Kontext: Für Fußballspieler*innen ist eine regelmäßige physiotherapeutische Behandlung wichtig, da ihre Körper ständig einer hohen Belastung, als auch Überlastungsverletzungen ausgesetzt sind. Ein Kreuzbandriss verstärkt diesen Umstand um so mehr, da die Stabilität des Kniegelenkes wieder aufgebaut werden muss. Aus diesem Grund wurden im Rahmen dieser Arbeit österreichische Profifußballer, die während ihrer Karriere einen Kreuzbandriss erlitten hatten zu ihren physiotherapeutischen Behandlungen befragt. Die Ergebnisse sollen Aufschluss darüber geben wie man den Bedürfnissen von Kreuzbandrisspatient*innen im Profifußball am besten gerecht werden und die Qualität der Versorgung verbessern kann. Methode: Um die Forschungsfrage zu beantworten, wurde ein qualitatives Forschungsdesign gewählt. Im Rahmen der Datenerhebung wurden fünf leitfadengestützte Interviews mit männlichen Fußballspielern, welche zum Zeitpunkt ihres vorderen Kreuzbandrisses in österreichischen Profiligen spielten, geführt. Die Interviews wurden mittels qualitativer Inhaltsanalyse nach Mayring ausgewertet. Hauptergebnisse: Der Kreuzbandriss war die schwerste Verletzung der Betroffenen, die sie in ihren Karrieren am meisten zurückwarf und für die Heilung und den Wiedereinstieg in den Fußball die längste Zeit benötigte. Nach einer Kreuzbandruptur spielte die Physiotherapie eine entscheidende Rolle, um einen guten Heilungserfolg zu erreichen, als auch bestmöglich wieder in den Fußballsport einsteigen zu können. Physiotherapeut*innen kommt im Falle einer Verletzung eine besondere Rolle zu. Sie gelten als Expert*innen in der Heilungs- und Wiederaufbauphase und werden für den Heilungserfolg als verantwortlich angesehen. Schlussfolgerung: Physiotherapeut*innen wird viel Vertrauen entgegengebracht, aber auch die Erwartungshaltung dieses nicht zu enttäuschen. Für die physiotherapeutische Arbeit in der Praxis ist es förderlich professionelle Leistungs- und Wettkampfsportler als eine eigene Patient*innengruppe, mit besonderen Bedürfnissen und Wünschen, zu betrachten und ihnen die nötige Aufmerksamkeit zukommen zu lassen. So sollte für Physiotherapeut*innen im Rahmen der Therapie nicht nur die körperliche Behandlung im Vordergrund stehen, sondern auch die psychische Befindlichkeit bedacht und in die Therapie miteinbezogen werden. Context: Regular physiotherapy treatment is important for soccer players as their bodies are constantly exposed to high levels of stress and overuse injuries. A cruciate ligament rupture exacerbates this situation all the more because the stability of the knee joint has to be rebuilt. For this reason, Austrian professional soccer players who had suffered a cruciate ligament rupture during their career were asked about their physiotherapeutic treatments as part of this study. The results should provide information on how to best meet the needs of cruciate ligament rupture patients in professional football and how to improve the quality of care. Method: A qualitative research design was chosen to answer the research question. As part of the data collection, five guided interviews were conducted with male soccer players who were playing in Austrian professional leagues at the time of their anterior cruciate ligament tear. The interviews were evaluated using qualitative content analysis according to Mayring. Main results: The cruciate ligament rupture was the most serious injury suffered by those affected, which set them back the most in their careers and required the longest time to heal and return to football. After a cruciate ligament rupture, physiotherapy plays a crucial role in order to achieve good healing success and to be able to get back into football in the best possible way. Physiotherapists have a special role to play in the event of an injury. They are considered experts in the healing and reconstruction phase and are seen as responsible for the healing success. Conclusion: There is a lot of trust in physiotherapists, but also the expectation not to disappoint them. For physiotherapeutic work in practice, it is beneficial to regard professional competitive athletes as a separate group of patients with special needs and wishes and to give them the necessary attention. For physiotherapists, not only the physical treatment should be the focus of the therapy, but also the mental state should be considered and included in the therapy.

