Threatened shark species are caught in large numbers by artisanal and commercial fisheries and traded globally. Monitoring both which shark species are caught and sold in fisheries, and the export of CITES-restricted products, are essential in reducing illegal fishing. Current methods for species identification rely on visual examination by experts or DNA barcoding techniques requiring specialist laboratory facilities and trained personnel. The need for specialist equipment and/or input from experts means many markets are currently not monitored. We have developed a paper-based Lab-on-a-Chip (LOC) to facilitate identification of three threatened and CITES-listed sharks, bigeye thresher (Alopias superciliosus), pelagic thresher (A. pelagicus) and shortfin mako shark (Isurus oxyrinchus) at market source. DNA was successfully extracted from shark meat and fin samples and combined with DNA amplification and visualisation using Loop Mediated Isothermal Amplification (LAMP) on the LOC. This resulted in the successful identification of the target species of sharks in under an hour, with a working positive and negative control. The LOC provided a simple "yes" or "no" result via a colour change from pink to yellow when one of the target species was present. The LOC serves as proof-of-concept (PoC) for field-based species identification as it does not require specialist facilities. It can be used by non-scientifically trained personnel, especially in areas where there are suspected high frequencies of mislabelling or for the identification of dried shark fins in seizures.

Understanding the link between seamounts and large pelagic species (LPS) may provide important insights for the conservation of these species in open water ecosystems. The seamounts along the Cocos Ridge in the Eastern Tropical Pacific (ETP) ocean are thought to be ecologically important aggregation sites for LPS when moving between Cocos Island (Costa Rica) and Galapagos Islands (Ecuador). However, to date, research efforts to quantify the abundance and distribution patterns of LPS beyond the borders of these two oceanic Marine Protected Areas (MPAs) have been limited. This study used drifting-pelagic baited remote underwater video stations (BRUVS) to investigate the distribution and relative abundance of LPS at Cocos Ridge seamounts. Our drifting-pelagic BRUVS recorded a total of 21 species including elasmobranchs, small and large teleosts, dolphins and one sea turtle; of which four species are currently threatened. Depth of seamount summit was the most significant driver for LPS richness and abundance which were significantly higher at shallow seamounts (< 400 m) compared to deeper ones (> 400m). Distance to nearest MPA was also a significant predictor for LPS abundance, which increased at increasing distances from the nearest MPA. Our results suggest that the Cocos Ridge seamounts, specifically Paramount and West Cocos which had the highest LPS richness and abundance, are important aggregation sites for LPS in the ETP. However, further research is still needed to demonstrate a positive association between LPS and Cocos Ridge seamounts. Our findings showed that drifting pelagic BRUVS are an effective tool to survey LPS in fully pelagic ecosystems of the ETP. This study represents the first step towards the standardization of this technique throughout the region.

Ocean sunfishes, with their peculiar morphology, large size, and surface habits, are valuable assets in ecotourism destinations worldwide. This study investigates site fidelity and long-range movements of short ocean sunfish, Mola ramsayi (Giglioli 1883), at Punta Vicente Roca (PVR) off Isabela Island in the Galapagos Islands. Five individuals were tracked between 32 and 733 days using ultrasonic receivers and transmitters. Two of the 5 were also tracked with towed pop-off satellite tags. One travelled to the equatorial front covering 2700 km in 53 days, with dive depths in the upper 360 m at temperatures between 9.2°C and 22°C. During its westward travel, dives extended to 1112 m (the deepest depth yet recorded for Molidae) into temperatures ranging between 4.5°C and 23.2°C. The remaining four individuals demonstrated site fidelity to PVR and were detected at the site between 128–1361 times for a total of 3557 reports. Forty-eight percent of the reports occurred during daytime hours and 52% after dark. Presumed cleaning session durations had a median of 15 minutes and a maximum of nearly 100 minutes. No other ultrasonic arrays around Galapagos or in the Eastern Pacific regional network recorded the presence of tagged individuals. These data are combined with tourist vessel sightings and submersible observations to confirm Punta Vicente Roca as an important sunfish hotspot.

Abstract Habitat loss is a major cause of species loss and is expected to increase. Loss of habitat is often associated with fragmentation of remaining habitat. Whether species can persist in fragmented landscapes may depend on their movement behavior, which determines their capability to respond flexibility to changes in habitat structure and spatial distribution of patches. Movement is frequently generalized to describe a total area used, or segmented to highlight resource use, often overlooking finer‐scale individual behaviors. We applied hidden Markov models (HMM) to movement data from 26 eastern bettongs (Bettongia gaimardi) in fragmented landscapes. HMMs are able to identify distinct behavior states associated with different movement patterns and discover how these behaviors are associated with habitat features. Three distinct behavior states were identified and interpreted as denning, foraging, and fast‐traveling. The probability of occurrence of each state, and of transitions between them, was predicted by variation in tree‐canopy cover and understorey vegetation density. Denning was associated with woodland with low canopy cover but high vegetation density, foraging with high canopy cover but low vegetation density, and fast‐traveling with low canopy cover and low vegetation density. Bettongs did move outside woodland patches, often fast‐traveling through pasture and using smaller stands of trees as stepping stones between neighboring patches. Males were more likely to fast‐travel and venture outside woodlands patches, while females concentrated their movement within woodland patches. Synthesis and applications: Our work demonstrates the value of using animal movement to understand how animals respond to variation in habitat structure, including fragmentation. Analysis using HMMs was able to characterize distinct habitat types needed for foraging and denning, and identify landscape features that facilitate movement between patches. Future work should extend the use of individual movement analyses to guide management of fragmented habitat in ways that support persistence of species potentially threatened by habitat loss.
