1. Analysis of the Patent of a Protective Cover for Vertical-Axis Wind Turbines (VAWTs): Simulations of Wind Flow

This paper presents a numerical and experimental analysis of the patent of a device to be used in vertical-axis wind turbines (VAWTs) under extreme wind conditions. The device consists of two hemispheres interconnected by a set of conveniently implemented variable section ducts through which the wind circulates to the blades. Furthermore, the design of the cross-section of the ducts allows the control of the wind speed inside the device. These ducts are intended to work as diffusers or nozzles, depending on the needs of the installation site. Simulations were performed for the case of high-speed external wind, for which the ducts act as diffusers to reduce wind speed and maintain a well-functioning internal turbine. Four different patent designs were analyzed, focusing on turbine performance and generated power. The results indicate that the patent allows the generation of electric power for a greater range of wind speeds than with a normal wind turbine. The results support that this patent may be a good alternative for wind power generation in geographic areas with extreme weather conditions or with maintained or strong gusty wind. Experimental tests were carried out on the movement of the blades using the available model. Finally, the power curve of the model of this wind turbine was obtained., Comment: 22 pages, 15 figures

Abstract

Abstract A new method to assess the quality of additive manufacturing (AM) metal powders using the triboelectric charging concept is demonstrated using CpTi, Ti6Al4V, AlSi10Mg, IN 738, and SS 316L powders. For each powder tested, the surface chemical composition was first analyzed using X-ray photoelectron spectroscopy (XPS) to determine the composition of the passivation layer. Some modifications to the current GranuCharge™ setup, developed by GranuTools™, were then performed by incorporating a flow rate measuring tool to assess how tribocharging is affected as a function of flow rate. Variations in the tribocharging response have been found with the flow rate of CpTi, AlSi10Mg and SS 316L powders. Moreover, results suggest that the tribocharging behavior might not be the same even with powders fabricated with the same passivation process. Finally, the compressed exponential model of Trachenko and Zaccone was used to reproduce the tribocharging behavior of the powders. The models were found to work best when the stretch constant β = 1.5, which is identical to the value found in other systems such as structural glasses, colloidal gels, entangled polymers, and supercooled liquids, which experience jamming when motion of individual particles become restricted, causing their motion to slow down.

In this article we consider the following fractional semilinear elliptic equation \[(-\Delta)^su+|x|^2u =\omega u+|u|^{2\sigma}u \quad \text{ in } \mathbb{R}^N,\] where \(s\in (0,1)\), \(N\gt 2s\), \(\sigma\in (0,\frac{2s}{N-2s})\) and \(\omega\in (0, \lambda_1)\). By using variational methods we show the existence of a symmetric decreasing ground state solution of this equation. Moreover, we study some continuity and differentiability properties of the ground state level. Finally, we consider a bifurcation type result.

Abstract

Abstract Smoldering multiple myeloma (SMM) precedes multiple myeloma (MM). The risk of progression of SMM patients is not uniform, thus different progression-risk models have been developed, although they are mainly based on clinical parameters. Recently, genomic predictors of progression have been defined for untreated SMM. However, the usefulness of such markers in the context of clinical trials evaluating upfront treatment in high-risk SMM (HR SMM) has not been explored yet, precluding the identification of baseline genomic alterations leading to drug resistance. For this reason, we carried out next-generation sequencing and fluorescent in-situ hybridization studies on 57 HR and ultra-high risk (UHR) SMM patients treated in the phase II GEM-CESAR clinical trial (NCT02415413). DIS3, FAM46C, and FGFR3 mutations, as well as t(4;14) and 1q alterations, were enriched in HR SMM. TRAF3 mutations were specifically associated with UHR SMM but identified cases with improved outcomes. Importantly, novel potential predictors of treatment resistance were identified: NRAS mutations and the co-occurrence of t(4;14) plus FGFR3 mutations were associated with an increased risk of biological progression. In conclusion, we have carried out for the first time a molecular characterization of HR SMM patients treated with an intensive regimen, identifying genomic predictors of poor outcomes in this setting.

