We aimed to evaluate the treatment with estradiol benzoate (EB) or 17β-estradiol (E2) associated with progesterone (P4) for resynchronization of ovulation 14 days after timed artificial insemination (TAI). In Experiment 1 (Exp. 1), Nelore heifers were submitted to TAI (D0). On D14, the animals received an intravaginal P4 device and were randomly assigned to one of three groups: control (no treatment; n = 17); EB (1 mg EB; n = 17); and E2+P4 (1 mg E2 + 9 mg P4; n = 18). Ultrasonography evaluations were performed daily from D14 to D22 to map follicular and luteal dynamics. On D22, the P4 devices were removed and non-pregnant (NP) animals were determined using corpus luteum blood flow Doppler ultrasonography. In Exp. 2, 1295 beef heifers were resynchronized and randomly allocated to the same experimental groups as described in Exp. 1. On D22, the largest follicle (LF) was measured in NP and a second TAI was performed on D24. In a subset of heifers (n = 337), an estrus detection patch was used between D22 and D24 to monitor estrus expression and the LF was measured at D24. Confirmatory diagnosis of pregnancy was performed between D37-67 and D43-67 after first and second TAI, respectively. In Exp 1, the proportion of heifers with a synchronized follicular wave emergence (from 3 to 5 days after treatment) was greater (P 0.05) in the EB group (93.8%) than in the control (62.5%) and E2+P4 (64.7%) groups. Structural luteolysis occurred earlier (P 0.05) in the EB and E2+P4 groups than in the controls. The pregnancy rate after first TAI did not differ (P 0.1) among the groups at D22 and at confirmatory diagnosis in both experiments. In Exp 2, the potential pregnancy loss between D22 and D37-67 was similar (P 0.1) in the control (19% [36/185]), EB (15% [28/182]) and E2+P4 (15% [28/184]) groups. The LF diameter (mm) on D22 was greater (P 0.05) in the control group (11.9 ± 0.1) than in EB (11.3 ± 0.1) and E2+P4 (11.5 ± 0.1). No difference (P 0.1) was observed in the proportion of heifers detected in estrus, but LF growth rate (mm/day) between D22 and D24 was greater (P 0.05) in EB group (0.9 ± 0.08) than in control (0.6 ± 0.07) and E2+P4 (0.7 ± 0.09) groups. The pregnancy rate for the second TAI was greater (P 0.05) in the EB group (47% [94/200]) than in the control (37% [76/203]), but did not differ (P 0.1) from the E2+P4 group (43% [93/214]). In conclusion, the treatment with 1 mg EB or 1 mg E2 + 9 mg P4 at 14 days post-TAI anticipates luteolysis in NP heifers but does not compromise pregnancy. The EB treatment induces a new synchronized follicle wave emergence and increases the pregnancy rate of resynchronized NP heifers.

Palladium nanoparticles were evaluated as a potential chemical modifier for lead determination by high-resolution continuum source graphite furnace atomic absorption spectrometry. Hydrophilic palladium nanocubes and palladium nanospheres were synthesised with sizes varying from 3 to 22 nm. Positive effects in terms of sensitivity and thermal stability upon the use of palladium nanoparticles were observed, especially using the 3 nm spherical-shaped. The mass of modifier influenced the thermal stability and relative signal intensity of the analyte, although the effectiveness of the modifier appeared to be suppressed to an important extent in the presence of matrix components of the leached extracts obtained from petroleum wastes. The optimised pyrolysis and atomisation temperatures were, respectively, 800 °C and 2300 °C, using 200 ng of 3 nm palladium nanoparticles. The limit of detection (4 µg L−1) and the precision (better than 18 %) were considered satisfactory for the proposed analysis. The method accuracy, evaluated by recovery tests and by comparison with the results obtained using inductively coupled plasma mass spectrometry, was proven adequate to enable the determination of Pb in leached extracts from petroleum waste. The method was successfully applied to determine lead in leached extracts obtained from oily sludge and drill cuttings and the obtained concentrations ranged from −1.

