Among other bioactive molecules, milk contains high amounts of sialylated milk oligosaccharides (MOs) that influence numerous processes in the offspring. For instance, sialylated MOs inhibit the invasion of pathogens and positively influence the gut microbiome to support the optimal development of the offspring. For these reasons, sialylated MOs are also used in infant formula as well as food supplements and are potential therapeutic substances for humans and animals. Because of the high interest in sialylated bovine MOs (bMOs), we used several analytical approaches, such as gas and liquid chromatography in combination with mass spectrometry, to investigate in detail the profile of sialylated bMOs in the milk of Holstein Friesian cows during early lactation. Most of the 40 MOs identified in this study were sialylated, and a rapid decrease in all detected sialylated bMOs took place during the first day of lactation. Remarkably, we observed a high variance within the sialylation level during the first two days after calving. Therefore, our results suggest that the content of sialylated MOs might be an additional quality marker for the bioactivity of colostrum and transitional milk to ensure its optimized application for the production of milk replacer and food supplements. The aim of this study was to utilise mid‐infrared spectroscopy with fatty acid (FA) predictions to characterise the seasonality‐associated changes to the FA profile of milk from Irish spring‐calving pasture‐based dairy cow production systems. A total of 546 454 individual milk records were collected from the Years 2015 to 2020, from 264 003 cows in 2400 commercial dairy farms. Significant changes occur to the predicted FA profile of milk across the year, such as increases in unsaturated FAs including C18:1 n‐9 until April. This can be associated with the increasing proportion of fresh pasture in the cows' diet, and a decrease between September and October as the cows begin to transition indoors for the winter period. The opposite trend was noted for saturated FAs and in particular C16:0, where proportions decreased until April and increased between September and November. Milk produced between April and September also coincided with the lowest spreadability index, indicating a more beneficial milk FA profile for more spreadable butter texture during this period. Background: Coagulase-negative Staphylococcus species are an emerging cause of intramammary infection, posing a significant economic and public health threat. The aim of this study was to assess the occurrence of coagulase-negative Staphylococcus species in bovine milk and dairy farms in Northwestern Ethiopia and to provide information about their antibiotic susceptibility and virulence gene profiles. Methods: The cross-sectional study was conducted from February to August 2022. Coagulase-negative Staphylococcus species were isolated from 290 milk samples. Species isolation and identification were performed by plate culturing and biochemical tests and the antimicrobial susceptibility pattern of each isolate was determined by the Kirby-Bauer disc diffusion test. The single-plex PCR was used to detect the presence of virulent genes. The STATA software version 16 was used for data analysis. The prevalence, proportion of antimicrobial resistance and the number of virulent genes detected from coagulase-negative Staphylococcus species were analyzed using descriptive statistics. Results: Coagulase-negative Staphylococcus species were isolated in 28.6%, (95% CI: 23.5–34.2) of the samples. Of these, the S. epidermidis, S. sciuri, S. warneri, S. haemolyticus, S. simulans, S. chromogens, S. cohnii, and S. captis species were isolated at the rates of 11, 5.2, 3.4, 3.1, 3.1, 1, 1, and 0.7% respectively. All the isolates showed a high percentage (100%) of resistance to Amoxicillin, Ampicillin, and Cefotetan and 37.5% of resistance to Oxacillin. The majority (54.2%) of coagulase-negative isolates also showed multidrug resistance. Coagulase-negative Staphylococcus species carried the icaD, pvl, mecA, hlb, sec, and hla virulent genes at the rates of 26.5%, 22.1%, 21.7%, 9.6%, 9.6% and 8.4% respectively. Conclusion: The present study revealed that the majority of the isolates (54.2%) were found multidrug-resistant and carriage of one or more virulent and enterotoxin genes responsible for intramammary and food poisoning infections. Thus, urgent disease control and prevention measures are warranted to reduce the deleterious impact of coagulase-negative species. To the best of our knowledge, this is the first study in Ethiopia to detect coagulase-negative Staphylococcus species with their associated virulent and food poisoning genes from bovine milk. Milk is a biological fluid with a dynamic composition of micronutrients and bioactive molecules that serves as a vital nutrient source for infants. Milk composition is affected by multiple factors, including genetics, geographical location, environmental conditions, lactation phase, and maternal nutrition, and plays a key role in dictating its microbiome. This study addresses a less-explored aspect, comparing the microbial communities in human breast milk with those in mature milk from species that are used for milk consumption. Since mature animal milk is used as a supplement for both the infant (formula) and the child/adolescent, our main aim was to identify shared microbial communities in colostrum and mature human milk. Using 16S rRNA metagenomic sequencing, we focused on characterizing the milk microbiota in the Northern Greek population by identifying shared microbial communities across samples and comparing the relative abundance of prevalent genera. We analyzed ten human milk samples (from five mothers), with five collected three days postpartum (colostrum) and five collected thirty to forty days postpartum (mature milk) from corresponding mothers. To perform an interspecies comparison of human milk microbiota, we analyzed five goat and five bovine milk samples from a local dairy industry, collected fifty to seventy days after birth. Alpha diversity analysis indicated moderate diversity and stability in bovine milk, high richness in goat milk, and constrained diversity in breast milk. Beta diversity analysis revealed significant distinctions among mammalian species, emphasizing both presence/absence and abundance-based clustering. Despite noticeable differences, shared microbial components underscore fundamental aspects across all mammalian species, highlighting the presence of a core microbiota predominantly comprising the Proteobacteria, Firmicutes, and Actinobacteriota phyla. At the genus level, Acinetobacter, Gemella, and Sphingobium exhibit significant higher abundance in human milk compared to bovine and goat milk, while Pseudomonas and Atopostipes are more prevalent in animal milk. Our comparative analysis revealed differences and commonalities in the microbial communities of various mammalian milks and unraveled the existence of a common fundamental milk core microbiome. We thus revealed both species-specific and conserved microbial communities in human, bovine, and goat milk. The existence of a common core microbiome with conserved differences between colostrum and mature human milk underscores fundamental similarities in the microbiota of milk across mammalian species, which could offer valuable implications for optimizing the nutritional quality and safety of dairy products as well as supplements for infant health. This study investigated the effects of isochoric freezing (IF) on the shelf-life and quality of raw bovine milk over a 5-week period. The results were compared with conventional refrigeration (RF) and refrigeration after pasteurization (HTST). The IF treatment process entailed storing liquid raw milk in isochoric chambers in thermodynamic equilibrium at -5 °C/77 MPa and -10 °C/96 MPa. Several parameters were analyzed, including microbiology count, physicochemical properties, indigenous enzyme activity, protein content, volatile organic compounds profile, and lipid degradation. Both raw and pasteurized milk experienced increases in the microbial level past the acceptable threshold (≥5.5 log CFU/mL) after 2 weeks and 5 weeks, respectively, leading to the deterioration of other parameters during storage. In comparison, microbiology count decreased significantly during storage for both IF treatment conditions but was more pronounced for the higher pressure (96 MPa) treatment, leading to undetectable levels of microorganism after 5 weeks. IF treatment maintained stable pH, titratable acidity, viscosity, lipid oxidation, volatile profiles, total protein content, and lactoperoxidase activity throughout the storage period. Color was preserved during IF treatment at -5 °C/77 MPa; however, color was impacted during IF treatment at -10 °C/96 MPa. Protein structures were also modified during pressurized storage in both IF treatments. Overall, the study demonstrated that isochoric freezing could significantly increase the shelf-life of milk by reducing microbiology activity, whilst maintaining its nutritional content. These results underscore the potential role of isochoric freezing as a valuable tool in eliminating pathogens while maintaining quality characteristics similar to raw milk over long storage periods.