Abstract

Abstract This study examines the electrical properties of isolated brain microtubules (MTs), which are long hollow cylinders assembled from αβ-tubulin dimers that form cytoskeletal structures engaged in several functions. MTs are implicated in sensory functions in cilia and flagella and cellular activities that range from cell motility, vesicular traffic, and neuronal processes to cell division in the centrosomes and centrioles. We determined the electrical properties of the MTs with the loose patch clamp technique in either the presence or absence of the MT stabilizer Paclitaxel. We observed electrical oscillations at different holding potentials that responded accordingly in amplitude and polarity. At zero mV in symmetrical ionic conditions, a single MT radiated an electrical power of 10–17 W. The spectral analysis of the time records disclosed a single fundamental peak at 39 Hz in the Paclitaxel-stabilized MTs. However, a richer oscillatory response and two mean conductances were observed in the non-Paclitaxel MTs. The findings evidence that the brain MTs are electrical oscillators that behave as "ionic-based" transistors to generate, propagate, and amplify electrical signals.

Abstract

The primary cilium is a non-motile sensory organelle that transduces environmental cues into cellular responses. It comprises an axoneme, a core of nine doublet microtubules (MTs) coated by a specialized membrane populated by receptors, and a high density of ion channels. Dysfunctional primary cilia generate the pathogenesis of several diseases known as ciliopathies. However, the electrical role of MTs in ciliary signaling remains largely unknown. Herein, we determined by the patch clamp technique the electrical activity of cytoplasmic and axonemal MTs from wild-type LLC-PK1 renal epithelial cells. We observed electrical oscillations with fundamental frequencies at ∼39 Hz and ∼93 Hz in sheets of cytoplasmic MTs. We also studied in situ and isolated, intact and Triton X-permeabilized primary cilia, observing electrical oscillations with peak frequencies at either 29–49 Hz (non-permeabilized) or ∼40–49 Hz (permeabilized) and ∼93 Hz (both). We applied Empirical Mode Decomposition (EMD), Continuous Wavelet Transform (CWT), and Cross-Correlation Analysis (CCA) to assess the differences and the coherence in the Time-Frequency domains of electrical oscillations between cytoplasmic and axonemal MTs. The data indicate that axonemal and cytoplasmic MTs show different patterns of electrical oscillations preserving coherence at specific frequency peaks that may serve as electromagnetic communication between compartments. Further, the electrical behavior of axonemal MTs was modified by siRNA deletion of polycystin-2 (PC2), which lengthens primary cilia, thus linking ciliary channels to the morphological and electrical behavior of cilia in ciliopathies. The encompassed evidence indicates that the primary cilium behaves as an electrical antenna, with an excitable MT structure that produces electrical oscillations whose synchronization and propagation constitute a novel cell signaling mechanism.

Abstract

Abstract Background Well-functioning competitive markets are key to controlling generic drug prices. This is important since over 90% of all drugs sold in the US are generics. Recently, there have been examples of large price increases in the generic market. Methods This paper examines price trajectories for generic drugs using a group-based trajectory modelling approach (GBTM). We fit the model using quarterly price information in the IBM MarketScan claims database for the past decade. Results We identify three dominant price trajectories for this period: rapid increase trajectories, slow decline and rapid decline. Most generic drugs show a slow or a rapid decline in price trajectories. However, around 17% of all generic drugs show rapid price increase trajectories. Conclusions As Congress is exploring an excise tax on drugs whose list price increases faster than the rate of inflation, we discuss what drugs would be most likely to be affected by this law.

