This classroom practice article explores the integration of TalkAbroad, an online language conversation platform, in an undergraduate World Languages Education program to support pre-service language teachers' oral proficiency development. Two pre-service teachers, facing challenges like family responsibilities and limited study abroad opportunities, engaged in four 30-minute conversations on topics including media, future plans, current events, and cultural comparisons. Through structured post-conversation activities involving transcription, self-assessment, goal setting, and feedback, students exhibited increased self-awareness and confidence in their linguistic abilities. This article emphasizes the platform's positive impact on student learning and highlights TalkAbroad's potential to support language proficiency development, offering valuable insights for future language education practices.

Refractive errors remain a global health concern, as a large proportion of the world's population is myopic. Current ablative approaches are costly, not without risks, and not all patients are candidates for these procedures. Electromechanical reshaping (EMR) has been explored as a viable cost-effective modality to directly shape tissues, including cartilage. In this study, stromal collagen structure and fibril orientation was examined before and after EMR with second-harmonic generation microscopy (SHG), a nonlinear multiphoton imaging method that has previously been used to study native corneal collagen with high spatial resolution. EMR, using a milled metal contact lens and potentiostat, was performed on the corneas of five extracted rabbit globes. SHG was performed using a confocal microscopy system and all images underwent collagen fibril orientation analysis. The collagen SHG signal in controls is uniform and is similarly seen in samples treated with pulsed potential, while continuous EMR specimens have reduced, nonhomogeneous signal. Collagen fibril orientation in native tissue demonstrates a broad distribution with suggestion of another peak evolving, while with EMR treated eyes a bimodal characteristic becomes readily evident. Pulsed EMR may be a means to correct refractive errors, as when comparing its SHG signal to negative control, preservation of collagen structures with little to no damage is observed. From collagen fiber orientation analysis, it can be inferred that simple DC application alters the structure of collagen. Future studies will involve histological assessment of these layers and multi-modal imaging analysis of dosimetry.

Stress can affect teachers' mental and physical health, student-teacher relationships, and energy levels. During the COVID-19 pandemic, educators around the world experienced stress in terms of worry for their students and for their own well-being in terms of energy levels, sleep and anxiety. The purpose of this study was to examine female music educators' stress, stressors, and well-being during 1 year in the height of the COVID-19 pandemic. We examined these factors with participants, approximately 1 year into the pandemic in spring 2021 and this study explored the same educators' stressors 1 year later in spring 2022. Through this comparative case study, eight self-identified female music educators completed a questionnaire and participated in focus groups. We approached data collection through the theory of wellbeing and uncovered four themes; (a) self-care and mental health; (b) pressure and frustration; (c) scheduling and reprioritization; and (d) accommodation. Results suggest female participants experienced personal and professional stressors during the COVID-19 pandemic that impacted their well-being, with feelings of burnout, anxiety, and desires to possibly leave the teaching profession.

Educators across the country utilize technology in innovative ways to bridge the gap between education and practice. This emphasis is prevalent within helping professions that rely heavily on field placements for further development of students. Digital pedagogy can be used by human services educators to bridge the gap between education and human services practice. In this study, we utilized the Catalyst Framework (Eynon & Gambino, 2017) to delineate student perceptions of their experiences with ePortfolio, demonstrating how digital pedagogy can support human services education. Through qualitative methods, ePortfolios were explored with undergraduate human services students to capture students' experiences and skill development. Three core themes emerged from the findings, suggesting that ePortfolio provides a necessary challenge in the education of human services students. The challenges students reported were meaningful, as ePortfolio increased ownership over their education. Students cultivated a digital space for creativity and authenticity in their education. Implications for human services education are proposed based on the research findings.

