Abstract

Together with environmental factors, physiological maturity at birth is a major determinant for neonatal survival and postnatal development in mammalian species. Maturity at birth is the outcome of complex mechanisms of intra-uterine development and maturation during the end of gestation. In pig production, piglet preweaning mortality averages 20% of the litter and thus, maturity is a major welfare and economic concern. Here, we used both targeted and untargeted metabolomic approaches to provide a deeper understanding of the maturity in a model of lines of pigs divergently selected on residual feed intake (RFI), previously shown to have contrasted signs of maturity at birth. Analyses were conducted on plasma metabolome of piglets at birth and integrated with other phenotypic characteristics associated to maturity. We confirmed proline and myo-inositol, previously described for their association with delayed growth, as potential markers of maturity. Urea cycle and energy metabolism were found more regulated in piglets from high and low RFI lines, respectively, suggesting a better thermoregulation ability for the low RFI (with higher feed efficiency) piglets. Abstract

Abstract The aim of this paper was to describe, and when possible compare, the multivariate methods used by the participants in the EADGENE WP1.4 workshop. The first approach was for class discovery and class prediction using evidence from the data at hand. Several teams used hierarchical clustering (HC) or principal component analysis (PCA) to identify groups of differentially expressed genes with a similar expression pattern over time points and infective agent (E. coli or S. aureus). The main result from these analyses was that HC and PCA were able to separate tissue samples taken at 24 h following E. coli infection from the other samples. The second approach identified groups of differentially co-expressed genes, by identifying clusters of genes highly correlated when animals were infected with E. coli but not correlated more than expected by chance when the infective pathogen was S. aureus. The third approach looked at differential expression of predefined gene sets. Gene sets were defined based on information retrieved from biological databases such as Gene Ontology. Based on these annotation sources the teams used either the GlobalTest or the Fisher exact test to identify differentially expressed gene sets. The main result from these analyses was that gene sets involved in immune defence responses were differentially expressed.

Abstract

Abstract A large variety of methods has been proposed in the literature for microarray data analysis. The aim of this paper was to present techniques used by the EADGENE (European Animal Disease Genomics Network of Excellence) WP1.4 participants for data quality control, normalisation and statistical methods for the detection of differentially expressed genes in order to provide some more general data analysis guidelines. All the workshop participants were given a real data set obtained in an EADGENE funded microarray study looking at the gene expression changes following artificial infection with two different mastitis causing bacteria: Escherichia coli and Staphylococcus aureus. It was reassuring to see that most of the teams found the same main biological results. In fact, most of the differentially expressed genes were found for infection by E. coli between uninfected and 24 h challenged udder quarters. Very little transcriptional variation was observed for the bacteria S. aureus. Lists of differentially expressed genes found by the different research teams were, however, quite dependent on the method used, especially concerning the data quality control step. These analyses also emphasised a biological problem of cross-talk between infected and uninfected quarters which will have to be dealt with for further microarray studies.

Abstract

International audience; Foie gras is a traditional dish in France that contains 50 to 60% of lipids. The high-fat content of the liver improves the organoleptic qualities of foie gras and reduces its technological yield at cooking (TY). As the valorization of the liver as foie gras products is strongly influenced by the TY, classifying the foie gras in their potential technological quality before cooking them is the main challenge for producers. Therefore, the current study aimed to identify hepatic biomarkers of foie gras qualities like liver weight (LW) and TY. A group of 120 male mule ducks was reared and overfed for 6-12 days, and their livers were sampled and analyzed by proton nuclear magnetic resonance (1 H-NMR). Eighteen biomarkers of foie gras qualities were identified, nine for LW and TY, five specific to LW, and four specific to TY. All biomarkers were strongly negatively correlated to the liver weights and positively correlated to the technological yield, except for the lactate and the threonine, and also for the creatine that was negatively correlated to foie gras technological quality. As a result, in heavy livers, the liver metabolism was oriented through a reduction of carbohydrate and amino acid metabolisms, and the plasma membrane could be damaged, which may explain the low technological yield of these livers. The detected biomarkers have been strongly discussed with the metabolism of the liver in nonalcoholic steatohepatitis.

