Abstract

This paper extends previous numerical results of the Flexible Equal Employment Opportunity (FEEO) model, a goal programing model (developed by A. Charnes, W. W. Cooper, K. A. Lewis, and R. J. Niehaus) consisting of Markoff transition elements imbedded in a goal programing framework with priorities that allow for element alteration to provide the organizational flexibility (in policies of promotion, recruitment, and training) necessary to achieve long-range equal employment opportunity (EEO) goals while still preserving the high priority of meeting the shorter range day-to-day operating goals of the organization. In the first section of the paper EEO goal-setting procedures are documented. Focus is on demographic considerations (making goals realistic from the labor market point of view) and upward mobility (consideration of affirmative action policies for the internal staff). A second section provides a numerical example with actual Navy data of the extended version of the model through which internal promotion and upward mobility considerations can be evaluated explicitly. A discussion is then provided of the implementation and continuing research possibilities. (JT)

Bluetongue virus (BTV) is a species of the Orbivirus within the Reoviradae group. X-ray crystallography has been used to determine the structure of the core particle of BTV. The atomic structure gives an insight into how around 1,000 protein components self-assemble using both the classical mechanism of quasi-equivalent contacts and geometrical quasi-equivalence.

New modules, bulk data cards and DMAP sequence were added to NASTRAN to aid in the seismic analysis of nuclear power plant structures. These allow input consisting of acceleration time histories and result in the generation of acceleration floor response spectra. The resulting system contains numerous user convenience features, as well as being reasonably efficient.

Abstract

A comprehensive computer program (SIMWEST) developed for the modeling of wind energy/storage systems utilizing any combination of five types of storage (pumped hydro, battery, thermal, flywheel, and pneumatic) is described. Features of the program include: a precompiler which generates computer models (in FORTRAN) of complex wind source/storage/application systems, from user specifications using the respective library components; a program which provides the techno-economic system analysis with the respective I/O the integration of system dynamics, and the iteration for conveyance of variables; and capability to evaluate economic feasibility as well as general performance of wind energy systems. The SIMWEST operation manual is presented and the usage of the SIMWEST program and the design of the library components are described. A number of example simulations intended to familiarize the user with the program's operation is given along with a listing of each SIMWEST library subroutine.

Abstract

A study was conducted to examine the structure of the bluetongue virus core serotype 10. Using X-Ray crystallography, it was revealed that the blue tongue virus' core contains a highly-ordered double-stranded RNA genome. Measurements provided by power diffraction rings showed that the double-stranded RNA maintains a 30 A separation between strands. The study also found that genome segments' local ordering occur during genetic transcription.

Abstract

The bluetongue virus core is a molecular machine that simultaneously and repeatedly transcribes mRNA from 10 segments of viral double-stranded RNA, packaged in a liquid crystalline array. To determine how the logistical problems of transcription within a sealed shell are solved, core crystals were soaked with various ligands and analyzed by X-ray crystallography. Mg2+ ions produce a slight expansion of the capsid around the 5-fold axes. Oligonucleotide soaks demonstrate that the 5-fold pore, opened up by this expansion, is the exit site for mRNA, whilst nucleotide soaks pinpoint a separate binding site that appears to be a selective channel for the entry and exit of substrates and by-products. Finally, nucleotides also bind to the outer core layer, providing a substrate sink. [ABSTRACT FROM AUTHOR]

47. Blood Enzymes in Relation to Porcine Muscle Properties

Abstract

Three experiments utilizing 46 pigs were conducted to study the possible relationship of blood enzymes, as influenced by stress, to postmortem porcine muscle glycolysis. Animals were stressed 40 min. antemortem by transportation, physical handling and restraint for 15 minutes. Postmortem decline in pH and protein solubility and color-structure were determined on longissimusmuscle. Pigs from Experiment 1 (19 Poland China) exhibited more rapid pH decline than those from Experiments 2 (11 Hamp × York) and 3 (16 heterogeneous). Significant correlation coefficients were noted between enzyme activity and muscle properties in all experiments. Correlations were much higher, however, in Experiments 2 and 3 as both blood enzyme activities and muscle properties were more variable. In Experiment 2, the activities of lactate dehydrogenase (LDH), aldolase (ALD) and creatine kinase (CPK) were correlated with muscle pH at death (r = −.83, P<.01; r = −.78, P<.01; r = −.63, P<.05) and 1 hr. (r = −.72, P< .01; r = −.86, P<.01, r = −.57, P<.05) postmortem. LDH, ALD and CPK were correlated with decrease in sarcoplasmic and myofibrillar protein solubility (between 0 and 24 hr postmortem) as follows: 0.59, P<.05; 0.56, P<.05; 0.39, P<0.5 and 0.80, P<.01; 0.76, P<.01; 0.64, P<.05. In Experiment 3, LDH and CPK were correlated with 45 min. pH (r=−.96 and r=−.95, P<.01) and significantly related to muscle protein solubility parameters (P<.01 and P<.05). The response to thermal stress (43 C for 10 min.) in terms of blood enzyme activity was highly repeatable in pigs from Experiment 3.

48. Crystallization and Preliminary X-ray Analysis of the Core Particle of Bluetongue Virus

Abstract

Core particles of bluetongue virus serotype 1 (South Africa) have been crystallized. The crystals, which grow up to 0.8 mm in diameter, belong to a primitive orthorhombic space group and have point group symmetry 222. The unit cell dimensions are 754 × 796 × 823 Å3 and the crystallographic asymmetric unit contains one-half of a core particle. The best crystals diffract strongly to 4.8 Å Bragg spacings, which is the maximum resolution to which we can measure data with the detectors available, suggesting that useful diffraction extends well beyond this. Core particles of serotype 10 have also been crystallized but the crystals have yet to be analyzed by X-ray diffraction. Copyright 1995, 1999 Academic Press

50. Multiple proteases in foot-and-mouth disease virus replication

Abstract

Translation of foot-and-mouth disease virus RNA in a rabbit reticulocyte lysate for short time intervals resulted in the production of the peptides P20a , P16, and P88 (Lab, Lb, and P1) (R. R. Rueckert , Recommendations of the 3rd European Study Group on Molecular Biology of Picornavirus, Urbino , Italy, 1983). If further translation was prevented, the structural protein precursor P88 was not cleaved, even after prolonged incubation. This result indicates that the mechanism of the cleavage between P20a -P16 and P88 and of that between P88 and P52 (P2) differs from the mechanism of the secondary cleavages which produce the structural proteins. Furthermore, treatment of foot-and-mouth disease virus-infected cells with the protease inhibitor D-valyl phenylalanyl lysyl chloromethyl ketone prevented the in vivo cleavage between P20a -P16 and P88 but had no effect on any of the other cleavage events. These results suggest that the cleavage of the foot-and-mouth disease virus polyprotein utilizes two different host proteases.
