The influence of radical-disulfide exchange on the polymerization and relaxation dynamics of thiol–ene–disulfide networks is investigated.

Microparticle-mediated nucleic acid delivery is a popular strategy to achieve therapeutic outcomes via antisense gene therapy. However, current methods used to fabricate polymeric microparticles suffer from suboptimal properties such as particle polydispersity and low encapsulation efficiency. Here, a new particulate delivery system based on step-growth thiol-Michael dispersion polymerization, is reported in which a low polydispersity microparticle is functionalized with a synthetic nucleic acid mimic, namely click nucleic acids (CNA). CNA oligomers, exhibiting an average length of approximately four nucleic acid repeat units per chain for both adenine and thymine bases, were successfully conjugated to excess thiols present in the microparticles. Effective DNA loading was obtained by simple mixing; and, up to 6 ± 2 pmol of complementary DNA/mg of particle was achieved, depending on the length of DNA used. In addition, DNA loading was orders of magnitude less for noncomplementary sequences and sequences containing an alternating base mismatch. The DNA release properties were evaluated, and it was found that release could be triggered by sudden changes in temperature but was unaffected over a range of pH. Finally, phagocytosis of loaded microparticles was observed by confocal microscopy and corroborated by an increase in cellular metabolic activity up to 90%. Overall, this work suggests that CNA functionalized microparticles could be a promising platform for controlled DNA delivery.

The effect of thiol substitution in radical thiol–ene reactions has been studied by using model, monofunctional thiols as well as multifunctional thiol monomers along with the assessment of their subsequent polymerization reactions and polymer mechanical behavior. FT-IR was used to monitor the polymerization rate and quantify the overall conversion. While the total conversion was observed to range from 70% to 100%, the polymerization rate was found to decrease by as much as 10-fold as the thiol substitution was changed from primary to tertiary. Analogous multi-thiol monomers of similar structure but varying substitution were synthesized to observe the effect of substitution type on polymerization kinetics and polymer behavior. Methylation at the α-carbon was varied from primary to tertiary to observe these differences. Mechanical properties were assessed by using dynamic mechanical analysis and water sorption experiments, where the glass transition temperatures were found to be within 1–2 °C as thiol substitution varied. Furthermore, primary thiol films absorbed 1–3% more water than secondary thiol films. Resin shelf stability experiments were performed by using rheometry to measure storage time-dependent viscosity changes, and it was found that secondary thiol films remained relatively stable for up to 100 times longer than their primary counterparts. It was concluded that while there are differences under relatively slow initiation conditions, at typical initiation rates all three thiol substitutions may be made to react at similar rates for both monofunctional and polymeric systems.

Background Neuroimaging studies suggest that the inferior frontal operculum (IFO) is part of a neuronal network involved in facial expression processing, but the causal role of this region in emotional face discrimination remains elusive. Objective We used cathodal (inhibitory) tDCS to test whether right (r-IFO) and left (l-IFO) IFO play a role in discriminating basic facial emotions in healthy volunteers. Specifically, we tested if the two sites are selectively involved in the processing of facial expressions conveying high or low arousal emotions. Based on the Arousal Hypothesis we expected to find a modulation of high and low arousal emotions by cathodal tDCS of the r-IFO and the l-IFO, respectively. Methods First, we validated an Emotional Faces Discrimination Task (EFDT). Then, we targeted the r-IFO and the l-IFO with cathodal tDCS (i.e. the cathode was placed over the right or left IFO, while the anode was placed over the contralateral supraorbital area) during facial emotions discrimination on the EFDT. Non-active (i.e. sham) tDCS was a control condition. Results Overall, participants manifested the “happy face advantage”. Interestingly, tDCS to r-IFO enhanced discrimination of faces expressing anger (a high arousal emotion), whereas, tDCS to l-IFO decreased discrimination of faces expressing sadness (a low arousal emotion). Conclusions Our findings revealed a differential causal role of r-IFO and l-IFO in the discrimination of specific high and low arousal emotions. Crucially, these results suggest that cathodal tDCS might reduce the neural noise triggered by facial emotions, improving discrimination of high arousal emotions but disrupting discrimination of low arousal emotions. These findings offer new insights for treating clinical population with deficits in processing facial expressions.

