Summary: Background: Cellulitis is a commonly recognised condition in first opinion equine practice yet there is little published information regarding its diagnosis, treatment and management. Objectives: To describe current approaches of veterinary surgeons working in the United Kingdom (UK) to the diagnosis, treatment and management of distal limb cellulitis in horses. Study design: Cross‐sectional survey. Methods: An online questionnaire was distributed via email and social media sites for a period of 12 weeks. Descriptive statistics were produced and chi‐square or Fisher's exact tests were used to assess associations between categorical variables, with critical probability set at 0.05. Results: Distal limb cellulitis is frequently encountered in the UK, with 83.5% (n = 224/268) of respondents having treated >5 cases in the preceding 12 months. Diagnosis was predominantly made based on clinical signs alone (89.2%; n = 239/268). The majority of respondents (95.5%; n = 256/268) treated cases with nonsteroidal anti‐inflammatory drugs (NSAIDs) and antimicrobials. Phenylbutazone (93.8%; n = 240/256) and trimethoprim sulphonamide (TMPS) (72.3%; n = 185/256) were most frequently prescribed and 61.7% (n = 148/240) of respondents prescribed these drugs in combination. Duration of treatment was most frequently five days for NSAIDs (33.5%; n = 64/191), TMPS (84.4%; n = 119/141) and doxycycline (62.5%; n = 25/40), the longest prescribed duration of any treatments being 10 days. Corticosteroids were administered by 41.0% of respondents (n = 110/268). Management recommendations included cold hosing/ice packing of the affected limb (79.4%; n = 212/267) with light exercise (turnout or hand walking) if the horse was comfortable enough to do so (97.4%; n = 260/267). Main limitations: Small sample size (n = 268) and low level of item omission for a small number of questions. Conclusions: Cellulitis is common in the UK and is usually treated with NSAIDs and antimicrobials for 5–10 days, plus cold hosing and light exercise. Further study on the topics of diagnosis and treatment, with particular emphasis on the aetiology and necessity of antimicrobials, is warranted. [ABSTRACT FROM AUTHOR]

Squids of the family Onychoteuthidae are ecologically important in pelagic food webs and have been reported from every ocean except the Arctic. Although they are abundant and caught frequently as bycatch in fisheries, the biogeography of many species remains poorly understood. Species identification within the Atlantic Ocean is usually restricted to two species: Onychoteuthis banksii and Onykia carriboea. Here, we report the occurrence of four species of the family Onychoteuthidae (Onychoteuthis cf. banksii, Onykia carriboea, Walvisteuthis jeremiahi, and Onychoteuthis sp. AL 2) from the Sargasso Sea in the western Atlantic, identified using DNA barcoding (cytochrome c oxidase subunit I) and morphology. Our results have doubled the known onychoteuthid biodiversity in the Sargasso Sea, which has implications for the ecology of this oceanic region. [ABSTRACT FROM AUTHOR]

Phylogenies for Octopoda have, until now, been based on morphological characters or a few genes. Here we provide the complete mitogenomes and the nuclear 18S and 28S ribosomal genes of twenty Octopoda specimens, comprising 18 species of Cirrata and Incirrata, representing 13 genera and all five putative families of Cirrata (Cirroctopodidae, Cirroteuthidae, Grimpoteuthidae, Opisthoteuthidae and Stauroteuthidae) and six families of Incirrata (Amphitretidae, Argonautidae, Bathypolypodidae, Eledonidae, Enteroctopodidae, and Megaleledonidae) which were assembled using genome skimming. Phylogenetic trees were built using Maximum Likelihood and Bayesian Inference with several alignment matrices. All mitochondrial genomes had the 'typical' genome composition and gene order previously reported for octopodiforms, except Bathypolypus ergasticus, which appears to lack ND5, two tRNA genes that flank ND5 and two other tRNA genes. Argonautoidea was revealed as sister to Octopodidae by the mitochondrial protein-coding gene dataset, however, it was recovered as sister to all other incirrate octopods with strong support in an analysis using nuclear rRNA genes. Within Cirrata, our study supports two existing classifications suggesting neither is likely in conflict with the true evolutionary history of the suborder. Background: Dental caries is one of the most prevalent non-communicable disease globally and can have serious health sequelae impacting negatively on quality of life. In the UK most adults experience dental caries during their lifetime and the 2009 Adult Dental Health Survey reported that 85% of adults have at least one dental restoration. Conservative removal of tooth tissue for both primary and secondary caries reduces the risk of failure due to tooth-restoration, complex fracture as well as remaining tooth surfaces being less vulnerable to further caries. However, despite its prevalence there is no consensus on how much caries to remove prior to placing a restoration to achieve optimal outcomes. Evidence for selective compared to complete or near-complete caries removal suggests there may be benefits for selective removal in sustaining tooth vitality, therefore avoiding abscess formation and pain, so eliminating the need for more complex and costly treatment or eventual tooth loss. However, the evidence is of low scientific quality and mainly gleaned from studies in primary teeth., Method: This is a pragmatic, multi-centre, two-arm patient randomised controlled clinical trial including an internal pilot set in primary dental care in Scotland and England. Dental health professionals will recruit 623 participants over 12-years of age with deep carious lesions in their permanent posterior teeth. Participants will have a single tooth randomised to either the selective caries removal or complete caries removal treatment arm. Baseline measures and outcome data (during the 3-year follow-up period) will be assessed through clinical examination, patient questionnaires and NHS databases. A mixed-method process evaluation will complement the clinical and economic outcome evaluation and examine implementation, mechanisms of impact and context. The primary outcome at three years is sustained tooth vitality. The primary economic outcome is net benefit modelled over a lifetime horizon. Clinical secondary outcomes include pulp exposure, progession of caries, restoration failure; as well as patient-centred and economic outcomes., Discussion: SCRiPT will provide evidence for the most clinically effective and cost-beneficial approach to managing deep carious lesions in permanent posterior teeth in primary care. Validated assessment protocols have been developed to quantify welfare states for intensively managed sport, pleasure, and working horses. There are few protocols for extensively managed or free-roaming populations. Here, we trialed welfare indicators to ascertain their feasibility, reliability, and repeatability using free-roaming Carneddau Mountain ponies as an example population. The project involved (1) the identification of animal and resource-based measures of welfare from both the literature and discussion with an expert group; (2) testing the feasibility and repeatability of a modified body condition score and mobility score on 34 free-roaming and conservation grazing Carneddau Mountain ponies; and (3) testing a prototype welfare assessment template comprising 12 animal-based and 6 resource-based welfare indicators, with a total of 20 questions, on 35 free-roaming Carneddau Mountain ponies to quantify inter-assessor reliability and repeatability. This pilot study revealed that many of the indicators were successfully repeatable and had good levels of inter-assessor reliability. Some of the indicators could not be verified for reliability due to low/absent occurrence. The results indicated that many animal and resource-based indicators commonly used in intensively managed equine settings could be measured in-range with minor modifications. This study is an initial step toward validating a much-needed tool for the welfare assessment of free-roaming and conservation grazing ponies.