Ülkeler Gümrük Tarifeleri ve Ticaret Genel Anlaşması (GATT) ve Dünya Ticaret Örgütü (DTÖ) nezdinde yürütülen görüşmeler ile bu kuruluşlar nezdinde verdikleri birtakım taahhütler neticesinde gümrük vergilerinde indirime giderek, bir noktaya kadar ticaretin serbestleştirilmesini sağlamıştır. Ancak bu durum hiç de göründüğü gibi değildir, zira bir taraftan DTÖ nezdindeki taahhütler doğrultusunda gümrük vergileri azaltılırken, diğer taraftan uygulanan Tarife Dışı Önlemlerin (TDÖ) sayısı arttırılmıştır. Günümüzde TDÖ’ler ticaret maliyetlerinin önemli bir bölümünü oluşturmaktadır. TDÖ’lerin ticaret akışları üzerindeki etkilerinin tahmin edilmesinde kullanılan en etkili araçlardan birisi ise çekim modeli olup, TDÖ’lerin etkilerinin tahmin edilmesinde yaygın olarak kullanılmaktadır. Belirtilen nedenlerle, bu çalışmanın temel amacı, TDÖ’lerin ticaret akışını ne yönde etkilediğine ilişkin çekim modeli kullanılarak analiz edilmesinde kullanılan yöntemleri, bu alandaki literatürde kaydedilen son gelişmeler ışığında ele almak ve ticaret politikası analizinin nasıl yapılacağına ilişkin çeşitli öneriler sunmaktır. Countries have strengthened the liberalization of trade up to a point as a result of some negotiations with General Agreement on Tariffs and Trade (GATT) and World Trade Organization (WTO), and one consequence of the commitments they have made with these institutions is a reduction in customs duties. However, this situation is not at all as it seems, because on the one hand, while customs duties were reduced in line with the commitments before WTO, on the other hand, the number of applied Non-Tariff Measures (NTMs) were increased. Today, NTMs constitute a significant part of trade costs. One of the most effective tools used to predict the effects of the NTMs on trade flows is the Gravity Model, and it is widely used to predict the effects of the NTMs. For the stated reasons, the main purpose of this study is to discuss the methods used in analyzing the NTMs by showing how the Gravity Model affects the trade flow in the light of recent developments in the literature and to provide various suggestions on how to conduct a trade policy analysis.

Bu araştırmada, konut kullanıcılarının döşemeli mobilyalarını kullanım süreleri, değiştirme nedenleri, satın alma şekilleri ve satın alırken dikkate aldığı hususların belirlenmesi amaçlanmıştır. Bu maksatla hazırlanmış olan ankete toplam 105 konut kullanıcısı katılmış olup, elde edilen sonuçlar SPSS programıyla değerlendirilmiştir. Araştırma sonuçlarına göre, konut kullanıcılarının yarısından fazlasının döşemeli mobilyalarını 3-8 yıl kullandığı, %50’den fazlasının ürünlerin eskimesi nedeniyle değiştirmek istediği ve kullanıcıların %82’sinin döşemeli mobilyalarını mağazadan satın aldığı tespit edilmiştir. Ayrıca, konut kullanıcılarının döşemeli mobilyaları satın alırken %17,2’si demonte olmasına, %14,7’si malzeme kalitesine, %12,8’i ölçü uyumuna, %23,5’i koltuk kılıfının yıkanabilir olmasına, %16,7’si kullanışlı olmasına ve %16,9’u ise modaya uygun olmasına dikkate aldığı belirlenmiştir., In this study, it was aimed to determine the usage time of the upholstered furniture, the reasons for changing, the ways of purchasing and the issues that are taken into consideration while purchasing. A total of 105 residential users participated in the survey prepared for this purpose, and the results were evaluated with the SPSS program. According to the results of the research, it has been determined that more than half of the residential users use their upholstered furniture for 3-8 years, more than 50% want to replace them due to the aging of the products, and 82% of the users buy their upholstered furniture from the store. In addition, when purchasing upholstered furniture for residential users, 17.2% disassemble, 14.7% material quality, 12.8% size fit, 23.5% seat cover washable, %16.7 useful and 16.9% being trendy.

