Abstract

GPT-4o is an autoregressive omni model that accepts as input any combination of text, audio, image, and video, and generates any combination of text, audio, and image outputs. It's trained end-to-end across text, vision, and audio, meaning all inputs and outputs are processed by the same neural network. GPT-4o can respond to audio inputs in as little as 232 milliseconds, with an average of 320 milliseconds, which is similar to human response time in conversation. It matches GPT-4 Turbo performance on text in English and code, with significant improvement on text in non-English languages, while also being much faster and 50\% cheaper in the API. GPT-4o is especially better at vision and audio understanding compared to existing models. In line with our commitment to building AI safely and consistent with our voluntary commitments to the White House, we are sharing the GPT-4o System Card, which includes our Preparedness Framework evaluations. In this System Card, we provide a detailed look at GPT-4o's capabilities, limitations, and safety evaluations across multiple categories, focusing on speech-to-speech while also evaluating text and image capabilities, and measures we've implemented to ensure the model is safe and aligned. We also include third-party assessments on dangerous capabilities, as well as discussion of potential societal impacts of GPT-4o's text and vision capabilities.

Abstract

There exist a wide range of options for field robotics research using ground and aerial mobile robots, but there are comparatively few robust and research-ready uncrewed surface vessels (USVs). This workshop paper starts with a snapshot of USVs currently available to the research community and then describes "OtterROS", an open source ROS 2 solution for the Otter USV. Field experiments using OtterROS are described, which highlight the utility of the Otter USV and the benefits of using ROS 2 in aquatic robotics research. 10. Efficient selection of gravitationally lensed OH megamasers with MeerKAT and the Square Kilometre Array

Abstract

There has been a recent resurgence in hydroxyl (OH) megamaser research driven by Square Kilometre Array (SKA) precursor/pathfinder telescopes. This will continue in the lead-up to the SKA mid-frequency array, which will greatly expand our view of OH megamasers and their cosmic evolution over $\gtrsim80$ per cent of the age of the universe. This is expected to yield large scientific returns as OH megamasers trace galaxy mergers, extreme star formation, high molecular gas densities, and potentially binary/dual supermassive black hole systems. In this paper, we predict the distortion to the OH luminosity function that a magnification bias will inflict, and in turn, predict the distortion on the OH megamaser number counts as a function of redshift. We identify spectral flux density thresholds that will enable efficient lensed OH megamaser selection in large spectral line surveys with MeerKAT and SKA. The surface density of lensed galaxies that could be discovered in this way is a strong function of the redshift evolution of the OH megamaser luminosity function, with predictions as high as $\sim$1 lensed OH source per square degree at high redshifts ($z \gtrsim 1$) for anticipated SKA spectral line survey designs. This could enable efficient selection of some of the most highly-obscured galaxies in the universe. This high-redshift selection efficiency, in combination with the large survey speed of the SKA at $\lesssim$1 GHz frequencies and the high magnifications possible with compact OH emission regions ($\mu_{\rm OH} \gg 10$), will enable a transformational view of OH in the universe., Comment: 13 pages, 6 figures, 2 tables; Accepted in MNRAS

Abstract

XIST noncoding RNA promotes the initiation of X chromosome silencing by recruiting the protein SPEN to one X chromosome in female mammals. The SPEN protein is also called SHARP (SMRT and HDAC-associated repressor protein) and MINT (Msx-2 interacting nuclear target) in humans. SPEN recruits N-CoR2 and HDAC3 to initiate histone deacetylation on the X chromosome, leading to the formation of repressive chromatin marks and silencing gene expression. We dissected the contributions of different RNA and protein regions to the formation of a human XIST-SPEN complex in vitro and identified novel sequence and structure determinants that may contribute to X chromosome silencing initiation. Binding of SPEN to XIST RNA requires RRM 4 of the protein, in contrast to the requirement of RRM 3 and RRM 4 for specific binding to SRA RNA. Measurements of SPEN binding to full-length, dimeric, trimeric, or other truncated versions of the A-repeat region revealed that high-affinity binding of XIST to SPEN in vitro requires a minimum of four A-repeat segments. SPEN binding to XIST A-repeat RNA changes the accessibility of the RNA at specific nucleotide sequences, as indicated by changes in RNA reactivity through chemical structure probing. Based on computational modeling, we found that inter-repeat duplexes formed by multiple A-repeats can present an unpaired adenosine in the context of a double-stranded region of RNA. The presence of this specific combination of sequence and structural motifs correlates with high-affinity SPEN binding in vitro. These data provide new information on the molecular basis of the XIST and SPEN interaction.

