Reading difficulties are prevalent worldwide, including in economically developed countries, and are associated with low academic achievement and unemployment. Longitudinal studies have identified several early childhood predictors of reading ability, but studies frequently lack genotype data that would enable testing of predictors with heritable influences. The National Child Development Study (NCDS) is a UK birth cohort study containing direct reading skill variables at every data collection wave from age 7 years through to adulthood with a subsample (final n = 6431) for whom modern genotype data are available. It is one of the longest running UK cohort studies for which genotyped data are currently available and is a rich dataset with excellent potential for future phenotypic and gene-by-environment interaction studies in reading. Here, we carry out imputation of the genotype data to the Haplotype Reference Panel, an updated reference panel that offers greater imputation quality. Guiding phenotype choice, we report a principal components analysis of nine reading variables, yielding a composite measure of reading ability in the genotyped sample. We include recommendations for use of composite scores and the most reliable variables for use during childhood when conducting longitudinal, genetically sensitive analyses of reading ability.

To report temporal trends in venovenous extracorporeal membrane oxygenation (ECMO) use for neonatal respiratory failure in U.S. centers before and after functional venovenous cannula shortage due to withdrawal of one dual lumen venovenous cannula from the market in 2018.Retrospective cohort study.ECMO registry of the Extracorporeal Life Support Organization.Infants who received neonatal (cannulated prior to 29 d of age) respiratory ECMO at a U.S. center and had a record available in the ECMO registry from January 1, 2010 to July 20, 2021.None.Primary outcome was receipt of venovenous ECMO (vs venoarterial or other), and secondary outcomes were survival to hospital discharge and adverse neurologic outcomes. Using an interrupted time series design, we fit multivariable mixed effects logistic regression models with receipt of venovenous ECMO as the dependent variable, treatment year modeled as a piecewise linear variable using three linear splines (pre shortage: 2010-2014, 2014-2018; shortage: 2018-2021), and adjusted for center clustering and multiple covariates. We evaluated trends in venovenous ECMO use by primary diagnosis including congenital diaphragmatic hernia, meconium aspiration, pulmonary hypertension, and other. Annual neonatal venovenous ECMO rates decreased after 2018: from 2010 to 2014, adjusted odds ratio (aOR) for yearly trend 0.98 (95% CI 0.92-1.04), from 2014 to 2018, aOR for yearly trend 0.90 (95% CI 0.80-1.01), and after 2018, aOR for yearly trend 0.46 (95% CI 0.37-0.57). We identified decreased venovenous ECMO use after 2018 in all diagnoses evaluated, and we failed to identify differences in temporal trends between diagnoses. Survival and adverse neurologic outcomes were unchanged across the study periods.Venovenous ECMO for neonatal respiratory failure in U.S. centers decreased after 2018 even after accounting for temporal trends, coincident with withdrawal of one of two venovenous cannulas from the market.

Prophylactic athletic tape is often used to provide additional ankle support and reduce potential injury in both wet and dry environments. The purpose of this study is to determine the effect of prophylactic athletic tape on ankle range of motion following exposure to moisture and exercise. Following exercise, range of motion increased slightly but remained below baseline and postexercise control values for all tape conditions. The novel synthetic tape maintained greater ankle support following the exercise period for wet condition in inversion and eversion. Cotton and novel synthetic adhesive tapes provided improved ankle stability following exercise, whether wet or dry, as compared with the use of no tape.

