A comprehensive investigation was carried out todetermine the phytochemical properties of Fraxinus americana and Fraxinus excelsior leavesextracts. In addition, infusion extraction method using only water as theextraction solvent and ultrasound-assisted extraction technique were followedcomparatively to extract the phenolic compounds from the leaves to estimate theefficacy of the traditional hot water infusion method. Initially, total antioxidant capacities, total phenolic contents, and radicalscavenging activities of the extracts were determined. Afterwards, 34 potentialphenolic compounds were analyzed by LC-ESI-MS/MS technique. The paper gives acomprehensive insight to the literature about the phytochemical properties of Fraxinus americana and Fraxinus excelsior leaves extracts.

Bu çalışmada, zeytin, zeytinyağı ve zeytin yaprağında bulunan bazı fenolik bileşiklerin tayinleri için elektrokimyasal ve kromatografik yöntemlerin geliştirilmesi amaçlanmaktadır.Dünya genelinde doğal antioksidanlara (gıdaların biyoaktif bileşenleri gibi) karşı giderek artan bir ilgi oluşmaktadır. Zeytin (Olea europaea L.) meyvesi, yağı ve yaprağı ile birlikte antioksidan kaynağı olan fenolik bileşikler bakımından oldukça zengindir. Diğer taraftan, zeytin ağacı ülkemizin bulunduğu coğrafi konum ve sahip olduğu iklim özellikleri sayesinde ülkemizde bolca yetişmektedir. Bu nedenle mükemmel derecede antioksidan kaynakları olan ve ülkemiz açısından önemli ticari değere sahip sofralık zeytin, zeytinyağı ve zeytin yapraklarında söz konusu fenolik bileşiklerin nicel tayinleri için analitik yöntemlerin geliştirilmesi önem arz etmektedir.Güçlü bir antioksidan olan oleuropein yalnızca oleaceae familyasına ait bitkilerde bulunur. Çalışmada öncelikle, oleuropein tayini için çok duvarlı karbon nanotüp ile modifiye edilmiş camımsı karbon elektrot kullanılarak gerçekleştirilen bir elektrokimyasal yöntem önerilmiştir. Yalın camımsı karbon elektrot ile karşılaştırıldığında, oleuropein yükseltgenme pik akımı, modifiye elektrot üzerinde yaklaşık 340 kez artmıştır. Önerilen teknik valide edilmiş ve valide yöntem bir zeytin yaprağı ekstraktında oleuropeinin tayini için başarıyla uygulanmıştır. Elde edilen sonuçlar, LC-ESI-MS/MS yöntemi ile elde edilen veriler ile uyum içerisindedir. Çalışmada daha sonra, 31 fenolik bileşiğin analizleri için LC-ESI-MS/MS kullanılarak gerçekleştirilen basit bir yöntem valide edilmiştir. Valide yöntemin analitik özellikleri, söz konusu yöntemin zeytin yaprağı örneklerinde bulunan fenolik bileşiklerin rutin analizlerinde oldukça kullanışlı olduğunu açıkça göstermektedir. In this study, it is aimed to develop electrochemical and chromatographic methods for the determination of phenolic compounds in table olive, olive oil and olive leaves.There is a rising interest in natural antioxidants as bioactive components of foods. Olive (Olea europaea L.) is quite abundant in terms of phenolic compounds which are antioxidant sources together with fruit, oil and leaf. On the other hand, the olive tree grows abundantly in our country thanks to the geographical location of our country and its climate characteristics. Therefore, it is important to develop analytical methods for the quantitative determination of phenolic compounds in table olive, olive oil and olive leaves, which are excellent antioxidant resources and have significant commercial value for our country.Oleuropein, a powerful antioxidant, is found only in plants belonging to the oleaceae family. In present study, a multi-walled carbon nanotube modified glassy carbon electrode was proposed to prepare an electrochemical sensing platform for the determination of oleuropein. Compared to the bare glassy carbon electrode, the oxidation peak current of oleuropein increased about 340 times at the modified electrode. The proposed technique was validated and the validated method was successfully applied to the determination of oleuropein in an olive leaf extract. The results obtained were in agreement with those obtained by LC-ESI-MS/MS. Moreover a simple method was validated for the analysis of 31 phenolic compounds using LC-ESI-MS/MS. Analytical properties of the validated method was satisfied the routine analysis of phenolic compounds in olive leaves samples. 94

