4. Transferencia de genes entre especies. Herencia horizontal y transmisión vertical de genes adquiridos

Colección Biblioteca de Ciencias; 478 págs; 17 x 24 cm; EAN: 9788433870452., El 20 de julio de 1822 nace en Heinzendorf (actual Hyncice, República Checa) Gregor Johann Mendel, un hombre que cambiaría para siempre el rumbo de la biología. En 1843, ingresa como fraile agustino en la abadía de Santo Tomás de Brno lo que le daría acceso al conocimiento científico más actual de su momento. Durante los casi 42 años que vivió allí fue un investigador incansable, miembro y fundador de diversas sociedades científicas, con un profundo interés por la meteorología y la astronomía y, sobre todo, por la agronomía. Sus estudios cruzando variedades de guisante, publicados entre 1865 y 1866, le permitieron sentar las bases de la herencia, hecho por el cual es considerado el Padre de la Genética. Aunque el reconocimiento no le llegó en vida, siempre mantuvo la convicción de estar trabajando en una empresa relevante. Como él mismo diría: Mi momento llegará . Y su momento llegó. La prueba más fehaciente de ello la tenemos en el auge de la genómica al que estamos asistiendo en la actualidad, y cuyas bases se anclan directamente en la genética mendeliana. Este libro se edita con motivo de la conmemoración del bicentenario del nacimiento de Mendel. En él se repasan los acontecimientos más importantes de su vida, el legado que ha dejado a la genética actual, así como los retos presentes y futuros que esta disciplina afronta. En esta obra coral han participado casi una treintena de biólogos y genetistas de reconocido prestigio internacional. Cada capítulo está ilustrado por el dibujante José Luis Prats (Ozeluí). Además, se incluye una nueva traducción del manuscrito de Mendel, la primera realizada directamente desde el alemán (idioma original en que está escrito).

Colección Cuadernos de Innovación Docente (CID); Serie Biociencias, 6; 64 pp., 17x24 cm., El desarrollo de técnicas de análisis genómico ha puesto de manifiesto que la transferencia horizontal o lateral de genes (HGT o LGT) ha jugado un importante papel en la evolución del mundo vivo, principalmente de los procariotas (bacterias y arqueas) pero también en la evolución de los eucariotas, contribuyendo a la adquisición de rasgos concretos que habrían facilitado adaptaciones fisiológicas importantes para la diversificación de estos grupos.

Abstract

Horizontal gene transfer (HGT) is widespread among prokaryotes driving their evolution. In this paper, we review the potential impact in humans of the HGT between prokaryotes living in close association with humans in two scenarios: horizontal transfer in human microbiomes and transfer between microbes living in human managed environments. Although our vision is focused on the possible impact of these transfers in the propagation of antibiotic resistance genes or pathogenicity determinants, we also discuss possible human physiological adaptations via gene transfer between resident and occasional bacteria in the human microbiome. Abstract

[EN]: The eukaryotic origin is one of the great transitions in the history of the life. Today is accepted that eukaryote domain is the fruit of the interaction between two prokaryotic organisms beloing to Bacteria and Archaea life domains. In the present article I review the different models and scenarios proposed to explain the eukaryotic origin., [ES]: El origen de los eucariotas constituye una de las grandes transiciones en la historia de la vida. Hoy existe un amplio consenso en que éste se produce por la interacción entre dos organismos procarioticos pertenecientes a los dominios bacteriano y archaeano. Sin embargo, diferentes modelos y escenarios que implican diferentes asunciones han sido propuestos para explicar este origen. En el presente artículo reviso sucintamente el estado actual de nuestro conocimiento en relación a como surgieron los eucariotas.

Abstract

Horizontal gene transfer (HGT) is widespread among prokaryotes driving their evolution. In this paper, we review the potential impact in humans of the HGT between prokaryotes living in close association with humans in two scenarios: horizontal transfer in human microbiomes and transfer between microbes living in human managed environments. Although our vision is focused on the possible impact of these transfers in the propagation of antibiotic resistance genes or pathogenicity determinants, we also discuss possible human physiological adaptations via gene transfer between resident and occasional bacteria in the human microbiome.

