak Test facility (DTT), under construction in Frascati (DTT 2019 DTT interim design report (2019)) Italy, and with the description of the main target plasma scenarios of the device. The main goal of the facility will be the study of the power exhaust, intended as a fully integrated core-edge problem, and eventually to propose an optimized divertor for the European DEMO plant. The approach used to design the facility is described and their main features are reported, by using simulations performed by state-of-the-art codes both for the bulk and edge studies. A detailed analysis of MHD, including also the possibility to study disruption events and Energetic Particles physics is also reported. Eventually, a description of the ongoing work to build-up a Research Plan written and shared by the full EUROfusion community is presented.

9. Exploring blog narratives of parental loneliness: A thematic network analysis

Abstract: UK-based national surveys and international longitudinal studies have shown that around a third of parents experience chronic or persistent loneliness. There is limited research about the experience of loneliness in parenthood, however blogs authored by parents, sharing their personal experiences about loneliness offer a potential rich data source. The purpose of this study was to identify and analyse blog narratives written by parents who had experienced loneliness to increase understanding of their experiences. One hundred and ninety-six relevant blog posts were identified, of which 157 had contact details to request permission to use the blog post in the study. Twenty-two parent bloggers gave their permission. Thematic network analysis was used to open code the blogs and 4 organising themes and subthemes emerged, which centred on a global theme of disconnection. Disconnection underpinned themes relating to a sense that being a parent was overwhelming, changes in identity linked to becoming a parent, difficulties in sharing feelings of loneliness with others, and a need for social connection. Findings point to parents being unprepared for the transition to parenthood, with implications for perinatal education and support, including further opportunities for parents to connect to reduce social isolation.

10. Satisfaction with Social Connectedness Is Associated with Depression and Anxiety Symptoms in Neurodiverse First-Semester College Students

Abstract: Social difficulties and mental health are primary behavioral health concerns in autistic young adults, perhaps especially during key life transitions such as entering college. This study evaluated how dissatisfaction with social connectedness may predict and/or maintain depression and anxiety symptoms in neurodiverse, first-semester, undergraduate students (N = 263; n = 105 with diagnosed or suspected autism). Participation included a baseline survey battery, a brief survey completed twice per week across 12 weeks, and an endpoint survey battery. Social dissatisfaction at baseline was prospectively associated with biweekly ratings of depression symptoms, when controlling for baseline depressive symptoms. Social dissatisfaction was synchronously related to elevated sadness, anhedonia, and anxiety throughout the semester. These relationships were generally consistent across levels of baseline social motivation; however, there was one significant moderation effect--the negative relationship between baseline social satisfaction and anxiety was strongest for more socially motivated participants. More autistic traits were related to lower social satisfaction at baseline and greater mood concerns across timepoints. In contrast, greater autistic traits at baseline were related to greater satisfaction with social connectedness throughout the semester. Results support ongoing efforts to address mental health in autistic college students by highlighting the importance of social satisfaction.

11. 3D numerical study of neutral gas dynamics in the DTT particle exhaust using the DSMC method

Abstract: Recently the design of the divertor tokamak test (DTT) Facility divertor has been modified and consolidated. The new divertor design presents significant geometrical differences compared to the previous ITER-like one, including the presence of a more flattened dome shape. This paper presents a complete 3D numerical analysis of the neutral gas dynamics inside the DTT subdivertor area for the latest divertor design. The analysis has been performed based on the direct simulation Monte Carlo method by applying the DIVGAS simulator code. SOLEDGE2D-EIRENE plasma simulations have been performed for a deuterium plasma scenario at the maximum additional power in partially detached condition achieved by neon impurity seeding and the extracted information about the neutral particles has been imposed as incoming boundary conditions. The pumping efficiency of the DTT divertor is examined by considering various cases with respect to the pumping probability and the effect of the toroidal and poloidal leakages is quantified. The results show that a significant percentage of the incoming flux of neutrals returns back to the plasma site through the entry gaps (60% for deuterium and 40% for neon), and, consequentially, only a small percentage (∼2%–15%) of the incoming flux can be pumped out from the system. The toroidal leakages affect significantly the pumping performance of the divertor causing a significant decrease in the pumped flux (and also in the pressure at the pumping opening) about 37%–47% and 43%–56% for deuterium and neon respectively. It is discussed how many pumping ports are needed depending on the achievable pumping performance per port. The number can be reduced by closing the toroidal gaps. The analysis shows that enlarging the poloidal gaps by a factor of two causes a significant increase in the poloidal flux losses by a factor 1.7. It is also illustrated how the presence of the cooling pipes leads to conductance losses.

