Developing culturally responsive science teachers requires changes in one's knowledge and dispositions, as well as one's instructional practices. Multicultural science instruction (MSI) provides educators with a framework for strengthening and disrupting the formal curriculum in ways that infuse critical, historical, and culturally responsive connections. Practitioners need not only to see MSI modeled by teacher educators and their colleagues, but also to have concrete resources provided on their journey. While MSI has been defined and researched in academic literature for decades, there is not yet a clear understanding of how it is depicted in authoritative practitioner spaces--such as the National Science Teachers Association (NSTA)--where 40,000 members regularly seek out instructional resources. This potentially limits a major avenue for science teacher education toward inclusive, equitable ends. We conducted a review of 29 articles in the NSTA journals "Science Scope" and "The Science Teacher" to examine the nature and scope of MSI in secondary settings. NSTA publishes nine issues per journal each year, with approximately 15-30 entries per issue. Results indicated, on average, fewer than three MSI-focused articles were published per journal per year for the 10-year period under review. The characterization of MSI within articles was consistent with published scholarship. However, generally inclusive pedagogies dominated, rather than targeted approaches that advance the critical and emancipatory aims of MSI. Articles featuring family collaboration and sociopolitical consciousness represented 2% and 10%, respectively, of all observed MSI. Implications for supporting science teachers' multicultural instruction with authoritative resources are discussed.

ABSTRACT Legionella pneumophila grows within membrane-bound vacuoles in phylogenetically diverse hosts. Intracellular growth requires the function of the Icm/Dot type-IVb secretion system, which translocates more than 300 proteins into host cells. A screen was performed to identify L. pneumophila proteins that stimulate mitogen-activated protein kinase (MAPK) activation, using Icm/Dot translocated proteins ectopically expressed in mammalian cells. In parallel, a second screen was performed to identify L. pneumophila proteins expressed in yeast that cause growth inhibition in MAPK pathway-stimulatory high-osmolarity medium. LegA7 was shared in both screens, a protein predicted to be a member of the bacterial cysteine protease family that has five carboxyl-terminal ankyrin repeats. Three conserved residues in the predicted catalytic triad of LegA7 were mutated. These mutations abolished the ability of LegA7 to inhibit yeast growth. To identify other residues important for LegA7 function, a generalizable selection strategy in yeast was devised to isolate mutants that have lost function and no longer cause growth inhibition on a high-osmolarity medium. Mutations were isolated in the two carboxyl-terminal ankyrin repeats, as well as an inter-domain region located between the cysteine protease domain and the ankyrin repeats. These mutations were predicted by AlphaFold modeling to localize to the face opposite from the catalytic site, arguing that they interfere with the positive regulation of the catalytic activity. Based on our data, we present a model in which LegA7 harbors a cysteine protease domain with an inter-domain and two carboxyl-terminal ankyrin repeat regions that modulate the function of the catalytic domain.IMPORTANCELegionella pneumophila grows in a membrane-bound compartment in macrophages during disease. Construction of the compartment requires a dedicated secretion system that translocates virulence proteins into host cells. One of these proteins, LegA7, is shown to activate a stress response pathway in host cells called the mitogen-activated protein kinase (MAPK) pathway. The effects on the mammalian MAPK pathway were reconstructed in yeast, allowing the development of a strategy to identify the role of individual domains of LegA7. A domain similar to cysteine proteases is demonstrated to be critical for impinging on the MAPK pathway, and the catalytic activity of this domain is required for targeting this path. In addition, a conserved series of repeats, called ankyrin repeats, controls this activity. Data are provided that argue the interaction of the ankyrin repeats with unknown targets probably results in activation of the cysteine protease domain.

