Abstract

Cancer neuroscience is a rapidly growing multidisciplinary field that conceptualizes tumors as tissues fully integrated into the nervous system. Recognizing the complexity and challenges in this field is of fundamental importance to achieving the goal of translational impact for cancer patients. Our commentary highlights key scientific priorities, optimal training settings, and roadblocks to translating scientific findings to the clinic in this emerging field, aiming to formulate a transformative and cohesive path forward.

2. Special Educators' Attrition and Retention in the Post-COVID-19 Era: A Systematic Literature Review

Abstract

Special education faces persistent challenges with teacher attrition and retention in the ever-changing landscape of education post COVID-19. Using empirical research and scholarly insights, this systematic review investigates the factors that influence whether special educators choose to continue or leave their jobs following the pandemic. Central to this investigation are the aspects that influence educators' career decisions pre pandemic, such as job satisfaction, working conditions, support systems and opportunities for professional growth. Through this exploration, we gain insights into the reasons why special education teachers choose to stay or leave their roles. The study emphasizes the pressing need to address the issue of losing education teachers and finding replacements. Teacher turnover not only affects student success by reducing teacher effectiveness, but it also disrupts collaborations and negatively impacts those who remain. Drawing on Bronfenbrenner's ecological systems theory framework adds depth to our understanding of these dynamics by providing information that can help improve teacher training programs and assist policymakers in addressing the pressing challenges of special education teacher attrition and retention. This study contributes to a deeper understanding of the factors post COVID-19 that are crucial for making empirical-based changes to support and retain education teachers. Although this study sheds light on the factors influencing teacher turnover and retention it acknowledges its limitations in establishing causal relationships among these factors. Nevertheless, the study's findings lay a groundwork for research and interventions based on evidence to strengthen the special education workforce in the aftermath of the pandemic. Abstract

EXPORTS -EXport Processes in the Ocean from RemoTe Sensing is NASA’s large field campaign focusing on development of a predictive understanding of the export, fate and carbon cycle impacts of the global net primary production. This co-funded program (NSF, private funding1, and international participation) was conceived in 2013, with EXPORTS Science plan published in 2016 (Siegel et al 2016, EXPORTS Writing Team 2015) and its implementation plan finalized in 2016 (EXPORTS Science Definition Team 2016). More details are presented in Siegel et al (2021). EXPORTS campaign is structured as a multiyear effort (Figure 1). It started with “Pre-EXPORTS” modeling and data-mining activity followed by a first phase with two major field programs and a second synthesis and modeling phase. The “Pre-EXPORTS” projects, total of 6 of them (Table 1), funded under A.3 Ocean Biology and Biogeochemistry 2015 call, helped to plan the field campaign (Resplandy et al 2019, Rousseaux & Gregg 2017), and supporting further global synthesis with datasets mined from the literature (e.g. Bisson et al (2020), Bisson et al (2018), Kramer and Siegel (2019)), directly responding to objectives outlined in Science Plan (see section 6 in Team (2015)) and Implementation team (see Figure 1 in Team (2016)). This document presents a compilation of the final reports of the Pre-EXPORTS funded projects, in hope of synthesizing the outcomes, and insuring the legacy of this program. Each of these reports contains a list of published papers, and reader should refer to them to see results in details.

