Tese de doutoramento em Engenharia de Tecidos, Medicina Regenerativa e Células Estaminais, In cutaneous wounds, neovascularization is critical for tissue repair, influencing the healing cascade from the early onset to the remodeling phase. When vascularization is affected by alterationsin angiogenic signaling, the healing process is impaired and the quality of the neotissue formed is affected. Several strategies have been explored to promote neovascularization in cutaneous wounds; however, their clinical translation is still to be achieved. Therefore, this thesis aimed to explore innovative strategies to promote in vitro vascularization of tissue engineered (TE) constructs for wound healing. To this end, we took advantage of gellan gum-based spongy-like hydrogels to explore i) the modulation of a fibroblast-driven angiogenic microenvironment fostering endothelial cell (ECs) potential, and ii) the ability of the stromal vascular fraction (SVF) from adipose tissue to generate a self-regulatory angiogenic microenvironment in the absence of extrinsic growth factors. To minimize batch-to-batch variability associated to the processing of dried polymeric networks/spongy-like hydrogels, we designed two freezing devices and mapped the effect of the thermal parameters on the properties of the obtained structures. We were then able to fabricate highly reproducible materials with fine-tuned microarchitecture according to our specific needs. A bilayered structure was used as a template to explore the distinct functions of the dermal fibroblast’s subpopulations, providing evidence that pre-selection of papillary and reticular fibroblasts is relevant for promoting the in vitro prevascularization of skin TE constructs. We further explored the delivery of genes encoding angiogenic factors to fibroblasts, generating a microenvironment prone to vascularization while the spongy-like hydrogel offered structural support for new matrix deposition. Finally, we exploited specific integrins found in ECs present in SVF by designing integrin-specific spongy-like hydrogels to retain those cells, to ultimately triggering vasculogenesis. This in vitro vasculogenesis platform benefited neovascularization upon implantation by fostering inosculation with the host vasculature, but negatively impacted the healing of cutaneous wounds. Overall, the works herein reported provide valuable contributions in the field of vascularization of TE constructs, proposing complementary and alternative approaches to the use of ECs, taking a step towards the development of better therapies for cutaneous wounds., Em feridas cutâneas, a neovascularização é crítica para a reparação do tecido, influenciando a cicatrização do início à fase de remodelação. Quando a vascularização é afetada por alterações na sinalização angiogénica, o processo de cicatrização é prejudicado, afetando a qualidade do neotecido formado. Várias estratégias têm sido exploradas para promover a neovascularização em feridas cutâneas; no entanto, vários fatores dificultam a sua translação para a clínica. Esta tese, teve como objetivo explorar estratégias inovadoras de vascularização in vitro de estruturas de engenharia de tecidos (ET) para cicatrização de feridas. Para tal, tirámos partido dos hidrogéis esponjosos à base de goma gelana para explorar i) a modulação do microambiente angiogénico mediado por fibroblastos para promover o potencial das células endoteliais (CEs), e ii) a capacidade da fração vascular estromal (FVE) do tecido adiposo em gerar um microambiente autorregulador angiogénico sem fatores de crescimento extrínsecos. Para minimizar a variabilidade associada ao processo de preparação de redes poliméricas secas/hidrogéis esponjosos, desenvolvemos dois dispositivos de congelamento e mapeamos o efeito dos parâmetros térmicos nas propriedades das estruturas obtidas. Isto permitiu fabricar materiais com características altamente reprodutíveis com microarquitetura ajustada de acordo com nossas necessidades específicas. Uma estrutura baseada em bicamada foi utilizada como modelo para explorar as funções típicas de cada subpopulação de fibroblastos da derme, demonstrando que a pré-seleção dos fibroblastos papilares e reticules promove a pré-vascularização in vitro de estruturas de ET de pele. Explorámos ainda a entrega de genes codificadores de fatores de crescimento angiogénicos aos fibroblastos para gerar um microambiente propenso à vascularização, enquanto o hidrogel esponjoso ofereceu suporte estrutural para a deposição de nova matriz. Finalmente, explorámos integrinas específicas encontradas nas CEs presentes na FVE e desenhámos hidrogéis esponjosos integrina específicos capazes de reter essas células e desencadear a vasculogénese in vitro. Esta plataforma de vasculogénese in vitro beneficiou a neovascularização in vivo após a implantação, promovendo a inosculação com a vasculatura hospedeira, mas teve um impacto negativo na cicatrização de feridas cutâneas. Orientador: Joyce Maria Annichino Bizzacchi Tese (doutorado) - Universidade Estadual de Campinas, Faculdade de Ciências Médicas Resumo: A Trombose Venosa Profunda (TVP) é definida como um processo patológico resultante da alteração da parede do vaso, do fluxo sanguíneo, e/ou da composição sanguínea, o que leva à formação de um trombo dentro do vaso. Apesar do conhecimento atual, ainda há muito a se avançar no conhecimento dos fatores de risco associados à TVP. O endotélio vascular ao responder ativamente a estímulos fisiopatológicos, pode alterar seu padrão antitrombótico, e com a perda de sua integridade, liberar células endoteliais circulantes (CECs), que vêm sendo consideradas como possíveis marcadores para diversas doenças. Além disso, as células endoteliais formadoras de colônia (CEFCs) são um subtipo de células endoteliais progenitoras com notável potencial clonogênico e capacidade de vascularização in vivo, e devido a essas características de natureza endotelial, vem ganhando destaque a sua utilização como modelo ex vivo na investigação de doenças. Contudo, apesar do endotélio possuir um papel-chave na hemostasia, nenhum estudo avaliou as CEFCs na TVP aguda. Além disso, a impossibilidade de uma análise direta do endotélio, dificulta a compreensão do seu real papel na doença. Sendo assim, o objetivo principal deste trabalho foi caracterizar e avaliar a função celular das CEFCs de pacientes com TVP aguda em comparação à indivíduos saudáveis (CN). Os pacientes foram selecionados após a confirmação do diagnóstico de TVP de membros inferiores no período de julho de 2019 a setembro de 2021. A casuística foi constituída de 25 pacientes com TVP aguda (idade média 45 ± 13 anos), sendo que 20 retornaram para a segunda coleta (TVP >6 meses) e 38 CN (idade média 36 ±10 anos). Segregamos as avaliações em 2 grupos: (1) avaliação descritiva e laboratorial de pacientes com TVP aguda, TVP >6 meses e controles normais (CN) e (2) avaliação da função endotelial e parâmetros biológicos, com caracterização das células por imunofenotipagem e porcentagem de células senescentes, proliferação, apoptose, migração celular e capacidade de geração de trombina (TGT), comparadas ao CN. Também realizamos uma subanálise para a identificação de marcadores biológicos envolvidos no sucesso da obtenção de CEFCs previamente isoladas a partir do sangue periférico e expandidas em cultura. Como resultados, no grupo 1 demonstramos um aumento plasmático nas interleucinas (IL) pró-inflamatórias IL-6, IL-8, do fator de crescimento derivado plaquetário (PDGF-AA/AB) e da molécula de adesão P-selectina, e uma maior frequência de CECs nos pacientes com TVP aguda em relação aos CNs. Observamos que na TVP >6 meses a IL-6, IL-8, P-selectina e CECs estavam diminuídos em comparação à TVP aguda, porém observamos um aumento da Molécula de adesão intercelular solúvel 1 (sICAM-1) e Molécula de adesão vascular solúvel 1 (sVCAM-1). Quanto ao grupo 2, o sucesso na obtenção das CEFCs foi de 37% no grupo CN, 20% na TVP aguda e 25% na TVP >6 meses. Os testes de função celular das CEFCs em pacientes com TVP aguda vs. CN revelou células com menor capacidade proliferativa e de migração, e mais células em apoptose. Já o TGT demonstrou células com propriedades similares entre os grupos. As análises do sobrenadante celular evidenciaram o aumento da expressão de IL-6 nas CEFCs de pacientes com TVP aguda. Ainda não temos a finalização desses testes na TVP >6 meses para comparação da TVP em dois momentos distintos. Por fim, alguns fatores parecem estar envolvidos no surgimento e manutenção do cultivo celular, como o aumento de sICAM-1 e sVCAM-1 tanto no plasma como no sobrenadante das células que foram isoladas. Como conclusão, definimos que é possível estabelecer o isolamento das CEFCs em pacientes com TVP aguda, no entanto com baixas taxas de sucesso, e isso pode ser atrelado aos achados deste trabalho em relação a alteração das propriedades funcionais, principalmente na capacidade replicativa e de reparo celular. No mais, acreditamos que as células também podem ser afetadas com o ambiente extracelular na fase aguda frente à estímulos pró-inflamatórios Abstract: Deep Vein Thrombosis (DVT) is defined as a pathological process resulting from changes in the vessel walls, blood flow, and/or blood composition, which leads to the formation of a thrombus within the vessel. Despite current knowledge, much remains to be understood about the risk factors associated with DVT. When the vascular endothelium actively responds to pathophysiological stimuli, the antithrombotic pattern of this endothelium can change; and with the loss of integrity, occurs a release of endothelial cells (CECs), which have been considered possible markers for several diseases. In addition, endothelial colony-forming cells (ECFCs) are a subtype of endothelial progenitor cells with remarkable clonogenic potential and in vivo vascularization capacity, and due to these characteristics of endothelial nature, are being used as an ex vivo model in disease investigation. However, despite the endothelium playing a key role in hemostasis, no study has evaluated the ECFCs in acute DVT. In addition, the impossibility of a direct analysis of the endothelium makes the actual role of the endothelium in the disease difficult to understand. Therefore, the main objective of this work was to characterize and evaluate the cellular function of the ECFCs of patients with acute DVT compared to healthy individuals (controls, CN). Patients were selected after confirmation of the diagnosis of DVT of the lower limbs from July 2019 to September 2021. The groups consisted of 25 patients with acute DVT (mean age 45 ± 13 years), with 20 follow-ups (DVT>6 months) and 38 HI (mean age 36 ±10 years). We divided the assessments into 2 groups: (1) descriptive and laboratory assessment of patients with acute DVT, DVT >6 months, and HI; and (2) evaluation of endothelial function and biological parameters, with the characterization of cells by immunophenotyping and percentage of senescent cells, proliferation, apoptosis, cell migration, and thrombin generation capacity (TGT), compared to HI. In addition, we performed a sub analysis for the identification of biological markers involved in the success of obtaining ECFCs previously isolated from peripheral blood and expanded in culture. As a result, in group 1 we demonstrated a plasmatic increase of the pro-inflammatory interleukins (IL) IL-6, IL-8, platelet-derived growth factor (PDGF-AA/AB), and P-selectin adhesion molecule, and a higher frequency of ECs in patients with acute DVT compared to HI. Thus, we observed that in DVT >6 months IL-6, IL-8, P-selectin, and CECs were decreased compared to acute DVT, however, we observed an increase in Soluble Intercellular Adhesion Molecule 1 (sICAM-1) and Vascular Adhesion Molecule soluble 1 (sVCAM-1). In group 2, success in obtaining CEFCs was 37% in the HI group, 20% in the acute DVT, and 25% in the DVT >6 months. Cell function tests of ECFCs in patients with acute DVT vs. HI revealed cells with lower proliferative and migratory capacity, and more cells in apoptosis. TGT showed cells with similar properties between groups. Cell supernatant analyzes showed an increase in IL-6 expression in the ECFCs of patients with acute DVT. We have yet to complete these tests at DVT >6 months to compare DVT at two different times. Finally, some factors appear to be involved in the emergence and maintenance of cell culture, such as the increase in sICAM-1 and sVCAM-1 both in plasma and in the supernatant of cells that were isolated. In conclusion, we defined that it is possible to establish the isolation of ECFCs in patients with acute DVT, however with low success rates, and this can be linked to the findings of this work regarding the change in functional properties, especially in the replicative and regenerative capacity. O endotélio corneano é uma monocamada de células poligonais justapostas que recobrem a superfície posterior da córnea. É uma estrutura fundamental para a manutenção da transparência corneana. A espécie suína é bastante empregada como modelo experimental na oftalmologia. No entanto, os estudos avaliando o endotélio da córnea com microscópio especular nessa espécie são escassos. Objetivou-se avaliar a densidade endotelial e o percentual de células hexagonais nas diferentes regiões do endotélio corneano de suínos utilizando a microscopia especular de contato. Foram estudados 24 bulbos oculares de 12 suínos (Sus scrofa domesticus), machos, com seis meses de idade e da raça Large White. A microscopia especular de contato foi realizada nas regiões central, superior, inferior, lateral e medial da córnea. A densidade endotelial média na região central foi de 1865 células/mm², na região superior foi de 1877,3 células/mm², na região inferior foi de 1854,9 células/mm², na região lateral foi de 1847 células/mm² e na região medial foi de 1831,7 células/mm². Na região central, a porcentagem média de células hexagonais foi de 53,3%, na região superior foi de 53,8%, na região inferior foi de 54,4%, na região lateral foi de 53,8%, na região medial foi de 53,7%. Não foram observadas diferenças significativas na densidade celular e na hexagonalidade nas diferentes regiões analisadas da córnea. Os valores da densidade endotelial e da hexagonalidade da região central do endotélio podem ser extrapolados para as demais regiões da córnea. The corneal endothelium is a monolayer of juxtaposed polygonal cells that cover the posterior surface of the cornea. It is a fundamental structure for the maintenance of corneal transparency. The swine species is widely used as an experimental model in ophthalmology. However, studies evaluating the corneal endothelium with a specular microscope in this species are scarce. The objective was to evaluate the endothelial density and cell morphology of different regions of the porcine corneal endothelium using specular contact microscopy. Twenty-four eyeballs from 12 male, six-month-old, Large White pigs (Sus scrofa domesticus) were studied. Contact specular microscopy was performed in the central, superior, inferior, lateral and medial regions of the cornea. The mean endothelial density in the central region was 1865 cells/mm², in the upper region it was 1877.3 cells/mm², in the lower region it was 1854.9 cells/mm², in the lateral region it was 1847 cells/mm² and in the medial was 1831.7 cells/mm². In the central region, the average percentage of hexagonal cells was 53.3%, in the upper region it was 53.8%, in the lower region it was 54.4%, in the lateral region it was 53.8%, in the medial region was 53.7%. No significant differences were observed in cell density and hexagonality in the different analyzed regions of the cornea. The endothelial density and hexagonal values of the central region of the endothelium can be extrapolated to the other regions of the cornea.

