20. Microbiome variation in corals with distinct depth distribution ranges across a shallow–mesophotic gradient (15–85 m)

Abstract

Mesophotic coral ecosystems (MCEs) are generally poorly studied, and our knowledge of lower MCEs (below 60 m depth) is largely limited to visual surveys. Here, we provide a first detailed assessment of the prokaryotic community associated with scleractinian corals over a depth gradient to the lower mesophotic realm (15-85 m). Specimens of three Caribbean coral species exhibiting differences in their depth distribution ranges (Agaricia grahamae, Madracis pharensis and Stephanocoenia intersepta) were collected with a manned submersible on the island of Curaçao, and their prokaryotic communities assessed using 16S rRNA gene sequencing analysis. Corals with narrower depth distribution ranges (depth-specialists) were associated with a stable prokaryotic community, whereas corals with a broader niche range (depth-generalists) revealed a higher variability in their prokaryotic community. The observed depth effects match previously described patterns in Symbiodinium depth zonation. This highlights the contribution of structured microbial communities over depth to the coral's ability to colonize a broader depth range.

25. Learning from Multiple Cases: A New Paradigm for Investigating the Effects of Clinical Experience on Knowledge Restructuring and Knowledge Acquisition.

Abstract

The effects of experience with a series of similar cases on the knowledge restructuring and learning from text were studied in a longitudinal design. Two groups of fourth-year medical students were confronted with a series of cases, part of them having the same underlying disease. The cases were interspersed with fillers, and each set of cases had one filler from the other set. Students were asked to diagnose the cases and to try to learn as much as possible, and they were asked to learn as much as possible from a text. Case processing time showed that students who had worked in a series of similar cases needed less time for the same case than students who did not have that experience. Results show that one series of cases, those dealing with polycystic ovaries syndrome, was more intensively processed than the other series, and that those students showed more traces of knowledge construction, although it was not clear why. Prior processing of a series of cases led to better text recall of the part of the text that concerned these specific diseases, but did not affect reading time. Results do suggest that students learned case content from processing multiple cases, and that they integrated it into their prior knowledge. It is hypothesized that the theory on construction and integration of W. Kintsch explains these outcomes. (Contains 1 table, 4 figures, and 10 references.) (SLD)

27. The Relation between the Nature of Prior Knowledge Activated and Information Processing: To Elaborate or To Infer?

Abstract

Effects of mobilizing prior knowledge on information processing were studied with 3 groups of 12 adult subjects each. The assumption that activating different kinds of prior knowledge would induce different information processing activities during subsequent text processing (inferencing or elaborating) was tested using passages about fishing policy or tourism in the European Economic Community. It was hypothesized that inferences are induced by activating prior knowledge that could have evolved into a schema through experiences in a particular domain, and that they instantiate the schema. Elaborations, on the other hand, would result from activating limited prior knowledge, represented in an associative network. Elaborations would be required during subsequent text processing to bridge gaps between conceptual nodes in the text and nodes in the associative network. Findings suggested that inferences were mostly drawn when the activated prior knowledge had evolved into a schema, but elaborations were most frequent when limited prior knowledge was activated, represented in an associative net. 28. Balance & Perspective: 25 Years of Dutch Applied Linguistics.

Abstract

Revised texts of five papers from a conference on Dutch applied linguistics are presented. "Learning a Foreign Language in a Natural Acquisition Context Without Instruction" (Wolfgang Klein) advocates observing and analyzing language learning in progress in natural language learning contexts and presents data from a European project on second language acquisition by adult immigrants. "Linguistics and Second Language Acquisition" (Peter Jordens) examines the relationships between linguistics and second language learning. The discussion touches on contrastive analysis as an explanatory model, then looks at the interlanguage model and the possible relevance of markedness theory, and gives attention to universal grammar. In "Ethnic Minorities, Language Diversity, and Educational Implications: A Case Study on the Netherlands" (Guus Extra), cross-national trends in Western Europe, especially ethnic minorities in the Netherlands are noted and the consequences of language diversity in elementary, secondary, and adult education are examined. "Three Decades of Foreign Language Teaching in the Netherlands" (Arthur van Essen) reviews developments in Dutch foreign language teaching in the past few decades, drawing from experience and existing literature. "Policy-Making in Foreign Language Teaching" (Theo van Els) discusses the role applied linguists may play in policy formation in foreign language instruction, based on recent Dutch educational history. Abstract

