Next-generation COVID-19 vaccines are being developed to expand the breadth of coverage against existing and future variants and to extend the duration of protection. Prime-2-CoV_Beta is an orf virus (ORFV) based multi-antigen COVID-19 vaccine that co-expresses Spike (S) and Nucleocapsid (N) antigens. The safety and immunogenicity of Prime-2-CoV_Beta is investigated in a phase 1 first-in-human (FIH) dose-finding trial (ORFEUS study, ClinicalTrials.gov: NCT05367843). Participants of two age groups (18–55 and 65–85 years) who previously completed at least two doses of mRNA vaccines were enrolled and sequentially assigned to different dose groups to receive one intramuscular dose of 3 × 105, 3 × 106, 1.5 × 107, or 3 × 107 plaque-forming units (PFU) of Prime-2-CoV_Beta on day 1 and a second dose on day 29. Here, we report safety and immunogenicity data collected up to 6 months after the first study vaccination. Prime-2-CoV_Beta is safe and well tolerated and elicits immune responses at higher dose levels in participants aged 18–55. A single dose of 3 × 107 PFU boosted binding and cross-neutralizing antibody responses that are maintained through 6 months after the first booster vaccination. Polyfunctional S-specific CD4+ and CD8+ T cell responses are observed after vaccination. No pre-existing or vaccine-induced neutralizing anti-vector antibodies are detected. Our findings highlight the potential of the ORFV vector as a safe platform for future vaccine design, which provides the ability to deliver multiple antigens and allows for repeat immunization.

Abstract The recent chromosome-based genome assembly and the newly developed 70K single nucleotide polymorphism (SNP) array for American mink (Neogale vison) facilitate the identification of genetic variants underlying complex traits in this species. The objective of this study was to evaluate the association between consensus runs of homozygosity (ROH) with growth and feed efficiency traits in American mink. A subsample of two mink populations (n = 2,986) were genotyped using the Affymetrix Mink 70K SNP array. The identified ROH segments were included simultaneously, concatenated into consensus regions, and the ROH-based association studies were carried out with linear mixed models considering a genomic relationship matrix for 11 growth and feed efficiency traits implemented in ASReml-R version 4. In total, 298,313 ROH were identified across all individuals, with an average length and coverage of 4.16 Mb and 414.8 Mb, respectively. Background: Irritability affects up to 20% of youth and is a primary reason for referral to pediatric mental health clinics. Irritability is thought to be associated with disruptions in processing of reward, threat, and cognitive control; however, empirical study of these associations at both the behavioral and neural level have yielded equivocal findings that may be driven by small sample sizes and differences in study design. Associations between irritability and brain connectivity between cognitive control and reward- or threat-processing circuits remain understudied. Furthermore, better inhibitory control has been linked to lower irritability and differential neural functioning among irritable youth, suggesting that good inhibitory control may serve as a protective factor. Methods: We hypothesized that higher irritability scores would be associated with less positive (or negative) connectivity between cognitive control and threat-processing circuits and between cognitive control and reward-processing circuits in the Healthy Brain Network dataset (release 10.0; N = 4135). We also hypothesized that these associations would be moderated by inhibitory control such that weaker associations between irritability and connectivity would be detected in youths with better than with worse inhibitory control. Regression models were used to test whether associations between irritability and between-network connectivity were moderated by inhibitory control. Results: Counter to our hypothesis, we detected higher irritability associated with reduced connectivity between threat- and reward-processing and cognitive control networks only in 5- to 9-year-old boys. Inhibitory control did not moderate associations of irritability with between-network connectivity. Conclusions: Exploratory findings indicate that reduced between-network connectivity may underlie difficulty regulating negative emotions, leading to greater irritability.

Summary: Immune cells express an incredible variety of proteins; by measuring combinations of these, cell types influencing disease can be precisely identified. We developed terraFlow, a platform that defines cell subsets exhaustively by combinatorial protein expression. Using high-parameter checkpoint-focused and function-focused panels, we studied classical Hodgkin’s lymphoma (cHL), where systemic T cells have not been investigated in detail. terraFlow revealed immune perturbations in patients, including elevated activated, exhausted, and interleukin (IL)-17+ phenotypes, along with diminished early, interferon (IFN)γ+, and tumor necrosis factor (TNF)+ T cells before treatment; many perturbations remained after treatment. terraFlow identified more disease-associated differences than other tools, often with better predictive power, and included a non-gating approach, eliminating time-consuming and subjective manual thresholds. It also reports a method to identify the smallest set of markers distinguishing study groups. Our results provide mechanistic support for past reports of immune deficiency in cHL and demonstrate the value of terraFlow in immunotherapy and biomarker studies.