The effects of 17β-estradiol (E2) or estradiol benzoate (EB) on PGF2α release were studied in bred-non-pregnant and pregnant Nelore beef heifers. The day of timed artificial insemination (TAI) was designated day 0 (D0), and a single treatment was given on D14. All heifers also received an intravaginal P4 device on D14, and were randomly assigned to three groups: Control (C, P4 device only, n = 12); E2 (1 mg E2 + 9 mg P4, n = 10); or EB (1 mg, n = 10). Blood samples were collected hourly for 8 hours after treatment (Hours 0-8) to measure plasma concentrations (pg/mL) of a PGF2α metabolite (PGFM). The P4 device was removed on D22 and pregnancy was diagnosed on D28. Pregnancy rate was not different among groups (C, n = 7/12; E2, n = 5/10; EB, n = 5/10). More (P0.05) heifers had a CV-identified prominent PGFM pulse (peak of100 pg/mL) in E2 group (6/10) than in EB (1/10) and C (0/12) groups. Hourly concentration of PGFM for Hours 0 to 8 showed significant effects of group and hour and an interaction of group by hour but did not show an interaction of group or hour with pregnancy status. In preliminary post-hoc analyses, PGFM concentrations during Hours 0 to 8 and pulse characteristics were analyzed within each pregnancy status. For the non-pregnant heifers, a group-by-hour interaction was detected tentatively indicating an increase (P0.005) in PGFM concentrations in E2 group from Hours 4 to 6 and in EB group at Hours 5 and 6. Maximum PGFM concentration during Hours 0 to 8 did not differ (P0.1) between E2 (124 ± 23) and EB (110 ± 30) groups, but was greater (P0.05) in each group than in C (32 ± 3). Furthermore, PGFM concentrations of pulses at the peak, amplitude, and area under pulse curve (pg/mL/h) were greater (P0.05) in E2 group than in C group whereas the EB group did not differ (P0.1) from the other groups. For pregnant heifers, no effects of group, hour, or their interaction were detected in PGFM concentrations during the hourly sessions, except that maximum PGFM concentration was greater (P0.05) in E2 than in EB and C groups. In addition, the number of prominent pulses was greater in E2 group than in Control or EB groups. In conclusion, PGFM increased earlier and in greater concentration combined for bred-non-pregnant and pregnant heifers treated 14 days after TAI with 1 mg E2 plus 9 mg P4 than with 1 mg EB. Tentatively, a positive effect for each of E2 and EB on PGFM concentrations was attenuated in pregnant heifers.

Marine mammals are considered excellent ocean health sentinels and are ubiquitously exposed to chemical contaminants worldwide. The Guiana dolphin (Sotalia guianensis) is a near-threatened dolphin species from Brazil with unknown population size data. This indicates the need for assessments regarding deleterious effects that may arise from exposure to chemical contamination, especially metals. After entry in the organism, these compounds are subject to internal subcellular compartmentalization, which in turn alters their bioavailability. However, almost no assessments regarding subcellular metal contents in marine mammals are available. In this context, metal compartmentalization was determined in three subcellular fractions for three toxic elements, Cd, Hg and Pb, by inductively coupled plasma mass spectrometry (ICP-MS) in Guiana dolphin kidney and liver samples from Southeastern Brazil. Differential metal-detoxification mechanisms were observed for the three elements, where metallothionein (MT) detoxification was postulated for only for Pb, while Cd and Hg were poorly associated to MT, and mostly present in the insoluble fraction, indicating low bioavailability. This is the first report on subcellular metal compartmentalization in Guiana dolphins and indicates that critical biochemical detoxification data is obtained through subcellular fraction analyses in marine mammals. This indicates an emerging study field for this type of assessment, which may, in turn, aid in conservation efforts.

