Abstract

EFSA and EMA have jointly reviewed measures taken in the EU to reduce the need for and use of antimicrobials in food-producing animals, and the resultant impacts on antimicrobial resistance (AMR). Reduction strategies have been implemented successfully in some Member States. Such strategies include national reduction targets, benchmarking of antimicrobial use, controls on prescribing and restrictions on use of specific critically important antimicrobials, together with improvements to animal husbandry and disease prevention and control measures. Due to the multiplicity of factors contributing to AMR, the impact of any single measure is difficult to quantify, although there is evidence of an association between reduction in antimicrobial use and reduced AMR. To minimise antimicrobial use, a multifaceted integrated approach should be implemented, adapted to local circumstances. Recommended options (non-prioritised) include: development of national strategies; harmonised systems for monitoring antimicrobial use and AMR development; establishing national targets for antimicrobial use reduction; use of on-farm health plans; increasing the responsibility of veterinarians for antimicrobial prescribing; training, education and raising public awareness; increasing the availability of rapid and reliable diagnostics; improving husbandry and management procedures for disease prevention and control; rethinking livestock production systems to reduce inherent disease risk. A limited number of studies provide robust evidence of alternatives to antimicrobials that positively influence health parameters. Possible alternatives include probiotics and prebiotics, competitive exclusion, bacteriophages, immunomodulators, organic acids and teat sealants. Development of a legislative framework that permits the use of specific products as alternatives should be considered. Abstract

Splicing of the FGFR2 K-SAM exon is repressed by hnRNP A1 bound to the exon and activated by TIA-1 bound to the downstream intron. Both proteins are expressed similarly by cells whether they splice the exon or not, so it is important to know which one is dominant. To answer this question, we used bacteriophage PP7 and bacteriophage MS2 coat fusions to tether hnRNP A1 and TIA-1 to distinct sites on the same pre-mRNA molecule. hnRNP A1 fused to one coat protein was tethered to a K-SAM exon containing the corresponding coat protein's binding site. TIA-1 fused to the other coat protein was tethered to the downstream intron containing that coat protein's binding site. This led to efficient K-SAM exon splicing. Our results show that TIA-1 is dominant for K-SAM exon splicing control and validate the combined use of PP7 and MS2 coat proteins for studying posttranscriptional events.

Abstract

International audience; We have isolated, from a human tumor cDNA library, a gene encoding a putative receptor-like protein-tyrosine kinase that we call TK14. The amino acid sequence of the TK14 protein is closely related to the available partial sequence of the mouse protein bek, and more distantly related to the sequences of a chicken basic fibroblast growth factor receptor (73% sequence homology) and the apparent human equivalent of this receptor, the FLG protein (encoded by the fms-like tyrosine kinase gene). Overexpression of the TK14 protein by transfection of COS-1 cells with the corresponding cDNA in a simian virus 40-based expression vector leads to the appearance of new cell-surface binding sites for both acidic and basic fibroblast growth factors. This has been demonstrated by specific binding assays and chemical cross-linking experiments using 125I-labeled growth factors. It appears, therefore, that the human genome contains at least two distinct genes, for TK14 and FLG, that code for related fibroblast growth factor receptors.

Abstract

A 2.3-kb cDNA probe for the human bek fibroblast growth factor receptor was used to determine the chromosomal localization of the corresponding gene by in situ hybridization. The results show that this gene, a form of which is amplified in some poorly differentiated stomach cancers, is localized on chromosome region 10q26. The two previously identified fibroblast growth factor receptor genes are thus not on the same chromosome, as the related flg ("fms-like gene") fibroblast growth factor receptor gene has previously been mapped to human chromosome region 8p12.