ÖZET Dünya Ticaret Örgütü'nün (DTÖ/GATT) nezdinde gerçekleştirilen müzakerelerin ardından, geleneksel ticaret engeli olarak kabul edilen gümrük tarifesi oranlarında ciddi indirimler sağlanmış ve bu kapsamda tarife dışı önlemler daha da önem kazanmıştır. Tarife Dışı Önlemlerin (TDÖ) ticaret politikası aracı olarak kullanımının artmasına yönelik oluşan bu yeni eğilimle birlikte 2009 küresel ekonomik ve mali krizinden bu yana dünya ticaretindeki durgun büyüme, yatırım faaliyetleri ile ticareti azaltan yeni korumacı programların artacağına dair endişeleri arttırmıştır. Söz konusu bu endişeler, TDÖ’lerin ticaret politikası aracı olarak kullanılmasına yönelik tartışmaları da beraberinde getirmiştir. Ticaret politikası aracı olarak TDÖ’lerin kullanımının yayılma hızı, bu alandaki literatüre TDÖ’lerin ekonomik etkilerine yönelik yapılan araştırmalar şeklinde yansımıştır. Bu noktadan hareketle, bu çalışmada Türkiye’nin otomotiv sektörü ithalatına uygulamış olduğu TDÖ’lerin etkileri yapısal çekim modeli kullanılarak analiz edilmiştir. Söz konusu analizde, çekim modeli literatüründe yer alan en güncel bilgiler de dikkate alınarak, Türkiye’nin seçilmiş 149 ülkeden 1995-2018 yılları arasında gerçekleştirmiş olduğu otomotiv ana ve yan sanayii ithalat verileri kullanılarak, TDÖ’lerin otomotiv sektörüne olan etkileri araştırılmıştır. Sektörü oluşturan eşyalar özelinde ve sektörel düzeyde yapılan analizler sonucunda her bir TDÖ için AVE hesaplamaları gerçekleştirilerek örtük koruma oranları hesaplanmıştır. Gerçekleştirilen analizler sonucunda, uygulanan TDÖ’lerin genel olarak otomotiv sektörü üzerinde olumsuz etki yarattığı görülmüştür. Bu itibarla eşyalar özelinde mikro ölçekte yürütülen analizlerde sektör ithalatına en çok engel teşkil eden önlem türlerinin sırasıyla, Ticarette Teknik Engeller, anti damping ve telafi edici vergileri de kapsayan Korunma Önlemleri ile Fiyat Kontrol Önlemleri olduğu sonucuna varılmıştır. Bunun yanında sektörel düzeyde yürütülen makro ölçekli analizde ise yalnızca Korunma Önlemlerinin sektör ithalatına dair genel bir koruma sağladığı sonucuna varılmıştır. Anahtar Kelimeler: Uluslararası Ticaret, Otomotiv Sektörü, Çekim Modeli, Tarife Dışı Önlemler, Sayısallaştırma ABSTRACT Following the negotiations held with the World Trade Organization (WTO / GATT), significant reductions were achieved in tariff rates, which are considered traditional trade barriers, and in this context, Non-Tariff Measures (NTMs) gained more importance. The sluggish growth in world trade since the 2009 global economic and financial crisis together with this new trend towards the increase in the use of NTMs as a trade policy tool, has raised concerns that investment activities and new protectionist programs will reduce trade. These concerns brought about discussions on the use of NTMs as a trade policy tool. The rate of the spread of the use of NTMs as a trade policy tool has been reflected in the literature in this field in research on the economic effects of NTMs. From this point on, in this study the effects of NTMs that Turkey has applied to automotive sector imports were analyzed using the structural gravity model. In this analysis, the effects of NTMs on the automotive industry were investigated taking into account the latest information contained in the gravity model literature and using the automotive and supply industry import data that was gathered about 149 selected countries including Turkey for 1995-2018. As a result of the analyses made at a sectoral level and specific to the items that make up the sector, the implicit protection rates were calculated by performing AVE calculations for each NTM. As a result of the analyses carried out, it has been seen that the applied NTMs generally have a negative effect on the automotive sector. In this respect, in the micro-scale analyses conducted for the goods, it has been concluded