2. Prevalence of childhood hypertension and associated factors in Zhejiang Province: a cross-sectional analysis based on random forest model and logistic regression

Abstract

Abstract With childhood hypertension emerging as a global public health concern, understanding its associated factors is crucial. This study investigated the prevalence and associated factors of hypertension among Chinese children. This cross-sectional investigation was conducted in Pinghu, Zhejiang province, involving 2,373 children aged 8–14 years from 12 schools. Anthropometric measurements were taken by trained staff. Blood pressure (BP) was measured in three separate occasions, with an interval of at least two weeks. Childhood hypertension was defined as systolic blood pressure (SBP) and/or diastolic blood pressure (DBP) ≥ age-, sex-, and height-specific 95th percentile, across all three visits. A self-administered questionnaire was utilized to collect demographic, socioeconomic, health behavioral, and parental information at the first visit of BP measurement. Random forest (RF) and multivariable logistic regression model were used collectively to identify associated factors. Additionally, population attributable fractions (PAFs) were calculated. The prevalence of childhood hypertension was 5.0% (95% confidence interval [CI]: 4.1–5.9%). Children with body mass index (BMI) ≥ 85th percentile were grouped into abnormal weight, and those with waist circumference (WC) > 90th percentile were sorted into central obesity. Normal weight with central obesity (NWCO, adjusted odds ratio [aOR] = 5.04, 95% CI: 1.96–12.98), abnormal weight with no central obesity (AWNCO, aOR = 4.60, 95% CI: 2.57–8.21), and abnormal weight with central obesity (AWCO, aOR = 9.94, 95% CI: 6.06–16.32) were associated with an increased risk of childhood hypertension. Childhood hypertension was attributable to AWCO mostly (PAF: 0.64, 95% CI: 0.50–0.75), followed by AWNCO (PAF: 0.34, 95% CI: 0.19–0.51), and NWCO (PAF: 0.13, 95% CI: 0.03–0.30). Our results indicated that obesity phenotype is associated with childhood hypertension, and the role of weight management could serve as potential target for intervention.

4. Policy-making as an emotionally-charged arena: the emotional geographies of urban cultural policy-making

Analysis of the role emotions play in a range of social processes has increased significantly, but is neglected in the context of cultural policy-making. Recent literatures in feminist and emotional geographies draw attention to how emotions are emergent in, and play a role in shaping, a broad range of social contexts and processes, while other literatures stress the need to ‘personalise’ the expert and consider the emotional aspects of planning. Inspired by these literatures we deploy the notion of ‘affective urbanism’ to study how emotions are interwoven with cultural policy spaces in the city and explore the ‘emotional regimes’ that incorporate emotions with the multi-scalar politics that is shaping urban cultural policy-making. This is undertaken through an analysis of emotions in the working lives and political contexts of cultural policy-makers in Stockholm (Sweden), Gdańsk (Poland) and Manchester (UK). Overall the paper seeks to develop a research agenda that places emotions centrally in studies of cultural policy formation and implementation.

6. Intra-urban connectedness, policy mobilities and creative city-making: national conservatism vs. urban (neo)liberalism

This article aims to advance the literature on policy mobility by decentring the primacy of mobility itself and focusing on understanding what cities do in order to ‘arrive at’ localized versions of urban policy in relation to globally circulating ideas around creativity. The paper explores the performance of a particular local ‘creative economy’ in terms of institutional and strategic adjustments, key drivers and individuals and events, and the role of long-term local, national and international influences on ‘creative cityness’. It does this through an analysis of cultural and creativity policy and local stakeholders in the cultural policy scene in Gdańsk, Poland, focusing on the local performative aspects of mobile policies and arguing the need to understand the formation of a ‘common local project’ as a form of intra-urban connectedness alongside inter-urban connectedness. The paper extends the range of contexts in which the ‘creative city’ has been analysed to include post-socialist, post-European Union accession Central and Eastern Europe, thus making an original contribution by studying these issues in the context of the complex multi-scalar relations between the city, national government and the supranational European Union and the ideological conflict between national authoritarian neoliberalism and urban and supranational scale (neo-)liberalism.