An understanding the organic matter (OM) in primitive interplanetary materials can provide us with important constraints on both the early solar system carbon cycle and incipient prebiotic synthesis before the origin of life. As a window to the past, primitive chondrites preserve the most pristine record of parent body, nebular and interstellar components and the occurrence of OM in them has been shown in both soluble (SOM) (1) and insoluble (IOM) (2) form. Total organic carbon (TOC) abundance reaches ~3-4 wt% in the most primitive carbonaceous chondrite (CCs) (3), such as Ivuna-type chondrites (CIs) – thus making them highly desirable for the OM studies, and relevant to the study of Asteroid 162173 Ryugu samples from the Hayabusa-2 mission.A combination of both SOM and IOM analysis of organic bulk meteorite separates together with in-situ analysis of OM have provided a comprehensive account of chondritic OM (4). In the case of in-situ analysis, the combination of both scanning (SEM) and transmission electron microscopy (TEM) together with soft X-Ray scanning transmission microscopy (STXM) have shown the presence of micron to submicron distinctive organic particles (OPs) (5). Carbon K-edge X-ray absorption near edge structure (XANES) has shown the aromatic-carbonyl-carboxyl chemical nature of these organic particles (5). In addition, aromatic-poorer and carboxylic-richer diffuse OM (6) within both amorphous and phyllosilicate occurs as well.As observation techniques are getting better, aberration corrected TEM coupled with electron energy loss spectroscopy (EELS) might provide the same results as carbon XANES, but with higher image magnification, rapid data acquisition and better accessibility. In this context, we present the results of a comparative carbon K-edge XANES and EELS study of CI meteorite Ivuna. An approximately 100 nm lamella of the Ivuna meteorite was prepared using focused ion beam (FIB)-SEM with the Helios 5 Hydra DualBeam (CEITEC, Masaryk University, Czechia) and analysed by TEM-EELS with the JEOL ARM200CF (ePSIC, Diamond Light Source, UK) and STXM-XANES at Beamline BL19A of the KEK Photon Factory, Japan. We observed that (I) XANES on samples that did not experience TEM-EELS are in agreement with the previous studies of aromatic-carbonyl-carboxylic macromolecular OPs and IOM, while (II) the TEM-EELS of OPs show aromatic-carbonyl functional chemistry but with amorphous carbon convoluting the carboxylic peak, and aromatic-poor spectra with a sharp carbonate peak in diffuse OM. The difference between XANES and EELS particularly in the diffuse OM can be interpreted by electron-beam damage. Thickness and e-beam damage leads to amorphous C formation in the OPs. Phyllosilicate minerals in the carbonaceous chondrites provide insights into processes in primitive parent bodies of the early Solar System. It is widely agreed that the CM- and CI-type carbonaceous chondrites underwent aqueous alteration on their parent bodies, resulting in phyllosilicate-rich matrices, where the dominant mineral phase is serpentine. There are many previous studies investigating phyllosilicate structure in carbonaceous chondrites, however, the presence of sulfur in these minerals and its effect on crystal lattice structure has not been studied in detail. We are investigating how the presence of sulfur (up to ≃9-10 wt% SO3) in serpentine phyllosilicate regions effects basal lattice spacing measurements of serpentine-like minerals in CM- and CI-type chondritic and related asteroidal material.Four specimens are being studied for this work: Winchcombe and Aguas Zarcas (CM-type), and Ryugu samples (A0058-C2001-08, A0104-00200502 and A0104-01700602) from Hayabusa2 and Ivuna (CI-type). All samples are TEM wafers. We have used a multi-technique approach to study the samples, with the E01 JEOL ARM200CF and E02 JEOL ARM300CF electron microscopes at the ePSIC facility at Diamond Light Source in Harwell, UK. EDS compositional data has been collected using the E01 microscope, whilst HRTEM and HAADF imaging data has been collected at E02. At E02 we are also applying a new 4D-STEM nano-diffraction technique in order to collect lattice spacing data to correlate with our other HRTEM results. Fe-K XANES analyses on Winchcombe and Ryugu have been carried out using the I18 microprobe and I14 hard x-ray nanoprobe respectively, also at Diamond Light Source, to constrain Fe3+/ΣFe. By combining these techniques we aim to better understand the physical and chemical structure of serpentine-like minerals in carbonaceous chondrites.Initial analyses have shown that sulfur presence in carbonaceous chondrite phyllosilicates reduces the basal lattice spacings of serpentine-like minerals. In these sulfur-bearing regions, we have been finding lattice spacings in the range ~0.60-0.74nm for the CM-type chondrites. For the CI-type, these range between ~0.65-0.76nm. Differences in the reduced lattice spacing ranges are likely related to the redox state of the sulfur. In Ryugu and other carbonaceous chondrites the sulfur appears reduced; its content in serpentine is low and we see FeS grains. Comparatively, in Winchcombe (and others) more of the sulfur seems to be in the serpentine structure.We can conclude that in serpentine-like minerals, the presence of sulfur appears to reduce basal lattice spacing values compared to the expected d-spacing value of 0.70nm for serpentine. Possible reasons for this include further investigations into the valency of the sulfur ions, the bonding environment within serpentine layers, and the location of sulfur in either the octa- or tetrahedral lattice sites.

