Characterization of honeys produced in the region of Djelfa steppe land in Algeria. Description of the subject. This paper deals with the quality of honeys produced in the steppe areas of Algeria and discusses the possibility of their valorization. Objectives. The objective was to characterize and compare the physical and chemical quality of honeys and to determine their pollen composition according to their geographical origin in three areas of the Djelfa semi-arid region of Algeria. Method. Thirty-eight samples of honey produced in 11 localities in the north, centre and south of the Djelfa semi-arid steppe region were harvested in July for two consecutive years. Pollen analyses were performed and the following properties of the honey samples were determined: water content, pH level, electrical conductivity, color, hydroxymethylfurfural content, saccharase index, diastase index and carbohydrate profile. Results. The results of the pollen analyses identified 34 taxa of pollen. The most abundant pollens were from the Ziziphus lotus, which were present in 97.12% of the samples. The pollens from this shrub were dominant in 27 of the honey samples tested, with a pollen percentage of greater than 45%. Over 55% of the pollen frequencies found came from plants belonging to Asteracae, Brassicaceae, Cistaceae and more particularly to Euphorbia bupleuroides, Peganum harmala, Thapsia garganica, Echium sp. and Retama retam. Conclusions. The physicochemical parameters of the honey samples studied comply with European and international quality standards, which opens up perspectives for their valorization.