Abstract

Abstract General and central obesity are suggested to be associated with elevated blood pressure (BP), whereas few studies have investigated their combined associations with hypertension in children. This study aimed to assess the associations of combinations of general obesity and central obesity with hypertension in Chinese children, including its stages and phenotypes. A total of 5430 children aged 7–17 years in Zhejiang Province were enrolled. General obesity was evaluated by body mass index (BMI), while central obesity was by waist circumference (WC). Then all children were sorted into three mutually exclusive groups: normal weight with or with no central obesity (NW), abnormal weight with no central obesity (AWNCO), and abnormal weight with central obesity (AWCO). Hypertension was defined as either a systolic or diastolic BP ≥ 95th percentile, and further classified into stage 1 hypertension, stage 2 hypertension, isolated systolic hypertension (ISH), isolated diastolic hypertension (IDH), and systolic diastolic hypertension (SDH). Logistic regression was used. AWNCO and AWCO were associated with stage 1 hypertension (AWNCO, odds ratio [OR] = 1.94, 95% confidence interval [CI]: 1.59‐2.37; AWCO, 2.67, 2.20‐3.25), stage 2 hypertension (AWNCO, 2.35, 1.33‐4.13; AWCO, 4.53, 2.79‐7.37), ISH (AWNCO, 2.50, 1.96‐3.18; AWCO, 3.95, 3.15‐4.95), and SDH (AWNCO, 2.48, 1.75‐3.52; AWCO, 2.78, 1.94‐3.99). Children with AWCO were more likely to have stage 1 and stage 2 hypertension, as well as ISH and SDH. The combined measurement of general and central obesity is suggested as an appropriate screening tool for hypertension among children and adolescents.

19. Artists and creative city policy: resistance, the mundane and engagement in Stockholm, Sweden

© 2016 Elsevier Ltd.Much of the literature around notions of the 'creative class' and the 'creative city' has placed artists as a central, typical creative group. However, that literature has often placed artists in a conceptual dichotomy - either they are seen as uncritical champions of creative city policy (because it boosts their profile and markets) or they are placed in radical opposition to it. This paper explores the attitudes of a sample of artists in Stockholm, Sweden to open this dichotomy up to a more nuanced critique. The analysis considers the diversity of views, attitudes and perceptions of these artists towards creative city policy. While opposition and resistance to the application of creative city policy can certainly be found, the paper seeks to move beyond this to examine how the lack of accord between creative producers and policy-makers can be the outcome of more mundane, everyday practices. In addition, artists join together in specific projects and loose, ephemeral networks to address the issues surrounding the implementation of creative city policy in ways which oppose it but also seek alternatives through engaging planners and the public. Overall the paper calls for an understanding of artists which goes beyond the enthusiast/opponent dichotomy towards developing an understanding of the diverse range of artist responses and engagement with creative city policy.

Abstract

Cell surface mucins configure the cell surface by presenting extended protein backbones that are heavily O-glycosylated. The glycopeptide structures establish physicochemical properties at the cell surface that enable and block the formation of biologically important molecular complexes. Some mucins, such as MUC1, associate with receptor tyrosine kinases and other cell surface receptors, and engage in signal transduction in order to communicate information regarding conditions at the cell surface to the nucleus. In that context, the MUC1 cytoplasmic tail (MUC1CT) receives phosphorylation signals from receptor tyrosine kinases and serine/threonine kinases, which enables its association with different signaling complexes that conduct these signals to the nucleus and perhaps other subcellular organelles. We have detected the MUC1CT at promoters of over 500 genes, in association with several different transcription factors, and have shown that promoter occupancy can vary under different growth factor conditions. However, the full biochemical nature of the nuclear forms of MUC1 and its function at these promoter regions remain undefined. I will present evidence that nuclear forms of the MUC1CT include extracellular and cytoplasmic tail domains. In addition, I will discuss evidence for a hypothesis that the MUC1CT possesses a novel catalytic function that enables remodeling of the transcription factor occupancy of promoters, and thereby engages in regulation of gene expression.

21. Artists and creative city policy: Resistance, the mundane and engagement in Stockholm, Sweden

Abstract

Much of the literature around notions of the ’creative class’ and the ‘creative city’ has placed artists as a central, typical creative group. However, that literature has often placed artists in a conceptual dichotomy - either they are seen as uncritical champions of creative city policy (because it boosts their profile and markets) or they are placed in radical opposition to it. This paper explores the attitudes of a sample of artists in Stockholm, Sweden to open this dichotomy up to a more nuanced critique. The analysis considers the diversity of views, attitudes and perceptions of these artists towards creative city policy. While opposition and resistance to the application of creative city policy can certainly be found, the paper seeks to move beyond this to examine how the lack of accord between creative producers and policy-makers can be the outcome of more mundane, everyday practices. In addition, artists join together in specific projects and loose, ephemeral networks to address the issues surrounding the implementation of creative city policy in ways which oppose it but also seek alternatives through engaging planners and the public. Overall the paper calls for an understanding of artists which goes beyond the enthusiast/opponent dichotomy towards developing an understanding of the diverse range of artist responses and engagement with creative city policy.