Abstract

(1) Background: The entomopathogenic fungus Metarhizium anisopliae sensu lato forms a species complex, comprising a tight cluster made up of four species, namely M. anisopliae sensu stricto, M. pinghaense, M. robertsii and M. brunneum. Unambiguous species delineation within this “PARB clade” that enables both the taxonomic assignment of new isolates and the identification of potentially new species is highly solicited. (2) Methods: Species-discriminating primer pairs targeting the ribosomal intergenic spacer (rIGS) sequence were designed and a diagnostic PCR protocol established. A partial rIGS sequence, referred to as rIGS-ID800, was introduced as a molecular taxonomic marker for PARB species delineation. (3) Results: PARB species from a validation strain set not implied in primer design were clearly discriminated using the diagnostic PCR protocol developed. Using rIGS-ID800 as a single sequence taxonomic marker gave rise to a higher resolution and statistically better supported delineation of PARB clade species. (4) Conclusions: Reliable species discrimination within the Metarhizium PARB clade is possible through both sequencing-independent diagnostic PCR and sequencing-dependent single marker comparison, both based on the rIGS marker.

15. Identification of intima-to-media signals for flow-induced vascular remodeling using correlative gene expression analysis

Abstract

Abstract Changes in blood flow can induce arterial remodeling. Intimal cells sense flow and send signals to the media to initiate remodeling. However, the nature of such intima-media signaling is not fully understood. To identify potential signals, New Zealand white rabbits underwent bilateral carotid ligation to increase flow in the basilar artery or sham surgery (n = 2 ligated, n = 2 sham). Flow was measured by transcranial Doppler ultrasonography, vessel geometry was determined by 3D angiography, and hemodynamics were quantified by computational fluid dynamics. 24 h post-surgery, the basilar artery and terminus were embedded for sectioning. Intima and media were separately microdissected from the sections, and whole transcriptomes were obtained by RNA-seq. Correlation analysis of expression across all possible intima-media gene pairs revealed potential remodeling signals. Carotid ligation increased flow in the basilar artery and terminus and caused differential expression of 194 intimal genes and 529 medial genes. 29,777 intima-media gene pairs exhibited correlated expression. 18 intimal genes had > 200 medial correlates and coded for extracellular products. Gene ontology of the medial correlates showed enrichment of organonitrogen metabolism, leukocyte activation/immune response, and secretion/exocytosis processes. This demonstrates correlative expression analysis of intimal and medial genes can reveal novel signals that may regulate flow-induced arterial remodeling.

Abstract

Abstract Background Pediatric emergency medicine training is in its infancy in India. Simulation provides an educational avenue to equip trainees with the skills to improve pediatric care. We hypothesized that a simulation-based curriculum can improve Indian post-graduate emergency medicine (EM) trainees’ self-efficacy, knowledge, and skills in pediatric care. Methods We designed a simulation-based curriculum for management of common pediatric emergencies including sepsis, trauma, and respiratory illness and pediatric-specific procedures including vascular access and airway skills. Training included didactics, procedural skill stations, and simulation. Measures included a self-efficacy survey, knowledge test, skills checklist, and follow-up survey. Results were analyzed using the Wilcoxon signed-rank test and paired-samples t test. A 6-month follow-up survey was done to evaluate lasting effects of the intervention. Results Seventy residents from four academic hospitals in India participated. Trainees reported feeling significantly more confident, after training, in performing procedures, and managing pediatric emergencies (p < 0.001). After the simulation-based curriculum, trainees demonstrated an increase in medical knowledge of 19% (p < 0.01) and improvement in procedural skills from baseline to mastery of 18%, 20%, 16%, and 19% for intubation, bag-valve mask ventilation, intravenous access, and intraosseous access respectively (p < 0.01). At 6-month follow-up, self-efficacy in procedural skills and management of pediatric emergencies improved from baseline. Conclusions A simulation-based curriculum is an effective and sustainable way to improve Indian post-graduate EM trainees’ self-efficacy, knowledge, and skills in pediatric emergency care.

