Abstract

The UCNA experiment was designed to measure the neutron β-asymmetry parameter A0 using polarized ultracold neutrons (UCN). UCN produced via downscattering in solid deuterium were polarized via transport through a 7 T magnetic field, and then directed to a 1 T solenoidal electron spectrometer, where the decay electrons were detected in electron detector packages located on the two ends of the spectrometer. A value for A0 was then extracted from the asymmetry in the numbers of counts in the two detector packages. We summarize all of the results from the UCNA experiment, obtained during run periods in 2007, 2008–2009, 2010, and 2011–2013, which ultimately culminated in a 0.67% precision result for A0.

Abstract

In January, 2018, Fornal and Grinstein proposed that a previously unobserved neutron decay branch to a dark matter particle (χ) could account for the discrepancy in the neutron lifetime observed in two different types of experiments. One of the possible final states discussed includes a single χ along with an e+e− pair. We use data from the UCNA (Ultracold Neutron Asymmetry) experiment to set limits on this decay channel. Coincident electron-like events are detected with ∼ 4π acceptance using a pair of detectors that observe a volume of stored Ultracold Neutrons (UCNs). We use the timing information of coincidence events to select candidate dark sector particle decays by applying a timing calibration and selecting events within a physically-forbidden timing region for conventional n → p + e- + ν̅e decays. The summed kinetic energy (Ee+e−) from such events is reconstructed and used to set limits, as a function of the χ mass, on the branching fraction for this decay channel.

Abstract

Systemic sclerosis (SSc) is an autoinflammatory, fibrotic condition of unknown aetiology. The presence of detectable autoantibodies against diverse nuclear antigens, as well as strong HLA associations with disease, suggest autoimmune involvement, however the links between endogenous and exogenous risk factors and SSc pathology remain undetermined. We have conducted a genetic analysis of HLA inheritance in two independent and meta-analysed cohorts of 1,465 SSc cases and 13,273 controls, including stratified association analyses in clinical and autoantibody positive subgroups of disease. Additionally, we have used patient genotypes to impute gene dosages across the KIR locus, encoding paired activating and inhibitory lymphocyte receptors for Class I HLA ligands, to conduct the largest analysis of KIR-HLA epistatic interactions in SSc to date. We confirm previous Class II HLA associations with SSc risk and report a new Class I association with haplotype HLA-B*44:03-HLA-C*16:01 at genome-wide significance (GWS). We further report statistically significant HLA associations with clinical and serological subtypes of disease through direct case-case comparison, and report a new association of HLA-DRB1*15:01, previously shown to bind topoisomerase-1 derived peptides, with anti-topoisomerase (ATA) positive disease. Finally, we identify genetic epistasis between KIRs and HLA class I ligands, suggesting genetic modulation of lymphocyte activation may further contribute to an individual's underlying disease risk. Abstract

Objective: We undertook this study to examine the X chromosome complement in participants with systemic sclerosis (SSc) as well as idiopathic inflammatory myopathies. Method(s): The participants met classification criteria for the diseases. All participants underwent single-nucleotide polymorphism typing. We examined X and Y single-nucleotide polymorphism heterogeneity to determine the number of X chromosomes. For statistical comparisons, we used chi2 analyses with calculation of 95% confidence intervals. Result(s): Three of seventy men with SSc had 47,XXY (P = 0.0001 compared with control men). Among the 435 women with SSc, none had 47,XXX. Among 709 men with polymyositis or dermatomyositis (PM/DM), seven had 47,XXY (P = 0.0016), whereas among the 1783 women with PM/DM, two had 47,XXX. Of 147 men with inclusion body myositis (IBM), six had 47,XXY, and 1 of the 114 women with IBM had 47,XXX. For each of these myositis disease groups, the excess 47,XXY and/or 47,XXX was significantly higher compared with in controls as well as the known birth rate of Klinefelter syndrome or 47,XXX. Conclusion(s): Klinefelter syndrome (47,XXY) is associated with SSc and idiopathic inflammatory myopathies, similar to other autoimmune diseases with type 1 interferon pathogenesis, namely, systemic lupus erythematosus and Sjogren syndrome.Copyright © 2022 The Authors. Abstract