Abstract Copy number variations (CNVs) are structural variants consisting of duplications and deletions of DNA segments, which are known to play important roles in the genetics of complex traits in livestock species. However, CNV-based genome-wide association studies (GWAS) have remained unexplored in American mink. Therefore, the purpose of the current study was to investigate the association between CNVs and complex traits in American mink. A CNV-based GWAS was performed with the ParseCNV2 software program using deregressed estimated breeding values of 27 traits as pseudophenotypes, categorized into traits of growth and feed efficiency, reproduction, pelt quality, and Aleutian disease tests. The study identified a total of 10,137 CNVs (6968 duplications and 3169 deletions) using the Affymetrix Mink 70K single nucleotide polymorphism (SNP) array in 2986 American mink. The association analyses identified 250 CNV regions (CNVRs) associated with at least one of the studied traits. These CNVRs overlapped with a total of 320 potential candidate genes, and among them, several genes have been known to be related to the traits such as ARID1B, APPL1, TOX, and GPC5 (growth and feed efficiency traits); GRM1, RNASE10, WNT3, WNT3A, and WNT9B (reproduction traits); MYO10, and LIMS1 (pelt quality traits); and IFNGR2, APEX1, UBE3A, and STX11 (Aleutian disease tests). Overall, the results of the study provide potential candidate genes that may regulate economically important traits and therefore may be used as genetic markers in mink genomic breeding programs.

Abstract Background Prior findings relating secondhand tobacco smoke (SHS) exposure and internalizing problems, characterized by heightened anxiety and depression symptoms, have been equivocal; effects of SHS on neurodevelopment may depend on the presence of other neurotoxicants. Early life stress (ELS) is a known risk factor for internalizing symptoms and is also often concurrent with SHS exposure. To date the interactive effects of ELS and SHS on children’s internalizing symptoms are unknown. We hypothesize that children with higher exposure to both prenatal SHS and ELS will have the most internalizing symptoms during the preschool period and the slowest reductions in symptoms over time. Methods The present study leveraged a prospective, longitudinal birth cohort of 564 Black and Latinx mothers and their children, recruited between 1998 and 2006. Cotinine extracted from cord and maternal blood at birth served as a biomarker of prenatal SHS exposure. Parent-reported Child Behavior Checklist (CBCL) scores were examined at four timepoints between preschool and eleven years-old. ELS exposure was measured as a composite of six domains of maternal stress reported at child age five. Latent growth models examined associations between SHS, ELS, and their interaction term with trajectories of children’s internalizing symptoms. In follow-up analyses, weighted quintile sum regression examined contributions of components of the ELS mixture to children’s internalizing symptoms at each time point. Results ELS interacted with SHS exposure such that higher levels of ELS and SHS exposure were associated with more internalizing symptoms during the preschool period (β = 0.14, p = 0.03). The interaction between ELS and SHS was also associated with a less negative rate of change in internalizing symptoms over time (β=-0.02, p = 0.01). ABSTRACTGlaciers in the Hindu Kush-Karakoram-Himalayas (HKH) are a major source of freshwater in the agriculture-dependent economy of Pakistan. In recent decades, mountain glaciers have been threatened by global warming. In this study, we estimated the Equilibrium Line Altitude (ELA) and geodetic mass balance of fifteen representative glaciers from Hunza (Karakoram), Chitral (Hindu Kush), and Astore (Himalaya) using Landsat satellite images and Advanced Space-borne Thermal Emission and Reflection Radiometer (ASTER) DEMs between 2000 and 2018. The climatic trends (temperature and precipitation) in the 19 years (1995–2013) time series of the three regions were assessed using the non-parametric Mann-Kendall test and Sen’s Slope. The ELA of the observed glaciers except for two Atrak (Chitral) and Gulkin (Hunza) is shifted upward, while the mass balance indicates a heterogeneous pattern ranging from −0.23 ± 0.05 to −0.01 ± 0.015 m w.e a−1, −0.13 ± 0.05 to+0.17 ± 0.11 m w.e a−1 and −0.03 ± 0.02 to+0.23 ± 0.09 m w.e a−1 in Chitral, Hunza, and Astore basin, respectively. A significant or slight increasing or decreasing trend was found in the mean annual temperature, mean total precipitation, and mean discharge of the studied basins. This study will be a good contribution to understanding snow and glacier dynamics in HKH, the factors that influence them, and their interaction with the environment.