Background: The endothelium is a layer fundamental to maintaining corneal transparency. In ophthalmology, sheep eyes have been used as a model in research related to corneal transplantation. Different techniques have been used to evaluate the corneal endothelium. Concerning vital dyes, corneal endothelial cell analyses have not yet been studied in ovines. The purpose of the present study was to evaluate the morphology of endothelial cells from different regions of the cornea of sheep after staining with alizarin red and trypan blue using an optical microscope. Materials, Methods & Results: Twenty healthy eyes of 10 male sheep obtained from a licensed commercial slaughterhouse were studied. The study was approved by the Research Committee of the Faculty of Veterinary at UFRGS and followed the ethical standards of the Association for Research in Vision and Ophthalmology (ARVO). Immediately after the slaughter, the eyes were enucleated and underwent eye examination. The corneal endothelium was stained with trypan blue and alizarin red and examined and photographed using an optical microscope. The central, superior, inferior, nasal and temporal areas of the cornea were evaluated for cell morphology. Data were compared by t-tests. Differences were considered statistically significant at P < 0.05. Immediately after staining the corneal endothelium, it was possible to examine with an optical microscope, obtain images and analyse the shape of endothelial cells from all regions of the sheep cornea. Polygonal, uniform and continuous cells were observed in all samples studied. Considering all the corneas analysed, cells with 6 sides (75.11%), 5 sides (12.76%) and 4 sides (12.12%) were found. In the central region of the cornea 75.91% of cells with 6 sides, 12.6% of cells with 5 sides and 11.48% with 7 sides were found. In the superior region of the cornea 76.07% of cells with 6 sides, 13.25% with 5 sides and 10.68% with 7 sides were found. In the lower region were found 74.72% of cells with 6 sides, 13% with 5 sides and 12.27% with 7 sides. In the temporal region, 74.14% were 6-sided cells, 11.42% had 5 sides, and 14.43% had 7 sides. Furthermore, in the nasal region, 74.72% of the cells had 6 sides, 13.54% had 5 sides, and 11.73% had 7 sides. No significant differences were found between cell morphology in all corneal regions evaluated. In addition, no significant difference was found when comparing the right eye with the left eye. Discussion: Different methods are used for the analysis of corneal endothelium. For ex vivo research optical microscopy after endothelial staining is an alternative low-cost technique that allows the analysis of all regions of the cornea. Quan¬titative analyses must characterise the endothelial parameters of the different species. The analysis of the morphology of corneal endothelium with an optic microscope after staining with alizarin red has been described as an effective, rapid and cost-efficient method, since this dye blends with the borated cells, allowing identification. In the present study, using optical microscopy and coloration with alizarin red it was possible to explore and obtain images of the ovine endothelium of all regions of the cornea. In the current study, the endothelium had a predominance of cells will 6 sides in all regions studied. This study allowed us to obtain images of the endothelium as well as quantitative data on the morphology of the different regions of the sheep cornea. This study demonstrated that morphology did not differ between the central and peripheral regions. The findings of this study represent a further source of reproducible data that should be considered when using sheep cornea as ex vivo model for experimental research.

No. of bitstreams: 0 Previous issue date: 2016-04-02 Although the role of soluble fms-like tyrosine kinase 1 (sFLT-1) in preeclampsia is well-established, the mechanism related to its synthesis remains poorly understood. We evaluated the association among the circulating microRNAs (miRs) and sFLT-1 levels in preeclampsia pregnant women. For this purpose, we measured the plasma sFLT-1 levels in 24 preeclampsia women and selected from these, three subjects with the lowest and three with the highest levels of sFLT-1 in order to screen for potential miRs associated with plasmatic sFLT-1 concentrations using a polymerase chain reaction-array (PCR-array) methodology. From screening results, we found three statistically different expressed miRs with fold change (FC-high/low levels) ≥3.0: miR-195-5p (FC = 5.2 increase in group with high sFLT-1 levels), miR-16-5p (FC = 3.2; increase in group with high sFLT-1 levels), and miR-375 (FC = −3.0; decrease in group with high sFLT-1 levels) which were later validated in all samples (n = 24). To correlate these miRs and plasma sFLT-1 levels, we used two extremes of analysis. In one part, we compared 12 preeclampsia women with the lowest sFLT-1 levels (L-50% group) to 12 with the highest levels (50% H group); and in the second analysis, 6 preeclampsia women (L-25%) from the L group to 6 preeclampsia women from the H group (H-25%). Our results showed increased expression of miR-195-5p in the H group, considering both the analyses with 50%, FC = 2.1 and 25%, FC = 4.2. Regarding other miRs, lack of correlation was found in both analyses (50% and 25%). In conclusion, our data demonstrate an association of higher levels of sFLT-1 with increased expression of miR-195-5p in preeclampsia pregnant women. Orientador: Henrique Marques Barbosa de Souza Dissertação (mestrado) - Universidade Estadual de Campinas, Instituto de Biologia Resumo: A regeneração tecidual é um processo biológico inteiramente dependente do suporte de oxigênio e nutrientes e ocorre pelo brotamento de uma nova vasculatura no local da lesão através da angiogênese. Aqui, nós propomos que a expressão de Nf2f2 é um pré-requisito para a regeneração, precisamente através do seu papel no controle da angiogênese. Primeiro, nós encontramos 2.222 genes co-regulados com assinatura gênica de Nr2f2 na condição regenerativa transiente do coração camundongo, sugerindo que o NR2F2 pode regular a resposta regenerativa cardíaca. Ao analisar o conjunto de dados de RNA-seq e ChIP-seq disponíveis publicamente, descobrimos uma forte correlação entre a expressão de Nr2f2 e os tecidos capazes de regeneração epimórfica. Ao procurar por genes potencialmente induzidos e reprimidos por Nr2f2 em uma rede regulação gênica, nós encontramos muitos genes alvos diretos e indiretos de NR2F2 enriquecidos em termos de ontologia gênica (GO) para angiogênese e vias de sinalização pro-angiogênicas já conhecidas. Juntos, nossos resultados fornecem evidências fortes de que a maioria, se não todos, os processos de regeneração tecidual em mamíferos dependem da expressão de Nr2f2 em células endoteliais, para promover a angiogênese e restabelecer a homeostase do tecido Abstract: Tissue regeneration is a biological process entirely dependent on support of oxygen and nutrients by the sprouting of a new vasculature in local injury through angiogenesis. Here, we propose that Nf2f2 expression is prerequisite to regeneration, precisely through its role in controlling angiogenesis. First, we found 2.222 co-regulated genes with Nr2f2 signature in heart transient regenerative condition in mouse, suggesting that NR2F2 may regulate cardiac regenerative response. By analyzing RNA-seq and ChIP-seq dataset publicly available, we discovered a strong correlation between Nr2f2 expression and tissues capable of epimorphic regeneration. By looking for potential genes induced and repressed by Nr2f2 in a gene regulatory network, we found many NR2F2 direct and indirect target genes enriched in angiogenesis GO terms and known pro-angiogenic signaling pathways. Orientadores: Nicola Amanda Conran Zorzetto, Fernando Ferreira Costa Tese (doutorado) - Universidade Estadual de Campinas, Faculdade de Ciências Médicas Resumo: A anemia falciforme (AF) é uma doença causada por uma mutação de ponto (troca do aminoácido glutâmico pela valina), que resulta na síntese de uma hemoglobina anômala, a hemoglobina S (HbS). A principal causa de morbidade para portadores de AF é a vaso-oclusão, que resulta da adesão anormal de células vermelhas e brancas ao endotélio, e consequêntemente diminui o fluxo sanguíneo. As plaquetas de pacientes AF apresentam um aumento das suas propriedades adesivas, e por isso, sugere-se a sua participação no processo de vaso-oclusão. Nossos dados mostraram que essas plaquetas são capazes de ativarem células endoteliais, in vitro, em ensaios de co-culturas com células endoteliais de veia umbilical humana (HUVEC) com plaquetas. A expressão da molécula de adesão intercelular 1 (ICAM-1) e a da E-selectina das células HUVEC aumentou significativamente na sua superfície, após a sua co-incubação com plaquetas de pacientes AF. A ativação das moléculas de adesão depende do contato físico entre plaquetas e células HUVEC, pois a utilização de insertos de transwell inibiu significativamente a expressão de ICAM-1 e E-selectina na superfície das HUVEC. A co-incubação de plaquetas de pacientes AF com as células HUVEC resultou em uma maior produção e liberação de mediadores inflamatórios (citocinas); a interleucina 8 (IL-8) e da interleucina 1 beta (IL-1?) quantificadas do sobrenadante das co-culturas com essas células. As plaquetas desses pacientes possuem propriedades inflamatórias, pois liberaram mais fator plaquetário 4 (PF4), uma quimiocina pró-agregante, quantificada do sobrenadante de cultura dessas plaquetas, em relação às plaquetas de indivíduos saudáveis. O aumento na expressão das moléculas de adesão, integrina ?IIb?3 e P-selectina, na superfície das plaquetas de pacientes AF, podem indicar que há um aumento na sua ativação plaquetária, favorecendo a sua adesão ao endotélio inflamado. O fator nuclear-?B (NF-?B) é um fator de transcrição nuclear muito importante envolvido na ativação de genes, como genes que codificam de moléculas de adesão endotelial, e os nossos resultados de expressão gênica mostraram que há um aumento na expressão do gene NF?BIA (subunidade p50 do NF-?B), em associação com o aumento na expressão do gene ICAM1 (que codifica a molécula de adesão ICAM-1) em HUVEC, após co-cultura dessas células com plaquetas de pacientes AF. A presença do inibidor BAY 11-7082 (inibe a via de sinalização NF-?B), diminuiu a expressão das moléculas de adesão ICAM-1 e E-selectina na superfície das células HUVEC, quando co-incubadas com plaquetas de pacientes AF. Os nossos dados são importantes e corroboram com a nossa hipótese de que as plaquetas participam no processo de vaso-oclusão na anemia falciforme, pois apresentam a capacidade de ativar células endoteliais (in vitro) tornando-as inflamatórias e mais adesivas. Portanto, sugerimos que as plaquetas representam um importante alvo para novas abordagens terapêuticas na anemia falciforme Abstract: Sickle cell anemia (SCA) is a disease caused by a point mutation (causing the exchange of glutamic acid for valine), which results in the synthesis of an abnormal hemoglobin, hemoglobin S (HbS). The principal cause of morbidity in patients with SCA is vaso-occlusion, which results from the abnormal adhesion of red blood cells and white cells to blood vessel walls, leading to decreases in blood flow. Platelets from SCA patients present an increase in their adhesive properties and, therefore, we have previously suggested that they may participate in the vaso-occlusive process. Our current data show that platelets from SCA patients are able to activate endothelial cells, in vitro, in co-culture assays using Human Umbilical Vein Endothelial Cells (HUVEC) that were co-incubated with platelets from SCA patients. The expression of intercellular adhesion molecule 1 (ICAM-1) and E-selectin was significantly increased on the surface of HUVEC cells after their co-incubation with platelets from SCA patients (SCA platelets). The activation of adhesion molecule expression observed depended on the physical contact between platelets and HUVEC cells, as transwell inserts were able to significantly inhibit the expression of ICAM-1 and E-selectin on HUVEC following their culture in the presence of SCA platelets. The co-incubation of SCA platelets with HUVEC cells also resulted in a higher production and release of inflammatory mediators (cytokines), such as interleukin 8 (IL-8) and interleukin 1 beta (IL-1?) in the supernatant of these co-cultures. Platelets from SCA patients have inflammatory