Apart from essential nutrients, bovine milk also contains bioactive compounds, including a microbiota derived from milk itself rather than external sources of contamination. Aim of review: Recognizing the profound impact of bovine milk microorganisms on future generations, our review focuses on exploring their composition, origins, functions, and applications. Key scientific concepts of review: Some of the primary microorganisms found in bovine milk are also present in human milk. These microorganisms are likely transferred to the mammary gland through two pathways: the entero-mammary pathway and the rumen-mammary pathway. We also elucidated potential mechanisms by which milk microbiota contribute to infant intestinal development. The mechanisms include the enhancing of the intestinal microecological niche, promoting the maturation of immune system, strengthening the intestinal epithelial barrier function, and interacting with milk components (e.g., oligosaccharides) via cross-feeding effect. However, given the limited understanding of bovine milk microbiota, further studies are necessary to validate hypotheses regarding their origins and to explore their functions and potential applications in early intestinal development.

The most reliable methods for pregnancy diagnosis in dairy herds include rectal palpation, ultrasound examination, and evaluation of plasma progesterone concentrations. However, these methods are expensive, labor-intensive, and invasive. Thus, there is a need to develop a practical, non-invasive, cost-effective method that can be implemented on the farm to detect pregnancy. This study suggests employing microwave dielectric spectroscopy (MDS, 0.5–40 GHz) as a method to evaluate reproduction events in dairy cows. The approach involves the integration of MDS data with information on milk solids to detect pregnancy and identify early embryonic loss in dairy cows. To test the ability to predict pregnancy according to these measurements, milk samples were collected from (i) pregnant and non-pregnant randomly selected cows, (ii) weekly from selected cows (n = 12) before insemination until a positive pregnancy test, and (iii) daily from selected cows (n = 10) prior to insemination until a positive pregnancy test. The results indicated that the dielectric strength of Δε and the relaxation time, τ, exhibited reduced variability in the case of a positive pregnancy diagnosis. Using principal component analysis (PCA), a clear distinction between pregnancy and nonpregnancy status was observed, with improved differentiation upon a higher sampling frequency. Additionally, a neural network machine learning technique was employed to develop a prediction algorithm with an accuracy of 73%. These findings demonstrate that MDS can be used to detect changes in milk upon pregnancy. Abstract Renneting is an enzymatic process that turns milk into curd which is then transformed into cheese. Rennet‐induced coagulation of caseins (CNs) is the critical step during this process and the key is the primary hydrolysis of κ‐CN's Phe105‐Met106 bond by chymosin. This article comprehensively reviews the existing data on the extent/degree of κ‐CN hydrolysis during renneting of bovine milk and critically evaluates its determination methods. The data show that under normal cheese‐making conditions, milk gelation occurs at a degree of κ‐CN hydrolysis

Due to the lack of effective assessment method, overheated milk commodities are often marketed as pasteurized milk on the market, which was sold in high price by fraud. Thus, this article aims to establish an approach based on metabolomics to monitor thermal processing temperature of bovine milk. Metabolomics data of bovine milk samples heated at temperatures ranging from 60 °C to 150 °C were achieved by ultra-performance liquid chromatography-high-resolution mass spectrometry (UPLC-HRMS) platform, followed by multivariate data analysis. A regular variation pattern of chemical composition as temperature rises was pictured, furthermore, Nε-carboxymethyl lysine (CML), Nε-carboxyethyl lysine (CEL), pentosidine, pyrraline and lysinoalanine (LAL) were identified as 5 of the most contributed compounds to discriminate pasteurized and ultra-high-temperature (UHT) milk. By the comprehensive study on their content changes, we concluded that the optimal temperature range was 90−100 °C for the generation of CML and CEL in this experiment, moreover, 110−120 °C for LAL, 80−100 °C for pentosidine and 130−140 °C for pyrraline. Finally, a predicted rule to discriminate pasteurized and UHT milk was preliminarily established based on the ratios of CML/CEL, CEL/pentosidine and CML/pentosidine, which could be applied in food labelling authentication of commercial bovine milk after further validation.