that the biggest obstacles to the imports of the sector are Technical Barriers to Trade, Contingent Trade-Protective Measures including antidumping, countervailing, and safeguard measures and Price Control Measures, respectively. In addition, in the macro-scale analysis carried out at sectoral level, it was concluded that only the Contingent Trade-Protective Measures provided a general protection for the imports of this sector. Bu çalışma beslenme, uyku süresi, uyku kalitesi,depresyon ve dinlenme metabolizma hızı arasındaki ilişkiyi incelemek amacıyla 19-25 yaş aralığındaki gönüllü 215 birey üzerinde gerçekleştirilmiştir. Bireylerin genel bilgileri ve beslenme alışkanlıkları ile ilgili sorular bir anket formu ile yüz yüze görüşme yöntemi kullanılarak kaydedilmiştir. Bireylerin antropometrik ölçümleri ve vücut bileşimleri biyoelektrik impedans analizi ile ölçülmüştür. Ayrıca bireylere besin tüketim sıklığı ( kafein ve triptofan), 24 saatlik besin tüketim kaydı, Pittsburg Uyku Kalitesi İndeksi ve Beck Depresyon Ölçeği formu uygulanmıştır. Çalışmaya katılan 215 bireyden iyi uyku kalitesine sahip 45 bireyin ve kötü uyku kalitesine sahip 45 bireyin Fitmate-Pro cihazı ile dinlenme metabolizma hızı ölçülmüştür. Çalışmaya katılan erkek bireylerin uyku kalitesi skoru, kadın bireylerin uyku kalitesi skorundan anlamlı olarak yüksek bulunmuştur (p0,05). Bu çalışmada PUKİ sınıflaması ile uyku süresi sınıflaması arasında anlamlı ilişki bulunmuştur (p0,05). İyi uyku kalitesine sahip olan kadın bireylerin DMH değerleri, kötü uyku kalitesine sahip olan kadınların DMH değerlerinden istatistiksel olarak anlamlı düzeyde daha yüksektir (p0,05). Sonuç olarak, uyku kalitesinin DMH, uyku süresi, depresyon ile ilişkisi olduğu belirlenmiştir. Bu ilişkinin açıklanabilmesi için daha geniş örneklemli çalışmaların yapılması gerekmektedir.AnahtarKelimeler : Uyku kalitesi, beslenme, dinlenme metabolizma hızı,depresyon This study, conducted on 215 individuals aging between 19 and 25, aims to examine the correlation between nutrition, sleep duration, sleep quality, depression and resting metabolic rate. Participants were requested to answer the questions regarding their nutritional habits and general information through face-to-face interview. Anthropometric measures and body composition data of individuals were measured via bioelectircal impedance analysis. Individuals were also applied food frequency questionnaire (caffeine and tryptophan), 24-hour food consumption record, Pittsburg Sleep Quality Index and Beck Depression Inventory. Resting metabolic rates of 45 individuals with good sleep quality and 45 individuals with poor sleep quality were measured by means of Fitmate-Pro. Sleep quality scores of male participants were found to be significantly higher than female participants (p0.05).However, in this study, there was a significant correlation between PSQI classification and sleep duration classification (p0.05).RMR values of female participants with good sleep quality were significantly higher than that of female individuals with poor sleep quality (p0,05). As a consequence, it was found out the sleep quality is correlated with RMR, sleep duration and depression. Buçalışma, bir ilkokulda okul öncesi eğitime devam eden çocukların sağlıklı besinve aktivite seçimleri ile beslenme durumlarının ilişkisini değerlendirmekamacıyla planlanıp yürütülmüştür. Araştırma okul öncesi eğitimine devam eden, 3-7 yaş arasıailesinden izin alınan 70 çocuk (36 kız ve 34 erkek) ve ebeveynleriyle yürütülmüştür.Verilerin toplanmasında kullanılan anket formu, çocukların ve ailelerin genelbilgileri, sosyodemografik özellikleri, ailelerin çocuklarına yönelik bedenalgısını sorgulayan sorular ile araştırmacılar tarafından benzerçalışmalar göz önünde bulundurularak geliştirilen, 64 adet besinden oluşanSağlıklı ve Sağlıksız Besin Seçim Kataloğu ve 8 adet resimden oluşan AktiviteSeçim