1. Validation of a 60K SNP chip for caribou (Rangifer tarandus) for use in wildlife forensics, conservation, and management

Abstract

Large-scale genotyping platforms are currently being developed for several wild species. By querying thousands of polymorphic loci, genomics can be a useful ecological tool for describing and monitoring populations. Genomics is becoming increasingly useful as a forensic tool because of its ability to identify population of origin for purposes of enforcing anti-poaching laws. Our aim was to test the new SNP chip for caribou/reindeer (Rangifer tarandus) (Illumina iSelect caribou 60 K) under recommended and non-optimal sample conditions. Impact on signal detection (call rate) and error rate were assessed using reference samples. The SNP chip was shown to be robust, highly sensitive, reliable, and accurate at more than 10-fold below the recommended DNA input. Biological source of DNA had minor impact, even with fecal pellets given sufficient amount of host DNA. Hybridization of non-Rangifer samples as well as samples bearing DNA from two Rangifer samples both showed a drop in call rate and shifted levels of heterozygosity. Based on a population-targeted subset of SNPs included in the chip design, reassignment of 981 samples to a functional group (here to a caribou ecotype) was highly accurate (99.59 %) and the relative probability of reassignment error was estimated using the logarithm of odds score. Overall, the SNP chip is suitable for analysis of caribou/reindeer genomes even with suboptimal sampling and hence useful for population management and forensics.

Abstract

Quantifying the consequences of winter ticks (Dermacentor albipictus) on the body condition and life‐history traits of moose (Alces alces) is a challenge due to several confounding factors. We experimentally reduced tick load on moose calves by testing the effectiveness of 2 acaricide treatments: one using topical permethrin (5%) alone and the other a combination of a more concentrated topical permethrin (44%) and orally administered fluralaner (25 mg/kg). We evaluated changes in tick load, body mass, hematocrit, and hair loss severity and occurrence, from recaptured or resighted moose calves over winter in Québec and New Brunswick, Canada. Nearly all untreated moose (94%, n = 41) experienced hair loss compared to calves that received the combination of permethrin (44%) and fluralaner (41%, n = 37). Of treated moose that exhibited hair loss, only 22% had more than 5% damage and some already had hair loss at capture. Capturing moose later likely increased the probability of observing hair loss when resighting treated moose, although hair loss essentially remained lower for treated calves than for untreated calves. In untreated moose, tick load at capture tended to drive hair loss, but calendar date mostly drove hair loss severity, especially during April. There was no clear effect of topical permethrin (5%) on tick load, body mass, and hematocrit. Body condition simply decreased from January captures to spring recaptures, regardless of treatment. Our results suggested that combining permethrin (44%) and fluralaner effectively reduced tick load based on hair loss severity and occurrence. We cannot, however, disentangle the individual effects of permethrin (44%) and fluralaner. We discuss research implications and considerations of using such a treatment for reducing winter tick load. Abstract

Density dependence is often assumed in population dynamics, but its importance in small, isolated populations has been questioned. We evaluated the relative influence of density dependence, environmental conditions, and sporadic events (disease outbreaks and specialist predators) on annual population growth rate, annual female reproduction, and annual survival of juveniles and adult females in three populations of mountain ungulates. We analyzed long-term (30–47 years) individual-based data on two bighorn sheep populations and one mountain goat population in Alberta, Canada. The effect of cougar predation episodes and pneumonia epizootics on annual population growth rate was twice as strong as that of population density. While pneumonia reduced adult female and juvenile survival and predation episodes decreased all demographic rates, high density lowered only juvenile survival. Long-term studies are pivotal for understanding the dynamics of large herbivore populations, but they are rarely duplicated. Our analysis of three mountain ungulate populations with similar life history and ecological characteristics provides evidence that infrequent sporadic events can have a greater relative influence on annual population growth than density-dependent factors in isolated populations. This result contrasts with studies of larger, well-connected populations, highlighting the importance of considering sporadic events in the management and conservation of isolated populations. Abstract