Ponencia Invitada Inaugural del Congreso de Espeleología "EspeleoMeeting Ciudad de Villacarrillo" (Villacarrillo (Jaén), 13, 14 y 15 de agosto de 2016)., [ES] En este trabajo se describe a través de la citología, histología y secuenciación del gen de la rodopsina, la única población de peces encontrada en una cueva de la Península Ibérica. Esta población pertenece a la especie Squalius alburnoides (Steindachner, 1866) (Cyprinidae) y tiene la particularidad de tener un origen híbrido con individuos y poblaciones de diferentes ploidias (2n=50; 3n=75 y 4n=100) y distintos mecanismos de reproducción. Los peces de la cueva de Peal del Becerro I presentaron una degeneración incipiente de la retina, con menor cantidad de fotorreceptores y pigmentación que los peces de la misma especie situados en el exterior de la cueva (Embalse de La Bolera). Todos resultaron ser tetraploides simétricos AAPP lo que sugiere que este biotipo favorece de algún modo la viabilidad de poblaciones de esta especie permitiendo recuperar la bisexualidad y un genoma balanceado. Sin embargo, es necesario realizar estudios en profundidad desde la genómica y transcriptómica y tener un mejor conocimiento de la biología y ecología de esta población cavernícola., [EN] In this study we described through a multidisciplinary approach (cytology, histology and rhodopsin gene sequencing) the only cave population of freshwater fish found in the Iberian Peninsula. This population belong to the species Squalius alburnoides (Steindachner, 1866) (Cyprinidae), which has the singularity of being a species of hybridogenetic origin with individuals and populations of different ploidies (2n=50; 3n=75 y 4n=100) and diverse mechanisms of reproduction. The individuals of S. alburnoides found in the Peal del Becerro I cave showed an incipient degeneration of the retina, with lower amount of photoreceptor cells and pigmentation relative to individuals outside the cave (de la Bolera reservoir). All individuals from the cave were symmetric tetraploids AAPP, which suggests that this biotope is favouring the viability of this population through the recovery of bisexuality and a balanced genome. Nonetheless, further deeper studies using genomics and transcriptomics approaches are required to achieve a better understanding of the biology and ecology of this cave-dweller population.

27. Horizontal gene transfer in evolution

Abstract

The concept of horizontal or lateral gene transfer (the non‐genealogical transmission of genetic material between organisms) was introduced in 1990s to explain the observed incongruence in phylogenetic reconstructions using different genes. Today, horizontal gene transfer is accepted as an important force modulating evolution of Bacteria, Archaea and unicellular eukaryotes, and evidence is rising for their role in the evolution of pluricellular eukaryotes (fungi, plants and animals). The realisation that horizontal gene transfer plays an important role in evolution of both, prokaryotes and eukaryotes, put in question the metaphor of the tree of life and the traditional view of evolution as a slow process. A more pluralistic approach to evolution is emerging that encompasses different evolutionary mechanisms operating at different levels of complexity.

Abstract

Studies on experimental evolution of microorganisms, on their in vivo evolution (mainly in the case of bacteria producing chronic infections) as well as the availability of multiple full genomic sequences are placing bacteria in the playground of evolutionary studies. This review focuses on the study of the relative contribution of processes as gene modification, gene acquisition and gene loss at the evolution of bacterial genomes in different ecological scenarios. In particular, we review how the different processes contribute to evolution in microbial communities, in free-living bacteria or for bacteria living in isolation. In addition, we discuss the temporal constraints in the evolution of bacterial genomes, considering bacterial evolution from the perspective of processes of short-sighted evolution and punctual acquisition of evolutionary novelties followed by long stasis periods.

Abstract

A new microsatellite locus (SAS1) for Squalius alburnoides was obtained through cloning by serendipity. The possible usefulness of this new species-specific microsatellite in genetic studies of this hybrid-species complex, was explored. The polymorphism exhibited by SAS1 microsatellite is an important addition to the set of microsatellites previously used in genetic studies in S. alburnoides complex, that mostly relied in markers described for other species. Moreover, the SAS1 microsatellite could be used to identifythe parental genomes of the complex, complementing other methods recently described for the same purpose..