Over the last decade, significant progress been made in elucidating the role of membrane transporters in altering drug disposition, with important toxicological consequences due to changes in localized concentrations of compounds. The topic of "Transporters and Toxicity" was recently highlighted as a scientific session at the International Transporter Consortium (ITC) Workshop 4 in 2021. The current white paper is not intended to be an extensive review on the topic of transporters and toxicity but an opportunity to highlight aspects of the role of transporters in various toxicities with clinically relevant implications as covered during the session. This includes a review of the role of solute carrier transporters in anticancer drug-induced organ injury, transporters as key players in organ barrier function, and the role of transporters in metal/metalloid toxicity.

Gastroesophageal reflux disease (GERD) is the retrograde flow of gastric contents into the distal esophagus and may be treated medically or surgically. Magnetic sphincter augmentation surgery using LINX has recently demonstrated comparable results to Nissen fundoplication. We aimed to evaluate preoperative patient risk factors that were associated with LINX removal rates or postoperative EGD with dilation rates (POEGDD). This is a single institution retrospective review of patients undergoing LINX between 2015 and 2021. One hundred and twelve patients were reviewed, 106 included within the study; those excluded had prior foregut surgery or device fracture. Variables including age, sex, BMI, size of device, DeMeester score, manometry, GERD Health-Related Quality of Life (GERD HRQL) questionnaires, POEGDD, and removal rates were recorded. Comparing removal and dilation status, the chi-square or Fisher’s exact test and the Mann-Whitney U test were used to analyze categorical and continuous variables, respectively. A P < .05 was considered to be statistically significant. Eleven LINX devices were removed (10%); of these, 9 (81%) underwent POEGDD ( P = .0023). There was no difference in DeMeester scores, size of device, or BMI in patients requiring LINX removal compared to those not removed or POEGDD rates. Patients who required LINX removal had higher GERD HRQL scores both preoperatively (median 34 vs 28) and postoperatively at all visits compared to those patients who did not undergo removal ( P = .032). Manometry and DeMeester scores were not associated with LINX removal suggesting a less invasive GERD HRQL questionnaire may be a better predictor of patients who will succeed with LINX surgery.

Pediatric patients with ARDS will on occasion need venovenous extracorporeal membrane oxygenation (VV-ECMO) for organ support. As these patients recover, they may benefit from lung recruitment maneuvers including flexible bronchoscopy (FB). The objective of this study was to assess the clinical course of patients who underwent FB while on VV-ECMO for ARDS.This was a secondary analysis of a retrospective multi-center cohort at 10 United States pediatric academic quaternary care centers. Data were collected on 204 subjects age 14 d-18 y on VV-ECMO.271 FBs were performed on 129 (63%) subjects. Pre-FB tidal volume was 1.8 mL/kg compared to 2.22 mL/kg following FB (FB can be performed on patients while anticoagulated on VV-ECMO with a low incidence of complications. FB may be beneficial especially when performed early in the course of VV-ECMO.