Abstract

Meningeal sporotrichosis is rare and occurs predominantly in immunosuppressed individuals. This retrospective study explored clinical and laboratory characteristics, treatment, and prognosis of patients with disseminated sporotrichosis who underwent lumbar puncture (LP) at a Brazilian reference center from 1999 to 2020. Kaplan–Meier and Cox regression models were used to estimate overall survival and hazard ratios. Among 57 enrolled patients, 17 had meningitis. Fifteen (88.2%) had HIV infection, and in 6 of them, neurological manifestations occurred because of the immune reconstitution inflammatory syndrome (IRIS). The most frequent symptom was headache (88.2%). Meningeal symptoms at first LP were absent in 7/17 (41.2%) patients. Sporothrix was diagnosed in cerebrospinal fluid either by culture or by polymerase chain reaction in seven and four patients, respectively. All but one patient received prolonged courses of amphotericin B formulations, and seven received posaconazole, but relapses were frequent. Lethality among patients with meningitis was 64.7%, with a higher chance of death compared to those without meningitis (HR = 3.87; IC95% = 1.23;12.17). Meningeal sporotrichosis occurs mostly in people with HIV and can be associated with IRIS. Screening LP is indicated in patients with disseminated disease despite the absence of neurological complaints. Meningitis is associated with poor prognosis, and better treatment strategies are needed.

Abstract

Among pharmaceuticals, cytostatics are a category of emerging contaminants that are of particular environmental concern, because of their mutagenic and carcinogenic characteristics. Concern about the presence of these drugs in the environment has increased in the last decades because of their potential harm to aquatic organisms and human health and their long-term cumulative impact on the environment, even at low concentrations in the order of ppb to ppt (ixg/L to ng/L). In this paper we provide of an overview of the use of ozone and (or) advanced oxidation processes to eliminate cytostatics from the pharmaceutical wastewaters focusing on studies undertaken in Cuba. The review revealed that these treatments are suitable to degrade several antineoplastic drugs of different chemical structures and to safety dispose of wastewaters to the environment. In addition, we describe the analytical methods used for the determination of the fundamental by-products of applied treatments. Les preoccupations concernant la presence de ces medicaments dans l'environnement ont augmente au cours des dernieres decennies en raison de leurs effets nocifs potentiels sur les organismes aquatiques, la sante humaine et leurs effets cumulatifs a long terme sur l'environnement, meme a de faibles concentrations de l'ordre de ppb a ppt (ixg/L a ng/L). Dans cette revue, nous presentons un apercu de l'utilisation de l'ozone et (ou) des procedes d'oxydation avancee pour eliminer les cytostatiques des eaux usees ayant des traces de produits pharmaceutiques en nous concentrant sur les etudes entreprises a Cuba. Abstract

Microtubules (MTs) are important structures of the cytoskeleton in neurons. Mammalian brain MTs act as biomolecular transistors that generate highly synchronous electrical oscillations. However, their role in brain function is largely unknown. To gain insight into the MT electrical oscillatory activity of the brain, we turned to the honeybee (Apis mellifera) as a useful model to isolate brains and MTs. The patch clamp technique was applied to MT sheets of purified honeybee brain MTs. High resistance seal patches showed electrical oscillations that linearly depended on the holding potential between ± 200 mV and had an average conductance in the order of ~9 nS. To place these oscillations in the context of the brain, we also explored local field potential (LFP) recordings from the Triton X-permeabilized whole honeybee brain unmasking spontaneous oscillations after but not before tissue permeabilization. Frequency domain spectral analysis of time records indicated at least two major peaks at approximately ~38 Hz and ~93 Hz in both preparations. The present data provide evidence that MT electrical oscillations are a novel signaling mechanism implicated in brain wave activity observed in the insect brain.

Abstract

Rapid eye movement (REM) sleep is a sleep phase characterised by random eye movements for which the underlying motor commands are yet to be revealed. The authors describe that a cluster of medulla oblongata neurons in the Nucleus papiliocontributes to the control of eye movements during REM sleep.

Abstract

The objective of this work is to further update the records of Argentinean Entomophthoralean fungal species and their geographical distribution. Our survey includes five new records of Zoophthora radicans infecting insect hosts belonging to the orders Diptera, Hemiptera, and Lepidoptera from Pampasic and Littoral regions of Argentina. These new records not only increase the host range of Entomophthoralean fungi but also their geographical distribution around the world.