Background: Congenital mirror movements (CMM) is a rare neurodevelopmental disorder characterized by involuntary movements from one side of the body that mirror voluntary movements on the opposite side. To date, five genes have been associated with CMM, namely DCC, RAD51, NTN1, ARHGEF7, and DNAL4., Objective: The aim of this study is to characterize the genetic landscape of CMM in a large group of 80 affected individuals., Methods: We screened 80 individuals with CMM from 43 families for pathogenic variants in CMM genes. In large CMM families, we tested for presence of pathogenic variants in multiple affected and unaffected individuals. In addition, we evaluated the impact of three missense DCC variants on binding between DCC and Netrin-1 in vitro., Results: Causal pathogenic/likely pathogenic variants were found in 35% of probands overall, and 70% with familial CMM. The most common causal gene was DCC, responsible for 28% of CMM probands and 80% of solved cases. RAD51, NTN1, and ARHGEF7 were rare causes of CMM, responsible for 2% each. Penetrance of CMM in DCC pathogenic variant carriers was 68% and higher in males than females (74% vs. 54%). The three tested missense variants (p.Ile164Thr; p.Asn176Ser; and p.Arg1343His) bind Netrin-1 similarly to wild type DCC., Conclusions: A genetic etiology can be identified in one third of CMM individuals, with DCC being the most common gene involved. Two thirds of CMM individuals were unsolved, highlighting that CMM is genetically heterogeneous and other CMM genes are yet to be discovered. © 2024 The Authors. Background and Objectives: Somatic and germline pathogenic variants in genes of the mammalian target of rapamycin (mTOR) signaling pathway are a common mechanism underlying a subset of focal malformations of cortical development (FMCDs) referred to as mTORopathies, which include focal cortical dysplasia (FCD) type II, subtypes of polymicrogyria, and hemimegalencephaly. Our objective is to screen resected FMCD specimens with mTORopathy features on histology for causal somatic variants in mTOR pathway genes, describe novel pathogenic variants, and examine the variant distribution in relation to neuroimaging, histopathologic classification, and clinical outcomes., Methods: We performed ultra-deep sequencing using a custom HaloPlex HS Target Enrichment kit in DNA from 21 resected fresh-frozen histologically confirmed FCD type II, tuberous sclerosis complex, or hemimegalencephaly specimens. We mapped the variant alternative allele frequency (AAF) across the resected brain using targeted ultra-deep sequencing in multiple formalin-fixed paraffin-embedded tissue blocks. We also functionally validated 2 candidate somatic MTOR variants and performed targeted RNA sequencing to validate a splicing defect associated with a novel DEPDC5 variant., Results: We identified causal mTOR pathway gene variants in 66.7% (14/21) of patients, of which 13 were somatic with AAF ranging between 0.6% and 12.0%. Moreover, the AAF did not predict balloon cell presence. Favorable seizure outcomes were associated with genetically clear resection borders. Individuals in whom a causal somatic variant was undetected had excellent postsurgical outcomes. In addition, we demonstrate pathogenicity of the novel c.4373_4375dupATG and candidate c.7499T>A MTOR variants in vitro. We also identified a novel germline aberrant splice site variant in DEPDC5 (c.2802-1G>C)., Discussion: The AAF of somatic pathogenic variants correlated with the topographic distribution, histopathology, and postsurgical outcomes. Moreover, cortical regions with absent histologic FCD features had negligible or undetectable pathogenic variant loads. By contrast, specimens with frank histologic abnormalities had detectable pathogenic variant loads, which raises important questions as to whether there is a tolerable variant threshold and whether surgical margins should be clean, as performed in tumor resections. In addition, we describe 2 novel pathogenic variants, expanding the mTORopathy genetic spectrum. Background: Cannabidiol (CBD), one major nonintoxicating phytocannabinoid from Cannabis sativa demonstrated anti-inflammatory effects in animal models of several inflammatory conditions, including arthritis. However, it is still unknown which cell types mediate these anti-inflammatory effects of CBD, and, since CBD binds to a plethora of receptors and enzymes, it is complicated to pinpoint its mechanism of action. In this study, we elucidate the effects of CBD on B cells and peripheral blood mononuclear cells (PBMCs) in respect to survival, calcium mobilization, drug uptake, and cytokine (IL-6, IL-10, and TNF) and immunoglobulin production. Methods: Modulation of intracellular calcium and drug uptake in B cells was determined by using the fluorescent dyes Cal-520 and PoPo3, respectively. Cytokine and immunoglobulin production was evaluated by enzyme-linked immunosorbent assay. PBMC composition and B cell survival after CBD treatment was assessed by flow cytometry. Results: B cells express two major target receptors for CBD, TRPV2 (transient receptor potential vanilloid 2) and TRPA1 (transient receptor potential ankyrin 1), which are not regulated by B cell activation. CBD increased intracellular calcium levels in mouse and human B cells, which was accompanied by enhanced uptake of PoPo3. These effects were not dependent on transient receptor potential channel activation. CBD increased the number of early apoptotic B cells at the expense of viable cells and diminished interleukin (IL)-10 and tumor necrosis factor (TNF) production when activated T cell independently. In PBMCs, CBD increased IL-10 production when B cells were activated T cell dependent, while decreasing TNF levels when activated T cell independently. In PBMC/rheumatoid synovial fibroblast cocultures, CBD reduced IL-10 production when B cells were activated T cell independently. Immunoglobulin M production was augmented by CBD when B cells were activated with CpG. Conclusion: CBD is able to provide pro- and anti-inflammatory effects in isolated B cells and PBMCs. This is dependent on the activating stimulus (T cell dependent or independent) and concentration of CBD. Therefore, CBD might be used to dampen B cell activity in autoimmune conditions such as rheumatoid arthritis, in which B cells are activated by specific autoantigens.

