The glycocalyx is a complex layer of carbohydrate and protein molecules that surrounds the cell membrane of many types of mammalian cells. It serves several important functions, including cell adhesion and communication, and maintain cell shape and stability, especially in the case of erythrocytes. Alteration of glycocalyx composition represents a cardiovascular health threatening. For example, in diabetes mellitus glycocalyx of erythrocytes and of endothelial cells is known to be impaired, a potential source of blood occlusion in microcirculation, which may lead to blindness, and renal failure of patients. The impact of glycocalyx impairment on erythrocyte aggregation remains a largely unexplored research area. We conduct here in vitro-experiments in microfluidic devices in order to investigate erythrocytes aggregation incubated with amylase, an enzyme that partially breaks down glycocalyx molecules. It is found that incubation of erythrocytes by amylase leads to a dramatic increase of their aggregation and stability and alters the aggregates morphologies. Confocal microscopy analysis reveals a significant degradation of the glycocalyx layer, correlated with enhanced erythrocytes aggregation. An increased erythrocyte aggregation in vivo should affect oxygen and other metabolites delivery to organs and tissues. This study brings new elements about elucidation of microscopic origins of erythrocyte aggregation and their potential impact on cardiovascular pathologies.

The adsorption of mixtures of charged polymers onto solid surfaces presents a big interest in different technological and industrial fields, and in particular, in cosmetics. This requires to deepen on the most fundamental physico-chemical bases governing the deposition, which is generally correlated to the interactions occurring in solution. This work explores the interaction in solution of model polymer mixtures formed by a cationic homopolymer (poly(diallyl-dimethyl-ammonium chloride), PDADMAC) and a zwitterionic copolymer (copolymer of acrylic acid, 3-Trimethylammonium propyl methacrylamide chloride and acrylamide, Merquat 2003), and the adsorption of such mixtures onto negatively charged surfaces. The analysis of the interactions occurring in solution between both polymers performed using dynamic light scattering (DLS), electrophoretic mobility and viscosity measurements, combined with the study of the deposition of the layers of mixtures containing different weight fractions of each polymer using ellipsometry and quartz crystal microbalance with dissipation monitoring (QCM-D) has shown that the interpolymer complexes formed in solution, and their composition, governs the deposition onto the solid surface and the tribological properties of the adsorbed layers as shown the Surface Force Apparatus (SFA) experiments, allowing for a control of the physico-chemical properties and structure of the layers. Furthermore, the use of Self Consistent Mean Fields Calculations (SCF) confirms the picture obtained from the experimental studies of the adsorbed layers, providing a prediction of the distribution of the polymer chains within the adsorbed layers. Lift forces are widespread in hydrodynamics. These are typically observed for big and fast objects, and are often associated with a combination of fluid inertia (i.e. large Reynolds numbers) and specific symmetry-breaking mechanisms. In contrast, the properties of viscosity-dominated (i.e. low Reynolds numbers) flows make it more difficult for such lift forces to emerge. However, the inclusion of boundary effects qualitatively changes this picture. Indeed, in the context of soft and biological matter, recent studies have revealed the emergence of novel lift forces generated by boundary softness, flow gradients and/or surface charges. The aim of the present review is to gather and analyse this corpus of literature, in order to identify and unify the questioning within the associated communities, and pave the way towards future research.

Reduced blood flow, as occurring in ischemia or resulting from exposure to microgravity such as encountered in space flights, induces a decrease in the level of shear stress sensed by the endothelial cells forming the inner part of blood vessels. In the present study, we use a microvasculature-on-a-chip device in order to investigate in vitro the effect of such a reduction in shear stress on shear-adapted endothelial cells. We find that, within one hour of exposition to reduced wall shear stress, human umbilical vein endothelial cells undergo a reorganization of their actin skeleton, with a decrease in the number of stress fibers and actin being recruited into the cells' peripheral band, indicating a fairly fast change in cells' phenotype due to altered flow.