Glacier monitoring has been internationally coordinated for more than 125 years. Despite this long history, there is no authoritative answer to the popular question: ‘Which glaciers are the largest in the world?’ Here, we present the first systematic assessment of this question and identify the largest glaciers in the world – distinct from the two ice sheets in Greenland and Antarctica but including the glaciers on the Antarctic Peninsula. We identify the largest glaciers in two domains: on each of the seven geographical continents and in the 19 first-order glacier regions defined by the Global Terrestrial Network for Glaciers. Ranking glaciers by area is non-trivial. It depends on how a glacier is defined and mapped and also requires differentiating between a glacier and a glacier complex, i.e. glaciers that meet at ice divides such as ice caps and icefields. It also depends on the availability of a homogenized global glacier inventory. Using separate rankings for glaciers and glacier complexes, we find that the largest glacier complexes have areas on the order of tens of thousands of square kilometers whereas the largest glaciers are several thousands of square kilometers. The world's largest glaciers and glacier complexes are located in the Antarctic, Arctic and Patagonia.

Abstract Background Copy number variations (CNVs) represent a major source of genetic diversity and contribute to the phenotypic variation of economically important traits in livestock species. In this study, we report the first genome-wide CNV analysis of American mink using whole-genome sequence data from 100 individuals. The analyses were performed by three complementary software programs including CNVpytor, DELLY and Manta. Results A total of 164,733 CNVs (144,517 deletions and 20,216 duplications) were identified representing 5378 CNV regions (CNVR) after merging overlapping CNVs, covering 47.3 Mb (1.9%) of the mink autosomal genome. Gene Ontology and KEGG pathway enrichment analyses of 1391 genes that overlapped CNVR revealed potential role of CNVs in a wide range of biological, molecular and cellular functions, e.g., pathways related to growth (regulation of actin cytoskeleton, and cAMP signaling pathways), behavior (axon guidance, circadian entrainment, and glutamatergic synapse), lipid metabolism (phospholipid binding, sphingolipid metabolism and regulation of lipolysis in adipocytes), and immune response (Wnt signaling, Fc receptor signaling, and GTPase regulator activity pathways). Furthermore, several CNVR-harbored genes associated with fur characteristics and development (MYO5A, RAB27B, FGF12, SLC7A11, EXOC2), and immune system processes (SWAP70, FYN, ORAI1, TRPM2, and FOXO3). Conclusions This study presents the first genome-wide CNV map of American mink. We identified 5378 CNVR in the mink genome and investigated genes that overlapped with CNVR. The results suggest potential links with mink behaviour as well as their possible impact on fur quality and immune response. Overall, the results provide new resources for mink genome analysis, serving as a guideline for future investigations in which genomic structural variations are present.

ABSTRACT: Provision of light during incubation has shown the potential to enhance hatching traits and affect posthatch productivity, physiology, and behavior. In this study, 2 repeated trials were conducted to investigate the effect of photoperiod and strain on the embryo temperature, hatching traits and posthatch growth performance of 2 commercial strains of broilers (Ross 308 and Cobb 500). In each trial, hatching eggs were randomly distributed into 6 incubators with 3 photoperiod treatments: blue LED light for 12 h d−1 (12L:12D) or 18 h d−1 (18L:6D) during entire incubation were compared with no illumination condition (DARK). Data were analyzed as a 3 × 2 factorial arrangement with the trial as the blocking factor. Embryos incubated under 12L:12D and 18L:6D had lower air cell temperature (P < 0.05) than the DARK embryos from d 13 of incubation onward except on the day of candling. The response of air cell temperature to periodic illumination differed between 2 strains. Cobb embryos had lower air cell temperature in 12L:12D than those incubated with 18L:6D from d 16 of incubation onward, whereas lower air cell temperature was found in Ross embryos when illuminated with 18L:6D photoperiod compared to those under 12L:12D. The 12L:12D treatment was associated with improved (P < 0.05) navel closure condition of hatchlings. There were no differences in hatchability, embryo mortality, body weight, or length at hatch among photoperiod groups or its combination with strain. No differences in production parameters were found between DARK and illuminated groups. However, 12L:12D had heavier (P < 0.05) body weight on d 14 of age and higher (P < 0.05) body weight gain than 18L:6D from d 7 to 14 of age. The results of this study indicate that providing blue LED light up to 18 h d−1 has no detrimental effect on production of broilers, however, 12L:12D light regime improved chick quality at hatch compared to DARK and resulted in heavier birds by d 14 compared to 18L:6D.

