Purpose: Many studies have found a correlation between overall usage rates of nonmainstream forms and reading scores, but less is known about which dialect differences are most predictive. Here, we consider different methods of characterizing African American English use from existing assessments and examine which methods best predict literacy achievement. Method: Kindergarten and first-grade students who speak African American English received two assessments of dialect use and two assessments of decoding at the beginning and end of the school year. Item-level analyses of the dialect-use assessments were used to compute measures of dialect usage: (a) an overall feature rate measure based on the Diagnostic Evaluation of Language Variation-Screening Test, (b) a subscore analysis of the Diagnostic Evaluation of Language Variation-Screening Test based on items that pattern together, (c) an alternative assessment where children repeat and translate sentences, and (d) "repertoire" measures based on a categorical distinction of whether a child used a particular feature of mainstream American English. Results: Models using feature rate measures provided better data--model fit than those with repertoire measures, and baseline performance on a sentence repetition task was a positive predictor of reading score at the end of the school year. For phonological subscores, change from the beginning to end of the school year predicted reading at the end of the school year, whereas baseline scores were most predictive for grammatical subscores. Conclusions: The addition of a sentence imitation task is useful for understanding a child's dialect and anticipating potential areas for support in early literacy. We observed some support for the idea that morphological dialect differences (i.e., irregular verb morphology) have a particularly close tie to later literacy, but future work will be necessary to confirm this finding.

The Washington University School of Medicine Knight Alzheimer Disease Research Center's "African American Participation in Alzheimer Disease Research: Effective Strategies" Workshop convened to address a major limitation of the ongoing scientific progress regarding Alzheimer's disease and related dementias (ADRD): participants in most ADRD research programs overwhelmingly have been limited to non-Hispanic white persons, thus precluding knowledge as to how ADRD may be represented in non-white individuals. Factors that may contribute to successful recruitment and retention of African Americans into ADRD research were discussed and organized into actionable next steps as described within this report.

T cell cytokines contribute to immunity against Staphylococcus aureus, but the predominant T cell subsets involved are unclear. In an S. aureus skin infection mouse model, we found that the IL-17 response was mediated by γδ T cells, which trafficked from lymph nodes to the infected skin to induce neutrophil recruitment, proinflammatory cytokines IL-1α, IL-1β, and TNF, and host defense peptides. RNA-seq for TRG and TRD sequences in lymph nodes and skin revealed a single clonotypic expansion of the encoded complementarity-determining region 3 amino acid sequence, which could be generated by canonical nucleotide sequences of TRGV5 or TRGV6 and TRDV4 However, only TRGV6 and TRDV4 but not TRGV5 sequences expanded. Finally, Vγ6+ T cells were a predominant γδ T cell subset that produced IL-17A as well as IL-22, TNF, and IFNγ, indicating a broad and substantial role for clonal Vγ6+Vδ4+ T cells in immunity against S. aureus skin infections.

Purpose: Dialect-shifting has shown promise as an effective way to improve academic outcomes of students who speak nonmainstream dialects such as African American English (AAE); however, limited studies have examined the impacts of an interprofessional approach with multiple instructional methods. In this study, we developed a dialect-shifting curriculum for early elementary school students who speak AAE and evaluated the curriculum for feasibility and preliminary impacts. Method: Forty-one kindergarten, first-, second-, and third-grade students and their teachers in one elementary school participated in a 7-week dialect-shifting instruction co-taught by the classroom teachers and a speech-language pathology graduate clinician. Students' use of dialect-shifting and dialect density was measured by calculating dialect density measures in retells presented in AAE and mainstream American English and responses to situational dialect-shifting and applied dialect-shifting tasks. Teacher surveys and interviews about the feasibility and perceived impacts were conducted. Results: Initial impacts of the curriculum demonstrated increased dialect awareness for all students, with grade-level differences when students were asked to explicitly dialect-shift. In particular, second- and third-grade students were more proficient at dialect-shifting AAE features included in the curriculum. Additionally, high rates of administrator, teacher, and student satisfaction, teacher generalization, and maintenance of incorporating contrastive analysis instruction into class activities were reported. Conclusions: Literacy and play-based instruction are feasible methods to create a dialect-shifting curriculum tailored to younger students. Furthermore, the feasibility and effectiveness of the curriculum were supported by an interprofessional approach.

