Cannabis plants have been used in medicine since ancient times. They are well known for their anti-diabetic, anti-inflammatory, neuroprotective, anti-cancer, anti-oxidative, anti-microbial, anti-viral, and anti-fungal activities. A growing body of evidence indicates that targeting the endocannabinoid system and various other receptors with cannabinoid compounds holds great promise for addressing multiple medical conditions. There are two distinct avenues in the development of cannabinoid-based drugs. The first involves creating treatments directly based on the components of the cannabis plant. The second involves a singular molecule strategy, in which specific phytocannabinoids or newly discovered cannabinoids with therapeutic promise are pinpointed and synthesized for future pharmaceutical development and validation. Although the therapeutic potential of cannabis is enormous, few cannabis-related approved drugs exist, and this avenue warrants further investigation. With this in mind, we review here the medicinal properties of cannabis, its phytochemicals, approved drugs of natural and synthetic origin, pitfalls on the way to the widespread clinical use of cannabis, and additional applications of cannabis-related products.

Abstract Background Cellular senescence, a permanent state of replicative arrest in otherwise proliferating cells, is a hallmark of aging and has been linked to aging-related diseases. Many genes play a role in cellular senescence, yet a comprehensive understanding of its pathways is still lacking. Results We develop CellAge ( http://genomics.senescence.info/cells ), a manually curated database of 279 human genes driving cellular senescence, and perform various integrative analyses. Genes inducing cellular senescence tend to be overexpressed with age in human tissues and are significantly overrepresented in anti-longevity and tumor-suppressor genes, while genes inhibiting cellular senescence overlap with pro-longevity and oncogenes. Furthermore, cellular senescence genes are strongly conserved in mammals but not in invertebrates. We also build cellular senescence protein-protein interaction and co-expression networks. Clusters in the networks are enriched for cell cycle and immunological processes. Network topological parameters also reveal novel potential cellular senescence regulators. Using siRNAs, we observe that all 26 candidates tested induce at least one marker of senescence with 13 genes (C9orf40, CDC25A, CDCA4, CKAP2, GTF3C4, HAUS4, IMMT, MCM7, MTHFD2, MYBL2, NEK2, NIPA2, and TCEB3) decreasing cell number, activating p16/p21, and undergoing morphological changes that resemble cellular senescence. Conclusions Overall, our work provides a benchmark resource for researchers to study cellular senescence, and our systems biology analyses reveal new insights and gene regulators of cellular senescence.

Phlomis viscosa Poiret (an evergreen shrub) represents a valuable source of medicinal compounds. In this study, we discovered compounds with antimicrobial and antiviral properties. The aim of this study was to identify compounds of P. viscosa and estimate the antimicrobial and antiviral activity of its phytochemicals. The volatile compounds were identified using gas chromatography/mass spectrometry (GC/MS) analysis. For the identification of nonvolatile components of the extracts, high-performance liquid chromatography (HPLC), liquid chromatography–electrospray ionization-mass spectrometry (LC-ESI-MS) and matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry (MALDI-TOF-MS) were applied. Quercetin 3-O-rutinoside and hesperidin caused a significant decrease in the bacterial concentration of Agrobacterium tumefaciens, Xylella fastidiosa and Pseudomonas syringae (p < 0.001). The growth of drug-resistant microorganisms (Escherichia coli, Klebsiella pneumoniae, Acinetobacter baumannii, Serratia marcescens and Salmonella enteritidis) was inhibited by quercetin 3-O-rutinoside, quercetin 3-O-arabinoside and hesperidin. In addition, these compounds demonstrated antiquorum-sensing properties. Diosmin, hesperidin and quercetin 3-O-arabinoside significantly inhibited varicella zoster virus (VZV) (p < 0.001). Quercetin 3-O-rutinoside and quercetin 3-O-arabinoside were effective against herpes simplex virus 1 (HSV-1), including mutant strains.

Anchusa strigosa is a widespread weed in Greece, Syria, Turkey, Lebanon, Israel, Jordan, and Iran. The purpose of this study was to identify the phytochemicals of Anchusa strigose and estimate the pro-wound healing (pro-WH) and antimicrobial activities of its active compounds. An identification of volatile compounds was performed by GC/MS analysis; HPLC, LC-ESI-MS, and MALDI-TOF-MS were also applied. Our results demonstrate that two specific combinations of compounds from A. strigosa extract significantly enhanced WH (p < 0.001). Several flavonoids of the plant extract, including quercetin 3-O-rutinoside, kaempferol, kaempferol 3-O-β-rhamnopyranosyl(1→6)-β-glucopyranoside, and kaempferol 3-O-α-rhamnopyranosyl(1→6)-β-galactopyranoside, were effective against drug-resistant microorganisms. In addition, all the above-mentioned compounds had antibiofilm activity against Escherichia coli and Salmonella enteritidis.

Improper land management, such as over-grazing in arid areas, has negative effects on the local ecosystems for both the short and the long term time periods. An effective rehabilitation scheme requires human interference by introducing ecosystem engineering organisms together with activities that encourage the spreading and the reproduction of the local plant and animal species. Most of the former studies in arid lands focused on shrubs as engineering species, and much less on other organisms. The major focus of this study was on assessing the impact of Messor ebeninus and M. arenarius on the micro-topographic patterns of arid areas using unique spatial statistical tools designed solely for this purpose. As a case study, the nests’ sizes and their distribution were compared between two adjacent shrublands with similar geographic outlines during 2008 and 2015.One of the shrublands was moderately grazed for the last 20 years (at the far past it was exposed to over-grazing), while the other one is still exposed to over-grazing. The results collected in 2014 at the shrublands and at the adjacent loess area demonstrate the spatial ecosystem ability of the Messor sp. to engineer and beneficially modify their environment by enlarging the water conserving area, increasing the soil fertility and vegetative productivity, and finally accelerating the whole area rehabilitation.