12. Vocal Emotion Recognition in Autism: Behavioral Performance and Event-Related Potential (ERP) Response

Abstract: Autistic youth display difficulties in emotion recognition, yet little research has examined behavioral and neural indices of vocal emotion recognition (VER). The current study examines behavioral and event-related potential (N100, P200, Late Positive Potential [LPP]) indices of VER in autistic and non-autistic youth. Participants (N = 164) completed an emotion recognition task, the Diagnostic Analyses of Nonverbal Accuracy (DANVA-2) which included VER, during EEG recording. The LPP amplitude was larger in response to high intensity VER, and social cognition predicted VER errors. Verbal IQ, not autism, was related to VER errors. An interaction between VER intensity and social communication impairments revealed these impairments were related to larger LPP amplitudes during low intensity VER. Taken together, differences in VER may be due to higher order cognitive processes, not basic, early perception (N100, P200), and verbal cognitive abilities may underlie behavioral, yet occlude neural, differences in VER processing.

15. A Partnership Pathway: Developing a Process, a Structure, and an Evaluation Tool for the Digital Communication Technology Partnership

Abstract: The mere existence of an articulation agreement does not constitute success for any stakeholders; success should be determined through evaluation (Payne et al., 2021). Wilmington University, a private, non-sectarian university with an enrollment of more than 20,000 students located in Wilmington, Delaware, utilizes multiple types of articulation agreements to establish academic partnerships. In 2021, Wilmington University and the Delaware Department of Education established the Digital Communication Technology MOU, which allows students who have completed the Digital Communication Technology Pathway in a Delaware high school the ability to articulate 18 college credits into Wilmington University. Due to a number of factors, Wilmington University has a limited ability to evaluate the Digital Communication Technology (DCT) Partnership. The purpose of this study was to create an evaluation tool that Wilmington University personnel can use to monitor and gauge the areas of success and needed improvements for the DCT Partnership. Qualitative research was conducted with identified stakeholders throughout Academic Affairs, Academic Partnerships, and the Recruiting Department within Wilmington University to inform the development of the DCT Evaluation Tool. Griggs' (2020) Conceptual Framework for Educational Partnerships which focuses on three areas of importance when evaluating educational partnerships - cooperation, coordination, and collaboration - was also utilized to inform the structure of the DCT evaluation tool and measure the current status of the DCT Partnership. Results indicate the DCT Partnership is operating in the cooperation phase of partnerships. The results show evaluation is one of many issues placing the DCT Partnership at risk. It is recommended Wilmington University implement a process, structure, and evaluation tool for the DCT Partnership. This EPP can be used as an implementation guide for Wilmington University stakeholders to optimize, monitor, and evaluate the DCT Partnership. Abstract: Background. Up to a quarter of inpatients in high-income countries (HICs) self-report beta-lactam allergy (BLA), which if incorrect,increases the use of alternative antibiotics, worsening individual health outcomes and driving bacterial resistance. In HICs, up to 95% ofself-reported BLAs are incorrect. The epidemiology of BLA in low- and middle-income African countries is unknown.Objectives. To describe the epidemiology and de-labelling outcomes of self-reported BLA in hospitalised South African (SA) patients.Methods. Point-prevalence surveys were conducted at seven hospitals (adult, paediatric, government and privately funded, district andtertiary level) in Cape Town, SA, between April 2019 and June 2021. Ward prescription records and in-person interviews were conductedto identify and risk-stratify BLA patients using the validated PEN-FAST tool. De-labelling was attempted at the tertiary allergy clinic atGroote Schuur Hospital.Results. A total of 1 486 hospital inpatients were surveyed (1 166 adults and 320 children). Only 48 patients (3.2%) self-reported a BLA,with a higher rate in private than in government-funded hospitals (6.3% v. 2.8%; p=0.014). Using the PEN-FAST tool, only 10.4% (n=5/48)of self-reported BLA patients were classified as high risk for true penicillin hypersensitivity. Antibiotics were prescribed to 70.8% (n=34/48)of self-reported BLA patients, with 64.7% (n=22/34) receiving a beta-lactam. Despite three attempts to contact patients for de-labelling atthe allergy clinic, only 3/36 underwent in vivo testing, with no positive results, and 1 patient proceeded to a negative oral challenge.Conclusion. Unlike HICs, self-reported BLA is low among inpatients in SA. The majority of those who self-reported BLA were low risk fortype 1 hypersensitivity, but outpatient de-labelling efforts were largely unsuccessful.

