BACKGROUND: Neuropsychiatric symptoms in SLE and other systemic autoimmune rheumatic diseases (SARDs) are challenging to diagnose, attribute and manage. We investigated the timings of onset of a broad range of neuropsychiatric (NP) symptoms in relation to timing of SLE onset. In addition, we explored whether NP symptoms may be a prodrome to SARD onset and to subsequent flares. METHODS: We collected patient reports of the timing of their first episode of 29 NP symptoms relative to SLE non-NP symptom onset. Surveys (n = 676 SLE patients and n = 400 clinicians) and interviews (n = 50 clinicians; and n = 69 SARD patients, including 27 SLE patients) were completed from 2022 to 2023, and analysed using mixed methods. FINDINGS: The majority of NP symptoms did not first present around the time of SLE onset, contrary to the prevailing view among many rheumatology participants and in the literature. For example, among patients who experienced hallucinations, 54% reported first presentation >1 year after disease onset. Patient interviews also revealed that a range of NP symptoms may be a prodrome to SLE/SARDs onset and later flares, including symptoms not represented in existing classification criteria. Evidence of a possible prodromal syndrome was elicited from those patients who experienced hallucinations. Of these, 61% (SLE) and 34% (other SARDs) reported increasingly disrupted dreaming sleep (usually nightmares) prior to their hallucinations. In-depth interviews revealed that progression of symptoms in flares showed a high degree of inter-patient variation, whilst symptom progression was often similar in individual patients recurrent flares. INTERPRETATION: Neuropsychiatric symptoms can first present at any stage in the SLE disease course. Attributional decisions should evaluate timings of NP symptoms in relation to timing of SLE/SARD symptom onset rather than time of diagnosis due to frequent diagnostic delays. Greater recognition of prodromal/early NP symptoms indicating impending SLE flares (and potentially other SARD flares) could enable quicker flare identification and treatment. OBJECTIVE: Calcium pyrophosphate deposition (CPPD) disease is prevalent and has diverse presentations, but there are no validated classification criteria for this symptomatic arthritis. The American College of Rheumatology (ACR) and EULAR have developed the first-ever validated classification criteria for symptomatic CPPD disease. METHODS: Supported by the ACR and EULAR, a multinational group of investigators followed established methodology to develop these disease classification criteria. The group generated lists of candidate items and refined their definitions, collected de-identified patient profiles, evaluated strengths of associations between candidate items and CPPD disease, developed a classification criteria framework, and used multi-criterion decision analysis to define criteria weights and a classification threshold score. The criteria were validated in an independent cohort. RESULTS: Among patients with joint pain, swelling, or tenderness (entry criterion) whose symptoms are not fully explained by an alternative disease (exclusion criterion), the presence of crowned dens syndrome or calcium pyrophosphate crystals in synovial fluid are sufficient to classify a patient as having CPPD disease. In the absence of these findings, a score>56 points using weighted criteria, comprising clinical features, associated metabolic disorders, and results of laboratory and imaging investigations, can be used to classify as CPPD disease. These criteria had a sensitivity of 92.2% and specificity of 87.9% in the derivation cohort (190 CPPD cases, 148 mimickers), whereas sensitivity was 99.2% and specificity was 92.5% in the validation cohort (251 CPPD cases, 162 mimickers). CONCLUSION: The 2023 ACR/EULAR CPPD disease classification criteria have excellent performance characteristics and will facilitate research in this field.

Background: Osseointegration depends on primary stability, and further, implant surface modifications may improve secondary stability. This randomized clinical trial evaluates whether adding a fluoride solution enhances the implant stability. Materials and Methods: Stability of dental implants with two different types of surface treatments was compared utilizing resonance frequency analysis (RFA). Twelve patients were recruited: eight females and four males with an average age of 58.3 years and they received either double acid etched implants (control: Porus Implant, Sistema Conexão) and double acid etched implants containing fluoride on their surfaces (test: Porus Nano Implant, Sistema Conexão). Each patient received one implant from the test category and another implant from the control category, resulting in 24 implants in this study. RFA, utilizing Osstell was used to assess the implant stability, immediately after the placement of implant and 15, 30 and 45 days post-implant placement. Results: At the initial time, there was no difference between the test and control groups regarding the ISQ. However, after 15, 30, and 45 days, ISQ values for the test group were significantly higher than the control group. When using implants without fluoride addition, it was observed that after 15 days, the ISQ values were lower than those found at the initial time, but after 30 days, values matched the initial. Concerning the test group results, the ISQ values from T0 (immediately after implant placement) to T3 (45 days after implant placement) always increased from every period of evaluation. Conclusions: It was concluded that double acid etched implants with surfaces containing fluoride solution increases the implant stability quotient, compared to implants with fluoride-free double acid etched surfaces.