Abstract The scale of cumulative cultural evolution (CCE) is a defining characteristic of humans. Despite marked scientific interest in CCE, the cognitive underpinnings supporting its development remain understudied. We examined the role cognitive flexibility plays in CCE by studying U.S. children’s (N = 167, 3–5-year-olds) propensity to relinquish an inefficient solution to a problem in favor of a more efficient alternative, and whether they would resist reverting to earlier versions. In contrast to previous work with chimpanzees, most children who first learned to solve a puzzlebox in an inefficient way switched to an observed, more efficient alternative. However, over multiple task interactions, 85% of children who switched reverted to the inefficient method. Moreover, almost all children in a control condition (who first learned the efficient method) switched to the inefficient method. Thus, children were keen to explore an alternative solution but, like chimpanzees, are overall conservative in reverting to their first-learned one.

The Inner Shell Spectroscopy (ISS) beamline on the 8-ID station at the National Synchrotron Light Source II (NSLS-II), Upton, NY, USA, is a high-throughput X-ray absorption spectroscopy beamline designed for in situ, operando, and time-resolved material characterization using high monochromatic flux and scanning speed. This contribution discusses the technical specifications of the beamline in terms of optics, heat load management, monochromator motion control, and data acquisition and processing. Results of the beamline tests demonstrating the quality of the data obtainable on the instrument, possible energy scanning speeds, as well as long-term beamline stability are shown. The ability to directly control the monochromator trajectory to define the acquisition time for each spectral region is highlighted. Examples of studies performed on the beamline are presented. The paper is concluded with a brief outlook for future developments.

Biomimetics has an immense potential to drive the next generation of technologies forward by propounding competent solutions from nature. For decades, the nonsmooth topography of most living creatures has been critical to their existence and survival. Contrary to human-made smooth surfaces, when adapted for fluid flow applications, nonsmooth surfaces can enhance the overall aerodynamic performance by reducing the drag force. Recently, the bioinspired scale structure of fish has been identified as a key biomimetic derivative for improving the aerodynamic efficiency in various cross-domain applications. This study investigates the aerodynamics of a fish scale array (FSA) NACA 0021 model at a specific Reynolds number (Re) of approximately 2.46 × 105 that is meant for laminar flow conditions using Computational Fluid Dynamics (CFD) tools and a subsonic wind tunnel facility. A 3D printed biomimetic FSA film is developed and affixed on the NACA 0021 wing profile for the experimental investigation, followed by flow visualization through the smoke tunnel facility. As proved qualitatively under specific aerodynamic conditions, the creation of velocity streaks has a major role in the drag reduction process. To obtain a clear perspective of flow across the overlapping fish scale structures, the results are focused on the central and overlapping regions of the FSA structure. The experiment has proved the existence of a combined formation of low and high velocity streaks in central and overlapping regions. The FSA 0021 model showed a maximum drag reduction of 9.57%, which was attributed to the streak formation phenomenon observed in the overlapping FSA configuration.

