Nosemosis, one of the most important honeybee diseases, causes colony losses and low productivity worldwide. A national study were carried out to presence of nosemosis throughout Türkiye, determine the prevalence of Nosema cereane and Nosema apis, and perform the network analyses of the parasites. Specimens were collected from hives with no clinical signs. A total of 1194 European honey bee colonies in 400 apiaries from 40 provinces of Türkiye were examined by microscopic and molecular techniques. Nosemosis were found in all of 40 provinces. The prevalence ratio were 64.3 ± 3,0, 95%CI in apiaries and 40.5 ± 2,9, 95%CI in hives. N. ceranae DNA were detected all of positive hives, while N. ceranae and N. apis co-infection in only four colonies. In the last two decades, it has been observed that nozemosis has spread to all provinces and its prevalence has increased in all regions of Türkiye. All of the N. ceranae or N. apis samples examined were 100% identical within themselves. Network analysis showed that they were within largest haplotype reported worldwide.

Acarapis woodi (Rennie, 1921) (Acari: Tarsonemidae) is one of the mites that settles in the respiratory system of honeybees (Apis mellifera L. (Hymenoptera, Apidae)) and distributed throughout the world. It causes significant economic losses on honey production. In Türkiye, studies on the existence of A. woodi are very limited and so far, no studies on the molecular diagnosis and phylogenetic of it have been reported in Türkiye. This study was conducted to investigate the prevalence of A. woodi in Türkiye, especially in areas where beekeeping is intense. Diagnosis of A. woodi was performed using both microscopic and molecular methods using specific PCR primers. Adult honeybee samples were collected from 1.193 hives in 40 provinces of Türkiye between 2018 and 2019. Based on identification studies, the presence of A. woodi was detected in a total of 3 hives (0.5%) in 2018 and 4 hives (0.7%) in 2019. This is the first report for determination of A. woodi in Türkiye.

The aim of this study is to determine the effect of growing media on antioxidant properties and antimicrobial activities of Pleurotus ostreatus(Jacq. ex Fr.) P. Kumm. In the study, growing substrates were prepared with five tree sawdust (80%) and waste (20% Tea Waste or Wheat Bran) at different ratios (80% Sawdust + 20% TW, 80% Sawdust + 20% GW). The antimicrobial activities of methanol extracts from 19 P. ostreatus against microorganisms were tested by agar well diffusion technique. The total phenolic content was measured using the Folin-Ciocalteau procedure. The antioxidant capacity was determined using ferric reducing antioxidant power (FRAP) and free radical scavenging activity of DPPH. It was observed that the addition of 20% tea waste in the substrate had a positive effect on antimicrobial activity. It was determined that the methanol extract of mushrooms obtained from the 80QS+20TW substrate was the most effective extract against to all of microorganisms investigated in this study. In the gastrointestinal tract of honey bees (Apis mellifera), there is a unique collection of microorganisms, called microflora or microbiota, which have various mgeyietabolic activities in the bee crops, which can protect against various microbial threats, and is linked to powerful mechanisms of action in the bee immune system. The adult honey bees especially have a rich microflora. Their microbiome is considered to be one of the most important and remarkable factors in maintaining bee health. Like humans, honey bees (Apis mellifera) are social creatures and work cooperatively to ensure the continuity of life in the hive. In productive colonies, the increasing diversity of microbiota depends on various factors, from the developmental age of the bee to its feeding, from the geographical location of the hive to the local climate. The lack of diversity of the bacterial community may negatively affect the productivity of the bee hive. This review describes the diversity of the honey bee intestinal core microflora, which are of great importance for bee life, the ways in which they acquire the bacteria, how they colonize in the gastrointestinal tract of bee and their associated beneficial mechanisms for honey bee survival.