Abstract

Hilary Putnam once suggested that "the actual existence of sets as 'intangible objects' suffers... from a generalization of a problem first pointed out by Paul Benacerraf... are sets a kind of function or are functions a sort of set?" Sadly, he did not elaborate; my aim, here, is to do so on his behalf. There are well-known methods for treating sets as functions and functions as sets. But these do not raise any obvious philosophical or foundational puzzles. For that, we first need to provide a full-fledged function theory. I supply such a theory: it axiomatizes the iterative notion of function in exactly the same sense that ZF axiomatizes the iterative notion of set. Indeed, this function theory is synonymous with ZF. It might seem that set theory and function theory present us with rival foundations for mathematics, since they postulate different ontologies. But appearances are deceptive. Set theory and function theory provide the very same judicial foundation for mathematics. They do not supply rival metaphysical foundations; indeed, if they supply metaphysical foundations at all, then they supply the very same metaphysical foundations.

14. Gastrojejunostomy junction perforation resulting from Dobhoff tube insertion in a patient with a history of Roux-en-Y surgery: a case report

Abstract

Abstract Background Gastrojejunostomy junction perforation is a rare yet critical complication associated with enteral tube placement, presenting unique challenges in patients with a history of Roux-en-Y gastric bypass surgery. Case presentation A 63-year-old white female with a complex medical history, including heart failure, atrial fibrillation, stage 4 chronic kidney disease, and prior Roux-en-Y gastric bypass surgery in 2015, experienced a significant decline in her health. She was discharged to a skilled nursing facility after a fall but deteriorated rapidly in the 2 weeks before admission. She presented with symptoms of failure to thrive, abdominal/back pain, inability to eat or drink, constipation, and stool incontinence. Lab tests showed anemia, electrolyte imbalances, and acute kidney injury. Imaging confirmed Roux-en-Y gastric bypass anatomy and a small hiatal hernia. Despite treatment attempts, her condition worsened. Nutrition discussions led to a temporary Dobhoff tube placement, considering her Roux-en-Y gastric bypass history, with plans for a gastrostomy tube. However, Dobhoff tube placement posed challenges, and imaging later revealed perforation near the gastrojejunostomy junction. After consulting with the family, the decision was made to transition the patient to comfort care due to her overall condition. Yearly education of staff about Roux-en-Y gastric bypass anatomy and updated Dobhoff placement protocol was implemented with physician oversight. Further imaging protocol in a patient who had had a Roux-en-Y gastric bypass was updated to include fluoroscopic guidance when endoscopic placement was unavailable. Conclusions This case highlights the intricacies of managing patients with Roux-en-Y gastric bypass history and underscores the need for meticulous planning and consideration of anatomical variations when performing procedures involving the gastrointestinal tract and the importance of involving multiple healthcare disciplines in complex decision-making and preventive measures to enhance patient safety in similar cases.

15. Lack of optimistic bias during social evaluation learning reflects reduced positive self-beliefs in depression and social anxiety, but via distinct mechanisms

Abstract Processing social feedback optimistically may maintain positive self-beliefs and stable social relationships. Conversely, a lack of this optimistic bias in depression and social anxiety may perpetuate negative self-beliefs and maintain symptoms. Research investigating this mechanism is scarce, however, and the mechanisms by which depressed and socially anxious individuals respond to social evaluation may also differ. Using a range of computational approaches in two large datasets (mega-analysis of previous studies, n = 450; pre-registered replication study, n = 807), we investigated how depression (PHQ-9) and social anxiety (BFNE) symptoms related to social evaluation learning in a computerized task. Optimistic bias (better learning of positive relative to negative evaluations) was found to be negatively associated with depression and social anxiety. Structural equation models suggested this reflected a heightened sensitivity to negative social feedback in social anxiety, whereas in depression it co-existed with a blunted response to positive social feedback. Computational belief-based learning models further suggested that reduced optimism was driven by less positive trait-like self-beliefs in both depression and social anxiety, with some evidence for a general blunting in belief updating in depression. Recognizing such transdiagnostic similarities and differences in social evaluation learning across disorders may inform approaches to personalizing treatment.