properties, releasing more platelet factor 4 (PF4), a pro-aggregating chemokine, than platelets from healthy subjects. The increased expression of the adhesion molecules, integrin ?IIb?3 and P-selectin, on the surface of platelets from SCA patients, may also indicate an increased platelet activation, favoring their adhesion to the inflamed endothelium. Nuclear factor-?B (NF-?B) is an important nuclear transcription factor involved in the activation of genes, including endothelial adhesion molecules genes. We found an increased gene expression of NF?BIA (encoding the p50 subunit of NF-kB), in association with increased gene expression of ICAM1 (encodes ICAM-1) in HUVEC, following the co-culture of these cells with platelets from SCA patients. The presence of the BAY 11-7082 inhibitor (inhibits the NF-?B signaling pathway) decreased the expressions of the ICAM-1 and E-selectin adhesion molecules on the surface of HUVEC cells when co-incubated with platelets from SCA patients. Our data corroborate our hypothesis that platelets probably participate in the process of vaso-occlusion in sickle cell anemia, since they have the ability to activate endothelial cells (in vitro), in turn, making them more inflammatory and adhesive. Orientador: Sara Teresinha Olalla Saad Tese (doutorado) - Universidade Estadual de Campinas, Faculdade de Ciências Médicas Resumo: Importante passo para a compreensão dos processos fisiopatológicos das neoplasias é a identificação de genes diferencialmente expressos e das funções biológicas de cada proteína codificada por estes genes. Rho GTPase activating protein 21 (ARHGAP21) é uma proteína pertencente à família RhoGAP, que interage com ARF-GTPases e com alfa-catenina, modulando a dinâmica da actina associada às membranas do Golgi e à integridade das junções aderentes. O objetivo geral do presente estudo foi investigar as funções da proteína ARHGAP21 em células endoteliais e de adenocarcinoma de próstata com relação à viabilidade celular, ciclo celular, migração, adesão e expressão gênica. Neste trabalho foi demonstrado que ARHGAP21 possui atividade RhoGAP para RhoA e RhoC. Em células HUVECs (Human Umbilical Vein Endothelial Cells), foi observada uma localização nuclear e citoplasmática de ARHGAP21 e sua depleção induziu alterações no ciclo celular. Além disso, ensaios de microarranjos de DNA em HUVECs demonstraram modulação de genes como PAI-1, BNIP3, staniocalcina 1 e podocalyxin. Em linhagens celulares de adenocarcinoma de próstata (LNCaP e PC-3), também foi observada uma localização nuclear e citoplasmática de ARHGAP21. A diminuição da expressão desta proteína em células PC-3 resultou em redução da viabilidade e da migração celular em fibronectina. Com relação à viabilidade celular, a inibição de ARHGAP21 tem efeito adicional ao agente quimioterápico cisplatina. Em células LNCaP, por sua vez, foi observada uma menor adesão em matrigel e fibronectina das células submetidas à inibição da expressão de ARHGAP21, em comparação às células controle. Experimentos de microarranjos de DNA e RT-PCR quantitativo em tempo real em células de adenocarcinoma de próstata submetidas à inibição de ARHGAP21 demonstraram expressão alterada dos genes TGF-beta induced e dos genes BNIP3, staniocalcina 1 e podocalyxin, que também foram modulados nas células HUVECs com inibição da expressão de ARHGAP21. Em conclusão, o presente estudo identificou ARHGAP21 como uma proteína com importantes funções em células endoteliais e de adenocarcinoma de próstata, através da regulação da viabilidade, adesão e migração celular. Os achados aqui descritos sugerem que ARHGAP21 pode ser uma molécula alvo para a terapia de neoplasias, provavelmente através da sua função como reguladora da atividade de RhoA e RhoC Abstract: One step in the path towards building a comprehensive molecular portrait of human cancer is the definition of differentially expressed genes and the function of their coding proteins. Rho GTPase activating protein 21 (ARHGAP21) is a RhoGAP protein, which interacts with ARF-GTPases and alpha-catenin, controlling actin dynamics on Golgi membranes and the integrity of adherens junctions, respectively. The aim of the present study was to investigate ARHGAP21 functions on endothelial and prostate cancer cells regarding to cell viability, cell cycle, migration, adhesion and gene expression. This study demonstrated that ARHGAP21 has RhoGAP activity for RhoA and RhoC. ARHGAP21 localized in the nucleus and cytoplasm of HUVECs (Human Umbilical Vein Endothelial Cells) and its depletion alteres the cell cycle phases. Furthermore, microarrays assays in HUVECs ARHGAP21 knockdown demonstrated modulation of genes such as PAI-1, BNIP3, stanniocalcin 1 and podocalyxin. In the prostate adenocarcinoma cell lines (LNCaP and PC-3), ARHGAP21 is also located in the nucleus and cytoplasm. Depletion of this protein in PC-3 cells resulted in decrease of cell viability and migration in fibronectin. Regarding to cell viability, inhibition of ARHGAP21 has an additional effect on cisplatin chemotherapeutic agent. In LNCaP cells, a lower adhesion was observed in matrigel and fibronectin of cells subjected to inhibition of ARHGAP21 expression compared to control cells. DNA microarray and quantitative RT-PCR experiments on prostate adenocarcinoma cells ARHGAP21 knockdown showed modulation of TGF-beta induced gene expression. The expression of BNIP3, stanniocalcin 1 and podocalyxin was also altered in HUVECs cells ARHGAP21 knockdown. In conclusion, this study identified ARHGAP21 as a protein with important functions in endothelial cells and prostate adenocarcinoma, by regulating cell viability, adhesion and migration. A sua capacidade de promover a reendotelização ou inibir a formação da neointima após injúria vascular é reconhecida nos últimos anos devido a sua eficácia em atuar na proliferação e na migração de células endoteliais, bem como na sua capacidade de inibir a adesão e agregação plaquetária. A neointima intra-stent pode ser combatida com o desenvolvimento de stents eluidores de drogas (SED). Os nitrosilos complexos de rutênio se destacam dentre os doadores de NO, devido a sua capacidade de liberar o NO de forma controlada e incorporados a veículos. Este estudo se propôs padronizar as condições de cultivo das células endoteliais umbilicais humana (HUVEC) "saudáveis", verificar a margem de segurança do composto trans-[Ru(NO)Cl(cyclam)](PF6)2, nomeado Ru(cyclam)NO, em células HUVEC por meio de ensaios de citotoxicidade, verificar sua capacidade de promover proliferação/migração celular e a inibição da adesão de plaquetas humanas, e, também verificar a biocompatibilidade deste composto quando incorporados em plataformas de aço. Expressão de proteínas fenotípicas da célula endotelial foi utilizada para padronização de cultivo de HUVEC. A concentração de 0,5mM foi definida como ótima nas condições experimentais. A ação do composto foi caracterizada pelas respostas de manutenção da proliferação (incorporação de timidina triciada - 3HT) e no aumento da migração celular (ensaio transwell), entretanto foi capaz de reduzir as ações estimulatórias do fator de crescimento epidermal (EGF). A citotoxicidade do complexo e das plataformas de incorporação foi determinada por ensaio de redução do 3-brometo de 4,5-dimetil-2-tiazolil-2,5-difenil-tetrazólio (MTT). Em relação ao nitroprussiato de sódio (SNP) a toxicidade induzida pelo composto nas concentrações de 0,5mM a 2,0mM foi reduzida, mesmo na maior concentração após 24 horas de exposição (MTT: 59% ± 4,42). O complexo mostrou ser capaz de reduzir a adesão ao fibrinogênio de plaquetas humanas ativadas ou não. A biocompatibilidade total das plataformas de inox, matriz de siloxano e matriz de siloxano incorporado com Ru(cyclam) foram demonstradas pelo ensaio de redução de MTT e a funcionalidade pelo ensaio de migração transwell. A margem de segurança do Ru(cyclam)NO e de suas respectivas plataformas de imobilização foram caracterizadas como baixa e ausente citotoxicidade. As avaliações funcionais evidenciaram que o Ru(cyclam)NO manteve a proliferação celular, estimulou a migração das HUVEC e inibiu a adesão de plaquetas humanas. Estas evidências constituem-se de pontos fundamentais para se obter a reendotelização Abstract: Nitric oxide (NO) is a multifunctional biological agent that has been the subject of several studies. In recent years, its ability to promote re-endothelialization or inhibit neointimal formation after vascular injury is recognized due to its efficacy in stimulates proliferation and migration of endothelial cells, as well as its ability to inhibit platelet adhesion and aggregation. The in-stent neointimal can be prevented with the development of drug-eluting stents (DES). Nitrosyl ruthenium complexes highlighted from the NO donors, due to their ability to release NO in a controlled manner and incorporated into vehicles. This study aimed to: 1) standardize the cultivation conditions of " healthy " human umbilical endothelial cells (HUVEC); 2) verify the safety margin of the compound trans - [Ru(NO)Cl(cyclam)](PF6)2, named Ru(cyclam) O, in HUVEC cells by cytotoxicity assay; 3) determine its ability to promote proliferation/migration cell and inhibition adhesion of human platelets; and also 4) verify biocompatibility of this compound when incorporated in slides. Phenotypic expression of the endothelial cell protein was used to standardize cultured HUVEC. A concentration of 0.5 mM was set to the optimal experimental conditions. The Ru(cyclam)NO action was characterized by responses of the proliferation maintenance (tritiated thymidine incorporation - 3HT ) and migration cell increased (transwell assay), however was able to reduce the stimulatory actions of epidermal growth factor (EGF). The cytotoxicity of the compound and its incorporation slides was determined by reduction of 3- bromide, 4,5-dimethyl -2-thiazolyl -2 ,5 -diphenyl tetrazolium bromide (MTT). In relation to sodium nitroprusside (SNP), Ru(cyclam)NO showed reduced toxicity from 0.5 mM to 2.0 mM, even at the highest concentration that was 59% ± 4.42 of the cell viability. The Ru(cyclam)NO was shown to reduce adhesion to fibrinogen of human platelets activated or not. The total biocompatibility of stainless slide with siloxane matrix and siloxane matrix incorporated with Ru(cyclam)NO were demonstrated by MTT reduction assay and their functionality by transwell migration assay. The safety margin of Ru(cyclam)NO and their respective slide incorporated were characterized as low or absent cytotoxicity, respectively. Functional evaluations showed that the Ru(cyclam)NO maintained cell proliferation and stimulated the migration of HUVEC, well as well, inhibited adhesion of human platelets. Em condições de hipóxia, a HbS sofre polimerização, alterando a forma e as propriedades da superfície dos eritrócitos. Indivíduos homozigotos para o alelo da hemoglobina S são portadores de anemia falciforme (AF), uma grave condição decorrente de manifestações clínicas, como ocorrência de hemólise e fenômenos vaso-oclusivos. A terapia transfusional é uma intervenção fundamental na diminuição da morbidade e mortalidade em pacientes com AF. Nos pacientes em regime de transfusão regular, níveis de HbS abaixo de 30% estão associados com menor frequência de agudizações da doença. O processo vaso-oclusivo e a lesão tecidual representam as principais manifestações da doença. O endotélio na anemia falciforme pode sofrer ativação e disfunção endotelial decorrente da hemólise intravascular e da inflamação crônica e, por interagir com hemácias, leucócitos e plaquetas no processo vaso-oclusivo. As células endoteliais ativadas induzem a via de sinalização inflamatória levando ao aumento da produção de mediadores inflamatórios e da expressão das moléculas de adesão. O objetivo desse estudo foi avaliar as propriedades adesivas e inflamatórias entre células endoteliais e plaquetas de pacientes com anemia falciforme em regime de transfusão regular em comparação a indivíduos controles sadios. Orientador: Cristina Pontes Vicente Dissertação (mestrado) - Universidade Estadual de Campinas, Instituto de Biologia Resumo: As células progenitoras endoteliais (CPEs) foram descritas por Asahara et al. (1997), isoladas a partir do sangue periférico e são células originadas da medula óssea. Estas células podem ser reconhecidas pela expressão de marcadores como o CD34, CD133, VEGFR2 e o CD31, pela capacidade de internalização de acLDL e de formar estruturas semelhantes a vasos quando