Whereas politicians broker peace deals, it falls to the public to embrace peace and help sustain it. The legacy of conflicts can make it difficult for people to support reconciling and reintegrating with former enemies. Here we create a five-minute media intervention from interviews we conducted with Colombian Revolutionary Armed Forces (FARC) ex-combatants in a Colombian demobilization camp and non-FARC Colombians in neighbouring communities. We show that exposure to the media intervention humanizes FARC ex-combatants and increases support for peace and reintegration. These effects persisted at least three months post-exposure, were replicated in an independent sample of non-FARC Colombians and affected both attitudes (for example, support for reintegration policies) and behaviour (for example, donations to organizations supporting ex-combatants). As predicted, the intervention's effects were mediated by changing conflict-associated cognitions-reducing the belief that ex-combatants are unwilling and unable to change-beyond affective pathways (for example, increased empathy or reduced prejudice)., (© 2022. Racism remains a pervasive force around the world with widespread and well documented harmful consequences for members of marginalized racial groups. The psychological biases that maintain structural and interpersonal racism begin to emerge in early childhood, but with considerable individual variation-some children develop more racial bias than others. The present study (N = 116; 4-year-old children) provides novel insights into the developmental mechanisms underlying the emergence of racial bias by longitudinally documenting how two psychological processes-normative beliefs about interracial friendships and explanatory beliefs about racial inequalities-developmentally predict the emergence of pro-White/anti-Black racial bias during early childhood. In a 6-month, three-wave, longitudinal study, we found that 4-year-old children's beliefs that their parents and peers do not value interracial friendships predicted increased racial bias in and across time and that children's endorsement of essentialist over extrinsic explanations for racial inequalities predicted the developmental trajectory of racial bias over time. These findings suggest that children's foundational beliefs about the social world developmentally predict the emergence of racial bias in early childhood and speak to the importance of early and persistent intervention efforts targeting children's normative beliefs about interracial friendships and explanatory beliefs about racial inequalities., (© 2021 John Wiley & Sons Ltd.)

In 16 independent samples from five countries involving ~7,700 participants, we employ a mixture of cross-sectional, longitudinal, and quasi-experimental methods to examine the effect of intergroup contact on (a) the blatant dehumanization of outgroups, and (b) the perception that outgroup members dehumanize the ingroup (meta-dehumanization). First, we conduct a meta-analysis across 12 survey samples collected from five countries regarding eight different target groups (total N = 5,388) and find a consistent effect of contact quality on dehumanization and meta-dehumanization. Second, we use a large longitudinal sample of American participants ( N = 1,103) to show that quality of contact with Muslims at Time 1 predicts dehumanization of Muslims and meta-dehumanization 6 months later. Finally, we show that sustained semester-long "virtual contact" between American and Muslim college students is associated with reduced American students' ( N = 487) dehumanization of, and perceived dehumanization by, Muslims.

Research suggests that some people, particularly those on the political right, tend to blatantly dehumanize low-status groups. However, these findings have largely relied on self-report measures, which are notoriously subject to social desirability concerns. To better understand just how widely blatant forms of intergroup dehumanization might extend, the present article leverages an unobtrusive, data-driven perceptual task to examine how U.S. respondents mentally represent "Americans" versus "Arabs" (a low-status group in the United States that is often explicitly targeted with blatant dehumanization). Data from 2 reverse-correlation experiments (original N = 108; preregistered replication N = 336) and 7 rating studies ( N = 2,301) suggest that U.S. respondents' mental representations of Arabs are significantly more dehumanizing than their representations of Americans. Furthermore, analyses indicate that this phenomenon is not reducible to a general tendency for our sample to mentally represent Arabs more negatively than Americans. Finally, these findings reveal that blatantly dehumanizing representations of Arabs can be just as prevalent among individuals exhibiting low levels of explicit dehumanization (e.g., liberals) as among individuals exhibiting high levels of explicit dehumanization (e.g., conservatives)-a phenomenon into which exploratory analyses suggest liberals may have only limited awareness. Taken together, these results suggest that blatant dehumanization may be more widespread than previously recognized and that it can persist even in the minds of those who explicitly reject it. Pollen stored by bees undergoes a fermentation marked by the presence of lactic acid bacteria and yeasts. It results in bee bread. Past studies have singled out Starmerella ( Candida ) magnoliae as the most common yeast species in honey bee-stored bee bread. Starmerella species are ecological specialists with potential biotechnological value. The rarity of recent studies on yeasts in honey bees prompted us to generate new information on yeast diversity during the conversion of bee-collected pollen to bee bread. Bees and stored pollen from two apiaries in Belgium were sampled, a yeast isolation protocol was developed, yeast isolates were grouped according to their macro- and micromorphology, and representative isolates were identified using DNA sequences. Most of the 252 identified isolates belonged to the genera Starmerella , Metschnikowia , and Zygosaccharomyces . The high abundance of yeasts in fresh bee bread decreased rapidly with the storage duration. Starmerella species dominated fresh bee bread, while mostly Zygosaccharomyces members were isolated from aged bee bread. Starmerella ( Candida ) apis , a rarely isolated species, was the most frequent and abundant species in fresh bee bread. Yeasts from the bee's honey stomach and from pollen pellets obtained from bees hind legs were dominated by Metschnikowia species. The distinctive communities from pollen pellets over fresh bee bread to aged bee bread indicate a non-random distribution of these yeasts.