Kataloğu’ndan oluşmaktadır.Her çocuğun vücut ağırlığı ve boy uzunluğu ölçümleri de tekniğine uygun olarakalınmıştır. Çocukların%17,1’inin zayıf, %64,3’ünün normal ağırlıkta, %5,8’inin kilolu ve %12,8’ininşişman olduğu belirlenmiştir. Ailesi tarafından şişman olarak değerlendirilençocukların %13,3’ü normal ağırlıktadır. Zayıf olan çocukların %66,7’si, kiloluveya şişman olanların %76,9’unun ailesi çocuğunu uygun ağırlıkta görmektedir.Zayıf çocukların tamamı yeşil ve turuncu gruptan toplam 12 adet besini sağlıklıolarak değerlendirirken, kilolu/şişman çocuklar ise sadece yeşil gruptan 3 adetbesini sağlıklı olarak belirtmişlerdir. Tüm çocuklar en fazla ev içiaktiviteleri tercih etmiştir. Çocukların BKI sınıfları ile besin ve aktivitetercihleri arasında istatistiksel olarak anlamlı ilişki bulunmamıştır (p>0,05).Ailelerin geliri ile çocukların sağlıklı besinleri doğru değerlendirmeleriarasında pozitif yönlü istatistiksel olarak anlamlı ilişki tespit edilmiştir(r=0,256; p

Serbest Ticaret Anlaşmaları (STA) (Free Trade Agreements-FTA),ülkelerin, kendi aralarında ikili veya çok taraflı tercihli ticaret anlaşması (TTA) yapmak suretiyle düzenlemeye giderek, ürettikleri ürünleri birbirlerinden tavizli bir şekilde ticarete konu ettikleri “Serbest Ticaret Alanı” tesis eden anlaşmalardır. Başka bir ifadeyle STA’lar, günümüzde çok taraflı ticaret sisteminin yeni pazar açılımları konusunda yetersiz kalan Dünya Ticaret Örgütü (DTÖ) düzenlemelerinin yetersizliğini aşmakta kullandıkları bir araç olarak da tanımlanabilir. Diğer taraftan Türkiye'nin STA ile ilgili politik karar ve tercihlerinde temel belirleyici unsur ise Avrupa Birliği (AB) ile imzalanan Gümrük Birliğidir (GB). Zira Türkiye, Avrupa Topluluğu Ortaklık Konseyi Kararı'nın 16. maddesi uyarınca AB'nin ortak ticaret politikasını üstlenmiş ve bu konudaki uyum yükümlülüğünü kabul etmiştir. Bu bağlamda çalışmamızda öncelikle Türkiye’nin imzaladığı STA’lar Türkiye ile AB ilişkileri bağlamında değerlendirilmiştir. Bu çerçevede Türkiye, AB ile imzaladığı GB anlaşması uyarınca üstlendiği uyum yükümlülüğünün (ortak ticaret politikası) bir parçası olarak Ürdün Haşimi Krallığı ile 2009 yılında bir Serbest Ticaret Antlaşması imzalamıştır. Bu doğrultuda çalışmanın son bölümünde söz konusu anlaşmanın Türkiye ile Ürdün’ün ikili ticareti üzerine etkileri, anlaşmanın yürürlüğe girdiği tarih öncesi ve sonrasında ortaya çıkan veriler ışığında, gerek ihracat ve ithalat rakamları gerekse iki ülkenin pazar payları açısından değerlendirilerek bazı çıkarımlarda bulunulmuştur. Free trade agreements (FTA’s) are agreements that establish "free trade area" in which countries make trade-offs between one another with products via bilateral or multilateral preferential trade agreements (PTA’s). In other words, FTA’S can also be described as a tool which overcomes the inadequacy of the multilateral trading system of World Trade Organization (WTO) regulations, which are insufficient for new markets. On the other hand, the main determinant of Turkey's political decisions and preferences for FTA’s is the Customs Union (CE) signed with the European Union (EU). According to the Article 16 of the European Community Association Council decision, Turkey has undertaken a common trade policy that of the EU and has accepted compliance to the obligations in this respect. In this publication, FTA’s signed by Turkey are assessed in the context of EU / Turkey relations. In this frame, Turkey signed a free trade agreement with the Jordanian kingdom in 2009 as part of the harmonization obligation (common trade policy) it undertook in accordance with the customs union agreement signed with the EU. In the final part of the publication, the pre and post effects of the treaty in means of bilateral trade between Turkey and Jordan were assessed, export and import figures were evaluated in terms of market shares of the two countries.