Abstract: Rising oceanic temperatures create more frequent coral bleaching events worldwide and as such there exists a need for rapid, non-destructive survey techniques to gather greater and higher definition information than that offered by traditional spectral based monitoring systems. Here, we examine thermally induced laboratory bleaching of Montipora capricornis and Montipora confusa samples, utilising hyperspectral data to gain an understanding of coral bleaching from a spectral standpoint. The data revealed several characteristic spectral peaks that can be used to make health determinations. The fluorescence peaks are attributed to fluorescent proteins (FPs) and Chlorophyll-a fluorescence. The reflectance peaks can be attributed to Chlorophyll absorption and accessory pigments such as Peridinin and Diadinoxanthin. Each characteristic spectral peak or ‘marker’ allows for observation of each aspect of coral health and hence, simultaneous monitoring of these markers using hyperspectral imaging techniques provides an opportunity to better understand the processes occurring during bleaching and the rates at which they occur relative to one another.

33. Immediate sequential bilateral cataract surgery: patient perceptions and preferences

Abstract: BACKGROUND: Recent national data suggests that less than 0.5% of NHS cataract patients undergo immediate sequential bilateral cataract surgery (ISBCS). Since ISBCS improves service efficiency, increasing its practice may help tackle the ever-growing burden of cataract in the UK, and reduce the COVID-19 cataract backlog. Surgeon attitudes are known to be a significant barrier to increasing the practice of ISBCS. However, little is known about patient perceptions of ISBCS. METHODS: Patients at cataract clinics across three NHS hospital sites were recruited to complete an investigator-led structured questionnaire. Open-ended and closed-ended questions were used to assess awareness of ISBCS, willingness to undergo ISBCS and attitudes towards ISBCS. RESULTS: Questionnaires were completed by 183 patients. Mean participant age was 70.5 (9.9) years and 58% were female. Forty-three percent were aware of ISBCS, chiefly via clinic staff. Just over a third would choose ISBCS if given the choice, and participants that perceived they were recommended ISBCS were more likely to opt for it. The most common motivator and barrier to uptake of ISBCS was convenience and the perceived risk of complications in both eyes respectively. Concerns related to the recovery period were common, including misunderstandings, such as the need to wear eye patches that obscure both eyes. CONCLUSIONS: Our study indicates that significantly more NHS patients would be willing to undergo ISBCS if given the choice. The reluctance of surgeons to recommend ISBCS and patient misunderstandings regarding the recovery period may be limiting its uptake.

Abstract: Limb buds develop as bilateral outgrowths of the lateral plate mesoderm and are patterned along three axes. Current models of proximal to distal patterning of early amniote limb buds suggest that two signals, a distal organizing signal from the apical epithelial ridge (AER, Fgfs) and an opposing proximal (retinoic acid [RA]) act early on pattern this axis.Transcriptional analysis of stage 51 Xenopus laevis hindlimb buds sectioned along the proximal-distal axis showed that the distal region is distinct from the rest of the limb. Expression of capn8.3, a novel calpain, was located in cells immediately flanking the AER. The Wnt antagonist Dkk1 was AER-specific in Xenopus limbs. Two transcription factors, sall1 and zic5, were expressed in distal mesenchyme. Zic5 has no described association with limb development. We also describe expression of two proximal genes, gata5 and tnn, not previously associated with limb development. Differentially expressed genes were associated with Fgf, Wnt, and RA signaling as well as differential cell adhesion and proliferation.We identify new candidate genes for early proximodistal limb patterning. Our analysis of RA-regulated genes supports a role for transient RA gradients in early limb bud in proximal-to-distal patterning in this anamniote model organism.

