The purpose of this study is to establish the rapid discrimination system of cybrid callus lines by Fourier transform infrared (FT-IR) spectroscopy without genetic fingerprinting analysis. Genomic DNA isolated from two parental lines (Brassica oleracea var. capitata and Raphanus sativus L.) and their cybrid callus lines were analyzed by FT-IR spectroscopy in the spectral region from 4000 to 400 cm−1. Several spectral differences between the two parental lines were detected in the frequency regions of N–H stretching (amide I), C=O stretching vibrations (amide II), and PO2− ionized asymmetric and symmetric stretching. Principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA) were used to discriminate cybrid cell lines from the two parental species at the genomic DNA level. PLS-DA analysis provided more clear discrimination between the two parental lines and their progeny cybrid lines in the score plot. PCA loading values also showed that obvious spectral differences played a significant role in discrimination between the two parental lines and their cybrid lines. These spectral differences might be directly related to subtle changes in the base functional groups and backbone structures of genomic DNA. Considering these results, this technique could provide a research foundation for the FT-IR spectral-based diagnosis, selection, and discrimination of parental lines and their cybrids. Furthermore, this technique could be applied in the hybrid seed industry for rapid screening with high heterogeneity.

This study aimed to establish a rapid system for discriminating the cultivation origins and cultivars of dry persimmons, using metabolite fingerprinting by Fourier transform infrared (FT-IR) spectroscopy combined with multivariate analysis. Whole-cell extracts from the sepals of four Korean cultivars and two different Chinese dry persimmons were subjected to FT-IR spectroscopy. Principle component analysis (PCA) and partial least squares discriminant analysis (PLS-DA) of the FT-IR spectral data successfully discriminated six dry persimmons into two groups depending on their cultivation origins. To determine whether or not FT-IR spectroscopy could be used for taxonomic and metabolic discrimination of African yam lines, tuber samples from African and Asian yam species were subjected to FT-IR. Most remarkable spectral differences between African and Asian yams were found in the 1750-1700 cm(-1) region, polysaccharide (1200-900 cm(-1)) and protein/amide I and II (1700-1500 cm(-1)) regions of FT-IR spectra. A hierarchical dendrogram based on partial least square-discriminant analysis (PLS-DA) of FT-IR data from 7 African yam species show phylogenetic relationship. In addition, the content of dioscin, a steroidal saponin found in yam tuber, was predicted using a PLS regression model with regression coefficient R(2)=0.7208 indicated that prediction model had average accuracy. Thus, considering these results we suggest that FT-IR combined with multivariate analysis could be applied as a novel tool for metabolic evaluation and high-throughput screening of African yam lines with higher content of dioscin.

To determine whether Fourier transform (FT)-IR spectral analysis combined with multivariate analysis of whole-cell extracts from ginseng leaves can be applied as a high-throughput discrimination system of cultivation ages and cultivars, a total of total 480 leaf samples belonging to 12 categories corresponding to four different cultivars (Yunpung, Kumpung, Chunpung, and an open-pollinated variety) and three different cultivation ages (1 yr, 2 yr, and 3 yr) were subjected to FT-IR. The spectral data were analyzed by principal component analysis and partial least squares-discriminant analysis. A dendrogram based on hierarchical clustering analysis of the FT-IR spectral data on ginseng leaves showed that leaf samples were initially segregated into three groups in a cultivation age-dependent manner. Then, within the same cultivation age group, leaf samples were clustered into four subgroups in a cultivar-dependent manner. The overall prediction accuracy for discrimination of cultivars and cultivation ages was 94.8% in a cross-validation test. These results clearly show that the FT-IR spectra combined with multivariate analysis from ginseng leaves can be applied as an alternative tool for discriminating of ginseng cultivars and cultivation ages. Therefore, we suggest that this result could be used as a rapid and reliable F1 hybrid seed-screening tool for accelerating the conventional breeding of ginseng.