To develop effective management to maintain or restore populations of large herbivores, wildlife managers require sound empirical data on their variations in size and associated parameters. Many studies have highlighted links between morphological traits of individuals and population density; however, less attention has been devoted to whether or not morphological traits can reliably inform on population size in years when no population estimates are available. We evaluated the relationships between three morphological traits (hind foot length, body mass, and body fat) and population size interpolated over three decades, for four migratory caribou (Rangifer tarandus) herds in northern Canada and Alaska. Our sample included 8865 measurements of 4473 individuals. We used a Bayesian modeling approach to evaluate the relationships between morphology and population size across different sex and age classes, considering different temporal scales and, when possible, phases of population growth or decline. We found that morphological traits were not consistently linked to population size. Statistically significant relationships existed for some combinations of herd and age classes, but weak to absent relationships were more common. Our study suggests that morphological traits alone cannot replace data obtained from aerial surveys to approximate population size when population trends are unknown. We discuss the usefulness of morphological traits to explain population size, and recognize their role as complementary metrics to inform the management and conservation of large herbivores, but conclude that morphological data should not be used to predict population size without information on population trends. Abstract

Climate change may modify species distribution to higher latitudes, resulting in potential changes of parasite diversity and transmission dynamics in areas where animals might not be locally adapted to these new parasite species. In addition, climate change may increase the frequency and severity of infestations of parasites that are already present in a region, by promoting the development and survival of infectious stages. Over the last decades, the number of moose ( Alces americanus ) infested by winter ticks ( Dermacentor albipictus ) has increased in eastern Canada, possibly because milder climatic conditions are increasing winter tick survival. Our main objective was to determine which meteorological variables are more likely to influence winter tick load on moose. We compiled several weather variables that may limit winter tick survival and explored which weather variables, or their interactions, influenced the winter tick load of 4,100 hunted moose from 2013 to 2019 in Québec, Canada along a latitudinal gradient. Winter tick load in fall decreased with the maximum number of consecutive days in spring with average daily temperatures below −15°C and with the number of consecutive days in summer with a relative humidity <80% when snowmelt in spring was earlier. These results suggest that cold temperatures and prolonged periods of low humidity, amplified by early snowmelt, limit the survival of adult female ticks and eggs, thus limiting their subsequent load on moose during the following fall. With climate change, precipitation increases and warm temperatures occur earlier in spring and are more frequent in summer. Our results suggest that climate change may have a positive long‐term influence on winter tick abundance in the environment and thereby increase winter tick load on moose, which could lead to a significant decrease in moose body condition and survival.

Abstract

Wild animals are infected with a large diversity and abundance of parasites that can affect their behavior, growth, body condition, and ultimately their survival. Although the adverse effects of parasites and the mechanisms involved in the interactions between a host and its parasites are generally well studied, much less is known about the additive or synergistic effects of multiple parasite species on a host. Moose populations in eastern Canada are infected by several species of endoparasites. In the last decades, the intensity of infestations by winter ticks, an ectoparasite, on moose have increased as a result of increased moose densities and favorable weather conditions that benefit winter tick survival. We aimed to document the diversity, intensity, prevalence, and distribution of different parasite species of moose in southern Quebec, Canada. We then evaluated the potential interaction between winter tick and endoparasites of moose, and we evaluated the effect of the simultaneous presence of ticks and endoparasites on moose body condition. To do so, we collected organs to identify and count endoparasite species, estimate winter tick abundance, and measure subcutaneous fat thickness from 174 hunted moose in fall 2019 in 8 regions of Quebec. Our results showed that the prevalence and intensity of winter tick and gastrointestinal parasites differed among regions, as well as the prevalence of the heart parasite Taenia krabbei and the intensity of lung parasite Echinoccocus granulosus . Moose body condition, however, was not influenced by the simultaneous presence of winter tick and endoparasites. The documentation of the interactive effects of multiple parasite species on a host is fundamental given that future environmental conditions in temperate climate will favor the reproduction, development, and survival of several parasite species, which could affect parasite diversity and abundance in the environment and modify host–parasite dynamics.

Abstract

Growing white‐tailed deer populations in Canada's boreal forest are linked to declines of threatened boreal caribou populations. Recent work by Dickie et al. (2024) shows that climate plays a stronger role than anthropogenic habitat alteration in impacting deer white‐tailed deer density, however we contend that these findings are not robust due to insufficient data and the analysis used is sensitive to data scaling approach. We demonstrate approximately equal but opposite effects of climate and alteration on white‐tailed deer population density, and emphasize the importance of robust analyses involving contentious conservation issues.