16. 'Snow Days Are the Best Days.' Exploring Teachers' and Administrators' Perceptions of Weather-Related School Disruptions

Abstract

Extreme weather events are becoming increasingly common and have the potential to impact the school day. This study aimed to explore teachers' and administrators' perspectives on weather-related school closures. Semi-structured interviews were conducted with ten key informants and analyzed using content analysis. Informants took a strengths-based approach and discussed the benefits of weather-related disruptions for student mental health and planning time. However, informants did mention that if these days continued to rise, it might be a cause for concern. School boards need to begin monitoring the impact of weather events.

17. A new multi-metric approach for quantifying global biodiscovery and conservation priorities reveals overlooked hotspots for amphibians

Abstract

Undocumented species represent one of the largest hurdles for conservation efforts due to the uncertainty they introduce into conservation planning. Until the distribution of earth's biodiversity is better understood, substantial conjecture will continue to be required for protecting species from anthropogenic extinction. Therefore, we developed a novel approach for identifying regions with promising biodiscovery prospects, linked to integrative conservation priorities, which we illustrate using amphibians. Our approach builds on previous estimates of biodiscovery priorities by simultaneously (1) considering linkages between spatio-environmental variables and biodiversity, (2) accounting for the negative relationship between past sampling intensity and future biodiscovery potential, (3) incorporating a priori knowledge about global species distribution patterns, (4) addressing spatial autocorrelation in community composition, and (5) weighting theoretical undocumented species by their predicted levels of conservation need. Using boosted regression trees and 50km^2 map pixels spread across the global range of amphibians, we identified several regions likely to contain many undocumented amphibian species and conservation needs, including the Southeast Asian Archipelago, humid portions of sub-Saharan Africa, and undersampled portions of the Amazon, Andes Mountains, and Central America. We also ranked top-scoring ecoregions by their mean and maximum biodiscovery potential and found that the top-20 ranked ecoregions were most concentrated in the Southeast Asian Archipelago and tropical Africa for undocumented species richness, and in tropical Africa and tropical South America for integrative undocumented amphibian conservation needs. However, high-scoring pixels tended to be widely distributed across different ecoregions for both biodiscovery scoring approaches.

Consider a variant of the usual story about the iterative conception of sets. As usual, at every stage, you find all the (bland) sets of objects which you found earlier. But you also find the result of tapping any earlier-found object with any magic wand (from a given stock of magic wands). By varying the number and behaviour of the wands, we can flesh out this idea in many different ways. This paper's main Theorem is that any loosely constructive way of fleshing out this idea is synonymous with a ZF-like theory. We give necessary and sufficient conditions under which a quasi-action of any group on an arbitrary metric space can be reduced to a cobounded isometric action on some bounded valence tree, following a result of Mosher, Sageev and Whyte. Moreover if the quasi-action is metrically proper and quasi-orbits are quasi-isometric to trees then the group is virtually free.

Abstract

We report the development of GPT-4, a large-scale, multimodal model which can accept image and text inputs and produce text outputs. While less capable than humans in many real-world scenarios, GPT-4 exhibits human-level performance on various professional and academic benchmarks, including passing a simulated bar exam with a score around the top 10% of test takers. GPT-4 is a Transformer-based model pre-trained to predict the next token in a document. The post-training alignment process results in improved performance on measures of factuality and adherence to desired behavior. A core component of this project was developing infrastructure and optimization methods that behave predictably across a wide range of scales. Abstract