Toplam Verimli Bakım (TVB), çalışanların kullandıkları donanım hakkındaki bilgilerini arttırma, düzenli bakım yapma, düzenli işyeri tasarlama konularındaki problemlerin kaynağına inip yeniden tekrarlanmayacak şekilde çözme yaklaşımlarıyla önce sıfır duruşu; sonra sıfır hatayı hedefleyen bir yönetim sistemidir. TVB, altı büyük kayıp olarak adlandırılan; arıza kayıplarını, hazırlık ve ayar kayıplarını, kısa süreli duruş ve boşta çalışma kayıplarını, hız kayıplarını, kalite kayıplarını, ürün kayıplarını yok etmeyi; hiç olmazsa azaltmayı hedefler. Yapılan çalışmalar, TVB?ın kararlı bir şekilde uygulanması halinde; üretim giderlerini, arızaları, müşteri şikâyetlerini ve daha önemlisi iş kazalarını azalttığı, verimliliği ve toplam donanım etkinliğini arttırdığını göstermektedir.Bu çalışmada öncelikle bakım, TVB, uçak bakım kavramları ele alınmış ve TVB?ın tarihsel gelişimi ile uygulamaları özetlenmiştir. Ayrıca, kayıtlarında F?16 Blok 30 uçakları bulunan bir askeri havacılık kuruluşunda uçuş hattı seviyesi bakım işlemlerinde TVB uygulamaları sonucu elde edilen kazanımlar değerlendirilmiştir. Yapılan TVB uygulamaları sonucunda uçak bataryası ile, Düşük Basınç Svici (DBS) [Low Pressure Switch, LPS] ünitesinden kaynaklanan arıza oranlarında ve kayıp malzeme kaynaklı olay sayısında azalma meydana geldiği gözlemlenmiştir. TPM is a management system which aims firstly zero stop afterwards zero defect with increasing the employee?s knowledge about the equipment, orderly maintenance, orderly workshop and descending problems origin and solving it never to repeat again. TPM aims to ignore or reduce the effects of ?six big losses? as breakdowns, set up times, lost time due to idling and minor stoppages, adjustment times, reduced speed and process defects. The studies show that a successfully applied TPM may reduce the manufacturing costs, sudden stops, customer complaints and labour accidents; increases the productivity and the total equipment efficiency.In this study, the terms of maintenance, total productive maintenance and aviation maintenance was introduced and the implementation steps and history of TPM was explained. Finally, the results of implementing TPM in flight line (organizational) level maintenance operations in a military aviation foundation with F-16 Block 30 aircrafts were evaluated. The results showed that the failure rates of aircraft battery and low pressure switch and the number of lost item events were reduced. 98

Bu çalışmada, buğdayın en tahrip edici patojenlerden biriolan sarı pasa karşı savunmada kullandığı genleri tanımlama amacınayoğunlaşılmştır. Farklılık gösterim ve Mikroarray yöntemleri kullanılarak, buğdayıniki farklı hattında (Avocet Yr10 ve Avocet Yr1) hastalığa sebep olan ve bitkidedirençlilik oluşturan patojen suşları kullanmı (Pst: PST17, PST45, 169E136 and232E137) ile ortaya çıkan gen ifadesindeki değişmeler saptanması amaçlanmıştır.Farklılık gösterim tekniği ile 90 primer kombinasyonu sonucunda bulunan genlerden14 tanesi gerçek zamanlı polimeraz zincir reaksiyonu ile de doğrulanmıştır. Bugenlerden biri hariç hepsi buğdayda daha önce tanımlanmamış yeni genlerdir. Bugenlerden iki tanesi tam olarak proteine dönüşebilecek şekilde üç üstü ve beş üstüuçları da dahil olmak üzere tanımlanmıştır . Bunlardan bir tanesi cylophilin (olasıviantifungal) benzeri bir gen diğeri ise ubikütin conjugating E2 tipi enzim olaraktanımlanmıştır. Tanımlanan genlerin homoloji çalışmaları sonucunda ubikütine bağlıprotein parçalanması, programlanmış hücre ölümü, hastalık dirençlilik gibimekanizmalarda rol alan genlere karşılık geldikleri saptanmıştır.Farklılık gösterim tekniğine ek olarak, mikroarray yöntemi kullanılarakbitkide hastalığa karşı direnç oluşumu sırasında ifade düzeyleri değişen 93 tane gensaptanmıştır. Bunun yanı sıra hastalığın bitkide oluşumu esnasında, yani dirençliliğinolmadığı durum da ifade düzeyleri değişen 75 tane gene saptanmıştır. Bunlarda 15tanesi gerçek zamanlı polimeraz zincir reaksiyonu ile çalışılıp gen ifadelerindekideğişimler doğrulanmıştır.Tanımlanan genlerin ileri düzeyde karakterize edilebilmeleri için, yanihastalık esnasında ne gibi roller üstlendiklerini anlamak için fonksiyonel testlereihtiyaç vardır. Biz bu sebeple bu tezde BSMV'ye (Barley Stripe Mosaic Virus) bağlıvirus ile gen susturma yöntemini buğday için optimize ettik. Bitkide bulunan PDSgenini başarıyla susturduk. Bu gen susturalarak tanımlamış olduğumuz genlerepozitif kontrol geni olarak kullanılıp , susturma deneylerinin izlenmesisağlanabilecektir. Thecurrent available information is mostly on model organisms and their plant-pathogeninteractions. However, this study is focused on the identification of genes involved inthe