We introduce a method utilizing single laser-generated cavitation bubbles to stimulate cellular mechanotransduction in dermal fibroblasts embedded within 3D hydrogels. We demonstrate that fibroblasts embedded in either amorphous or fibrillar hydrogels engage in Ca2+ signaling following exposure to an impulsive mechanical stimulus provided by a single 250 µm diameter laser-generated cavitation bubble. We find that the spatial extent of the cellular signaling is larger for cells embedded within a fibrous collagen hydrogel as compared to those embedded within an amorphous polyvinyl alcohol polymer (SLO-PVA) hydrogel. Additionally, for fibroblasts embedded in collagen, we find an increased range of cellular mechanosensitivity for cells that are polarized relative to the radial axis as compared to the circumferential axis. By contrast, fibroblasts embedded within SLO-PVA did not display orientation-dependent mechanosensitivity. Fibroblasts embedded in hydrogels and cultured in calcium-free media did not show cavitation-induced mechanotransduction; implicating calcium signaling based on transmembrane Ca2+ transport. This study demonstrates the utility of single laser-generated cavitation bubbles to provide local non-invasive impulsive mechanical stimuli within 3D hydrogel tissue models with concurrent imaging using optical microscopy. STATEMENT OF SIGNIFICANCE: Currently, there are limited methods for the non-invasive real-time assessment of cellular sensitivity to mechanical stimuli within 3D tissue scaffolds. We describe an original approach that utilizes a pulsed laser microbeam within a standard laser scanning microscope system to generate single cavitation bubbles to provide impulsive mechanostimulation to cells within 3D fibrillar and amorphous hydrogels. Using this technique, we measure the cellular mechanosensitivity of primary human dermal fibroblasts embedded in amorphous and fibrillar hydrogels, thereby providing a useful method to examine cellular mechanotransduction in 3D biomaterials. Moreover, the implementation of our method within a standard optical microscope makes it suitable for broad adoption by cellular mechanotransduction researchers and opens the possibility of high-throughput evaluation of biomaterials with respect to cellular mechanosignaling.

The efficient operation of solid oxide fuel cells for renewable energy generation requires solid electrolytes with high ionic conductivity, thermal stability, and chemical durability. Metal oxide perovskites of the form ABO3 are promising candidates, especially when doped with cations that incorporate interstitial oxygen or oxygen vacancies to enhance ionic conductivity. However, doping can lead to complex and poorly understood crystallographic structures. In this study, we investigate the effects of Fe3+ doping on the orthorhombic Pbnm CaTiO3 perovskite, forming Ca(Ti0.8Fe0.2)O3−δ, and demonstrate the complexity of its phase transitions at operational temperatures. Using synchrotron X-ray diffraction and thermogravimetric analysis, we show that this material undergoes significant oxygen uptake at elevated temperatures, leading to an irreversible expansion of the unit cell and changes in polyhedral coordination upon cooling. These structural modifications are attributed to the partial oxidation of Fe3+ to Fe4+, providing insight into the inconsistencies observed in previous studies of oxygen-deficient perovskites. Our findings highlight the critical role of thermal history and oxygen availability in determining the structural stability and long-term performance of perovskite-based solid electrolytes in solid oxide fuel cells.

Background:. This study assesses the effectiveness of large language models (LLMs) in simplifying complex language within orthopaedic patient education materials (PEMs) and identifies predictive factors for successful text transformation. Methods:. We transformed 48 orthopaedic PEMs using GPT-4, GPT-3.5, Claude 2, and Llama 2. The readability, quantified by the Flesch-Kincaid Reading Ease (FKRE) and Flesch-Kincaid Grade Level (FKGL) scores, was measured before and after transformation. Analysis included text characteristics such as syllable count, word length, and sentence length. Statistical and machine learning methods evaluated the correlations and predictive capacity of these features for transformation success. Results:. All LLMs improved FKRE and FKGL scores (p < 0.01). GPT-4 showed superior performance, transforming PEMs to a seventh-grade reading level (mean FKGL, 6.72 ± 0.99), with higher FKRE and lower FKGL than other models. GPT-3.5, Claude 2, and Llama 2 significantly shortened sentences and overall text length (p < 0.01). Importantly, correlation analysis revealed that transformation success varied substantially with the model used, depending on original text factors such as word length and sentence complexity. Conclusions:. LLMs successfully simplify orthopaedic PEMs, with GPT-4 leading in readability improvement. This study highlights the importance of initial text characteristics in determining the effectiveness of LLM transformations, offering insights for optimizing orthopaedic health literacy initiatives using artificial intelligence (AI). Clinical Relevance:. This study provides critical insights into the ability of LLMs to simplify complex orthopaedic PEMs, enhancing their readability without compromising informational integrity. By identifying predictive factors for successful text transformation, this research supports the application of AI in improving health literacy, potentially leading to better patient comprehension and outcomes in orthopaedic care.