22. Conceptual export and theory mobilities: exploring the reception and development of the “creative city thesis” in the post-socialist urban realm

Abstract

This paper addresses the limited contribution of scholarship from within/on the post-socialist urban arena to global urban studies, a phenomenon attributed to the influence of a hegemonic Anglo-American academic complex. We seek to present a more nuanced account by considering scholarship on the “creative city” in a post-socialist context. A numerical analysis of English language publications confirms the lack of impact of scholarship from/on post-socialist areas, though we do identify literature which may be “theory exporting” and emphasize the temporal dimension of the development of scholarship. We then consider the interaction of three global mobilities to present a more nuanced account of this pattern – the “creative city” thesis as globally mobile urban policy, the neoliberalization of universities as a globally mobile restructuring of the context in which these inequalities in knowledge-production are produced, and urban studies theorizing itself as a set of globally mobile concepts and practices. We therefore explore the dynamic interaction of a particular urban phenomenon (“creative city” policy) with academic knowledge production. Adopting this perspective allows us to emphasize other factors such as path dependencies within post-socialist areas and to give due emphasis to agency within the region and how these interact with global processes of neoliberalizing academia.

Abstract

Background. Rotavirus-induced diarrhea poses a worldwide medical problem in causing substantial morbidity and mortality among children in developing countries. We therefore developed a system for passive immunotherapy in which recombinant lactobacilli constitutively express neutralizing variable domain of llama heavy-chain (VHH) antibody fragments against rotavirus. Methods. VHH were expressed in Lactobacillus paracasei, in both secreted and cell surface-anchored forms. Electron microscopy was used to investigate the binding efficacy of VHH-expressing lactobacilli. To investigate the in vivo function of VHH-expressing lactobacilli, a mouse pup model of rotavirus infection was used. Results. Efficient binding of the VHH antibody fragments to rotavirus was shown by enzyme-linked immunosorbent assay and scanning electron microscopy. VHH fragments expressed by lactobacilli conferred a significant reduction in infection in cell cultures. When administered orally, lactobacilli-producing surface-expressed VHH markedly shortened disease duration, severity, and viral load in a mouse model of rotavirus-induced diarrhea when administered both fresh and in a freeze-dried form. Conclusions. Transformed lactobacilli may form the basis of a novel form of prophylactic treatment against rotavirus infections and other diarrheal diseases. © 2006 by the Infectious Diseases Society of America. Abstract

Adherence of Actinomyces naeslundii ATCC 12104 to hydroxyapatite beads coated with protein fractions of parotid saliva, obtained by gel filtration on S-200 HR columns, showed Ga1NAcβ1-3Ga1α-O-ethyl-inhibitable binding to high-molecular-weight proteins (Strömberg et al., 1992). The present study investigates the nature of these high-molecular-weight binding proteins and determines their specific ability to mediate adherence to representative strains of Actinomyces species. Strain ATCC 12104 bound specifically in a lactose-inhibitable manner to the heavy chain of secretory immunoglobulin A (S-IgA), contained within a high-molecular- weight parotid protein fraction separated on SDS-PAGE and transferred to a solid membrane support. Lactose-inhibitable binding to the heavy chain of S-IgA from human colostrum was also demonstrated. Peanut agglutinin bound to the heavy chain of parotid and colostrum S-IgAs contained on solid support membranes, confirming the presence of Ga1β1-3Ga1NAc residues on these molecules. Both salivary and colostrum S-IgA aggregated with strain ATCC 12104 in a Ga1NAcβ-3Ga1α-O-ethyl-inhibitable fashion. Further separation of high-molecular-weight salivary proteins on S-500 HR columns showed Ga1NAcβ-3Ga1α-O-ethyl-inhibitable binding to both mucin- and S-IgA-containing fractions. The presence of S-IgA in salivary pellicles formed in vivo on teeth was demonstrated by Western blot analysis of pellicle extracts with anti-IgA antibodies. Among strains representing A. naeslundii genospecies 1 and 2 and A. odontolyticus, only those of genospecies 1 with a particular adherence profile showed efficient Ga1NAcβ-3Ga1α-O-ethyl-inhibitable binding to S-IgA. Thus, oligosaccharides on S-IgA may promote bacterial aggregation (or adherence) and provide a mechanism by which S-IgA can interact with bacteria without prior immunological challenge. Abstract