Abstract Background Four out of five people living with osteoarthritis (OA) also suffer with at least one other long-term health condition. The complex interaction between OA and multiple long-term conditions (MLTCs) can result in difficulties with self-care, restricted mobility, pain, anxiety, depression and reduced quality of life. The aim of the MulTI-domain Self-management in Older People wiTh OstEoarthritis and Multi-Morbidities (TIPTOE) trial is to evaluate the clinical and cost-effectiveness of the Living Well self-management support intervention, co-designed with people living with OA, integrated into usual care, in comparison to usual care alone. Methods TIPTOE is a multi-centre, two-arm, individually randomised controlled trial where 824 individuals over 65 years old with knee and/or hip joint pain from their OA affected joint and at least one other long-term health condition will be randomised to receive either the Living Well Self-Management support intervention or usual care. Eligible participants can self-refer onto the trial via a website or be referred via NHS services across Wales and England. Those randomised to receive the Living Well support intervention will be offered up to six one-to-one coaching sessions with a TIPTOE-trained healthcare practitioner and a co-designed book. Participants will be encouraged to nominate a support person to assist them throughout the study. All participants will complete a series of self-reported outcome measures at baseline and 6- and 12-month follow-up. The primary outcome is symptoms and quality of life as assessed by the Musculoskeletal Health Questionnaire (MSK-HQ). Routine data will be used to evaluate health resource use. A mixed methods process evaluation will be conducted alongside the trial to inform future implementation should the TIPTOE intervention be found both clinically and cost-effective. An embedded ‘Study Within A Project’ (SWAP) will explore and address barriers to the inclusion of under-served patient groups (e.g. oldest old, low socioeconomic groups, ethnic groups). Discussion TIPTOE will evaluate the clinical and cost-effectiveness of a co-designed, living well personalised self-management support intervention for older individuals with knee and/or hip OA and MLTCs. The trial has been designed to maximise inclusivity and access. Trial registration ISRCTN 16024745 . Registered on October 16, 2023.

Abstract

Abstract The aim of this meta-meta-analysis was to systematically review randomised controlled trial (RCT) evidence examining the effectiveness of e- and m-Health interventions designed to improve physical activity, sedentary behaviour, healthy eating and sleep. Nine electronic databases were searched for eligible studies published from inception to 1 June 2023. Systematic reviews with meta-analyses of RCTs that evaluate e- and m-Health interventions designed to improve physical activity, sedentary behaviour, sleep and healthy eating in any adult population were included. Forty-seven meta-analyses were included, comprising of 507 RCTs and 206,873 participants. Interventions involved mobile apps, web-based and SMS interventions, with 14 focused on physical activity, 3 for diet, 4 for sleep and 26 evaluating multiple behaviours. Meta-meta-analyses showed that e- and m-Health interventions resulted in improvements in steps/day (mean difference, MD = 1329 [95% CI = 593.9, 2065.7] steps/day), moderate-to-vigorous physical activity (MD = 55.1 [95% CI = 13.8, 96.4] min/week), total physical activity (MD = 44.8 [95% CI = 21.6, 67.9] min/week), sedentary behaviour (MD = −426.3 [95% CI = −850.2, −2.3] min/week), fruit and vegetable consumption (MD = 0.57 [95% CI = 0.11, 1.02] servings/day), energy intake (MD = −102.9 kcals/day), saturated fat consumption (MD = −5.5 grams/day), and bodyweight (MD = −1.89 [95% CI = −2.42, −1.36] kg). Analyses based on standardised mean differences (SMD) showed improvements in sleep quality (SMD = 0.56, 95% CI = 0.40, 0.72) and insomnia severity (SMD = −0.90, 95% CI = −1.14, −0.65). Most subgroup analyses were not significant, suggesting that a variety of e- and m-Health interventions are effective across diverse age and health populations. These interventions offer scalable and accessible approaches to help individuals adopt and sustain healthier behaviours, with implications for broader public health and healthcare challenges.

Abstract

Manipulation of the gut microbiome using live biotherapeutic products shows promise for clinical applications but remains challenging to achieve. Here, we induced dysbiosis in 56 healthy volunteers using antibiotics to test a synbiotic comprising the infant gut microbe, Bifidobacterium longum subspecies infantis (B. infantis), and human milk oligosaccharides (HMOs). B. infantis engrafted in 76% of subjects in an HMO-dependent manner, reaching a relative abundance of up to 81%. Changes in microbiome composition and gut metabolites reflect altered recovery of engrafted subjects compared with controls. Engraftment associates with increases in lactate-consuming Veillonella, faster acetate recovery, and changes in indolelactate and p-cresol sulfate, metabolites that impact host inflammatory status. Furthermore, Veillonella co-cultured in vitro and in vivo with B. infantis and HMO converts lactate produced by B. infantis to propionate, an important mediator of host physiology. These results suggest that the synbiotic reproducibly and predictably modulates recovery of a dysbiotic microbiome.