INTRODUCTION: Neighborhood disadvantage may be an important determinant of cardiometabolic health and cognitive aging. However, less is known about relationships among individuals with mild cognitive impairment (MCI). METHODS: The objective of this study is to investigate the relationship between neighborhood disadvantage measured by national Area Deprivation Index (ADI) rank with measures of cardiometabolic health and cognition among Wake Forest (WF) Alzheimer's Disease Research Center (ADRC) participants, with and without MCI. RESULTS: ADI was positively associated with blood pressure and cardiometabolic index (CMI), and negatively associated with global and Preclinical Alzheimer's Cognitive Composite (PACC5) scores, in cognitively unimpaired (CU) individuals. ADI was only positively associated with hemoglobin A1c (HbA1c) in MCI. DISCUSSION: Neighborhood disadvantage is associated more strongly with measures of cardiometabolic health and cognition among CU individuals rather than MCI. These findings demonstrate a need for structural solutions to address social determinants of health in an attempt to reduce cardiometabolic and cognitive risks. [ABSTRACT FROM AUTHOR]

ObjectiveTo identify a causative variant(s) that may contribute to Alzheimer disease (AD) in African Americans (AA) in the ATP-binding cassette, subfamily A (ABC1), member 7 (ABCA7) gene, a known risk factor for late-onset AD.MethodsCustom capture sequencing was performed on ∼150 kb encompassing ABCA7 in 40 AA cases and 37 AA controls carrying the AA risk allele (rs115550680). Association testing was performed for an ABCA7 deletion identified in large AA data sets (discovery n = 1,068; replication n = 1,749) and whole exome sequencing of Caribbean Hispanic (CH) AD families.ResultsA 44-base pair deletion (rs142076058) was identified in all 77 risk genotype carriers, which shows that the deletion is in high linkage disequilibrium with the risk allele. The deletion was assessed in a large data set (531 cases and 527 controls) and, after adjustments for age, sex, and APOE status, was significantly associated with disease (p = 0.0002, odds ratio [OR] = 2.13 [95% confidence interval (CI): 1.42-3.20]). An independent data set replicated the association (447 cases and 880 controls, p = 0.0117, OR = 1.65 [95% CI: 1.12-2.44]), and joint analysis increased the significance (p = 1.414 × 10(-5), OR = 1.81 [95% CI: 1.38-2.37]). The deletion is common in AA cases (15.2%) and AA controls (9.74%), but in only 0.12% of our non-Hispanic white cohort. Whole exome sequencing of multiplex, CH families identified the deletion cosegregating with disease in a large sibship. The deleted allele produces a stable, detectable RNA strand and is predicted to result in a frameshift mutation (p.Arg578Alafs) that could interfere with protein function.ConclusionsThis common ABCA7 deletion could represent an ethnic-specific pathogenic alteration in AD.