7. Spatial and temporal variation in ambient vibratory noise and its impact potential on a common urban arthropod.

Abstract

Recent, rapid arthropod declines have been tied to human activity. Yet, the propensity for human‐produced noise to disturb the substrate‐borne (vibratory) channel remains virtually untested despite arthropods' ubiquitous use of vibratory information. Characterizing the vibratory landscape at biologically relevant scales is an essential initial step. We aimed to measure vibratory noise across space and time to assess its potential overlap with the urban–rural range and season of a funnel‐weaving spider, Agelenopsis pennsylvanica (C.L. Koch) (Araneae: Agelenidae), a common urban arthropod. We assessed 24‐h ambient vibrations (hereafter vibratory "noise") at 23 sites across urban–rural Lincoln, Nebraska (NE, USA), during the adult season of A. pennsylvanica (August–October). Vibratory noise (amplitude, Leq) varied by ~15 dB across sites overlapping with frequencies within A. pennsylvanica's sensitivity (20–1000 Hz). Urban vibratory noise was positively correlated with the principal component containing estimates of traffic‐induced vibrations (e.g., traffic volume, road proximity, and impervious cover), whereas rural noise levels correlated with probable harvest times. Our findings indicate spatial variation in noise in urban areas and seasonal noise variability in rural areas. We also tested how A. pennsylvanica—collected from four distinct noise survey sites—use their vibratory microhabitat. We assessed daily spider position and dry silk mass within a microcosm that played loud and quiet vibrations (white noise differing by ~15 dB) in separate but connected chambers. Age affected the chamber choice (hereafter "microhabitat use") of spiders collected from the loudest site, as younger adults associated with and used more silk in the loud chamber, but this effect decreased with age. As our data demonstrate that vibratory noise varies with A. pennsylvanica's microhabitat and season with a high potential for impacting behavior, we hypothesize that this environmental noise likely impacts other arthropods as well. Abstract

If environments stay relatively constant over an individual's lifetime, a juvenile that accurately perceives environmental conditions, like population density, may adjust adult traits to better match their environment, thereby increasing success. While previous studies have explored how adult exposure to population density affects physiological and behavioural plasticity, the influence of juvenile density experience on adult traits is less studied.Using the common house cricket, Acheta domesticus, we explored whether perceived acoustic population density during development affected adult physiology and behaviour. We simulated high‐ and low‐densities using live ambient male song. Upon maturation, we measured metabolic (resting respiration) rate, reproductive investment (testes and accessory gland masses), calling song characteristics and aggressive behaviours from pairwise contests between males from different densities.Male rearing density did not affect resting metabolic rates or reproductive organ masses. However, high‐density males had significantly faster, longer chirps, with more pulses—known to be preferred by females—and higher dominant frequency. Low‐density males won more aggressive contests and sang the aggressive song more. Initiation of aggressive behaviours or song and singing more aggressive song were the only other significant predictors of contest outcome.These results suggest that males may plastically adjust calling song characteristics and aggressive behaviour, but not physiology, based on perceived density during development. We hypothesize that alternative mating tactics—that is, territory guarding versus attractive song production—may underlie these observed patterns.Overall, our study highlights the significant influence of early‐life biotic environments on adult behavioural decisions to enhance success in diverse environments. Abstract

The bacterial phytopathogen Pantoea stewartii subsp. stewartii causes leaf blight and Stewart’s wilt disease in susceptible corn varieties. A previous RNA-Seq study examined P. stewartii gene expression patterns during late-stage infection in the xylem, and a Tn-Seq study using a P. stewartii mutant library revealed genes essential for colonization of the xylem. Based on these findings, strains with in-frame chromosomal deletions in the genes encoding seven transcription factors (NsrR, IscR, Nac, Lrp, DSJ_00125, DSJ_03645, and DSJ_18135) and one hypothetical protein (DSJ_21690) were constructed to further evaluate the role of the encoded gene products during in vitro and in planta growth. Assays for capsule production and motility indicate that Lrp plays a role in regulating these two key physiological outputs in vitro. Single infections of each deletion strain into the xylem of corn seedlings determined that Lrp plays a significant role in P. stewartii virulence. In planta xylem competition assays between co-inoculated deletion and the corresponding complementation or wild-type strains as well as in vitro growth curves determined that Lrp controls functions important for P. stewartii colonization and growth in corn plants, whereas IscR may have a more generalized impact on growth. Defining the role of essential transcription factors, such as Lrp, during in planta growth will enable modeling of key components of the P. stewartii regulatory network utilized during growth in corn plants.