Pseudomonas aeruginosa is becoming an increasingly prevalent pathogen, capable of causing numerous life threatening infections in immunocompromised patients. The three hierarchically arranged quorum sensing (QS) systems, namely las, rhl, and pqs play key roles in coordinating virulence expression in P. aeruginosa. However, the regulatory mechanisms of the pqs system have not been fully elucidated. To identify new genes involved in synthesis of the Pseudomonas quinolone signal (PQS), a transposon mutagenesis library was constructed. PA1429 was found to inhibit PQS biosynthesis. The PA1429 deletion mutant also exhibited increased bacterial motility, biofilm formation, and virulence in a mouse model of acute lung infection. Interestingly, it also displayed reduced tolerance to oxidative stress. Mutated pqsH in the PA1429 deletion background restored bacterial susceptibility to H2O2. In addition, our data showed that PA1429 repressed the expression of las and rhl systems. Overall, these results provide new insights into the complex regulatory networks of quorum-sensing and virulence expression in P. aeruginosa.

45. Properties of a transfer RNA lacking modified nucleosides.

Abstract

A transfer RNA complete devoid of modified nucleosides was synthesized by in vitro transcription, and some of its properties in aminoacylation and protein synthesis in vitro were studied. For this purpose, a plasmid was constructed which contained a glycine tRNA gene from Mycoplasma mycoides under the promoter of the T7 RNA polymerase, as well as a BstNI restriction site at the 3′-end of the tRNA gene. Cleavage of plasmid DNA with BstNI followed by T7 RNA polymerase transcription in vitro yielded an RNA which was processed with M1 RNA, the catalytic subunit of ribonuclease P, to give a tRNA of mature length. The tRNA synthesized in this manner can be esterified with glycine in vitro, and the rate of aminoacylation is the same as when using the corresponding fully modified glycine tRNA from M. mycoides. Furthermore, in protein synthesis in vitro, the tRNA lacking modified nucleosides was essentially as efficient as the corresponding normal glycine tRNA. However, the Escherichia coli extract used in our protein-synthesizing system introduced one modification, pseudouridine, into the in vitro-synthesized tRNA, and it cannot be excluded that this modification has an essential role in protein synthesis.

46. Unconventional reading of the glycine codons.

Abstract

We have used a protein-synthesizing in vitro system programmed with the phage message MS2-RNA to investigate the ability of glycyl-tRNAs with different anticodons to read the glycine codons. Under conditions of no competition, when the glycyl-tRNA analyzed was the only source of glycine for protein synthesis, each of the isoacceptors tested, tRNA1Gly (anticodon CCC), tRNA2Gly (anticodon N/UCC), tRNA3Gly (anticodon GCC) from Escherichia coli, and tRNAGly (anticodon UCC) from Mycoplasma mycoides, could read all of the glycine codons in the MS2 coat protein cistron (GGU, GGC, GGA, and GGG). However, tRNA1Gly seemed to have difficulties reading through the whole cistron. Experiments in which two glycyl-tRNAs competed for the same codon showed that the mycoplasma tRNAGly (anticodon UCC) was almost as efficient in the unorthodox reading of the codons GGU and GGC as it was in conventional reading. It would seem to be the only tRNAGly present in Mycoplasma mycoides and our results are consistent with this finding since the mycoplasma tRNAGly appears to have been designed to read all four glycine codons with approximately equal efficiency. The competition experiments furthermore showed that E. coli tRNA1Gly (anticodon CCC) reads the codon GGA more efficiently than it reads GGU and GGC suggesting that the mispair C . A between the wobble position of the anticodon and the third codon position might have appreciable stability.