Taurine bulls are highly susceptible to heat stress, leading to increased oxidative stress (OS) and impaired sperm viability. Polyunsaturated fatty acids (PUFAs) supplementation can be an alternative to improve semen quality, which also results in more sperm susceptibility to lipid peroxidation. Moreover, this deleterious effect can be exacerbated in animals affected by heat stress. Vitamin E is a key antioxidant that counteracts lipid peroxidation of sperm membrane caused by OS. Thus, combining PUFAs with vitamin E may improve sperm quality. In this context, this study aimed to evaluate the effect of interaction between PUFAs and vitamin E on sperm quality in Bos taurus bulls under testicular heat stress. Sixteen taurine bulls under testicular heat stress were randomly assigned in four groups: Control, Vitamin E, PUFA, and PUFA + Vitamin E. All groups lasted for 60 days. Samples were cryopreserved/thawed and analyzed for motility variables (CASA), membrane and acrosome integrity, mitochondrial activity, susceptibility to oxidative stress, DNA integrity, and sperm-binding capacity. Results showed that vitamin E had a beneficial effect on some sperm characteristics, whereas PUFA supplementation had an adverse effect when the two treatments were evaluated separately. Finally, the association between PUFAs and vitamin E did not improve sperm quality.

Com a busca por alimentos mais saudaveis, ha a importância de um produto de baixos indices calorificos que associe sabor ao bem-estar. Sendo assim, como se da a entrada de pequenas empresas produtoras de requeijao saborizado light ao mercado das grandes industrias? Objetivamos analisar o comportamento de mercado do produto em questao e quais sao suas dificuldades de modo que esta pesquisa sirva de embasamento a pequenos novos produtores do ramo de laticinios. Realizamos uma analise qualitativa de dados por meio de pesquisas bibliograficas, como artigos cientificos e entrevista, visando abordar o conteudo de maneira mais consistente possivel. A analise mercadologica em questao e fruto de uma pesquisa da Universidade Federal do Rio Grande do Norte – UFRN. Dito isso, concluimos que a busca por inovacoes por parte das empresas – micro e macro – surte efeitos positivos ao mercado, como no caso da adequada porcentagem de Extrato Seco Desengordurado (ESD) do requeijao cremoso light, que garantem textura, cor e sabor associados ao baixo teor de gordura necessario.

SummaryThe aim of this study was to compare different concentrations of soy lecithin (LEC0.01%, LEC0.05% and LEC0.1%) with egg yolk (Control) in cooling extenders during the storage of semen at 5ºC for 5 days. Twelve dogs (n = 12) were selected, and semen was cooled and assessed after 2, 24, 48, 72, 96 or 120 h. At each time point, sperm were analyzed for kinetic patterns (using computer-assisted sperm analysis), mitochondrial activity (3′3- diaminobenzidine assay), lipid peroxidation (TBARS assay), DNA fragmentation (SCSA®) and plasma and acrosome membrane integrity (eosin/nigrosin and fast green/rose Bengal stains, respectively). The Control group (1814.4 ± 197.2) presented the highest rates of lipid peroxidation at 120 h. Conversely, progressive motility (42.8 ± 4%), linearity (45.4 ± 1%), and VAP (88 ± 3%) were higher in the Control group. In addition, there was lower mitochondrial activity in the Control group at 72 h. Therefore, our data show that lecithin used at these concentrations was not able to maintain sperm viability at as high qualities as would egg yolk. Moreover, the decrease in high mitochondrial activity and the persistence of sperm motility may indicate a compensatory mechanism in canine spermatozoa (i.e., glycolytic pathway). Furthermore, these higher lipid peroxidation indexes could indicate the necessity for future therapy using extenders and antioxidants over a long cooling time for dog sperm.