Abstract Background The rapid authorization and widespread rollout of COVID-19 vaccines in the United States demonstrated a need for additional data on vaccine side effects, both to provide insight into the range and severity of side effects that might be expected in medically-diverse populations as well as to inform decision-making and combat vaccine hesitancy going forward. Here we report the results of a survey of 4825 individuals from southcentral Kentucky who received two doses of either the Pfizer-BioNTech (BNT162b2) or Moderna (mRNA-1273) vaccine between December 14, 2020 and May 1, 2021. As new versions of the vaccine are rolled-out, local initiatives such as this may offer a means to combat vaccine hesitancy in reference to COVID-19, but are also important as we face new viral threats that will necessitate a rapid vaccine rollout, and to combat a growing public distrust of vaccines in general. Methods Individuals that received two doses of either BNT162b2 or mRNA-1273 between December 14, 2020 and May 1, 2021 were sent a survey, created by the research team. Respondents were asked to rate the incidence and severity of 15 potential side effects and two related outcomes following each of their two doses of the vaccine. All statistical analyses were carried out using SYSTAT, version 13. The data were analyzed utilizing a range of statistical tests, including chi-square tests of association, Cohen’s h, Kruskal-Wallis test one-way nonparametric ANOVA, least-squares regression, and Wilcoxon signed-ranks test. Significance was assessed using Bonferroni-adjusted criteria within families of tests. Results In general, the pattern and severity in side effects was similar to both clinical trial data as well as other published studies. Responses to the mRNA-1273 vaccine were more severe than to BNT162b2, though all were generally in the mild to moderate category. Individuals who reported having previously tested positive for COVID-19 reported stronger responses following the first dose of either vaccine relative to COVID-naïve individuals. The reported severity to the COVID-19 vaccine was positively correlated with self-reported responses to other vaccines. Conclusions Our findings allow broad-scale estimates of the nature and severity of reactions one might expect following vaccination within a clinically-diverse community, and provide a context for addressing vaccine hesitancy in communities such as ours, where locally-generated data and communication may be more influential than national trends and statistics in convincing individuals to become vaccinated. Further, we argue this community-based approach could be important in the future in three key ways: 1) as new boosters and modified vaccines re-volatilize vaccine hesitancy, 2) as new vaccines receive similar testing and rapid authorization, and 3) to combat vaccine hesitancy in other arenas (e.g., annual vaccines, childhood vaccines).

Background: Gut microbiota (GM) modulation has been considered a nutritional approach to manage obesity. Reduced Firmicutes/Bacteroidetes ratio (F/B) is associated with reduced energy harvesting capacity from the diet, ameliorates endotoxemia and inflammation, and restores gut hormone signaling related to hypothalamic control of energy homeostasis. As anti-obesogenic and anti-inflammatory properties have been attributed to Ginkgo biloba extract (GbE), the present study investigated whether GbE supplementation for two weeks modulates the GM composition of obese rats. Method: Fifty-six 2-month-old male Wistar rats were submitted to a lard-rich diet-induced obesity protocol for 60 days (high-fat diet, HFD). Following the obesity-inducing period, rats were gavaged daily with GbE at 500 mg/kg (HFD+G group), or saline (HFD group), for 14 days. A 3rd group (pair-fed group, PF) was performed by mimicking the HFD group (saline administration) but with its food intake matched to the HFD+G group. Rats were euthanized on the 14th supplementation day. Stool DNA was extracted and amplified with V3–V4 region primers of the 16S rRNA gene. Results: In comparison to both HFD and PF groups, GbE supplementation increased the number of Bacteroidetes colon community and concomitantly reduced the abundance of Firmicutes, reducing the F/B ratio. Hierarchical clustering showed that communities of the HFD+G group were less likely related to HFD and PF groups. Conclusion: As GbE modulated the GM structure and diversity in GbE-supplemented obese rats, our results show that GbE possesses phytotherapeutic potential to modulate obesity by improving GM and lessening the consequences of obesity-related GM dysbiosis.

Abstract There remains tremendous interest in developing liquid biopsy assays for detection of cancer‐specific alterations, such as mutations and DNA methylation, in cell‐free DNA (cfDNA) obtained through noninvasive blood draws. However, liquid biopsy analysis is often challenging due to exceedingly low fractions of circulating tumor DNA (ctDNA), necessitating the use of extended tumor biomarker panels. While multiplexed PCR strategies provide advantages such as higher throughput, their implementation is often hindered by challenges such as primer‐dimers and PCR competition. Alternatively, digital PCR (dPCR) approaches generally offer superior performance, but with constrained multiplexing capability. This paper describes development and validation of the first multiplex digital methylation‐specific PCR (mdMSP) platform for simultaneous analysis of four methylation biomarkers for liquid‐biopsy‐based detection of non‐small cell lung cancer (NSCLC). mdMSP employs a microfluidic device containing four independent, but identical modules, housing a total of 40 160 nanowells. Analytical validation of the mdMSP platform demonstrates multiplex detection at analytical specificities as low as 0.0005%. The clinical utility of mdMSP is also demonstrated in a cohort of 72 clinical samples of low‐volume liquid biopsy specimens from patients with computed tomography (CT)‐scan indeterminant pulmonary nodules, exhibiting superior clinical performance when compared to traditional MSP assays for noninvasive detection of early‐stage NSCLC.