Lilium candidum L., known as Madonna, meadow, or white lily, is a bulbous plant from the Liliaceae family, originating in the Middle East. L. candidum has been abundantly used in folk medicine since ancient times to relieve a variety of ailments, including age-related diseases, burns, ulcers, and coughs. The aim of this article is to investigate the anti-inflammatory and anti-diabetic activities of L. candidum extracts and its active phytochemicals. Some active volatile phytochemicals were identified using gas chromatography–mass spectrometry (GC-MS) analysis. Significant (p < 0.001) anti-diabetic properties of the extracts kaempferol, linalool, citronellal, and humulene were demonstrated by an elevation in glucose uptake by adipocytes. The significant (p < 0.01) effect of the plant extracts kaempferol, citronellal, and humulene on the secretion of pro-inflammatory cytokines interleukin 6 (IL-6) and interleukin 8 (IL-8) was demonstrated using enzyme-linked immunosorbent assay. Altogether, L. candidum and its rich collection of phytochemicals hold promising medicinal potential, and further investigations of its therapeutic prospects are encouraged.

One of the consequences of soil erosion in arid and semi-arid environments is the emergence of rocky shrublands. While their existence is well documented, gaps exist in our understanding of processes affecting the soil fertility inside them and in their surrounded environment which is crucial for their successive sustainably management. Our aims were as follows: (i) assessing the impact of various parameters (geographical, chemical and physical) on the herbaceous cover in shrublands located in arid rocky areas; (ii) assessing the impact of the rocky topography on the fertility parameters at the inter-patches and surrounded matrix areas. Geographically, the site of study is located in private family farm at Chiran area, northern Negev, Israel. It has semi - arid climate (precipitation 200 mm year-1) with hilly rocky topography. Ecologically the area is sustainably grazed shrubland characterized by patch-matrix patterns.For analyzing the impact of the above mentioned parameters we chose 24 patches belonging to different geographic groups: “Inside the rock”, “Adjacent to rock”, “Parallel to rock” and “Slopped”). From each patch we took soil and herbaceous biomass samples from predefined locations based on the geographical patterns of each group. Four methodologies were implemented for analysis as follows: (i) comparisons between the actual values; (ii) ranking the differences between the patches” sub-plots; (iii) correlating the soil parameters with the herbaceous biomass using regression analysis; (iv) spatial analysis of the different parameters based on kriging. The results demonstrate that the most influential parameter in the plots inside the patch were the Soil Organic Matter and clay content. The soil moisture in this study did not affect the area fertility. The rocky topography, together with the patch spatial patterns had high impact on the values of the examined parameters, even when compared to the surrounding matrix. Altogether, the presented results indicate that the patch fertility is affected by combination of different soil parameters and the geographic topography of a given patch. Additionally, reciprocal effects between the patches and their surrounding environment are also important determinants of fertility. The techniques and methodologies demonstrated here could be applied to other landscapes as well.

Plants growing in the Judea region are widely used in traditional medicine of the Levant region. Nevertheless, they have not so far been sufficiently analyzed and their medicinal potential has not been evaluated. This study is the first attempt to fill the gap in the knowledge of the plants growing in the region. Comprehensive data mining of online botanical databases and peer-reviewed scientific literature including ethno-pharmacological surveys from the Levant region was applied to compile a full list of plants growing in the Judea region, with the focus on their medicinal applications. Around 1300 plants growing in the Judea region were identified. Of them, 25% have medicinal applications which were analyzed in this study. Screening for chemical-protein interactions, together with the network-based analysis of potential targets, will facilitate discovery and therapeutic applications of the Judea region plants. Such an approach could also be applied as an integrative platform for further searching the potential therapeutic targets of plants growing in other regions of the world.

Although different aspects of cancer immunity are a subject of intensive investigation, an integrative view on the possible molecular links between immunoregulators and cancer-associated genes has not yet been fully considered. In an attempt to get more insights on the problem, we analyzed these links from a network perspective. We showed that the immunoregulators could be organized into a miRNA-regulated PPI network-the immunoregulatory network. This network has numerous links with cancer, including (i) cancerassociated immunoregulators, (ii) direct and indirect protein-protein interactions (through the common protein partners), and (iii) common miRNAs. These links may largely determine the interactions between the host's immunity and cancer, supporting the possibility for co-expression and post-transcriptional co-regulation of immunoregulatory and cancer genes. In addition, the connection between immunoregulation and cancer may lie within the realm of cancer-predisposing conditions, such as chronic inflammation and fibroproliferative repair. A gradual, age-related deterioration of the integrity and functionality of the immunoregulaory network could contribute to impaired immunity and generation of cancer-predisposing conditions.

Background & objectives: Trypanosoma lewisi is a common, flagellated parasite of the rat. Ourprevious study showed that rabbits injected with serum collected from rats infected with Trypanosomalewisi and treated with cyclophosphamide (CyI) produced high levels of antibodies against a newprotein in the CyI rat serum.Results: In the present study, this protein was characterised as α2 macroglobulin (α2M) and thekinetics of its production and its influence on the malignancy of the disease were determined. In ratsinfected with T. lewisi, α2M was first demonstrated and peaked on the second day post-infection(972 μg/ml) and then reduced gradually, reaching a level of 32 μg/ml on the eighth day post-infection.However, in the CyI rats the level of α2M was gradually increased as the disease progressed,reaching a level of 890 μg/ml on the eighth day post-infection. Injection of both crude and purifiedα2M into rats infected with T. lewisi led to increased parasitaemia.Interpretation & conclusion: The present study suggests that increased levels of α2M in the CyI ratscontribute to the malignancy of the disease.