Abstract Extracellular vesicles (EVs) in bovine milk confer beneficial physiologic effects to consumers. Industrial processing treatments may affect the amount or bioactivity of EVs intrinsic to bovine milk. We investigated how the content and concentration of EVs were affected by homogenization and thermal processing of raw bovine milk. Raw milk was processed by homogenization, low‐temperature (LT) heat, or pasteurization [high‐temperature short‐time (HTST) and ultra‐high‐temperature (UHT)] in a pilot processing facility. EVs were isolated from the raw and processed bovine milk using differential ultracentrifugation and quantified using a nanoparticle tracking analyzer. Bovine milk EVs were assessed for total miRNA and protein concentrations standardized to particle count using a fluorometric assay. There were 1.01 × 1010 (±3.30 × 109) EV particles per ml of bovine milk. All industrial processing treatments caused >60% decrease in EV concentration compared to the raw bovine milk. Homogenization and heat treatments independently and additively reduced the content of EVs in bovine milk. The averages of total miRNA/particle and total protein/particle concentrations were elevated threefold by low‐temperature heat‐processing treatment relative to HTST and UHT pasteurizations. The average diameter of EVs was reduced by 11%–16% by low temperature compared to raw milk (127 ± 13 nm). Homogenization and pasteurization indiscriminately reduce the EV concentration of bovine milk. Smaller EVs with higher protein content resist degradation when processing bovine milk at sub‐pasteurization temperature. This new foundational knowledge may contribute to food product development on the preservation of EVs in processed dairy products, including bovine milk‐based infant formulas that some newborns are dependent on for adequate growth and development.

Lactoperoxidase (LPO) is a crucial enzyme found in milk, tears, and saliva, protecting newborns from harmful microorganisms. This study examined the effects of phenolic substances (naringin, morin, esculin, homovanillic acid, and phloridzin) on purified LPO. LPO was purified from bovine milk using affinity chromatography, yielding 66.6 % with a purity of 387.5‐fold. Three methodologies were employed to assess inhibitory properties: spectrophotometric techniques, molecular docking, and MM‐GBSA. The compounds′ IC50 values were 0.0024 μM, 0.0315 μM, 0.0373 μM, 0.0506 μM, and 0.0221 μM, while their Ki values were 0.0059±0.0012 μM, 0.0672±0.0247 μM, 0.0973±0.0369 μM, 0.0664±0.0190 μM, and 0.0470±0.0159 μM, respectively. Naringin exhibited the most potent inhibitory effect, competitively inhibiting LPO. Therefore, naringin could serve as a novel reversible LPO inhibitor. [ABSTRACT FROM AUTHOR]

Renneting is an enzymatic process that turns milk into curd which is then transformed into cheese. This article comprehensively reviews the existing data on the extent/degree of κ‐CN hydrolysis during renneting of bovine milk and critically evaluates its determination methods. The data show that under normal cheese‐making conditions, milk gelation occurs at a degree of κ‐CN hydrolysis <80%, which varies due to several factors including analytical and estimation approaches. The common approach involves isolating the macropeptides released, by precipitating whey proteins and residual CN in 1%–12% trichloroacetic acid (TCA), then assuming that the maximum amount obtained is 100% κ‐CN hydrolysis. The drawback is that the estimated degree of κ‐CN hydrolysis may be higher than the actual value as TCA partially precipitates the macropeptide fractions. Moreover, macropeptide isolation seems unnecessary based on current advances in chromatographic and electrophoretic techniques. The present work proposes a simple mass balance‐based approach that will provide accurate estimates in future studies. The accuracy of measuring the degree of κ‐CN hydrolysis has implications on the precision of the data in relation to its partitioning (% distribution between the curd and whey) which is essential for improving whey quality. A2 milk is characterized by a difference in the amino acid at position 67 of the β-casein polypeptide chain, which releases much smaller amounts of bioactive opioid peptide β-casomorphin - 7 upon digestion. Milk consumption may lead to discomfort due to the presence of A1 beta-casein protein. A2 milk is the preferable alternative for people with mild lactose intolerance or lactose sensitivity because it does not include A1 protein, which is thought to be kinder to the digestive system. The need for healthy and minimally processed dairy products, the growing awareness of dairyrelated sensitivities, as well as the growing interest in organic and natural food options are some of the major reasons driving the expansion of the A2 milk market. ABSTRACT In low-microbial biomass samples such as bovine milk, contaminants can outnumber endogenous bacteria. Because of this, milk microbiome research suffers from a critical knowledge gap, namely, does non-mastitis bovine milk contain a native microbiome? In this study, we sampled external and internal mammary epithelia and stripped and cisternal milk and used numerous negative controls, including air and sampling controls and extraction and library preparation blanks, to identify the potential sources of contamination. Two algorithms were used to mathematically remove contaminants and track the potential movement of microbes among samples. Results suggest that the majority (i.e., >75%) of sequence data generated from bovine milk and mammary epithelium samples represents contaminating DNA. Contaminants in milk samples were primarily sourced from DNA extraction kits and the internal and external skin of the teat, while teat canal and apex samples were mainly contaminated during the sampling process. After decontamination, the milk microbiome displayed a more dispersed, less diverse, and compositionally distinct bacterial profile compared with epithelial samples. Similar microbial compositions were observed between cisternal and stripped milk samples, as well as between teat apex and canal samples. Staphylococcus and Acinetobacter were the predominant genera detected in milk sample sequences, and bacterial culture showed growth of Staphylococcus and Corynebacterium spp. in 50% (7/14) of stripped milk samples and growth of Staphylococcus spp. in 7% (1/14) of cisternal milk samples. Our study suggests that microbiome data generated from milk samples obtained from clinically healthy bovine udders may be heavily biased by contaminants that enter the sample during sample collection and processing workflows.IMPORTANCEObtaining a non-contaminated sample of bovine milk is challenging due to the nature of the sampling environment and the route by which milk is typically extracted from the mammary gland. Furthermore, the very low bacterial biomass of bovine milk exacerbates the impacts of contaminant sequences in downstream analyses, which can lead to severe biases. Our finding showed that bovine milk contains very low bacterial biomass and each contamination event (including sampling procedure and DNA extraction process) introduces bacteria and/or DNA fragments that easily outnumber the native bacterial cells. This finding has important implications for our ability to draw robust conclusions from milk microbiome data, especially if the data have not been subjected to rigorous decontamination procedures. Based on these findings, we strongly urge researchers to include numerous negative controls into their sampling and sample processing workflows and to utilize several complementary methods for identifying potential contaminants within the resulting sequence data. These measures will improve the accuracy, reliability, reproducibility, and interpretability of milk microbiome data and research.