cultivas em matrizes tridimensionais. Quando ocorrem lesões endoteliais, elas podem ser mobilizadas da medula migrando para o sangue periférico e atuar no local da lesão arterial, podendo se diferenciar em células endoteliais maduras e promover a re-endotelização do vaso lesionado. Contudo, estas células estão presentes na medula óssea e no sangue periférico em quantidade muito pequena, 0,1 e 0,01% do total de células respectivamente, o que leva diversos grupos de pesquisa buscarem selecionar e cultivar estas CPEs visando aumentar seu número para melhor caracterizá-las in vitro e in vivo. Em nosso trabalho, buscamos estabelecer um meio de cultura capaz de sustentar o crescimento, proliferação e diferenciação das células progenitoras endoteliais isoladas a partir de medula óssea de camundongos C57bl/6, e de caracterizar seus vários estágios de diferenciação destas células em diferentes tempos de cultura. Para tal, células mononucleares foram isoladas da medula de camundongos C57bl/6 por gradiente de Ficoll-Paque PLUS e cultivadas por 3, 7 e 14 dias em meios de cultura diferentes acrescidos de diferentes fatores de crescimento (VEGF, IGF, bFGF e EGF) com concentrações variadas. Dentre todos os meios testados, o DMEM1-M1 (DMEM com vermelho de fenol acrescido de VEGF, IGF e bFGF) foi o que apresentou melhores resultados para os ensaios de proliferação, de viabilidade celular por DAPI, Azul de Tripan e MTT. A partir deste meio foram realizados testes para verificar a expressão de marcadores celulares (CD133, CD34, VEGFR-2 e CD31) por imunocitoquímica, imunofluorescência, Western Blotting e citometria de fluxo. Os resultados de imunocitoquímica e imunofluorescência foram positivos para todos os marcadores, em todos os períodos analisados. A análise por Western Blotting confirmou a expressão destes marcadores, com aumento na expressão de 3 para 7 dias de cultura. Por citometria de fluxo analisamos o perfil destas células em cultura de 7 dias comparando com as células in vivo, e conseguimos visualizar um aumento principalmente na freqüência de células que expressam VEGFR-2, um indicativo de que o meio utilizado está selecionando células progenitoras. A internalização de acLDL que também indica CPEs foi positiva após 7 e 14 dias. As células cultivadas em Matrigel® formaram estruturas semelhantes a vasos após 7 dias de cultura. Nossos resultados indicam que fomos capazes de estabelecer um meio de cultura eficiente na seleção e proliferação de CPEs, inibindo sua diferenciação inicial em células endoteliais, o que nos permitirá utilizar esta técnica de cultura para expandir o número de CPEs obtidas da medula óssea e poder utilizá-las no tratamento de lesões arteriais e em diferentes processos de terapia celular Abstract: Endothelial progenitor cells (EPCs) were described by Asahara et al. (1997) isolated from peripheral blood, these cells are derived from bone marrow. EPCs can be characterized by the expression of cellular markers like CD34, CD133, VEGFR2 and CD31, internalization of acLDL and formation of structures similar to vessels when culture in tridimensional matrices. In the presence of endothelial lesion, they can be mobilized from bone marrow, migrating to peripheral blood and subsequently the endothelial lesion site. They can differentiate into mature endothelial cells and participate in the re-endothelization of the damaged vessel. However these cells are rare in bone marrow and peripheral blood 0,1 and 0,01%, respectively, several research groups are trying to develop strategies to isolate and cultivate these cells to better determine their characteristics in vitro and in vivo. In our study we tried to establish a culture medium able to sustain growth and proliferation of EPC obtained from C57bl/6 mice bone marrow and characterize the stages of differentiation that they could present in different times of culture. In order to do it, we isolated mononuclear cell from bone marrow using a Ficoll-Paque PLUS gradient and cultivated then for 3, 7 and 14 days using culture mediums with different compositions and concentrations of growth factors (VEGF, IGF, bFGF, EGF). Cellular growth determined by DAPI, trypan blue counting and MTT was more significant with the medium we named DMEM1-M1 (with VEGF, IGF and bFGF). Results obtained with immunocitochemistry and immunofluorescence with this medium was positive in all culture times. Western blotting demonstrated the expression of VEGFR2, CD34 and CD133 in 3 e 7 day, with a litte increase after 7 days. Flow citometry analysis of cells after 7 days in culture demonstrated an increase in cells expressing VEGFR2, indicating the selection of EPC with this medium. Internalization of acLDL and formation of structures similar to vessels were observed after 7 and 14 days in culture and confirms differentiation of mononuclear cells into EPC. So, in conclusion, our results indicate that we developed a culture medium able to sustain proliferation and differentiation of EPCs, inhibiting their terminal differentiation what will allow us to use this culture technique to increase the number of EPC from bone marrow obtained and in the future use these cells in the treatment of arterial lesions in different process of cellular therapy Mestrado Biologia Celular Mestre em Biologia Celular e Estrutural

While the neuronal underpinnings of autism spectrum disorder (ASD) are being unraveled, vascular contributions to ASD remain elusive. Here, we investigated postnatal cerebrovascular development in the 16p11.2df/+ mouse model of 16p11.2 deletion ASD syndrome. We discover that 16p11.2 hemizygosity leads to male-specific, endothelium-dependent structural and functional neurovascular abnormalities. In 16p11.2df/+ mice, endothelial dysfunction results in impaired cerebral angiogenesis at postnatal day 14, and in altered neurovascular coupling and cerebrovascular reactivity at postnatal day 50. Moreover, we show that there is defective angiogenesis in primary 16p11.2df/+ mouse brain endothelial cells and in induced-pluripotent-stem-cell-derived endothelial cells from human carriers of the 16p11.2 deletion. Finally, we find that mice with an endothelium-specific 16p11.2 deletion (16p11.2ΔEC) partially recapitulate some of the behavioral changes seen in 16p11.2 syndrome, specifically hyperactivity and impaired motor learning. By showing that developmental 16p11.2 haploinsufficiency from endothelial cells results in neurovascular and behavioral changes in adults, our results point to a potential role for endothelial impairment in ASD.

Although often beneficial, the inflammatory process is closely involved in the pathophysiology of several diseases. The use of anti-inflammatory drugs available on the market is still a problem for patients, as the prolonged use is associated with severe side effects. Therefore, is relevant to identify new molecules with therapeutic potential for the treatment of inflammatory disorders. Cyclic peptides have attracted interest from the pharmaceutical industry for drug development that more than 40 cyclic peptide-based drugs have been approved by the FDA for clinical use in the past decade. In addition to neuroprotective effects, the cyclic dipeptide cyclo (Glycine-Proline, CGP), has been described by our research group as having analgesic and anti-inflammatory effects by inhibiting hypernociception and the accumulation of leukocytes in inflamed tissue. From these results, our main objective was to identify cellular and molecular targets that could account for the anti-inflammatory action of CGP. Initially, we found that GCP does not present cytotoxicity in all tested concentrations (10-300 μM). Our results also showed that CGP significantly inhibited the adhesion of neutrophils to endothelial cells at all concentrations tested (10-50 μM). We verified that CGP did not affect the expression of ICAM-1 and E-selectin. Similarly, CGP also did not significantly affect the production of IL-6, IL-8, IL-12p70 and reactive oxygen species (ROS) by endothelial cells stimulated with TNF-α. When assessing the motility the endothelium, our results show that CGP did not affect cell migration. However, CGP induced a significant increase in calcium influx in endothelial cells stimulated with histamine, a phenomenon that was not observed when cells were stimulated with CGP alone. In this regard, evaluating the competition of CGP for binding to receptors coupled to protein G was evaluated by the NanoBRET technique, we found that CGP promoted an increase in the interaction between H1 and β2 receptors and their respective natural ligands, a phenomenon that was not observed for other receivers, such as A1, A2A, A2B, A3, VEGFR2, CXCR4 and CXCR7. Taken together, we demonstrated, for the first time, that CGP may interact with H1 and β2 receptors and their ligands in which it may be associated with the effects of this dipeptide on the endothelium. Os peptídeos cíclicos têm despertado interesse da indústria farmacêutica para o desenvolvimento de fármacos, tanto que mais de 40 medicamentos à base de peptídeos cíclicos foram aprovados pela agência reguladora de fármacos norte-americana FDA para uso clínico na última década. O dipeptídeo cíclico ciclo(Glicina-Prolina, CGP) além do efeitos neuroprotetores, tem sido descrito, pelo nosso grupo de pesquisa, como possuidor de efeitos analgésicas e anti-inflamatórias por inibir a hipernocicepção e o acúmulo de leucócitos no tecido inflamado. A partir destes resultados, buscamos identificar alvos celulares e moleculares que poderiam responder pela ação anti-inflamatória do CGP. Incialmente, constatamos a ausência de citotoxicidade do GCP, em todas as concentrações testadas (10-300 μM), o que garantiu segurança para interpretar os resultados obtidos. Em seguida, verificamos que a adesão de neutrófilos às células endoteliais tratadas com CGP foi inibida de forma significativa em todas as concentrações testadas (10-50 μM). Ao avaliar o conteúdo de moléculas de adesão nas células endoteliais verificamos que o CGP não afetou a expressão de ICAM-1 e E-selectina. De forma semelhante, o CGP também não afetou de forma significativa a produção de IL-6, IL-8, IL-12p70 e espécies reativas de oxigênio (ROS) por células endoteliais estimuladas com TNF-α. Ao avaliar a motilidade das células endoteliais verificamos que o CGP não afetou a migração celular. Entretanto, o CGP induziu um aumento significativo no influxo de cálcio em células endoteliais estimuladas com histamina, fenômeno que não foi observado quando as células foram estimuladas apenas com CGP. A este respeito, avaliando a competição do CGP pela ligação em receptores acoplados à proteína G foi avaliada pela técnica de NanoBRET, constatamos que o CGP promoveu um aumento na interação entre os receptores H1 e β2 e seus respectivos ligantes naturais, fenômeno que não foi observado para outros receptores, como A1, A2A, A2B, A3, VEGFR2, CXCR4 e CXCR7. Tomados em conjunto, demonstramos, pela primeira vez, que o CGP pode interagir com os receptores H1 e β2 e seus ligantes, de forma que tal interação pode estar associada aos efeitos deste dipeptideo sobre o endotélio.

Endothelial heterogeneity has important implications in health and disease. Molecular markers selectively expressed in the vasculature of different organs and tissues are currently being explored in targeted therapies with promising results in preclinical and clinical studies. Noteworthy is the role that combinatorial approaches such as phage display have had in identifying such markers by using phage as nanoparticles and surrogates for billions of different peptides, screening noninvasively the vascular lumen for binding sites. Here, we show that a new peptide motif that emerged from such combinatorial screening of the vasculature binds selectively to blood vessels in the brain in vivo but not to vessels in other organs. Peptides containing a conserved motif in which amino acids Phenylalanine-Arginine-Tryptophan (FRW) predominate could be visualized by transmission electron microscopy bound to the junctions between endothelial cells in all areas of the brain, including the optic nerve, but not in other barrier-containing tissues, such as intestines and testis. Remarkably, peptides containing the motif do not bind to vessels in the retina, implying an important molecular difference between these two vascular barriers. Furthermore, the peptide allows for in vivo imaging, demonstrating that new tools for studying and imaging the brain are likely to emerge from this motif.

