Background: This study aims to describe and test a tiered approach for assessing compliance to Environmental Quality standards (EQSs) for priority substances in biota in line with the European Water Framework Directive. This approach is based on caged gammarids and trophic magnification factors (TMFs) at the first tier, with fish analysed at the second tier at sites exceeding the EQS. A dataset was implemented by monitoring perfluorooctane (PFOS) in caged gammarids exposed at 15 sites in French rivers, and in fish muscle and rest-of- body from the same sites. Isotopic ratios (δ13C and δ15N) were also measured in gammarids and fish. Two scenarios were developed to compare measured PFOS concentrations in fish against predicted concentrations based on measures in caged gammarids and TMFs. Scenario (1) compared measured PFOS concentrations in fish fillets with predicted PFOS concentrations based on measured concentrations in caged gammarids and δ15N. Scenario (2) tested whether or not EQS exceedance was correctly predicted based on measured concentrations in caged gammarids and trophic levels (TLs) from wild fish and gammarid populations.Results: δ13C and δ15N variations showed that caged gammarids used local food resources during exposure in the field. PFOS concentrations in gammarids were fairly variable through time at each site. In fish, concentrations ranged from non-quantifiable to 250 ng g− 1 (wet weight). After adjustment to the TL at which the EQS is set, 12 sites were above the EQS for PFOS. In scenario (1), predicted concentrations were almost correct at 7 sites out of 15. Most incorrect predictions were overestimations that were slightly improved by applying a lower TMF. In scenario (2) we tested several variants for parameters involved in the predictions. The most efficient combination yielded two wrong predictions out of 15. This result was obtained with a higher TMF value, mean concentrations in gammarids from several field exposures, and a TL for gammarids at the median of the distribution in French rivers.Conclusion: The proposed tiered approach was thus efficient. However, the number of sites was relatively limited, and the dataset was biased towards EQS exceedance. The tiered approach warrants further validation.

TRANSKARST is an interdisciplinary research project with scientists and regional water resource administrators. This 3 years project (2019-2022) is set up on Arcier's karstic watershed used for the drinking water supply of 60 000 inhabitants of Besançon, France. Using this instrumental basin, as a part of Jurassic Karst French observation system, the project aims at defining pathways of mineral, organic and microbiological contamination in karstic system.The methodology used in the project, combines field expertise with the implementation of analytical tools related to conventional - dissolved phase chemistry (major ions, miners, traces, organic carbon), isotopes (oxygen-18, deuterium, carbon-13) – and prospective analysis - emerging pollutants such as pharmaceuticals and ETM, dissolved organic matter, suspended matter and microbiology (bacteria, fungal species) with a special focus on antibiotic resistance. The TRANSKARST project thus brings together a consortium of researchers from various disciplines: hydrogeology, chemistry, sedimentology, microbiology, geology and geophysic. The project is also highly associated to water managers as Arcier’s spring is used for drinking water supply.The three main under-objectives following by TRANSKARST could be summarized as follows. First, through geological and geophysical investigation, a conceptual and numerical geological model will be established under geomodeler and will be further used to constrain the pathways of karstic groundwater. The second point focus on the evaluation of karst media contamination: chemical (including emerging molecules) and microbiological. Then we expect to discriminate, by the use of ETM, dissolved, colloidal and particulate pathways of contamination. All the observations will be linked together to identify actions and feedback of different parameters and contaminants within karst hydrosystems.