25. Implementation of dietary methionine restriction using casein after selective, oxidative deletion of methionine

Abstract

Summary: Dietary methionine restriction (MR) is normally implemented using diets formulated from elemental amino acids (AA) that reduce methionine content to ∼0.17%. However, translational implementation of MR with elemental AA-based diets is intractable due to poor palatability. To solve this problem and restrict methionine using intact proteins, casein was subjected to mild oxidation to selectively reduce methionine. Diets were then formulated using oxidized casein, adding back methionine to produce a final concentration of 0.17%. The biological efficacy of dietary MR using the oxidized casein (Ox Cas) diet was compared with the standard elemental MR diet in terms of the behavioral, metabolic, endocrine, and transcriptional responses to the four diets. The Ox Cas MR diet faithfully reproduced the expected physiological, biochemical, and transcriptional responses in liver and inguinal white adipose tissue. Collectively, these findings demonstrate that dietary MR can be effectively implemented using casein after selective oxidative reduction of methionine.

28. Catálogo de la colección de cultivos de especies de hongos patógenos y simbiontes de insectos y otros artrópodos de la Argentina

Abstract

El presente trabajo, tiene como objetivo dar a conocer los datos actualizados e información relevante de las cepas de especies fúngicas entomopatógenas preservadas en la colección de cultivos del CEPAVE (Centro de Estudios Parasitológicos y de Vectores) CONICET-UNLP, Argentina. Los datos que se informan en este trabajo son de aislamientos nativos de diferentes regiones fitogeográficas de la Argentina. Para cada cepa se especifica: acrónimo, sustrato, especie de insecto hospedante, fecha de recolección, referencias de coordenadas geográficas y medio de cultivo en el que fue aislado. Los fines de esta Colección son de investigación, docencia y vinculación tecnológica a empresas. Su inicio fue en 1988 y, actualmente se encuentra registrada en la FELACC (Federación Latinoamericana de Colecciones de Cultivos) como Socio Institucional: Nº SI-06 y en la Federación Mundial de Colecciones de Cultivos (World Federation for Culture Collections – WFCC) (http://www.wfcc.info/). Siguiendo las pautas de organización de la WFCC esta colección está incorporada a: World Data Centre for Microorganisms (WDCM, Nº 973 CEP). Los métodos de preservación que se utilizan son: agua destilada estéril, papel, aceite mineral, sílica gel, freezer -20 °C y -70 °C y liofilización, siendo realizados rigurosos controles de calidad y de viabilidad de las cepas. Las cepas son caracterizadas por técnicas morfológicas y el 30 % de los aislamientos fúngicos han sido caracterizados, además, mediante técnicas de biología molecular. En la actualidad, la colección mantiene más de 650 cepas correspondientes en su mayoría a los géneros Beauveria, Metarhizium, Isaria, Lecanicillium y algunas especies de Entomophthorales.

Abstract

La hemorragia alveolar difusa es un hallazgo clínico patológico, consecuencia de una lesión en la microcirculación pulmonar que se presenta con sintomatología inespecífica e incluso en pacientes asintomáticos. Se diferencian tres diagnósticos histopatológicos, siendo la capilaritis el más común y secundario a patologías autoinmunes. Se presenta el caso de un paciente, con hallazgo imagenológico y en lavado broncoalveolar, compatible con hemorragia alveolar difusa en quien se descartó como causa, enfermedades autoinmunes e infecciosas; durante la anamnesis, se identificó inicio reciente de montelukast, con relación temporal, respuesta a suspensión, cambio imagenológico y escalas de probabilidad de reacción adversa de Naranjo que apoya el diagnóstico.