Abstract

Porcine deltacoronavirus (δ-CoV) is the object of extensive research in several countries including the United States. In contrast, the epidemiology of δ-CoVs in wild birds in the US is largely unknown. Our aim was to comparatively assess the prevalence of δ- and γ-CoVs in wild migratory terrestrial and aquatic birds in Arkansas, Illinois, Indiana, Maryland, Mississippi, Missouri, Ohio, Tennessee and Wisconsin. A total of 1236 cloacal/fecal swabs collected during the period 2015−2018 were tested for γ- and δ-CoVs using genus-specific reverse transcription-PCR assays. A total of 61 (4.99%) samples were γ-CoV positive, with up to 29 positive samples per state. In contrast, only 14 samples were positive for δ-CoV (1.14%) with only 1−4 originating from the same state. Thus, unlike previous reports from Asia, γ-CoVs are more prevalent than δ-CoVs in the US, suggesting that δ-CoVs may spread in birds with lower efficiency. This may indicate δ-CoV emerging status and incomplete adaptation to new host species limiting its spread. Phylogenetic analysis of the partial N gene revealed that the newly identified δ-CoV strains were most closely related to the HKU20 (wigeon) strain. Further studies are necessary to investigate the role of aquatic bird δ-CoVs in the epidemiology of δ-CoVs in swine and terrestrial birds.

The bacterial phytopathogen Pantoea stewartii subsp. stewartii causes leaf blight and Stewart's wilt disease in susceptible corn varieties. A previous RNA-Seq study examined P. stewartii gene expression patterns during late-stage infection in the xylem, and a Tn-Seq study using a P. stewartii mutant library revealed genes essential for colonization of the xylem. Based on these findings, strains with in-frame chromosomal deletions in the genes encoding seven transcription factors (NsrR, IscR, Nac, Lrp, DSJ_00125, DSJ_03645, and DSJ_18135) and one hypothetical protein (DSJ_21690) were constructed to further evaluate the role of the encoded gene products during in vitro and in planta growth. Assays for capsule production and motility indicate that Lrp plays a role in regulating these two key physiological outputs in vitro. Single infections of each deletion strain into the xylem of corn seedlings determined that Lrp plays a significant role in P. stewartii virulence. In planta xylem competition assays between co-inoculated deletion and the corresponding complementation or wild-type strains as well as in vitro growth curves determined that Lrp controls functions important for P. stewartii colonization and growth in corn plants, whereas IscR may have a more generalized impact on growth. Defining the role of essential transcription factors, such as Lrp, during in planta growth will enable modeling of key components of the P. stewartii regulatory network utilized during growth in corn plants.

Abstract

Maize is one of the world's most widely cultivated crops. As future demands for maize will continue to rise, fields will face ever more frequent and extreme weather patterns that directly affect crop productivity. Development of environmentally resilient crops with improved standability in the field, like wheat and rice, was enabled by shifting the architecture of plants to a short stature ideotype. However, such architectural change has not been implemented in maize due to the unique interactions between gibberellin (GA) and floral morphology which limited the use of the same type of mutations as in rice and wheat. Here, we report the development of a short stature maize ideotype in commercial hybrid germplasm, which was generated by targeted suppression of the biosynthetic pathway for GA. To accomplish this, we utilized a dominant, miRNA-based construct expressed in a hemizygous state to selectively reduce expression of the ZmGA20ox3 and ZmGA20ox5 genes that control GA biosynthesis primarily in vegetative tissues. Suppression of both genes resulted in the reduction of GA levels leading to inhibition of cell elongation in internodal tissues, which reduced plant height. Expression of the miRNA did not alter GA levels in reproductive tissues, and thus, the reproductive potential of the plants remained unchanged. As a result, we developed a dominant, short-stature maize ideotype that is conducive for the commercial production of hybrid maize. We expect that the new maize ideotype would enable more efficient and more sustainable maize farming for a growing world population.

This article describes students’ emotional intelligence (EI) development when participating in the Trans-Atlantic and Pacific Project (TAPP) in two technical and professional communication (TPC) courses. The researchers used modified grounded theory to compile the emotions used for coding students’ weekly reflections, and content analyzed how the TAPP experience affected students’ EI development. Overall, the article emphasizes the importance of supporting TPC students’ EI development in low-stakes environments since EI directly impacted their actions when collaborating.