Abstract Objective The aim of this study was to evaluate juxta-apical radiolucency (JAR) prevalence and its relationship with adjacent structures using cone-beam computed tomography (CBCT). Material and method CBCT scans were analyzed from a database from 2017 to 2021 of adult patients with mandibular third molars to visualize the presence of JAR. Third molars associated with JAR were classified according to impaction depth, angulation, and root development. JAR was also classified according to its relationship with the mandibular canal and its cortical area, size, and location. The chi-square test and the Mann-Whitney Test were performed to compare the age of patients and gender regarding the presence or absence of JAR. Result 1097 CBCT scans were collected from the database during the evaluation period, and after the application of the eligibility criteria, 155 scans were included in the analysis. Thirty-one third molars showed the presence of JAR with a 20% prevalence. Most cases were observed to be unilateral (84%), located mesially (53%), and medium-sized (53%), in contact with the mandibular canal with cortical presence (50%), visualized in third molars mostly mesioangular (36%), partially intraosseous (86%), and with complete root development (92%). No significant differences were detected between gender and age concerning the presence of JAR. Conclusion High JAR prevalence was observed, which was frequently in contact with the mandibular canal, being considered a potential risk for nerve damage.

ABSTRACT: The objective of the present study was to compare the effectiveness of dietary supplementation of muramidase (MUR) and 2 phytogenic additives on the growth performance, intestinal morphology, bacteria load, and production of short-chain fatty acids (SCFA) of broiler chickens raised under field-like conditions. A total of 6,400 day-old Ross 308 broiler chicks were randomly selected and distributed into 32 floor pens, with 200 chicks (100 males and 100 females)/pen. The treatment groups were an unsupplemented control, and the experimental groups supplemented with MUR at 35,000 LSU(F)/kg of feed, phytogenic 1 (Phyto 1, based on thymol) at 100g/ton feed, or phytogenic 2 (Phyto 2, based on alkaloids) at 60g/ton feed, for a total period of 41 d. A 4-phase feeding program was applied (starter, grower, finisher and withdrawal). The paramenters evaluated were: growth performance, carcass yield, concentration of muranic acid in the jejunum content and excreta, liver enzyme concentration, intestinal morphology, and bacteria enumeration and short and branch chain fatty acids (SCFA and BCFA) in the cecal content. Data were analyzed by ANOVA and Tukey's test was used to separate the means. Soluble muramic acid (MurN) in the jejunum increased with the supplementation of MUR and Phyto 2 when compared to the other groups (P = 0.0001), but only the supplementation of MUR increased the concentration of MurN in the excreta. The supplementation of all feed additives improved the body weight gain and the body weight corrected feed conversion ratio when compared to the control group (P = 0.0001). MUR increased villus heigh (VH) when compared to the control or the other supplemented groups (P = 0.0001), and led to the highest concentration of most SCFA, total BCFA, and total SCFA (P < 0.05). In conclusion, the supplementation of MUR and phytogenics to the diets of broiler chickens improved the growth performance, but MUR, only, was capable of effectively degrading peptidoglycans (PGNs) in both intestinal segments, as well as to increase the abundance of beneficial bacteria and SCFA production.

Diquat, glufosinate and the combination of the latter with carfentrazone or saflufenacil are believed to be as effective for pre-harvest desiccation of soybean plants as paraquat. This work was developed with the objective of evaluating the efficacy of paraquat, diquat, and glufosinate alone or at mixtures of carfentrazone or saflufenacil in pre-harvest desiccation of soybean. Soybean defoliation, maturity and yield were evaluated. Similar efficacy was observed between glufosinate and paraquat in soybean defoliation and maturity, in addition to a greater flexibility of use, in which it could be an anticipator and standardizing agent of the harvest. The application of diquat (200 g of active ingredient [a.i.] ha-1), paraquat (200 g a.i. ha-1) or glufosinate (400 or 500 g a.i. ha-1) was effective for pre-harvest desiccation of soybean at the R7.2 stage. As well, it was observed promising results for application of glufosinate + saflufenacil (300 + 24.5 g a.i. ha-1), glufosinate + carfentrazone (200 + 25 g a.i. ha-1), glufosinate + saflufenacil (200 + 35 g a.i. ha-1). Diquat and glufosinate can replace paraquat in pre-harvest desiccation of soybean plants, and glufosinate also provide greater management flexibility, in anticipating the harvest.