IntroductionAttention-Deficit/Hyperactivity Disorder (ADHD) is a neurodevelopmental disorder where hyperactivity often manifests as fidgeting, a non-goal-directed motoric action. Many studies demonstrate fidgeting varies under different conditions as a self-regulating mechanism for attention and alertness during cognitively demanding tasks. Fidgeting has also been associated with reaction time variability. However, a lack of standard variables to define and quantify fidgeting can lead to discrepancies in data and interpretability issues across studies. Furthermore, little is known about fidgeting in adults with ADHD compared to youth. This study aims to design a framework to quantify meaningful fidgeting variables and to apply them to test the relation between fidgeting and performance on a cognitive task, the Flanker, in adults with ADHD.MethodOur study included 70 adult participants diagnosed with ADHD, aged 18-50 years (30.5 ± 7.2 years). Screening included a structured clinical interview, childhood, current self and current observer ratings of ADHD symptoms. Actigraphy devices were attached to the left wrist and right ankle during completion of a cognitive control, attention task (the Flanker). Laboratory testing was subsequently completed on a single day. The relation between task performance, reaction time variability and fidgeting was examined.Results and discussionOur analysis revealed increased fidgeting during correct trials as defined by our new variables, consistent with previous observations. Furthermore, differences in fidgeting were observed between early and later trials while the percentage of correct trials were not significantly different. This suggests a relation between the role of fidgeting and sustaining attention. Participants with low reaction time variability, that is, those with more consistent reaction times, fidgeted more during later trials. This observation supports the theory that fidgeting aids arousal and improves sustained attention. Finally, a correlation analysis using ADHD-symptom rating scales validated the relevance of the fidget variables in relation to ADHD symptom severity. These findings suggest fidgeting may be a compensatory mechanism that aids in sustained attention for those with ADHD, although alternative explanations exist.ConclusionOur study suggests that fidgeting may aid in sustained attention during the attention-demanding, cognitive control processes for adults with ADHD, with more fidgeting observed during correct trials and among participants with lower reaction time variability. Furthermore, the newly defined fidget variables were validated through a significant correlation with ADHD rating scales. By sharing our implementation of fidget variables, we hope to standardize and encourage further quantitative research into the role of fidgeting in ADHD.

Abstract We include the quark to gluon and gluon to quark shock-wave transition operators into the small Bjorken-x evolution equations for helicity in the flavor-singlet channel derived earlier in [1–3]. While such transitions do not affect the large-N c version of the evolution equations for helicity, the large-N c & N f equations are affected. (N c and N f are the numbers of quark colors and flavors, respectively.) We derive the corresponding corrected large-N c & N f equations for the polarized dipole amplitudes contributing to the flavor-singlet quark and gluon helicity distributions in the double-logarithmic approximation (DLA), resumming powers of α s ln2(1/x) with α s the strong coupling constant. We solve these equations iteratively and extract the polarized splitting functions up to four loops. We show that our splitting functions agree with the fixed-order perturbative calculations up to and including the existing three-loops results [4–7]. Similar to the large-N c helicity evolution in the shock-wave approach [8], our large-N c & N f small-x splitting functions agree with those obtained in the infrared evolution equations framework from [9, 10] up to three loops, but appear to slightly disagree at four loops.

Abstract Microbubbles (MBs) combined with focused ultrasound (FUS) has emerged as a promising noninvasive technique to permeabilize the blood–brain barrier (BBB) for drug delivery into the brain. However, the safety and biological consequences of BBB opening (BBBO) remain incompletely understood. This study aims to investigate the effects of two parameters mediating BBBO: microbubble volume dose (MVD) and mechanical index (MI). High-resolution MRI-guided FUS was employed in mouse brains to assess BBBO by manipulating these two parameters. Afterward, the sterile inflammatory response (SIR) was studied 6 h post-FUS treatment. Results demonstrated that both MVD and MI significantly influenced the extent of BBBO, with higher MVD and MI leading to increased permeability. Moreover, RNA sequencing revealed upregulation of major inflammatory pathways and immune cell infiltration after BBBO, indicating the presence and extent of SIR. Gene set enrichment analysis identified 12 gene sets associated with inflammatory responses that were significantly upregulated at higher MVD or MI. A therapeutic window was established between therapeutically relevant BBBO and the onset of SIR, providing operating regimes to avoid damage from stimulation of the NFκB pathway via TNFɑ signaling to apoptosis. These results contribute to the optimization and standardization of BBB opening parameters for safe and effective drug delivery to the brain and further elucidate the underlying molecular mechanisms driving sterile inflammation.