Abstract

In response to the onset of the COVID-19 pandemic, many institutions of higher learning locked down their campuses and altered their ways of teaching. This article discusses changes made to courses at five highly varied public universities in New England participating in the multiyear Campuses for Environmental Stewardship (CES) program. The primary aim of the CES program is to integrate environmental service-learning (SL) into college curricula through workshops, faculty fellowships, and mentoring. We detail how teaching strategies were altered in fall 2020 to accommodate the threat of COVID-19 in the classroom. The authors transitioned significant portions of their instruction to online formats or outdoor classrooms. Specifics about the impacts of the shift to virtual teaching--learning are discussed, with particular focus on the impacts to the service-learning components of each of the courses.

Abstract

This paper is concerned with the everyday economic crimes of ordinary people. Although dishonesty is a core characteristic of these offences, we are not aware of any prior research that examines the direct, relationship between dishonest attitude and economic crime. Based on a survey of the British public, this article presents insights into this relationship. By profiling the integrity, dishonest disposition and everyday economic criminality of the British public, the study finds a strong relationship between attitudes and harmful dishonest behaviour such that, in a given year, 26 % of adults in the UK commit at least one economic crime. The higher levels of dishonesty in males and younger adults helps to explain their more prolific offending. Both the honesty and age-economic crime curves indicate that the maturation decline in offending is gradual compared to the sharp desistance in early adulthood of the traditional age-crime curve. Whilst this finding also contradicts the conventional view that economic crime is a middle-aged problem, it helps to explain this perception. By comparing the attitude results to a 2011 study, the research further found that the maturation effect is more pronounced due to a decline in integrity of the under 55 y population so that the younger generations now have a steeper moral hill to climb.

41. How Chemists Handle Not-Knowing in Reasoning about a Novel Problem

Abstract

The most obvious feature of expertise in chemistry is content knowledge, which defines the primary objectives of instruction. Research in chemistry education, and STEM education more broadly, has also devoted attention to students' developing scientific practices of reasoning, investigation, and learning. In this study, we set out to investigate how expert chemists reason about an unfamiliar question. We conducted semi-structured, think-aloud interviews with fourteen chemists, all of whom found the problem novel. In this article, we focus on how the chemists handled the situation of not-knowing. We analyzed the moments when they said ''I don't know'' (IDK), taking that as a clear, systematic marker of their not-knowing. The results elucidate two general dimensions of the chemists' reasoning and experience. First, their identifying what they do not know served substantive roles in their reasoning, including to mark that they needed to search for insight or information, or to mark a boundary to the problem space. Second, IDK statements served to help the chemists manage what they experienced socially and emotionally, such as to hedge or distance themselves from ideas they considered, or to forestall their own--or the interviewer's--negative judgments. We discuss both aspects of our findings, and we consider possible implications for instruction and for further research.

42. Paul's subversive leadership : gift-giving, patron-client relationships, and leadership in the Pauline correspondence

Abstract

This thesis presents the argument that Paul deliberately subverts normal practices associated with gift-giving and with patron-client relationships, which he expects to be adopted by the communities to whom he is writing. As such, contrary to other studies, I argue that there is no tension between Paul's egalitarian vision, and with the hierarchical language that he employs elsewhere. Whilst there is equality between members of the church, leadership and authority are necessary functions to help create healthy churches, and mature and Christ-like individual members. I bring together two distinct areas in this study. Primarily, this thesis is concerned with the social world in which the Pauline congregations lived, specifically customs related to gift-giving and the various relationships through which gifts were exchanged. The second chapter situates my argument within the wider body of literature offering both an analysis of contemporary scholarship, and Paul's historical context. Although the vertical dimension has been given significant attention, the horizontal dimension is less explored. The third chapter outlines the various ways in which Paul subverts the normal patterns of gift and reciprocity. I demonstrate this by considering the three gift lists - Romans 12:6-8 and 1 Corinthians 12:7-10, 27-28 - showing that Paul offers the congregations a different way of sharing gifts with each other. Chapters four and five address the apparent tension that exists between that subversive, egalitarian approach, with the hierarchical approach expressed elsewhere. Although Paul appears to remove status differences, and flattens hierarchy, he maintains a position of authority for himself (and others) at other times. Chapter four addresses the wider issues of power, authority, and leadership, arguing that Paul presents himself as a patron of the congregation, as well as shifting the focus away from the structures to the nature and character of leaders. Chapter five surveys several letters where issues of authority and leadership arise and I demonstrate that there too, Paul offers several subversive moves that transform the way his patronal position functioned. He is in no doubt, however, that such authority is necessary so that the Gospel is protected and so that the congregations continue to function as they should. The conclusion summarises the main findings ultimately showing that the concepts of equality and hierarchy, for Paul, work in harmony for the benefit of all. I also offer suggestions for further study, as well as offering thoughts on how some of the issues raised might inform conversations about church leadership today.