The kidney-specific gene UMOD encodes for uromodulin, the most abundant protein excreted in normal urine. Rare large-effect variants in UMOD cause autosomal dominant tubulointerstitial kidney disease (ADTKD), while common low-impact variants strongly associate with kidney function and the risk of chronic kidney disease (CKD) in the general population. It is unknown whether intermediate-effect variants in UMOD contribute to CKD. Here, candidate intermediate-effect UMOD variants were identified using large-population and ADTKD cohorts. Biological and phenotypical effects were investigated using cell models, in silico simulations, patient samples, and international databases and biobanks. Eight UMOD missense variants reported in ADTKD are present in the Genome Aggregation Database (gnomAD), with minor allele frequency (MAF) ranging from 10 −5 to 10 −3 . Among them, the missense variant p.Thr62Pro is detected in ∼1/1,000 individuals of European ancestry, shows incomplete penetrance but a high genetic load in familial clusters of CKD, and is associated with kidney failure in the 100,000 Genomes Project (odds ratio [OR] = 3.99 [1.84 to 8.98]) and the UK Biobank (OR = 4.12 [1.32 to 12.85). Compared with canonical ADTKD mutations, the p.Thr62Pro carriers displayed reduced disease severity, with slower progression of CKD and an intermediate reduction of urinary uromodulin levels, in line with an intermediate trafficking defect in vitro and modest induction of endoplasmic reticulum (ER) stress. Identification of an intermediate-effect UMOD variant completes the spectrum of UMOD -associated kidney diseases and provides insights into the mechanisms of ADTKD and the genetic architecture of CKD.

Abstract

CONTEXT: Pathogenic germline MAX variants are associated with pheochromocytoma and paraganglioma (PPGL), pituitary neuroendocrine tumors and, possibly, other endocrine and nonendocrine tumors. OBJECTIVE(S): To report 2 families with germline MAX variants, pheochromocytomas (PCs) and multiple other tumors. METHOD(S): Clinical, genetic, immunohistochemical, and functional studies at University hospitals in Australia on 2 families with germline MAX variants undergoing usual clinical care. The main outcome measures were phenotyping; germline and tumor sequencing; immunohistochemistry of PC and other tumors; functional studies of MAX variants. RESULT(S): Family A has multiple individuals with PC (including bilateral and metastatic disease) and 2 children (to date, without PC) with neuroendocrine tumors (paravertebral ganglioneuroma and abdominal neuroblastoma, respectively). One individual has acromegaly; immunohistochemistry of PC tissue showed positive growth hormone-releasing hormone staining. Another individual with previously resected PCs has pituitary enlargement and elevated insulin-like growth factor (IGF-1). A germline MAX variant (c.200C>A, p.Ala67Asp) was identified in all individuals with PC and both children, with loss of heterozygosity in PC tissue. Immunohistochemistry showed loss of MAX staining in PCs and other neural crest tumors. In vitro studies confirmed the variant as loss of function. In Family B, the proband has bilateral and metastatic PC, prolactin-producing pituitary tumor, multigland parathyroid adenomas, chondrosarcoma, and multifocal pulmonary adenocarcinomas. A truncating germline MAX variant (c.22G>T, p.Glu8*) was identified. CONCLUSION(S): Germline MAX mutations are associated with PCs, ganglioneuromas, neuroblastomas, pituitary neuroendocrine tumors, and, possibly, parathyroid adenomas, as well as nonendocrine tumors of chondrosarcoma and lung adenocarcinoma, suggesting MAX is a novel multiple endocrine neoplasia gene.Copyright © The Aut

