Introduction: Inbreeding depression (ID) in food-deceptive plants have been reported previously, however, it has not been often proven that selfed seeds germinate better than outbred ones or that selfing affects ID. To resolved these issues, food-deceptive related Dactylorhiza majalis, D. incarnata var. incarnata and D. fuchsii orchids were investigated. Methods: Hand pollination treatments and control pollination were conducted. Fruit set, number of seeds per fruit, seed length, number of well-developed seeds per fruit, and proportion of in vitro asymbiotic germination seeds, were analyzed in relation to inflorescence levels and used as fitness indicators for these orchids. The ID and pollen limitation were measured. Results: The lowest ID (d = -1.000) was in D. majalis, and present in combination with a high pollen limitation in its populations. D. fuchsii showed higher ID (d = 0.366), and D. incarnata var. incarnata weak ID (d = 0.065), although ID varied between its fitness components. The seed number per fruit differed significantly between the treatments and the inflorescence levels in treatments. Discussion: This study emphasizes that the breeding system rather than the flower position on the inflorescence shaped the quality and quantity of reproductive output. Plant species evolution is driven by many factors that have different roles in space and time. Using different field and laboratory methods, we studied reproductive patterns and their determinants in pure and mixed P. bifolia and P. chlorantha populations in different habitats. We also considered the probability of hybridisation between these two species and the role of intra-population processes in maintaining species integrity. Generally, we found a high level of reproductive success in both Platantherans. In both species, male (MRS) and female (FRS) reproductive success depended on floral display, and male reproductive success additionally on population structure. The flower traits were only weakly related to reproductive success. Moths' assemblages varied spatially and temporally, and their diversity and numbers were correlated with MRS in the year, when their abundance was markedly lower. Analysis of patterns of pollen transfer showed that pollen was transported up to 25 m (average 8.2 ± 4.83 m) and showed gene exchange between these two Platanthera species. The germination level of both species was significantly lower than seed viability, although P. bifolia seed germinated with higher frequency than P. chlorantha seeds. We noted differences in viability and germination of seeds developed as an effect of experimental interspecies crossings and those developed from natural pollination. The presence of intermediate ecotypes together with observations of spontaneous interspecies crosses in the field and viability of seeds produced in interspecies crossing suggest that both pre- and postzygotic reproductive barriers are not complete and do not prevent hybrid production. Abstract: We carried out translocations of three rare plants that inhabit mineral islands in the marshy Biebrza Valley in order to create alternative populations and facilitate connectivity between existing subpopulations. The species chosen were Iris aphylla and two orchids: Cypripedium calceolus and Cephalanthera rubra. Thirty soil monoliths with vegetative orchid plants or parts of I. aphylla rhizomes were dug out and transplanted to three different sites on new mineral islands (half in 2012 and half in 2013). Prior to translocation, we measured soil moisture and pH and took phytosociological characteristics in 68 potential recipient sites for orchids and 15 for I. aphylla. Then, we monitored the number of shoots for 4 years and retrospectively conducted principal component analysis (PCA) in order to compare the similarity of donor sites and chosen recipient sites. Three years after transplantation we found new C. calceolus populations in good condition, I. aphylla populations in moderate, while C. rubra transplants emerged only in the first and second season. All newly established populations of C. calceolus and I. aphylla survived. In the case of the first species, fruiting was observed in two populations, while a marked increase in shoot number was observed in one population. We investigated the phylogeographical structure of the boreal-montane orchid Malaxis monophyllos in its Eurasian geographical range. We analysed four sequences of plastid DNA ( trn L, trn L- trn F, rps16 and acc D- psa I), resulting in 19 haplotypes and revealing a high level of intraspecific diversity ( HD = 0.702 and π = 0.196 × 10−2), but showing a lack of phylogeographical structure. This pattern might be caused by multiple phenomena and processes, e.g. broad-fronted recolonization with accompanying multi-directional gene flow between populations and expansion from at least two refugial areas. Despite the lack of phylogeographical structure, three centres of haplotype diversity were indicated in the European part of the range of M. monophyllos. According to these data, alpine and lowland glacial refugia located between the ice sheets in the European Alps and the Scandinavian glaciers seem most likely to be in Europe. Moreover, models of climatically suitable areas during the Last Glacial Maximum ( LGM) confirmed the Alps as a possible refuge, and indicated an opportunity for the persistence of M. monophyllos populations in Beringia and parts of Siberia. Using two models [Model for Interdisciplinary Research on Climate ( MIROC) and Community Climate System Model ( CCSM)], we predicted a significant reduction in climatically suitable areas for M. monophyllos in the future (2080). Our study also demonstrated that the biological features of M. monophyllos, including breeding system and dispersal mode, seem to be crucial in understanding its phylogeographical pattern. We examined the genetic diversity of two orchid species, the nectar-rewarding Platanthera chlorantha and the nectarless Cephalanthera rubra, in north-eastern Poland. We found lower differences in genetic diversity between the species than we expected. The level of genetic variation at species level was lower in C. rubra ( PPOL = 14%, A = 1.14 and HE = 0.060) than in P. chlorantha ( PPOL = 25.5%, A = 1.35 and HE = 0.078). In the majority of populations of both species a high proportion of unique genotypes was noted. The overall FST values for all populations were moderate and similar for both species ( P. chlorantha: 0.251, P < 0.001; C. rubra: 0.267, P < 0.001). No relationship was found between genetic and geographical distances in either species ( P > 0.05, Mantel test). We discuss the breeding systems, small population size and population subdivision as the most important factors affecting the genetic diversity of this species. We suggest that conservation programmes should be initiated to maintain or even increase the fitness and genetic variation of populations of both species. © 2013 The Linnean Society of London [ABSTRACT FROM AUTHOR]
