Since pollen characters can be used to help distinguish species, our aim was to determine if palynological information has taxonomic significance for Gagea species from Xinjiang, China. Gagea is widely distributed in north temperate and the subtropical zones. The genus has limited taxonomic characteristics and large morphological variation, which results in difficulty of species classification. Pollen morphology of 16 species of this genus was examined comprehensively via light microscope (LM) and scanning electron microscope (SEM). One qualitative and nine quantitative traits of the pollen grains were surveyed, followed by hierarchical cluster analysis (HCA). The pollen grains were bilaterally symmetrical heteropolar monads with a mono-sulcus and they were oblate or peroblate (Polar diameter (P) / Equatorial diameter (E) = 0.36–0.73) in shape and medium to large (P = 17.17–34.64 μm, E = 27.63–81.65 μm) in size. Three types of exine ornamentation were observed: perforate, microreticulate and reticulate cristatum. The HCA divided the 16 species into two groups. This research provides new data on pollen morphology for Gagea (the pollen morphology of eight species was reported for the first time). Pollen morphology also can be used to identify species with similar external morphology, such as G. nigra and G. filiformis. Furthermore, the study of pollen morphology not only provides new data for palynology research on Gagea, but also provides a basis for future classification of this genus.

Plagiothecium mauiense was first described in 1927 by V.F. Brotherus, based on materials from Hawaii. It has, so far been, treated as a separate species. A detailed analysis of the original material housed in the New York Botanical Garden Herbarium (NY01256708) found the specimen to be characterised by a lack of metallic lustre; concave, asymmetrical, lanceolate to lanceolate-ovate leaves, shrunken in their dry condition; a straight, not denticulate, acute to apiculate apex; elongate-hexagonal cells in irregular transverse rows, 101–131 × 15–21 µm at mid-leaf; very lax areolation, with decurrencies composed of three rows of cells. These characteristics indicate that this species is identical to the original material of P. longisetum (e.g. H-SOL 1563 011; PC0132572). Hence, we propose that P. mauiense should be recognised as a new synonym of P. longisetum. In addition, a review of P. longisetum syntypes found one (H-SOL 1563 011) to have the same date of collection as the protologue, and to possess a quite abundant gametophyte turf with well-preserved sporophytes, indicating it to be fertile. Considering the above, we propose that specimen H-SOL 1563 011 be designated the lectotype of P. longisetum.

In 1859, William Mitten described Stereodon nemoralis (≡ Plagiothecium nemorale) based on the gathering of Sir J.D. Hooker from India. However, the protologue did not indicate any specific specimen or illustration. For the past 50 years, the original material (NY 913349) deposited at the NY Herbarium has been considered as the holotype. However, this assumption has since been found to be incorrect, because in the Herbarium of The Natural History Museum exists other original material of this species (BM 1030713), collected by Hooker. In addition, the specimen from NY Herbarium is in poor condition and its most important diagnostic characters are not visible. In contrast, the material from BM Herbarium is in very good condition, and therefore it is herein designated as the lectotype. Also, the paper describes the resolution of this type, a process complicated by changes that had occurred in the provisions of subsequent botanical Codes.

The present study reports the complete mitochondrial genome of Myurella julacea (Schwägr.) Schimp. (GenBank accession number MT850126); the genome size was 104,979 bp. The gene arrangement was found to be similar to that in other bryophytes, and the genome consisted of 40 protein-coding genes (PCGs), 3 ribosomal RNAs (rRNAs), and 24 transfer RNAs (tRNAs). The phylogenic relationship was analyzed by construction of a phylogenetic tree based on the mitogenome of M. julacea and 25 other bryophytes publicly available in GenBank. The complete mitogenome of M. julacea is expected to provide insights into the evolution of species belonging to the order Hypnales.