Abstract

Decreased milk production of beef cattle grazing endophyte-infected (EI) tall fescue (TF), an important cool season grass in the southeastern United States, can affect calf growth. The objective was to determine whether a thermal or heat-tolerant Bos taurus breed of cattle, Romosinuano (RO), would tolerate EI TF toxins relative to Angus (AN) cattle. Milk production and quality were examined on AN and RO cows grazing endophyte-free (EF; variety K-31; n = 36) or EI TF (variety K-31; n = 37) along with growth of their Charolais-sired calves in 2007 and 2008 in west central Arkansas. Cows calved between early February and late March (spring). Milk yield and quality, BW, rectal temperature, and serum prolactin were determined at 57.1 [+ or -] 2.6 d postpartum and every 28 d thereafter between April and August. Milk yield was greater in AN compared with RO cows (P < 0.001), but not influenced by forage type (P > 0.88). Percent milk fat was greater in cows grazing EF TF in April, but greater for cows grazing EI TF in July (forage x month, P < 0.001). Percent milk fat was greater for RO than AN cows (P < 0.001). Percent milk protein (P < 0.001) was greater and somatic cell counts (log-transformed; P < 0.001) were less in RO than AN cows. Milk lactose was greater for RO compared with AN cows in June through August (breed x month, P = 0.004). Adjusted weaning BW of calves was similar between EF and EI TF in 2007, but greater for calves from EF than EI TF in 2008 (forage x year, P = 0.03). Rectal temperature was similar between RO cows grazing EF and EI TF, but greater in AN cows grazing EI compared with EF TF in most months (forage x breed x month x year, P < 0.001). Serum prolactin was reduced in both breeds of cows grazing EI TF between April and July of both years and greatest in RO cows grazing EF TF (breed, P < 0.001; forage x month, P < 0.001). These data suggest that RO cows were more thermal-tolerant, but still susceptible to toxins in EI TF as shown by a reduction in serum concentrations of prolactin. However, milk production was not influenced by EI TF as previously observed, but milk fat percent was decreased in early lactation in this group of cows. Twenty-one mixed-parity (average 2.4 [+ or -] 0.49) crossbred sows and their offspring were used to determine whether sow milk leptin at farrowing was related to neonatal serum leptin and pig growth to weaning. During farrowing, pigs were randomly allotted to suckling (n = 99) or delayed suckling (n = 89) groups, with delayed suckling pigs placed in a group pen apart from the dam before suckling. Both groups had access to heat lamps. Colostrum samples were collected no more than 2 h after farrowing the first pig. Blood samples were collected from all pigs approximately 2 h after farrowing was complete; pigs were then ear notched and returned to their dams. Pig BW was recorded at 1.2 [+ or -] 0.04 d of age and again at weaning. Milk and blood serum were collected after centrifugation; leptin concentrations were estimated using RIA. Leptin was detected in colostral milk, as expected, and averaged 46.0 [+ or -] 1.1 ng/mL. Pig serum leptin (P < 0.02) was greater in suckling pigs than in delayed suckling pigs, averaging 0.69 [+ or -] 0.08 and 0.54 [+ or -] 0.07 ng/mL, respectively. Although male pigs were heavier (P < 0.01) at birth than female pigs (1,507 [+ or -] 52 vs. 1,381 [+ or -] 43 g), ADG to weaning and weaning weights were similar for both sexes, averaging 229 [+ or -] 14 g and 5,829 [+ or -] 323 g, respectively, for all pigs; serum leptin concentrations were not affected by sex of the pig. Milk serum leptin was not associated with litter size, parity, pig birth weight, ADG to weaning, or weaning weight. Suckling status did not influence ADG to weaning or weaning weight of pigs; neonatal pig serum leptin was not related to birth weight, weaning weight, or ADG to weaning. These results indicate that leptin is not directly related to early neonatal growth in the pig; however, more in-depth studies are needed to determine possible indirect or long-term effects of early leptin exposure. The cDNA-encoding sequences for yak metallothionein isoforms I (MT-I) and II (MT-II) were amplified and cloned by reverse-transcription PCR to characterize the nucleotide sequence and protein structure of metallothionein in the yak. The cDNA sequences of MT-I and MT-II were subjected to BLAST searching at the National Center for Biotechnology Information, and the results indicated that the nucleotide sequences of yak MT-I and MT-II, when compared among different species of mammals, are highly conserved. The yak open reading frames have a length of 183 nucleotides, which encode for yak MT-I and MT-II proteins of 61 AA, respectively. Analysis of hydrophobicity, transmembrane region, and signal peptides suggested that metallothioneins of the yak are nonsecretory proteins. There were several conserved tripeptide sequences, such as C-X-C, C-C-X-C-C, and C-X-X-C (X designates AA excluding cysteine in MT-I and MT-II), and they are highly conserved in their evolution. By homologous comparative modeling, we predicted the molecular spatial structures of yak MT-I and MT-II, which are composed of [alpha]- and [beta]-domains that are linked by the conserved tripeptide [Lys.sup.30]-[Lys.sup.30] -[Ser.sup.32] (KKS). Abstract