The creation and curation of environmental data present numerous challenges and rewards. In this study, we reflect on the increasing amount of freely available glacier data (inventories and changes), as well as on related demands by data providers, data users, and data repositories in-between. The amount of glacier data has increased significantly over the last two decades as remote sensing techniques have improved and free data access is much more common. The portfolio of observed parameters has increased as well, which presents new challenges for international data centers, and fosters new expectations from users. We focus here on the service of the Global Terrestrial Network for Glaciers (GTN-G) as the central organization for standardized data on glacier distribution and change. Within GTN-G, different glacier datasets are consolidated under one umbrella, and the glaciological community supports this service by actively contributing their datasets and by providing strategic guidance via an Advisory Board. To assess each GTN-G dataset, we present a maturity matrix and summarize achievements, challenges, and ambitions. The challenges and ambitions in the democratization of glacier data are discussed in more detail, as they are key to providing an even better service for glacier data in the future. Most challenges can only be overcome in a financially secure setting for data services and with the help of international standardization as, for example, provided by the CoreTrustSeal. Therefore, dedicated financial support for and organizational long-term commitment to certified data repositories build the basis for the successful democratization of data. In the field of glacier data, this balancing act has so far been successfully achieved through joint collaboration between data repository institutions, data providers, and data users. However, we also note an unequal allotment of funds for data creation and projects using the data, and data curation. Considering the importance of glacier data to answering numerous key societal questions (from local and regional water availability to global sea-level rise), this imbalance needs to be adjusted. In order to guarantee the continuation and success of GTN-G in the future, regular evaluations are required and adaptation measures have to be implemented.

Despite the significant improvement of feed efficiency (FE) in pigs over the past decades, feed costs remain a major challenge for producers profitability. Improving FE is a top priority for the global swine industry. A deeper understanding of the biology underlying FE is crucial for making progress in genetic improvement of FE traits. This review comprehensively discusses the topics related to the FE in pigs including: measurements, genetics, genomics, biological pathways and the advanced technologies and methods involved in FE improvement. We first provide an update of heritability for different FE indicators and then characterize the correlations of FE traits with other economically important traits. Moreover, we present the quantitative trait loci (QTL) and possible candidate genes associated with FE in pigs and outline the most important biological pathways related to the FE traits in pigs. Finally, we present possible ways to improve FE in swine including the implementation of genomic selection, new technologies for measuring the FE traits, and the potential use of genome editing and omics technologies.

Abstract Background The BRCA1 c.3331_3334delCAAG founder mutation has been reported in hereditary breast and ovarian cancer families from multiple Hispanic groups. We aimed to evaluate BRCA1 c.3331_3334delCAAG haplotype diversity in cases of European, African, and Latin American ancestry. Methods BC mutation carrier cases from Colombia (n = 32), Spain (n = 13), Portugal (n = 2), Chile (n = 10), Africa (n = 1), and Brazil (n = 2) were genotyped with the genome-wide single nucleotide polymorphism (SNP) arrays to evaluate haplotype diversity around BRCA1 c.3331_3334delCAAG. Additional Portuguese (n = 13) and Brazilian (n = 18) BC mutation carriers were genotyped for 15 informative SNPs surrounding BRCA1. Data were phased using SHAPEIT2, and identical by descent regions were determined using BEAGLE and GERMLINE. DMLE+ was used to date the mutation in Colombia and Iberia. Results The haplotype reconstruction revealed a shared 264.4-kb region among carriers from all six countries. The estimated mutation age was ~ 100 generations in Iberia and that it was introduced to South America early during the European colonization period. Conclusions Our results suggest that this mutation originated in Iberia and later introduced to Colombia and South America at the time of Spanish colonization during the early 1500s. We also found that the Colombian mutation carriers had higher European ancestry, at the BRCA1 gene harboring chromosome 17, than controls, which further supported the European origin of the mutation. Understanding founder mutations in diverse populations has implications in implementing cost-effective, ancestry-informed screening.

ABSTRACT: The search for alternatives to antibiotics in poultry production is still on-going and has been directed towards investigation of the efficacy of different potential alternatives. However, it is important that the sought alternatives are cost-efficient and have no negative impact on meat quality, for ease of adoption and profit maximization. This study aimed at exploiting an agro-industrial waste, grape pomace (GP) as an alternative to in-feed antibiotics and assessing the effects on growth, intestinal morphology, ceca microbiota, ceca short-chain fatty acid (SCFA) concentration, blood biochemical parameters, and breast muscle myopathies of broiler chickens. A total of 576 one-day-old Cobb-500 broiler chicks were randomly allotted to 3 dietary treatments – Negative control (NC, a corn-wheat soybean-based diet), NC + 0.05% bacitracin methylene disalicylate (BMD), and NC + 2.5% GP. Each treatment was assigned to 8 replicate pens with 25 birds per pen. Body weight (BW), feed intake (FI), and feed conversion ratio (FCR) were determined weekly. On d 36, 2 chickens/pen were euthanized for measuring blood biochemical parameters, ceca SCFA, and ceca microbiota. White striping (WS) and wooden breast (WB) incidence were assessed in 4 chickens/pen on d 42. The GP diet increased (P < 0.05) average FI throughout the feeding phases compared to the other treatments, but overall FCR was similar. Birds in the GP treatment had higher (P < 0.05) villus height (VH) and increased VH:crypt depth ratio in the duodenum and jejunum compared to other treatments. The level of ceca SCFA and the incidence of WS and WB was the same for all treatments. Plasma Ca and P were significantly higher (P < 0.05) in birds fed GP and BMD, compared to the NC. Birds in the GP treatment had significantly reduced (P < 0.05) plasma aspartate transaminase than other treatments. Birds receiving GP had a higher (P < 0.05) relative abundance of the phylum Bacteroidetes and reduced (P < 0.05) Firmicutes compared to other treatments. The relative abundance of Bacteroides and Lactobacillus genera were higher (P < 0.05) among birds fed GP compared to other treatments. Inclusion of 2.5% GP in broiler chicken diets improved gut morphology and modified the cecal bacterial community and blood biochemical profiles with no adverse effect on growth performance and meat quality.