Semen biotechniques may impair sperm quality due to excessive production of reactive oxygen species (ROS). Additionally, products of the oxidative reaction, especially involving lipids (e.g., malondialdehyde - MDA), may be even more harmful to sperm. Carnosine, previously reported to be present in seminal plasma of several species, may be a key factor on sperm tolerance to biotechniques by counterattacking the deleterious influence of MDA. Therefore, the aim of this study was to measure the levels of carnosine present in equine seminal plasma and relate these findings with sperm function and oxidative status during cooling and cryopreservation. Thus, semen samples were collected from 40 stallions in duplicate (N = 80) and then submitted to cooling and cryopreservation. Samples were then allocated into groups of high and low tolerance to refrigeration and cryopreservation (bad cooler and good cooler/bad freezer and good freezer, respectively), and in groups of different concentrations of carnosine (High, Medium-high, Medium-low and Low carnosine). Samples were evaluated for sperm kinetics patterns, function of sperm structures and oxidative status. In good cooler samples, it was observed higher concentrations of carnosine (Good cooler: 224.98 ± 19.16 ng/mL; Bad cooler: 159.72 ± 15.99 ng/mL; p = 0.0056), ROS production (Good cooler: 26.40 ± 18.33%; Bad cooler: 18.33 ± 1.84%; p = 0.001) and lipid peroxidation rates (Good cooler: 193.23 ± 18.22 ng/mL; Bad cooler: 131.92 ± 12.25; p = 0.0064). Groups of samples with higher carnosine concentrations had lower levels of malondialdehyde (High: 79.33 ± 6.72 ng/mL; Medium-high: 140.45 ± 11.70 ng/mL; Medium-low: 202.57 ± 16.30 ng/mL and Low: 231.02 ± 32.35 ng/mL; p 0.05), demonstrating that carnosine was effective in removing lipid peroxidation products. Due to the removal of seminal plasma during the cryopreservation process, no differences occurred in carnosine levels between bad and good freezer groups. In this context, this study provides relevant data for future therapies using carnosine during cryopreservation, aiming to replace the levels lost due to the necessary removal of seminal plasma.

Semen cryopreservation is an essential biotechnology in canine reproduction and during the cryopreservation process commonly egg yolk are used. The discrepancy in the egg yolk composition and the potential risk of disease dissemination are obstacles for semen exportation and use. Therefore, studies aiming to substitute egg yolk are extremely important. In this context, soy lecithin contains a low-density lipoprotein fraction, is an interesting alternative. Thus, the objective of this study was to compare extenders based on soy lecithin (several concentrations and forms) with egg yolk during the cryopreservation process of dog sperm. For this purpose, we used twelve dogs. Semen was evaluated at different time points (after refrigeration, glycerolization, and thawing), by motility analysis (CASA) and functional tests (e.g., membrane integrity-eosin/nigrosin, acrosome integrity-fast green/Bengal rose, mitochondrial activity-3'3 diaminobenzidine, Chromatin susceptibility to acid-induced denaturation-SCSA, and susceptibility to oxidative stress-thiobarbituric acid reactive substances). The results indicated that egg yolk and lower concentrations of lecithin had similar effects on mitochondrial activity and motility. Thus, soy lecithin is a potentially viable alternative to egg yolk for the cryopreservation of dog semen.