O objetivo deste estudo consistiu em avaliar o potencial de utilização das espécies presentes em áreas campestres e florestais localizadas na Fazenda Santa Rita, com cerca de 200 hectares, situada na região da Coxilha Rica, Lages – SC. Para o levantamento florístico foi adotado o método de coletas por caminhamento, no período de março de 2021 a dezembro de 2022. Os espécimes coletados em campo, todos férteis, foram catalogados e incorporados ao acervo do Herbário LUSC, da Universidade do Estado de Santa Catarina. Foram identificadas 110 espécies de Asteraceae. Para a etapa de verificação dos usos das espécies, foram pesquisados artigos científicos publicados em periódicos digitais disponíveis nos bancos de dados da Scielo, CAPES e Web of Science. Além disso, foram consultados livros impressos e artigos publicados em revistas específicas da área de botânica, para a seleção de trabalhos relevantes que tratavam sobre o potencial de uso de espécies de Asteraceae. As palavras-chave utilizadas na pesquisa foram: "uso econômico da Família Asteraceae", "Asteraceae de uso ornamental", "Asteraceae de uso medicinal", "Asteraceae de uso melífero e apícola", "Asteraceae de uso alimentício convencional e não convencional". Também houve a substituição do nome “Asteraceae” por “Compositae” em cada uma das utilidades, para localização dos artigos, bem como a análise individual das espécies, sendo, posteriormente, as mesmas avaliadas quanto ao seu potencial de uso. Das espécies confirmadas na área de estudo, 60 (54,55%) delas possuem informações sobre o uso, enquanto para as outras 50 (45,45%) espécies não foi possível localizar na literatura o seu potencial de uso. Essa pesquisa evidencia a necessidade de estudos básicos sobre espécies nativas e a importância de aprofundar o conhecimento sobre suas funcionalidades, para a manutenção dos ecossistemas, podendo oferecer diversas aplicações potenciais.

Background: The Myrtaceae family represents one of the most diverse groups in South America. Yet, there are few studies on their reproductive characteristics. Among the species with limited information on their reproductive behavior are Myrceugenia euosma (O. Berg) D. Legrand and Siphoneugena reitzii D. Legrand. Questions: What are the floral attractions and resources of M. euosma and S. reitzii for pollinators? Who are the pollinators of these species? What is the predominant reproductive system in both species? Studied species: M. euosma and S. reitzii, two sympatric Myrtaceae from southern Brazil. Study site and dates: Urubici, Painel, and Rio Rufino, Santa Catarina state, southern Brazil. November 2021 - February 2022. Methods: Information was obtained on morphological aspects, anthesis time, stigmatic viability, floral resources, structures attractive to visitors, olfactory testing, observation and identification of floral visitors, and reproductive system testing. Results: The flowers of M. euosma opens between 11 am and 17 pm (- 20 pm), with full opening occurring mostly at night; S. reitzii flowers open between 4 a.m. and 6 a.m., with a highest peak by 8 a.m. Flowers of both species have receptive stigmas until senescence. Pollen is the primary floral reward and the scent coming from the anthers combined with the floral display attract native bees and Apis mellifera, legitimate pollinators of both species. The lack of fruit development in the self-pollination tests support cross reproduction in both taxa. Conclusions: Both species exhibit traits consistent with melittophily syndrome and potential self-incompatibility. These results are essential for conservation efforts.

Abstract The objective of this work was to evaluate the effectiveness of herbicides when applied alone and in combinations at sourgrass (Digitaria insularis) post-emergence, as well as to identify a substitute to paraquat in sequential application. Field and greenhouse experiments were conducted during the 2019/2020 and 2020/2021 crop seasons. The herbicides applied alone and in combinations were: atrazine, clethodim, clodinafop, diquat, glufosinate, haloxyfop, imazapic, imazapyr, mesotrione, nicosulfuron, paraquat, glyphosate, saflufenacil, tembotrione, and tepraloxydim. In the 2019/2020 crop season, in the experiment conducted in the field, the control of sourgrass was considered low due to the dry weather condition and to the full flowering of the plants. In the greenhouse, a satisfactory control above 80% was observed at 28 days after herbicide application for most treatments. In the 2020/2021 crop season, under field conditions, the application of glyphosate combined with haloxyfop, with a sequential application of glufosinate, resulted in the highest weed control. In the greenhouse, most treatments were effective and, of these, all contained glufosinate. The sequential application of glufosinate or in combinations favors a better control of sourgrass. However, diquat and glufosinate do not differ in efficacy in sequential application and are an option for the control of the weed.