Abstract: Bortolanite (IMA 2021–040a), ideally Ca2(Ca1.5Zr0.5)Na(NaCa)Ti(Si2O7)2(FO)F2, is a rinkite-group (seidozerite supergroup) TS-block mineral from Poços de Caldas massif, Minas Gerais, Brazil. Associated minerals are götzenite, nepheline, alkali feldspar, aegirine, natrolite, analcime, and manganoan pectolite. Bortolanite shows complex compositional zoning with götzenite and is visually indistinguishable from götzenite. Bortolanite is pale-yellow to brown and has a vitreous luster. Cleavage is perfect parallel to {100}. Mohs hardness is 5. Bortolanite fluoresces weak yellow under ultraviolet light (100–280 nm). Dcalc. = 3.195 g/cm3. Bortolanite is biaxial (+) with refractive indices (λ = 589.3 nm) α = 1.673(2), β = 1.677(2), γ = 1.690(2); 2Vmeas. = 56(2)°, 2Vcalc. = 58.4°. Chemical analysis by electron microprobe gave Nb2O5 1.07, HfO2 0.20, ZrO2 6.70, TiO2 9.94, SiO2 32.49, Gd2O3 0.12, Nd2O3 0.37, Ce2O3 1.25, La2O3 0.65, Y2O3 0.31, FeO 0.59, MnO 1.46, CaO 31.15, Na2O 8.36, F 6.95, O=F –2.93, sum 98.68 wt.%. The empirical formula based on 18 (O + F) apfu is (Ca1.88La0.03Ce0.06Nd0.02Gd0.01)Σ2[Ca1.56(Zr0.41Hf0.01Y0.02)Σ0.44]Σ2(Na0.85Ca0.15)Σ1(Na1.18Ca0.60Mn0.16Fe2+0.06)Σ2(Ti0.94Nb0.06)Σ1(Si4.07O14)(O1.24F0.76)Σ2F2, Z = 1. The crystal structure of bortolanite is a framework of TS (Titanium-Silicate) blocks [structure type B3(RG)], where the TS block consists of HOH sheets (H-heteropolyhedral, O-octahedral). The TS block exhibits linkage and stereochemistry typical for the rinkite group where Ti (+ Nb + Zr) = 1 apfu. The O sheet is composed of Ti-dominant MO(1) octahedra, [8]Na-dominant MO(2) polyhedra and (Na,Ca) MO(3) octahedra. In the H sheet in bortolanite, Si2O7 groups link to (Ca1.5Zr0.5) MH and Ca-dominant AP octahedra. Along a, TS blocks link directly through common edges of MH and AP polyhedra and common vertices of MH, AP, and Si polyhedra of the H sheets belonging to two TS blocks. The name bortolanite is after the locality: the Bortolan quarry in the Poços de Caldas massif, Brazil. Bortolanite is isostructural with three rinkite-group minerals: fogoite-(Y), hainite-(Y), and götzenite.

Abstract: Pollen development, from unicellular microspores to anthesis, is a complex process involving the coordinated specification, differentiation and functions of different cell types. Key to understanding this development is identifying the genes expressed at precise stages of development. However, transcriptomic studies on pollen prior to anthesis are complicated by the inaccessible nature of pollen developing in the anther and the resistant pollen wall. To assist with understanding gene expression during pollen development we have developed a protocol to perform RNA-Seq on pollen isolated from a single anther (SA RNA-Seq). The protocol involves removing pollen from a single anther for analysis and viewing the remaining pollen to determine the developmental stage. The isolated pollen is chemically lysed and mRNA isolated from the lysate using an oligo-dT column before library preparation. Here, we report on the development and testing of our method and the generation of a transcriptome for three stages of pollen development from Arabidopsis (Arabidopsis thaliana) and two stages from male kiwifruit (Actinidia chinensis). This protocol enables the transcriptome of precise developmental stages of pollen to be analyzed, and uses a small number of plants, potentially facilitating studies that require a range of treatments or the analysis of the first generation of transgenic plants.

Abstract: Uvite, CaMg3(Al5Mg)(Si6O18)(BO3)3(OH)3(OH), is a new mineral of the tourmaline supergroup. It occurs in the Facciatoia quarry, San Piero in Campo, Elba Island, Italy (42°45′04.55″N, 10°12′50.89″E) at the centre of a narrow (2–3 cm wide) vein composed of aggregates of dark brown to black tourmaline, penetrating (magnesite + dolomite)-rich hydrothermally altered metaserpentinite. Crystals are euhedral and up to 1 cm in size, brown with a vitreous lustre, conchoidal fracture and grey streak. Uvite has a Mohs hardness of ~7½, a calculated density of 3.115 g/cm3 and is uniaxial (–). Uvite has trigonal symmetry, space group R3m, a = 15.9519(10) Å, c = 7.2222(5) Å, V = 1597.3(1) Å3 and Z = 3. The crystal structure was refined to R1 = 1.77% using 1666 unique reflections collected with MoKα X-rays. The closest end-member compositions of valid tourmaline species are fluor-uvite and feruvite, to which uvite is related by the substitutions W(OH)– ↔ WF– and YMg2+ ↔ YFe2+, respectively. The occurrence of a solid-solution between uvite and magnesio-lucchesiite, according to the substitution ZMg2+ + W(OH)– ↔ ZAl3+ + WO2–, is supported by experimental data. The new mineral was approved by the IMA–CNMNC (IMA 2019-113). Uvite from Facciatoia formed by the reaction between B-rich fluids, released during the crystallisation process of LCT pegmatites, and the surrounding metaserpentinites, altered by contact metamorphism in the aureole of the Miocene Mt. Capanne monzogranitic pluton.