This review examines metal enhanced fluorescence (MEF) using gold (Au) nanostructures, with a focus on biomedical applications of this phenomenon, namely fluorescence biosensing and imaging. The mechanism by which MEF occurs will be briefly discussed before a thorough review of recent investigations, particularly surrounding cancer detection and imaging. Work in this field has primarily focused on substrate-based MEF and more recently has explored solution-based enhancement. Therefore, this review will examine both the different nanostructured substrates as well as the colloidal nanomaterials that have been synthesized, along with their interactions with fluorescent molecules. The potential applications of plasmonic probes for optical imaging, particularly in the near-infrared II (NIR-II) window, are also discussed. By incorporating MEF into diagnostic and imaging techniques, it is expected that there will be dramatic improvements to patient survival rates via early diagnosis and efficient treatment.

Under-skin browning (USB) is an unsightly physiological disorder that afflicts ‘Honey Gold’ mango fruit. Under-skin browning symptoms develop after harvest upon the interaction of physical abrasion and physiological chilling stresses. Less understood preharvest and/or harvest factors may also influence fruit susceptibility to USB. In this study, we examined the impact of harvest time during the diurnal cycle and fruit sap components on USB development. Fruits were harvested at 4- to 6-h intervals, lightly abraded with sandpaper to simulate vibration damage during refrigerated road transport, held at 12 ± 1°C for 6 days, transported to the research facilities and ripened before USB assessment. Spurt and ooze sap from the fruit were collected at each harvest time. The samples were separated and analysed by gas chromatography–mass spectrometry. Fruit harvested at 10:00, 14:00 and 18:00 h had 3- to 5-fold higher incidence of USB than did those picked at 22:00, 2:00 and 6:00 h. Sap concentrations of the key aroma volatile compounds 2-carene, 3-carene, α-terpinene, p-cymene, limonene and α-terpinolene were higher for fruit harvested at 14:00 h compared to those picked at other times. In the fruits harvested in the afternoon, abraded skin treated with spurt sap sampled at 14:00 h had 14.3- and 29.0-fold higher incidence and severity, respectively, of induced browning than did those treated with sap collected at 6:00 h. The results showed that fruit harvested in the afternoon were more susceptible to USB than those picked at night or in early morning. The diurnal variation in fruit sensitivity was evidently associated with specific compositional differences in sap phytotoxicity. Topical application to the fruit skin of pure terpinolene and limonene resulted in induced USB damage, whereas pure carene and distilled water did not. Microscopy examination showed that while skin damage caused by pure terpinolene and limonene was not identical to USB per se, similarities suggested that sap components cause USB under inductive commercial conditions. Considered collectively, these findings suggest that night and early morning harvesting will reduce USB and thus improve the postharvest quality of Honey Gold mango fruit.

Global demand for avocados has risen rapidly in recent years, yet supplying fruit that consistently meets consumer expectations for quality remains a challenge in the industry. Body rots in avocado fruit are a leading cause of consumer dissatisfaction. Anecdotal evidence suggests that body rot development may be promoted by mechanical injury at harvest and packing, despite the fruit being hard, green and mature (i.e., unripe) at these stages. Here, ‘Hass’ avocado fruit, harvested across multiple fruiting seasons from commercial orchards, were subjected to controlled impact from drop heights of 15−60 cm at the time of harvest or packing. With increasing drop height, body rot development at eating ripe stage generally occurred more frequently and produced larger lesions at the impact site and, in some experiments, elsewhere on the fruit. These findings refute a general belief that green mature avocado fruit can tolerate a degree of rough physical handling without ripe fruit quality being compromised. Ideally, best avocado harvesting and packing practice should recognize that unripe fruit must not experience drop heights of 30 cm or higher.

Millions of people worldwide currently suffer from serious neurological diseases and injuries for which there are few, and often no, effective treatments. The paucity of effective interventions is, no doubt, due in large part to the complexity of the disorders, as well as our currently limited understanding of their pathophysiology. The bleak picture for patients, however, is also attributable to avoidable impediments stemming from quality concerns in preclinical research that often escape detection by research regulation efforts. In our essay, we connect the dots between these concerns about the quality of preclinical research and their potential ethical impact on the patients who volunteer for early trials of interventions informed by it. We do so in hopes that a greater appreciation among preclinical researchers of these serious ethical consequences can lead to a greater commitment within the research community to adopt widely available tools and measures that can help to improve the quality of research.