We conducted DNA methylation association analyses using Illumina 450K data from whole blood for an Australian amyotrophic lateral sclerosis (ALS) case–control cohort (782 cases and 613 controls). Analyses used mixed linear models as implemented in the OSCA software. We found a significantly higher proportion of neutrophils in cases compared to controls which replicated in an independent cohort from the Netherlands (1159 cases and 637 controls). The OSCA MOMENT linear mixed model has been shown in simulations to best account for confounders. When combined in a methylation profile score, the 25 most-associated probes identified by MOMENT significantly classified case–control status in the Netherlands sample (area under the curve, AUC = 0.65, CI95% = [0.62–0.68], p = 8.3 × 10−22). The maximum AUC achieved was 0.69 (CI95% = [0.66–0.71], p = 4.3 × 10−34) when cell-type proportion was included in the predictor.

Milk yield from 160 Brangus cows sired by 65 Brangus bulls was measured over a 3-yr period with a single-cow milking machine to estimate the relationship of actual milk yield of daughters and their calves' BW with cow sire EPD for milk during the preweaning period. Milk yield was measured six times per year at an average 49, 78, 109, 138, 168, and 198 d postpartum. The regression of daughters' milk yield on sire milk EPD was quadratic (P < 0.01), and the initial linear portion of the curve differed among months (P < 0.05) at an average cow BW. Similarly, the regression of 6-mo average 24-h milk yield on sire milk EPD was curvilinear (P < 0.05). When cow BW was fitted as a covariate in the regression of 6-mo average 24-h milk yield on sire milk EPD, there was an interaction of cow BW with linear sire milk EPD and quadratic sire milk EPD (P < 0.10). The associated response surface suggested that the regression was primarily linear in cows weighing [less than or equal to] 520 kg and curvilinear in cows weighing >520 kg. A trend existed for the regression of calf 205-d weight on grandsire milk EPD to be curvilinear (P < 0.21); however, the regression of calf 205-d weight on milk yield of their dam was linear (P < 0.01). Results from these data suggest that genetic potential for milk yield, and possibly the associated effects on calf BW transmitted through the grandsire, may have a practical maximum because of nutritional limitations that prevent the expression of genetic potential beyond that level, particularly in heavier cows, which suggests the need to match sire milk EPD and cow BW with production environment. 42. Influence of milk production potential on forage dry matter intake by multiparous and primiparous Brangus females