Evaluating quality and microbiological parameters in bovine milk processing centers is essential for ensuring public health and product quality and positively impacting the local economy. This study aimed to evaluate the physicochemical and sanitary parameters of bovine milk in the Amazon region. For this purpose, 31 samples were collected from milk processing centers in the region. The following quality parameters were evaluated: somatic cell count; physicochemical parameters: fat, protein, nonfat solids, lactose, ash, pH, and density; and microbiological parameters: mesophilic aerobic bacteria and total coliforms in the raw milk. For data analysis, mean comparison tests were performed using Kruskal‒Wallis and Tukey tests, and cluster and principal component analysis was performed using R software version 4.3.2 and InfoStat version 2018p. The results for the 31 milk processing centers showed significant differences in all the parameters evaluated; fat, somatic cell count, and density were within the technical specifications of the Supreme Decree N° 007-2017-MINAGRI. Microbiological analyses revealed significant variability in total coliforms and mesophilic aerobic bacteria. Cluster analysis revealed five groups with similar characteristics, with Group 1 exhibiting high somatic cell, lactose, and ash contents and Group 3 exhibiting higher fat contents. Principal component analysis highlighted the significant contributions of total coliforms, aerobic bacteria, and somatic cells to the degradation of milk quality. Although somatic cell counts meet specific standards, elevated levels of coliforms and aerobic bacteria are of concern. These findings are essential for ensuring food safety and improving milk quality.

ABSTRACT: This study investigated the effect of feeding seaweed (Ascophyllum nodosum) to dairy cows on milk mineral concentrations, feed-to-milk mineral transfer efficiencies, and hematological parameters. Lactating Holstein cows (n = 46) were allocated to 1 of 2 diets (n = 23 each): (1) control (CON; without seaweed) and (2) seaweed (SWD; replacing 330 g/d of dried corn meal in CON with 330 g/d dried A. nodosum). All cows were fed the CON diet for 4 wk before the experiment (adaptation period), and animals were then fed the experimental diets for 9 wk. Samples included sequential 3-wk composite feed samples, a composite milk sample on the last day of each week, and a blood sample at the end of the study. Data were statistically analyzed using a linear mixed effects model with diet, week, and their interaction as fixed factors; cow (nested within diet) as a random factor; and data collected on the last day of the adaptation period as covariates. Feeding SWD increased milk concentrations of Mg (+6.6 mg/kg), P (+56 mg/kg), and I (+1,720 μg/kg). It also reduced transfer efficiency of Ca, Mg, P, K, Mn, and Zn, and increased transfer efficiency of Mo. Feeding SWD marginally reduced milk protein concentrations, whereas there was no effect of SWD feeding on cows' hematological parameters. Feeding A. nodosum increased milk I concentrations, which can be beneficial when feed I concentration is limited or in demographics or populations with increased risk of I deficiency (e.g., female adolescents, pregnant women, nursing mothers). However, care should also be taken when feeding SWD to dairy cows because, in the present study, milk I concentrations were particularly high and could result in I intakes that pose a health risk for children consuming milk.

Among other bioactive molecules, milk contains high amounts of sialylated milk oligosaccharides (MOs) that influence numerous processes in the offspring. For instance, sialylated MOs inhibit the invasion of pathogens and positively influence the gut microbiome to support the optimal development of the offspring. For these reasons, sialylated MOs are also used in infant formula as well as food supplements and are potential therapeutic substances for humans and animals. Because of the high interest in sialylated bovine MOs (bMOs), we used several analytical approaches, such as gas and liquid chromatography in combination with mass spectrometry, to investigate in detail the profile of sialylated bMOs in the milk of Holstein Friesian cows during early lactation. Most of the 40 MOs identified in this study were sialylated, and a rapid decrease in all detected sialylated bMOs took place during the first day of lactation. Remarkably, we observed a high variance within the sialylation level during the first two days after calving. Therefore, our results suggest that the content of sialylated MOs might be an additional quality marker for the bioactivity of colostrum and transitional milk to ensure its optimized application for the production of milk replacer and food supplements.

Milk is a biological fluid with a dynamic composition of micronutrients and bioactive molecules that serves as a vital nutrient source for infants. Milk composition is affected by multiple factors, including genetics, geographical location, environmental conditions, lactation phase, and maternal nutrition, and plays a key role in dictating its microbiome. This study addresses a less-explored aspect, comparing the microbial communities in human breast milk with those in mature milk from species that are used for milk consumption. Since mature animal milk is used as a supplement for both the infant (formula) and the child/adolescent, our main aim was to identify shared microbial communities in colostrum and mature human milk. Using 16S rRNA metagenomic sequencing, we focused on characterizing the milk microbiota in the Northern Greek population by identifying shared microbial communities across samples and comparing the relative abundance of prevalent genera. We analyzed ten human milk samples (from five mothers), with five collected three days postpartum (colostrum) and five collected thirty to forty days postpartum (mature milk) from corresponding mothers. To perform an interspecies comparison of human milk microbiota, we analyzed five goat and five bovine milk samples from a local dairy industry, collected fifty to seventy days after birth. Alpha diversity analysis indicated moderate diversity and stability in bovine milk, high richness in goat milk, and constrained diversity in breast milk. Beta diversity analysis revealed significant distinctions among mammalian species, emphasizing both presence/absence and abundance-based clustering. Despite noticeable differences, shared microbial components underscore fundamental aspects across all mammalian species, highlighting the presence of a core microbiota predominantly comprising the Proteobacteria, Firmicutes, and Actinobacteriota phyla. At the genus level, Acinetobacter, Gemella, and Sphingobium exhibit significant higher abundance in human milk compared to bovine and goat milk, while Pseudomonas and Atopostipes are more prevalent in animal milk. Our comparative analysis revealed differences and commonalities in the microbial communities of various mammalian milks and unraveled the existence of a common fundamental milk core microbiome. We thus revealed both species-specific and conserved microbial communities in human, bovine, and goat milk. The existence of a common core microbiome with conserved differences between colostrum and mature human milk underscores fundamental similarities in the microbiota of milk across mammalian species, which could offer valuable implications for optimizing the nutritional quality and safety of dairy products as well as supplements for infant health.