Abstract

Activation of c-Fos, by using an inducible c-Fos estrogen receptor fusion protein, triggers the epitheliofibroblastoid cell conversion of mouse mammary epithelial cells. We show that this change in phenotype is accompanied by a definitive switch of the fibroblast growth factor receptor 2 from K-SAM to BEK. This splicing switch occurs a few hours after estrogen stimulation. Our data suggest that Fos proteins could be important in modulating the FGFR-2 splicing choice. Moreover, these observations reinforce previous evidence that the BEK/K-SAM choice is strictly tissue-specific: the K-SAM exon is expressed exclusively in epithelial cells, the BEK exon in cells of the fibroblastic type.

Abstract

Various markers of the inflammatory response were measured in peripheral blood mononuclear cells (PBMCs) and polymorphonuclear neutrophils (PMNs) from 31 dogs with atopic dermatitis (AD). The variables assayed included chemiluminescence, acid hydrolase enzyme concentrations, leukotriene B4 (LTB4) production and complement C3 conversion. The results were compared to those derived from a population of clinically healthy dogs. Dogs with AD exhibited a significant increase in median LTB4 production in PMNs compared to controls (0.94 versus 0.00 ng/ 106 cells; P < 0.01). Significant increases in the median concentrations of intracellular β-galactosidase (PBMC fraction -- 0.42 versus 0.25 mU/106 cells; P < 0.05) (PMN fraction -- 0.47 versus 0.12 mU/106 cells; P < 0.01) and β-glucuronidase (PBMC fraction -- 0.52 versus 0.27 mU/106 cells; P < 0.05) were also evident in the AD group. Although median maximum chemiluminescence values for both leucocyte sub-populations were higher in controls, the differences recorded were not significant (P > 0.05). However, the median time taken to reach maximum chemiluminescence was significantly shorter in the PMN fraction of dogs with AD (7.00 versus 10.00 min; P < 0.01). Atopic dogs had a significant increase in the median percentage conversion of complement C3 to C3b (66.0 versus 57.3%; P < 0.01). The results of this study indicate a priming of the inflammatory response in dogs with AD. The role of LTB4 in the pathogenesis of canine AD and the potential efficacy of leukotriene antagonists in the treatment of this disorder warrant further investigation. [ABSTRACT FROM AUTHOR]

Abstract

Transformed cells obtained after transfecting FR3T3 rat fibroblasts with DNA of bovine papilloma virus type 1 (BPV1) maintained only free copies of the viral genome. Transfection with BPV1 DNA inserted in a bacterial plasmid (pBR322 or pML2) did not produce transformants at a detectable rate, unless the viral sequences had been first excised from the plasmid. In contrast, transfer of the same plasmids by polyethylene glycol‐induced fusion of bacterial protoplasts with FR3T3 rat or C127 mouse cells led to significant transformation frequencies. A total of eight cell lines were studied, three rat and five mouse transformants, obtained with various BPV1 ‐ pML2 recombinants. In all cell lines, both BPV1 and plasmid sequences were maintained as non‐integrated molecules, predominantly as oligomeric forms of the transforming DNA. In the three rat transformants and in two of the mouse lines, parts of the non‐transforming viral region and some bacterial sequences were deleted. In the remaining three mouse lines, the monomeric repeat was a non‐rearranged plasmid molecule which could be re‐established as a plasmid in Escherichia coli after cleavage with “one‐cut” restriction endonucleases and circularization of the molecule.

50. Synthetic donor and acceptor splice sites function in an RNA polymerase B (II) transcription unit.

Abstract

We have synthesised a 32‐bp oligonucleotide containing sequences conforming to the consensus sequences for donor and acceptor splice sites. The oligonucleotide has been inserted into an RNA polymerase B (II) transcription unit and the resulting recombinant used to study the splicing mechanism. Our findings are as follows: (i) the synthetic sites function when separated by several different prokaryotic or eukaryotic DNA fragments providing bulk intron sequence, (ii) intron size need not be greater than 29 bp, (iii) an AG dinucleotide 11 bp upstream from the invariant AG of an acceptor splice site renders the latter non‐functional, and (iv) sequence changes distant from splice sites can affect the efficiency of their utilisation.