Eastern hemlock, Tsuga canadensis (L.) Carriere, is an important component of riparian zones in Appalachian forests. Tree mortality caused by hemlock woolly adelgid, Adelges tsugae Annand (Hemiptera: Adelgidae) could impact aquatic macroinvertebrate populations. Our study examined larval populations of Ephemeroptera, Plecoptera, and Trichoptera occurring in the headwaters of two creeks in the Sumter National Forest (Oconee Co., SC). Larval populations were initially sampled in King Creek (2006) and Crane Creek (2007) in separate studies. When those studies were conducted, there was no noticeable decline in hemlock health along either creek. However, by 2009 hemlock mortality along both creeks was obvious, and the Ephemeroptera, Plecoptera, and Trichoptera community was resurveyed on both creeks. This is the first study comparing aquatic macroinvertebrate populations before and after A. tsugae infestation. Ephemeroptera, Plecoptera, and Trichoptera larvae were identified to genus, and analyzed at the functional feeding group level. In both creeks, collector filterers and scrapers were significantly more abundant in the initial survey than in 2009. Generic diversity within each creek was examined using the total Ephemeroptera, Plecoptera and Trichoptera community, and was based on the indices of: richness (D0), Shannon exponential (D1), and inverse Simpson (D2). The only diversity measure that differed significantly between the original survey and 2009 was the Shannon exponential index for Crane Creek which was significantly higher in 2009 than 2006

International audience; We have investigated several particles collected during each of two touchdowns of the Hayabusa2 spacecraft at the surface of the C-type asteroid 162173 Ryugu using various electron microscope techniques. Our detailed transmission electron microscopy study shows the presence of magnetite with various morphologies coexisting in close proximity. This is characteristic of CI chondrite-like materials and consistent with the mineral assemblages and compositions in the Ryugu parent body. We describe the microstructural characteristics of magnetite with different morphologies, which could have resulted from the chemical conditions (growth vs. diffusion rate) during their formation. Furthermore, we describe the presence of magnetites with a spherulitic structure composed of individual radiating fibers that are characterized by pervasive, homogeneously distributed euhedral to subhedral pores that have not been described in previous chondrite studies. This particular spherulitic structure is consistent with crystallization under nonequilibrium conditions. Additionally, the presence of a high density of defects within the magnetite fibers, the high surface/volume ratio of this morphology, and the presence of amorphous materials in several pores and at the edges of the acicular fibers further support their formation under nonequilibrium conditions. We suggest that the growth processes that lead to this structure result from the solution reaching a supersaturated state, resulting in an adjustment to a lower free energy condition via nucleation and rapid growth.

The Winchcombe meteorite is a CM chondrite breccia composed of eight distinct lithological units plus a cataclastic matrix. The degree of aqueous alteration varies between intensely altered CM2.0 and moderately altered CM2.6. Although no lithology dominates, three heavily altered rock types (CM2.1–2.3) represent >70 area%. Tochilinite–cronstedtite intergrowths (TCIs) are common in several lithologies. Their compositions can vary significantly, even within a single lithology, which can prevent a clear assessment of alteration extent if only TCI composition is considered. We suggest that this is due to early alteration under localized geochemical microenvironments creating a diversity of compositions and because later reprocessing was incomplete, leaving a record of the parent body's fluid history. In Winchcombe, the fragments of primary accretionary rock are held within a cataclastic matrix (~15 area%). This material is impact-derived fallback debris. Its grain size and texture suggest that the disruption of the original parent asteroid responded by intergranular fracture at grain sizes

Without a protective atmosphere, space-exposed surfaces of airless Solar System bodies gradually experience an alteration in composition, structure and optical properties through a collective process called space weathering. The return of samples from near-Earth asteroid (162173) Ryugu by Hayabusa2 provides the first opportunity for laboratory study of space-weathering signatures on the most abundant type of inner solar system body: a C-type asteroid, composed of materials largely unchanged since the formation of the Solar System. Weathered Ryugu grains show areas of surface amorphization and partial melting of phyllosilicates, in which reduction from Fe3+ to Fe2+ and dehydration developed. Space weathering probably contributed to dehydration by dehydroxylation of Ryugu surface phyllosilicates that had already lost interlayer water molecules and to weakening of the 2.7 µm hydroxyl (–OH) band in reflectance spectra. For C-type asteroids in general, this indicates that a weak 2.7 µm band can signify space-weathering-induced surface dehydration, rather than bulk volatile loss.