Amino acid‐based surfactants are valuable compounds for cosmetic formulations. The chemical synthesis of acyl amino acids is conventionally performed by the Schotten–Baumann reaction using fatty acyl chlorides, but aminoacylases have also been investigated for use in biocatalytic synthesis with free fatty acids. Aminoacylases and their properties are diverse; they belong to different peptidase families and show differences in substrate specificity and biocatalytic potential. Bacterial aminoacylases capable of synthesis have been isolated from Burkholderia, Mycolicibacterium, and Streptomyces. Although several proteases and peptidases from S. griseus have been described, no aminoacylases from this species have been identified yet. In this study, we investigated two novel enzymes produced by S. griseus DSM 40236T. We identified and cloned the respective genes and recombinantly expressed an α‐aminoacylase (EC3.5.1.14), designated SgAA, and an ε‐lysine acylase (EC3.5.1.17), designated SgELA, in S. lividans TK23. The purified aminoacylase SgAA was biochemically characterized, focusing on its hydrolytic activity to determine temperature‐ and pH optima and stabilities. The aminoacylase could hydrolyze various acetyl amino acids at the Nα‐position with a broad specificity regarding the sidechain. Substrates with longer acyl chains, like lauroyl amino acids, were hydrolyzed to a lesser extent. Purified aminoacylase SgELA specific for the hydrolysis of Nε‐acetyl‐l‐lysine was unstable and lost its enzymatic activity upon storage for a longer period but could initially be characterized. The pH optimum of SgELA was pH 8.0. While synthesis of acyl amino acids was not observed with SgELA, SgAA catalyzed the synthesis of lauroyl‐methionine. [ABSTRACT FROM AUTHOR]

Abstract

Subtilisins from microbial sources, especially from the Bacillaceae family, are of particular interest for biotechnological applications and serve the currently growing enzyme market as efficient and novel biocatalysts. Biotechnological applications include use in detergents, cosmetics, leather processing, wastewater treatment and pharmaceuticals. To identify a possible candidate for the enzyme market, here we cloned the gene of the subtilisin SPFA from Fictibacillus arsenicus DSM 15822T (obtained through a data mining‐based search) and expressed it in Bacillus subtilis DB104. After production and purification, the protease showed a molecular mass of 27.57 kDa and a pI of 5.8. SPFA displayed hydrolytic activity at a temperature optimum of 80 °C and a very broad pH optimum between 8.5 and 11.5, with high activity up to pH 12.5. SPFA displayed no NaCl dependence but a high NaCl tolerance, with decreasing activity up to concentrations of 5 m NaCl. The stability enhanced with increasing NaCl concentration. Based on its substrate preference for 10 synthetic peptide 4‐nitroanilide substrates with three or four amino acids and its phylogenetic classification, SPFA can be assigned to the subgroup of true subtilisins. Moreover, SPFA exhibited high tolerance to 5% (w/v) SDS and 5% H2O2 (v/v). The biochemical properties of SPFA, especially its tolerance of remarkably high pH, SDS and H2O2, suggest it has potential for biotechnological applications. [ABSTRACT FROM AUTHOR]

46. Native-Like Attainment of Dummy Subjects in Dutch and the Role of the L1

Abstract

In this study, we test the prediction, derived from the Critical Period Hypothesis, that a native level in L2 grammar cannot be attained by learners who start acquiring a second language after childhood. We selected 43 very advanced late learners of Dutch (native speakers of German, French and Turkish) and compared their performance on a grammar test with that of 44 native speakers of Dutch. The test consisted of an elicited imitation task and a sentence preference task. In these tasks, participants were tested on their knowledge of dummy subject constructions. These construction types are known to be very hard to acquire for L2-learners of Dutch and are hardly covered in Dutch grammars or L2 Dutch textbooks. The results show that it is possible to attain a native level of proficiency for learners who start acquiring an L2 after puberty, even for learners with a typologically distant L1.

Two picture-word interference experiments investigated whether or not words from a first and more dominant language are activates during lexical access in a foreign and less dominant language. Native speakers of Dutch were instructed to name pictures in English. Revealed that the Dutch name of a picture is activated during initial stages of the process of lexical access to English.