We study experimentally the motion of non-deformable microbeads in a linear shear flow close to a wall bearing a thin and soft polymer layer. Combining microfluidics and 3D optical tracking, we demonstrate that the steady-state bead/surface distance increases with the flow strength. Moreover, such lift is shown to result from flow-induced deformations of the layer, in quantitative agreement with theoretical predictions from elastohydrodynamics. This study thus provides the first experimental evidence of "soft lubrication" at play at small scale, in a system relevant {\it e.g.} to the physics of blood microcirculation.

We describe a microscope-based optical setup that allows us to perform space-and time-resolved measurements of the spectral reflectance of transparent substrates coated with ultrathin films. This technique is applied to investigate the behavior in water of thermosensitive polymer brushes made of poly(N-isopropylacrylamide) grafted on glass. We show that spectral reflectance measurements yield quantitative information about the conformation and axial structure of the brushes as a function of temperature. We study how molecular parameters (grafting density, chain length) affect the hydra-tion state of a brush, and provide one of the few experimental evidence for the occurrence of vertical phase separation in the vicinity of the lower critical solution temperature of the polymer. The origin of the hysteretic behavior of poly(N-isopropylacrylamide) brushes upon cycling the temperature is also clarified. We thus demonstrate that our optical technique allows for in-depth characterization of stimuli-responsive polymer layers, which is crucial for the rational design of smart polymer coatings in actuation, gating or sensing applications., Comment: Revised version. The unique ability of a red blood cell to flow through extremely small microcapillaries depends on the viscoelastic properties of its membrane. Here, we study in vitro the response time upon flow startup exhibited by red blood cells confined into microchannels. We show that the characteristic transient time depends on the imposed flow strength, and that such a dependence gives access to both the effective viscosity and the elastic modulus controlling the temporal response of red cells. A simple theoretical analysis of our experimental data, validated by numerical simulations, further allows us to compute an estimate for the two-dimensional membrane viscosity of red blood cells, $\eta_{mem}^{2D}\sim 10^{-7}$ N$\cdot$s$\cdot$m$^{-1}$. By comparing our results with those from previous studies, we discuss and clarify the origin of the discrepancies found in the literature regarding the determination of $\eta_{mem}^{2D}$, and reconcile seemingly conflicting conclusions from previous works.

We describe the design of micropatterned surfaces for single cell studies, based on thermoresponsive polymer brushes. We show that brushes made of poly(N-isopropylacrylamide) grafted at high surface density display excellent protein and cell anti-adhesive properties. Such brushes are readily patterned at the micron scale via deep UV photolithography. A proper choice of the adhesive pattern shapes, combined with the temperature-dependent swelling properties of PNIPAM, allow us to use the polymer brush as a microactuator which induces cell detachment when the temperature is reduced below 32degC.

We probe the rheology of the model liquid octamethylcyclotetrasiloxane (OMCTS) confined into molecularly thin films, using a unique Surface Forces Apparatus allowing to explore a large range of shear rates and confinement. We thus show that OMCTS under increasing confinement exhibits the viscosity enhancement and the non-linear flow properties characteristic of a sheared supercooled liquid approaching its glass transition. Besides, we study the drainage of confined OMCTS via the propagation of "squeeze-out" fronts. The hydrodynamic model proposed by Becker and Mugele [Phys. Rev. Lett. {\bf 91}, 166104 (2003)] to describe such front dynamics leads to a conclusion in apparent contradiction with the dynamical slowdown evidenced by rheology measurements, which suggests that front propagation is not controlled by large scale flow in the confined films.

We probe, using the Surface Forces Apparatus, the thermal response of poly(N-isopropylacrylamide) (PNIPAM) brushes of various grafting densities, grown from plasma-activated mica by means of surface-initiated polymerization. We thus show that dense thermoresponsive brushes collapse gradually as temperature is increased, and that grafting density greatly affects their ability to swell: the swelling ratio of the brushes, which characterizes the thickness variation between the swollen and the collapsed state, is found to decrease from $\sim 7$ to $\sim 3$ as the number of grafted chains per unit area increases. Such a result, obtained with an unprecedent resolution in grafting density, provides qualitative support to calculations by Mendez {\it et al.} [{\it Macromolecules} {\bf 2005} {\it 38}, 174]. We further show that, in contrast to swelling, adhesion between two PNIPAM brushes appears to be rather insensitive to their molecular structure.