Mercury is a heavy metal toxicant that is prevalent throughout the environment. Organic forms of mercury, such as methylmercury (MeHg), can cross the placenta and can lead to lasting detrimental effects in the fetus. The toxicological effects of MeHg on the placenta itself have not been clearly defined. Therefore, the purpose of the current study was to assess the transport of MeHg into placental syncytiotrophoblasts and to characterize the mechanisms by which MeHg exerts its toxic effects. Cultured placental syncytiotrophoblasts (BeWo) were used for these studies. The transport of radioactive MeHg was measured to identify potential mechanisms involved in the uptake of this compound. The toxicological effects of MeHg on BeWo cells were determined by assessing visible pathological change, autophagy, mitochondrial viability, and oxidative stress. The findings of this study suggest that MeHg compounds are transported into BeWo cells primarily by sodium-independent amino acid carriers and organic anion transporters. The MeHg altered mitochondrial function and viability, decreased mitophagy and autophagy, and increased oxidative stress. Exposure to higher concentrations of MeHg inhibited the ability of cells to protect against MeHg-induced injury. The findings show that MeHg is directly toxic to syncytiotrophoblasts and may lead to disruptions in the fetal/maternal transfer of nutrients and wastes.

The effects of drill and broadcast planting methods on cover crop biomass production depend on various environmental and operational factors. We investigated whether drilling and broadcasting result in different amounts of biomass production by crimson clover (Trifolium incarnatum L.) and hairy vetch (Vicia villosa Roth) in the upstate of South Carolina, and results vary when seeding rates are increased by 50% from the standard value (22.4 kg ha−1). Field trials were conducted during the fall–winter of 2019–2020 (season one) and 2020–2021 (season two) at the Piedmont Research and Education Center in Pendleton, SC, USA. Cover crop (hairy vetch, crimson clover), planting method (broadcast, drill), and seeding rate (standard, high) treatments were arranged as a 2 × 2 × 2 factorial in both years. Aboveground biomass was measured after 22.5 weeks from planting. At standard seeding rates, crimson clover produced a higher biomass when drilled, rather than broadcasted, whereas biomass production did not vary for hairy vetch. Even with 50% higher seeding rates, broadcasting did not always produce the same biomass as that of drilling for crimson clover. Our results suggest that the advantage of drilling over broadcasting depends upon the cover crop species, as crimson clover responds well to drilling, whereas hairy vetch does not.

International audience; Aeolian processes have shaped and contributed to the geological record in Gale crater, Mars, long after the fluviolacustrine system existed ∼3 Ga ago. Understanding these aeolian deposits, particularly those which have been lithified and show evidence for aqueous alteration, can help to constrain the environment at their time of deposition and the role of liquid water later in Mars' history. The NASA Curiosity rover investigated a prominent outcrop of aeolian sandstone within the Stimson formation at the Greenheugh pediment as part of its investigation of the Glen Torridon area. In this study, we use geochemical data from ChemCam to constrain the effects of aeolian sedimentary processes, sediment provenance, and diagenesis of the sandstone at the Greenheugh pediment, comparing the Greenheugh data to the results from previous Stimson localities situated 2.5 km north and >200 m lower in elevation. Our results, supported by mineralogical data from CheMin, show that the Stimson formation at the Greenheugh pediment was predominately sourced from an olivine-rich unit that may be present farther up the slopes of Gale crater's central mound. Our results also suggest that the Greenheugh pediment Stimson formation was cemented by surface water runoff such as that which may have formed Gediz Vallis. The lack of alteration features in the Stimson formation at the Greenheugh pediment relative to those of the Emerson and Naukluft plateaus suggests that groundwater was not as available at this locality compared to the others. However, all sites share diagenesis at the unconformity