As the beef industry moves towards efficient animal production to improve sustainability in agriculture, new production and management approaches are emerging. Among the many facets of the beef industry, cow–calf operations have the most opportunity for efficiency improvement, including improvements in fertility. This project accounts for measures and methods of (1) twinning reproductive technologies and (2) twin calf perinatal care and pre-weaning rearing. The overall objective was to produce twin calves using two reproductive technologies—embryo transfer and artificial insemination. The subobjectives were to determine accuracy of twin pregnancies embryo/fetal losses using ultrasonography, evaluate parturition and dystocia, and determine the effects of different twin-raising methods on neonatal behavior and growth. A fixed-time artificial insemination (FTAI) protocol was applied to 77 multiparous Angus-cross cows from a commercial beef herd in northcentral South Dakota during the summer of 2019. Cows were assigned to two different treatments groups: only artificially inseminated (AI) or received an embryo transfer following artificial insemination (ET + AI). They were estrous-synchronized, artificially inseminated (AI) with black Angus semen at day 0, and received and embryo transfer (ET) at day 7. Ultrasound examination detected 56% pregnancy risk for both groups, with sensitivity, specificity, and accuracy of 75%, 100%, and 90.5%, respectively, for bilateral twin detection. Calves were born during spring 2020. Twin calves (n = 34) and singleton calves (n = 11) were assigned to one of three raising methods: (1) twin born and twin raised (TT; n = 16), (2) twin born and single raised (TS; n = 18), and (3) single born and single raised (S; n = 11). Neonatal nursing behavior and birth weights were recorded, and adjusted day 200 and day 280 were calculated measures of vitality and growth. Blood samples were collected at age 24 h for colostrum intake measures (total serum protein, IgG1, and IgM). Twin calves were born 20% (p < 0.05) lighter in body weight than singletons; however, weights did not differ at day 280 between TT and S calves. TS calves had the shortest average latency to stand, but immunoglobulin concentrations did not differ among treatments. At weaning, cows that had birthed and raised twins produced more kilograms of live weight per pregnancy than cows birthing and raising singletons. Using ET + AI proved to increase twinning rate, and growth was maintained when raising both twins with their dam.

Abstract Background The intravesical instillation of the paclitaxel-hyaluronan conjugate ONCOFID-P-B™ in patients with bacillus Calmette-Guérin (BCG)-unresponsive bladder carcinoma in situ (CIS; NCT04798703 phase I study), induced 75 and 40% of complete response (CR) after 12 weeks of intensive phase and 12 months of maintenance phase, respectively. The aim of this study was to provide a detailed description of the tumor microenvironment (TME) of ONCOFID-P-B™-treated BCG-unresponsive bladder CIS patients enrolled in the NCT04798703 phase I study, in order to identify predictive biomarkers of response. Methods The composition and spatial interactions of tumor-infiltrating immune cells and the expression of the most relevant hyaluronic acid (HA) receptors on cancer cells, were analyzed in biopsies from the 20 patients enrolled in the NCT04798703 phase I study collected before starting ONCOFID-P-B™ therapy (baseline), and after the intensive and the maintenance phases. Clinical data were correlated with cell densities, cell distribution and cell interactions. Associations between immune populations or HA receptors expression and outcome were analyzed using univariate Cox regression and log-rank analysis. Results In baseline biopsies, patients achieving CR after the intensive phase had a lower density of intra-tumoral CD8+ cytotoxic T lymphocytes (CTL), but also fewer interactions between CTL and macrophages or T-regulatory cells, as compared to non-responders (NR). NR expressed higher levels of the HA receptors CD44v6, ICAM-1 and RHAMM. The intra-tumoral macrophage density was positively correlated with the expression of the pro-metastatic and aggressive variant CD44v6, and the combined score of intra-tumoral macrophage density and CD44v6 expression had an AUC of 0.85 (95% CI 0.68–1.00) for patient response prediction. Conclusions The clinical response to ONCOFID-P-B™ in bladder CIS likely relies on several components of the TME, and the combined evaluation of intra-tumoral macrophages density and CD44v6 expression is a potentially new predictive biomarker for patient response. Overall, our data allow to advance a potential rationale for combinatorial treatments targeting the immune infiltrate such as immune checkpoint inhibitors, to make bladder CIS more responsive to ONCOFID-P-B™ treatment.