Paenibacillus larvae (P. larvae) is a pathogenic bacterium causing American Foulbrood Disease (AFB) in honeybee larvae. It is necessary to develop alternative strategies for the control of this disease due to the serious honeybee colonies loses and the use of antibiotics. Recent studies are aimed at the investigating natural fighting agents against P. larvae. In our study, it was aimed to demonstrate potential antibacterial efficacy of ethanol extract of Anatolian Propolis (EAP) against P. larvae strains PB35 and SV35 which were isolated in Turkey. The total phenolic content (TPC) and flavonoid content (TFC) of EEAP were determined as 181.73±5.20 mg Gallic Acid Equivalents (GAE)/g, and 42.33±1.40 mg Quercetin Equivalents (QE)/g, respectively. It was found that EAP contains different amounts of ferulic, caffeic, coumaric acids, rutin, and caffeic acid phenethyl ester (CAPE). The antibacterial activity of the EAP was determined by using agar-well diffusion, microdilution, and Bioscreen C techniques. The Minimal Inhibition Concentration (MIC) values of the EAP were determined as 74.87 μg/ml against strain PB35 and SV35 using both microdilution and Bioscreen C teqnique. In both techniques, Minimal Bactericidal Concentration (MBC) values of the EAP were evaluated as 149 and 598.4 μg/ml against strain PB35 and SV35, respectively. The fact that EAP shows low concentrations of bacteriostatic (MIC) and bactericide (MBC) activity values against P. larvae strains, spore-forming bacilli, which are agents of AFB disease, suggests that it may be a potential source in AFB disease control. Çalışmamızda Anadolu propolisi etanolik ekstresinin (EAP) Türkiye'de izole edilen P. larvae suşları PB35 ve SV35'e karşı potansiyel antimikrobiyal etkinliğinin ortaya konması amaçlanmıştır. EAP, toplam fenolik madde (TPC), flavonoid madde miktarları (TFC) ve bazı fenolik bileşikler açısından karakterize edilmiştir. Analizlere göre, EAP'nin total fenolik madde miktarı ve flavonoid madde miktarları sırasıyla 181,73±5,20 mg Gallik Asit Eşdeğeri (GAE)/g TFC olarak 42,33±1,40 mg Kuersetin Eşdeğeri (QE)/g ve farklı miktarlarda ferulik, kafeik, kumarik asitler, rutin ve kafeik asit fenetil ester (CAPE) içerdiği bulundu. EAP'nin antimikrobiyal aktivitesi, agar kuyu difüzyon, mikrodilüsyon ve Bioscreen C teknikleri kullanılarak belirlendi. EAP'nin Minimal İnhibisyon Konsantrasyon (MIC) değerleri, hem mikrodilüsyon hem de Bioscreen C tekniği kullanılarak PB35 ve SV35 suşuna karşı 74,87 μg/ml olarak belirlendi. Her iki teknikte de PB35 ve SV35 suşlarına karşı EAP'nin minimal bakterisidal konsantrasyon (MBC) değerleri sırasıyla 149 ve 598,4 μg/ml olarak belirlendi.

The goal of the work is to determine the bioactive pharmaceutical metabolites produced by the Fusarium oxysporum YP9B isolate. Ten new natural compounds were isolated from the ethyl acetate extract of the F. oxysporum YP9B strain. Their structures were elucidated by spectroscopic methods using 1D and 2D NMR, UV, FT-IR, and mass spectra (LC-QTOF MS and GC-FID/MS). Identified compounds were named as; (1-benzyl-2-methoxy-2-oxoethyl)-2-hydroxy-3-methylbutanoate (1), 2-oxo-8-azatricyclo[9.3.1.13,7]-hexadeca-1(15),3(16),4,6,11,13-hexaen-10-one (2), 2,3-dihydroxypropanoic, hexadecanoic anhydride (3a), 2,3-dihydroxypropanoic (9Z)-octadecenoic anhydride (3b), 2,3-dihydroxy-propanoic (9Z,12Z)-octadecadienoic anhydride (3c), 2,3-dihydroxypropanoic (11Z)-octadecenoic anhydride (4a), 2,3-dihydroxypropanoic, (9E,12E)-octadecadienoic anhydride (4b), 3-hydroxy-1,2,6,10-tetramethylundecyl hexzadecanoate (5a), 3-hydroxy-1,2,6,10-tetramethylundecyl (9E)-octadecaenoate (5b), and 3-hydroxy-1,2,6,10-tetramethylundecyl octadecanoate (5c). Antimicrobial activities of the isolates obtained from the YP9B strain were determined. Cytotoxic and antiviral activities were tested for the isolates against VERO, MCF-7, PC-3, and A549. Compounds 5a-c, 1, and 3a-c showed bacteriostatic activity at low concentrations, and 4a-b and 2 were found to be bactericides. MIC and MBC values against Mycobacterium smegmatis for the compounds 5a-c and 1 were determined to be ​​