People's actions toward a competitive outgroup can be motivated not only by their perceptions of the outgroup, but also by how they think the outgroup perceives the ingroup (i.e., meta-perceptions). Here, we examine the prevalence, accuracy, and consequences of meta-perceptions among American political partisans. Using a representative sample ( n = 1,056) and a longitudinal convenience sample ( n = 2,707), we find that Democrats and Republicans equally dislike and dehumanize each other but think that the levels of prejudice and dehumanization held by the outgroup party are approximately twice as strong as actually reported by a representative sample of Democrats and Republicans. Overestimations of negative meta-perceptions were consistent across samples over time and between demographic subgroups but were modulated by political ideology: More strongly liberal Democrats and more strongly conservative Republicans were particularly prone to exaggerate meta-perceptions. Finally, we show that meta-prejudice and meta-dehumanization are independently associated with the desire for social distance from members of the outgroup party and support for policies that harm the country and flout democratic norms to favor the ingroup political party. This research demonstrates that partisan meta-perceptions are subject to a strong negativity bias with Democrats and Republicans agreeing that the shadow of partisanship is much larger than it actually is, which fosters mutual intergroup hostility.

Recent behavioral work demonstrates that many people view low-status groups as less "evolved and civilized" than high-status groups. Are these people using blatant expressions of dehumanization simply to express strong dislike toward other groups? Or is blatant dehumanization a process distinct from other negative assessments? We tested these competing hypotheses using functional neuroimaging. Participants judged 10 groups (e.g., Europeans, Muslims, rats) on four scales: blatant dehumanization, dislike, dissimilarity and perceived within-group homogeneity. Consistent with expectations, neural responses when making ratings of dehumanization diverged from those when judging the same targets on the other related dimensions. Specifically, we found regions in the left inferior parietal cortex (IPC) and left inferior frontal cortex (IFC) that were selectively parametrically modulated by dehumanization ratings. The pattern of responses in the left IFC was also consistent with animalistic dehumanization: high responses to low-status human groups and animals, and lower responses to high-status human groups. By contrast, a region in the posterior cingulate cortex was parametrically sensitive specifically to liking. We therefore demonstrate a double dissociation between brain activity associated with judgments of blatant dehumanization and judgments of dislike. Synthetic fungicides are pesticides widely used in agriculture to control phytopathogenic fungi. The systemicity, persistency and intense application of some of these fungicides, such as boscalid, leads to long periods of exposure for honeybees via contaminated water, pollen and nectar. We exposed adult honeybees in the lab to food contaminated with boscalid for 33 days instead of the standard 10-day test. Most of the toxic effects were observed after 10 days. The median time to death (LT 50 ) ranged from 24.9 days (lowest concentration) to 7.1 days (highest concentration) and was significantly shorter in all cases than with the control (32.0 days). The concentration and dietary doses of boscalid inducing 50% mortality (LC 50 and LDD 50 , respectively) decreased strongly with the time of exposure: LC 50  = 14,729 and 1,174 mg/l and LDD 50  = 0.318 and 0.0301 mg bee -1 day -1 at days 8 and 25, respectively. We found evidence of reinforced toxicity when exposure is prolonged, but with an unusual pattern: no cumulative toxicity is observed until 17-18 days, when a point of inflexion appears that suggests a reduced capacity of bees to deal with the toxicant. Our results show the importance of time-to-death experiments rather than fixed-duration studies for evaluating chronic toxicity.