We aimed to compare three methods to detect pregnancy based on interferon-tau stimulated gene expression in peripheral blood polymorphonuclear cells and Doppler ultrasonography (Doppler-US) 20 days after timed AI and the concentrations of pregnancy associated glycoproteins (PAGs) 25 days after timed AI. Our second objective was to compare interferon-tau stimulated genes and PAGs as early embryo loss (EEL) predictors. Nelore cows (n=144) and heifers (n=103) were submitted to timed AI (Day (D) 0). On D20, polymorphonuclear cells were isolated from blood samples by Ficoll gradient (GE Healthcare), and the RNA was extracted. Expression of the interferon-tau stimulated gene (OAS-1) was quantified by qPCR and normalized to reference genes (GAPDH and ACTB). On D25, blood was collected from the jugular vein and PAG concentrations were measured by an in-house enzyme-linked immunosorbent assay. Pregnancy diagnoses were performed on D20 by luteolysis detection with Doppler-US and on D30 by detection of an embryo by heartbeat. Animals were classified as pregnant (P; fetus on D30), non-P (NP; no active corpus luteum on D20), and EEL (active corpus luteum on D20 but NP on D30). Expression of OAS-1 and PAG concentrations were analysed by analysis of variance using PROC MIXED of SAS considering the effects of group, category, and their interaction. This study characterised the expression of interferon (IFN)-τ-stimulated genes (ISGs) and Type I IFN receptors in circulating polymorphonuclear cells (PMNs) of beef heifers and compared it with expression in peripheral blood mononuclear cells (PBMCs) up to Day 20 of gestation. Nelore heifers (n=26) were subjected to fixed-time AI (FTAI) on Day 0. PMNs and PBMCs were isolated on Days 0, 10, 14, 16, 18 and 20 after FTAI. The abundance of target transcripts (ubiquitin-like protein (ISG15), 2′-5′-oligoadenylate synthetase 1 (OAS1), myxovirus resistance 1 (MX1), myxovirus resistance 2 (MX2), IFN receptor I (IFNAR1) and IFN receptor 2 (IFNAR2)) was determined using real-time quantitative polymerase chain reaction and compared between pregnant (n=8) and non-pregnant (n=9) females. In both PBMCs and PMNs, ISG15 and OAS1 expression was greater in pregnant than non-pregnant heifers on Days 18 and 20. There were no significant differences in the expression of ISGs between PBMCs and PMNs. A time effect on expression was found for IFNAR1 in PBMCs and IFNAR2 in PMNs, with decreased expression of both genes on Days 18 and 20. When the expression of these genes was compared between cell types only in pregnant heifers, IFNAR2 expression in PMNs had an earlier decrease when compared to its expression in PBMCs, starting from Day 18. In conclusion, PMNs do not respond earlier to the conceptus stimulus, and ISG15 and OAS1 expression in both PMNs and PBMCs can be used as a suitable marker for pregnancy diagnosis on Days 18 and 20. In addition, gestational status did not affect IFNAR1 and IFNAR2 expression, but IFNAR2 showed a distinct response between PMNs and PBMCs of pregnant heifers.

Seminal cryopreservation provides several advantages in horse breeding. However, improvement on post-thaw sperm survival is still necessary. One of the main factors known to impair post-thaw quality of stallion's sperm is the oxidative damage caused by reactive oxygen species (ROS). In this context, the aim of this study was evaluated the effects of ROS on stallion sperm and the possible influence of seminal plasma (SP). Toward this aim, 13 ejaculates from adult stallions (n = 13) were divided into two aliquots which were centrifuged (600g/10 minutes), and the SP was removed and reserved. Pellets were suspended in physiological saline solution (A) or SP (B). Each of these solutions was divided into four aliquots and subjected to challenge with different ROS (superoxide anion, hydrogen peroxide, and hydroxyl radical [OH−]) and the toxic by-product of lipid peroxidation (malondialdehyde [MDA]). Samples were then evaluated for the susceptibility to lipid peroxidation (thiobarbituric acid reactive substances) plasma membrane integrity (eosin–nigrosin), acrosome integrity (Fast Green/rose bengal), mitochondrial activity (3′3 diaminobenzidine), and DNA integrity. Samples incubated in the presence of SP were highly impaired by OH− with regard to motility, plasma membrane integrity, mitochondrial activity, and DNA integrity. On the other hand, in the absence of SP, MDA was highly deleterious, especially with regard to motility, plasma membrane integrity, and mitochondrial activity. Thus, these results indicate that SP may have an important role on the protection of stallion sperm against the damages caused by MDA, an important product of lipid peroxidation.