Fourier transform infrared spectroscopy (FT-IR) spectroscopy in combination with multivariate analysis was used to discriminate two different F1 hybrid lines from their parental inbred lines. Genomic DNA was isolated from leaves of three parental lines and two F1 hybrid lines of Brassica campestris subsp. pekinensis. Purified genomic DNA was analyzed by FT-IR spectroscopy in the spectral region from 4,000 to 400 cm−1. FT-IR spectra confirmed typical spectral differences between the frequency regions of N–H stretching (amide I) and C=O stretching vibrations (amide II) as well as PO2− ionized asymmetric and symmetric stretching. Principal component analysis was able to discriminate between F1 hybrid progenies depending on their parental lineages, even though they share the same maternal background. Partial least squares discriminant analysis gave a more clear discrimination between the two parental lines and their hybrid progenies. These FT-IR spectral differences might be directly related to subtle changes in the base functional group and backbone structures of genomic DNA. Considering these results, this technique could provide a solid research foundation for FT-IR spectral-based rapid diagnosis, selection, and discrimination of parental lines from their progenies. Furthermore, this technique could be applied to test purity in the hybrid seed industry.

This study reports high-frequency plant regeneration from immature zygotic embryo cultures of Houttuynia cordata Thunb via somatic embryogenesis. Numerous green globular structures were directly formed on the surfaces of cotyledons and radicles from 2-week-old immature zygotic embryos at a frequency of 42.1 % when cultured on Murashige and Skoog (MS) medium supplemented with 2 mg l−1 of α-naphthaleneacetic acid (NAA) and 1 mg l−1 of 6-benzyladenine (BA). In comparison, white globular structures and pale-yellow calluses were formed simultaneously at a frequency of 28.3 % when cultured on MS medium supplemented with 2,4-dichlorophenoxyacetic acid (2,4-D). The pale-yellow calluses were transferred to MS liquid medium supplemented with 2,4-D to establish embryogenic cell suspension cultures consisting of round, isodiametric cells that formed cell aggregates. Upon plating of these cell aggregates on half-strength MS medium without growth regulators under light conditions, cell aggregates gave rise to numerous globular embryos at a frequency of 56 %. Of the globular embryos, 15 % were successfully converted into cotyledonary embryos when cultured on half-strength MS medium under light conditions. The plant regeneration system of H. cordata established in this study will be useful for the selection, genetic transformation, and mass proliferation of elite clones with medicinal potential.

본 실험에서는 상추 연산홍에 apoptosis 관련 유전자인 Bcl-2 유전자를 도입하여 내병성 상추의 육성을 하기 위한 목적으로 형질전환을 수행하였다. 상추의 자엽조직을 NPTII-35S Promoter와 Bcl-2 유전자가 삽입된 Agrobacterium GV 3101과 공동배양 한 후 0.1 mg/L NAA, 0.5 mg/L BAP, 100 mg/L Kanamycin, 300 mg/L Lilacillin이 첨가된 MS 배지에서 식물체가 유기되었다. Kanamycin 내성을 가진 식물체들을 PCR, Southern blot 분석을 통해 Bcl-2 유전자가 안정적으로 식물체 genome 안에 삽입되었음을 확인하였다. 100개의 형질전환식물체가 확인되었으며 T1식물체를 채종하였다. T1 종자를 파종하여 Sclerotinia sclerotiorum. 균주를 접종하여 내병성 검정을 실시하였고 그 중 2개의 line에서 내병성을 확인하였다. 이러한 결과를 통하여 인간의 apoptosis에 관련하는 유전자가 식물체 내에서 안정된 발현을 통하여 내병성을 증가시켰음을 밝혔다. 【Transgenic lettuce plants were successfully obtained from hypocotyl explants inoculated with Agrobacterium tumefaciens, which harbored a binary vector plasmid with Bcl-2 gene, related to apoptosis. After culture and selection on MS medium a number of kanamycin-resistant plantlets were regenerated. Polymerase chain reaction, Southern blot analysis and Northern blot analysis were used to identify and characterize the transgenic plants with the integrated Bcl-2 gene. Over 100 transgenic plants have been established in soil and flowered in the greenhouse. T1 progeny of 100 transgenic lettuce inbred lines were inoculated with Sclerotinia sclerotiorum. Expression of the Bcl-2 peptide in transgenic lettuce plants provides high levels of field resistance against Sclerotinia sclerotiorum, causal agent of the agronomically important fungal disease of lettuce.】