Caesalpinioideae is the second largest subfamily of legumes (Leguminosae) with ca. 4680 species and 163 genera. It is an ecologically and economically important group formed of mostly woody perennials that range from large canopy emergent trees to functionally herbaceous geoxyles, lianas and shrubs, and which has a global distribution, occurring on every continent except Antarctica. Following the recent re-circumscription of 15 Caesalpinioideae genera as presented in Advances in Legume Systematics 14, Part 1, and using as a basis a phylogenomic analysis of 997 nuclear gene sequences for 420 species and all but five of the genera currently recognised in the subfamily, we present a new higher-level classification for the subfamily. The new classification of Caesalpinioideae comprises eleven tribes, all of which are either new, reinstated or re-circumscribed at this rank: Caesalpinieae Rchb. (27 genera / ca. 223 species), Campsiandreae LPWG (2 / 5–22), Cassieae Bronn (7 / 695), Ceratonieae Rchb. (4 / 6), Dimorphandreae Benth. (4 / 35), Erythrophleeae LPWG (2 /13), Gleditsieae Nakai (3 / 20), Mimoseae Bronn (100 / ca. 3510), Pterogyneae LPWG (1 / 1), Schizolobieae Nakai (8 / 42–43), Sclerolobieae Benth. & Hook. f. (5 / ca. 113). Although many of these lineages have been recognised and named in the past, either as tribes or informal generic groups, their circumscriptions have varied widely and changed over the past decades, such that all the tribes described here differ in generic membership from those previously recognised. Importantly, the approximately 3500 species and 100 genera of the former subfamily Mimosoideae are now placed in the reinstated, but newly circumscribed, tribe Mimoseae. Because of the large size and ecological importance of the tribe, we also provide a clade-based classification system for Mimoseae that includes 17 named lower-level clades. Fourteen of the 100 Mimoseae genera remain unplaced in these lower-level clades: eight are resolved in two grades and six are phylogenetically isolated monogeneric lineages. In addition to the new classification, we provide a key to genera, morphological descriptions and notes for all 163 genera, all tribes, and all named clades. The diversity of growth forms, foliage, flowers and fruits are illustrated for all genera, and for each genus we also provide a distribution map, based on quality-controlled herbarium specimen localities. A glossary for specialised terms used in legume morphology is provided. This new phylogenetically based classification of Caesalpinioideae provides a solid system for communication and a framework for downstream analyses of biogeography, trait evolution and diversification, as well as for taxonomic revision of still understudied genera.

The objective of the present study was to evaluate the effect of a precision biotic (PB; Symphiome®, dsm-firmenich) on the growth performance, nutrient digestibility, litter quality, blood uric acid (UA) concentration, cecal short-chain fatty acids (SCFA) concentration, and ileal and cecal microbiome of broiler chickens fed corn or wheat-based diets. One-day-old broiler chicks were placed in a completely randomized design (2 × 2 factorial scheme) in 4 treatments, 8 replicates/treatment, and 16 birds/replicate. The treatments were a corn or wheat and soybean meal-based diet without or with supplementation of 0.9 kg/MT of PB. All the birds were vaccinated against coccidiosis at the time of placement (Paracox 5). Growth performance was evaluated weekly and at the end of the experiment. At d 35, blood was collected to determine UA concentration and cecal content for SCFA concentration analysis. Ileal digesta was also collected for nutrient digestibility analysis, and ileal digesta and cecal content were collected for microbiome analysis. The data were submitted to two-way ANOVA (P ≤ 0.05), and LSM was used to separate the means in case of interaction. During the starter and grower phases of the study, a better body weight gain (P = 0.0008 and P = 0.04, respectively) was observed in birds fed wheat compared to corn-based diets. From 28 to 36 d of age, the supplementation of PB increased feed intake (P = 0.05), and tended (P = 0.06) to improve the feed conversion ratio by 4.3 points vs non-supplementated birds. Birds fed wheat-based diets had higher (P = 0.02) blood UA than corn based-diets fed birds, and the supplementation of PB led to a reduction (P = 0.02) of blood UA compared to non-supplemented birds. The supplementation of PB changed the abundance of core metabolic pathways of the microbiome, mostly related to protein metabolism, which led to a reduction in blood UA concentration and increase of cecal SCFA concentrations. In conclusion, by beneficially modulating the microbiome, the supplementation of PB was translated into improved growth performance of broiler chickens fed corn or wheat-based diets.