The species of genera Uroderma and Artibeus are medium-sized bats belonging to the family Phyllostomidae and subfamily Stenodermatinae (Mammalia, Chiroptera) from South America. They have a wide distribution in the Neotropical region, with two currently recognized species in Uroderma and approximately 20 species in Artibeus. These two genera have different rates of chromosome evolution, with Artibeus probably having retained the ancestral karyotype for the subfamily. We used whole chromosome paint probe sets from Carollia brevicauda and Phyllostomus hastatus on Uroderma magnirostrum, Uroderma bilobatum, and Artibeus obscurus. With the aim of testing the previous phylogenies of these bats using cytogenetics, we compared these results with published painting maps on Phyllostomidae. The genome-wide comparative maps based on chromosome painting and chromosome banding reveal the chromosome forms that characterize each taxonomic level within the Phyllostomidae and show the chromosome evolution of this family. Based on this, we are able to suggest an ancestral karyotype for Phyllostomidae. Our cladistic analysis is an independent confirmation using multidirectional chromosome painting of the previous Phyllostomidae phylogenies.

A moving bed bioreactor containing cubes of polyether foam immersed in a synthetic wastewater (an aqueous mixture of meat extract, yeast extract, dextrose, meat peptone, ammonium chloride, potassium chloride, sodium chloride, sodium bicarbonate, potassium mono-hydrogen-phosphate and magnesium sulphate) was used to evaluate bacterial growth and biomass yield parameters based on Monod's equation. The wastewater was supplied in the bottom of the equipment flowing ascending in parallel with a diffused air current that provided the mixing of the reactor content. Suspended and attached biomass concentration was measured through gravimetric methods. Good agreement was found between experimental kinetic parameters values and those obtained by other researchers. The only significant difference was the high global biomass content about 2 times the values obtained in conventional processes, providing high performance with volumetric loading rates up to 5.5 kg COD/m³/d.

1. The present study evaluated the participation of tumour necrosis factor-alpha (TNF-alpha) in the inflammatory and nociceptive responses evoked by carrageenan in the mouse paw. 2. The intraplantar injection of carrageenan (300 microg paw-1) induced a marked and biphasic paw oedema formation (peaks at 6 and 72 h), which was accompanied by a long-lasting mechanical allodynia (that remained elevated for up to 72 h) and a significant increase of myeloperoxidase (MPO) activity (peak at 6 h) in both Swiss and C57/BL6 mice. 3. The paw oedema, the elevation of MPO activity and to a lesser extent the mechanical allodynia elicited by carrageenan were found to be significantly reduced in TNF-alpha p55 receptor knockout mice. 4. Of interest, the systemic administration of an anti-TNF-alpha antibody produced a significant inhibition of paw oedema, mechanical allodynia and MPO activity. A noteworthy decrease in inflammatory and nociceptive responses caused by carrageenan was also observed when mice were previously treated with the preferential inhibitor of TNF-alpha synthesis, thalidomide. 5. The present results clearly indicate that the proinflammatory cytokine TNF-alpha plays a critical role in the oedema formation, as well as in the mechanical allodynia and the neutrophil migration, following carrageenan administration into the mouse paw. Intraplantar injection of carrageenan in mice could constitute a useful model for assessment of the in vivo effects of potential inhibitors of TNF-alpha-related pathways.

The overhead distribution lines with bare conductors for medium and low voltage circuits are being substituted due to their failure rate and security level, and not meeting customer needs, particularly in adverse areas, i.e., heavy trees and high isokeraunic levels. The failure rate in CEMIG's medium voltage overhead distribution lines, in urban areas, is 0.9 failure/km-year. More and more customers ask for better services and lower costs. These types of systems are greatly influenced by external causes such as lightning, rain, trees, collision, and vandalism. In Brazil, based on the US experience, the aerial spacer cable system has been shown as the best system to improve its reliability. The low maintenance cost, easy operation, high security level and the attractive cost-benefits have justified its use, mainly in urban areas, by CEMIG, a Brazilian Utility. In the MV spacer cable system were included other technologies, such as CSP transformer, SF/sub 6/ switch, medium and low voltage polymeric arresters, low voltage multiplex cable, and wedge and piercing connectors. CEMIG's spacer cable system has been getting an average failure rate of 0.04 failure/km-year. This paper describes the line design, equipment, standards and also, the maintenance methods in live-line and field experiences of spacer cable system in our company, with the technical support of Hendrix Wire & Cable and Wirex Cable, Brazil. Other utilities in Brazil are now using this system, to meet one of the government's requirements of improving reliability. It is a good alternative for companies searching for high reliability for their distribution electrical system.