Communication orale, résuméCommunication orale, résumé; Le nombre et la diversité des contaminants organiques (pesticides, HAP, PCB, médicaments,…) potentiellement présents dans l’environnement est une réalité à laquelle sont confrontées les recherches portant sur le devenir et les impacts de ces substances. Selon les estimations, entre 30 000 et 100 000 substances sont concernées, il est donc impossible de les étudier au cas par cas. Cette difficulté se rencontre notamment dans le contexte de la Directive Européenne REACH (Registration, Evaluation, Authorisation and Restriction of Chemical substances) qui vise à mettre en place un système d’enregistrement de 30 000 substances chimiques pour lesquelles des informations précises sur la dispersion de ces substances dans l’environnement et leurs impacts écotoxicologiques sont exigées. Parallèlement à ces besoins en lien direct avec le travail d’évaluation des risques, notre communauté scientifique ressent fortement la nécessité de classer les contaminants organiques en fonction de leurs propriétés essentielles dans les mécanismes responsables de leur comportement dans l’environnement, de leur passage dans les organismes vivants et de leurs effets toxicologiques. Nous avons récemment développé une méthodologie permettant de classer des contaminants organiques selon des propriétés directement liées aux processus d’intérêt environnementaux, par exemple leur rétention irréversible, leur dégradation biotique ou abiotique, leur transfert vers l’air ou les eaux, leur passage dans les organismes vivants ou encore leurs effets toxicologiques. Cet outil, appelé TYPOL, est basé à la fois sur une approche «in silico» calculant des descripteurs moléculaires et sur des méthodes de classification combinant descripteurs moléculaires et paramètres comportementaux. Les classes sont construites à partir d’une analyse statistique (type PLS) des propriétés déduites de la prise en compte des caractéristiques structurales des molécules considérées et des paramètres environnementaux. Le choix des relations entre paramètres et descripteurs est en grande partie basée sur des relations de type QSAR (structure-activité). De par son mode de construction, l’outil est adaptable à différentes questions de recherche et concerne l’ensemble des contaminants organiques existants ou potentiels. Il vise à extrapoler des connaissances au sein d’une classe de composés à partir des résultats obtenus sur quelques molécules. Il est composé d'une base de données en MySQL accessible sur un serveur commun. L'ajout de nouvelles données se fait soit par transfert d'un fichier tableur (type Excel), soit par modification de la base à l'aide d'un outil web (phpMyAdmin). Les interfaces d'interrogation de la base de données et des traitements statistiques sont programmées dans le langage R qui intègre de nombreuses bibliothèques de méthodes de traitements statistiques des données. Les sorties graphiques sont aussi réalisées par le logiciel R. Des perspectives d’améliorations sont envisagées : - Ajouts de paramètres environnementaux. Un travail important sera à réaliser autour des données manquantes pour combler une partie des manques. Au sein de l’ANSES, des contacts dans les services BIOCIDE et REACH sont envisagés. Pour les pesticides, il sera possible d’insérer des données extraites de la base AGRITOX pour notamment pour des paramètres écotoxicologiques. Des possibilités non explorées existent auprès de l’INERIS. Des travaux en cours au Cemagref de Lyon pourraient potentiellement se raccrocher à ce travail. - Ajouts de descripteurs moléculaires. Pour d’autres pesticides ou des contaminants émergents, la phase d’obtention des descripteurs pourra être améliorée en réalisant l’analyse conformationnelle pour localiser le minimum global grâce à l’utilisation de méthodes comme la dynamique moléculaire ou des méthodes de type Monte Carlo. - Travailler sur les liens entre molécules parent et métabolites Il serait intéressant de les prévoir pour par exemple faire apparaître les filiations lors des changements de classes suite à des processus de biodégradation. Ce travail a démarré en 2011 dans le cadre du projet BIODECHLORD « Etudes exploratoires de la dégradation microbienne de la chlordécone » (financement AIP DEMICHLORD et coordination Fabrice Martin (INRA Dijon) où TYPOL sera utilisé pour tester des scénarios de biodégradation de la chlordécone et de cyclodiènes apparentés (approche in silico).

In recent years, metal oxides such as titania have been commercially available as chromatographic beds that can potentially be used to achieve novel separations of polar compounds. For example β blockers, which are more often encountered in environmental sciences, have a wide range of polarity, and their basic character leads to difficult sample treatment and separation on conventional silica-based sorbents. The contribution of titania to the selective analysis of nine β blockers was evaluated in terms of retention mechanisms observed in hydrophilic interaction LC using acetonitrile/water mobile phases with various additives. The mobile phase additives enabled to control the β blocker charge as well as the titania surface charge. Depending on their respective ionic state, various retention mechanisms were identified at low water contents (40%), including mainly adsorption mixed with hydrophilic interaction LC partition, ion exchange and ion exclusion. An unexpected retention was also observed for high water content and high pH, changing the selectivity of the support.