Abstract

Early detection of arterial hypotension during cesarean delivery under spinal anesthesia is important. This study aims to compare the validity of NexfinTM as beat-to-beat noninvasive blood pressure monitoring with conventional intermittent oscillometric measurement of blood pressure during elective cesarean delivery. This open prospective observational bicentric study was performed between January 2013 and December 2015. We simultaneously recorded arterial blood pressure with both techniques in pregnant women undergoing elective cesarean delivery under spinal anesthesia. The primary outcome was a Bland–Altman analysis of systolic blood pressure measurement comparing NexfinTM and a conventional method. The secondary outcomes were the time to detect the first relevant hypotensive episode and the comparison of both devices using a four-quadrant graph. One hundred and seventy-four parturients completed the study, and 2640 pairs of systolic blood pressure measurements were analyzed. Bias was -10 mmHg with upper and lower limits of agreement of -61 and +41 mmHg. In 73.9% of the cases, the two techniques provided the same information (normotension or hypotension), but the conventional method missed 20.8% of measurements, with NexfinTM detecting 16.2% more hypotensive measurements. Abstract

Abstract cDNA clones from a pig granulosa cell cDNA library were isolated by differential hybridisation for follicle stimulating hormone (FSH) regulation in granulosa cells in a previous study. The clones that did not match any known sequence were studied for their expression in granulosa cells (treated or not by FSH) and in fresh isolated ovarian follicles mainly by comparative RT-PCR analysis. These results give functional data on genes that may be implicated in follicular growing. These ESTs have been localised on the porcine genome, using a somatic cell hybrid panel, providing new type I markers on the porcine map and information on the comparative map between humans and pigs.

Abstract

International audience; The foie gras is an emblematic product of French gastronomy composed of waterfowl fatty liver. The organoleptic qualities of this product depend on the liver characteristics such as liver weight (LW) and technological yield (TY) at cooking. One of the main issues for producers is to classify the foie gras with high or low technological quality before cooking them. Thus the study aims at identifying biomarkers of these characteristics with non-invasive biomarkers in duck. 1H-NMR (nuclear magnetic resonance of the proton) analyses were performed on plasma of male mule ducks at different time points during the overfeeding period to obtain a large range of liver characteristics so as to identify plasmatic biomarkers of foie gras. We used two methods, one based on bucket data from the 1H-NMR spectra and another one based on the fingerprints of several metabolites. PLS analyses and Linear models were performed to identify biomarkers. We identified 18 biomarkers of liver weight and 15 biomarkers of technological yield. As these two quality parameters were strongly correlated (−0.82), 13 biomarkers were common. The lactate was the most important biomarker, the other were mainly amino acids. Contrary to the amino acids, the lactate increased with the liver weight and decreased with the technological yield. We also identified 5 biomarkers specific to LW (3 carbohydrates: glucuronic acid, mannose, sorbitol and 2 amino acids: glutamic acid and methionine) that were negatively correlated to liver weight. It was of main interest to identify 2 biomarkers specific to the technological yield. Contrary to the isovaleric acid, the valine was negatively correlated to the technological yield.

Abstract

Objectives for this study were, first, to describe individual differences in risk taking among scuba divers. Differences were examined on personality dimensions and psycho-affective variables, including positive and negative affect, as well as alexithymia. In addition, the study examined contributors to two types of behavior associated with scuba diving--deliberate risk taking and controlled participation in a high-risk sport (non-risk-taking). A cross-sectional design was used, and 131 participants were assessed on extraversion-neuroticism, affectivity, and alexithymia. The broad dimensions of personality and affectivity explained risk taking among divers. Alexithymia differentially predicted two types of risktaking behavior (direct or short-term and indirect or long-term) and was associated significantly with short-term risk-taking behavior. Abstract

Abstract Background Domestic animal breeding and product quality improvement require the control of reproduction, nutrition, health and welfare in these animals. It is thus necessary to improve our knowledge of the major physiological functions and their interactions. This would be greatly enhanced by the availability of expressed gene sequences in the databases and by cDNA arrays allowing the transcriptome analysis of any function. The objective within the AGENAE French program was to initiate a high-throughput cDNA sequencing program of a 38-tissue normalised library and generate a diverse microarray for transcriptome analysis in pig species. Results We constructed a multi-tissue cDNA library, which was normalised and subtracted to reduce the redundancy of the clones. Expressed Sequence Tags were produced and 24449 high-quality sequences were released in EMBL database. The assembly of all the public ESTs (available through SIGENAE website) resulted in 40786 contigs and 54653 singletons. At least one Agenae sequence is present in 11969 contigs (12.5%) and in 9291 of the deeper-than-one-contigs (22.8%). Sequence analysis showed that both normalisation and subtraction processes were successful and that the initial tissue complexity was maintained in the final libraries. A 9K nylon cDNA microarray was produced and is available through CRB-GADIE. It will allow high sensitivity transcriptome analyses in pigs. Conclusion In the present work, a pig multi-tissue cDNA library was constructed and a 9K cDNA microarray designed. It contributes to the Expressed Sequence Tags pig data, and offers a valuable tool for transcriptome analysis.