Bacterial diseases, such as American Foulbrood (AFB) and European Foulbrood (EFB), are known to have catastrophic effects on honey bees (if left to spread, can wipe out entire colonies), leading to severe financial losses in the beekeeping industry. The aim of this study was to evaluate the pharmacological properties of methanol extract and its fractions (ethyl acetate, hexane, water) derived from Dicranum scoparium Hedw., which could be utilized as a potential drug to prevent the bacterial diseases (AFB and EFB) affecting the honey bees. For this purpose, crude methanol extract and ethyl acetate/hexane/water fractions were prepared from the aerial part of D. scoparium, collected from Trabzon province. Bio-guided fractionation of the extract and its fractions led to the first-time isolation of five compounds. The structure of all compounds was elucidated by nuclear magnetic resonance (NMR) spectroscopy, ultraviolet (UV) spectral analysis, Fourier-transform infrared spectroscopy (FT-IR), liquid chromatography quadrupole time-of-flight mass spectroscopy (LC-QToF-MS), and by comparison of their NMR data with that of literature. The analysis of these compounds revealed significant antibacterial and sporicidal activities against bacteria causing larval diseases in honey bees. The antibacterial activity of these compounds ranged from 0.6 to 60 μg/mL against AFB and EFB causing bacteria. Therefore, the natural raw extract and fractions of D. scoparium could be used as potential therapeutic agents against bacterial agents affecting honey bees.

Çalışmada çay toprağından izole edilen Trichoderma harzianum ID11C suşunun bazı enzim aktivitesi, ağır metallere toleransı, tohum (domates, mısır ve fasulye) çimlenmesi üzerine etkinliği ve fasulyede Rhizoctonia solani B227 (AG-4)'nin biyotik stresine karşı biyokontrol aktivitesinin araştırılması amaçlandı. T. harzianum ID11C suşunun bazı enzim aktiviteleri kromojenik yöntemle incelendi. Sonuçlar farklı konsantrasyonlarda ağır metal (Cu, Pb, Zn ve Cd) ve tuz toleransı olduğunu gösterdi.T. harzianum ID11C Rhizoctonia solani B227(AG4) izolatının fasulye gelişimine karşın patojenite testi ve T. harzianum ID11C suşunun tohum çimlenme başarısına olan etkinliği, su agar metodu ile incelendi. T. harzianum ID11C ve R. solani B227 varlığında fasulye gelişim parametreleri saksı deneyi ile araştırıldı. Fizyolojik (ana kök ve gövde uzunluğu, saçaklanma sayısı, yaprak sayısı, yaş ve kuru ağırlıkları) parametreler ölçüldü ve istatistiksel analizleri yapıldı.Trichoderma harzianum ID11C suşu, yüksek konsantrasyonlarda ağır metallere (100-400 mM) ve tuza (250 mM) karşı toleranslıdır. Bitki gelişimini teşvik eden enzim aktivitelerinin var olduğu gözlendi. T. harzianum suşunun tohumların çimlenme başarısı üzerinde kontrole kıyasla olumsuz bir etkisinin olmadığı tespit edildi. Saksı deneyinde, kontrol ile R. solani B227 grupları birbirine kıyaslandığında, gövde ve ana kök uzunluğu, saçaklanma sayısı ve kökte lezyon skala değerleri ile yapılan istatistik analizi sonucu aralarında anlamlı farkın (p< 0.05, Tukey) olduğu belirlendi. R. solani B227 ve T. harzianum ID11D’nin ayrı ayrı fasulye gelişimi üzerine olan etkileri, kontrol grubuna göre iyi olmadığı belirlendi. Ancak ID11C ve B227+ID11C grupları B227 grubu ile kıyaslandığında, patojenik etkinliği azalttığı belirlendi. R. solani B227 (AG4) izolatı, fasulyede (Zulbiye) patojeniteye neden olduğu, T. harzianum ID11C suşunun ise istatistiksel olarak olumlu yönde anlamlı bir fark oluşturmamasına rağmen, tüm parametrelerde patojeniteyi azalttığı belirlendi. T. harzianum ID11C suşunun fasulye tarımında Rhizoctonia solani’ye karşı biyokontrol ve bitki destekleyici ajan olarak kullanılabileceği sonucuna varıldı.