Bu çalışmanın ilk bölümünde, Manisa Bağcılık Araştırma İstasyonundan temin edilen yaş ve kuru üzüm örneklerinde pestisit kalıntı düzeyleri araştırılmıştır. Bu örneklerde kalıntısı araştırılan 360 adet pestisit, QuEChERS ekstraksiyonunu takiben GC-MS ve LC-MS/MS cihazları ile nitel olarak izlenmiştir. Söz konusu üzümlerde kullanıldığı tespit edilen, GC grubu pestisitlerinden boscalidin, üzümlerde kurutma öncesi uygulanan işleme [kurutma öncesi K2CO3 (%5, a/a) çözeltisine daldırma yapıldıktan sonra güneş altında kurutma ve daldırma yapılmadan kurutma] ve bekleme süresine bağlı olarak nicel tayini gerçekleştirilmiştir. Elde edilen sonuçlar, söz konusu pestisitin daldırılmış örneklerdeki bozunma hızının, daldırılmamış örneklerdekine göre daha yüksek olduğunu göstermiştir.Çalışmanın ikinci bölümü ise, elde edilen daldırılmış ortamdaki yüksek bozunma hızının, ortamdaki K2CO3?ın sağladığı bazlık etkisiyle ilişkisinin araştırılması için, 3 farklı derişimde (2, 4 ve 6 mg/L), su ve 5 farklı pH?de (pH=7, 8, 9, 10, 11) tamponlanmış K2CO3 çözeltileri ile boscalid standardı kullanılarak hazırlanan pestisitlerin, bozunma hızlarının 30 gün boyunca incelenmesini içermektedir. Bu amaçla, boscalid standart çözeltileri (1, 2, 4, 6 ve 7 mg/L) ile söz konusu ortamlar için yapılan doğrusallık çalışmalarında, korelasyon katsayısı 0.9947 ile 0.9999 arasında değişen kalibrasyon eğrileri elde edilmiştir. Boscalid miktarındaki değişimler, su ve söz konusu tampon ortamlarında farklanmak üzere 234.66 ile 244.99 nm arasında değişen dalga boylarında, hazırlanan çözeltilerin, hazırlandığı gün (ilk gün), 3. gün, 7. gün, 15. gün ve 30. günlerdeki absorpsiyonlarının UV-Vis spektrofotometresi kullanılarak ölçülmesi ile hesaplanmıştır. Sonuçlar, boscalid?in bozunma hızının K2CO3 ile hazırlanan tampon ortamlarında daha yüksek olduğunu göstermektedir. Özellikle pH=8, 9 ve 10 tamponlarında pestisit miktarındaki azalma, pH=7 ve 11 tamponlarındakine göre daha yüksek oranda gerçekleşmiştir. In the first part of this study, pesticide residue levels in grape and raisin samples obtained from Manisa Viticulture Research Station were investigated. 360 pesticides monitored qualitatively with GC-MS and LC-MS/MS following the QuEChERS extraction to determine the types of pesticide residues in these samples. Quantitative determination of boscalid that determined as used on grapes was carried out depending on the process before drying [drying in the sun after immersion in K2CO3 (%5 w/w) solution before drying and without immersion] and for the future time. The results obtained showed that degradation rate of the pesticide in immersed samples is higher than those of not immersed.However, the second part of the study includes the analysis of pesticide degradation rates that prepared at 3 different concentrations (2, 4 ve 6 mg/L) using water and K2CO3 solutions buffered at 5 different pH (pH=7, 8, 9, 10, 11) with boscalid standart, during 30 days to investigate the relationship between the high degradation rate of the pesticide in immersed media and the effect of alkalinity provided by K2CO3. Correlation coefficients of the calibration curves obtained ranging from 0.9947 to 0.9999 in linearity studies with prepared boscalid standart solutions (1, 2, 4, 6 ve 7 mg/L) for this purpose. Changes in the amount of pesticide was calculated with measuring the absorptions of the prepared solutions ranging from at 234.66 nm to 244.99 nm wavelenghts for the different media using UV-Vis spectrophotometer in preparation day (first day), 3nd day, 7th day, 15th day and 30th day. The results showed that boscalid degradation rate is higher in buffer media. Decreases in the amount of pesticide occured higher especially at pH=8, 9 and 10 buffers than pH=7 and 11. 147