Orientador: Erich Vinicius de Paula Dissertação (mestrado) - Universidade Estadual de Campinas, Faculdade de Ciências Médicas Resumo: É sabido que o heme extracelular livre ativa vários compartimentos da imunidade inata, atuando como um padrão molecular associado ao perigo (DAMP) ou alarmina, em doenças associadas à hemólise. Embora a interrupção regulada e localizada da barreira endotelial (BE) seja uma parte importante da ativação da imunidade inata, que permite que os leucócitos circulantes atinjam os tecidos inflamados, a perda de localização e/ou da regulação da integridade da BE pode levar a aumento da permeabilidade microvascular generalizado e a lesão tecidual secundária, como observado em choque séptico. Em camundongos, foi demonstrado que o heme livre causa quebra da BE (in vivo e em culturas de células endoteliais) e uma forma de lesão pulmonar aguda que se assemelha à Síndrome Torácica Aguda (STA), que é a complicação mais grave da anemia falciforme (AF). Embora os efeitos da quebra da BE pelo heme também tenham sido recentemente mostrados em culturas de células endoteliais humanas, até onde sabemos, nenhum estudo avaliou o efeito do soro de pacientes com AF na integridade da BE. Neste estudo, exploramos o efeito de heme e soro de pacientes com AF na integridade da BE usando monocamadas de células endoteliais humanas, em condições experimentais destinadas a mimetizar eventos que normalmente desencadeiam complicações agudas da AF como a STA. A integridade da BE foi avaliada em células endoteliais da veia umbilical humana (HUVECs) por método baseado na detecção elétrica de impedância celular (Electric Cell Impedance Sensing), na presença de heme, LPS e soro de pacientes com AF (n=20) ou voluntários saudáveis (n=10). O heme causou uma diminuição dose-dependente (de 20 a 100 µM) na resistência normalizada das monocamadas, consistente com a redução da integridade da BE. O LPS exerceu efeito aditivo com o heme em relação à redução da integridade da BE em HUVECs. O soro de pacientes com AF induziu uma diminuição transitória da resistência, não observada com o soro de voluntários saudáveis. A adição do heme ao soro de pacientes com AF foi capaz de induzir uma perda significativa da integridade da BE, indicando que os efeitos do heme sobre a BE também ocorrem na presença de soro. Curiosamente, este efeito não foi observado quando o heme foi adicionado ao soro de voluntários saudáveis. Nossos resultados apoiam o conceito de que o heme livre participa da patogênese da STA, ao agregar dados gerados com células humanas e com amostras de pacientes, e sugerem que o ambiente inflamatório, presente nos soros de pacientes com AF, seja necessário para esse efeito do heme Abstract: Free extracellular heme has been shown to activate several compartments of innate immunity, acting as a danger-associated molecular pattern in diseases associated with hemolysis. Although localized endothelial barrier (EB) disruption is an important part of innate immunity activation, that allows circulating leukocytes to reach inflamed tissues, loss of localization and/or regulation of EB disruption can lead to widespread microvascular hyperpermeability and secondary tissue damage, such as observed in septic shock. In mice, free heme has been shown to cause EB disruption (both in vivo and in endothelial cell cultures), and a form of acute lung injury that closely resembles acute chest syndrome (ACS), the most severe complication of sickle cell disease (SCD). Although the barrier-disrupting effects of heme have also been recently shown in human endothelial cell cultures, to our knowledge no studies evaluated the effect of serum from patients with SCD in EB integrity. Here we explored the effect of heme and sera from SCD patients on EB integrity using human endothelial cell monolayers, in experimental conditions aimed to mimic events that normally trigger acute SCD complications such as ACS. EB integrity was assessed in human umbilical vein endothelial cells (HUVECs) by electric cell-substrate impedance sensing, in the presence of heme, LPS and sera from SCD patients (n=20) or healthy volunteers (n=10). Heme caused a dose-dependent (20 to 100 µM) decrease in transendothelial electrical resistance, consistent with EB disruption. LPS caused an additive effect with heme on EB integrity in HUVECs. Sera from SCD patients induced a transient resistance decrease not observed with serum from healthy volunteers. Finally, heme caused transient EB disruption even in the presence of human serum, but this effect could only be observed with SCD patients serum, but not healthy volunteers. O presente estudo teve como objetivo avaliar o efeito da Terapia Fotodinâmica (PDT) e da Fotobiomodulação (PBM) como terapias complementares ao protocolo de revascularização pulpar, em cultura celular primária de células da papila apical humana (APCs) e em células endoteliais (HUVECs). Dois primeiros pré-molares inferiores com um canal foram seccionados, simulando dentes com rizogênese incompleta, e um dos canais foi irrigado com 1mL de azul de metileno 0.005%. A seguir, os dois canais foram irrigados com 20mL de Hipoclorito de sódio (NaOCl) 1.5% e 20mL de EDTA. Antes de cada ensaio, as soluções químicas obtidas foram diluídas em meios de cultura na concentração de 0.5% (NaOCl + EDTA 0.5%; NaOCl + EDTA + MB 0.5%). O efeito da PDT e da PBM em cultura de células foi avaliado por meio dos seguintes grupos: CTG (meio de cultura); ISG (NaOCl + EDTA 0.5%); MBG (MB + NaOCl + EDTA 0.5%); PBMG (NaOCl + EDTA 0.5% + laser); PDTG (NaOCl + EDTA + MB 0.5% + laser). Foram realizados os ensaios de citotoxicidade em APCs, de índice de polaridade em APCs, migração celular em APCs e angiogênese em HUVECs. Os dados obtidos foram analisados pelo ANOVA, seguido do teste de Tuckey (P≤0.05). A PDT e a PBM aumentaram a viabilidade celular em comparação com os grupos ISG e MBG, respectivamente. Todos os protocolos testados reduziram a migração celular em comparação com CTG. A PBMG mostrou maior distância de migração das HUVECs em comparação a ISG, evidenciando um potencial de angiogênese. Conclui-se que a PBM e a PDT têm potencial para serem empregadas como terapias coadjuvantes ao protocolo de revascularização pulpar em dentes com rizogênese incompleta. The aim of this study was to evaluate photodynamic therapy (PDT) and photobiomodulation therapy (PBM) as complementary therapies on a pulp revascularization protocol using primary culture of cells from apical papilla (APCs) and endothelial cells (HUVECs). In order to simulate tooth with open apex, two mandibular first premolars with a single canal were sectioned and one of the root canal was irrigated with 1mL of 0.005% methylene blue (MB). Next, the root canal was irrigated with 20mL of 1.5% sodium hypochlorite (NaOCl) and 20mL of 17% EDTA. The chemical solution obtained was diluted in cell culture media previous to each experiment (NaOCl + EDTA 0.5%; NaOCl + EDTA + MB 0.5%). The effect of PDT and PBM was evaluated accordingly to these groups: CTG (cell culture media); ISG (NaOCl + EDTA 0.5%); MBG (MB + NaOCl + EDTA 0.5%); PBMG (NaOCl + EDTA 0.5% + laser); PDTG (NaOCl + EDTA + MB 0.5% + laser). It was performed cytotoxic assay, polarity index and migration analysis in APCs and sprouting assay in HUVECs. All data was analyzed by ANOVA followed by Tuckey post-hoc test (P≤0.05). PDT and PBM favored viability compared to ISG and MBG, respectively. All tested protocols reduced cell migration compared to the CTG. PBMG demonstrated increased migration distance of HUVECs when compared to ISG, what might be related to an angiogenesis potential. PBM and PDT have potential to be employed as complementary therapies on pulp revascularization protocol of necrotic tooth with open apex.

Dissertação de Mestrado em Investigação Biomédica apresentada à Faculdade de Medicina As doenças cardiovasculares (DCV) permanecem a principal causa de morte a nível global. Apesar de uma significativa melhoria em prevenção e no desenvolvimento de novas terapias, o número total de mortes devido as DCVs aumentaram em 12,5% na última década. Este aumento é devido a fatores de risco individuais, como o sedentarismo, abuso de álcool e/ou outras substâncias. No geral a combinação destes fatores pode levar à ocorrência de um enfarte agudo do miocárdio (EAM).Um EAM é caracterizado por redução ou bloqueio de perfusão no miocárdio, culminando, eventualmente em morte celular irreversível devido as condições isquémicas. De maneira a minimizar o dano no coração, é essencial restabelecer o fluxo sanguíneo, no entanto, esta reperfusão leva a danos adicionais a um coração já danificado, este fenómeno é chamado de lesão de isquemia/reperfusão (I/R). Sendo assim, terapias inovadoras capazes de melhorar e/ou resolver estas complicações são necessárias.Vesiculas extracelulares (VEs) são importantes mediadores de comunicação celular. A sua composição, tanto na sua superfície como o seu conteúdo, retrata o estado da célula que os libertou, e VEs de diferentes origens celulares e até de diferentes fluidos biológicos são usados como biomarcadores e agentes terapêuticos. Presentemente, acredita-se que os principais componentes responsáveis por esta bioatividade nos EVs, são RNAs não codificantes, em particular microRNAs (miRNA).Numa primeira fase desta tese, testamos a bioatividade de VEs isolados de urina de pacientes que sofreram um EAM, em células endoteliais, e numa segunda parte, exploramos o papel de um novo miRNA, previamente identificado no nosso grupo como capaz de aumentar a sobrevivência em células endoteliais em condições de isquemia. Para entregar este miRNA de interesse (a partir de agora, denominado miR-B) nas células de interesse, exploramos duas estratégias: complexação do miRNA com uma lipídica formulação comercial (lipofectamina RNAiMAX) e complexação do miRNA com VEs isolados de urine de dadores saudáveis. Em última análise, testamos estas abordagens na sobrevivência de células endoteliais expostas a condições isquémicas e mostramos que os VEs-modulados com miRNA eram mais eficazes em aumentar a sobrevivência endotelial do que a lipofectamina. Com estes resultados, concluímos que o miR-B aumenta a sobrevivência de células endoteliais expostas a condições de isquemia e que os VEs de dadores saudáveis serviram como o veículo ideal para a entrega do miRNA às células. Cardiovascular diseases (CVD) remain the leading cause of death globally. Despite significant improvements in prevention and in the development of novel therapeutic strategies, the total number of deaths due to CVD has increased 12.5% in the past decade. This increase is likely due to individual risk factors, like physical inactivity, alcohol and/or other substances abuse as well as environmental factors such as pollution. Overall, the combination of these factors can ultimately lead to an acute myocardial infarction (AMI).An AMI is characterized by a reduction or blockage of myocardial perfusion, eventually culminating in irreversible cell death due to the ischemic conditions that ensue. To minimize the damage to the heart, it is essential to restore the blood flow as quickly as possible. However, this reperfusion will lead to additional harm to the already injured heart, a phenomenon known as ischemia/reperfusion injury. Thus, novel therapies capable of ameliorating and/or resolve these complications are much needed. Extracellular vesicles (EVs) are important mediators of intercellular communication. Their composition, both the surface as well as the cargo, portray the state and composition of the releasing cell, and EVs from many different cellular origins and from different biological fluids have been used as biomarkers and/or therapeutic agents. Currently, it is believed that the major components driving the bioactivity of EVs are non-coding RNAs, in particular microRNAs (miRNA). In the first part of this thesis we tested the bioactivity of EVs isolated from urine of AMI patients in endothelial cells (ECs) and in the second part we explored the role of a novel miRNA, previously identified in our group as capable of enhancing the survival of EC in ischemic conditions. To deliver this miRNA of interest (from here on denoted miR-B) into the cells we explored two strategies: complexation of the miRNA with a commercial lipidic formulation (lipofectamine RNAiMax) and complexation of miRNA with EVs isolated from the urine of healthy donors. Ultimately, we tested these approaches on the survival of ECs exposed to ischemic conditions and showed that miRNA-modulated EVs were more effective on enhancing EC survival than lipofectamine. Based on the above-mentioned results, we concluded that miR-B enhances the survival of ECs exposed to ischemia and that urine EVs from healthy donors can be used as an effective vehicle to deliver miRNA into the cells. O objetivo do presente estudo foi avaliar, através de experimentos in vitro, o efeito da fotobiomodulação com laser de baixa intensidade na proliferação e viabilidade de células endoteliais humanas. Células endoteliais da veia do cordão umbilical humano (HUVEC) foram cultivadas e divididas em quatro grupos: C - controle (sem irradiação) e grupos L1, L4 e L7.5 – células irradiadas com doses de 1; 4; e 7,5 J/cm², respectivamente, de laser diodo (InGaAlP; comprimento de onda de 660 nm; potência de 30 Mw, em dose única). A atividade metabólica das células foi avaliada nos intervalos de 24, 48 e 72 horas após a irradiação, através do ensaio do Alamar Blue. A viabilidade e a proliferação celular também foram avaliadas no intervalo de 72h, através dos ensaios de Live/Dead, Annexin V/PI, análise das fases do ciclo celular e imunomarcação da proteína Ki67. Um dos principais mecanismos que levam às doenças cardiovasculares é a disfunção do endotélio, associada com a diminuição da biodisponibilidade de óxido nítrico (NO) e a produção excessiva de espécies reativas de oxigênio (ROS). Do ponto de vista de biomarcadores de estresse oxidativo/antioxidante e de envelhecimento biológico, enzimas com propriedades antioxidantes e o comprimento do telômero podem apresentar efeitos na modulação do sistema vascular. O presente estudo inclui dois manuscritos, sendo o primeiro relacionado a via de regulação NRF2/HO-1 e o segundo ao comprimento dos telômero em onívoros (ONI) e vegetarianos (VEG). No primeiro manuscrito, o objetivo foi verificar a concentração de HO-1 circulante, bem como investigar o efeito da incubação do plasma de ONI e VEG em células endoteliais sob a modulação da via NRF2/HO-1 e a produção de NO. Dos 745 indivíduos inicialmente recrutados, 44 ONI e 44 VEG do sexo masculino aparentemente saudáveis foram incluídos no estudo. A concentração de HO-1 circulante foi mensurado usando o ensaio de ELISA. As células endoteliais foram incubadas com amostras de plasma de ONI e VEG. Nós observamos que a concentração de HO-1 circulante foi maior nos ONI em relação aos VEG. A incubação das células endoteliais com o plasma de ONI induziu o aumento da expressão gênica/proteica do NRF2 e HO-1, bem como a atividade do ARE e a produção de NO, quando comparado com o grupo VEG. Estes achados indicam que a produção de HO-1 circulante nos ONI pode estar relacionada a um maior status pro-oxidativo. A ativação da via HO-1/NRF2 e a produção de NO em cultura de células endoteliais incubadas com o plasma de ONI, parece refletir um mecanismo adaptativo de proteção destas células contra possíveis