We present herein clinical, histological and magnetic resonance imaging (MRI) findings in a patient with Fukuyama-type congenital muscular dystrophy (FCMD). He is the first case report in the Japanese population living in Brazil.The child presented with neonatal hypotonia, delayed motor abilities and speech, seizures, cerebral and cerebellar gyrus abnormalities with signal intensity change in the white matter by MRI, high serum level of creatinephosphokinase (CK), and dystrophic skeletal muscle with normal merosin, alpha-sarcoglycan and dystrophin expression. The fukutin gene study showed one founder 3-kb retrotransposal insertion in the 3'-non-coding region, and in the other allele no mutation was detected after screening all exons and flanking introns by sequencing.This case report emphasizes the importance to consider FCMD in Japanese people living in other countries.

In Brazil, more than 80% of venomous snakebites are caused by Bothrops and about 10% by Crotalus. This study evaluated 133 reported cases that occurred between 1994 and 1996 in the 52 municipalities covered by the Pouso Alegre Regional Health Center in southern Minas Gerais State. Most of the patients were male (89.5%). The most frequently attacked age bracket was that of people in their twenties, and the most frequently bitten anatomical regions were the lower limbs (77.7%), principally the feet (34.6%). Of the 124 cases stating the snake genus, 62.9% were caused by Crotalus and 37.1% by Bothrops. The conclusion of this study is that although the epidemiology of snakebites in Minas Gerais State is similar to other regions of the country, the percentage of Crotalus bites is much higher.

