2. Modelling sediment assimilative capacity and organic carbon degradation efficiency at marine fish farms

Abstract

The eutrophication of sediments underlying marine fish farms is one of the major environmental concerns of the industry and regulatory agencies. In this study, a mechanistic model of sediment geochemistry was developed to: (1) determine maximum organic loads that can be degraded without leading to eutrophicated conditions, (2) predict the transition to suboxic conditions as a result of organic enrichment, and (3) predict sulfide levels in surface sediments, a key regulatory variable in Canada. A new definition is proposed for sediment assimilative capacity (AC) of marine fish farms, as the gross deposition rate of organic wastes that maximizes total organic carbon (Corg) degradation rates while preventing sediment sulfide accumulations in surface sediments (upper 2 cm) above regulatory limits (AC-H2S). Model results were consistent with empirical observations and highlight the influence that organic loading history, hydrodynamics, and microbial activity have on assimilative capacity. AC-H2S varied between 0.6 to 22.1 g Corg m-2 d-1 in poorly flushed environments, with no upper limit defined in environments exposed to mean tidal currents >9.5 cm s-1 (dispersion scenarios), where most fish farm organic wastes are dispersed to the far-field or resuspended after deposition below fish cages. The combination of diagenetic modelling and geochemical indicators may contribute significantly to the development of more effective tools for site selection and environmental management of marine fish farms. At this stage, the model should be considered as a proof of concept, developed with the purpose of verifying the utility of assimilative capacity for real-world application. Further validation is still required.

Abstract

The ExoClock project has been created with the aim of increasing the efficiency of the Ariel mission. It will achieve this by continuously monitoring and updating the ephemerides of Ariel candidates over an extended period, in order to produce a consistent catalogue of reliable and precise ephemerides. This work presents a homogenous catalogue of updated ephemerides for 450 planets, generated by the integration of $\sim$18000 data points from multiple sources. These sources include observations from ground-based telescopes (ExoClock network and ETD), mid-time values from the literature and light-curves from space telescopes (Kepler/K2 and TESS). With all the above, we manage to collect observations for half of the post-discovery years (median), with data that have a median uncertainty less than one minute. In comparison with literature, the ephemerides generated by the project are more precise and less biased. More than 40\% of the initial literature ephemerides had to be updated to reach the goals of the project, as they were either of low precision or drifting. Moreover, the integrated approach of the project enables both the monitoring of the majority of the Ariel candidates (95\%), and also the identification of missing data. The dedicated ExoClock network effectively supports this task by contributing additional observations when a gap in the data is identified. These results highlight the need for continuous monitoring to increase the observing coverage of the candidate planets. Finally, the extended observing coverage of planets allows us to detect trends (TTVs - Transit Timing Variations) for a sample of 19 planets. Abstract

Stand density management diagrams (SDMDs) are robust decision-support tools available to forest managers under limited information. SDMDs which are based on empirical models at stand level, graphically represent the temporal relationships among stand density, and different stand variables such as quadratic mean diameter, dominant height, and mean tree volume. They are used to define initial planting spacing or thinning interventions, to meet various management objectives. Nowadays, there is a growing interest in mixed-species forests as an option for adaptive forest management, where they are considered a guarantor to safeguarding a wide variety of ecosystem services within the framework of sustainability. But there is still a lack of knowledge and efficient tools and models for mixed stands such as SDMDs. This study aims to develop an SDMD for Pinus sylvestris L. and Pinus pinaster Ait. mixed stands in the Sierra de la Demanda (Spain) using data from the third Spanish National Forest Inventory. Both species are two of the most important conifers in Europe and the western Mediterranean basin. Different variables can be used to develop an SDMD. In this case, quadratic mean diameter, dominant height, total stand volume, number of trees per hectare, and stand density index were used. These equations were fit by simultaneous fitting including a new variable representing the proportion of both species in the mixed stand. The results of the simultaneous fitting showed the new variable representing the proportion of both species was not significant. Based on that, the SDMD was constructed without including mixture degree. This SDMD can be used by forest managers as an efficient tool to plan thinning operations.

10. Plant conservation in protected areas in Vietnam: an analysis from the Threatened Species Lists

Abstract

In order to guide national conservation policies, lists of nationally threatened plant species are a useful tool. To facilitate the national biodiversity commitments, Vietnam published a conservation management guide. We evaluated the application of these guidelines, focusing on the conservation programs within protected areas across the country. We sent a survey to management offices of 32 protected areas to assess the extent to which the threatened plant species lists have been used in official conservation efforts. We found that the IUCN Red List, the Vietnam Red Data Book and the national decrees are the principal guidelines for conservation prioritization. Besides describing characteristics of conservation programs for threatened plant species, we discovered that the species selected for the programs did not always adhere to the guidelines but showed more or less a rule of thumb in selecting species from lists. We highlight that this is not necessarily a bad implementation if taking into account the shortage of finance and maximizing the number of protected species. Thus, to enhance the utility of the lists for the conservation of threatened plant species, we advise users to recognize that the lists are not always updated, and to employ them as a reasonable reference rather than as the sole guideline.

