Günümüzde işletme ve organizasyonların ekonomik dalgalanmalar, teknolojik gelişmeler ve değişen müşteri talepleri nedeniyle gittikçe karmaşıklaşan çevre ve rekabet ortamı içerisinde ayakta kalabilmeleri için kurumsallaşmış bir yönetime ve etkin bir iç kontrol sistemine sahip olmaları beklenmektedir. Kurumsallaşmayı yakalamış demokratik ve katılımcı bir anlayışla yönetilen kurumlar, kurumsallaşmanın günümüz çalışan ve müşteri beklentilerine cevap verebilecek öğeleri ve göstergeleri sayesinde amaç ve hedeflerine ulaşabilmektedirler. Eski yönetim modellerinin yerini alan kurumsal yönetim, işletmeleri sadece kâr amacı güden organizasyonlar olmalarının yanısıra çalışan ve hizmet alan memnuniyetinin önemsendiği kurumlara dönüştürmüştür. Son yıllarda, kurumsallaşma ve iç kontrol sisteminin, özel sektörde faaliyet gösteren kurumlar kadar devlet kurumlarının da çalışan motivasyonunu sağlayarak amaç ve hedeflerine ulaşabilmeleri ve varlıklarını sürdürebilmeleri için son derece önemli olduğu anlaşılmaktadır. Buradan yola çıkılarak bu çalışmada Karadeniz Teknik Üniversitesi kurumu düzeyinde kurumsallaşma ve iç kontrol sistemi odaklı bir araştırma yapılarak bu kavramların aralarında bir ilişki olup olmadığı ile kurumdaki etkinlikleri belirlenmek istenmiştir. Bunun için Üniversite bünyesine bağlı birimlerin bütçe ve faaliyet durumları araştırılmış ve kurum statüsünde olan 38 birim tespit edilmiştir. Bunlardan 37 birim yöneticisinin her birine nicel bir yöntem olan anket yöntemi uygulanarak tam sayım araştırması yapılmıştır. Mümkün olduğunca yüzyüze görüşmeler de yapılmaya çalışılmıştır. Elde edilen bulgular neticesinde Karadeniz Teknik Üniversitesinde kurumsallaşa süreci ve iç kontrol sistemi arasında yüksek derecede pozitif yönlü anlamlı bir ilişki olduğu ve iç kontrol sürecinin kurumsallaşmaya pozitif yönde etki ettiği ortaya konmuştur. Üniversitede iç kontrol sisteminin etkinliğinin 3,75 puanla, kurumsallaşma seviyesinin 3,98 puanla yüksek seviyede olduğu anlaşılmıştır. Today, enterprises and organizations are expected to have an institutionalized management and an effective internal control system in order to survive in an increasingly complex environment and competitive environment due to economic fluctuations, technological developments and changing customer demands. The institutions managed with a democratic and participatory approach that has achieved institutionalization can achieve the goals and objectives of institutionalization thanks to the elements and indicators that can meet the needs of today's employees and customers. The corporate governance, which replaces the old management models, has transformed the enterprises into the organizations where employee and service-buyer satisfaction are cared as well as being just profit-oriented organizations. In recent years, it has been understood that institutionalization and internal control system is highly essential to reach the goals and objectives and to sustain their presence by providing employee satisfaction for state institutions inasmuch as for the instutions operating in private sector. In this study, it was aimed to determine whether there is a relationship between these concepts and to identify their effectiveness in the institution by conducting a research on institutionalization and internal control system at the level of the institution of Karadeniz Technical University. For this purpose, the budget and the activity of the units affiliated to the University were investigated and 38 units with the status of institution were determined. Out of these 38 units, a quantitative method was applied to each of the manager of the 37 units and a full census survey was conducted. Face-to-face meetings were tried to be made as much as possible. As a result of the findings, it has been revealed that there is a highly significant positive relationship between the institutionalization process and the internal control system at Karadeniz Technical University and the internal control process has a positive effect on institutionalization. It has been understood that the effectiveness of the internal control system at the university has been 3.75 and the institutionalization level has been 3.98. 10. Immune modulatory effects of pediococcus pentosaceus derived membrane vesicles: Mechanism of action and therapeutic applications