Bos taurus bulls, when raised under tropical conditions, are highly susceptible to heat stress, which leads to impaired semen quality, leading to significant economical losses because, in these regions, the reproductive mounting season occurs mainly during the summer. Previous studies have indicated that oxidative stress (i.e. attack by reactive oxygen species) may be the main mechanism of sperm damage in such conditions. Therefore, treatment with antioxidants may be an important alternative to improve semen quality in heat-stressed B. taurus bulls. The objective of the present study was to evaluate whether the treatment with vitamin E, an important antioxidant, could improve sperm quality in insulated bulls. Towards this aim, eight adult Holstein bulls were submitted for semen collection, and the sperm was submitted for motility evaluation by computer-assisted sperm analysis (Ivos, Hamilton Thorne Inc., Beverly, MA, USA), examination of membrane and acrosomal integrity (eosin/nigrosin and fast green/bengal rose stain, respectively), mitochondrial activity (diaminobenzidine stain; full mitochondrial activity or no mitochondrial activity), and sperm susceptibility to oxidative stress (thiobarbituric acid-reactive substances). However, studies indicate that cryopreservation may lead to increased oxidative stress which may cause structural damage to biomolecules, DNA, lipids, carbohydrates, and proteins, as well as other cellular components. Previous studies performed by our group indicate fresh goat sperm is highly susceptible to the attack of hydrogen peroxide. Therefore, the treatment with hydrogen peroxide scavengers would be an alternative to improve post-thaw sperm quality in goats. The aim of the present study was to evaluate the efficiency of catalase, an important hydrogen peroxide scavenger, to improve post-thaw quality in cryopreserved goat semen samples. Sperm recovery from the caudae epididymides can be advantageous for preserving semen of endangered animal species. In this context, the domestic cat is a suitable model for the study of sperm physiology in endangered feline species and the research on epididymal sperm preservation combined with the use of reproductive biotechnologies including intracytoplasmic sperm injection (ICSI). The aim of the present study was to examine the sperm collected from the cauda and caput of the cat epididymis using functional tests. Testicles and epididymides from 5 adult tomcats were collected by orchiectomy and maintained at 4°C for 4 h, until semen collection. Due to the importance of dogs to humans, there is increasing interest in breeders in the use of reproductive biotechnologies. However, most of the biotechnologies would require the removal or dilution of the seminal plasma, which is known to exert both beneficial and deleterious effects on sperm quality. One of the beneficial effects of seminal plasma would be the antioxidant protection because sperm are particularly susceptible to oxidative stress, mainly due to the reduced cytoplasm and the high content of polyunsaturated fatty acids in their membrane. An alternative to overcome the injuries caused by oxidative stress is the antioxidant treatment, which requires the identification of those reactive oxygen species (ROS) that are the most deleterious. Mangalarga, due to its marching abilities, is the mostly widespread and numerous equine breed in Brazil. Furthermore, previous studies indicate that the semen of these horses is particularly susceptible to cryo-injuries. Artificial insemination results can be improved using fixed-timed AI (FTAI). Currently, progesterone release devices, indispensable in FTAI procedures, such as DIB® (Schering-Plough, Kenilworth, NJ, USA), are made of silicone, a nonbiodegradable biocompatible polymer. Recently, biodegradable progesterone (P4)-releasing devices made of poly(3-hydroxy)butyrate-valerate copolymer (PHB-V) and poly-ϵ-caprolactona (PCL) have been developed. To investigate the efficiency on pregnancy rate of FTAI programs using a new intravaginal device made of PHB-V (Progestar®, Innovare, São Paulo, Brazil), 240 Nellore (Bos indicus) cows, between 3 and 8 years of age, were randomly assigned to 2 experiments. In experiment 1, new P4 devices DIB (control group, DIB1; n = 60) were compared with PHB-V (Progestar1; n = 76); in experiment 2, previously used DIB (DIB2; n = 52) and PHB-V (Progestar2; n = 52) devices were compared. All groups were treated with 2 mg of estradiol benzoate i.m. (Innovare) and received the intravaginal device according to group (Day 0). Together with P4 device withdrawal (Day 8) all animals received 300IU of eCG i.m. (Novormon®, Schering-Plough) and 150 μg i.m. of D-cloprostenol (Croniben®, Biogenesis-Bagó, Buenos Aires, Argentina). On Day 9, all cows were treated with 1 mg of estradiol benzoate and the FTAI was performed 30 h later. The data were submitted to ANOVA (PROC GLM of SAS, SAS Institute, Cary, NC, USA), separating as causes of variance the effect of treatment, postpartum period (PPP1-45 until 70 days and PPP2-71 until 90 days), body condition score (BCS4, BCS5, and BCS6 using a 1 to 9 scale), cyclicity (cycling and noncycling) and clean-up bulls (5 bulls nominated with numbers 1 to 5). Significant differences were considered to have P < 0.05. In experiment 1, interactions were not significant, there was no effect of treatment on FTAI (DIB1: 66.7% v. Progestar1: 65.8%) or breeding season pregnancy rates (BSPR; DIB1: 95.0% v. Progestar1: 94.7%) and no effect of other variables on FTAI pregnancy rates (FTAIPR) and BSPR. In experiment 2 there was no effect of treatment on FTAIPR (DIB2: 61.6% v. Progestar2: 55.8%) and BSPR (DIB2: 94.2% v. Progestar2: 88.5%). As ANOVA indicated a significant effect of clean-up bulls on BSPR, Tukey test was used to split it up. A bull of the Progestar2 group had lower performance (41.7%) causing the difference (albeit nonsignificant) on BSPR. This experiment showed that Progestar and DIB had the same efficiency on pregnancy rate and both treatments have the potential to produce FTAIPR >50% and BSPR >90%.