Abstract

The notion of risk-taking implies a cognitive process that determines the level of risk involved in a particular activity or task. This risk appraisal process gives rise to emotional responses, including anxious arousal and changes in mood, which may play a significant role in risk-related decision making. This study examines how emotional responses to the perceived risk of a scuba-diving injury contribute to divers' behavior, as well as the ways that risk taking or non-risk taking behavior, in turn, affects emotional states. The study sample consisted of 131 divers (risk takers and non-risk takers), who either had or had not been in a previous diving accident. Divers' emotional states were assessed immediately prior to diving, as well as immediately following a dive. Results indicated presence of subjective emotional experiences that are specific to whether a risk has been perceived and whether a risk has been taken. Important differences in emotion regulation were also found between divers who typically take risks and those who do not. [ABSTRACT FROM AUTHOR]

Abstract

National audience; La sélection pour plus de prolificité et plus de croissance maigre chez le porc s’est accompagnée d’une augmentation de la mortalité néonatale des porcelets. Cette mortalité est une source importante de perte économique pour la production porcine européenne et pose un problème éthique relatif au bien-être animal. Les facteurs qui contrôlent la survie des porcelets sont nombreux et dépendent de caractères inhérents à la mère et au porcelet. Le poids de naissance est généralement proposé en tant que bon indicateur de survie des porcelets (Leenhouwers et al. 2001) . Ce poids reflète l’ensemble du développement fœtal et des processus spécifiques de maturation qui se mettent en place en fin de gestation (van der Lende et al. 2001). Les porcelets sélectionnés pour le taux de survie sont aussi plus mature à la naissance et mieux préparés à la vie extra utérine (Leenhouwers et al. 2001; Leenhouwers et al. 2002). Le poids et la maturité sont donc des bons marqueurs de survie. Cependant il n’existe pas de lien évident entre le poids à la naissance et le taux de survie entre les races puisque les porcelets Meishan (MS) sont plus petits à la naissance mais moins sujets à la mortalité que les porcelets de race Large White (LW) (Canario et al. 2014). Les porcelets MS sont physiologiquement plus matures que les porcelets LW. La maturité est définie ici comme l’état de plein développement permettant la survie à la naissance. Les processus d’acquisition de cette maturité au cours du dernier mois de gestation, et leurs éventuelles variations en relation avec la génétique et l’environnement maternel utérin, jouent donc un rôle déterminant dans la mortalité précoce du porcelet. La survie des porcelets est, par exemple, corrélée positivement avec l’efficacité placentaire (Leenhouwers et al. 2002). Dans ce contexte, la régulation par le placenta de l’allocation des nutriments de la mère vers le jeune impacte grandement la croissance fœtale (et en conséquence la maturité), la santé et les performances des animaux. Le placenta est aussi le siège d’un conflit entre le génome maternel et paternel. Un dialogue utéro-placentaire s’instaure donc, faisant intervenir des mécanismes épigénétiques encore mal connus. L’objectif de du projet PENELOPE est d’évaluer, à partir d’une sélection de gènes (40 gènes) connus dans d’autres espèces ou d’expression préférentielle nouvelle, le statut de l’empreinte chez le porc en fin de gestation. Nous disposons pour cela des 2 tissus adjacents le placenta et l’endomètre pour 4 génotypes fœtaux (LW (lignée à mortalité néonatale importante), MS (peu de mortalité), et les croisés réciproques) issus du projet PORCINET. Un séquençage au débit des ADN et ADNc est en cours pour identifier les SN! P informatifs puis évaluer le statut mono allèlique et mono parental de l’expression de ces gènes. Ces premiers résultats permettront d’évaluer plasticité de ces gènes dans le placenta et l’endomètre.
