Background: Rapid and accurate detection of viral respiratory infections is important for infection control measures. This study compares the analytical and clinical performance of the Xpert® Xpress CoV-2/Flu/RSV plus test ("Xpert", Cepheid) and the STANDARD™ M10 Flu/RSV/SARS-CoV-2 test ("M10", SD Biosensor). Both tests are quadruplex RT-PCR assays for rapid diagnosis of SARS-CoV-2, influenza A/B and RSV., Study Design: Analytical sensitivities were determined by limit of detection for SARS-CoV-2, influenza A, influenza B and RSV, respectively. Additionally, the clinical performance of the Xpert and the M10 tests was evaluated against standard-of-care RT-PCR by testing of 492 clinical specimens., Results: The analytical sensitivities for Xpert versus M10 test was 10, 50, 50 and 300 versus 300, 200, 800 and 1500 copies/mL for SARS-CoV-2, influenza A, influenza B and RSV, respectively. Clinical sensitivity for the Xpert test was superior across all four pathogens compared to the M10 test. Xpert showed clinical sensitivity of 100 % in all Ct-ranges for all four pathogens whereas M10 showed clinical sensitivity of 100 % in the 25-30 Ct-range, 84-100 % in the 30-35 Ct-range and 47-67 % in the >35 Ct-range across the four pathogens. Translating into real-life clinical sensitivity, the Xpert would detect 100 % of all four pathogens, whereas M10 would detect 92.1, 92.4, 84.8 and 94.7 % for SARS-CoV-2, influenza A, influenza B and RSV., Conclusion: This study demonstrates improved analytical and clinical performance of Xpert Xpress CoV-2/Flu/RSV plus compared to STANDARD M10 Flu/RSV/SARS-CoV-2, which is important for ensuring accuracy of diagnosis at all stages of a respiratory infection., Competing Interests: Declaration of competing interest The authors declare that they have no known competing financial interests or personal relationships that could have appeared to influence the work reported in this paper., (Copyright © 2024 The Author(s). Although biochar application to soils has been found to increase soil quality and crop yield, the biochar dispersion extent and its impacts on native soil organic carbon (SOC) has received relatively little attention. Here, the vertical and lateral migration of fine, intermediate and coarse-sized biochar (<0.5, 0.5-1 and 1-5 mm, respectively), applied at low and high doses (1.5-2 and 3-4% w/w, respectively), was tracked using stable isotope methods, along with its impact on native SOC stocks. Biochar was homogeneously mixed into the surface layer (0-7 cm depth) of a loamy sandy Acrisol in Zambia. After 4.5 y, 38-75% of the biochar carbon (BC) was lost from the applied layer and 4-25% was detected in lower soil layers (7-30 cm). Estimating BC mineralization to be no more than 8%, 25-60% was likely transported laterally out of the experimental plots. This conclusion was supported by observations of BC in the control plot and in soils up to 2 m outside of the experimental plots. These processes were likely progressive as recovery of BC in similar plots 1 year after application was greater in both surface and lower soil layers than after 4.5 y. Fine and intermediate-sized BC displayed the greatest downward migration (25.3 and 17.9%, respectively), particularly when applied at lower doses, suggesting its movement through soil inter-particle spaces. At higher dosages, fine and intermediate-sized particles may have clogged pore, so coarse biochar displayed the greatest downward migration when biochar was applied at higher doses. In the BC treatment plot soil profiles, native SOC stocks were reduced by 2.8 to 24.5% (18.4% on average), i.e. positive priming. However, some evidence suggested that the soils may switch to negative priming over time. In March 2022, we observed samples with a negative fluorescent signal (60.5%, n = 43) for the influenza A matrix gene and a stronger positive signal for subtype A(H3N2). Forty-three samples were positive in InfA (H3N2) (mean Cq 30.9, range 23.9-35.1), and 26 of the 43 samples were negative in InfA matrix (mean Cq 28.0, range 23.2-30.6). Our multiplex test is a laboratory-developed four-target, four-color influenza A reverse-transcription PCR assay targeting the matrix gene, subtypes A(H3N2) and A(H1N1)pdm09. Several samples were negative when retested on commercial influenza Point-of-Care assays. The present work was the first exploration of the use of industrial byproducts from iron and titanium processing as sorbents for the stabilization of soil contamination. The main aim was to test slag waste and iron-rich charred fossil coal ("Fe-char"), as sorbents for per- and polyfluorinated alkyl substances (PFASs), as well as lead (Pb) and antimony (Sb), in four soils from a firefighting training area (PFASs) and a shooting range (Pb and Sb). Adding slag (10-20%) to shooting range soils decreased the leaching of Pb and Sb up to 50-90%. Fe-char amendment