This study explores the effects of microsatellite size homoplasies in the reconstruction of phylogenetic relationships and estimates of population parameters as Fixation index (FST) using as a case study a truncated microsatellite from the picote splitfin Zoogoneticus quitzeoensis. The results suggest that the use of imperfect microsatellites may have only a minor effect in phylogenetic and population studies.

10 paginas, 6 figuras, 4 tables, The present taxonomic status of the so called fracticornis–opacicollis complex of the genus Onthophagus, Onthophagus fracticornis (Preyssler, 1790), Onthophagus similis (Scriba, 1790) and Onthophagus opacicollis Reitter, 1892 is still controversial. Discrepancies mainly involve the status of O. similis and O. opacicollis. A genetic analysis of nine populations of this complex using eight loci is presented, indicating that: (i) O. fracticornis is a good species, and (ii) the genetic distances between O. opacicollis and O. similis populations as a function of the biogeographic scenery (allopatry versus sympatry), indicate that they are not completely reproductively isolated across their entire geographic range, thus suggesting a still unfinished speciation in different areas. The data demonstrate both the existence of some gene flow between O. opacicollis and O. similis in one place (‘El Barraco’; Central Spain) and isolation in others (‘Sª Nevada’; South-east Spain). Geographic areas hosting populations of both taxa with intermediate genetic distances (‘Doñana’; South-west Spain) also exist in the Iberian Peninsula., This research was funded by grants PB89 041 and PB81 9010 from the Spanish D.G.IC.Y.T

37. Horizontal gene transfer in evolution: facts and challenges

Abstract

The contribution of horizontal gene transfer to evolution has been controversial since it was suggested to be a force driving evolution in the microbial world. In this paper, I review the current standpoint on horizontal gene transfer in evolutionary thinking and discuss how important horizontal gene transfer is in evolution in the broad sense and particularly in prokaryotic evolution. I review recent literature asking firstly which processes are involved in the evolutionary success of transferred genes and secondly about the extent of horizontal gene transfer toward different evolutionary times. Moreover, I discuss the feasibility of reconstructing ancient phylogenetic relationships in the face of horizontal gene transfer. Finally, I discuss how horizontal gene transfer fits in the current neodarwinian evolutionary paradigm and conclude there is a need for a new evolutionary paradigm that includes horizontal gene transfer as well as other mechanisms in the explanation of evolution.

[EN] In this paper I present a survey for molecular markers available for Vertebrate species inhabiting the Comunidad de Madrid, with the aim of being useful to researchers working on the characterization of animal biodiversity in this area., [ES] En este trabajo presento los resultados de una búsqueda en bases de datos de marcadores moleculares para especies de Vertebrados presentes en la Comunidad de Madrid, en la esperanza de que este trabajo pueda ser de utilidad para aquellos investigadores involucrados en la caracterización de biodiversidad animal en esta Comunidad.

44. Nucleotide Sequence and Deduced Functions of a Set of Cotranscribed Genes of Streptomyces coelicolor A3(2) Including the Polyketide Synthase for the Antibiotic Actinorhodin*

Abstract

13 páginas, 6 figuras, 3 tables et al, A 5.3-kb region of the Streptomyces coelicolor actinorhodin gene cluster, including the genes for polyketide biosynthesis, was sequenced. Six identified open reading frames (ORF1-6) were related to genetically characterized mutations of classes actI, VII, IV, and VB by complementation analysis. ORF1-6 run divergently from the adjacent act111 gene, which encodes the polyketide synthase (PKS) ketoreductase, and appear to form an operon. The deduced gene products of ORF1-3 are similar to fatty acid synthases (FAS) of different organisms and PKS genes from other polyketide producers. The predicted ORF5 gene product is similar to type I1 &lactamases of Bacillus cereus and Bacteroides fragilis. The ORF6 product does not resemble other known proteins. combining the genetical, biochemical, and similarity data, the potential activities of the products of the six genes can be postulated as: 1) condensing enzyme/acyl transferase (ORF1 + ORF2); 2) acyl carrier protein (ORF3); 3) putative cyclase/dehydrase (ORF4); 4) dehydrase (ORF5); and 5) “dimerase” (ORF6). The data show that the actinorhodin PKS consists of discrete monofunctional components, like that of the Escherichia coli (Type 11) FAS, rather than the multifunctional polypeptides for the macrolide PKSs and vertebrate FASs (Type I)., * This research was supported in part by grants (to F. 46. Mitochondrial DNA variability in viviparous and ovoviviparous populations of the urodele Salamandra salamandra