22. Effect of temperature and nucleotide on the binding of BiP chaperone to a protein substrate

Abstract

BiP (immunoglobulin heavy-chain Binding Protein) is a Hsp70 monomeric ATPase motor that plays broad and crucial roles maintaining proteostasis inside the cell. Structurally, BiP is formed by two domains, a nucleotide-binding domain (NBD) with ATPase activity connected by a flexible hydrophobic linker to the substrate-binding domain (SBD). While the ATPase and substrate binding activities of BiP are allosterically coupled, the latter is also dependent on nucleotide binding. Recent structural studies have provided new insights into BiP's allostery; however, the influence of temperature on the coupling between substrate and nucleotide binding to BiP remains unexplored. Here we study BiP's binding to its substrate at the single molecule level using thermo-regulated optical tweezers which allows us to mechanically unfold the client protein and explore the effect of temperature and different nucleotides on BiP binding. Our results confirm that the affinity of BiP for its protein substrate relies on nucleotide binding, by mainly regulating the binding kinetics between BiP and its substrate. Interestingly, our findings also showed that the apparent affinity of BiP for its protein substrate in the presence of nucleotides remains invariable over a wide range of temperatures, suggesting that BiP may interact with its client proteins with similar affinities even when the temperature is not optimal. Thus, BiP could play a role as a "thermal buffer" in proteostasis. This article is protected by copyright. All rights reserved.

Abstract

The beneficial health effects of (poly)phenol-rich foods such as red grapes mainly depend on both the type and concentration of (poly)phenols. Since fruit (poly)phenol content is influenced by growing conditions, the study examines the seasonal effects of red grapes (Vitis vinifera L.), grown under various cultivation conditions, on metabolic markers of adipose tissue in healthy rats.For this purpose, Fischer 344 rats are exposed into three different light-dark cycles and daily supplemented with 100 mg kg-1 of either conventionally or organically grown red grapes for 10 weeks (n = 6). Seasonal consumption of organic grapes (OGs), which are richer in anthocyanins, increases energy expenditure (EE) of animals exposed to long photoperiod and enhances uncoupling protein 1 (UCP1) protein expression in brown adipose tissue of animals under standard photoperiod. Additionally, red grape consumption affects the gene expression profile of white adipose tissue (WAT), upregulating browning markers of subcutaneous WAT in 12 h light (L12) and 18 h light (L18) photoperiods, and downregulating adipogenic and lipolytic markers of visceral WAT in 6 h light (L6) and L12 photoperiods.These results clearly show that bioactive compounds of grapes can modulate the metabolic markers of white and brown adipose tissues in a photoperiod and depot-dependent manner, partly affecting EE when consumed out of season.© 2023 The Authors. 27. miR15a and miR16 in Chilean type 1 diabetes patients: possible association with apoptosis, inflammatory, or autoimmunity markers

Abstract

Type 1 diabetes mellitus (T1D) is an autoimmune disease characterized by the progressive destruction of β cells, mediated by the interaction between T cells and several cytokines. The pathogenesis of T1D has established its possible relationship with miRNAs. In this study, we analyze the expression profile of miR-15a and miR-16 in peripheral blood mononuclear cells (PBMCs) and their possible association with apoptosis, inflammation, or autoimmunity markers. 38 T1D patients and 41 control subjects were recruited. mRNAs were analyzed by means of qPCR and TaqMan probes. PBMCs were treated with different concentrations of glucose (baseline, 11 and 25 mM) with or without an inflammatory stimulus as TNF-α (10 ng/ml). A decrease in the levels of the miR-15a expression in basal conditions is observed in T1D patients compared to healthy control subjects (relative units 0.5 vs. 1.8, p< 0.05). This change in miR-15a and miR-16 is not affected by the addition of TNF-α. No association is observed with inflammatory markers (IL-6, TNF-α, vCAM) or apoptosis (bcl2 expression). The relationship with immunological markers shows an interaction effect between miR16 and IA-2 (p< 0.03). TNF-α does not affect the expression profile of miR-15a and miR16 in PBMCs. A weak correlation is observed between miR-16 and with the autoimmunity profile (IA-2 autoantibody).

Abstract

A large (M w 7.6) megathrust earthquake occurred on 2016 December 25 in Southern Chile, south of the Chiloé Island (74.2°W, 43.3°S) in the South America–Nazca subduction zone. This earthquake was the first large event in this seismotectonic segment since the M w 9.5 1960 Valdivia megathrust earthquake and broke a ~50-km-long segment of the southern part of its rupture zone. Source parameters are inferred from teleseismic broad-bands, strong motions, GPS, cGPS, InSAR and tide gauge data. We show that the joint inversion significantly improves the resolution of the slip distribution, taking advantage of each data set. Our slip models predict a single slip patch of 70 km × 60 km with a maximum slip of 3.2 ± 0.8 m and a moment magnitude of 7.64. The hypothesis from previous studies that the Chiloé earthquake released energy accumulated before the Valdivia earthquake is not supported by the ensemble of geodetic, seismological and tsunami data. Hence, the Chiloé earthquake most likely released all the strain accumulated in the rupture area since the 1960 earthquake. [ABSTRACT FROM AUTHOR]

Abstract

The recent decision No. 26972 of the Italian Supreme Court (United Sections) of November 11, 2008 fixed the correct interpretation of law about “non pecuniary loss” and the principles that must be applied by the other Italian courts. Nevertheless some aspects of the interpretation provided by the Italian Supreme Court are not so easily applied and the first cases after the decision named above present some problems. This study analyses the critical aspects and the questions presented by the decision of the United Sections’ Italian Supreme Court and the subsequent decisions of the Italian judges.