The potential influences of turbulence on planktonic processes such as nutrient uptake, grazing, predation, infection, and mating have been explored in hundreds of laboratory and theoretical studies. However, the turbulence levels used may not represent those experienced by oceanic plankton, bringing into question their relevance for understanding planktonic dynamics in the ocean. Here, we take a data-centric approach to understand the turbulence climate experienced by plankton in the ocean, analyzing over one million turbulence measurements acquired in the open ocean. Median dissipation rates in the upper 100 m were < 10−8 W kg−1, with 99% of the observations < 10−6 W kg−1. Below mixed layers, the median dissipation rate was ~ 10−10 W kg−1, with 99% of the observations < 10−7 W kg−1. Even in strongly mixing layers the median dissipation rates rarely reached 10−5 W kg−1, decreasing by orders of magnitude over 10 m or less in depth. Furthermore, episodes of intense turbulence were transient, transitioning to background levels within 10 min or less. We define three turbulence conditions in the ocean: weak (< 10−8 W kg−1), moderate (10−8–10−6 W kg−1), and strong (> 10−6 W kg−1). Even the strongest of these is much weaker than those used in most laboratory experiments. The most frequent turbulence levels found in this study are weak enough for most plankton—including small protists—to outswim them, and to allow chemical plumes and trails to persist for tens of minutes. Our analyses underscore the primary importance of planktonic behavior in driving individual interactions.

Abstract Misfolded protein aggregates may cause toxic proteinopathy, including autosomal dominant tubulointerstitial kidney disease due to uromodulin mutations (ADTKD-UMOD), a leading hereditary kidney disease. There are no targeted therapies. In our generated mouse model recapitulating human ADTKD-UMOD carrying a leading UMOD mutation, we show that autophagy/mitophagy and mitochondrial biogenesis are impaired, leading to cGAS-STING activation and tubular injury. Moreover, we demonstrate that inducible tubular overexpression of mesencephalic astrocyte-derived neurotrophic factor (MANF), a secreted endoplasmic reticulum protein, after the onset of disease stimulates autophagy/mitophagy, clears mutant UMOD, and promotes mitochondrial biogenesis through p-AMPK enhancement, thus protecting kidney function in our ADTKD mouse model. Conversely, genetic ablation of MANF in the mutant thick ascending limb tubular cells worsens autophagy suppression and kidney fibrosis. Together, we have discovered MANF as a biotherapeutic protein and elucidated previously unknown mechanisms of MANF in the regulation of organelle homeostasis, which may have broad therapeutic applications to treat various proteinopathies.

Abstract Type 2 diabetes (T2D) and its complications can have debilitating, sometimes fatal consequences for afflicted individuals. The disease can be difficult to control, and therapeutic strategies to prevent T2D-induced tissue and organ damage are needed. Here we describe the results of administering a potent and selective inhibitor of Protein Kinase C (PKC) family members PKCα and PKCβ, Cmpd 1, in the ZSF1 obese rat model of hyperphagia-induced, obesity-driven T2D. Although our initial intent was to evaluate the effect of PKCα/β inhibition on renal damage in this model setting, Cmpd 1 unexpectedly caused a marked reduction in the hyperphagic response of ZSF1 obese animals. This halted renal function decline but did so indirectly and indistinguishably from a pair feeding comparator group. However, above and beyond this food intake effect, Cmpd 1 lowered overall animal body weights, reduced liver vacuolation, and reduced inguinal adipose tissue (iWAT) mass, inflammation, and adipocyte size. Taken together, Cmpd 1 had strong effects on multiple disease parameters in this obesity-driven rodent model of T2D. Further evaluation for potential translation of PKCα/β inhibition to T2D and obesity in humans is warranted.