Abstract

Brangus cows (n = 29) were used in three experiments to evaluate the effects of parity (multiparous vs. primiparous) and potential genetic merit for milk production (high vs. low) on forage intake during late gestation, early lactation, and late lactation. Cows were selected for milk production based on their sire's EPD for milk production (MEPD). Cows had ad libitum access to (130% of previous 2-d average intake) low-quality hay (5.3% CP and 76% NDF), and cottonseed meal was supplemented to ensure adequate degradable intake protein. All females were adapted to diets for at least 7 d, and individual intake data were collected for 9 d. During the lactation trials, actual milk production was determined using a portable milking machine following a 12-h separation from calves. During late gestation, multiparous cows consumed 24% more (P = 0.01) forage DM (kg/d) than primiparous cows; however, parity class did not influence forage intake when intake was expressed relative to BW. Furthermore, MEPD did not influence forage intake during late gestation. During early lactation, multiparous cows produced 66% more (P < 0.001) milk than primiparous cows, and high MEPD tended (P = 0.10) to produce more milk than low MEPD. Multiparous cows consumed 19% more (P < 0.0001) forage DM than did primiparous cows when expressed on an absolute basis, but not when expressed on a BW basis. High-MEPD cows consumed 8% more (P < 0.05) forage DM than did low-MEPD cows. During late lactation, multiparous cows produced 84% more milk than primiparous cows, although MEPD did not influence (P = 0.40) milk yield. In addition, multiparous cows consumed 17% more (P < 0.01) forage DM per day than primiparous cows, but when intake was expressed relative to BW, neither parity nor MEPD influenced forage DMI during late lactation. Milk yield and BW explained significant proportions of the variation in forage DMI during early and late lactation. Each kilogram increase in milk yield was associated with a 0.33- and 0.37-kg increase in forage DMI for early and late lactation, respectively. Results suggest that multi- and primiparous cows consume similar amounts of low-quality forage DM, expressed per unit of BW, during late gestation and lactation. Mature weight and rate of maturing were estimated in 177 Angus, Brahman, and reciprocal-cross cows grazing bermudagrass or endophyte-infected tall fescue over a 4-yr period to evaluate genotype x environment interactions. Data were collected every 28 d until cows were approximately 18 mo of age and then at prebreedihg, postcalving, and weaning of calf. All cows with weight data to at least 42 mo of age were included in the analysis. Mature weight and rate of maturing were estimated using the three-parameter growth curve model described by Brody (1945). Data were pooled over year and analyzed by the general linear model procedure of SAS. Included in the models for mature weight and rate of maturing were the independent variables of genotype, environment, and genotype X environment interaction. There was a genotype X environment interaction (P < 0.01) for mature body weight (BW) but not for rate of maturing. Angus cows grazing fescue pastures had greater (P < 0.01) mean mature BW than Angus x Brahman cows grazing bermudagrass (611 [+ or -] 17 vs 546 [+ or -] 16 kg). Angus X Brahman cows grazing bermudagrass had lower (P < 0.05) mean mature BW than Brahman X Angus cows grazing bermudagrass or endophyte-infected fescue and Brahman cows grazing bermudagrass (546 [+ or -] 16 vs 624 [+ or -] 19, 614 [+ or -] 22 and 598 [+ or -] 20 kg, respectively). Brahman cows grazing endophyte-infected fescue had smaller (P < 0.05) mean mature BW than all genotype X forage combinations except for Angus X Brahman cows grazing bermudagrass. Angus cows had a smaller (P < 0.05) mean rate of maturing than Angus X Brahman and Brahman X Angus cows (0.039 + 0.002 vs 0.054 + 0.002 and 0.049 [+ or -] 0.002%/mo, respectively), respectively, and Angus x Brahman cows had a larger (P < 0.05) mean rate of maturing than Brahman x Angus and Brahman cows (0.054 [+ or -] 0.002 vs 0.049 [+ or -] 0.002 and 0.041 [+ or -] 0.002 %/mo, respectively). There was a direct breed x forage interaction (P < 0.05) for mature BW. These data suggest that the choice of breed type is important in maintaining a crossbreeding program, in that mature BW and rate of maturing are critical to the matching of animal requirements to available production resources. Interactions of the regression of preweaning ADG on dam milk yield and quality with breed group and forage environment were evaluated in a two-phase study. Phase I consisted of milk yield and quality and calf gain records from 1989 to 1991 for purebred Angus (n = 64) and Brahman (n = 62) cows mated to sires of both breeds. Phase II consisted of milk yield and quality and calf gain records from 1991 to 1997 for Angus (n = 94), Brahman (n = 85), Angus x Brahman (n = 86) and Brahman x Angus (n = 93) mated to Polled Hereford sires. In Phase I, forage environments included common bermudagrass and endophyte-infected tall fescue. In Phase II, forage environments included common bermudagrass and endophyte-infected tall fescue (1991 to 1995) and a rotational system of both forages (1995 to 1997) in which each forage was grazed during its appropriate growing season, usually June through October for bermudagrass and November through May for tall fescue. Milk yield was estimated monthly six times during lactation from spring through fall and converted to a 24-h basis. Milk fat, milk protein, and somatic cell count were analyzed by a commercial laboratory. In Phase I, the relation of preweaning ADG to milk yield, milk fat yield, and protein yield was greater (P < 0.05) in Brahman cows on bermudagrass than Angus on bermudagrass. The regression of preweaning ADG on milk yield in Phase I was greater (P < 0.05) for cows on tall fescue than cows which grazed bermudagrass. In Phase II, the relation of preweaning ADG to milk yield, milk fat yield, and milk protein yield was greater or tended to be greater (P < 0.01, P < 0.11, P < 0.01, respectively) in purebred cows compared to reciprocal-cross cows. The regression of preweaning ADG on milk yield and milk protein yield was greater (P < 0.05) on tall fescue than bermudagrass in Phase II. These results suggest that the influence of milk yield and quality on calf growth may differ among breed types and production system, and the efficacy of genetic improvements in milk traits may depend on the breed type and forage environment. Abstract

