Abstract

© 2019 International Medical Informatics Association (IMIA) and IOS Press. Pre-eclampsia is a pregnancy condition affecting 5-10% of pregnancies, and it is the leading cause of death in pregnancy associated with increased risk of cardiovascular disease later in life. Despite research, the pathogenesis of pre-eclampsia is still poorly understood. In this paper, we investigate the overlapping pathogenic mechanisms between pre-eclampsia and adult hypertension using an integrative biomedical informatics strategy that combined text mining techniques to identify genes and proteins, with geneset analyses, generating knowledge on the pathways and mechanisms involved in these conditions. We identified several overlapping pathogenic pathways/systems including metabolic pathways, developmental biology pathways, immune system, haemostasis, tyrosine kinase pathways, extracellular matrix and oxidative stress pathways. This bioinformatics approach could be applied for investigating mechanistic pathways of other disorders.

Abstract

An aerodynamic analysis system based on potential theory at subsonic and/or supersonic speeds and impact type finite element solutions at hypersonic conditions is described. Three dimensional configurations having multiple nonplanar surfaces of arbitrary planform and bodies of noncircular contour may be analyzed. Static, rotary, and control longitudinal and lateral directional characteristics may be generated. The analysis was implemented on a time sharing system in conjunction with an input tablet digitizer and an interactive graphics input/output display and editing terminal to maximize its responsiveness to the preliminary analysis problem. The program provides an efficient analysis for systematically performing various aerodynamic configuration tradeoff and evaluation studies.

Abstract

In order for the Department of Safeguards to provide credible assurance that states are honoring their safeguards obligations, quantitative conclusions regarding states' nuclear material inventories and activities are needed. The statistical analysis used to reach these conclusions requires that each measurement method undergo uncertainty quantification (UQ). This paper describes current work aimed at improved understanding of uncertainty sources and provides two example UQ applications, one using a top-down approach and the other using a bottom-up approach. We explore possible improvement of current top-down analysis, through a new calculation that accommodates multiplicative error models to compare operator and inspector data. To demonstrate the benefit of bottom-up analysis, we examine individual uncertainty components associated with uranium neutron coincidence collar (UNCL) measurements. Improving safeguards effectiveness is an on-going challenge and incorporating predicted uncertainties from bottom-up analyses into top-down verification measurement performance evaluations is a useful path forward to comprehensive UQ. Abstract

Advancements have occurred in transonic numerical simulation that place aerodynamic performance design into a relatively well developed status. Efficient broad band operating characteristics can be reliably developed at the conceptual design level. Recent aeroelastic and separated flow simulation results indicate that systematic consideration of an increased range of design problems appears promising. This emerging capability addresses static and dynamic structural/aerodynamic coupling and nonlinearities associated with viscous dominated flows.

Abstract

A subsonic/supersonic/hypersonic aerodynamic analysis was developed by integrating the Aerodynamic Preliminary Analysis System (APAS), and the inviscid force calculation modules of the Hypersonic Arbitrary Body Program. APAS analysis was extended for nonlinear vortex forces using a generalization of the Polhamus analogy. The interactive system provides appropriate aerodynamic models for a single input geometry data base and has a run/output format similar to a wind tunnel test program. The user's manual was organized to cover the principle system activities of a typical application, geometric input/editing, aerodynamic evaluation, and post analysis review/display. Sample sessions are included to illustrate the specific task involved and are followed by a comprehensive command/subcommand dictionary used to operate the system.

Computerization of aerodynamic theory has progressed to state where analysis of complete aircraft configurations can be performed in single program. Aerodynamic Preliminary Analysis System, APAS, is comprehensive aerodynamic analysis system, based on linearized potential theory. Three-dimensional configurations (with or without jet flaps) having multiple nonplanar surfaces of arbitrary planform and open or closed slender bodies of noncircular contour may be analyzed with APAS. As preliminary design aid, APAS allows designer to survey systematically large number of alternative configurations and component geometries economically.

Abstract

The quantitive relationship of cruise drag and nacelle shape was investigated for a representative advanced supersonic transport configuration. Nacelle shape parameters were systematically varied, and the effects of these variations on wave and friction drag were determined. The effects of changes in vehicle drag, propulsion weight, and specific fuel consumption on vehicle takeoff gross weight were computed. Generally, it was found that nacelle shapes such that the maximum cross-sectional area occurred at or near the nozzle exit resulted in the lowest wave drag. In fact, nacelle shapes were found that produce favorable interference effects (drag reduction) of such magnitude as to nearly offset the friction drag of the nacelle.