Collectively blaming groups for the actions of individuals can license vicarious retribution. Acts of terrorism by Muslim extremists against innocents, and the spikes in anti-Muslim hate crimes against innocent Muslims that follow, suggest that reciprocal bouts of collective blame can spark cycles of violence. How can this cycle be short-circuited? After establishing a link between collective blame of Muslims and anti-Muslim attitudes and behavior, we used an "interventions tournament" to identify a successful intervention (among many that failed). The "winning" intervention reduced collective blame of Muslims by highlighting hypocrisy in the ways individuals collectively blame Muslims-but not other groups (White Americans, Christians)-for individual group members' actions. After replicating the effect in an independent sample, we demonstrate that a novel interactive activity that isolates the psychological mechanism amplifies the effectiveness of the collective blame hypocrisy intervention and results in downstream reductions in anti-Muslim attitudes and anti-Muslim behavior.

In the current work, we experimentally examined the effect of exposure to a narrative of nonviolent resistance on third-party attitudes toward and support for a disempowered group involved in asymmetric conflict. Across three experiments, we found that Americans exposed to a brief video about Palestinian nonviolent resistance consistently registered more favorable attitudes toward Palestinians than people who watched a film trailer either unrelated to the Israeli-Palestinian conflict or a trailer to a Palestinian-made film about sympathetic Palestinians violently opposing Israelis. Americans' attitudes toward Palestinians and behavior supporting Palestinian collective action persisted weeks after exposure to nonviolent resistance and were mediated by decreased perceptions that Palestinians are inherently violent. Importantly, positive attitudes toward Palestinians did not result in increased negativity toward Israelis. Historically, dehumanization has enabled members of advantaged groups to 'morally disengage' from disadvantaged group suffering, thereby facilitating acts of intergroup aggression such as colonization, slavery and genocide. But is blatant dehumanization exclusive to those at the top 'looking down', or might disadvantaged groups similarly dehumanize those who dominate them? We examined this question in the context of intergroup warfare in which the disadvantaged group shoulders a disproportionate share of casualties and may be especially likely to question the humanity of the advantaged group. Specifically, we assessed blatant dehumanization in the context of stark asymmetric conflict between Israelis (Study 1; N = 521) and Palestinians (Study 2; N = 354) during the 2014 Gaza war. We observed that (a) community samples of Israelis and Palestinians expressed extreme (and comparable) levels of blatant dehumanization, (b) blatant dehumanization was uniquely associated with outcomes related to outgroup hostility for both groups, even after accounting for political ideologies known to strongly predict outgroup aggression, and (c) the strength of association between blatant dehumanization and outcomes was similar across both groups. This study illuminates the striking potency and symmetry of blatant dehumanization among those on both sides of an active asymmetric conflict.

To evaluate the risks of pesticides for pollinators, we must not only evaluate their toxicity but also understand how pollinators are exposed to these xenobiotics in the field. We focused on this last point and modeled honey bee exposure to pesticides at the landscape level. Pollen pellet samples (n = 60) from 40 Belgian apiaries were collected from late July to October 2011 and underwent palynological and pesticide residue analyses. Areas of various crops around each apiary were measured at 4 spatial scales. The most frequently detected pesticides were the fungicides boscalid (n = 19, 31.7%) and pyrimethanil (n = 10, 16.7%) and the insecticide dimethoate (n = 10, 16.7%). We were able to predict exposure probability for boscalid and dimethoate by using broad indicators of cropping intensity, but it remained difficult to identify the precise source of contamination (e.g. specific crops in which the use of the pesticide is authorized). For pyrimethanil, we were not able to build any convincing landscape model that could explain the contamination. Our results, combined with the late sampling period, strongly suggest that pesticides applied to crops unattractive to pollinators, and therefore considered of no risk for them, may be sources of exposure through weeds, drift to neighboring plants, or succeeding crops.

Research suggests that members of advantaged groups who feel dehumanized by other groups respond aggressively. But little is known about how meta-dehumanization affects disadvantaged minority group members, historically the primary targets of dehumanization. We examine this important question in the context of the 2016 U.S. Republican Primaries, which have witnessed the widespread derogation and dehumanization of Mexican immigrants and Muslims. Two initial studies document that Americans blatantly dehumanize Mexican immigrants and Muslims; this dehumanization uniquely predicts support for aggressive policies proposed by Republican nominees, and dehumanization is highly associated with supporting Republican candidates (especially Donald Trump). Two further studies show that, in this climate, Latinos and Muslims in the United States feel heavily dehumanized, which predicts hostile responses including support for violent versus non-violent collective action and unwillingness to assist counterterrorism efforts. Our results extend theorizing on dehumanization, and suggest that it may have cyclical and self-fulfilling consequences.