danos. No segundo manuscrito, nosso objetivo foi avaliar o comprimento do telômero em leucócitos (LTL, do inglês leukocyte telomere length) nos ONI e VEG e sua associação com biomarcadores clássicos de risco cardiovascular. O LTL foi mensurado em 39 amostras de ONI e 41 de VEG através da reação em cadeia da polimerase (PCR) quantitativa em tempo real (RT-QPCR). Apesar de não haver diferença no comprimento do telômero entre ONI e VEG, uma forte correlação negativa entre LTL e a espessura intima-média da carótida (IMT, do inglês intima-media thickness) foi observada nos ONI, mas não no grupo VEG. Além disso, os ONI que foram classificados com o LTL curto apresentaram maior IMT comparado com VEG. Nossos achados sugerem que o comprimento do telomero pode ser utilizado como um marcador de aterosclerose subclinico no grupo de onívoros. Several studies report that a vegetarian diet lowers the probability of developing cardiovascular diseases (CVDs). The endothelial dysfunction is one of the main mechanism that leads to CVDs, associated with decreased nitric oxide (NO) bioavailability and excessive production of reactive oxygen species (ROS). Regarding oxidative/antioxidant stress and biological aging biomarkers, enzymes with antioxidant properties and telomere length may have effects on vascular system modulation. The present study includes two manuscripts related to: 1) regulation of NRF2/HO-1 pathway and 2) telomere length in omnivorous (OMN) and vegetarians (VEG). In the first manuscript, our objectives were to verify circulating HO-1 levels and the effect of plasma incubation from omnivorous and vegetarians in endothelial cells on modulating of NRF2/HO-1 pathway and NO production. From 745 participants initially recruited, 44 omnivorous and 44 vegetarian men apparently healthy were included in this study and circulating HO-1 was measured using ELISA assay. Endothelial cells were incubated plasma samples from OMN and VEG. We found higher circulating HO-1 production in omnivorous compared to vegetarian. Moreover, the plasma collected from omnivorous was able to increase the gene/protein NRF2/HO-1 expression, ARE activity, and NO production in endothelial cells culture compared to vegetarian group. We suggest that HO-1 induction in omnivorous may indicate a pro-oxidative. Activation of the HO-1 / NRF2 pathway and NO production in endothelial cell culture incubated with ONI plasma seems to reflect an adaptive mechanism of protection of these cells against possible damage. In the second manuscript, our objectives were to evaluate leukocyte telomere length (LTL) in VEG and OMN subjects and its association with classical cardiovascular risk biomarkers. LTL was measured in 39 omnivorous and 41 vegetarians by Real-Time Quantitative PCR reaction. Although telomere length was not different between omnivorous and vegetarians, we found a strong negative correlation between LTL and intima-media thickness (IMT) in omnivorous, but not in vegetarian group. In addition, omnivorous who were classified with short telomere length had higher carotid IMT compared to vegetarians. Our data suggest that telomere length can be a marker of subclinical atherosclerosis in the omnivorous group. FAPESP: 2015/20669-8

A interação entre os trofoblastos extravilosos humanos e as células endoteliais desempenha um importante papel nos processos de implantação embrionária e placentação. Alterações nos padrões de comunicação entre estas duas populações celulares estão associadas com complicações gestacionais, neste contexto, a infecção por Toxoplasma gondii pode ser um problema em potencial para essa relação. Assim, o objetivo desse estudo foi avaliar os fenômenos de invasão, migração e morte celular de células trofoblásticas extravilosas (linhagem HTR-8/SVneo) sob a influência de células endoteliais venosas (linhagem HUVEC) infectadas por T. gondii. As células HTR-8/SVneo foram co-cultivadas com as células HUVEC infectadas ou não por T. gondii, no sentido de avaliar a produção de citocinas detectadas pelo método de ELISA, bem como funções biológicas das células trofoblásticas extravilosas. A capacidade invasiva das células trofoblásticas foi analisada diante a utilização de insertos de transwell previamente cobertos com Matrigel, a capacidade migratória dessas células foi verificado pelo ensaio de scratch e o índice de morte celular foi mensurado através do método de citometria de fluxo. A interação entre as células HTR-8/SVneo e HUVEC induziram a secreção de IL-6 e IL-8 pelas células trofoblásticas. Já a secreção de MIF foi diminuída pelas células HTR-8/SVneo após a co-cultura com as células HUVEC não infectadas, e maiores níveis dessa citocina foram observados após a co-cultura com células HUVEC infectadas. O estímulo da co-cultura com células HUVEC não infectadas reduziu a capacidade invasiva e aumentou a migração de células HTR-8/SVneo. De forma contrária, as condições de células HUVEC infectadas aumentaram a capacidade invasiva, reduziram a migração e induziram o índice de morte celular das células trofoblásticas. O tratamento com ISO-1, inibidor altamente específico de MIF, alterou os eventos relacionados com a capacidade invasiva e morte celular das células HTR-8/SVneo quando comparados com cada condição respectiva sem o tratamento com inibidor de MIF. A comunicação entre as células trofoblásticas extravilosas e células endoteliais venosas foi modificada pela infecção por T. gondii e a indução da produção de MIF pelas células HTR-8/SVneo juntamente com a alteração dos índices de morte celular, podem ter sido alguns dos fatores responsáveis pela variação das funções trofoblásticas relacionadas à capacidade invasiva e migratória das células trofoblásticas extravilosas. The interaction between human extravillous trophoblast and human endothelial cells has an important role in embryonic implantation and placentation. Changes in communication patterns between these two cell populations are associated with pregnancy complications, in this context, Toxoplasma gondii infection can be a potential problem in this crosstalk. Therefore, the aim of this study was to evaluate the occurrence of invasion, migration and cell death of extravillous trophoblast cells (HTR-8/SVneo cell line) under influence of infected human venous endothelial cell (HUVEC) by T. gondii. HTR-8/SVneo cells were co-cultured with T. gondii-infected or uninfected HUVECs in order to analyze cytokine production by ELISA as well biological functions of extravillous trophoblast cells. The invasive capacity of trophoblastic cells was analyzed using transwell inserts previously covered with Matrigel, the migratory capacity of these cells was verified by scratch method and the cell death index was measured by the flow cytometry. HTR-8/SVneo cells and HUVECs interaction induced secretion of IL-6 and IL-8 by trophoblastic cells. MIF secretion was downregulated by HTR-8/SVneo cells after co-culture with uninfected HUVECs, but upregulated MIF levels were detected after co-culture with infected HUVECs. Stimuli of uninfected HUVECs co-culture reduced invasive capacity and increased migration of HTR-8/SVneo cells. Otherwise, infected HUVECs condition enhanced invasive capability, reduced migration, induced cell death index of trophoblastic cells. The treatment with ISO-1, highly specific MIF inhibitor, altered the events related to invasive capacity and cell death of HTR-8/SVneo when compared to each conditions without treatment with MIF inhibitor. The extravillous trophoblast-venous endothelial cells crosstalk was changed by T. gondii infection and induction of MIF production by HTR-8/SVneo cells together with changes in cell death index, might be some of the factors responsible for alteration in trophoblastic functions related to migratory and invasive capacity of extravillous trophoblastic cells. Por sua vez, o sistema esquelético abriga um conjunto de células que, de maneira hierárquica, sustentam a formação do osso ao longo da vida, sendo sua capacidade regenerativa comprometida durante o processo de envelhecimento. Com o aumento da expectativa de vida, a população idosa vem crescendo nos últimos anos e com ela o aumento eminente de fraturas ósseas. Sabe-se que o desenvolvimento e regeneração ósseos são eventos complexos e controlados por mecanismos parácrinos de sinalização intercelulares, destacando que a osteogênese está acoplada, principalmente, à angiogênese. Embora relatado, este mecanismo de comunicação entre células endoteliais e células osteoprogenitoras não está bem elucidado, sobretudo considerando o repertório de moléculas tróficas envolvidas. A fim de compreender melhor estes mecanismos, o objetivo deste trabalho foi desenvolver metodologias capazes de mimetizar o microambiente endotelial-ósseo, bem como desvendar eventos acoplados entre os diferentes tipos celulares envolvidos no processo, sobretudo gerando tecido endotelial in vitro, equivalente ao original. Para isso, fizemos uso de células humanas primárias, as quais foram submetidas a diferentes protocolos experimentais, onde a mesma densidade de células endoteliais arterial (HCAEC) e venosa (HUVEC) e de musculatura lisa (AoSMC) (cultivo misto) foi plaqueada em tubos cônicos sem tratamento e, após 72 horas, os esferóides gerados foram tecnicamente processados para caracterização bioquímica (expressão das proteínas CD31 e α-SMA pelas células endoteliais e musculares lisa, respectivamente), estrutural (através de microscopia eletrônica) e morfológica (avaliações histológicas). Os resultados obtidos demonstram uma sequência experimental capaz de gerar esferóides de tecido endotelial com propriedades funcionais. Como nota, essas análises preliminares mostram uma concentração de células endoteliais (positivas para CD31+) ao centro do esferóide, enquanto células de musculatura lisa concentram-se perifericamente (positivas para α-SMA), estabelecendo uma distribuição hierárquica-estrutural entre as duas linhagens primárias. Os dados moleculares aqui obtidos evidenciam que estes eventos de homeostase do tecido requerem processamento de microRNAs (miRs) e envolvimento do fator indutor de hipóxia (HIF-1α), o qual foi significantemente mais expresso por células de musculatura lisa, localizada na periferia do tecido gerado. Os resultados mostram ainda que os esferóides gerados expressam biomarcadores osteogênicos (RUNX 2, OTX, ALP), podendo, assim, contribuir com eventos de diferenciação osteoblástica. Variando estratégias experimentais através da modulação da atividade de HIF-1α, mostramos haver efeito diferencial entre esferóides arteriais e venosos, em processos de diferenciação osteoblástica a partir de células indiferenciadas (CTM-MS). With the progressive increase in the life expectancy of the population, bone defects became a public health problem. In turn, the skeletal system houses a set of cells that, in a hierarchical way, support the formation of the bone throughout life, and its regenerative capacity is compromised during the aging process. With the increase in life expectancy, the elderly population has been increasing in recent years and with it the eminent increase of bone fractures. It is known that bone development and regeneration are complex events and controlled by paracrine mechanisms of intercellular signaling, emphasizing that osteogenesis is mainly coupled with angiogenesis. Although reported, this mechanism of crosstalk between endothelial cells and osteoprogenitor cells is not well elucidated, especially considering the repertoire of involved trophic molecules. In order to better understand these mechanisms, the objective of this work was to develop methodologies capable of mimicking the endothelial-bony microenvironment, as well as unveiling the coupled events between the different cell types involved in the process, especially generating endothelial tissue in vitro, equivalent to the original one. For this, we used primary human cells, which were submitted to different experimental protocols, where the same density of arterial (HCAEC) and venous (HUVEC) endothelial cells and smooth muscle cell (AoSMC) (mixed culture) were plated in tubes (s), and after 72 hours the generated spheroids were technically processed for biochemical characterization (expression of CD31 and α-SMA proteins by smooth muscle and endothelial cells, respectively), structural (through electron microscopy) and morphological (histological evaluations). The results obtained demonstrate an experimental sequence capable of generating endothelial tissue spheroids with functional properties. As a note, these preliminary analyzes show a concentration of endothelial cells (positive for CD31) at the center of the spheroid, while smooth muscle cells concentrate peripherally (positive for α-SMA), establishing a hierarchical-structural distribution between the two primary lineages . The molecular data obtained here evidenced that these tissue homeostasis events require the processing of microRNAs (miRs) and involvement of the hypoxia inducing factor (HIF-1α), which was significantly more expressed by smooth muscle cells located at the periphery of the tissue generated. The results also show that the generated spheroids express osteogenic biomarkers (RUNX 2, OTX, ALP), thus contributing to osteoblastic differentiation events. By varying experimental strategies by modulating the activity of HIF-1α, we show a differential effect between arterial and venous spheroids in processes of osteoblastic differentiation from undifferentiated cells (CTM-MS). Together, the datas demonstrate that the methodology investigated provides an experimental flow capable of generating a model of 3D culture of venous and arterial equivalent endothelial tissue, with defined cellular, biochemical and ultrastructural properties, capable of contributing in a way to the osteogenic phenotype with dynamic and differential involvement of HIF-1α. Como uma interface entre o sangue e a parede do vaso, as ECs ocupam um local único diretamente exposto ao shear stress (SS), a força mecânica de atrito lateral produzido pelo fluxo de sangue na membrana apical da célula endotelial, que pode influenciar o comportamento de ambas ECs e AoSMCs. Geralmente, AoSMCs não sofrem diretamente às forcas de cisalhamento, no entanto, estas são diretamente expostas ao fluxo sanguíneo quando ocorre alguma injúria vascular, como por exemplo em algumas lesões ateroscleróticas ou por técnicas invasivas, como a angioplastia. As forças hemodinâmicas influenciam as propriedades funcionais do endotélio, porém estas não são profundamente compreendidas quanto aos mecanismos bioquímicos de respostas de células endoteliais e de musculatura lisa. Assim, a proposta desta dissertação foi estabelecer um modelo de cultivo in vitro que mimetize as forças tensionais de cisalhamento (shear stress), buscando compreender mecanismos celulares, bioquímicos e epigenéticos. Cultura de células primárias endoteliais e de musculatura lisa humanas foram obtidas da empresa LONZA e mantidas conforme recomendações do fabricante. Estas células foram mantidas rotineiramente em condições convencionais em incubadora de CO2. Para mimetizar o fluxo sanguíneo, estas células, separadamente, foram semeadas em anel periférico de uma placa de petri (100mm de diâmetro), modificada de acordo estudos anteriores, e mantidas sob fluxo laminar por 72 horas, quando as células foram coletadas e encaminhadas para as diferentes