Environmental change, especially frequent droughts, is predicted to detrimentally impact the North American perennial grasslands. Consistent dry spells will affect plant communities as well as their associated rhizobiomes, possibly altering the plant host performance under environmental stress. Therefore, there is a need to understand the impact of drought on the rhizobiome, and how the rhizobiome may modulate host performance and ameliorate its response to drought stress. In this study, we analyzed bacterial and fungal communities in the rhizospheres of three ecotypes (dry, mesic, and wet) of a dominant prairie grass,Andropogon gerardii. The ecotypes were established in 2010 in a common garden design and grown for a decade under persistent dry conditions at the arid margin of the species’ range in Colby Kansas. The experiment aimed to answer whether and to what extent do the different ecotypes maintain or recruit distinct rhizobiomes after ten years in an arid climate. In order to answer this question, we screened the bacterial and fungal rhizobiome profiles of the ecotypes under the arid conditions of western KS as a surrogate for future climate environmental stress using 16S rRNA and ITS2 metabarcoding sequencing. Under these conditions, bacterial communities differed compositionally among theA. gerardiiecotypes, whereas the fungal communities did not. The ecotypes were instrumental in driving the differences among bacterial rhizobiomes, as the ecotypes maintained distinct bacterial rhizobiomes even after ten years at the edge of the host species range. This study will aid us to optimize plant productivity through the use of different ecotypes under future abiotic environmental stress, especially drought.

Abstract

Complex-terrain locations often have repeatable near-surface wind patterns, such as synoptic gap flows and local thermally forced flows. An example is the Columbia River Valley in east-central Oregon-Washington, a significant wind-energy-generation region and the site of the Second Wind-Forecast Improvement Project (WFIP2). Data from three Doppler lidars deployed during WFIP2 define and characterize summertime wind regimes and their large-scale contexts, and provide insight into NWP model errors by examining differences in the ability of a model [NOAA’s High-Resolution Rapid-Refresh (HRRR-version1)] to forecast wind-speed profiles for different regimes. Seven regimes were identified based on daily time series of the lidar-measured rotor-layer winds, which then suggested two broad categories. First, in three regimes the primary dynamic forcing was the large-scale pressure gradient. Second, in two regimes the dominant forcing was the diurnal heating-cooling cycle (regional sea-breeze-type dynamics), including the marine intrusion previously described, which generates strong nocturnal winds over the region. The other two included a hybrid regime and a non-conforming regime. For the large-scale pressure-gradient regimes, HRRR had wind-speed biases of ~1 m s−1 and RMSEs of 2-3 m s−1. Errors were much larger for the thermally forced regimes, owing to the premature demise of the strong nocturnal flow in HRRR. Thus, the more dominant the role of surface heating in generating the flow, the larger the errors. Major errors could result from surface heating of the atmosphere, boundary-layer responses to that heating, and associated terrain interactions. Measurement/modeling research programs should be aimed at determining which modeled processes produce the largest errors, so those processes can be improved and errors reduced.

Abstract

Post-translational modifications (PTMs) occur during or after protein biosynthesis and increase the functional diversity of proteome. They comprise phosphorylation, acetylation, methylation, glycosylation, ubiquitination, sumoylation (among many other modifications), and influence all aspects of cell biology. Mass-spectrometry (MS)-based proteomics is the most powerful approach for PTM analysis. Despite this, it is challenging due to low abundance and labile nature of many PTMs. Hence, enrichment of modified peptides is required for MS analysis. This review provides an overview of most common PTMs and a discussion of current enrichment methods for MS-based proteomics analysis. The traditional affinity strategies, including immunoenrichment, chromatography and protein pull-down, are outlined together with their strengths and shortcomings. Moreover, a special attention is paid to chemical enrichment strategies, such as capture by chemoselective probes, metabolic and chemoenzymatic labelling, which are discussed with an emphasis on their recent progress. Finally, the challenges and future trends in the field are discussed.