Background and Objectives: Prenatal exposure to environmental tobacco smoke (ETS) is associated with increased attention problems in children, however, the effects of such exposure on children’s brain structure and function have not been studied. Herein, we probed effects of prenatal ETS on children’s cognitive control circuitry and behavior. Methods: Forty-one children (7–9 years) recruited from a prospective longitudinal birth cohort of non-smoking mothers completed structural and task-functional magnetic resonance imaging to evaluate effects of maternal ETS exposure, measured by maternal prenatal urinary cotinine. Attention problems and externalizing behaviors were measured by parent report on the Child Behavior Checklist. ABSTRACT: Providing lighting schedule during incubation has been shown to improve chick quality and reduce stress posthatch. This study was conducted to evaluate the effects of providing light of different colors during incubation on embryo development, air cell temperature, the spread of hatch, and hatching performance. Four batches of eggs (n = 2,176, 1,664, 1,696 and 1,600) from Ross 308 broiler breeders were used in the experiment. In each trial, eggs were randomly distributed into 4 lighting treatments. The incubation lighting treatments included: incubated under dark as control, illuminated with white, red or blue lights for 12 h daily. There were no incubation lighting treatment differences in embryo development, the spread of hatch, hatchability, embryo mortality, hatch weight, chick length, navel closure quality, yolk-free body weight, or relative spleen weight. However, embryos incubated under red light had lower average air cell temperature than those in dark, white or blue light treatments. This finding may suggest higher melatonin secretion during the scotophase when illuminated with red light. Male chicks incubated under dark had a higher bursa of Fabricius weight than males illuminated with blue light. In conclusion, these results suggest that the red, white and blue light stimulation during incubation had no negative effects on hatchability, embryo mortality, spread of hatch or day-old chick quality, but may have potential impacts on immunity and energy metabolism in broiler embryos.

ABSTRACT: Previous findings have reported that providing light during incubation can affect hatchability and chick quality. This study was conducted to investigate the effects of providing light during incubation on posthatch broiler production parameters, thermoregulation and immune response. Lights with different wavelengths were used over the course of four separate hatches. Ross 308 broiler hatching eggs were randomly distributed into 4 lighting treatments for each hatch. The incubation lighting treatments included: dark as control, white, red, or blue lights for 12 h d−1 (200 lux at egg level). Broilers hatched from each incubator with the same gender were placed into one of 8 sets of pens (3 pens/set) and raised under 18 h d−1 photoperiod. Six birds per pen were immunized intraocularly with AviPro ND-IB Polybanco vaccine on d 10 and 21 posthatch. Chicks hatched under white and blue lights had heavier (P < 0.05) body weight and higher (P < 0.05) feed consumption than the control group during the first 6 h postplacement. No differences in vent temperature were found among treatments at 24 h posthatch (P > 0.05). Chicks hatched with light stimulation however had more stable (P < 0.05) cloaca temperature at 36 h posthatch. No differences in average body weight gain, feed consumption or feed conversion ratio were found among lighting treatments between d 7 and d 35. On d 14 of age, birds hatched from red light had higher (P < 0.05) total IgG concentration than those hatched under dark, blue or white light. These results indicated that in ovo light stimulation with different wavelengths did not affect growth parameters of broilers at market age. Providing photoperiodic blue and white light during incubation improved the production parameters of broilers during the first week posthatch.