Abstract

Site-directed mutagenesis has been used to change the nucleotide C in the wobble position of tRNA(1Gly) (CCC) to U. The mutated tRNA was tested for its ability to read glycine codons in an in vitro protein-synthesizing system programmed with the phage message MS2-RNA that had been modified by site-directed mutagenesis so as to make it possible to monitor conveniently the reading of all four glycine codons. The results showed that while the efficiency of tRNA(1Gly) (UCC) was comparable to that of mycoplasma tRNA(Gly) (UCC) in the reading of the codon GGA, the mycoplasma tRNA(Gly) was far more efficient than the tRNA(1Gly) (UCC) in the reading of the codons GGU and GGC. Thus, the anticodon UCC, when present in the structural context of the tRNA(1Gly) molecule, behaved as predicted by the wobble rules while in the structural context of the mycoplasma tRNA(Gly) it read without discrimination between the nucleotides in the third codon position, in violation of the wobble restrictions. The result with the codon GGG showed that the anticodon UCC, when present in tRNA(1Gly), was considerably less efficient in reading this codon than it was in the structural context of the mycoplasma tRNA(Gly). It would therefore seem that the anticodon UCC, when present in a certain tRNA, can be an efficient wobbler, while in the molecular environment of another tRNA it is markedly restricted in its ability to wobble.

48. Apparent lack of discrimination in the reading of certain codons in Mycoplasma mycoides.

Abstract

We report a cluster of four tRNA genes from Mycoplasma mycoides as well as the sequence of the alanine, proline, and valine tRNAs and the serine tRNA reading the UCN codons (where N stands for G, A, C, or U). This brings the total number of tRNA genes that we have so far characterized in this organism to 14, 6 of which code for tRNAs that read the codons of family boxes. In each of these latter cases, we found only one gene per family box, and the gene sequence contains a thymidine in the position corresponding to the wobble nucleotide, with the exception of the arginine tRNA gene that has an adenosine in this position. Furthermore, all of the tRNA structures reported here have an unsubstituted uridine in the wobble position. These findings are similar to those reported for mitochondria, especially yeast mitochondria, that contain an arginine tRNA with the anticodon ACG. However, the resemblance is not complete since we have demonstrated the presence of two isoacceptor tRNAs for threonine having uridine and adenosine, respectively, in the wobble position. It is suggested that in the M. mycoides at least some of the family codon boxes are read by only one tRNA each, using an unconventional method without discrimination between the nucleotides in the third codon position.

Abstract

The strength of binding between the Helicobacter pyloriblood group antigen-binding adhesin (BabA) and its cognate glycan receptor, the Lewis b blood group antigen (Leb), was measured by means of atomic force microscopy. High-resolution measurements of rupture forces between single receptor–ligand pairs were performed between the purified BabA and immobilized Lebstructures on self-assembled monolayers. Dynamic force spectroscopy revealed two similar but statistically different bond populations. These findings suggest that the BabA may form different adhesive attachments to the gastric mucosa in ways that enhance the efficiency and stability of bacterial adhesion.

50. Antimicrobial Activity of Two Different Types of Silver Nanoparticles against Wide Range of Pathogenic Bacteria.

Abstract

The emergence of antibiotic-resistant bacteria, particularly the most hazardous pathogens, namely Enterococcus faecium , Staphylococcus aureus , Klebsiella pneumoniae , Acinetobacter baumannii , Pseudomonas aeruginosa , and Enterobacter spp. (ESKAPE)-pathogens pose a significant threat to global health. Current antimicrobial therapies, including those targeting biofilms, have shown limited effectiveness against these superbugs. Nanoparticles, specifically silver nanoparticles (AgNPs), have emerged as a promising alternative for combating bacterial infections. In this study, two types of AgNPs with different physic-chemical properties were evaluated for their antimicrobial and antibiofilm activities against clinical ESKAPE strains. Two types of silver nanoparticles were assessed: spherical silver nanoparticles (AgNPs-1) and cubic-shaped silver nanoparticles (AgNPs-2). AgNPs-2, characterized by a cubic shape and higher surface-area-to-volume ratio, exhibited superior antimicrobial activity compared to spherical AgNPs-1. Both types of AgNPs demonstrated the ability to inhibit biofilm formation and disrupt established biofilms, leading to membrane damage and reduced viability of the bacteria. These findings highlight the potential of AgNPs as effective antibacterial agents against ESKAPE pathogens, emphasizing the importance of nanoparticle characteristics in determining their antimicrobial properties. Further research is warranted to explore the underlying mechanisms and optimize nanoparticle-based therapies for the management of infections caused by antibiotic-resistant bacteria.