Abstract

Background: The Global Anticoagulant Registry in the FIELD–Atrial Fibrillation (GARFIELD-AF) is an ongoing prospective noninterventional registry, which is providing important information on the baseline characteristics, treatment patterns, and 1-year outcomes in patients with newly diagnosed non-valvular atrial fibrillation (NVAF). This report describes data from Indian patients recruited in this registry. Methods and results: A total of 52,014 patients with newly diagnosed AF were enrolled globally; of these, 1388 patients were recruited from 26 sites within India (2012–2016). Abstract

Background: Data on the benefits of rapid microbiological testing on antimicrobial consumption (AC) and antimicrobial resistance patterns (ARPs) are scarce. We evaluated the impact of a protocol based on rapid techniques on AC and ARP in intensive care (ICU) patients. Methods: A retrospective pre- (2018) and post-intervention (2019–2021) study was conducted in ICU patients. A rapid diagnostic algorithm was applied starting in 2019 in patients with a lower respiratory tract infection. The incidence of nosocomial infections, ARPs, and AC as DDDs (defined daily doses) were monitored. Results: A total of 3635 patients were included: 987 in the pre-intervention group and 2648 in the post-intervention group. The median age was 60 years, the sample was 64% male, and the average APACHE II and SOFA scores were 19 points and 3 points. The overall ICU mortality was 17.2% without any differences between the groups. An increase in the number of infections was observed in the post-intervention group (44.5% vs. 17.9%, p < 0.01), especially due to an increase in the incidence of ventilator-associated pneumonia (44.6% vs. 25%, p < 0.001). AC decreased from 128.7 DDD in 2018 to 66.0 DDD in 2021 (rate ratio = 0.51). An increase in Pseudomonas aeruginosa susceptibility of 23% for Piperacillin/tazobactam and 31% for Meropenem was observed. Conclusion: The implementation of an algorithm based on rapid microbiological diagnostic techniques allowed for a significant reduction in AC and ARPs without affecting the prognosis of critically ill patients.

38. Parents as Study Buddies: The Role of the Parents in the Implementation of Modular Learning Program

Abstract

This descriptive phenomenological study explored the role of parents as study buddies of their children in implementing the modular learning program. An in-depth interview was used to gather the lived experiences of 16 parents purposely chosen to participate in the study. The parents included are either male or female, with at least two children studying in public schools and have not gone to school or completed elementary level. Their personal narratives about their experiences, their coping mechanisms to address the struggles encountered, their learnings, and insights from their experiences as a study guide of their children in the modular learning were gathered using a semi-structured interview guide. The data collected were analyzed using Colaizzi's method for data analysis. Findings reveal that parents' learning experiences are varied; they are glad to engage in their children's learning and can learn while assisting them with their module activities. Parents face challenges in comprehension modules, time management, and parental literacy, but they overcome them by seeking assistance from relatives, using the internet, and praying for wisdom. Despite their challenges, they understand the value of education for their children's future and realize that education is the only weapon that can be used to face life's challenges.

Abstract

Purpose:Undetectable measurable residual disease (MRD) is a surrogate of prolonged survival in multiple myeloma. Thus, treatment individualization based on the probability of a patient achieving undetectable MRD with a singular regimen could represent a new concept toward personalized treatment, with fast assessment of its success. This has never been investigated; therefore, we sought to define a machine learning model to predict undetectable MRD at the onset of multiple myeloma.Experimental Design:This study included 487 newly diagnosed patients with multiple myeloma. The training (n = 152) and internal validation cohorts (n = 149) consisted of 301 transplant-eligible patients with active multiple myeloma enrolled in the GEM2012MENOS65 trial. Two external validation cohorts were defined by 76 high-risk transplant-eligible patients with smoldering multiple myeloma enrolled in the Grupo Español de Mieloma(GEM)-CESAR trial, and 110 transplant-ineligible elderly patients enrolled in the GEM-CLARIDEX trial.Results:The most effective model to predict MRD status resulted from integrating cytogenetic [t(4;14) and/or del(17p13)], tumor burden (bone marrow plasma cell clonality and circulating tumor cells), and immune-related biomarkers. Accurate predictions of MRD outcomes were achieved in 71% of cases in the GEM2012MENOS65 trial (n = 214/301) and 72% in the external validation cohorts (n = 134/186). The model also predicted sustained MRD negativity from consolidation onto 2 years maintenance (GEM2014MAIN). High-confidence prediction of undetectable MRD at diagnosis identified a subgroup of patients with active multiple myeloma with 80% and 93% progression-free and overall survival rates at 5 years.Conclusions:It is possible to accurately predict MRD outcomes using an integrative, weighted model defined by machine learning algorithms. Abstract