We investigate the effect of loading rate on drainage in molecularly thin films of a simple fluid made of quasi-spherical molecules (octamethylcyclotetrasiloxane, OMCTS). We find that (i) rapidly confined OMCTS retains its tendency to organize into layers parallel to the confining surfaces, and (ii) flow resistance in such layered films can be described by bulklike viscous forces if one accounts for the existence of one monolayer immobilized on each surfaces. The latter result is fully consistent with the recent work of Becker and Mugele, who reached a similar conclusion by analyzing the dynamics of squeeze-out fronts in OMCTS [T. Becker and F. Mugele, Phys. Rev. Lett. {\bf 91} 166104(2003)]. Furthermore, we show that the confinement rate controls the nature of the thinning transitions: layer-by-layer expulsion of molecules in metastable, slowly confined films proceeds by a nucleation/growth mechanism, whereas deeply and rapidly quenched films are unstable and undergo thinning transitions akin to spinodal decomposition.

We probe the temperature dependence of friction at the interface between a glassy poly(methylmethacrylate) lens and a flat substrate coated with a methyl-terminated self-assembled monolayer. The monolayer exhibits density defects which act as pinning sites for the polymer chains. We show that the shear response of such an interface supports the existence, at the surface of the glassy polymer, of a nanometer-thick layer of mobile chains. Friction can be ascribed to the interplay between viscouslike dissipation in this layer and depinning of chains adsorbed on the substrate. We further show that the pinning dynamics is controlled by \beta rotational motions localized at the interface.

We perform drainage experiments of a linear alkane fluid (n-hexadecane) down to molecular thicknesses, and focus on the role played by the confinement rate. We show that molecular layering is strongly influenced by the velocity at which the confining walls are approached: under high enough shear rates, the confined medium behaves as a structureless liquid of enhanced viscosity for film thickness below $\sim$10 nm. Our results also lead us to conclude that a rapidly confined film can be quenched in a metastable disordered state, which might be related with recent intriguing results on the shear properties of confined films produced at different rates [Zhu and Granick, Phys. Rev. Lett. {\bf 93}, 096101 (2004)].

We describe a surface force apparatus designed to probe the rheology of a nanoconfined medium under large shear amplitudes (up to 500 $\mu$m). The instrument can be operated in closed-loop, controlling either the applied normal load or the thickness of the medium during shear experiments. Feedback control allows to greatly extend the range of confinement/shear strain attainable with the surface force apparatus. The performances of the instrument are illustrated using hexadecane as the confined medium.

We present single contact friction experiments between a glassy polymer and smooth silica substrates grafted with alkylsilane layers of different coverage densities and morphologies. This allows us to adjust the polymer/substrate interaction strength. We find that, when going from weak to strong interaction, the response of the interfacial junction where shear localizes evolves from that of a highly viscous threshold fluid to that of a plastically deformed glassy solid. This we analyse as resulting from an interaction-induced ``interfacial glass transition'' helped by pressure.

We report on the frictional properties of a single contact between a glassy polymer lens and a flat silica substrate covered either by a disordered or by a self-assembled alkylsilane monolayer. We find that, in contrast to common belief, the Amontons proportionality between frictional and normal stresses does not hold. Besides, we observe that the velocity dependence of the sliding stress is strongly sensitive to the structure of the silane layer. Analysis of the frictional rheology observed on both disordered and self-assembled monolayers suggests that dissipation is controlled by the plasticity of a glass-like interfacial layer in the former case, and by pinning of polymer chains on the substrate in the latter one., Comment: submitted to Eur. We study the friction of a poly(dimethylsiloxane) (PDMS) rubber network sliding, at low velocity, on a substrate on which PDMS chains are end-tethered. We thus clearly evidence the contribution to friction of the pull-out mechanism of chain-ends that penetrate into the network. This interfacial dissipative process is systematically investigated by probing the velocity dependence of the friction stress and its variations with the grafting density and molecular weight of the tethered chains. This allows us to confirm semi-quantitatively the picture of arm retraction relaxation of the grafted chains proposed in models of slippage at a network/brush interface.