Reproductive and preweaning data on 190 Angus (A x A), Brahman (B x B), and reciprocal-cross cows (A x B and B x A) and 434 two- and three-breed-cross calves managed on common bermudagrass (BG), endophyte-infected tall fescue (E+), or a combination of both forages (ROT) were used to evaluate the interaction of forage type with individual and maternal heterosis and maternal and grandmaternal breed effects. Cows were born from 1988 to 1991, and calves sired by 13 Polled Hereford bulls were born from 1995 to 1997. Heterosis for calving rate was larger on E+ than on BG or ROT (P < .05), whereas maternal effects were larger on BG than on ROT (P < .10). Maternal heterosis for birth weight was negative on BG (P < .11) but positive on E+ and ROT (P < .10). Grandmaternal effects were evident on BG (P < .10) and E+ (P < .01) but not on ROT. Forage effects were generally substantial for 205-d weight, calf weaning hip height, and calf weaning weight:height ratio; BG was highest, ROT was intermediate, and E+ was lowest. Maternal heterosis for these traits was generally greater on E+ than on BG (P < .10). Grandmaternal effects for 205-d weight, hip height, and weight:height ratio were not important on any forage. Heterosis for weaning weight per cow exposed was substantial on all forages (P < .01) and was significantly greater on E+ (P < .01) than on BG or ROT, but maternal effects were not significant. Thus, we observed more advantage to Brahman-cross cows over purebreds on E+ than on BG. We also observed that moving cows and calves from E+ to BG in the summer will alleviate some, but not all, of the deleterious effects of E+ on calf growth, although it may be more beneficial for reproductive traits in purebred cows. Abstract

Data from 403 Polled Hereford-sired calves from Angus, Brahman, and reciprocal-cross cows were used to evaluate the effects of preweaning forage environment on postweaning performance. Calves were spring-born in 1991 to 1994 and managed on either endophyte-infected tall rescue (E+) or common bermudagrass (BG) during the preweaning phase. After weaning, calves were shipped to the Grazinglands Research Laboratory, El Reno, OK and stratified to one of two winter stocker treatments by breed and preweaning forage; stocker treatments were winter wheat pasture (WW) or native range plus supplemental CP (NR). Each stocker treatment was terminated in March, calves grazed cool-season grasses, and calves were then moved to a feedlot phase in June. In the feedlot phase, calves were fed to approximately 10 mm fat over the 12th rib and averaged approximately 115 d on feed. When finished, calves were weighed and shipped to Amarillo, TX for slaughter. Averaged over calf breed group, calves from E+ gained faster during the stocker phase (P < .10), had lighter starting and finished weights on feed (P < .01), lighter carcass weights (P < .01), and smaller longissimus muscle areas (P < .05) than calves from BG. Calves from E+ were similar to calves from BG in feedlot ADG, percentage kidney, heart, and pelvic fat, fat thickness over 12th rib, yield grade, marbling score, and dressing percentage. Maternal heterosis was larger in calves from E+ for starting weight on feed (P < .01), finished weight (P < .10), and carcass weight (P < . 16). These data suggest that few carryover effects from tall rescue preweaning environments exist, other than lighter, but acceptable, weights through slaughter. These data further suggest that the tolerance to E+ in calves from reciprocal-cross cows, expressed in weaning weights, moderated postweaning weight differences between E+ and BG compared to similar comparisons in calves from purebred cows.