Abstract

The effects of various propulsion system parameters on the characteristics of a supersonic transport were investigated. The effects of arbitrarily scaling engine size on wave drag, friction drag, drag-due-to-lift, wing sizing, airplane balance, and airplane weight were studied. These evaluations were made for two families of nacelle shapes, resulting from typical turbojet and turbofan installations. Also examined were effects of nacelle location, and the wing camber plane deformations required to cancel the nacelle interference pressure field at cruise Mach number (2.7 M) were determined. The most drag-sensitive parameter is found to be nacelle shape. Similarly, wing deformation requirements are found to be primarily affected by nacelle shape. Effects of engine size variations are noted primarily in airplane gross weight.

Abstract

Increases in supersonic cruise lift drag ratio were sought at Mach numbers 2.2 and 2.7 using wing body planform and thickness blending. Constrained twist and camber optimization was performed in the presence of nacelles. Wing and fuselage thickness distributions were optimized for either minimum volume wave drag or minimum total pressure wave drag. The zero leading edge suction lift drag ratios were determined for three wing planforms. The magnitude of the effect of leading edge suction on attainable lift drag ratio was defined on one planform and estimation of available leading edge suction was made.

A comprehensive aerodynamic analysis program based on linearized potential theory is described. The solution treats thickness and attitude problems at subsonic and supersonic speeds. Three dimensional configurations with or without jet flaps having multiple non-planar surfaces of arbitrary planform and open or closed slender bodies of non-circular contour may be analyzed. Longitudinal and lateral-directional static and rotary derivative solutions may be generated. The analysis was implemented on a time sharing system in conjunction with an input tablet digitizer and an interactive graphics input/output display and editing terminal to maximize its responsiveness to the preliminary analysis problem. Nominal case computation time of 45 CPU seconds on the CDC 175 for a 200 panel simulation indicates the program provides an efficient analysis for systematically performing various aerodynamic configuration tradeoff and evaluation studies.

46. Purification and characterization of Bacillus subtilis PyrR, a bifunctional pyr mRNA-binding attenuation protein/uracil phosphoribosyltransferase.

Abstract

Bacillus subtilis PyrR has been shown to mediate transcriptional attenuation at three separate sites within the pyrimidine nucleotide biosynthetic (pyr) operon. Molecular genetic evidence suggests that regulation is achieved by PyrR binding to pyr mRNA. PyrR is also a uracil phosphoribosyltransferase (UPRTase). Recombinant PyrR was expressed in Escherichia coli, purified to homogeneity, physically and chemically characterized, and examined with respect to both of these activities. Mass spectroscopic characterization of PyrR demonstrated a monomeric mass of 20,263 Da. Gel filtration chromatography showed the native mass of PyrR to be dependent on protein concentration and suggested a rapid equilibrium between dimeric and hexameric forms. The UPRTase activity of PyrR has a pH optimum of 8.2. The Km value for uracil is very pH-dependent; the Km for uracil at pH 7.7 is 990 +/- 114 muM, which is much higher than for most UPRTases and may account for the low physiological activity of PyrR as a UPRTase. Using an electrophoretic mobility shift assay, PyrR was shown to bind pyr RNA that includes sequences from its predicted binding site in the second attenuator region. Binding of PyrR to pyr RNA was specific and UMP-dependent with apparent Kd values of 10 and 220 nM in the presence and absence of UMP, respectively. The concentration of UMP required for half-maximal stimulation of binding of PyrR to RNA was 6 muM. The results support a model for the regulation of pyr transcription whereby termination is governed by the UMP-dependent binding of PyrR to pyr RNA and provide purified and characterized PyrR for detailed biochemical studies of RNA binding and transcriptional attenuation.

Abstract

A tactical supercritical flow research model was designed to achieve high aerodynamic efficiency at Mach number 0.90 and lift coefficient 0.50 using numerical transonic design procedures. A wind tunnel model was built and tested in the Rockwell International Trisonic Wind Tunnel at Mach numbers of 0.40, 0. 85, 0.90, 0.95. Lift-drag ratio at the design condition compared favorably with theoretical upper bound levels for the test arrangement and conditions. Six component force measurements for the joined wing are well behaved and near linear over an angle-of-attack range of -4< alpha <10 degrees. Stable stall is naturally achieved for the arrangement.