Abstract Background Bed bugs, Cimex lectularius, form day-time aggregations from which they depart at night to feed on human blood. Obtaining an initial blood meal is a critical step in the development of first instars. Previous research had shown that first instars had greater success in obtaining this essential meal when in the presence of adults than when they were alone. Methods Feeding by bed bugs was tested in upright vertical cylindrical chambers fitted with a paper ramp to aid in climbing toward a blood feeder suspended across the upper end of the cylinder. Feeding success by the first instars was tested when they were alone in the chamber or when they were in the presence of adult females, males, or both together. Results The mean proportions of the first instars that fed were significantly higher when they were confined with adults of both sexes or adult females than when they were confined alone or with males. Feeding by adult males was also enhanced by confinement with females. When first instars and adult females were confined together, the mean duration before first instars began feeding was longer than for females. There was no difference in feeding success by first instars confined with their mothers or nonmothers. Conclusions Elevated feeding by first instars and adult males in the presence of females may be adaptive traits that enhance fitness. First instars must feed to avoid dehydration and starvation and to obtain resources needed for development. Adult males would benefit not only by increased feeding success but also by greater likelihood of finding a recently engorged female with which to mate. The lack of any difference in feeding success of first instars in the presence of their mothers or nonmothers argues against parental care in this species. Graphical Abstract

The eukaryotic translation initiation factor eIF4E acts as a multifunctional factor that simultaneously influences mRNA processing, export, and translation in many organisms. Its multifactorial effects are derived from its capacity to bind to the methyl-7-guanosine cap on the 5’end of mRNAs and thus can act as a cap chaperone for transcripts in the nucleus and cytoplasm. In this review, we describe the multifactorial roles of eIF4E in major mRNA-processing events including capping, splicing, cleavage and polyadenylation, nuclear export and translation. We discuss the evidence that eIF4E acts at two levels to generate widescale changes to processing, export and ultimately the protein produced. First, eIF4E alters the production of components of the mRNA processing machinery, supporting a widescale reprogramming of multiple mRNA processing events. In this way, eIF4E can modulate mRNA processing without physically interacting with target transcripts. Second, eIF4E also physically interacts with both capped mRNAs and components of the RNA processing or translation machineries. Further, specific mRNAs are sensitive to eIF4E only in particular mRNA processing events. This selectivity is governed by the presence of cis-acting elements within mRNAs known as USER codes that recruit relevant co-factors engaging the appropriate machinery. In all, we describe the molecular bases for eIF4E’s multifactorial function and relevant regulatory pathways, discuss the basis for selectivity, present a compendium of ~80 eIF4E-interacting factors which play roles in these activities and provide an overview of the relevance of its functions to its oncogenic potential. Finally, we summarize early-stage clinical studies targeting eIF4E in cancer.

We present the last part of this special issue on Use of Artificial Intelligence in Communication: Ethical Implications for Media. This editorial examines the role that humans should play in the responsible use of generative artificial intelligence (GenAI) in the media. It provides an overview of transparency as an ethical obligation in relation to trust and truthfulness when using GenAI to create different kinds of content. Finally, it enters into the debate about the moral status of machines as we anticipate ethical questions in the study of ethics and GenAI in the media. Six articles complete this special issue with studies examining implications of GenAI for semiotics research, public relations, advertising, fact checking, information literacy and journalism education.

Introduction: Monitoring the atypical antipsychotic drug clozapine is crucial to ensure patient safety. This article showcases a high-throughput analytical method for measuring clozapine and its primary metabolite norclozapine (N-desmethylclozapine) in serum using paper spray mass spectrometry (PS-MS). Objectives: This study aimed to assess the viability of a PS-MS method for the rapid measurement of clozapine and norclozapine in human serum samples as an alternative to liquid chromatography mass spectrometry (LC-MS). Methods: Serum samples were processed by protein precipitation followed by deposition of the supernatant containing labelled internal standards onto paper spray substrates mounted in cartridges. Analytes were then analyzed using a triple quadrupole mass spectrometer equipped with a commercial paper spray ionization source. The results obtained from the patient samples were compared to those from a validated LC-MS assay. Results: PS-MS calibrations for clozapine and norclozapine were linear (R2 > 0.99) over five days. Between-run precision was below 8 %, and within-run precision did not exceed 10 %. When compared to a validated LC-MS method, the mean bias for 39 patient samples was −9% for clozapine and −1% for norclozapine, with no outliers. Mass spectrometry ion ratio comparisons indicated no interference for patient samples above the lower limit of quantification. There was less than 7 % change in the measured concentrations of both analytes over five days for samples dried on paper substrates. Notably, virtually no maintenance of the MS source was required during this study. Conclusion: This study illustrates the potential of PS-MS for serum drug monitoring in the clinical laboratory.