Mosses can be used as biomonitors to monitor radionuclide deposition and heavy metal pollution in cities, forests, and grasslands. The aims of this work were to determine the activity concentrations of natural (210Po, 210Pb or 210Pbex (excess 210Pb is defined as the activity of 210Pb minus the activity of 226Ra), 7Be, 40K, 226Ra, 238U, and 232Th) and anthropogenic radionuclides (137Cs) in moss body profiles and in situ underlying soils of moss samples and to assess/determine the distribution features and accumulation of these radionuclides. Activity concentrations of radionuclides in the samples were measured using a low-background gamma spectrometer and a low-background alpha spectrometer. Consistent with their source, the studied radionuclides in the moss samples and underlying soils were divided according to the principal component analysis (PCA) results into an airborne group (210Po, 210Pb (210Pbex), 7Be, and 137Cs) and a terrestrial group (40K, 238U, 226Ra, and 232Th). The activity concentrations of 210Po and 210Pbex in moss body profiles were mainly concentrated in the stems–rhizoid parts, in which we measured some of the highest 210Po and 210Pbex levels compared to the results in the literature. 7Be mainly accumulated in the leaves–stem parts. Different positive correlations were observed between 210Po and 210Pb and between 7Be and 210Pb, which indicated that the uptake mechanisms of 210Po, 210Pb, and 7Be by moss plants were different, to some extent. 137Cs was detected only in some moss samples, and the fraction of 137Cs in the underlying soils was much lower than that in the moss, suggesting that mosses were protecting the underlying soils from further pollution. Except for 40K, the terrestrial radionuclide (238U, 226Ra, and 232Th) content in mosses was predominantly at low levels, which indicated not only the inability of mosses to use those elements for metabolic purposes but also the rather poor capability of mosses to directly mobilize, absorb, and transport elements (U, Ra, or Th) not dissolved in water.

Transacting siRNA loci (TAS3-like) of a particular plant species are usually represented by several gene families. PCR-based approach was used as a phylogenetic profiling tool to probe genomic DNA samples from representatives of evolutionary distant Bryophyta taxa, namely, class Bryopsida (subclasses Bryidae and Dicranidae) and class Sphagnopsida. We found relatives of all four Physcomitrella patens (subclass Funariidae) TAS3-like loci in subclasses Bryidae and Dicranidae. Only representatives of subclass Bryidae encoded TAS3-like genes belonging to P. patens TAS3a and TAS3d families. On the other hand, only the members of order Grimmiales (subclass Dicranidae) encoded gene relatives of P. patens TAS3c family. These data indicate that moss ta-siRNA families have been long conserved during land plant evolution. However, P. patens TAS3-like loci were detected neither in two Sphagnum species from the earliest diverged moss class Sphagnopsida, nor in the Selaginella kraussiana from the earliest extant tracheophyta lineage, Lycopodiopsida.

The diplolepideous-alternate peristome, when most highly developed, has endostome segments attached to a basal membrane and positioned alternate to the outer exostome teeth, with cilia often present between the segments. This peristome type defines the Bryidae ( sensu Vitt et al. 1998), which includes four orders: the Bryales, Leucodontales, Hypnales, and Hookeriales, of which the latter three are mainly pleurocarpous in their growth form. Chloroplast (rbcL, rps4, and trnL-trnF) and nuclear (18S rRNA) gene sequences have been analyzed using the parsimony optimality criterion to elucidate relationships among the Bryidae. The analyses strongly support the paraphyly of the Bryidae, with the Splachnidae, and possibly the Orthotrichidae, having arisen from ancestors within the Bryidae. The Leucodontales, Hypnales, and Hookeriales form a monophyletic group, as do the pleurocarpous members of the Bryidae. However, the two pleurocarpous clades are not resolved as sister groups, although their non-monophy...

In contrast to animals, the slowly evolving mitochondrial nucleotide sequences of plants appear well suited to investigate phylogenetic relations between old taxonomic groups. Analysis ofnad5 gene sequences in 47 bryophytes, the living representatives of very early land plants, confirm this assessment. Statistically reliable phylogenetic trees are obtained with different mathematical approaches. A group I intron sequence conserved in thenad5 gene of all 30 mosses and 15 liverworts investigated supports a sister group relationship of the two classes. The intron sequence adds phylogenetic information for fine resolution on top of the conserved exon sequences down to the level of classically defined orders or families, respectively. This intron is not present in the hornwortsAnthoceros husnotii andA. punctatus. The results allow statements on diverging taxonomic interpretations and support the monophyly of the liverworts, mosses, Jungermanniidae, Marchantiidae and Bryidae, and allow recognition of subclasses like Hypnanae and Dicrananae. Among the mosses, the derived orders (subclass Bryidae) are confidently set apart from the Sphagnales, Andreaeales, Polytrichales and Tetraphidales with Buxbaumiales occupying a mediating position. Among the liverworts, full support is found for the classic separation of simple (jungermanniid) and complex thalloid (marchantiid) species with a strikingly low mitochondrial sequence divergence among the latter.