The Southern Hemisphere long-finned pilot whale (Globicephala melas edwardii) is a top predator in the New Zealand pelagic food web, feeding predominantly on arrow squids. This study quantified trace element concentrations (Al, As, Cd, Co, Cr, Cu, Fe, Hg, Ni, Mn, Pb, Se, V, Zn) in four tissues (blubber, kidney, liver, muscle) from 21 individuals from stranding sites in New Zealand. Maximum Cd and Hg concentrations were measured in liver and kidney, respectively. Selenium had a positive correlation with Cd and Hg, suggesting the involvement of Se in Cd and Hg detoxification. Arrow squids from the whales' stomach contents were DNA barcoded and identified as Nototodarus sloanii. Trace element concentrations were measured in squid samples from the whale stomach contents. The significant correlation for Hg between the squid tissue and the whale tissue suggests that arrow squids play a major role in trace element uptake by G. m. edwardii., (Copyright © 2021 Elsevier Ltd. The New Zealand arrow squids, Nototodarus gouldi and N. sloanii, play an important role in the marine food web, and are both economically important fishery species. This study compares trace element concentrations (As, Cd, Co, Cr, Cu, Fe, Hg, Ni, Mn, Pb, U, and Zn) in these animals from different fishing locations within New Zealand's Exclusive Economic Zone (Auckland Islands, Chatham Rise, Dunedin, Golden Bay, Taranaki, and West Coast of the South Island). Muscular mantle (the tissue usually consumed by humans) and digestive gland tissue (the primary organ for trace element accumulation) concentrations were compared among regions, revealing size, species, and sampling location effects. Overall, N. gouldi had higher concentrations of As and Zn. The Taranaki region had the highest concentrations for Cd, Co, Cu, and Zn in both tissues analysed. To assess the bioaccessibility of these trace elements to humans, an in-vitro digestion simulation experiment was conducted, revealing bioavailable concentrations overall below the maximum levels presently considered tolerable for human consumption. However, the in-vitro digestion had no observable effect upon Cd concentrations in arrow squid mantle tissue (implying that the measured concentrations are bioaccessible to humans), and revealed that Cd had the highest potential impact on marine food webs and human health. Background: Traditionally, patients at low risk and high risk of developing dental disease have been encouraged to attend dental recall appointments at regular intervals of six months between appointments. The lack of evidence for the effect that different recall intervals between dental check-ups have on patient outcomes, provider workload and healthcare costs is causing considerable uncertainty for the profession and patients, despite the publication of the NICE Guideline on dental recall. The need for primary research has been highlighted in the Health Technology Assessment Group's systematic review of routine dental check-ups, which found little evidence to support or refute the practice of encouraging 6-monthly dental check-ups in adults. The more recent Cochrane review on recall interval concluded there was insufficient evidence to draw any conclusions regarding the potential beneficial or harmful effects of altering the recall interval between dental check-ups. There is therefore an urgent need to assess the relative effectiveness and cost-benefit of different dental recall intervals in a robust, sufficiently powered randomised control trial (RCT) in primary dental care., Methods: This is a four year multi-centre, parallel-group, randomised controlled trial with blinded outcome assessment based in dental primary care in the UK. Practitioners will recruit 2372 dentate adult patients. Patient participants will be randomised to one of three groups: fixed-period six month recall, risk-based recall, or fixed-period twenty-four month recall. Outcome data will be assessed through clinical examination, patient questionnaires and NHS databases. Human fibroblast cultures derived from normal donors of different ages were examined for the proportion of spindle-shaped, sail-shaped, and large pleomorphic cells which constitute the cultures, and the capacities of these cells for unscheduled DNA synthesis (UDS). The large pleomorphic cells were most abundant in all cultures, followed in abundance by the sail-shaped and spindle-shaped cells. Major changes in the occurrence of the cells of different shape with increased donor age were not apparent, although the abundance of spindle-shaped cells decreased slightly with donor age. The large pleomorphic and sail-shaped cells exhibited similar UDS which was consistently greater than that of the spindle-shaped cells. No major changes in UDS were observed for either type of cell with increased donor age.