Background: For decades, the use of organochlorine (OC) pesticides has had a detrimental effect on the environment and human health. Contamination of soil, water, and air has also resulted in contaminated milk. Aim: The purpose of this study was to investigate if any OC residues dichlorodiphenyltrichloroethane (DDT, Dieldrin, Endrin, and Lindane) were present in raw bovine milk from West Delta, Egypt. Methods: 200 fresh raw cow milk samples (500 ml of each sample) collected from three different governorates, west Delta, Egypt, for determination of OC pesticides residues using gas chromatography with an Agilent 6890A model gas chromatograph equipped with a 63Ni microelectron capture detector. Results: The obtained results revealed that åDDT, dieldrin, endrin, and lindane were detected in Alexandria, Behera, and Matrouh at incidence levels (22.7%, 30.7%, and 10%), (20%, 20%, and 16%), (9.33%, 13.3%, and 16%), and (12%, 10.7%, and 14%) with mean values of 232.2 ± 163.6, 156.4 ± 134.6 and 100.4 ± 85.9; 91.3 ± 61.2, 95.3 ± 59.8 and 57.6 ± 3.33; 15.7± 3.86, 15.1 ± 3.96 and 20.1 ± 7.33; 33.7 ± 10.6, 36.9 ± 5.51 and 52.2 ± 21.8 ng/g fat, respectively. El-Behera was the most contaminated province with an incidence level of 53.3% with a mean value of 136.8 ± 128.0 ng/g fat, followed by Alexandria at 44% with a mean value of 173.7 ± 155.5 ng/g fat, and finally, Matrouh 40% with a mean value of 74.5 ± 56.5 ng/g fat. Conclusion: This research demonstrated that milk samples contain varying levels of OC pesticide residues, which can be hazardous to consumer health. Milk is a nutrient-rich food source, and among the various milks, breast milk is a nutrient source provided by mothers to newborns in many mammals. Exosomes are nano-sized membranous extracellular vesicles that play important roles in cell-to-cell communication. Exosomes originate from endogenous synthesis and dietary sources such as milk. Discovered through electron microscopy as floating vesicles, the existence of exosomes in human milk was confirmed owing to a density between 1.10 and 1.18 g/mL in a sucrose gradient corresponding to the known density of exosomes and detection of MHC classes I and II, CD63, CD81, and CD86 on the vesicles. To date, milk exosomes have been used for treating many diseases, including cancers, and are widely proposed as promising carriers for the delivery of chemotherapeutic agents. However, few studies on milk exosomes focus on geriatric health, especially sarcopenia and osteoporosis related to bone and muscle. Therefore, the present study focused on milk exosomes and their cargoes, which are potential candidates for dietary supplements, and when combined with drugs, they can be effective in treating musculoskeletal diseases. In this review, we introduce the basic concepts, including the definition, various sources, and cargoes of milk exosomes, and exosome isolation and characterization methods. Additionally, we review recent literature on the musculoskeletal system and milk exosomes. Since inflammation and oxidative stress underly musculoskeletal disorders, studies reporting the antioxidant and anti-inflammatory properties of milk exosomes are also summarized. Simple Summary: Prednisolone (PSL) is generally used as an anti-inflammatory and analgesic agent. To establish a withdrawal period using PSL in this study, PSL residual levels and withdrawal time (WT) were assessed in milk after PSL was administered to healthy cows. As a result, the LC-MS/MS method's recovery rate, relative standard deviation, and detection sensitivity met all required standards. At 24 h after treatment, the amount of PSL in milk containing 10 and 20 mg/mL of PSL was lower than the LOD and the LOQ. The time limit for withdrawal was set at 12 h. In summary, since the residual withdrawal period of PSL in milk is shorter than that of the test drug, it suggests that the administration of PSL preparations will produce less milk wastage than that of previous recommendations. We measured the levels of prednisolone (PSL) residues in milk of intramuscularly dosed dairy cows and established a withdrawal time (WT) of PSL in milk. Eight healthy Holstein cows were injected with 10 (PSL-1) or 20 (PSL-2) mL of 10 mg/mL of PSL, and then, their milk was sampled at 12 h intervals for five days. PSL residue concentrations in milk were determined using LC-MS/MS. The correlation coefficient of the calibration curve was 0.9976. The limit of detection (LOD) and the limit of quantification (LOQ) were 0.2 µg/kg and 0.6 μg/kg, respectively. Recoveries ranged from 96.5% to 110.0%, and the coefficient of variation was <5.64%. At 24 h after administration, PSL levels in PSL-1 and PSL-2 were below the LOQ in all milk samples. Although this study had a smaller sample size than the European Medicines Agency's recommendations (n = 20), it was based on the Animal and Plant Quarantine Agency guidelines of the Republic of Korea (n = 8) for the determination of withdrawal periods in milk. We established the withdrawal period for both PSL-1 and PSL-2 in milk at 12 h. In conclusion, we developed an analytical method that is sensitive and can reliably detect PSL in milk, and our estimated WT of PSL in bovine milk is shorter than the current 3-day withdrawal period of PSL in commercial PSL products. ABSTRACT: Two PPG1000 based temperature-sensitive magnetic ionic liquid were synthesized and characterized. The temperature-sensitive magnetic ionic liquid aqueous biphasic system combined with HPLC was applied for the continuous enrichment and trace analysis of tetracycline antibiotics (TC) in bovine milk for the first time. High enrichment factors were achieved and the detection was highly sensitive. The trace analysis of TC was rapid, free of organic solvent, recyclable and magnetically assisted for phase separation. Under optimum conditions, wide linear ranges of 0.25–300 ng/mL for all TC, high enrichment factors of 217.7–231.4, good precisions with relative standard deviation in the range of 0.74–3.97%, very low limits of detection of 0.031–0.067 ng/mL, limits of quantification of 0.103–0.223 ng/mL, and good recoveries of 94.28–99.76% were acquired for the proposed analytical method. Real milk analysis was satisfactory. This developed analytical method is showing great potential for trace analysis of targeted analytes in foods and drinks.