metodologias. Nossos resultados estão apresentados em 2 capítulos nessa dissertação, sendo o Cap 2 dedicado aos mecanismos envolvidos na adaptação de células endoteliais ao shear stress, e o Cap. 3, dedicado aos mecanismos desencadeados em células musculares lisa. De um modo geral, nossos resultados mostram respostas diferenciais entre células endoteliais e de musculatura lisa ao shear-stress, as quais são regidas por mecanismos epigenéticos no controle da expressão gênica desencadeados pelo remodelamento da matriz extracelular, avaliada aqui como um mecanismo importante de adaptação destas células às forças de cisalhamento. Endothelial cells (ECs) and smooth muscle cells (AoSMCs) are the major cellular components of the endothelium. Interactions between these cell types play roles in homeostasis and vascular structure. As an interface between the blood and the vessel wall, ECs occupy a single site directly exposed to shear stress (SS), the mechanical lateral friction force produced by blood flow on the apical membrane of the endothelial cell, which can influence behavior of both ECs and AoSMCs. Generally, AoSMCs do not directly undergo shear forces, however, they are directly exposed to blood flow when vascular injury occurs, for example in some atherosclerotic lesions or by invasive techniques such as angioplasty or surgical endarterectomy. Hemodynamic forces influence the functional properties of the endothelium, but these are not deeply understood as to the biochemical mechanisms of endothelial cell and smooth muscle responses. Thus, the purpose of this dissertation was to establish an in vitro culture model that mimics the shear forces, seeking to understand cellular, biochemical and epigenetic mechanisms. Culture of human endothelial and smooth muscle cells were obtained from the company LONZA and maintained according to the manufacturer's recommendations. These cells were routinely maintained under standard conditions in CO2 incubator. To mimic blood flow, these cells were separately seeded in a peripheral ring of a petri dish (100mm diameter), modified according to previous studies, and maintained under laminar flow for 72 hours, when the cells were collected and sent to the different methodologies. Our results are presented in 2 chapters in this dissertation, with Cap. 2 dedicated to the mechanisms involved in the adaptation of endothelial cells to shear stress, and Cap. 3, dedicated to the mechanisms triggered in smooth muscle cells. Overall, our results show differential responses between endothelial and smooth muscle cells at shear stress, which are governed by epigenetic mechanisms in the control of gene expression triggered by the remodeling of the extracellular matrix, evaluated here as an important mechanism of adaptation of these cells to shear forces. 2016/22270-8

Purpose: To evaluate the expression of endothelial and inducible NOS in addition to the miRNA-27b in the corpus cavernosum and peripheral blood of healthy rats, diabetic rats, alcoholic rats and rats with both pathologies. Methods: Forty eight Wistar rats were divided into four groups: control (C), alcoholic (A), diabetic (D) and alcoholic-diabetic (AD). Samples of the corpus cavernosum were prepared to study protein expressions of eNOS and iNOS by immunohistochemistry and expression of miRNA-27b in the corpus cavernosum and peripheral blood. Results: Immunohistochemistry for eNOS and iNOS showed an increase in cavernosal smooth muscle cells in the alcoholic, diabetic and alcoholic-diabetic groups when compared with the control group. Similarly, the mRNA levels for eNOS were increased in cavernosal smooth muscle (CSM) in the alcoholic, diabetic and alcoholic-diabetic groups and miRNA-27b were decreased in CSM in the alcoholic, diabetic and alcoholic-diabetic groups. Conclusion: The major new finding of our study was an impairment of relaxation of cavernosal smooth muscle in alcoholic, diabetic, and alcoholic-diabetic rats that involved a decrease in the nitric oxide pathway by endothelium-dependent mechanisms accompanied by a change in the corpus cavernosum contractile sensitivity.

Hypoxia and necrosis are fundamental features of glioma, and their emergence is critical for the rapid biological progression of this fatal tumor. The presence of vaso-occlusive thrombus is higher in grade IV tumors [glioblastoma multiforme (GBM)] compared with lower grade tumors, suggesting that the procoagulant properties of the tumor contribute to its aggressive behavior, as well as the establishment of tumor hypoxia and necrosis. Tissue factor (TF), the primary cellular initiator of coagulation, is overexpressed in GBMs and likely favors a thrombotic microenvironment. Phosphatase and tensin homolog (PTEN) loss and hypoxia are two common alterations observed in glioma that may be responsible for TF upregulation. In the present study, ST1 and P7 rat glioma lines, with different levels of aggressiveness, were comparatively analyzed with the aim of identifying differences in procoagulant mechanisms. The results indicated that P7 cells display potent procoagulant activity compared with ST1 cells. Flow cytometric analysis showed less pronounced levels of TF in ST1 cells compared with P7 cells. Notably, P7 cells supported factor X (FX) activation via factor VIIa, whereas no significant FXa generation was observed in ST1 cells. Furthermore, the exposure of phosphatidylserine on the surface of P7 and ST1 cells was investigated. The results supported the assembly of prothrombinase complexes, accounting for the production of thrombin. Furthermore, reverse transcription-quantitative polymerase chain reaction showed that CoCl2 (known to induce a hypoxic-like stress) led to an upregulation of TF levels in P7 and ST1 cells. Therefore, increased TF expression in P7 cells was accompanied by increased TF procoagulant activity. In addition, hypoxia increased the shedding of procoagulant TF-bearing microvesicles in both cell lines. Finally, hypoxic stress induced by treatment with CoCl2 upregulated the expression of the PAR1 receptor in both P7 and ST1 cells. In addition to PAR1, P7, but not ST1 cells, expressed higher levels of PAR2 under hypoxic stress. Thus, modulating these molecular interactions may provide additional insights for the development of more efficient therapeutic strategies against aggressive glioma.

Summary Background & aims Protein malnutrition (PM) affects hematopoiesis leading to bone marrow (BM) hypoplasia and arrests hematopoietic stem cells (HSC) in G0/G1 cell cycle phases, which cause anemia and leukopenia. Hematopoiesis is mainly regulated by BM niches where endothelial cells (EC) present a key regulatory role. Thus, our objective is to evaluate whether PM affects the modulatory capacity of EC on hematopoiesis. Methods C57BL/6 male mice received for 5 weeks a normal protein diet (12% casein) or a low protein diet (2% casein). MSC were isolated and differentiated in vitro into EC and the synthesis of SCF, Ang-1, CXCL-12, IL-11, TGF-β and G-CSF were evaluated. The HSC and hematopoietic progenitors were quantified and the EC capacity to modulate the hematopoietic system was also evaluated. Moreover, the ability of PM bone marrow to support hematopoieisis was assessed by proliferation of infused leukemic myelo-monoblasts cells. Results PM decreases HSC and hematopoietic progenitor pool and promotes cell cycle arrest and a lower proliferation rate of leukemic myelo-monoblasts. PM also committed hematopoietic regulatory characteristics from EC, resulting in the modification of both cell cycle pattern and hematopoietic differentiation. Conclusion BM microenvironment is compromised in PM, and since PM disturbs EC, it becomes one of the factors responsible for the hematopoietic cell cycle arrest and impairment of HSC differentiation.

Ramos is thankful to CAPES for financial support Abstract: Although redox processes closely interplay with mechanoresponses to control vascular remodeling, redox pathways coupling mechanostimulation to cellular cytoskeletal organization remain unclear. The peri/epicellular pool of protein disulfide isomerase-A1 (pecPDIA1) supports postinjury vessel remodeling. Using distinct models, we investigated whether pecPDIA1 could work as a redox-dependent organizer of cytoskeletal mechanoresponses. In vascular smooth muscle cells (VSMCs), pecPDIA1 immunoneutralization impaired stress fiber assembly in response to equibiaxial stretch and, under uniaxial stretch, significantly perturbed cell repositioning perpendicularly to stretch orientation. During cyclic stretch, pecPDIA1 supported thiol oxidation of the known mechanosensor beta(1)-integrin and promoted polarized compartmentalization of suifenylated proteins. Using traction force microscopy, we showed that pecPDIA1 organizes intracellular force distribution. The net contractile moment ratio of platelet-derived growth factor-exposed to basal VSMCs decreased from 0.90 +/- 0.09 (IgG-exposed controls) to 0.70 +/- 0.08 after pecPDIA1 neutralization (P < 0.05), together with an enhanced coefficient of variation for distribution of force modules, suggesting increased noise. Moreover, in a single cell model, pecPDIA1 neutralization impaired migration persistence without affecting total distance or velocity, whereas siRNA-mediated total PDIA1 silencing disabled all such variables of VSMC migration. Neither expression nor total activity of the master mechanotransmitter/regulator RhoA was affected by pecPDIA1 neutralization. However, cyclic stretch-induced focal distribution of membrane-bound RhoA was disrupted by pecPDI inhibition, which promoted a nonpolarized pattern of RhoA/caveolin-3 cluster colocalization. Accordingly, FRET biosensors showed that pecPDIA1 supports localized RhoA activity at cell protrusions versus perinuclear regions. Thus. pecPDI acts as a thiol redox-dependent organizer and noise reducer mechanism of cytoskeletal repositioning, oxidant generation, and localized RhoA activation during a variety of VSMC mechanoresponses. International audience; Head and neck squamous cell carcinoma (HNSCC) represent aggressive classes of tumors with a high mortality rate. The mammalian target of rapamycin (mTOR) pathway is instrumental in their initiation and expansion. Although results from pre-clinical models promise mTOR targeting as a potent novel therapeutic approach, its impact on the tumor microenvironment, such as endothelial cells is only scarcely investigated. Here, we first confirmed the effects of mTOR pharmacological inhibition on cell viability, clonogenicity, and proliferation in HNSCC human cell lines, HN26, and HN30. While Everolimus and Torin1 potently blunted mTOR-based proliferation of HN26 and HN30 lines, endothelial cells were left intact. To further explore the possibility of a paracrine bystander action of HNSCC-treated cells on endothelial cells, conditioned medium from Everolimus-and Torin1-challenged HN26 and HN30 cells were collected and applied to naive human endothelial cells. Although endothelial cell viability was again not modified, morphology and mobility were changed. Indeed, spreading of endothelial cells was altered upon challenge with mTOR-pretreated tumor conditioned-media, as measured via cell impedance and imagery. Interestingly, this was associated with an augmentation of focal adhesion kinase (FAK) active phosphorylation and enhanced migratory behavior. From a molecular standpoint, the production of vascular endothelial growth factor was elevated in treated HNSCC cells and might contribute to FAK phosphorylation. Although mTOR inhibition in tumor cells did hinder their growth, it also favors the release of factors that subsequently enable endothelial cell migration. Objective: To characterize the calcium influx pathways implicated in the sustained elevation of endothelial intracellular calcium concentration, required for the synthesis and release of relaxing factors. Methods: We evaluated the effect of the newly synthesized pyrazole derivatives, described as selective inhibitors for ORAI (BTP2/Pyr2 and Pyr6) and TRPC3 (Pyr3 and Pyr10) channels, upon endothelium- and extracellular calcium-dependent relaxations stimulated by acetylcholine and thapsigargin, in pre-constricted rat thoracic aortic rings. Results: Acetylcholine and thapsigargin responses were completely reverted by Pyr2 and Pyr6 (1 to 3μM). Pyr3 (0.3 to 3μM) caused a rapid reversal of acetylcholine (6.2±0.08mg.s−1) and thapsigargin (3.9±0.25mg.s−1) relaxations, whereas the more selective TRPC3 blocker Pyr10 (1 to 3μM) had no effect. The recently described TRPC4/5 selective blocker, ML204 (1 to 3μM), reverted completely acetylcholine relaxations, but minimally thapsigargin induced ones. Noteworthy, relaxations elicited by GSK1016790A (TRPV4 agonist) were unaffected by pyrazole compounds or ML204. After Pyr2 and Pyr6 pre-incubation, acetylcholine and thapsigargin evoked transient relaxations similar in magnitude and kinetics to those observed in the absence of extracellular calcium. Sodium nitroprusside relaxations as well as phenylephrine-induced contractions (denuded aorta) were not affected by any of pyrazole compounds (1 to 3μM). Conclusion: These observations revealed a previously unrecognized complexity in rat aorta endothelial calcium influx pathways, which result in production and release of nitric oxide. Pharmacologically distinguishable pathways mediate acetylcholine (ORAI/TRPC other than TRPC3/TRPC4 calcium-permeable channels) and thapsigargin (TRPC4 not required) induced calcium influx. RESUMO Objetivo: Caracterizar as vias do influxo de cálcio envolvidas no aumento sustentado da concentração intracelular de cálcio na célula endotelial, essencial para a síntese e a liberação de fatores relaxantes. Métodos: Analisamos o efeito de derivados pirazólicos sintetizados recentemente, descritos como inibidores seletivos para canais ORAI (BTP2/Pyr2 e Pyr6) e TRPC3 (Pyr3 e Pyr10), nos relaxamentos dependentes de endotélio e cálcio extracelular, produzidos por acetilcolina e tapsigargina, em anéis pré-contraídos da aorta torácica de rato. Resultados: As respostas de acetilcolina e tapsigargina foram completamente revertidas por Pyr2 e Pyr6 (1 a 3μM). Pyr3 (0,3 a 3μM) produziu reversão rápida dos relaxamentos de acetilcolina (6,2±0,08mg.s−1) e tapsigargina (3,9±0,25mg.s−1), enquanto o bloqueador mais seletivo para TRPC3, Pyr10 (1 a 3μM), não apresentou efeito. ML204 (1 a 3μM), bloqueador seletivo de TRPC4, descrito há pouco tempo, reverteu os relaxamentos induzidos por acetilcolina de forma completa, mas afetou minimamente aqueles produzidos por tapsigargina. Os derivados pirazólicos ou ML204 não afetaram os relaxamentos estimulados com GSK1016790A (TRPV4-agonista). Ainda, após pré-incubação com Pyr2 e Pyr6, acetilcolina e tapsigargina provocaram relaxamentos transitórios semelhantes em magnitude e cinética àqueles observados na ausência de cálcio extracelular. Os relaxamentos do nitroprussiato de sódio e as contrações induzidas pela fenilefrina (aorta sem endotélio) não foram afetados pelos compostos pirazólicos (1 a 3μM). Conclusão: Essas observações revelaram uma complexidade desconhecida das vias de influxo de cálcio no endotélio da aorta de rato, que resultam na produção e na liberação de óxido nítrico. Vias distinguíveis farmacologicamente medeiam o influxo estimulado por acetilcolina (ORAI TRPC, diferentes de TRPC3 TRPC4) e tapsigargina (TRPC4 não requerido).