Önceki çalışmalarımızda 5 farklı insan kommensal bakteri izolatından salgılanan membran keseciklerini (MV) karakterize edip etkinliklerini patojenik olmayan E.coli bakteri izolatından salgılanan membran kesecikleriyle karşılaştırdık. Sonuçlar, kommensal bakteri kökenli MV'lerin immün düzenleyici etkilerinin olduğunu, buna karşın patojenik olmayan E.coli bakteri izolatından salgılanan MV'lerin ise immün uyarıcı özellikleri olduğunu gösterdi. Bu tez çalışmasında, en yüksek immün düzenleyici etki gösteren Pediococcus pentosaceus izolatından salgılanan MV'lerin etkileri ayrıntılı bir şekilde incelenmiştir. OVA model antijeni immünizasyon modelinde Pediococcus pentosaceus kökenli MV'ler anti-OVA spesifik IgG1 ve IgG2c ile CTL yanıtlarını baskılamıştır. MV'lerin farklı hücre tipleri üzerindeki etkileri incelendiğinde immün düzenleyici aktivitenin M2 makrofajlarından ve myeloid kökenli baskılayıcı hücrelerden kaynaklandığı, ödüzenleyici T hücrelerin bir rolü olmadığı gözlemlendi. MV'lerin antienflamatuar etkileri farede farklı akut enflamasyon modelleri oluşturularak test edildi. Zymosan aracılı peritonit modelinde MV'ler şiddetli enflammasyon oluşumunu, nötrofillerin periton boşluğuna toplanmasını ve makrofajların enflamasyon yanıtından dolayı ölümlerini engellemiştir. Dextran sodyum sülfat aracılı akut kolit modelinde MV'lerle yapılan geç tedavinin (0. ve 3. günlerde) anti-enflamatuar koruyucu etkileri olduğu, kolon kısalmasını ve kript yapı bozulmasını engellediği gözlemlendi. Eksizyonel yara iyileşmesi modelinde, intraperitoneal MV uygulamasının yara bölgesine PD-L1 ifade eden miyeliod kökenli hücre toplanması aracılığıyla yara iyileşmesi sürecini hızlandırdığı bulunmuştur. Bütün bu sonuçlar Pediococcus pentosaceus izolatından salgılanan membran keseciklerinin baskılayıcı – düzenleyici hücrelerin aktivasyonu üzerinde etkileri olduğunu ve potansiyel antienflamatuar ajanlar olarak enflamatuar hastalıkların ya da otoimmün hastalıkların tedavisinde kullanılabileceklerini göstermektedir. In our previous studies, we characterized 5 different human gram positive commensal bacteria derived membrane vesicles (MVs) and compared their activity with non-pathogenic E.coli derived membrane vesicles. Results showed that commensal bacteria derived MVs had immunomodulatory properties whereas non-pathogenic E.coli derived membrane vesicles had immune stimulatory properties. In this thesis, we aimed to focus our attention to Pediococcus pentosaceus-derived MVs that displayed the highest immunomodulatory activity. In an immunization model, Pediococcus pentosaceus-derived MVs supressed anti-OVA specific IgG1 and IgG2c and CTL responses. Analysis of MV effect on different cell types showed that MVs exerted an immunomodulatory response by generating M2 macrophages and myeloid derived suppressor cells (MDSCs) but not regulatory T cells. MVs' anti-inflammatory effects were also tested in acute inflammation models established in mice. In zymosan induced peritonitis model, MVs ameliorated excessive inflammation by reducing neutrophil recruitment to peritoneal cavity and inhibiting macrophage loss caused by inflammation. In dextran sodium sulphate (DSS) induced acute colitis model, post-treatment with MVs (Day 0 and 3) prevented colon shortening and loss of crypt architecture. In an excisional wound healing model, intraperitoneal MV administration accelerated wound closure through recruitement of PD-L1 expressing myeloid cells to the wound site. Collectively, these results indicate that Pediococcus pentosaceus derived membrane vesicles activates suppressor – regulatory cell types and can be used as potent anti-inflammatory agents for the treatment of inflammatory or autoimmune diseases. 150

Immunomodulatory commensal bacteria modify host immunity through delivery of regulatory microbial-derived products to host cells. Extracellular membrane vesicles (MVs) secreted from symbiont commensals represent one such transport mechanism. How MVs exert their anti-inflammatory effects or whether their tolerance-inducing potential can be used for therapeutic purposes remains poorly defined. In this study, we show that MVs isolated from the human lactic acid commensal bacteria Pediococcus pentosaceus suppressed Ag-specific humoral and cellular responses. MV treatment of bone marrow–derived macrophages and bone marrow progenitors promoted M2-like macrophage polarization and myeloid-derived suppressor cell differentiation, respectively, most likely in a TLR2-dependent manner. Consistent with their immunomodulatory activity, MV-differentiated cells upregulated expression of IL-10, arginase-1, and PD-L1 and suppressed the proliferation of activated T cells. MVs’ anti-inflammatory effects were further tested in acute inflammation models in mice. In carbon tetrachloride–induced fibrosis and zymosan-induced peritonitis models, MVs ameliorated inflammation. In the dextran sodium sulfate–induced acute colitis model, systemic treatment with MVs prevented colon shortening and loss of crypt architecture. In an excisional wound healing model, i.p. MV administration accelerated wound closure through recruitment of PD-L1–expressing myeloid cells to the wound site. Collectively, these results indicate that P. pentosaceus–derived MVs hold promise as therapeutic agents in management/treatment of inflammatory conditions. [ABSTRACT FROM AUTHOR]