Dihydroflavonol 4-reductase (DFR) is a key enzyme of the flavonoid biosynthesis pathway which catalyses the NADPH-dependent reduction of 2R,3R-trans-dihydroflavonols to leucoanthocyanidins. In this study we describe cloning and expression of the genes encoding the flavonoid-bio- synthetic enzyme DFR in Gypsophila paniculata L. Inspec- tion of the 1279 bp long sequence revealed an open reading frame 1063 bp, including a 36 bp 5' leader region and 181 bp 3' untranslated region. Comparison of the coding region of this DFR cDNA sequence including the sequences of Arabidopsis thaliana, Citrus sinensis, Dianthus caryophyllus, Ipomoea batatas, Matthiola incana, Nierembergia sp, Pe- tunia hybrida, Solanum tuberosum, Vitis vinifera reveals an identity higher than 69% at the nucleotide level. The func- tion of this nucleotide sequences was verified by comparison with amino acid sequences of the amino-terminus and tryptic peptides from purified plant enzyme, by northern blotting with mRNA from wild type and mutant plants, by in vitro expression yielding and enzymatically active reductase, as indicated by the small leucopelargonidin peak. 【This study aimed to develop carnation cultivars with new coloring system. We used four genes of Petunia hybrida - chalcone synthase (CHS), flavanone 3-hydroxylase (FHT), dihydroflavonol 4-reductase (DFR), and anthocyanidin synthase (ANS) - as probes, in order to isolate four genes from carnations (Dianthus Caryophyllus). The isolated genes were used as probes in order to select mutants out of collected carnations, using Northern blot analysis. The Northern blot analysis revealed 10 DFR mutants - Gumbyul, Eunbyul, Ballatyne, Crystal, Eugenia, Koreno, Imp. White Sim, West Crystal, White Alpine, and White Charotte. Six among the selected 10 cultivarswere excluded from the target cultivars, because Eugenia, Imp. White Sim, and White Alpine were proved to be double mutants of DFR and ANS, Koreno was considered to be a double mutant of DFR and CHS, and Gumbyul and Ballatyne were proved to be double mutants of DFR and CHI (Chalcone isomerase). Consequently, we selected five DFR mutants, including Virginie, which was already selected as a DFR mutant. Finally, we measured DFR activities in order to confirm the selection, and the results showed that all of the five cultivars - Eunbyul, Crystal, West Crystal, White Charotte, and Virginie - had got no DFR activity.】

Protoplasts from cabbage and radish were isolated and fused symmetrically by PEG treatment. The PEG treated mixture of high concentrated protoplasts produced lots of micro-calli after weeks. The microcalli developed to normal calli and shoots were regenerated from the calli. A total of 218 shoots were regenerated, but none of them contained the NWB-CMS specific DNA marker, indicating that the transfer of the radish NWB-CMS character into cabbage did not occur. However, ISSR analysis revealed that the cell fusion between protoplasts from radish and cabbage was occurred (3 out of 208 plantlet). The fused regenerants possessed the characteristics of source plants used for protoplast fusion. After vernalization, three regenerants were flowered with white petal color as seen in radish. Only three seeds were able to obtain from one regenerant by backcrossing with the cabbage pollen.

L-Ascorbic acid (vitamin C) in vegetables is an essential component of human nutrition. The objective is to transform lettuce (Lactuca sativa L.) with GalUR gene that is involved in the vitamin C biosynthesis. The cotyledons of Hwoahong (Nongwoo Bio Co.) were used to induce the callus and shoot under the selection media with MS + 30 g/L Sucrose + 0.5 mg/L BAP + 0.1 mg/L NAA + 100 mg/L kanamycin + 200 mg/L lilacillin, pH 5.2. The shoot was developed from the cut side of the explants after 3 weeks on the selection media. We successfully transformed the lettuce with GaIUR gene and analyzed the levels of vitamin C. We found that some of the lettuce transgenic lines contained higher levels of vitamin C compared with the normal one (non-transformed). Especially, some of lettuces inserted by GalUR showed about times higher content of vitamin C compared to the non-transformed lettuce. This data support the previously work performed with GLOase transgenic lettuces from which several times higher content of vitamin C were identified. The lettuces with high content of vitamin C have been selected for further analysis.

