Valproic acid, one of the most used drugs for epilepsy patients, has some known side effects. Chard extract has many pharmacological activities. The study aims to evaluate whether valproic acid might interfere with oxidative metabolism in gastric tissue and whether chard ameliorates these effects. The Sprague Dawley rats were divided into four groups (n=8); control, chard-given control, valproic acid, and chard-given valproic acid. The aqueous extracts of chard leaves were given 1 h before the administration of valproic acid for 7 days. On the 8th day, the animals were sacrificed under anesthesia and gastric tissues were homogenized. When compared to the control group, valproic acid significantly increased malondialdehyde and catalase activity, while superoxide dismutase activity decreased. Chard administration increased glutathione and, sialic acid levels and decreased malondialdehyde levels and superoxide dismutase activity in the valproic acid group. Based on these findings, since chard increased gastric sialic acid levels, we may suggest that chard may protect gastric mucosa may be through its antioxidant effects.

The aim of our study was to examine the effects of exercise and calorie restriction on various tissue damage and antioxidant parameters in the brain and testis of rats in a natural aging model. For this purpose, male Sprague-Dawley rats were the control group (C), the elderly (A), the elderly with calorie restriction (ACR), the elderly who were exercised (AE) and the elderly who were exercised with calorie restriction (ACRE), they were divided into 5 groups. The control group was composed of three-month-old animals. The other group consisted of 15-month-old rats. Exercise and calorie restriction were applied for 6 weeks. At the end of the experiment, lipid peroxidation (LPO), nitric oxide (NO), glutathione (GSH) levels and superoxide dismutase (SOD), catalase (CAT), glutathione-S-transferase (GST) and tissue factor (TF) were determined in brain and testicular tissues homogenates. As a result of the study, the A group’s brain and testis LPO, NO levels and TF activity increased, GSH levels and SOD, CAT and GST activities decreased, when compared to the C group. As a result of our study, an increase in oxidant damage was observed with TF activity in the brain and testis in the natural aging model, and positive effects of exercise and calorie restriction on the antioxidant levels in the brain were determined, especially in aging. Amaç: Aritmi tedavisinde kullanılan amiodaronun çeşitli organlarda yan etkilere neden olduğu gösterilmiştir. Beyaz lahana (Brassica oleracea L. var. capitata) yaygın olarak tüketilen bir sebzedir. Amiodaron maruziyeti ve beyaz lahana tüketiminin kalp dokusu üzerindeki etkileri araştırılmamıştır. Beyaz lahana ekstraktının amiodaron uygulanan sıçanların kalp dokuları üzerindeki etkilerini araştırmayı amaçladık. Gereç ve Yöntem: Sıçanlara yedi gün boyunca gavajla beyaz lahana ekstresi ve amiodaron verildi. Beyaz lahana ekstresinin uygulanmasından 1 saat sonra amiodaron uygulandı. Sekizinci günde, hayvanların kalpleri alındı ve homojenize edildi. Lipit peroksidasyon (LPO), indirgenmiş glutatyon (GSH), siyalik asit (SA) düzeyleri ile glutatyon-S-transferaz (GST), katalaz, süperoksit dismutaz (SOD) ve doku faktörü (TF) aktiviteleri belirlendi. Bulgular: Amiodaron verilen grupta GSH, CAT, SOD aktiviteleri azalırken LPO ve SA seviyeleri arttı. Beyaz lahana, LPO ve SA miktarını azaltarak amiodaron zararını önledi. Ayrıca GSH düzeyleri ile SOD, CAT ve GST aktivitelerini artırarak antioksidan savunma sistemini güçlendirdi. Sonuç: Beyaz lahana tüketiminin, amiodaronun kalp dokusunda olumsuz etkilerini önlemek için alternatif bir yaklaşım olabileceği ileri sürülebilir., Objective: Amiodarone is used in the treatment of arrhythmia and has been shown to cause side effects in various organs. White cabbage (Brassica oleracea L. var. capitata) is a widely consumed vegetable. The effects of exposure to amiodarone and white cabbage consumption on heart tissue has not previously been investigated. We aimed to investigate the effects of white cabbage extract on amiodarone given to rats’ cardiac tissues. Material and Method: Rats were given white cabbage extract and amiodarone by gavage for seven days. Amiodarone was applied 1 hour after administration of the white cabbage extract. On the eighth day, the hearts of the animals were taken and homogenized. The levels of lipid peroxidation (LPO), reduced glutathione (GSH), sialic acid (SA) and activities of glutathione-S-transferase (GST), catalase, superoxide dismutase (SOD) and tissue factor (TF) were determined. Results: The GSH level, CAT and SOD activities decreased, while the LPO and SA levels increased in the amiodarone treated group. The White cabbage extract prevented any damage of the amiodarone by decreasing the amount of LPO and SA and also strengthened the antioxidant defence system by increasing the amount of the GSH and SOD activities and the CAT and GST activities. Conclusion: It is suggested that the consumption of white cabbage could be an alternative approach to the prevention of the adverse effects of amiodarone on heart tissue.