In the U.S., Mexican culture has been characterized as inherently traditional in its gender values. However, we aimed to highlight the heterogeneity of Mexican culture within the U.S. by testing whether Mexican-heritage women who reject traditional gender values necessarily disavow other cultural connections (i.e., practices, identity).Mexican-heritage young women (Latent profile analyses revealed five profiles based on cultural practices (either high [immersed] or moderate [participating] levels) and gender values (either traditional, moderate, or nontraditional): (a) immersed/nontraditional, (b) immersed/moderate, (c) immersed/traditional, (d) participating/nontraditional, and (e) participating/moderate. Regardless of gender values, the immersed profiles were generally stronger in Mexican ethnic identity than the participating profiles.Contrary to some prior views linking Mexican cultural practices with traditional gender values, many Mexican-heritage women rejected traditional gender values while maintaining strong immersion in cultural practices and strong ethnic identities. The aim of this paper is to study the structure of irreducible modules in the variety ℳ of commutative power-associative nilalgebras of nilindex ≤ 4. If A ∈ ℳ with dimension at most 5, then we prove that A 2 is contained in the annihilator of every irreducible A -module in the variety ℳ. Also, we consider the enveloping algebra of an algebra A in the variety ℳ and we obtain a new example of a commutative power-associative non-nilpotent nilalgebra of dimension 9. Abstract

The quantitative investigation of the scalar Bethe–Salpeter equation in Minkowski space, within the ladder-approximation framework, is extended to include the excited states. This study has been carried out for an interacting system composed by two massive bosons exchanging a massive scalar, by adopting (i) the Nakanishi integral representation of the Bethe–Salpeter amplitude, and (ii) the formally exact projection onto the null plane. Our analysis, on one hand, confirms the reliability of the method already applied to the ground state and, on the other one, extends the investigation from the valence distribution in momentum space to the corresponding quantity in the impact-parameter space, pointing out some relevant features, like (i) the equivalence between Minkowski and Euclidean transverse-momentum amplitudes, and (ii) the leading exponential fall-off of the valence wave function in the impact-parameter space.

Abstract

Natural sequence variation within mitochondrial DNA (mtDNA) contributes to human phenotypes and may serve as natural genetic markers in human cells for clonal and lineage tracing. We recently developed a single-cell multi-omic approach, called ‘mitochondrial single-cell assay for transposase-accessible chromatin with sequencing’ (mtscATAC-seq), enabling concomitant high-throughput mtDNA genotyping and accessible chromatin profiling. Specifically, our technique allows the mitochondrial genome-wide inference of mtDNA variant heteroplasmy along with information on cell state and accessible chromatin variation in individual cells. Leveraging somatic mtDNA mutations, our method further enables inference of clonal relationships among native ex vivo-derived human cells not amenable to genetic engineering-based clonal tracing approaches. Here, we provide a step-by-step protocol for the use of mtscATAC-seq, including various cell-processing and flow cytometry workflows, by using primary hematopoietic cells, subsequent single-cell genomic library preparation and sequencing that collectively take ~3–4 days to complete. We discuss experimental and computational data quality control metrics and considerations for the extension to other mammalian tissues. Overall, mtscATAC-seq provides a broadly applicable platform to map clonal relationships between cells in human tissues, investigate fundamental aspects of mitochondrial genetics and enable additional modes of multi-omic discovery.