A mannose selection system was adapted for use in the Agrobacterium-mediated transformation of Chinese cabbage. This system makes use of the pmi gene that encodes phosphomannose isomerase, which converts mannose-6-phosphate to fructose-6-phosphate. Hypocotyl explants from 4-5-day-old seedlings of Chinese cabbage inbred lines were pre-cultured for 2-3 days and then infected with Agrobacterium. Two genes (L: -guluno-gamma-lactone oxidase, GLOase, and jasmonic methyl transferase, JMT) were transformed into Chinese cabbage using the transformation procedure developed in this study. We found that supplementing the media with 7 g l(-1) mannose and 2% sucrose provides the necessary conditions for the selection of transformed plants from nontransformed plants. The transformation rates were 1.4% for GLOase and 3.0% for JMT, respectively. The Southern blot analysis revealed that several independent transformants (T (0)) were obtained from each transgene. Three different inbred lines were transformed, and most of the T (1) plants had normal phenotypes. The transformation method presented here for Chinese cabbage using mannose selection is efficient and reproducible, and it can be useful to introduce a desirable gene(s) into commercially useful inbred lines of Chinese cabbage.

Flavanone 3-hydroxylase (FHT) is an enzyme acting in the central part of the flavonoid biosynthesis pathway. FHT catalyses the hydroxylation of flavanone to dihydroflavonols in the anthocyanin pathway. In this paper we describe the cloning and expression of the genes encoding the flavonoid-biosynthetic enzyme FHT in Gypsophila paniculata L. A heterologous cDHA probe from Dianthus cavophyllus was used to isolate FHT-encoding cDHA clones from Gypsophila paniculata L.. Inspection of the 1471 bp long sequence revealed an open reading frame 1047 bp, including a 190 bp 5` leader region and 288 bp 3` untranslated region. Comparison of the coding region of this FHT cDHA sequence including the sequences of Arabidopsis thaliana, Citrus sinensis, Dianthus caryophyllus, Ipomoea batatas, Matthiola incana, Nierembergia sp, Petunia hybrida, Solanum tuberosum, Vitis vinifera reveals a identity higher than 69% at the nucleotide level. The function of this nucleotide sequences were verified by comparison with amino acid sequences of the amino-terminus and tryptic peptides from purified plant enzyme, by northern blotting with mRHA from wild type and mutant plants, by in vitro expression yielding and enzymatically active hydroxylase, as indicated by the small dihydrokaempferol peak. We have isolated a full-length cDNA, PPI1 (pepper-PMMV interaction 1), encoding a novel basic region-leucine zipper (bZIP) DNA-binding protein, from expressed sequence tags differentially expressed in Capsicum chinense PI257284 infected with Pepper mild mottle virus (PMMV). PPI1 encodes a predicted protein of 170 amino acids and contains a putative DNA-binding domain that shares significant amino acid identity with ACGT-binding domains of members of the bZIP DNA-binding protein family. PPI1 was localized in the nucleus and had transcriptional activation activity in yeast. Transcripts of the PPI1 gene were preferentially induced during an incompatible interaction by inoculation with PMMV, Pseudomonas syringae pv. syringae 61, and Xanthomonas campestris pv. vesicatoria race 3. However, the PPI1 gene was not induced by abiotic stressors that activate the plant defense-signaling pathway. Our data provide the first evidence that a bZIP transcription factor is preferentially induced by pathogen attack, suggesting that PPI1 may play a specific functional role in the regulation of expression of plant defense-related genes.