Although the act of dehumanizing an outgroup is a pervasive and potent intergroup process that drives discrimination and conflict, no formal research has examined the consequences of being dehumanized by an outgroup-that is, "metadehumanization." Across 10 studies (N = 3,440) involving several real-world conflicts spanning 3 continents, we provide the first empirical evidence that metadehumanization (a) plays a central role in outgroup aggression that is (b) mediated by outgroup dehumanization, and (c) distinct from metaprejudice. Studies 1a and 1b demonstrate experimentally that Americans who learn that Arabs (Study 1a) or Muslims (Study 1b) blatantly dehumanize Americans are more likely to dehumanize that outgroup in return; by contrast, experimentally increasing outgroup dehumanization did not increase metadehumanization (Study 1c). Using correlational data, Study 2 documents indirect effects of metadehumanization on Americans' support for aggressive policies toward Arabs (e.g., torture) via Arab dehumanization. In the context of Hungarians and ethnic minority Roma, Study 3 shows that the pathway for Hungarians from metadehumanization to aggression through outgroup dehumanization holds controlling for outgroup prejudice. Study 4 examines Israelis' metaperceptions with respect to Palestinians, showing that: (a) feeling dehumanized (i.e., metadehumanization) is distinct from feeling disliked (i.e., metaprejudice), and (b) metadehumanization uniquely influences aggression through outgroup dehumanization, controlling for metaprejudice. Studies 5a and 5b explore Americans' metaperceptions regarding ISIS and Iran. We document a dehumanization-specific pathway from metadehumanization to aggressive attitudes and behavior that is distinct from the path from metaprejudice through prejudice to aggression. In Study 6, American participants learning that Muslims humanize Americans (i.e., metahumanization) humanize Muslims in turn. Finally, Study 7 experimentally contrasts metadehumanization and metahumanization primes, and shows that resulting differences in outgroup dehumanization are mediated by (a) perceived identity threat, and (b) a general desire to reciprocate the outgroup's perceptions of the ingroup. In summary, our research outlines how and why metadehumanization contributes to cycles of ongoing violence and animosity, providing direction for future research and policy. Functional localizer tasks allow researchers to identify brain regions in each individual's brain, using a combination of anatomical and functional constraints. In this study, we compare three social cognitive localizer tasks, designed to efficiently identify regions in the "Pain Matrix," recruited in response to a person's physical pain, and the "Theory of Mind network," recruited in response to a person's mental states (i.e. beliefs and emotions). Participants performed three tasks: first, the verbal false-belief stories task; second, a verbal task including stories describing physical pain versus emotional suffering; and third, passively viewing a non-verbal animated movie, which included segments depicting physical pain and beliefs and emotions. All three localizers were efficient in identifying replicable, stable networks in individual subjects. The consistency across tasks makes all three tasks viable localizers. Nevertheless, there were small reliable differences in the location of the regions and the pattern of activity within regions, hinting at more specific representations. The new localizers go beyond those currently available: first, they simultaneously identify two functional networks with no additional scan time, and second, the non-verbal task extends the populations in whom functional localizers can be applied. Dehumanization is a central concept in the study of intergroup relations. Yet although theoretical and methodological advances in subtle, "everyday" dehumanization have progressed rapidly, blatant dehumanization remains understudied. The present research attempts to refocus theoretical and empirical attention on blatant dehumanization, examining when and why it provides explanatory power beyond subtle dehumanization. To accomplish this, we introduce and validate a blatant measure of dehumanization based on the popular depiction of evolutionary progress in the "Ascent of Man." We compare blatant dehumanization to established conceptualizations of subtle and implicit dehumanization, including infrahumanization, perceptions of human nature and human uniqueness, and implicit associations between ingroup-outgroup and human-animal concepts. Across 7 studies conducted in 3 countries, we demonstrate that blatant dehumanization is (a) more strongly associated with individual differences in support for hierarchy than subtle or implicit dehumanization, (b) uniquely predictive of numerous consequential attitudes and behaviors toward multiple outgroup targets, (c) predictive above prejudice, and (d) reliable over time. Finally, we show that blatant-but not subtle-dehumanization spikes immediately after incidents of real intergroup violence and strongly predicts support for aggressive actions like torture and retaliatory violence (after the Boston Marathon bombings and Woolwich attacks in England). This research extends theory on the role of dehumanization in intergroup relations and intergroup conflict and provides an intuitive, validated empirical tool to reliably measure blatant dehumanization., ((c) 2015 APA, all rights reserved).)