Background: Complicating the COVID-19 pandemic are the healthcare disparities experienced by ethnic minorities, especially those with comorbidities including cancer. The introduction of COVID-19 vaccines has been instrumental in blunting the morbidity and mortality from the pandemic; however, vaccine hesitancy, particularly among ethnic minorities, has been a major concern. Thus, we sought to evaluate the knowledge and perspectives of COVID-19 and vaccines among our ethnic minority cancer patient population. Methods: Following an IRB approved protocol, questionnaires were completed by patients in a predominantly ethnic minority population at a single institution between 1 February and 30 June 2021. Included were any adult cancer patients with either a solid or hematologic malignancy. Results: Among the 84 patients that were offered the questionnaires, 52 patients responded, with a median age of 63.5 years. Overall, 36% were non-Hispanic Blacks and 30% were Hispanics; 65% were receiving active treatment for their cancer. Seventy-nine percent believed COVID-19 to be dangerous or harmful to them, 61% were concerned about the side effects, yet 65% considered COVID-19 vaccines as safe. Among the seven patients that refused the vaccine, (71%, n = 5) cited side effects and/or (57%, n = 4) believed that the vaccine was not needed. Overall, there was a significantly higher chance of being vaccinated if patients were receiving active cancer treatment, believed COVID-19 was harmful, or that the vaccine was safe, and knew COVID-19 was a virus. Conclusions: This exploratory study demonstrates that most ethnic minority cancer patients are receptive to vaccines, with a majority being vaccinated. However, we also discovered various reasons why this group of patients may not want be vaccinated, including concerns about side effects and perception that COVID-19 is not harmful. These findings can help us further understand the complex nature of vaccine hesitancy in ethnic minority cancer patients, and aid in developing future vaccine awareness strategies as the COVID-19 pandemic continues to evolve.

# Background This paper estimates the costs and benefits of investing in education interventions and specific child marriage programs to reduce child marriage in India. Child marriage in India remains highly prevalent despite considerable progress in the last decade or more, associated, in particular, with a decline in poverty. The economic consequences of the COVID 19 pandemic are likely to reverse these gains, meaning that the interventions to reduce child marriage evaluated in this paper assume even greater importance. # Methods A literature search to identify interventions with costs and estimates of impact on reduction in child marriage for India, and other low and middle income countries was conducted. The costs and effectiveness of the interventions were modelled for India, and economic benefits of reducing school dropout rates and increasing secondary school completions were estimated. The ratio of the economic benefits to the intervention costs estimated gave the benefit-cost ratio. # Results The interventions resulted in an average benefit-cost ratio of 16.8. The interventions are projected to decrease the estimated marriage rate for girls aged 15–17 years for India from an estimated rate of 16.4% in 2020 by 7.5 percentage points by 2050, with education interventions targeting increased attendance being the larger factor. By 2030, the proportion of girls completing school is estimated to increase by 13 percentage points and productivity to have increased by 16.4%. # Conclusions The results indicate substantial economic and social gains to reducing child marriage by interventions that decrease school dropout rates, increase secondary school completions for girls, and address some of the broader social and cultural disadvantages they face in India. This has significant implications not only for their health, but also for the quality of the available economic opportunities and the possibility of attaining Sustainable Development Goal 5.

Background: Anticorrelated resting state connectivity between task-positive and task-negative networks in adults supports flexible shifting between externally focused attention and internal thought. Findings suggest that children show positive correlations between task-positive (frontoparietal; FP) and task-negative (default mode; DMN) networks. FP-DMN connectivity also associates with intellectual functioning across the lifespan. We investigated whether FP-DMN connectivity in healthy children varied with age and intelligence quotient (IQ). Methods: We utilized network-based statistics (NBS) to examine resting state functional connectivity between FP and DMN seeds in N = 133 7−25-year-olds (Mage = 15.80). Linear regression evaluated FP-DMN associations with IQ. Results: We detected NBS subnetworks containing both within- and between-network connections that were inversely associated with age. Four FP-DMN connections showed more negative connectivity between FP (inferior frontal gyrus and precentral gyrus) and DMN regions (frontal medial cortex, precuneus, and frontal pole) among older participants. Frontal pole-precentral gyrus connectivity inversely associated with IQ. Conclusions: FP-DMN connectivity was more anticorrelated at older ages, potentially indicating dynamic network segregation of these circuits from childhood to early adulthood. Youth with more mature (i.e., anticorrelated) FP-DMN connectivity demonstrated higher IQ. Our findings add to the growing body of literature examining neural network development and its association with IQ.