Reversibly programmable liquid crystal elastomer microparticles (LCEMPs), formed as a covalent adaptable network (CAN), with an average diameter of 7 μm ± 2 μm, were synthesized via a thiol-Michael dispersion polymerization. The particles were programmed to a prolate shape via a photoinitiated addition-fragmentation chain-transfer (AFT) exchange reaction by activating the AFT after undergoing compression. Due to the thermotropic nature of the AFT-LCEMPs, shape switching was driven by heating the particles above their nematic-isotropic phase transition temperature (TNI). The programmed particles subsequently displayed cyclable two-way shape switching from prolate to spherical when at low or high temperatures, respectively. Furthermore, the shape programming is reversible, and a second programming step was done to erase the prolate shape by initiating AFT at high temperature while the particles were in their spherical shape. Upon cooling, the particles remained spherical until additional programming steps were taken. Particles were also programmed to maintain a permanent oblate shape. Additionally, the particle surface was programmed with a diffraction grating, demonstrating programmable complex surface topography via AFT activation.

Of importance for adhesive materials, particularly pressure-sensitive adhesive (PSA) systems, is the ability to increase bulk toughness without reduction of adhesion. Previous approaches for increasing PSA durability sacrifice permanent cross-linking or adhesive potential, limiting performance. In this work, covalent adaptable networks (CANs) derived from thiol-thioester exchange (TTE) are utilized as a basis for adhesive films. Tensile and single-lap shear tests were conducted for adhesive materials containing no filler, 15 wt % nanoparticles functionalized with thioester-containing acrylate, or 15 wt % nanoparticles functionalized with nonthioester-containing acrylate. Additionally, fatigue experiments were conducted on unfilled adhesives. Results indicate that TTE improves toughness, adhesion, and fatigue in unfilled materials. Filled adhesives with activated TTE showed a nearly fourfold increase in adhesion with slightly reduced toughness compared to uncatalyzed filled specimens. This work has implications in many industries, from biomedical to automotive, as toughness and fatigue resistance are important considerations for adhesive applications.

Covalent adaptable networks (CANs), unlike typical thermosets or other covalently crosslinked networks, possess a unique, often dormant ability to activate one or more forms of stimuli-responsive, dynamic covalent chemistries as a means to transition their behavior from that of a viscoelastic solid to a material with fluid-like plastic flow. Upon application of a stimulus, such as light or other irradiation, temperature, or even a distinct chemical signal, the CAN responds by transforming to a state of temporal plasticity through activation of either reversible addition or reversible bond exchange, either of which allows the material to essentially re-equilibrate to an altered set of conditions that are distinct from those in which the original covalently crosslinked network is formed, often simultaneously enabling a new and distinct shape, function, and characteristics. As such, CANs span the divide between thermosets and thermoplastics, thus offering unprecedented possibilities for innovation in polymer and materials science. Without attempting to comprehensively review the literature, recent developments in CANs are discussed here with an emphasis on the most effective dynamic chemistries that render these materials to be stimuli responsive, enabling features that make CANs more broadly applicable.

The number of lymphocytes in the blood is constant, pointing to an effective control of circulating lymphocyte values. The mechanisms of this regulation are uncertain, although it is likely that the number of blood lymphocytes is conditioned by hormones, homing factors and cytokines whose production is at least partly restrained by genetic factors. Particularly genetic factors linked to major histocompatibility complex (MHC) appear to be involved. In human beings a decreased number of blood lymphocytes has been described in healthy subjects carrying the Human Leucocyte Antigens (HLA) haplotype HLA-B8,DR3. In the present study, to inquire into the mechanisms of this lymphocyte decreased number, we have performed an analysis of blood subset values in these subjects. When the absolute values of lymphocytes were analysed according to HLA phenotype, HLA-B8,DR3 positive subjects (N = 26) displayed significantly lower values as compared to HLA-B8,DR3 negative ones (N = 282). The analysis of lymphocyte subpopulations performed by flow cytometry in 72 subjects did not show significant changes in lymphocyte subset percentages between HLA-B8,DR3 positive subjects and negative ones. Thus, the decrease of circulating lymphocytes seems to be due to a reduction of cell number affecting all lymphocyte subsets rather than a single cell subpopulation. The analysis of in vitro spontaneous apoptosis performed by flow cytometry in a smaller sample of subjects showed a significant increase of spontaneous apoptosis in lymphocytes from HLA-B8,DR3 positive individuals suggesting a possible explanation for the deviation from normal lymphocyte count observed in these subjects. However it is intriguing that a decreased number of blood lymphocytes can be observed in healthy HLA-B8,DR3 positive subjects but also in autoimmune diseases linked to this haplotype like systemic lupus erythematosus and insulin-dependent diabetes. Furthermore, in our opinion, this finding is to be kept in mind in evaluating hematological parameters in healthy subjects.