Summary: Hepatocellular carcinoma (HCC) remains a leading cause of cancer-related deaths worldwide, and a large proportion is attributable to viral causes, including hepatitis B (HBV) and C viruses (HCV). The pathogenesis of viral-mediated HCC can differ between HBV and HCV, but it is unclear how much these differences influence the tumors’ final molecular and immune profiles. Additionally, there are known sex differences in the molecular etiology of HCC, but sex differences have not been explored in the context of viral-mediated HCC. To determine the extent to which the viral status and sex impact the molecular and immune profiles of HCC, we performed differential expression and immune cell deconvolution analyses. We identified a large number of differentially expressed genes unique to the HBV or HCV tumor:tumor-adjacent comparison. Pathway enrichment analyses demonstrated that changes unique to the HCV tumor:tumor-adjacent tissue were dominated by changes in immune pathways. Immune cell deconvolution demonstrated that HCV tumor-adjacent tissue had the largest immune cell infiltrate, with no difference in the immune profiles within HBV and HCV tumor samples. Overall, this work demonstrates the convergence of HBV- and HCV-mediated HCC on a similar transcriptomic landscape and immune profile despite differences in the surrounding tissue.

Clostridioides difficile is a spore-forming pathogen and the most common cause of healthcare-associated diarrhea and colitis in the United States. Besides producing the main virulence factors, toxin A (TcdA) and toxin B (TcdB), many of the common clinical strains encode the C. difficile transferase (CDT) binary toxin. The role of CDT in the context of C. difficile infection (CDI) is poorly understood. Inflammation is a hallmark of CDI and multiple mechanisms of inflammasome activation have been reported for TcdA, TcdB, and the organism. Some studies have suggested that CDT contributes to this inflammation through a TLR2-dependent priming mechanism that leads to the suppression of protective eosinophils. Here, we show that CDT does not prime but instead activates the inflammasome in bone marrow-derived dendritic cells (BMDCs). In bone marrow-derived macrophages (BMDMs), the cell binding and pore-forming component of the toxin, CDTb, alone activates the inflammasome and is dependent on K+ efflux. The activation is not observed in the presence of CDTa and is not observed in BMDMs derived from Nlrp3-/- mice suggesting the involvement of the NLRP3 inflammasome. However, we did not observe evidence of CDT-dependent inflammasome priming or activation in vivo. Mice were infected with R20291 and an isogenic CRISPR/Cas9-generated R20291 ΔcdtB strain of C. difficile. While CDT contributes to increased weight loss and cecal edema at 2 days post infection, the relative levels of inflammasome-associated cytokines, IL-1β and IL-18, in the cecum and distal colon are unchanged. We also saw CDT-dependent weightloss in Nlrp3-/- mice, suggesting that the increased weightloss associated with the presence of CDT is not a result of NLRP3-dependent inflammasome activation. This study highlights the importance of studying gene deletions in the context of otherwise fully isogenic strains and the challenge of translating toxin-specific cellular responses into a physiological context, especially when multiple toxins are acting at the same time.

Students in Yiddish supplementary schools used texts produced by educators steeped in a diaspora nationalist pedagogy that reflected the ideological coupling of Yiddish and Yiddishkeit: the Yiddish language informed one’s sense of Jewishness. By the 1960s, doodles students left in their schoolbooks challenged this coupling of language and identity. Though it is generally supposed that Yiddish primers ultimately tell us more about the aspirations of adults than they do about the experiences of children, reading these texts together reveals that children evolved their own relationship between Yiddish and Jewishness that was far more subtle than what they encountered in their textbooks. Postwar primers emphasize maintaining the Yiddish language on American soil, to the exclusion of the external culture; children’s doodles argue that more important than preserving the language was locating Yiddishkeit in the culture around them.