42. Mass Bias Corrections for Hydrogen and Oxygen Isotope Analysis of Tourmaline by Secondary Ion Mass Spectrometry

Abstract: Hydrogen (δ2H) and oxygen (δ18O) stable isotopes are used to trace fluid sources, metals, and contaminants in the environment and Earth's subsurface. Tourmaline-supergroup minerals provide an opportunity to quantify both δ2H and δ18O from the same grain using in situ analytical techniques (e.g., Secondary Ion Mass Spectrometry – SIMS). These minerals occur in a wide variety of geological environments and have a wide range of chemical compositions. However, large differences in chemical composition are problematic during SIMS analysis, as instrumental mass fractionation (IMF) often varies with the chemical composition of the mineral. Therefore, calibration models derived by analyzing tourmalines of different chemical composition must be developed for accurate analysis by SIMS. Hydrogen and oxygen isotope analysis was done on six reference tourmaline samples using a CAMECA 7f SIMS instrument operating at extreme energy filtering. Spot-to-spot repeatability for tourmalines was in the range 4–5‰ and 0.6–1.0‰ for δ2H and δ18O, respectively. There is a strong correlation between IMF and several elements (B, Si, Ca, Fe, and Fe#). Iron content is the most robust predictor of IMF, and we report two calibration curves for the correction of δ2H and δ18O measured by SIMS using reference tourmaline crystals with different Fe contents, ranging from 0.00 to 14.00 wt.% Fe. This is the first calibration curve used to correct for the fractionation of hydrogen isotope ratios in tourmaline as measured by SIMS. Tourmaline-supergroup minerals require a suite of at least three, with a range of Fe content, to ensure accurate and precise H and O analysis by SIMS. Crystallographic orientation effects were not observed for these tourmalines.

43. An evaluation of newborn hearing screening brochures and parental understanding of screening result terminology

Abstract: To assess the suitability of newborn hearing screening brochures by evaluating current state-level brochures and pregnant people’s understanding of screening result terminology. In Study 1, state-level brochures were evaluated based on readability, design, picture appropriateness, and use of the word “refer.” In Study 2, pregnant people completed a questionnaire that queried their understanding of and expected anxiety about three newborn hearing screening outcomes (“refer,” “did not pass,” and “pass”). In Study 1, 59 newborn hearing screening brochures were analysed. In Study 2, 43 pregnant people completed surveys during a prenatal appointment. Most of the brochures were found deficient on at least one element. Thirty percent of brochures used the word “refer” to indicate a hearing screening failure; yet, fewer than half of participants understood its meaning. Ratings of expected anxiety were highest in response to the term “did not pass.” Based on four study criteria of brochure suitability, 88% of available state-level newborn hearing screening brochures should be modified to make them readily understandable by a broad educational demographic. Discretion in use of the term “refer” should be made when indicating screening results, because the term is not readily understood.