As in many other locations in the world, honeybee colony losses and disorders have increased in Belgium. Some of the symptoms observed rest unspecific and their causes remain unknown. The present study aims to determine the role of both pesticide exposure and virus load on the appraisal of unexplained honeybee colony disorders in field conditions. From July 2011 to May 2012, 330 colonies were monitored. Honeybees, wax, beebread and honey samples were collected. Morbidity and mortality information provided by beekeepers, colony clinical visits and availability of analytical matrix were used to form 2 groups: healthy colonies and colonies with disorders (n = 29, n = 25, respectively). Disorders included: (1) dead colonies or colonies in which part of the colony appeared dead, or had disappeared; (2) weak colonies; (3) queen loss; (4) problems linked to brood and not related to any known disease. Five common viruses and 99 pesticides (41 fungicides, 39 insecticides and synergist, 14 herbicides, 5 acaricides and metabolites) were quantified in the samples.The main symptoms observed in the group with disorders are linked to brood and queens. The viruses most frequently found are Black Queen Cell Virus, Sac Brood Virus, Deformed Wing Virus. No significant difference in virus load was observed between the two groups. Three acaricides, 5 insecticides and 13 fungicides were detected in the analysed samples. A significant correlation was found between the presence of fungicide residues and honeybee colony disorders. A significant positive link could also be established between the observation of disorder and the abundance of crop surface around the beehive. According to our results, the role of fungicides as a potential stressor for honeybee colonies should be further studied, either by their direct and/or indirect impacts on bees and bee colonies.

People are often called upon to witness, and to empathize with, the pain and suffering of others. In the current study, we directly compared neural responses to others' physical pain and emotional suffering by presenting participants (n = 41) with 96 verbal stories, each describing a protagonist's physical and/or emotional experience, ranging from neutral to extremely negative. A separate group of participants rated "how much physical pain", and "how much emotional suffering" the protagonist experienced in each story, as well as how "vivid and movie-like" the story was. Although ratings of Pain, Suffering and Vividness were positively correlated with each other across stories, item-analyses revealed that each scale was correlated with activity in distinct brain regions. Even within regions of the "Shared Pain network" identified using a separate data set, responses to others' physical pain and emotional suffering were distinct. More broadly, item analyses with continuous predictors provided a high-powered method for identifying brain regions associated with specific aspects of complex stimuli - like verbal descriptions of physical and emotional events.

The physicochemical determinants governing the temperature-dependent adhesion of Streptococcus thermophilus to abiotic surfaces are identified under physiological condition for cells either lacking or not the Rgg0182 transcriptional regulator involved in their thermal adaptation. For that purpose, the wild type LMG18311 strain and Δrgg0182 mutant were imaged using highly resolved atomic force microscopy (AFM) at various cell growth temperatures (42 to 55 °C). The corresponding hydrophobic/hydrophilic balance of the cells was quantitatively addressed via the measurement by chemical force microcopy of their adhesion to a reference hydrophobic surface. Analysis of force-separation distance curves further allowed us to discriminate cell surfaces according to the presence or absence of biopolymers. These results were interpreted in relation to the measured adhesion of the Δrgg0182 mutant onto the hydrophobic wall of microwells in the temperature range from 46 to 52 °C. It is evidenced that the viscoelastic Δrgg0182 cell envelop behaves as a thermo-responsive film whose hydrophobicity increases with increasing temperature, thereby favoring cell attachment to hydrophobic surfaces. Regardless cell growth temperature, wild-type cells do not attach to hydrophobic surfaces and the presence of the Rgg0182 transcriptional regulator is associated with the synthesis of hydrophilic cell surface biopolymers. Throughout, the impact of electrostatics on bioadhesion is ruled out upon examination of electrohydrodynamic cell properties at 50 °C.