Abstract

Mitochondrial DNA analysis of 13 populations of S. salamandra along a transect across the North of the Iberian Peninsula showed values of divergence between haplotypes ranging from d= 0.41% to 5.91%. Phenetic and cladistic analysis grouped the isofemale lineages into two main clusters with contrasting phylogeographic patterns. The first group encompasses populations located at each extreme of the Iberian Peninsula. Despite the large geographic distance separating these populations, they exhibit only a minor degree of divergence among haplotypes. In contrast, much higher diversification, in both number of distinct haplotypes, and overall genetic divergence, was observed in the second group of phylogenetically related populations. Surprisingly, this process of radiation and divergence in mtDNA haplotypes occurred in populations in close geographic proximity. All populations sampled in this group are located within a 300 km range, in the central part of our transect across the Northern edge of the Peninsula. Most populations in the central range of our transect exhibit viviparous reproduction ~ which is derived and highly unusual among urodeles. The genetic distances measured among Asturian (central portion of our transect), viviparous populations are higher than the distances measured between the two main taxonomic clusters. A viviparous population showing an unusual level of mtDNA heterogenetiy is reported and the potential implications of this focus of localized variability are discussed. The dynamics of isofemale lineages among the two reproductive modes was further explored in combination with the previous allozyme data. Several nuclear markers suggest that major mtDNA divergences could be explained by long-term extrinsic barriers to gene flow. Isofemale lineages indicate a narrow secondary contact zone among populations with different reproductive patterns. 47. Fructose 2,6-bisphosphate in <em>Dictyostelium discoideum</em>.

Abstract

The occurrence of fructose 2,6-bisphosphate was detected in Dictyostelium discoideum. The levels of this compound were compared with those of cyclic AMP and several glycolytic intermediates during the early stages of development. Removal of the growth medium and resuspension of the organism in the differentiation medium decreased the content of fructose 2,6-bisphosphate to about 20% within 1 h, remaining low when starvation-induced development was fallowed for 8 h. The content of cyclic AMP exhibited a transient increase that did not correlate with the change in fructose 2,6-bisphosphate. If after 1 h of development 2% glucose was added to the differentiation medium, fructose 2,6-bisphosphate rapidly rose to similar levels to those found in the vegetative state, while the increase in cyclic AMP was prevented. The contents of hexose 6-phosphates, fructose 1,6-bisphosphate and those phosphates changed in a way that was parallel to that of fructose 2,6-bisphosphate, and addition of sugar resulted in a large increase in the levels of these metabolites. The content of fructose 2,6-bisphosphate was not significantly modified by the addition of the 8-bromo or dibutyryl derivatives of cyclic AMP to the differentiation medium. These results provide evidence that the changes in fructose 2,6-bisphosphate levels in D. discoideum development are not related to a cyclic-AMP-dependent mechanism but to the availability of substrate. Fructose 2,6-bisphosphate was found to inhibit fructose-1,6-bisphosphatase activity of this organism at nanomolar concentrations, while it does not affect the activity of phosphofructokinase in the micromolar range. 48. Short term regulation by lysine of ornithine decarboxylase activity in Dictyostelium discoideum

Abstract

A transitory increase in ornithine decarboxylase activity has been observed soon after food removal from Dictyostelium discoideumamoeba. This increase can be prevented by supplementation of the differentiation buffer with the 11 amino acids known for their ability to retard the development of this slime mold. Lysine can replace the amino acid mixture with an apparent inhibition constant of 50 micromolar. This inhibition by lysine, which was only observed in vivo, took place within 5 min and was readily reversed upon lysine removal.
