Abstract

International audience; The increase in egg metabolism triggered by fertilization initiates the meiotic exit and participates in organizing the maternal and paternal centrosomes of the first mitotic spindle. In the mouse, unfertilized oocytes have all constituents needed to organize a bipolar mitotic apparatus but we don't know whether changes in the reducing power of the NAD(P)H co-factor induced by parthenogenetic agents can generate aberrant centriole distribution in parthenogenetic cell lines. In the present experiment, we activated freshly ovulated mouse eggs by a series of Ca2+ signals either in the presence of lactate which is a high cytosolic reducing agent or, in the presence of pyruvate which is a high cytosolic oxidant.

27. Oocytes and Ovarian Follicles As Targets Of Endocrine Disrupters: Consequences For Reproductive Health.

Abstract

This chapter illustrates the physiological characteristics of oocytes and ovarian follicles in relation with their possible exposure to environmental contaminants. The total amount of oocytes present in the adult ovary is established shortly before birth. As soon as the primordial follicle store is established, follicle recruitment begins and it continues without halting for the rest of life or until the ovary is depleted. Between recruitment and ovulation, the oocyte goes through a deep transformation in order to become able to sustain embryonic development. The permanent nature of the oocyte population together with its essential role in supporting embryonic development makes it a sensitive target for the adverse effects of environmental contaminants. Amongst the different environmental contaminants we focussed our analysis on a range of chemicals known as endocrine disrupters (EDs) because of their widespread diffusion and the potential hazard they represent for reproductive health. Their sites of action include the hypothalamus-hypophyseal system, resulting in disruption of the normal pattern of gonadotropin secretion, and the ovary, resulting in destruction of the oocyte. In turn, oocyte destruction can result from directly impairing oocyte viability or from indirect mechanisms involving alterations within the follicular wall. Recent work performed in our laboratory on bovine and pig, has begun to elucidate some of the cellular and molecular mechanisms involved in EDs negative effects on oocyte developmental competence. Our results indicate that EDs perturb maternal mRNA stability and disrupt the physiological remodelling of the cytoplasm, taking place during oocyte maturation. [ABSTRACT FROM AUTHOR]

Abstract

The cytochrome P-450 enzyme system was studied in juvenile Acipenser naccarii (317.6±77 g initial live weight) reared at 23°C and at a standard diet over a period of two month before using for experiments (final weight 546.3±148.8 g). While determining normal levels of cytochrome P-450 in liver tissue its induction was also studied under exposure to β-naphthoflavone (BNF; single dose at 80 mg/Kg body weight 52 hrs prior to sampling). Control fish showed total cytochrome P-450 levels in liver microsomes around 0.40±0.12 nmoles/mg proteins (n=12) while in BNF exposed fish the values increased significantly (0.554±0.11 nmoles/mg proteins; n=12). Ethoxyresorufin-Odeethylase (EROD) in control fish was 0.015±0.02 nmoles/mg proteins/min (n=12); it was found to be NADPH dependent and inducible by BNF. Western Immunoblotting and enzyme-linkedimmunosorbent (ELISA) analyses of liver microsomes from untreated and BNF-exposed fish, using an antibody to rat cytochmme P-450 1Al isoform (P-450 1Al) (Nelson et al., 1996) suggested the presence of a constitutive and BNF-inducible cytochrome P-450 homologous to rat P-450 protein. Measurement of cytochrome P-450-dependent EROD activity and Po450 1A1 expression levels in A. naccarii may be a useful parameter in monitoring environmental pollution. [ABSTRACT FROM AUTHOR]