resistance mechanism of one of the most devastating diseases of wheat, yellowrust. The strategy undertaken was to use differential display method (DD) togetherwith microarray technology, on yellow rust differential lines of wheat (Avocet-Yr1and Avocet-Yr10) infected with the virulent and avirulent Puccinia striiformis f. sp.tritici races (Pst: PST17, PST45, 169E136 and 232E137) together with appropriatecontrol infections. DD primer combinations of ninety allowed the detection offourteen differentially expressed genes which were also confirmed by real-timeQRT-PCR analysis. All of but one were found to be novel sequences in wheatgenome. Among those, two very important genes were identified as full ORFincluding 5? and 3? end untranslated regions (UTR); namely cyclophilin like proteiniv(putative antifungal activity) and ubiquitin conjugating enzyme (E2). The sequencehomology analysis of the cloned gene fragments reveled that the genes detected haveroles in ubiquitinylation, programmed cell death (apoptosis), putative antifungalactivities, disease resistance, pathogen related responses, including a few with noknown function.In addition to DD analysis, using wheat Affymetrix ?GeneChip?, weidentified 93 differentially expressed ESTs of wheat in response to avirulentpathogen attack. We also investigated the differential expression profiles of wheatleaves during the virulent infections and determined 75 differentially regulated ESTs.1Selected ESTs were further analyzed using QRT-PCR analysis and 15 wereconfirmed to be differentially regulated.For the further characterization of the identified genes, such as determinationof their putative roles in disease response, functional studies have to be performed.For this purpose, BSMV (Barley Stripe Mosaic Virus) mediated virus induced genesilencing (VIGS) method is optimized in this thesis for wheat. We have successfullymanaged to silence the endogenous PDS gene (Phytoene desaturase) of wheat whichcan be used as a positive control for the monitoring of silencing of the genes we haveidentified. Fakat, modern bitki ıslahı çalışmalarının bir sonucu olarak genetik çeşitliliği azalmış ve hastalıklara karşı daha duyarlı bir hale gelmiştir. Puccinia striiformis'in sebep olduğu san pas, buğday için çok önemli bir hastalıktır. Türkiyede son 20 yıl boyunca buğday için en tahrip edici hastalık olmuştur. 100 den fazlasıtanımlanmış olan pas dirençlilik genlerinin hiç birisi bugüne kadar moleküler düzeyde karakterize edilememiştir. İki hücre tipinde farklı ifade olmuş genlerin tanımlanmasında kuvvetli bir araç olan mRNA `farklılık gösterim` teknolojisi, bu tezde kapsamlı bir şekilde kullanılmıştır. San pasa karşı bir majör ve iki minör gene sahip 'Pastor' varyetesi, iki minör gene sahip 'Avocet' varyetesi ve bunların eşleştirilmesinden olşmuş homozigot dirençli ve homozigot hassas bitkiler farklılık gösterim yöntemi analizlerinde san pas hastalığına karşı dirençlilik genlerini bulmak amacıyla kullanılmıştır. Farklı olarak ifade olduğu tanımlanan cDNA bantlarının nükleotid dizileri gen bankalarındaki sekanslarla karşılaştırılmıştır ve daha önceki klonlanmış olan dirençlilk genleriyle benzer protein motifleri bulunmuştur. Klonlarımızın amino asit sekanslarında dirençlilik genlerinde bulunan reseptör motifleri, protein kinaz motifleri, nükleotide bağlanma bölgesi motifleri ve gelişimsel sinyal protein motiflerine rastlanmıştır. However, as a result of modem plant breeding practices, reduced genetic diversity in wheat, made it increasingly vulnerable to diseases. Yellow rust caused by Puccinia striiformis is a major disease of wheat. It has been the most devastating disease in Turkey during the past 20 years. Of the 100 rust resistance genes identified, none of them have been characterized at the molecular level yet. mRNA differential display technique, which is a powerful tool to identify those genes that are differentially expressed between the two cell types has been extensively used in this study. inThe variety 'Pastor', with one major and two minor, 'Avocet' with two minor resistance genes to yellow rust as well as homozygote resistant and susceptible F5 plants produced from the mating of Pastor and Avocet have been utilized for differential display analysis for the detection of yellow rust disease resistance genes. The differentially expressed cDNA bands were cloned and sequenced. Nucleotide sequences of differentially expressed cDNA bands were compared with the sequences in the Genebank. Some protein motifs similar to protein motifs of the previously cloned resistance gene sequences have been identified. Translated sequences of our clones contained receptor motifs, protein kinase motifs, nucleotide