Abstract

The Ozone Profiling Atmospheric Lidar is a scanning four-wavelength ultraviolet differential absorption lidar that measures tropospheric ozone and aerosols. Derived profiles from the lidar data include ozone concentration, aerosol extinction, and calibrated aerosol backscatter. Aerosol calibrations assume a clear air region aloft. Other products include cloud base heights, aerosol layer heights, and scans of particulate plumes from aircraft. The aerosol data range from 280 m to 12 km with 5 m range resolution, while the ozone data ranges from 280 m to about 12 km with 100 m resolution. In horizontally homogeneous atmospheres, data from multiple-elevation angles is combined to reduce the minimum altitude of the aerosol and ozone profiles to about 20 m. The lidar design, the characterization of the photomultiplier tubes, ozone and aerosol analysis techniques, and sample data are described. Also discussed is a double-gating technique to shorten the gated turn-on time of the photomultiplier tubes, and thereby reduce the detection of background light and the outgoing laser pulse. 41. Efficient Polyhydroxyalkanoate Production by Rhodopseudomonas Palustris from Lignocellulosic Biomass

Abstract

Polyhydroxyalkanoates (PHAs) are biopolymers produced by bacteria with the potential to replace conventional plastics. However, the relatively high production costs of PHAs are keeping them from market acceptance, with approximately half of the production costs derived from the feedstock. Thus, engineering a microbe for PHA production from cheaper and renewable carbon sources is necessary to promote the valorization of PHAs. Lignocellulosic biomass is considered to be one of the most economic carbon sources in the world, and is thus an attractive candidate for cheaper production of bioplastics. Rhodopseudomonas palustris CGA009is a metabolically robust bacterium capable of catabolizing lignin breakdown products (LBPs),and also has the ability to produce several high-value bioproducts like bioplastics and biohydrogen. Thus, the goal of this research was aimed at producing and optimizing PHA production from R. palustris from LBPs. The first study produced poly-3-hydroxybutyrate(PHB) from R. palustris from the LBP p-coumarate with a PHB titer of 0.41 g/L and 68.4%carbon conversion efficiency. This study also optimized a high-throughput quantification method for PHB that employed flow cytometry. The second study produced poly(3-hydroxybutyrate-co3-hydroxyvalerate) (PHBV) from the LBPs p-coumarate and coniferyl alcohol, and utilized an integrated experimental and computational modeling approach to infer metabolic factors controlling PHB production that can be expanded to any PHB-producing microbe with similar metabolic features. The third study expressed the phaP1 phasin gene from the PHB-producing model bacterium Cupriavidus necator H16 in R. palustris for the overproduction of PHBV onLBPs. Expression of phaP1 yielded PHBV production from R. palustris aerobically (0.7 g/L),which does not occur in the wild type strain and provides more flexibility for industrial production. Abstract

In multiple sclerosis (MS) functional changes in connectivity due to cortical reorganization could lead to cognitive impairment (CI), or reflect a re-adjustment to reduce the clinical effects of widespread tissue damage. Such alterations in connectivity could result in changes in neural activation as assayed by executive function tasks. We examined cognitive function in MS patients with mild to moderate cognitive impairment and age-matched controls. We evaluated brain activity using functional magnetic resonance imaging (fMRI) during the successful performance of the Wisconsin-card sorting (WCS) task by MS patients, showing compensatory maintenance of normal function, as measured by response latency and error rate. To assess changes in functional connectivity throughout the brain, we performed a global functional brain network analysis by computing voxel by voxel correlations on the fMRI time series data and carrying out a hierarchical cluster analysis. We found that during the WCS task there is a significant reduction in the number of smaller size brain functional networks, and a change in the brain areas representing the nodes of these networks in MS patients compared to age-matched controls. There is also a concomitant increase in the strength of functional connections between brain loci separated at intermediate scale distances in these patients. These functional alterations might reflect compensatory neuroplastic reorganization underlying maintenance of relatively normal cognitive function in the face of white matter lesions and cortical atrophy produced by MS.