Childhood asthma is a prevalent and costly chronic condition. Optimal management enables secondary and tertiary prevention. The goal was to identify population health considerations for pediatric asthma in California to inform the development of quality improvement interventions. California Health Interview Survey 2011-2012 is a random-digit dial telephone survey conducted in 5 languages. It includes 44,000 households from all 58 counties in California. This study assessed factors related to symptom control and health care use in children ages 2-11 years with asthma. An estimated 492,385 (9.6%) of children in California currently have asthma. Urban and rural residents face comparable asthma disease burdens. School-age male children as well as Asian and African American children are disproportionately affected. Asthma causes significant morbidity, with poorer health status, high utilization of emergency care, and the need for daily medication use. Only 38% of children with asthma have a recent asthma management plan. Half of all children with asthma did not receive influenza immunization in the past year, although this reflects the overall low rate of influenza vaccination. Parents of children with asthma frequently utilize the Internet for health information and communication with their child's health care provider. Children with asthma in California face several population-level challenges, including poor health status, low influenza vaccination rates, high use of emergency care, and suboptimal use of health literacy tools. Focusing on improved care coordination and preventive care for high-risk groups is especially urgent given the expansion of public health insurance and impending shortages in the primary care workforce. Background: Vascular‐based dementia risk scores (VDRS) which reliably predict risk of Alzheimer's disease and related dementias (ADRDs), may be useful to identify at‐risk individuals for secondary prevention trials. Dementia risk scores have typically focused on predicting ADRD‐associated symptoms, with fewer studies assessing capacity for detecting individuals with underlying brain pathologies. We compare the predictive value of two vascular‐based risk scores (CAIDE and Reitz VDRSs) for discriminating AD‐related histopathological and structural abnormalities, further considering race and gender differences. Method: We included non‐demented (self‐identified) White and Black participants at the Wake Forest ADRC Clinical Core with baseline UDSv3 cognitive testing (used to calculate the Preclinical Alzheimer's Cognitive Composite ‐ PACC5), clinical adjudication, amyloid (PiB) PET, APOE genotype, and metabolic characterization for diabetes including prediabetes. WHICAP/Reitz scores were calculated using components of sex, education, age, race, ethnicity, hypertension, diabetes, WHR, HDL, smoking and APOE‐ε4 status. CAIDE scores included age, education, sex, systolic BP, BMI, physical activity, total cholesterol, and APOE‐ε4. We assessed differences in VDRS among cognitive, amyloid PET and racial groups. Result: 616 participants (Table 1) had baseline data available for analysis. Of 287 with amyloid PET, 33% were amyloid positive (CL>20). Median Reitz scores differed by cognitive status (Figure 1A), with higher mean scores noted for those with amnestic (aMCI) and non‐amnestic MCI (naMCI) compared to those with normal cognition (naMCI/aMCI: median 17, p<.001). Reitz scores were also significantly higher in amyloid positive vs negative participants (Figure 1C) as well as in Black vs White participants (Figure 2A) (median (IQR): 16 (2,34) vs 12 (0,28), 17 (0,33) vs 14 (0,40) respectively; p<.001), however no sex differences were noted. In contrast, we found no significant differences in CAIDE scores among cognitive and amyloid pathology groups (p>0.05, Figures 1B and 1D), except ethno‐racial groups (p<.001, Figure 2B). Conclusion: Significant differences in distinct VDRSs were found in relation to brain amyloid pathology levels and clinico‐cognitive status, with ethno‐racial influences potentially contributing to noted differences. Risk composites accounting for these factors will be useful for greater precision around pre‐selection of at‐risk populations towards targeted trials for disease modifying therapies and optimal management of clinical care. ImportanceMutations in known causal Alzheimer disease (AD) genes account for only 1% to 3% of patients and almost all are dominantly inherited. Recessive inheritance of complex phenotypes can be linked to long (>1-megabase [Mb]) runs of homozygosity (ROHs) detectable by single-nucleotide polymorphism (SNP) arrays.ObjectiveTo evaluate the association between ROHs and AD in an African American population known to have a risk for AD up to 3 times higher than white individuals.Design, setting, and participantsCase-control study of a large African American data set previously genotyped on different genome-wide SNP arrays conducted from December 2013 to January 2015. Global and locus-based ROH measurements were analyzed using raw or imputed genotype data. We studied the raw genotypes from 2 case-control subsets grouped based on SNP array: Alzheimer's Disease Genetics Consortium data set (871 cases and 1620 control individuals) and Chicago Health and Aging Project-Indianapolis Ibadan Dementia Study data set (279 cases and 1367 control individuals). We then examined the entire data set using imputed genotypes from 1917 cases and 3858 control individuals.Main outcomes and measuresThe ROHs larger than 1 Mb, 2 Mb, or 3 Mb were investigated separately for global burden evaluation, consensus regions, and gene-based analyses.ResultsThe African American cohort had a low degree of inbreeding (F ~ 0.006). In the Alzheimer's Disease Genetics Consortium data set, we detected a significantly higher proportion of cases with ROHs greater than 2 Mb (P = .004) or greater than 3 Mb (P = .02), as well as a significant 114-kilobase consensus region on chr4q31.3 (empirical P value 2 = .04; ROHs >2 Mb). In the Chicago Health and Aging Project-Indianapolis Ibadan Dementia Study data set, we identified a significant 202-kilobase consensus region on Chr15q24.1 (empirical P value 2 = .02; ROHs >1 Mb) and a cluster of 13 significant genes on Chr3p21.31 (empirical P value 2 = .03; ROHs >3 Mb). A total of 43 of 49 nominally significant genes common for both data sets also mapped to Chr3p21.31. Analyses of imputed SNP data from the entire data set confirmed the association of AD with global ROH measurements (12.38 ROHs >1 Mb in cases vs 12.11 in controls; 2.986 Mb average size of ROHs >2 Mb in cases vs 2.889 Mb in controls; and 22% of cases with ROHs >3 Mb vs 19% of controls) and a gene-cluster on Chr3p21.31 (empirical P value 2 = .006-.04; ROHs >3 Mb). Also, we detected a significant association between AD and CLDN17 (empirical P value 2 = .01; ROHs >1 Mb), encoding a protein from the Claudin family, members of which were previously suggested as AD biomarkers.Conclusions and relevanceTo our knowledge, we discovered the first evidence of increased burden of ROHs among patients with AD from an outbred African American population, which could reflect either the cumulative effect of multiple ROHs to AD or the contribution of specific loci harboring recessive mutations and risk haplotypes in a subset of patients. Sequencing is required to uncover AD variants in these individuals.

Performance appraisal of professional librarians in academic libraries is important because of the critical role these employees play. Professional librarians ensure that the library's resources and services are effective, relevant, and integrated within the parent institution. Performance appraisal and job feedback have been understudied in the library literature while, by comparison, these topics have generated great attention in other fields and in the corporate world. There have also been innovations in performance appraisal. Some large corporations have abandoned annual evaluations, substituting, for instance, quarterly performance snapshots and weekly check-ins with the supervisor. To investigate the current status of performance appraisal in academic libraries, we deployed a web-based survey in November 2013 to library directors in the United States. A national survey on this topic and with this population had not been conducted for 25 years. The results we report in this article relate to the following research objectives: The Snapshot research objective sought to identify the components of the performance appraisal systems currently being used. The Feedback research objective sought to identify who can give feedback during each performance appraisal event, the extent of peer-to-peer feedback, and whether there is sufficient feedback in library performance appraisal systems. We also report on programmatic effectiveness for libraries' annual evaluations as well as their overall performance appraisal system. A list of notes is included.