This study describes (i) spore morphology of 121 Alaskan species of true moss (Bryidae) and (ii) percentages of Bryidae and Sphagnum spores in mud-water interface samples from 46 Alaskan lakes and a late-Quaternary sediment core from Idavain Lake in southwestern Alaska. Bryidae spores were collected from herbarium specimens, prepared by standard palynological chemical treatment and described under light microscopy. An identification key based on spore shape, wall structure, and size recognized 11 major morphological groups. Although family, genus, or species identifications were not possible for most groups, spores of a few genera and species were distinctive. Bryidae spores occurred in 80% of the modern sediment samples, representing boreal forest and three areas of tundra (North Slope, Seward Peninsula, and southwestern Alaska). Bryidae and Sphagnum spore percentages showed greatest differences between the North Slope tundra and other vegetation types, whereas pollen percentages distinguished boreal forest from tundra regions as a group. Bryidae spores were present throughout the Idavain Lake record but were most common in late-glacial sediments. Variations in the abundance of Bryidae spores are consistent with or enhance paleoenvironmental interpretations based on pollen and other spore types.Key words: Bryidae, palynology, spores, Alaska, Quaternary, paleoenvironment.

The bioaccumulation of Sb, Sn, As, Fe, Ni, Mn, Pb, Cu, and Zn in mosses from polluted and unpolluted areas of the southern parts of Nigeria is presented. Results of the moss samples show strong variations in the concentration of metal ions with type of sampling sites. The concentration of accumulated heavy metals also differs in various moss species from the same biotope. Bryidae mnium is a better accumulator of heavy metals than Tetraphidae tetraphis, as evidenced from the interspecies calibration between the two moss samples.

The fractions of apoplasmic water of six moss species were estimated by comparison of the osmotic potentials of fully turgid living tissues (Ψπ(s)) and of killed shoots (Ψπ(k)). The values of Ψπ(s) were determined by pressure-volume analysis using thermocouple hygrometry, whereas those of Ψπ(k) were obtained by cryoscopy using extracts produced from dried materials and taking into consideration the original saturation water content of the shoots. Most of the tissues had Ψπ(s) values around-1 to -1.5 MPa and non-osmotic water fractions of roughly 20% of the total water content at full turgor. Quantitative analysis of a number of osmotically active cell constituents showed that about one-third of the osmotic potential resulted from the accumulation of sugars. The total free amino acids accounted for about 15–20% of the osmolality of the cell solution. Malic- and citricacid contents varied much more with species but, in general, both carboxylic acids together contributed nearly 10–20% to the osmotic potential. The contents of inorganic anions such as free chloride and phosphate in mosses were low.

We have investigated the presence of auxin and the ability of chloronema cells to synthesize indole-3-acetic acid (IAA) in axenic protonema cell cultures of the moss Funaria hygrometrica. The endogenous level of auxin activity was 4 and 7μg-IAA equivalents/kg in caulonema and chloronema cell types, respectively. Based on an indole-α-pyrone fluorometric assay, the level of putative IAA was observed to be 5.0 and 1.9.μg/kg in caulonema and chloronema cells, respectively. [3H]Tryptophan was metabolized into IAA via the indole-pyruvate pathway by intact chloronema cells and also by the cell free homogenates. More [3H]IAA accumulated when homogenates from cells pre-grown at low cell densities ( 0.5 mg/ml) were used. Since the activities of peroxidase and IAA-oxidase are known to be high at high cell densities, the lack of accumulation of radioactivity in IAA at high densities can be attributed to a high level of IAA-oxidizing enzymes. Our results suggest a possible relationship between IAA accumulation and caulonema differentiation.

High affinity cAMP phosphodiesterase (PDE) activity has been purified ( ca 474-fold) from the chloronema cells of the moss Funaria hygrometrica . The activity in the 15 000 g supernatant was precipitated at 40–80% saturation with (NH 4 ) 2 SO 4 and separated by DEAE-cellulose chromatography into two peaks of activity A and B with pH optima at pH 5.5 and 7.5, respectively. When rechromatographed on DEAE-cellulose, the PDE B activity redistributed into two peaks (A′and B′). The recovery of PDE activity in the latter peaks suggests a conversion of alkaline PDE into acid PDE. The phosphodiesterase activity B was further resolved by affinity chromatography on a cAMP—agarose column into PDE I and PDE II. The latter was free from nucleotidases, was Ca 2+ -dependent and showed only a high-affinity component towards cAMP as the substrate, the K m being 11.8 ± 4.8 μM.