The most reliable methods for pregnancy diagnosis in dairy herds include rectal palpation, ultrasound examination, and evaluation of plasma progesterone concentrations. However, these methods are expensive, labor-intensive, and invasive. Thus, there is a need to develop a practical, non-invasive, cost-effective method that can be implemented on the farm to detect pregnancy. This study suggests employing microwave dielectric spectroscopy (MDS, 0.5–40 GHz) as a method to evaluate reproduction events in dairy cows. The approach involves the integration of MDS data with information on milk solids to detect pregnancy and identify early embryonic loss in dairy cows. To test the ability to predict pregnancy according to these measurements, milk samples were collected from (i) pregnant and non-pregnant randomly selected cows, (ii) weekly from selected cows (n = 12) before insemination until a positive pregnancy test, and (iii) daily from selected cows (n = 10) prior to insemination until a positive pregnancy test. The results indicated that the dielectric strength of Δε and the relaxation time, τ, exhibited reduced variability in the case of a positive pregnancy diagnosis. Using principal component analysis (PCA), a clear distinction between pregnancy and nonpregnancy status was observed, with improved differentiation upon a higher sampling frequency. Additionally, a neural network machine learning technique was employed to develop a prediction algorithm with an accuracy of 73%. These findings demonstrate that MDS can be used to detect changes in milk upon pregnancy. The developed machine learning provides a broad classification that could be further enhanced with additional data.

Human milk, which provides personalized, comprehensive, and balanced nutrients, and immune protection, is the best nutritional source for 0~6 month-old infants, and it is the gold standard for infant food. A cohort study of human milk omics is a necessary prerequisite for comprehensively analyzing the composition and characteristics of Chinese human milk, establishing Chinese infant food norms and standards, developing infant nutrition guidelines, and creating infant foods. This article presents the research progress of the National Engineering Research Center of Dairy Health for Maternal and Child pertaining to the China Maternal and Infant Nutrition Health Birth Cohort Study (MINC) in terms of breast milk lipidomics, glycomics, proteomics, and microbiomics detection techniques, composition characteristics, differences from bovine milk, and industrial applications in infant food. This will lay a science and technical support for the omics research of Chinese human milk and provide the necessary data and demonstration for industrial application.