binding domains, and developmental signaling motifs, which are the signatures of plant disease resistance genes. Plants and animals both rely on nucleotide-binding, leucine-rich repeat proteins (NLRs) to provide disease resistance. Plant NLRs often co-operate as pairs or networks. A sensor NLR detects the pathogen and a helper NLR executes cell death and/or downstream signalling. NLRs are believed to localise to the cytoplasm, plasma membrane or nucleus. However, these localisation patterns were observed in the absence of infection, not during infection with relevant pathogens due to the HR triggered by the NLR making it impossible to perform microscopy. Many plant proteins re-locate within the cell upon infection by microbes. Therefore the localisation of NLRs during infection may be different to those without infection. Additionally, determining the localisation of NLRs can help us understand what function the NLR provides and how it detects effectors and elicits an immune response. Here I study the localisation of a solanaceous helper NLR, NRC4 during live cell imaging of Nicotiana benthamiana leaves with and without infection by the Irish potato famine pathogen, Phytophthora infestans. I use transient gene expression and confocal microscopy to show that during NRC4, accumulates from the cytoplasm to a membrane surrounding the pathogen finger-like appendages called haustoria. The extra-haustorial membrane is a newly formed plant membrane interface surrounding the pathogen haustoria. This reveals that NLRs can be motile and alter localisation during infection to accumulate at pathogen contact sites. I overcome the problem of NLRs triggering cell death when their cognate effector is present by either studying the helper NLR NRC4 in the absence of a sensor, or by disarming NRC4 by mutating the first alpha helix of the NLR’s cell death eliciting coiled-coil domain. When activated, we found that NRC4 forms puncta which associate with the EHM or in the absence of infection, the plasma membrane. These puncta may represent inflammasome-like ‘resistosomes’ or groups of resistosomes. A phylogenetically and functionally related NLR, ZAR1, was recently shown to form a resistosome and is proposed to execute cell death by possibly forming pores in the host membrane. Further evidence is required in the future to determine if NRC4 is forming resistosomes and whether these are eliciting cell death directly. Intriguingly, efforts to prevent NRC4 accumulation at the EHM, by mutation, truncation or chimeric NLR generation, led to loss of NRC4’s cell death function. As a whole, this thesis provides evidence that NRC4 accumulates to the host-pathogen interface to provide disease resistance against P. infestans. This thesis reveals that NLRs can alter their localisations upon infection and that they can even focally accumulate at the host-pathogen interface. Further understanding into how NRC4 focally accumulates during infection and exactly what it achieves by doing so may help us understand how resistance to fungi and oomycetes occurs. Ultimately, this may allow us to engineer disease resistant crops by, for instance, altering NRC4’s specificity to recognise other EHM localised effectors, without altering NRC4’s localisation. Plants and other organisms are able to tolerate stress, combat infection and maintain homeostasis through the conserved catabolic process of autophagy which involves sequestration of cytoplasmic material in double membraned vesicles for degradation and recycling. Understanding how autophagy is initiated and regulated is critical to gaining insight into many essential cellular processes such as development, aging and immunity against intracellular pathogens. In this study I show that a plant RabGTPase activating protein, Rab3GAPL, directly interacts with the autophagy adaptor protein ATG8CL through a canonical ATG8 interacting motif (AIM) to suppress autophagy in an AIM and catalytic Arg479 dependent manner. Furthermore, I show that Rab3GAPL interacts with and is a cognate GAP for the small RabGTPase Rab8a1 which plays a positive role in autophagosome biogenesis and flux. Finally, I elucidate that Rab3GAPL negatively affects immunity to the oomycete pathogen Phythophthora infestans but suppressing defence-related secretion immune responses mediated by Rab8a1. This thesis combines multiple approaches to functionally characterise a new plant RabGAP and its role in autophagy and immunity against invading pathogens. Furthermore, it provides evidence for extensive interactions between vesicle traffic components, autophagy and immunity in plants, paving the way for several new lines of inquiry into these processes.