PURPOSE: While there is growing evidence for resilience building programmes, to date research has not explored how professionals construct understanding of programme impact. RISE (resilience, integration, self-awareness, engagement), a 5-day yoga-based retreat programme, has been linked with positive wellness outcomes. This qualitative inquiry explores participants reflection and experience 3 months after programme completion. METHODS: Through a grounded constructivist lens, in-depth semi-structured phone interviews were conducted with 17 adult professionals in high-stress work environments who attended RISE. Initial hand-coding informed codebook development for systematic coding using directed content analysis using sensitizing structuring. RESULTS: Two integrated perceptions woven through five themes. Persistent threads of experiential learning and sense of permission provided structure for themes found. Five interrelated themes related to psychological health and workplace dynamics were (1) use of acquired behavioural skills and practices; (2) lived mindfulness; (3) resilience to stress and emotion regulation (4) self-care and self-compassion, and (5) sharing with others. CONCLUSION: Findings provide meaningful interpretation of previously reported programme efficacy by contextualizing perceived benefits within participants constructed understanding of change. Specifically, environmental, social, and experiential considerations have suggested implications for resilience building programmes.Abbreviations: RISE (resilience, integration, self-awareness, engagement).

Mutations in the MAPT gene that encodes tau lead to frontotemporal dementia (FTD) with pathology evident in both cerebral neurons and glia. Human cerebral organoids (hCOs) from individuals harboring pathogenic tau mutations can reveal the earliest downstream effects on molecular pathways within a developmental context, generating interacting neurons and glia. We found that in hCOs carrying the V337M and R406W tau mutations, the cholesterol biosynthesis pathway in astrocytes was the top upregulated gene set compared with isogenic controls by single-cell RNA sequencing (scRNA-seq). The 15 upregulated genes included HMGCR, ACAT2, STARD4, LDLR, and SREBF2. This result was confirmed in a homozygous R406W mutant cell line by immunostaining and sterol measurements. Cholesterol abundance in the brain is tightly regulated by efflux and cholesterol biosynthetic enzyme levels in astrocytes, and dysregulation can cause aberrant phosphorylation of tau. Our findings suggest that cholesterol dyshomeostasis is an early event in the etiology of neurodegeneration caused by tau mutations.

The COVID-19 pandemic affected educators around the world. The purpose of the study was to examine music educators' stress and stressors during the COVID-19 pandemic. We previously examined these factors with participants approximately one year into the pandemic from March to May 2021 (Koner et al., 2022). The current study explored these same music educators' stressors one year later, from March to April 2022, specifically exploring the research questions: (1) how do music educators describe the impact on their students' learning and creating music through the COVID-19 pandemic; and (2) how have music educators professionally navigated an academic school year through a global pandemic? We analyzed data through open coding (Gibbs, 2007), which yielded the themes of logistics, student engagement, student behavior, and administrative expectations. The findings suggest that music educators have a continued need for support from their peers and colleagues, parents and guardians, administrators, and the professional community. Implications for future research are also discussed.

The purpose of the study was to examine the impact of yoga as a mindfulness practice on music educators' stress and stress management. We designed this multiple-case study through the conceptual framework of practicing yoga to reduce teachers' stress because yoga reduces stress in some populations. Participants attended a weekly virtual yoga class for 45 min and answered weekly reflection questions from the "Five Facet Mindfulness Questionnaire" and the "Mindfulness Attention Awareness Scale." Findings indicated that yoga positively impacted participants in terms of improving mood, reducing stress, feeling more relaxed, remembering to breathe, and maintaining more focus while teaching. Participants also indicated that they were bringing yoga and mindfulness practices into their teaching and focusing on their breath to help with stress management.

Grounded in a desire to explore how the field of culturally responsive pedagogy (CRP) and its connected sub-fields is constructed/shaped in and through mathematics education research, the study reported on in this paper sought to understand how researchers/scholars situate, or position their research. The study began from a conceptualization of CRP as a term that encompasses the areas (or sub-fields) of Indigenous education [IE], equity-based research [E-b], critical mathematics [CM], ethnomathematics [EM], social justice [SJ], and language diversity [LD]. By asking mathematics education scholars to position their research within this conceptualization, this paper points to the different themes or 'noticings' that summarize the ways in which they described their research and/or the sub-fields. As mathematics education scholars ourselves, this paper is part of our journey to explore these varied and interconnected terms/fields and to come to a more nuanced understanding of how they interact so that we (and others) might act within our research and our classrooms with a clear aim to disrupt. In our efforts to portray some of the fluidity that we noticed, we each present a model that attempts to capture our thinking and positioning within the conceptualization, and we invite the reader to do the same.