Low childhood socioeconomic status (SES) predisposes individuals to altered trajectories of brain development and increased rates of mental illness. Brain connectivity at birth is associated with psychiatric outcomes. We sought to investigate whether SES at birth is associated with neonatal brain connectivity and if these differences account for socioeconomic disparities in infant symptoms at age 2 years that are predictive of psychopathology. Resting state functional MRI was performed on 75 full-term and 37 term-equivalent preterm newborns (n = 112). SES was characterized by insurance type, the Area Deprivation Index, and a composite score. Seed-based voxelwise linear regression related SES to whole-brain functional connectivity of five brain regions representing functional networks implicated in psychiatric illnesses and affected by socioeconomic disadvantage: striatum, medial prefrontal cortex (mPFC), ventrolateral prefrontal cortex (vlPFC), and dorsal anterior cingulate cortex. Lower SES was associated with differences in striatum and vlPFC connectivity. Striatum connectivity with frontopolar and medial PFC mediated the relationship between SES and behavioral inhibition at age 2 measured by the Infant-Toddler Social Emotional Assessment (n = 46). Striatum-frontopolar connectivity mediated the relationship between SES and externalizing symptoms. These results, convergent across three SES metrics, suggest that neurodevelopmental trajectories linking SES and mental illness may begin as early as birth.

Background: The once-daily, single-tablet regimen darunavir/cobicistat/emtricitabine/tenofovir alafenamide (D/C/F/TAF) 800/150/200/10 mg is approved for the treatment of HIV-1 infection. The 48-week efficacy and safety of D/C/F/TAF versus darunavir/cobicistat + emtricitabine/tenofovir disoproxil fumarate (control) in treatment-naïve adults were demonstrated in the phase 3 AMBER study. Objective: To describe AMBER outcomes across patient subgroups based on demographic and clinical characteristics at baseline. Methods: AMBER patients had viral load (VL) ≥1000 copies/mL, CD4+ cell count >50 cells/µL, and genotypic susceptibility to darunavir, emtricitabine, and tenofovir. Primary endpoint was the proportion of patients with virologic response (VL 50 years), gender, race (black/non-black), baseline VL (≤/>100,000 copies/mL), baseline CD4+ cell count (50 years and women, relative to their comparator groups, regardless of treatment arm (notably, sample sizes were small for patients >50 years and women). Improvements in eGFRcystC and stable bone mineral density were observed with D/C/F/TAF overall, and results were generally consistent across subgroups. Conclusions: For treatment-naïve patients in AMBER, initiating therapy with the D/C/F/TAF single-tablet regimen was an effective and well-tolerated option, regardless of demographic or clinical characteristics. Abstract Background Treatments for localized prostate cancer present challenging tradeoffs in the face of uncertain treatment benefits. These options are best weighed in a process of shared decision-making with the patient’s healthcare team. Minority men experience disparities in prostate cancer outcomes, possibly due in part to a lack of optimal communication during treatment selection. Decision aids facilitate shared decision-making, improve knowledge of treatment options, may increase satisfaction with treatment choice, and likely facilitate long-term quality of life. Methods/design This study will compare the effect of two evidence-based decision aids on patient knowledge and on quality of life measured one year after treatment, oversampling minority men. One decision aid will be administered prior to specialist consultation, preparing patients for a treatment discussion. The other decision aid will be administered within the consultation to facilitate transparent, preference-sensitive, and evidence-informed deliberations. The study will utilize a four-arm, block-randomized design to test whether each decision aid alone (Arms 1 and 2) or in combination (Arm 3) can improve patient knowledge and quality of life compared to usual care (Arm 4). The study, funded by the National Cancer Institute’s Community Oncology Research Program (NCORP), will be deployed within select institutions that have demonstrated capacity to recruit minority populations into urologic oncology trials. Discussion Upon completion of the trial, we will have 1) tested the effectiveness of two evidence-based decision aids in enhancing patients’ knowledge of options for prostate cancer therapy and 2) estimated whether decision aids may improve patient quality of life one year after initial treatment choice.