43. Envisioning the sign of God : the rhetorical figuration of the cross in the Epistle of Barnabas and the writings of Clement of Alexandria : semiosis, performance, and incarnation

Abstract

This thesis is a study of how and in what ways some early Christian writers came to envision the cross as a sign, and in this context, how the cross in its shape and form is seen to generate meaning. It focusses on the Epistle of Barnabas and the writings of Clement of Alexandria. It analyses examples from within these writings where the cross is presented as a sign, but in a manner which plays on the shape and materiality of the cruciform - a process referred to as the rhetorical figuration of the cross. It explores how the cross is presented as a phenomenal form which communicates divine meaning, and how it is utilised to encourage a re-orientation of life in the crucified and risen Christ. The thesis develops a novel methodology which draws on contemporary literature on signification, materiality, and performance, to encourage a fresh appreciation of the early Christian rhetorical imaging of the cross, which draws attention to its semiotic and performative dimensions. It suggests that the rhetorical figuration of the cross involves a semiosis and performance of the cross as form and thing. It argues that this semiosis reflects and participates in, the incarnational semiosis and performance of God, as Christ in becoming human enters the world of signs and signification and, in turn, the realm of embodied performativity. It relates the figuration of the cross to the theme of performance specifically, as Barnabas and Clement identify expressions of the cross in performative contexts, in bodily posture, gesture, and object, and in scenes from Greek epic and tragedy. This thesis argues that the imagining of the cross as a sign, and therein as a phenomenal geometric form which conveys theological meaning and even presence, engages the materiality of incarnation as a semiotic and performative happening.

Abstract

Abstract Therapeutic antibodies have been developed to target amyloid-beta (Aβ), and some of these slow the progression of Alzheimer’s disease (AD). However, they can also cause adverse events known as amyloid-related imaging abnormalities with edema (ARIA-E). We investigated therapeutic Aβ antibody binding to cerebral amyloid angiopathy (CAA) fibrils isolated from human leptomeningeal tissue to study whether this related to the ARIA-E frequencies previously reported by clinical trials. The binding of Aβ antibodies to CAA Aβ fibrils was evaluated in vitro using immunoprecipitation, surface plasmon resonance, and direct binding assay. Marked differences in Aβ antibody binding to CAA fibrils were observed. Solanezumab and crenezumab showed negligible CAA fibril binding and these antibodies have no reported ARIA-E cases. Lecanemab showed a low binding to CAA fibrils, consistent with its relatively low ARIA-E frequency of 12.6%, while aducanumab, bapineuzumab, and gantenerumab all showed higher binding to CAA fibrils and substantially higher ARIA-E frequencies (25–35%). An ARIA-E frequency of 24% was reported for donanemab, and its binding to CAA fibrils correlated with the amount of pyroglutamate-modified Aβ present. The findings of this study support the proposal that Aβ antibody-CAA interactions may relate to the ARIA-E frequency observed in patients treated with Aβ-based immunotherapies.

Abstract

Abstract This study explores the impact of the COVID-19 pandemic on Memorial University of Newfoundland (MUN) undergraduate and graduate students. Using the National College Health Assessment (third revision) (NCHA-III) survey, the relationship between COVID-related stressors and mental health was assessed after controlling for demographic, economic, and academic variables, and reported mental illness. A hierarchical regression revealed that psychological distress was higher for students who were undergraduate, female, of lower family income, with a pre-existing anxiety or depressive disorder. Psychological distress was also predicted by direct COVID stressors (e.g., fear of infection), as well as indirect stressors, including worries about employment and tuition, professor/instructor support, campus efforts to ensure safety, and discrimination/hostility due to race/ethnicity. Chi Square tests subsequently revealed that graduate students were more likely to be concerned about the threat COVID-19 posed to loved ones, separation from family/friends, and pandemic duration, while undergraduates were more likely concerned about returning to school, tuition, employment, and the legitimacy of their degree. Undergraduates were also more likely to have witnessed discrimination/hostility, and less likely to report professor/instructor support. Finally, independent t tests revealed that undergraduate students were significantly lower in overall social support, as well as for particular subdomains including ‘guidance’, ‘social integration’, and ‘reassurance of worth’. Interpretation of the findings and implications are considered.