Abstract: Shkatulkalite, ideally Na5TiNb2(Si2O7)2O3F(H2O)7, is a lamprophyllite-group (seidozerite supergroup) TS-block mineral from the Lovozero massif, Kola Peninsula, Russia. The crystal structure of shkatulkalite was refined as triclinic, space group P, a 5.464(1), b 7.161(1), c 15.573(1) Å, α 90.00(3), β 95.75(3), γ 90.00(3)°, V 606.3(4) Å3, R1 = 7.26%. The previously given chemical analysis has been modified to better fit the crystal structure: Nb2O5 24.15, TiO2 11.35, SiO2 27.22, BaO 1.15, SrO 2.20, MnO 1.68, CaO 0.46, K2O 0.29, Na2O 14.78, H2O 15.27, F 1.61, O = F −0.68, sum 99.48 wt.%; H2O was determined from structure-refinement results. The empirical formula was calculated on 25.27 (O + F) apfu (in accord with the crystal structure): (Na1.40Sr0.19Ba0.07K0.05)Σ1.71(Na2.86Mn0.10Ca0.07)Σ3.03(Nb1.62Ti1.27Mn0.11)Σ3Si4.03O24.52H15.11F0.75, Z = 1. The ideal structural formula is as follows: Na2Nb2Na3Ti(Si2O7)2O2(FO)(H2O)4(H2O)3. The crystal structure of shkatulkalite [basic structure type B5(LG)] is a combination of a TS (titanium-silicate) block and an I (intermediate) block. The TS block consists of HOH sheets (H-heteropolyhedral, O-octahedral). The TS block exhibits linkage and stereochemistry typical for the lamprophyllite group where Ti (+ Nb + Fe3+ + Mg) = 3 apfu. The O sheet is composed of Ti-dominant MO(1) and Na-dominant MO(2,3) octahedra. In the H sheet in shkatulkalite, Si2O7 groups link to Nb-dominant MH octahedra. The AP site occurs in the plane of the H sheet and splits into AP(1) and AP(2) sites, occupied by Na at 70% and Sr (less Ba and K) at 11%. The I block consists of H2O groups. The I block of shkatulkalite is topologically identical to those in the derivative structures of kazanskyite and nechelyustovite. The structure of the lamprophyllite-group mineral shkatulkalite has a counterpart structure in the murmanite group (Ti = 4 apfu): kolskyite, ideally Na2CaTi4(Si2O7)2O4(H2O)7 [basic structure type B7(MG)]: the two structures have TS blocks of different topology and similar I blocks, mainly occupied by H2O groups.

Abstract: Activation of the EGF receptors EGFR (ErbB1) and HER2 (ErbB2) drives the progression of multiple cancer types through complex mechanisms that are still not fully understood. In this study, we report that HER2 expression is elevated in bone metastases of prostate cancer independently of gene amplification. An examination of HER2 and NF-κB receptor (RANK) coexpression revealed increased levels of both proteins in aggressive prostate tumors and metastatic deposits. Inhibiting HER2 expression in bone tumor xenografts reduced proliferation and RANK expression while maintaining EGFR expression. In examining the role of EGFR in tumor-initiating cells (TIC), we found that EGFR expression was required for primary and secondary sphere formation of prostate cancer cells. EGFR expression was also observed in circulating tumor cells (CTC) during prostate cancer metastasis. Dual inhibition of HER2 and EGFR resulted in significant inhibition of tumor xenograft growth, further supporting the significance of these receptors in prostate cancer progression. Overall, our results indicate that EGFR promotes survival of prostate TIC and CTC that metastasize to bone, whereas HER2 supports the growth of prostate cancer cells once they are established at metastatic sites. Abstract: Using human tumor and cDNA microarray technology, we have recently shown that the ADAM15 disintegrin is significantly overexpressed during the metastatic progression of human prostate cancer. In the current study, we used lentiviral-based short hairpin RNA (shRNA) technology to down-regulate ADAM15 in the metastatic prostate cancer cell line, PC-3. ADAM15 down-regulation dramatically attenuated many of the malignant characteristics of PC-3 cells in vitro and prevented the s.c. growth of PC-3 cells in severe combined immunodeficient (SCID) mice. By inhibiting the expression of ADAM15 in PC-3 cells, we showed decreased cell migration and adhesion to specific extracellular matrix proteins. This was accompanied by a reduction in the cleavage of N-cadherin by ADAM15 at the cell surface. Fluorescence-activated cell sorting analysis revealed reduced cell surface expression of the metastasis-associated proteins αv integrin and CD44. Furthermore, matrix metalloproteinase 9 secretion and activity were abrogated in response to ADAM15 reduction. In an in vitro model of vascular invasion, loss of ADAM15 reduced PC-3 adhesion to, and migration through, vascular endothelial cell monolayers. Using an SCID mouse model of human prostate cancer metastasis, we found that the loss of ADAM15 significantly attenuated the metastatic spread of PC-3 cells to bone. Taken together, these data strongly support a functional role for ADAM15 in prostate tumor cell interaction with vascular endothelium and the metastatic progression of human prostate cancer.