INTRODUÇÃO: As doenças cardiometabólicas têm sido associadas ao elevado consumo de bebidas adoçadas. Estes produtos são responsáveis pela maior porção do consumo total de frutose na dieta e sugere-se que a ingestão excessiva deste monossacarídeo possa promover efeitos negativos na saúde, devido algumas particularidades de seu metabolismo. Entretanto, faltam dados na literatura demonstrando os efeitos deletérios do consumo diário de frutose sobre a função endotelial, influenciados pelas possíveis alterações nas concentrações de ácido úrico, e se o treinamento aeróbio é capaz de prevenir ou atenuar estes prejuízos em humanos. OBJETIVO: Verificar o efeito de 4 semanas de dieta rica em frutose associada ao treinamento aeróbio sobre fatores de risco para doenças cardiometabólicas, incluindo as concentrações de ácido úrico e marcadores relacionados a função endotelial. MÉTODOS: O presente estudo trata-se de um ensaio clínico randomizado controlado, cego e em paralelo. Vinte e um homens e mulheres adultos jovens, com idades entre 19 e 35 anos, foram randomizados em três grupos: Dieta Rica em Frutose (DRF), Dieta Rica em Glicose (DRG) e Dieta Rica em Frutose e Exercício (DRFE). Durante as 4 semanas de intervenção, os sujeitos foram submetidos à dieta personalizada, incluindo 1g/kg de massa corporal de frutose ou glicose por dia. O grupo DRFE realizou 3 sessões semanais de 60 minutos de exercício aeróbio em intensidade moderada (65% da frequência cardíaca obtida no consumo de oxigênio de pico) em bicicleta ergométrica. Antes e após este período, foram realizadas coletas de sangue, para verificar as concentrações de ácido úrico, nitrito/nitrato (NOx), endotelina-1 (ET-1), colesterol total (CT), triglicerídeos (TG), lipoproteína de baixa densidade (LDL), lipoproteína de alta densidade (HDL), glicose, insulina, proteína C-reativa (PCR) e substâncias reativas ao ácido tiobarbitúrico (TBARS), e foram avaliadas as variáveis dilatação mediada por fluxo (DMF), índice de resistência à insulina (HOMA2-IR), índice da capacidade funcional das células beta pancreáticas (HOMA2-%B), índice de sensibilidade à insulina (ISI) e pressão arterial de 24 horas (MAPA). As comparações foram realizadas através das Equações de Estimativas Generalizadas (GEE), adotando-se os fatores grupo e tempo. Foi utilizado o post-hoc de LSD para identificar diferenças. Todos os resultados foram expressos em média e desvio padrão e o nível de significância aceito foi 5%. Para todas as variáveis, o tamanho de efeito (TE) foi calculado (Cohen d). RESULTADOS: Observou-se efeito de interação entre grupo e tempo para as concentrações de ácido úrico (p=0,035), no qual o grupo DRF exibiu aumento (4,46±1,11 mg/dL para 4,74±1,22 mg/dL; TE: 0,22; p=0,040) da semana 0 para a semana 4. O grupo DRG (5,27±1,56 mg/dL para 5,40±1,32 mg/dL; TE: 0,08; p=0,573) e o grupo DRFE (5,40±0,87 mg/dL para 5,66±0,66 mg/dL; TE: 0,31; p=0,220) não apresentaram alterações significativas entre a semana 0 e a semana 4, porém DRFE aumentou as concentrações de ácido úrico entre a semana 2 e a semana 4 (p=0,013). As concentrações de glicose também apresentaram interação entre grupo e tempo (p≤0,001), sem alterações entre os momentos pré e pós. Porém foi encontrado aumento entre a semana 2 e a semana 4 no grupo DRF (p=0,037) e, no grupo DRFE, aumento da semana 0 para a semana 2 (p=0,019) seguido de redução da semana 2 para a semana 4 (p=0,003). Por outro lado, demonstrou-se interação para a pressão arterial sistólica (PAS) de 24h (p=0,029), na qual o grupo DRFE apresentou queda do momento pré para o momento pós intervenção (110,68±8,20 mmHg para 107,17±8,15 mmHg; TE: 0,40; p=0,018). Também foram observadas interações para as variáveis PAS de sono (p=0,010) e pressão arterial diastólica (PAD) de sono (p=0,008). O grupo DRF apresentou incrementos na PAS (98,98±9,13 mmHg para 104,99±9,58 mmHg; TE: 0,59; p=0,022) e na PAD (53,71±6,32 mmHg para 60,98±9,95 mmHg; TE: 0,80; p=0,009) de sono e o grupo DRG exibiu reduções na PAS (103,12±5,82 mmHg para 100,26±4,15 mmHg; TE: 0,52; p=0,028) e PAD de sono (56,29±6,43 mmHg para 54,80±5,56 mmHg; TE: 0,23; p=0,031) após as 4 semanas. CONCLUSÃO: O consumo de 1g/kg de massa corporal de frutose por dia, durante 4 semanas, é capaz de aumentar as concentrações de ácido úrico e promover incrementos na pressão arterial de sono em adultos saudáveis. Por outro lado, o exercício aeróbio associado ao consumo de frutose atenua as alterações nas concentrações de ácido úrico e impede os prejuízos causados pela frutose na pressão arterial de sono. INTRODUCTION: Cardiometabolic diseases have been associated with high consumption of sweetened beverages. These products are responsible for the largest portion of total dietary fructose consumption and it is suggested that excessive intake of this monosaccharide may promote negative health effects due to some particularities of its metabolism. However, there is a lack of data in the literature demonstrating the deleterious effects of daily fructose consumption on endotelial function, influenced by possible changes in uric acid concentrations, and whether aerobic training can prevent or mitigate these impairments in humans. AIM: To verify the effect of 4 weeks high fructose diet associated with aerobic training on risk factors for cardiometabolic diseases, including uric acid concentrations and endotelial function-related outcomes. METHODS: This study is a blinded, paralleled randomized controlled trial. Twenty-one young adult men and women, aged 19 to 35 years, were randomized into three groups: Fructose Rich Diet (DRF), Glucose Rich Diet (DRG) and Fructose Rich Diet and Exercise (DRFE). During the 4 weeks of intervention, subjects underwent a personalized diet, including 1 g / kg body weight of fructose or glucose per day. The DRFE group performed 3 weekly 60-minute sessions of moderate-intensity aerobic exercise (65% of heart rate obtained on peak oxygen uptake). Before and after this period, blood samples were taken to verify the concentrations of uric acid, nitrite / nitrate (NOx), endothelin-1 (ET-1), total cholesterol (TC), triglycerides (TG), low-density lipoprotein (LDL), high-density lipoprotein (HDL), glucose, insulin, C-reactive protein (CRP) and thiobarbituric acid-reactive substances (TBARS), and the variables flow-mediated dilatation (FMD), resistance index insulin (HOMA2-IR), pancreatic beta cell functional capacity index (HOMA2-%B), insulin sensitivity index (ISI) and 24-hour blood pressure. Comparisons were performed using Generalized Estimation Equations (GEE), adopting the group and time factors. The LSD post hoc was used to identify differences. All results were expressed as mean and standard deviation and the accepted significance level was 5%. For all variables, the effect size (ES) was calculated (Cohen d). RESULTS: An interaction effect between group and time was observed for uric acid concentrations (p = 0.035), in which the DRF group showed an increase (4.46 ± 1.11 mg / dL to 4.74 ± 1.22 mg / dL; ES: 0.22; p = 0.040) from pre to post intervention. The DRG group (5.27 ± 1.56 mg / dL to 5.40 ± 1.32 mg / dL; ES: 0.08; p = 0.573) and the DRFE group (5.40 ± 0.87 mg / dL to 5.66 ± 0.66 mg / dL; ES: 0.31; p = 0.220) showed no significant changes between the pre and post intervention, but DRFE increased the uric acid concentrations between the half of the intervention and the moment after intervention (p = 0.013). Glucose concentrations also showed interaction between group and time (p≤0.001), without changes between the pre and post moments. However, an increase was found between the half and the end of the intervention in the DRF group (p = 0.037) and, in the DRFE group, an increase from pre to half the intervention (p = 0.019) followed by a reduction from half to the post intervention moment (p = 0.003). On the other hand, an interaction was observed for systolic blood pressure (SBP) of 24h (p = 0.029), in which the DRFE group presented a decrease from pre to post intervention (110.68 ± 8.20 mmHg to 107.17 ± 8.15 mmHg; ES: 0.40; p = 0.018). Interactions were observed for nocturnal SBP (p = 0.010) and nocturnal diastolic blood pressure (DBP) (p = 0.008). The DRF group showed increases in nocturnal SBP (98.98 ± 9.13 mmHg to 104.99 ± 9.58 mmHg; ES: 0.59; p = 0.022) and nocturnal DBP (53.71 ± 6.32 mmHg to 60.98 ± 9.95 mmHg; ES: 0.80; p = 0.009) and the DRG group showed reductions in nocturnal SBP (103.12 ± 5.82 mmHg to 100.26 ± 4.15 mmHg; ES: 0.52; p = 0.028) and nocturnal DBP (56.29 ± 6.43 mmHg to 54.80 ± 5.56 mmHg; ES: 0.23; p = 0.031) after 4 weeks. CONCLUSION: The daily consumption of 1 g / kg body mass of fructose for 4 weeks is able to increase uric acid concentrations and promote increases in nocturnal blood pressure in healthy adults. On the other hand, aerobic exercise associated with fructose consumption attenuates changes in uric acid concentrations and prevents the deleterious effects of fructose in nocturnal blood pressure.