Bovine milk is a nutrient-dense food and a major component of the human diet. Therefore, understanding the factors that impact its composition is of great importance. Applications of metabolomics provide in-depth analysis of the metabolite composition of milk. The objective of this research was to examine the impact of lactation stage on bovine milk metabolite levels. Metabolomic analysis of bovine milk powder samples across lactation (N = 18) was performed using nuclear magnetic resonance (1H-NMR) spectroscopy and liquid chromatography–tandem mass spectrometry (LC-MS/MS). Forty-one metabolites were identified and quantified in the 1H-NMR spectra. Statistical analysis revealed that 17 metabolites were significantly different across lactation stages (FDR < 0.05), of which the majority had higher levels in early lactation. In total, 491 metabolites were measured using LC-MS/MS, of which 269 had significantly different levels across lactation (FDR < 0.05). Compound classes significantly affected by lactation stage included phosphatidylcholines (59%) and triglycerides (64%), of which 100% of phosphatidylcholines and 61% of triglycerides increased from early lactation onwards. Our study demonstrates significant differences in metabolites across the stages of lactation, with early-lactation milk having a distinct metabolomic profile. This study investigated whether heat treatments (raw, 63 °C for 30 min, and 85 °C for 5 min) affect protein hydrolysis by endogenous enzymes in the milk of ruminants (bovine, ovine, and caprine) using a self-digestion model. Self-digestion consisted of the incubation for six hours at 37 °C of the ruminants' milk. Free amino group concentration was measured by the o-phthaldialdehyde method, and peptide sequences were identified by chromatography-mass spectrometry. Results showed that heat treatments prior to self-digestion decreased the free NH2 by 59% in bovine milk heated at 85 °C/5 min, and by 44 and 53% in caprine milk heated at 63 °C/30 min and 85 °C/5 min, respectively. However, after self-digestion, only new free amino groups were observed for the raw and heated at 63 °C/30 min milk. β-Casein was the most cleaved protein in the raw and heated at 63 °C/30 min bovine milk. A similar trend was observed in raw ovine and caprine milk. Self-digestion increased 6.8-fold the potential antithrombin peptides in the bovine milk heated at 63 °C/30 min. Enhancing bioactive peptide abundance through self-digestion has potential applications in the industry for functional products. Overall, heat treatments affected the free amino groups according to the species and heat treatment applied, which was reflected in the varying degrees of cleaved peptide bonds and peptides released during self-digestion. Lactoferrin, IGF‐1 and IgG are three well‐known bioactive proteins found in bovine colostrum. We studied the effects of these three active ingredients on the levels of eight cytokines in serum and on the cellular communication networks in vivo by oral administration, intraperitoneal injection and intravenous injection. The results suggested that bioactive ingredients, especially biomacromolecules, can induce different effects, even opposite effects, on the immune system depending on the route of administration. These effects are related to both the functions of the bioactive ingredient in vivo and to its specific receptor binding in the gastrointestinal mucosa that activates different signalling pathways. The rise in prevalence and drug resistance in Staphylococcus aureus of dairy animals require an alternative to antibiotics. Nanoparticles particularly metallic nanoparticles are a wise approach to modulating drug resistance. The current study thus focuses on the replacement of antibiotics with ZnO, MgO, and Fe2O3 nanoparticles. To achieve this goal, 200 cattle milk samples were collected aseptically and screened for subclinical mastitis using the purposive sampling technique. Following Bergey's manual of determinative bacteriology, S.aureus was identified and antibiotic susceptibility was tested using clinical laboratory and standard institute guidelines. On the other hand, resistant strains were put to antibacterial testing assay against metallic nanoparticles both by well diffusion and broth microdilution method. To analyze the data, both probability and non-probability statistical tools were applied using SPSS version 22 of statistical software at 5% probability. The current study showed 24.56% of S. aureus positive from commercial dairy while the resistance of these isolates against gentamicin, enrofloxacin, levofloxacin, and vancomycin was found to be 50, 40, 30, 30%, respectively. On the other hand, a disc diffusion assay was concluded with 24.525±0.806 mm (ZnO) followed by 16.475±0.950mm (MgO) and 13.150±1.392mm (Fe2O3). MgO stood first to contribute lowest Minimum inhibitory concentration (MIC) (1.302±0.564 mg/mL) followed by Fe2O3 (2.930±1.691 mg/mL) and ZnO (3.906±0.000 mg/mL). MIC of ZnO was recorded to be 18.23±11.93 mg/mL, 15.63±0.0 mg/mL, 13.02±4.5 mg/mL, 7.81±6.77 mg/mL, 5.21±2.26 mg/mL, and (3.906±0.000 mg/mL) at 04, 08, 12, 16, 20 and 24th h of incubation. A similar trend was followed by other nanoparticles at different hours of incubation. Simple Summary: Bovine milk is important for human nutrition, especially for infants. However, per-fluoroalkyl substances (PFASs) are harmful chemicals that can contaminate milk and pose a threat to both humans and animals. This study evaluated how 14 different PFASs were distributed within milk by analyzing the content of whole milk, skim milk, and cream. Samples were taken from 23 cows in Northern Italy that were not exposed to known sources of PFASs. The most common PFAS found in all three fractions was perfluorobutanoic acid (PFBA), followed by perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA). PFOS was found in higher concentrations in cream compared to raw and skimmed milk. Multiparous cows had higher levels of PFASs in their milk. To fully understand the risks of consuming dairy products contaminated with PFASs and their potential impact on cattle health status, further investigation is necessary. Additionally, more studies are needed to uncover the mechanisms behind the excretion of these substances in milk. Bovine milk is a pillar of the human diet and plays a key role in the nutrition of infants. Perfluoroalkyl substances (PFASs) are well-recognized highly stable organic compounds that are able to pollute ecosystems persistently and threaten both human and animal health. The study aimed to analyze the distribution of 14 PFASs within the milk matrix by comparing their content in whole milk, and its skimmed and creamed fractions. Raw milk samples were individually collected from 23 healthy cows (10 primiparous and 13 multiparous) reared on a farm in Northern Italy not surrounded by known point sources of PFASs. Each sample was fractioned in whole, skim, and cream components to undergo PFAS analysis using liquid chromatography–high-resolution mass spectrometry. All samples contained at least one PFAS, with perfluorobutanoic acid (PFBA) being the primary contaminant in all three fractions, followed by perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA). PFOS was shown to be significantly (p < 0.001) more concentrated in cream than in raw and skimmed milk. Multiparous cows showed a higher frequency of positive samples in all analyzed fractions. Further research is necessary to assess the risk of dairy diets and high-fat dairy products and to investigate the toxicological effects of PFASs on cattle, even in environments without known PFAS sources. BackgroundThe rate of stomach emptying of milk from different ruminant species differs, suggesting that the small intestinal digestibility of nutrients could also differ across these milk types.ObjectiveTo determine the small intestinal amino acid (AA) digestibility of raw bovine, caprine, and ovine milk in the piglet as an animal model for the infant.MethodsSeven-day-old piglets (n = 12) consumed either bovine, caprine, or ovine milk diets for 15 days (n = 4 piglets/milk). On day 15, fasted piglets received a single meal of fresh raw milk normalized for protein content and containing the indigestible marker titanium dioxide. Entire gastrointestinal tract contents were collected at 210 min postprandially. Apparent AA digestibility (disappearance) in different regions of the small intestine was determined.ResultsOn average, 35% of the dietary AAs were apparently taken up in the small intestine during the first 210 min post-feeding, with 67% of the AA digestibility occurring in the first quarter (p ≤ 0.05) and 33% in the subsequent two quarters. Overall, except for isoleucine, valine, phenylalanine, and tyrosine, the small intestinal apparent digestibility of all AAs at 210 min postprandially in piglets fed ovine milk was, on average, 29% higher (p ≤ 0.05) than for those fed bovine milk. Except for lysine, there was no difference in the apparent digestibility (p > 0.05) of any AAs between piglets fed caprine milk or ovine milk. The apparent digestibility of alanine was higher (p ≤ 0.05) in piglets fed caprine milk than those fed bovine milk. When apparent digestibility was corrected for gastric AA retention, only small differences in the small intestinal apparent digestibility of AAs were observed across milk types.ConclusionBovine, caprine and ovine milk had different apparent small intestinal AA digestibility at 210 min postprandially. When corrected for gastric AA retention, the differences in apparent digestibility across species largely disappeared. The apparent AA digestibility differed across small intestinal locations.