Postnatal muscle growth is achieved through hypertrophy of the muscle fibers and is impacted by the activity of satellite cells, the quiescent muscle stem cell. Several miRNAs are preferentially expressed in skeletal muscle and could provide a mechanism for increasing muscle hypertrophy through satellite cell proliferation and/or differentiation. The objectives of this study were to: 1) Characterize the miRNA transcriptome of the longissimus thoracis et lumborum muscle at several developmental timepoints [gestational d 85 (PN1), 110 (PN2), 133 (PN3), postnatal d 42 (PW1), 65 (PW2), 243 (MAT)] during muscle hypertrophy in lambs, and 2) examine miR-29a, identified in sequencing to be differentially regulated across development, loss of function on satellite cell proliferation and differentiation. Muscle fiber characteristics showed drastic increases (p < 0.0001) in fiber size and alterations in muscle fiber type occur during pre and postnatal development. miRNA sequencing comparisons were performed in developmental order (PN1 vs. PN2, PN2 vs. PN3, PN3 vs. PW1, PW1 vs. PW2, PW2 vs. MAT). There were 184 differentially expressed (Padj < 0.05) miRNA, 142 unique miRNA, from all 5 comparisons made. The transitional stage (PN3 vs. PW1) had the largest number (115) of differentially expressed miRNA. Inhibition of miR-29a in satellite cell culture increased (p < 0.05) cell proliferation and differentiation capacity. Characterization of the miRNA transcriptome provides valuable insights into the miRNA involved in muscle fiber hypertrophy and the potential importance of the transitional period.

OBJECTIVE Our objective is to characterize the incidence of tracheostomy placement and of new requirement for long-term mechanical ventilation after extracorporeal membrane oxygenation (ECMO) among children with acute respiratory failure. We examine whether an association exists between demographics, pre-ECMO and ECMO clinical factors, and the placement of a tracheostomy or need for long-term mechanical ventilation. METHODS A retrospective multicenter cohort study was conducted at 10 quaternary care pediatric academic centers, including children supported with veno-venous (V-V) ECMO from 2011 to 2016. RESULTS Among 202 patients, 136 (67%) survived to ICU discharge. All tracheostomies were placed after ECMO decannulation, in 22 patients, with 19 of those surviving to ICU discharge (14% of survivors). Twelve patients (9% of survivors) were discharged on long-term mechanical ventilation. Tracheostomy placement and discharge on home ventilation were not associated with pre-ECMO severity of illness or pre-existing chronic illness. Patients who received a tracheostomy were older and weighed more than patients who did not receive a tracheostomy, although this association did not exist among patients discharged on home ventilation. ECMO duration was longer in those who received a tracheostomy compared with those who did not, as well as for those discharged on home ventilation, compared to those who were not. CONCLUSION The 14% rate for tracheostomy placement and 9% rate for discharge on long-term mechanical ventilation after V-V ECMO are important patient-centered findings. This study informs anticipatory guidance provided to families of patients requiring prolonged respiratory ECMO support, and lays the foundation for future research.

Lentil, a cool-season food legume, is rich in protein and micronutrients with a range of prebiotic carbohydrates, such as raffinose-family oligosaccharides (RFOs), fructooligosaccharides (FOSs), sugar alcohols (SAs), and resistant starch (RS), which contribute to lentil's health benefits. Beneficial microorganisms ferment prebiotic carbohydrates in the colon, which impart health benefits to the consumer. In addition, these carbohydrates are vital to lentil plant health associated with carbon transport, storage, and abiotic stress tolerance. Thus, lentil prebiotic carbohydrates are a potential nutritional breeding target for increasing crop resilience to climate change with increased global nutritional security. This study phenotyped a total of 143 accessions for prebiotic carbohydrates. A genome-wide association study (GWAS) was then performed to identify associated variants and neighboring candidate genes. All carbohydrates analyzed had broad-sense heritability estimates (H2) ranging from 0.22 to 0.44, comparable to those reported in the literature. Concentration ranges corresponded to percent recommended daily allowances of 2–9% SAs, 7–31% RFOs, 51–111% RS, and 57–116% total prebiotic carbohydrates. Significant SNPs and associated genes were identified for numerous traits, including a galactosyltransferase (Lcu.2RBY.1g019390) known to aid in RFO synthesis. Further studies in multiple field locations are necessary. Yet, these findings suggest the potential for molecular-assisted breeding for prebiotic carbohydrates in lentil to support human health and crop resilience to increase global food security.