Kalan ömür süreci ile ilgili uygulamalarda genellikle sürecin H ortalama değer fonksiyonu bilgisine ihtiyaç duyulur. Fakat, H fonksiyonu bazı özel durumlar dışında analitik olarak elde edilemez. Bundan dolayı bu fonksiyon için sınırlar büyük değere sahiptir. Bu çalışmada H ortalama kalan ömür fonksiyonunun düzgün, üstel ve Erlang gibi bazı dağılımlar için literatürde mevcut olan analitik ifadeleri ve genel halde bazı sınır fonksiyonları verildikten sonra H için bir integral denklem yardımıyla tanımlanan fonksiyonlar dizisinin H fonksiyonuna monoton olarak yakınsamasına bağlı olarak H için bazı sınır fonksiyonlarının elde edilmesi üzerinde durulur. H fonksiyonunun monotonluğu incelenir ve bu fonksiyonun kendi asimptotik ifadesi ve yalnızca yeni bir parça için geriye kalan ortalama ömür fonksiyonu ile olan sınırsal anlamdaki ilişkisi ortaya çıkartılmaya çalışılır. In the applications related to residual life process, generally it is needed to get information on the H mean value function of the process. But, H function cannot be obtained analitically except some specific situations. Therefore, bounds have high value for this function. In this study, already existing of some distributions such as uniform, exponential and Erlang of H mean residual life function, H is emhasized to obtain some bound functions depending on monotonous convergence of series of functions defined by an integral equation for H. Monotonity of H function is analysed and it is discovered boundary relation of this function between its own asymptotic expression and residual mean life function for only a new part. 87

Bone marrow adipose tissue (BMAT) has garnered significant attention due to its critical roles in leukemia pathogenesis, cancer metastasis, and bone marrow failure. BMAT is a metabolically active, distinct tissue that differs from other fat depots. Marrow adipocytes, closely interacting with hematopoietic stem/progenitor cells and osteoblasts, play a pivotal role in regulating their functions. However, standardized methods for isolating and defining human BMAT (hBMAT) remain unclear. In animal models, BMAT is commonly isolated directly from the bone marrow through flushing, enzymatic digestion, or mechanical disruption. In humans, BMAT isolation often involves the adipogenic induction of bone marrow mesenchymal stem/stromal cells (BM-MSCs) derived from bone marrow aspirates. However, this approach reflects cellular responses to chemical stimuli and may not accurately represent in vivo differentiation potential. Similarly, BMAT obtained from hip or knee replacement surgeries might not reflect basal physiological conditions due to inflammatory influences. Here, we describe a direct method for culturing BMAT from the fatty layer of bone marrow aspirates obtained from healthy transplant donors. This protocol employs centrifugation and washing steps using basic laboratory equipment, offering simple and non-enzymatic approach. 15. How Long Does Deep Brain Stimulation Give Patients Benefit?

Introduction: One of the most common questions patients ask when they are contemplating deep brain stimulation (DBS) is how long it will last. To guide physicians in answering this query, we performed a scoping review to assess the current state of the literature and to identify the gaps that need to be addressed., Materials and Methods: The authors performed a MEDLINE search inclusive of articles from January 1987 (advent of DBS literature) to June 2023 including human and modeling studies written in English. For longevity of therapy data, only studies with a mean follow-up of ≥three years were included. Using the Rayyan platform, two reviewers (JP and RM) performed a title screen. Of the 734 articles, 205 were selected by title screen and 109 from abstract review. Ultimately, a total of 122 articles were reviewed. The research questions we explored were 1) how long can the different components of the DBS system maintain functionality? and 2) how long can DBS remain efficacious in treating Parkinson's disease (PD), essential tremor (ET), dystonia, and other disorders?, Results: We showed that patients with PD, ET, and dystonia maintain a considerable long-term benefit in motor scores seven to ten years after implant, although the percentage improvement decreases over time. Stimulation off scores in PD and ET show worsening, consistent with disease progression. Battery life varies by the disease treated and the programming settings used. There remains a paucity of literature after ten years, and the impact of new device technology has not been classified to date., Conclusion: We reviewed existing data on DBS longevity. Overall, outcomes data after ten years of therapy are substantially limited in the current literature.