Abstract The intestinal epithelium comprises diverse cell types and executes many specialized functions as the primary interface between luminal contents and internal organs. A key function provided by the epithelium is maintenance of a barrier that protects the individual from pathogens, irritating luminal contents, and the microbiota. Disruption of this barrier can lead to inflammatory disease within the intestinal mucosa, and, in more severe cases, to sepsis. Animal models to study intestinal permeability are costly and not entirely predictive of human biology. Here we present a model of human colon barrier function that integrates primary human colon stem cells into Draper’s PREDICT96 microfluidic organ-on-chip platform to yield a high-throughput system appropriate to predict damage and healing of the human colon epithelial barrier. We have demonstrated pharmacologically induced barrier damage measured by both a high throughput molecular permeability assay and transepithelial resistance. Using these assays, we developed an Inflammatory Bowel Disease-relevant model through cytokine induced damage that can support studies of disease mechanisms and putative therapeutics.

We demonstrate an interferometric method to provide direct, single-shot measurements of cavitation bubble dynamics with nanoscale spatial and temporal resolution with results that closely match theoretical predictions. Implementation of this method reduces the need for expensive and complex ultra-high speed camera systems for the measurement of single cavitation events. This method can capture dynamics over large time intervals with sub-nanosecond temporal resolution and spatial precision surpassing the optical diffraction limit. We expect this method to have broad utility for examination of cavitation bubble dynamics, as well as for metrology applications such as optorheological materials characterization. This method provides an accurate approach for precise measurement of cavitation bubble dynamics suitable for metrology applications such as optorheological materials characterization.

Considerable clinical evidence supports that increased blood-brain barrier (BBB) permeability is linked to immune extravasation of CNS parenchyma during neuroinflammation. Although BBB permeability and immune extravasation are known to be provoked by vascular endothelial growth factor-A (i.e., VEGF-A) and C-X-C motif chemokine ligand 12 (CXCL12), respectively, the mechanisms that link both processes are still elusive. The interleukin-20 (i.e., IL-20) cytokine signaling pathway was previously implicated in VEGF-mediated angiogenesis and is known to induce cellular response by way of signaling through IL-20 receptor subunit β (i.e., IL-20RB). Dysregulated IL-20 signaling is implicated in many inflammatory pathologies, but it's contribution to neuroinflammation has yet to be reported. We hypothesize that the IL-20 cytokine, and the IL cytokine subfamily more broadly, play a key role in CNS neuroinflammation by signaling through IL-20RB, induce VEGF activity, and enhance both BBB-permeability and CXCL12-mediated immune extravasation. To address this hypothesis, we actively immunized IL-20RB-/- mice and wild-type mice to induce experimental autoimmune encephalomyelitis (EAE) and found that IL-20RB-/- mice showed amelioration of disease progression compared to wild-type mice. Similarly, we passively immunized IL-20RB-/- mice and wild-type mice with myelin-reactive Th1 cells from either IL-20RB-/- and wild-type genotype. Host IL-20RB-/- mice showed lesser disease progression than wild-type mice, regardless of the myelin-reactive Th1 cells genotype. Using multianalyte bead-based immunoassay and ELISA, we found distinctive changes in levels of pro-inflammatory cytokines between IL-20RB-/- mice and wild-type mice at peak of EAE. We also found detectable levels of all cytokines of the IL-20 subfamily within CNS tissues and specific alteration to IL-20 subfamily cytokines IL-19, IL-20, and IL-24, expression levels. Immunolabeling of CNS region-specific microvessels confirmed IL-20RB protein at the spinal cord microvasculature and upregulation during EAE. Microvessels isolated from macaques CNS tissues also expressed IL-20RB. Moreover, we identified the expression of all IL-20 receptor subunits: IL-22 receptor subunit α-1 (IL-22RA1), IL-20RB, and IL-20 receptor subunit α (IL-20RA) in human CNS microvessels. Notably, human cerebral microvasculature endothelial cells (HCMEC/D3) treated with IL-1β showed augmented expression of the IL-20 receptor. Lastly, IL-20-treated HCMEC/D3 showed alterations on CXCL12 apicobasal polarity consistent with a neuroinflammatory status. This evidence suggests that IL-20 subfamily cytokines may signal at the BBB via IL-20RB, triggering neuroinflammation.