to these soils resulted in a moderate reduction in Sb leaching (20-70%) and a slightly stronger effect on Pb (40-50%). The sorption is most likely explained by the presence of Fe oxyhydroxides. These are present in the highest concentrations in the slag, probably resulting in more effective metal binding to the slag than to the Fe-char. Fe-char but not slag proved to be a strong sorbent for PFASs (reducing PFAS leaching from the soil by up to 99.7%) in soil containing low total organic carbon (TOC; 1.2%) but not in high-TOC soil (34%). The sorption coefficient K D for Fe-char was high, in the range of 10 4.3 to 10 6.5 L/kg at 1 ng/L in the low-TOC soil. The K D value increased with increasing perfluorocarbon chain length, exceeding PFAS sorption to biochar in the low ng/L concentration range. This result indicates that the mechanism behind the strong PFAS sorption to Fe-char was mainly van der Waals dispersive interactions between the hydrophobic PFAS-chain and the aromatic π-electron systems on nanopore walls within the Fe-char matrix. Overall, this study indicates that industrial byproducts can provide sustainable and cost-effective materials for soil remediation. We estimated interim influenza A vaccine effectiveness (VE) following a late sharp rise in cases during an influenza A(H3N2)-dominated 2021/22 season, after lifting COVID-19 restrictions. In children aged 2-6 years offered a live attenuated influenza vaccine, adjusted VE was 62.7% (95% CI: 10.9-84.4) in hospitalised and 64.2% (95% CI: 50.5-74.1) in non-hospitalised children. In non-hospitalised patients aged 7-44 years, VE was 24.8% (95% CI: 12.8-35.2); VE was non-significant in remaining age groups and hospital/non-hospital settings.

From ontogenesis to homeostasis, the phenotypes of complex organisms are shaped by the bidirectional interactions between the host organisms and their associated microbiota. Current technology can reveal many such interactions by combining multi-omic data from both hosts and microbes. However, exploring the full extent of these interactions requires careful consideration of study design for the efficient generation and optimal integration of data derived from (meta)genomics, (meta)transcriptomics, (meta)proteomics, and (meta)metabolomics. In this perspective, we introduce the holo-omic approach that incorporates multi-omic data from both host and microbiota domains to untangle the interplay between the two. We revisit the recent literature on biomolecular host-microbe interactions and discuss the implementation and current limitations of the holo-omic approach. We anticipate that the application of this approach can contribute to opening new research avenues and discoveries in biomedicine, biotechnology, agricultural and aquacultural sciences, nature conservation, as well as basic ecological and evolutionary research., (Copyright © 2020 The Authors. Pyrolysis of organic waste or woody materials yields a stable carbonaceous product that can be mixed into soil and is often termed "biochar". During pyrolysis carbon-containing gases are emitted, mainly volatile organic carbon species, carbon monoxide and aerosols. In modern pyrolysis units, gases are after-combusted, which reduces emissions substantially. However, emission data for medium- to large-scale pyrolysis units are scant, both regarding gases, aerosols, heavy metals and polycyclic aromatic hydrocarbons (PAH). Making biochar from lightly contaminated waste timber (WT) is a promising waste handling option as it results in the potential valorization of such residues into e.g. sorbents for contaminant stabilization. For this process to be environmentally sustainable, emissions during the process need to be low and the resulting biochar of sufficient quality. To investigate both issues, we pyrolyzed three batches of WT and one reference batch of clean wood/leaves in a representative medium-scale pyrolysis unit (Pyreg-500, 750 t/year) with after-combustion of the pyrolysis gases, and measured the gas, aerosol, metal and PAH emissions, as well as the characteristics and contamination levels of the resulting biochar, including contaminant leaching. Mean emission factors for the WT were (g/kg biochar); CO = 7 ± 2, non-methane volatile organic compounds (NMVOC) = 0.86 ± 0.14, CH 4  = 0, aerosols (PM 10 ) = 0.6 ± 0.3, total products of incomplete combustion (PIC) = 9 ± 3, PAH-16 = (2.0 ± 0.2) · 10 -5 , As (most abundant metal) = (2.3 ± 1.9) · 10 -3 and NO X  = 0.65 ± 0.10. There were no significant differences in emission factors between the pyrolysis of WT and the reference respectively, except for PM 10 , NMVOC, and PAH-16, which were significantly lower for WT than for the clean wood/leaves. The WT biochar did not satisfy premium or basic European Biochar Certificate criteria due to high levels of zinc and PAH. However, leachable metal contents were <0.1% of total contents.