An increasing number of studies have pointed out the side effects of valproic acid (VPA), an antiepileptic drug used for the treatment of seizures in children and adults. The aim of this study is to evaluate whether VPA interferes with oxidative metabolism in the heart and whether edaravone, the novel free radical scavenger, ameliorates any such effects.Female rats were divided into four groups: intact control animals, VPA (0.5 g/kg/day), edaravone (30 mg/kg/day), and VPA+edaravone (0.5 g/kg/day+30 mg/kg/day) injected groups for seven days. On the 8(th) day the animals were sacrificed under ether anesthesia, and hearts were homogenized. Concentrations of malondialdehyde (MDA), sialic acid (SA), glutathione (GSH) and the activities of superoxide dismutase (SOD), catalase (CAT), glutathione -S- transferase (GST), glutathione peroxidase (GPx), myeloperoxidase (MPO), Na+K+ ATPase and tissue factor (TF) were evaluated in the homogenates.In the VPA group, increased MDA levels and decreased GPx activities indicated heart damage compared with the control group. On the other hand, edaravone treatment in the VPA group increased the activities of GST and SOD and decreased the activities of TF and ALP.Our study is the first to demonstrate the beneficial effects of edaravone on the impaired oxidant/antioxidant status of heart in VPA-induced toxicity.

Tezin başlığı : Moringa oleifera’nın valproik asit verilen --sıçanlarda tükürük bezleri üzerine olası koruyucu etkisinin incelenmesiAmaç: Bu çalışmada, M. oleifera ekstresinin valproik asit verilen Sprague Dawley sıçanlarında tükürük bezi üzerine olası koruyucu etkisinin araştırılması amaçlandı.Gereç ve Yöntem: Sprague Dawley dişi sıçanları; kontrol grubu (%0,9 g’lık NaCl verilen), kontrol + moringa grubu (0,3 g/kg/gün dozunda %70’lik etanollü Moringa oleifera ekstresi verilen), valproik asit grubu (0,5 g/kg/gün valproik asit verilen) ve valproik asit + moringa grubu (benzer dozlarda valproik asit ve Moringa oleifera ekstresi verilen) olmak üzere dört gruba ayrıldı. On altıncı günde aç bırakılan sıçanlar anestezi altında sakrifiye edildi. Parotis, submandibular ve sublingual tükürük bezleri alınarak %0,9 g’lık NaCl ile homojenize edildi ve deney gününe kadar -78 oC’de saklandı. Tüm tükürük bezlerinde çeşitli biyokimyasal antioksidan ve oksidan parametreler tayin edildi.Bulgular: Valproik asit verilen tüm tükürük bezlerinde glutatyon, glutatyon-S- transferaz, süperoksit dismutaz değerleri kontrol grubu sıçanlarına göre anlamlı olarak azaldı. Total antioksidan kapasite ise sadece parotis ve sublingual tükürük bezlerinde azaldı, submandibular bezde anlamlı olarak değişmedi. Lipid peroksidasyon, siyalik asit, nitrik oksit değerleri ise valproik asit verilen grupta kontrol grubuna göre anlamlı olarak arttı. Total oksidan kapasite ise sublingual bezde değişmedi ancak diğer dokularda anlamlı derecede arttı. Katalaz, doku faktörü aktivitesi ve total protein değerlerinde ise kontrol grubu ve valproik asit verilen grupta anlamlı olarak değişmedi. Moringa oleifera ekstresinin valproik asit grubuna uygulanması, çeşitli antioksidan ve terapötik özellikleri aracılığıyla valproik asidin neden olduğu değişen biyokimyasal parametreleri olumlu yönde etkiledi.Sonuç: Moringa oleifera ekstresi tükürük bezlerinde valproik aside bağlı oksidan hasarın gerilemesinde etkili oldu. Parotis, submandibular and sublingual salivary glands were taken and homogenized with 0,9 g % NaCl and stored at -78 °C until the day of the experiment. Various biochemical antioxidant and oxidant parameters were determined in all salivary glands.Results: Glutathione, glutathione-S-transferase and superoxide dismutase values in all salivary glands given valproic acid were significantly decreased compared to control group rats. Total antioxidant capacity decreased only in parotis and sublingual salivary glands. It did not change significantly in the submandibular gland. Lipid peroxidation, sialic acid and nitric oxide values increased significantly in the valproic acid group compared to the control group. Total oxidant capacity did not change in the sublingual gland, but increased significantly in the others. Catalase, tissue factor activity and total protein values did not change significantly between groups. Moringa oleifera extract was found to correct these biochemical deteriorations caused by valproic acid through its various antioxidant and therapeutic properties.Conclusion: Moringa oleifera extract was effective in the regression of valproic acid- induced oxidant damage in salivary glands. Objective: The aim of this in vitro study was to investigate the fluoride-releasing/recharging and antimicrobial effects of restorative materialsused in pediatric dentistry.Material and Method: High-viscosity and resin-modified glass-ionomercement, glass carbomer, compomer, giomer and composite weretested. Dental materials’ fluoride-release was measured in distilled water(DW) and artificial saliva (AS) for eight weeks using a fluoride-specificelectrode. At the end of eight weeks, specimens were divided intothree groups for recharging with acidulated phosphate fluoride gel andfluoride varnish. After recharge, fluoride release was measured again.Agar diffusion test was used to assess antimicrobial effects againstS. mutans and L. acidophilus. The statistical analysis was done withMann-Whitney U and Friedman tests using SPSS-22 software.Results: High-viscosity glass-ionomer cement showed the highestfluoride-release in both storage media, followed by glass carbomer,while giomer demonstrated the lowest. After recharge, the increasein the varnish group was significantly higher than others (p