Abstract Repetitive bouts of coughing expose the large airways to significant cycles of shear stress. This leads to the release of alarmins and the tussive agent adenosine triphosphate (ATP) which may be modulated by the activity of ion channels present in the human airway. This study aimed to investigate the role of the transient receptor potential subfamily vanilloid member 2 (TRPV2) channel in mechanically induced ATP release from primary bronchial epithelial cells (PBECs). PBECs were obtained from individuals undergoing bronchoscopy. They were cultured in vitro and exposed to mechanical stress in the form of compressive and fluid shear stress (CFSS) or fluid shear stress (FSS) alone at various intensities. ATP release was measured using a luciferin–luciferase assay. Functional TRPV2 protein expression in human PBECs was investigated by confocal calcium imaging. The role of TRPV2 inhibition on FSS-induced ATP release was investigated using the TRPV2 inhibitor tranilast or siRNA knockdown of TRPV2. Abstract

Background: Higher malnutrition risk is associated with mortality among patients with heart failure and heart transplantation (HT). We investigated which malnutrition risk scores – geriatric nutrition risk index (GNRI), prognostic nutritional index (PNI), or controlling nutritional status (CONUT) – have the strongest mortality association for HT candidates, and whether they are prognostic at the time of HT listing or HT operation. Methods: We reviewed adults listed for primary HT at a single center 1987–March 2022, with survival follow-up to November 2022. We constructed Cox proportional hazards models and restricted cubic spline curves for each risk score at both HT listing and HT operation and inputted HT operation as a time-varying covariate for models commencing at listing. Risk scores were assessed as continuous and binary variables. Weight change between listing and HT was evaluated similarly. Results: The 1,024 patients [76% male; median age 55 (46–61) years; HT operation n = 656] had median follow-up of 4.6 (interquartile range 1.6–8.7) years. As a continuous variable, lower GNRI at HT listing (greater malnutrition risk) was independently associated with higher mortality. As binary variables, higher risk GNRI, PNI or CONUT scores at listing were all associated with higher mortality. Conversely, risk scores at time of HT operation were not associated with subsequent mortality. There was a non-significant trend towards higher mortality with greater weight loss between listing and HT operation. Conclusion: Malnutrition risk scores showed prognostic utility only at time of HT listing. As a continuous variable, GNRI at HT listing was independently associated with subsequent mortality.

Abstract

We look at isometric actions on arbitrary hyperbolic spaces of generalised Baumslag - Solitar groups of arbitrary dimension (the rank of the free abelian vertex and edge subgroups). It is known that being a hierarchically hyperbolic group is not a quasi-isometric invariant. We show that virtually being a hierarchically hyperbolic group is not invariant under quasi-isometry either, and nor is property (QT).

ABSTRACTPrecision engineering of the gut microbiome holds promise as an effective therapeutic approach for diseases associated with a disruption in this microbial community. Engrafting a live biotherapeutic product (LBP) in a predictable, controllable manner is key to the consistent success of this approach and has remained a challenge for most LBPs under development. We recently demonstrated high-level engraftment of Bifidobacterium longum subsp. infantis (B. infantis) in adults when co-dosed with a specific prebiotic, human milk oligosaccharides (HMO). Here, we present a cellular kinetic-pharmacodynamic approach, analogous to pharmacokinetic-pharmacodynamic-based analyses of small molecule- and biologic-based drugs, to establish how HMO controls expansion, abundance, and metabolic output of B. infantis in a human microbiota-based model in gnotobiotic mice. Our data demonstrate that the HMO dose controls steady-state abundance of B. infantis in the microbiome, and that B. infantis together with HMO impacts gut metabolite levels in a targeted, HMO-dependent manner. We also found that HMO creates a privileged niche for B. infantis expansion across a 5-log range of bacterial inocula. These results demonstrate remarkable control of both B. infantis levels and the microbiome community metabolic outputs using this synbiotic approach, and pave the way for precision engineering of desirable microbes and metabolites to treat a range of diseases.