One of the salting methods in cheese production implies salting the milk before coagulation used in making Domiati-type cheeses and a variety of autochthonous "Lički Škripavac" cheese. The most used sodium replacer is potassium. This study investigated the influence of different added salt concentrations (1%, 1.5%, and 2%) and NaCl to KCl ratios (100%, 50:50%, 25:75%) on the rennet coagulation and curd firmness in bovine milk. The milk coagulation parameters were determined with a computerized renneting meter, Lactodinamograph. The results showed significant interactions between the salt concentrations and NaCl to KCl ratios (p < 0.0001, α = 0.05) by prolonging the beginning of coagulation (10–20 min) and curd firming rate (1–5 min) by an increase in salt concentration for all treatments. The 50:50 treatment values (RCT, k20, a30, a60, amax) were closest to the control (without salt) and had the best results among all treatments in the lower (1%) and medium (1.5%) salt concentration (p > 0.0001, α = 0.05) while in the highest salt concentration (2%) the treatment effect was nonsignificant (p > 0.05). Abstract Bovine milk peptides are the protein fragments with diverse bioactive properties having antioxidant, anticarcinogenic, other therapeutic and nutraceutical potentials. These peptides are formed in milk by enzymatic hydrolysis, gastrointestinal digestion and fermentation processes. They have significant health impact with high potency and low toxicity making them a suitable natural alternative for preventing and managing diseases. Antibiotic resistance has increased the quest for better peptide candidates with antimicrobial effects. This article presents a comprehensive review on well documented antimicrobial, immunological, opioid, and anti-hypertensive activities of bovine milk peptides. It also covers the usage of computational biology tools and databases for prediction and analysis of the food-derived bioactive peptides. In silico analysis of amino acid sequences of Bos taurus milk proteins have been predicted to generate peptides with dipeptidyl peptidase IV inhibitory and ACE inhibitory properties, making them favorable candidates for developing blood sugar lowering drugs and anti-hypertensives. In addition to the prediction of new bioactive peptides, application of bioinformatics tools to predict novel functions of already known peptides is also discussed. Overall, this review focuses on the reported as well as predicted biologically active peptide of casein and whey proteins of bovine milk that can be utilized to develop therapeutic agents. The study investigated potential changes in bioactive milk proteins in relation to the season, breed, farm and udder health status. Nine hundred and sixty bulk milk samples from 12 commercial farms in northwest China were collected during 2020–2021. Six bioactive proteins including fraction of β‐casein proteins, α‐lactalbumin, β‐lactoglobulin, lactoferrin (Lf) and osteopontin were evaluated. It was found that most proteins (with exception of β‐casein A1) of Jersey cows are significantly higher than Holstein cows. Most bioactive proteins demonstrated seasonal variation, and some exhibited geographical variation (with exception of whey albumins). Milk oligosaccharides are a complex class of carbohydrates that act as bioactive factors in numerous defensive and physiological functions, including brain development. Early nutrition can modulate nervous system development and can lead to epigenetic imprinting. We attempted to increase the sialylated oligosaccharide content of zebrafish yolk reserves, with the aim of evaluating any short-term effects of the treatment on mortality, locomotor behavior, and gene expression. Wild-type embryos were microinjected with saline solution or solutions containing sialylated milk oligosaccharides extracted from human and bovine milk. The results suggest that burst activity and larval survival rates were unaffected by the treatments. Locomotion parameters were found to be similar during the light phase between control and treated larvae; in the dark, however, milk oligosaccharide-treated larvae showed increased test plate exploration. Thigmotaxis results did not reveal significant differences in either the light or the dark conditions. The RNA-seq analysis indicated that both treatments exert an antioxidant effect in developing fish. Moreover, sialylated human milk oligosaccharides seemed to increase the expression of genes related to cell cycle control and chromosomal replication, while bovine-derived oligosaccharides caused an increase in the expression of genes involved in synaptogenesis and neuronal signaling. Buffalo milk contains more polyunsaturated fatty acids than bovine milk. However, it is not clear about the effects of buffalo milk and bovine milk on lipid metabolism. In this study, a mouse model was used to explore the effects of buffalo milk and bovine milk on lipid metabolism in mice. The experiment was divided into three groups: a control group on a normal diet; a bovine milk group infused with bovine milk; a buffalo milk group infused with buffalo milk. We fed three groups of mice (n = 6) for 6 weeks. These results showed that bovine milk and buffalo milk had no effect on body weight gain. Bovine milk increased the content of ApoA1, ApoB and glucose in serum, compared with the control group, but buffalo milk has no profound change in serum ApoB. Remarkably, buffalo milk decreased the content of total cholesterol (TC) and triglyceride (TG) in the liver lipid profile, and also downregulated the expression of the carnitine palmitoyltransferase 2 (Cpt2) gene involved in the fatty acid oxidation in the liver. This study also found that bovine milk and buffalo milk did not cause the expression of pro‐inflammatory factors in serum and colon tissues. This experiment proved that buffalo milk has beneficial effects on the regulation of lipid metabolism, and also does not affect the normal growth and pro‐inflammatory response of the colon in mice. Hemp press cake flour (HPCF) is a by-product of hemp oil production rich in proteins, carbohydrates, minerals, vitamins, oleochemicals, and phytochemicals. The purpose of this study was to investigate how the addition of HPCF to bovine and ovine plain yoghurts at concentrations of 0%, 2%, 4%, 6%, 8%, and 10% could change the physicochemical, microbiological, and sensory properties of the yoghurts, focusing on the improvement of quality and antioxidant activity, and the issue of food by-products and their utilisation. The results showed that the addition of HPCF to yoghurts significantly affected their properties, including an increase in pH and decrease in titratable acidity, change in colour to darker, reddish or yellowish hue, and a rise in total polyphenols and antioxidant activity during storage. Yoghurts fortified with 4% and 6% HPCF exhibited the best sensory properties, thus maintaining viable starter counts in the yoghurts during the study period. There were no statistically significant differences between the control yoghurts and the samples with 4% added HPCF in terms of overall sensory score while maintaining viable starter counts during the seven-day storage. These results suggest that the addition of HPCF to yoghurts can improve product quality and create functional products and may have potential in sustainable food waste management. The safety and success of an implant after surgery depend on many factors, some related to the implant's biocompatibility, properties, surface modification, design, and other factors related to surgical procedures, implant bed preparation, and drilling techniques. It is recognized that the success of implant dentistry depends on several factors that may be related to biochemical properties and modification in mechanical properties. The present study aimed to assess the effect of using bovine milk as an irrigant solution on implant osseointegration. The implant socket was prepared by drilling bone holes in 20 femurs of the rabbits at steady rotation speeds with different irrigate solutions (normal saline / commercial pasteurized bovine milk). Mechanical tests and histological investigation were performed to estimate the removal torque record and implant contact area, BIC. Findings illustrate that implant contact area (BIC) and removal torque mean values are higher in experimental compared to control with more bone apposition and maturation at 4&8 week measured periods.