We introduce laser cavitation rheology (LCR) as a minimally-invasive optical method to characterize mechanical properties within the interior of biological and synthetic aqueous soft materials at high strain-rates. We utilized time-resolved photography to measure cavitation bubble dynamics generated by the delivery of focused 500 ps duration laser radiation at λ = 532 nm within fibrin hydrogels at pulse energies of Ep = 12, 18 µJ and within polyethylene glycol (600) diacrylate (PEG (600) DA) hydrogels at Ep = 2, 5, 12 µJ. Elastic moduli and failure strains of fibrin and PEG (600) DA hydrogels were calculated from these measurements by determining parameter values which provide the best fit of the measured data to a theoretical model of cavitation bubble dynamics in a Neo-Hookean viscoelastic medium subject to material failure. We demonstrate the use of this method to retrieve the local, interior elastic modulus of these hydrogels and both the radial and circumferential failure strains.

Abstract Diverse phytoplankton exude polysaccharides that can form a viscous mucus layer surrounding their cells, facilitating complex chemical exchanges with bacteria. An unexplored ramification of mucus production is the influence of its viscosity on the diffusion of nutrients such as vitamins to the cell. Here, I use simulations to demonstrate that mucus viscosity increases the nutrient concentration gradient but always reduces the flux to the cell. Uptake is marginally improved during nutrient pulses, as the mucus acts like a sponge by retaining nutrients near the cell. Lower uptake in the presence of mucus presents a fitness cost that any ecological benefits of mucus must outweigh. I derive a relationship between mucus viscosity and nutrient uptake that can be used to test hypotheses of mutualistic interactions between phytoplankton and bacteria. This work emphasizes the need for empirical measurements of macronutrient diffusion through mucus layers and provides a framework for interpreting those results.