This study aims to establish the discrimination of F1 hybrid seeds from their parental lines and rapid selection of proteinrich lines from inbreeding lines of corn using FT-IR spectroscopy combined by multivariate analysis. Eight individual seeds from the maternal and paternal lines of Gwangpyeongok and their F1 progeny seeds were subjected to FT-IR spectroscopy. A total of 176 corn inbreeding lines including commercial corn cultivars were subjected to FT-IR spectroscopy. To establish the prediction model for total protein content from corn seed, 33 corn inbreeding lines out of 176 were randomly selected and total seed protein contents using Bradford assay were examined. PLS-DA (partial least square regression discriminant analysis) could clearly discriminate F1 hybrid seeds from their parental lines. PC (principal component) loading values show that 1,700 – 1,500 cm−1 and 1,200 – 900 cm−1 regions of FT-IR spectra are significantly important for discrimination of corn lines. The prediction model for total protein contents was established by PLS (partial least square regression) algorithm, and its accuracy was confirmed by cross-validation test (R2 = 0.94). After external validation fromexternal 25 corn inbreeding lines, regression coefficient (R2) was 0.78 which indicated that the prediction model had relatively good accuracy. Thus, considering these results we suggest that FT-IR combined with multivariate analysis could be applied as a novel tool for high-throughput screening of F1 hybrid seeds from their parental lines and protein-rich lines for breeding of silage corn cultivar.

In this study, we have investigated the cytoplasmic male sterility (CMS) of a novel male sterile radish line, designated NWB CMS. The NWB CMS was crossed with 16 fertile breeding lines, and all the progenies were completely male sterile. The degree of male sterility exhibited by NWB CMS is more than Ogura CMS from the Cruciferae family. The NWB CMS was found to induce 100% male sterility when crossed with all the tested breeding lines, whereas the Ogura CMS did not induce male sterility with any of the breeding lines. PCR analysis revealed that the molecular factor that influenced Ogura CMS, the orf138 gene, was absent in the NWB CMS line, and that the orf138 gene was not also expressed in this CMS line. In order to identify the cytoplasmic factors that confer male sterility in the NWB CMS line, we carried out RFLP analyses with 32 mitochondrial genes, all of which were used as probes. Fourteen genes exhibited polymorphisms between the NWB CMS line and other radish cultivars. Based on these RFLP data, intergenic primers were developed in order to amplify the intergenic regions between the polymorphic genes. Among these, a primer pair at the 3' region of the atp6 gene (5'-cgcttggactatgctatgtatga-3') and the 5' region of the nad3 gene (5'-tcatagagaaatccaatcgtcaa-3') produced a 2 kbp DNA fragment as a result of PCR. This DNA fragment was found to be specific to NWB CMS and was not present in other CMS types. It appears that this fragment could be used as a DNA marker to select NWB CMS line in a radish-breeding program.

We attempted to establish a new selection marker system from cauliflower without antibiotics or herbicide. This new selection marker system was based on the principle that only the transformed plants could convert mannose to fructose. A transformed plant could then use the fructose as carbon source by using pmi gene that can recognize phosphomannose-isomerase that converts mannose-6-phosphate into fructose-6-phosphate. Agrobacterium tumefaciens LBA4404 contained pNWB-HSP101 vector linked with HSP101. Five days after germination, the heat tolerant gene was inoculated into the cauliflower's hypocotyl. After culturing, selection was performed in MS medium that contained BAP, NAA, 0.4% (w/v) mannose and 2.0% (w/v) sucrose. PCR, Southern blot and Northern blot analyses were performed to determine whether HSP101 gene was inserted in the transformed plant. After acclimation, seeds were successfully collected from the transformed plants in the greenhouse. This is the first report for successful transformation of cauliflower plant using the mannose selection system. Moreover, the stable selection system established in this study could be applied an alternative reducing antibiotics for human health and environmental safety during the genetic transformation of higher plants.

Nitrogen fixing Enterobacter agglomerans strains, isolated from the innermost rhizosphere of wheat, contained all genes essential for nitrogen fixation on large (100–150 kb) indigenous plasmids. One of these plasmids, pEA9, was marked with either Tn1725 or Tn5, and studied with regard to its transmissibility by conjugation with other recipient strains. In a homologous system (cured parental strains as recipients) the rate of transfer was in the order of 10-5 per recipient cell. It could be drastically increased by introduction of a mob site (on Tn5) and offering plasmid RP4 as helper. However, the host range was narrow. To obtain nif-plasmids with a wide host range, the nif-genes have been cut out and ligated to a broad host range vector, pRK415, derivative of RP4. These studies are a prerequisite for later releasing experiments.