Background: Streptococcus thermophilus is an important starter strain for the production of yogurt and cheeses. The analysis of sequenced genomes of four strains of S. thermophilus indicates that they contain several genes of the rgg familly potentially encoding transcriptional regulators. Some of the Rgg proteins are known to be involved in bacterial stress adaptation., Results: In this study, we demonstrated that Streptococcus thermophilus thermal stress adaptation required the rgg0182 gene which transcription depends on the culture medium and the growth temperature. This gene encoded a protein showing similarity with members of the Rgg family transcriptional regulator. Our data confirmed that Rgg0182 is a transcriptional regulator controlling the expression of its neighboring genes as well as chaperones and proteases encoding genes. Therefore, analysis of a Δrgg0182 mutant revealed that this protein played a role in the heat shock adaptation of Streptococcus thermophilus LMG18311., Conclusions: These data showed the importance of the Rgg0182 transcriptional regulator on the survival of S. thermophilus during dairy processes and more specifically during changes in temperature.

Rapsyn, a cytoplasmic receptor-associated protein, is required for the clustering of acetylcholine receptors (AChRs). Although AChR dynamics have been extensively studied, little is known about the dynamics of rapsyn. Here, we used a rapsyn-green fluorescent protein (GFP) fusion protein and quantitative fluorescent imaging to study the dynamics of rapsyn in transfected C2C12 myotubes. First, we found that rapsyn-GFP expression at clusters did not alter AChR aggregation, function, or turnover. Quantification of rapsyn immunofluorescence indicated that the expression of rapsyn-GFP proteins at clusters does not increase the overall rapsyn density compared with untransfected myotube clusters. Using time lapse imaging and fluorescence recovery after photobleaching, we demonstrated that the recovery of rapsyn-GFP fluorescence at clusters was very fast, with a halftime of about approximately 1.5 h (approximately 3 times faster than AChRs). Inhibition of protein kinase C significantly altered receptor insertion, but it had no effect on rapsyn insertion. When cells were treated with the broad spectrum kinase inhibitor staurosporine, receptor insertion was decreased even further. However, inhibition of protein kinase A had no effect on insertion of either rapsyn or receptors. Finally, when cells were treated with neural agrin, rapsyn and AChRs were both directed away from preexisting clusters and accumulated together in new small clusters. These results demonstrate the remarkable dynamism of rapsyn, which may underlie the stability and maintenance of the postsynaptic scaffold and suggest that the insertion of different postsynaptic proteins may be operating independently.

In the CNS, receptor recycling is critical for synaptic plasticity; however, the recycling of receptors has never been observed at peripheral synapses. Using a novel imaging technique, we show here that nicotinic acetylcholine receptors (AChRs) recycle into the postsynaptic membrane of the neuromuscular junction. By sequentially labeling AChRs with biotin-bungarotoxin and streptavidin-fluorophore conjugates, we were able to distinguish recycled, preexisting, and new receptor pools at synapses in living mice. Time-lapse imaging revealed that recycled AChRs were incorporated into the synapse within hours of initial labeling, and their numbers increased with time. At fully functional synapses, AChR recycling was robust and comparable in magnitude with the insertion of newly synthesized receptors, whereas chronic synaptic activity blockade nearly abolished receptor recycling. Finally, using the same sequential labeling method, we found that acetylcholinesterase, another synaptic component, does not recycle. These results identify an activity-dependent AChR-recycling mechanism that enables the regulation of receptor density, which could lead to rapid alterations in synaptic efficacy.