Increasing depictions of kink in mainstream society are leading to a rise in people seeking to learn about and participate in bondage/discipline, domination/submission, and sadism/masochism (BDSM). For many, this initial education process is likely to focus on the Internet and pornography specifically. Existing research into the impact of pornography on sex education has been mixed, though predominantly negative. However, pornography's role in BDSM education has yet to be explored. To address this gap, 18 semi-structured interviews were conducted with BDSM community leaders in Canada and the USA to examine from where and what practitioners learn about BDSM. Participants drew upon their personal experiences and those they interacted with in their capacity as a community leader. Interviewees identified the benefits and risks associated with using pornography as a learning tool for BDSM participation. They stressed how pornography's value as a learning tool has evolved over time, how it can normalise BDSM and increase self-acceptance, teach skills, and reinforce negotiation and consent practices. However, if consumers cannot separate fantasy from reality, pornography can have negative effects. The impact of BDSM-themed pornography for education, especially in adolescents and young people, is discussed, as are directions for more in-depth investigations of this topic. A detailed variable temperature neutron total scattering study of the potential nuclear waste matrix TlTcO4 was conducted. The long-range average structure of TlTcO4 undergoes an orthorhombic Pnma to tetragonal I41/amd phase transition below 600 K, consistent with previous synchrotron X-ray diffraction studies. However, several anomalies were observed in the Rietveld refinements to the neutron powder diffraction data, such as large atomic displacement parameters at low temperature and a shortening of the Tc-O bond distance upon heating. Modelling the short-range local structure of both the low- and high-temperature data required a lowering of symmetry to the monoclinic P21/c model due to the stereochemical activity of the Tl+ 6s2 lone pairs. Density functional theory calculations also verified this model to have a lower ground state energy than the corresponding long-range average structure. It is concluded that at low temperatures, the Tl+ 6s2 lone pairs are 'frozen' into the structure. Upon heating, the rigid TcO4 tetrahedra begin to rotate, as governed by the ...3+ and M4+ modes. However, there is a disconnect between the two length scales, with the 6s² lone pair electrons remaining stereochemically active on the local scale, as observed in the neutron pair distribution function fits. The orthorhombic Pnma to tetragonal I41/amd phase transition is seemingly the result of a change in the correlation length of the Tl+ 6s² lone pairs, leading to a larger unit cell volume due to their uncorrelated displacements. Fundamental marine ecosystem dynamics such as mating, predation, and infection require individual plankton to move relative to one another. Ambient turbulence is often invoked as a mechanism to facilitate such interactions. The local intensity of turbulence is quantified as the dissipation rate of turbulent kinetic energy. While the dissipation rate is central to understanding large‐scale fluxes of heat, salt, and nutrients in the ocean, we show that it can be a poor descriptor of the turbulent environment experienced by individual plankton. A dissipation rate is a single integrated quantity representing all the complex motions in a turbulent region; the instantaneous turbulent environment of plankton may bear little resemblance to that predicted by the dissipation rate or quantities derived from it. Most importantly, the statistics (probabilities) of the relative motions of plankton in turbulence cannot be recovered from the dissipation rate or its spectrum: the probabilities of the plankton experiencing any given turbulent relative velocity are lost in the calculation. This presents a fundamental barrier to our understanding of the effects of ambient turbulence on planktonic ecosystem dynamics in the ocean. Rather than relying on dissipation rates, we show that quantifying the probability distributions of the microscale turbulent motions can provide much richer insights into the turbulent environment of individual plankton. Expanding such statistical analyses, and improving our understanding of the Lagrangian properties of ocean turbulence as experienced by plankton in the ocean will lead to significant increases in our ability to understand and quantify the effects of turbulence on plankton. Potato virus Y (PVY, Potyviridae) is among the most important viral pathogens of potato. The potato resistance gene Nytbr confers hypersensitive resistance to the ordinary strain of PVY (PVYO), but not the necrotic strain (PVYN). Here, we unveil that residue 247 of PVY helper component proteinase (HCPro) acts as a central player controlling Nytbr strain‐specific activation. We found that substituting the serine at 247 in the HCPro of PVYO (HCProO) with an alanine as in PVYN HCPro (HCProN) disrupts Nytbr recognition. Conversely, an HCProN mutant carrying a serine at position 247 triggers defence. Moreover, we demonstrate that plant defences are induced against HCProO mutants with a phosphomimetic or another phosphorylatable residue at 247, but not with a phosphoablative residue, suggesting that phosphorylation could modulate Nytbr resistance. Extending beyond PVY, we establish that the same response elicited by the PVYO HCPro is also induced by HCPro proteins from other members of the Potyviridae family that have a serine at position 247, but not by those with an alanine. Together, our results provide further insights in the strain‐specific PVY resistance in potato and infer a broad‐spectrum detection mechanism of plant potyvirus effectors contingent on a single amino acid residue. The thermal transformation of the tetragonal‐zircon (tz‐) to tetragonal‐scheelite (ts‐)BiVO4 was studied by in situ synchrotron X‐ray diffraction, thermogravimetric analysis, and Fourier‐transformed infrared spectroscopy. Upon heating, the tetragonal zircon polymorph of BiVO4 (tz‐BiVO4) transitioned to the ts‐polymorph between 693–773 K. Above 773 K, single phase ts‐BiVO4 was observed before transitioning to the monoclinic fergusonite (mf‐) polymorph upon cooling. An anomaly in thermal expansion was observed between 400–500 K, associated with the loss of intercalated H2O/NH4+ from the coprecipitation procedure. Heating tz‐BiVO4 resulted in contraction of the V−O bond distance and VO4 polyhedra volume, ascribed to rotation of the tetrahedra groups. Attempts to study this by neutron diffraction failed due to the large incoherent scatter from the hydrogenous species. Efforts to remove these species while maintaining the tz‐BiVO4 structure were unsuccessful, suggesting they play a role in stabilizing the tz‐polymorph. The local structure of both mf‐BiVO4 and tz‐BiVO4 were investigated by X‐ray pair distribution function analysis, revealing local distortions. [ABSTRACT FROM AUTHOR]
