1. Treatment of hemophilia B: focus on recombinant factor IX

In hemophilia B patients the severity of the bleeding phenotype is related to the degree of the FIX defect. Hemophilia B treatment has improved greatly in the last 20 years with the introduction first of plasma-derived and then of recombinant FIX concentrates. Replacement therapy may be administered through on-demand or prophylaxis regimens, but the latter treatment modality has been shown to be superior in prevention of hemophilic arthropathy and in improvement of patients' quality of life. The purpose of this narrative review is to summarize the current knowledge on treatment strategies for hemophilia B, focusing on recombinant FIX products either clinically used or in development. There is only one rFIX product that is licensed to treat hemophilia B patients; from the analysis of the literature data presented in this review, the authors conclude that this rFIX product has demonstrated an excellent safety profile and excellent clinical efficacy for halting and preventing bleeds in hemophilia B patients. While prophylaxis has emerged as the best therapeutic strategy for such patients because of its ability to prevent hemophilic arthropathy and to improve patients' quality of life, the pharmacokinetically tailored dosing of rFIX is another key point when planning hemophilia B treatment, as it allows optimization of the factor concentrate usage. Further clinical studies are needed to better assess the safety and efficacy (ie, the incidence of adverse reactions and inhibitor development) of newer rFIX products.Keywords: recombinant FIX products, plasma-derived FIX concentrate, bleeding, blood clotting disorder, on-demand treatment, prophylaxis treatment

3. The census of nuclear activity of late-type galaxies in the Virgo cluster

The first spectroscopic census of AGNs associated to late-type galaxies in the Virgo cluster is carried on by observing 213 out of a complete set of 237 galaxies more massive than M_dyn>10^{8.5} solar masses. Among them, 77 are classified as AGNs (including 21 transition objects, 47 LINERs and 9 Seyferts), and comprize 32% of the late-type galaxies in Virgo. Due to spectroscopic incompleteness at most 21 AGNs are missed in the survey, so that the fraction would increase up to 41%. Using corollary Near-IR observations, that enable us to estimate galaxies dynamical masses, it is found that AGNs are hosted exclusively in massive galaxies, i.e. M_dyn\gsim 10^{10} solar masses. Their frequency increases steeply with the dynamical mass from zero at M_dyn\approx10^{9.5} solar masses to virtually 1 at M_dyn>10^{11.5} solar masses. These frequencies are consistent with the ones of low luminosity AGNs found in the general field by the SDSS. Massive galaxies that harbor AGNs commonly show conspicuous r-band star-like nuclear enhancements. Conversely they often, but not necessarily contain massive bulges. Few well known AGNs (e.g. M61, M100, NGC4535) are found in massive Sc galaxies with little or no bulge. The AGN fraction seems to be only marginally sensitive to galaxy environment. We infer the black hole masses using the known scaling relations of quiescent black holes. 4. Star formation and dust extinction in nearby star forming and starburst galaxies

Abstract

We study the star formation rate and dust extinction properties of a sample of nearby star forming galaxies as derived from Halpha and UV (2000 A) observations and we compare them to those of a sample of starburst galaxies. The dust extinction in Halpha is estimated from the Balmer decrement and the extinction in UV using the FIR to UV flux ratio or the attenuation law for starburst galaxies of Calzetti et al. The Halpha and UV emissions are strongly correlated with a very low scatter for the star forming objects and with a much higher scatter for the starburst galaxies. The Halpha to UV flux ratio is found larger by a factor ~ 2 for the starburst galaxies. We compare both samples with a purely UV selected sample of galaxies and we conclude that the mean Halpha and UV properties of nearby star forming galaxies are more representative of UV selected galaxies than starburst galaxies. We emphasize that the Halpha to UV flux ratio is strongly dependent on the dust extinction: the positive correlation found between F{Halpha}/F{UV}$ and F{FIR}/F{UV} vanishes when the Halpha and UV flux are corrected for dust extinction. The Halpha to UV flux ratios converted into star formation rate and combined with the Balmer decrement measurements are tentatively used to estimate the dust extinction in UV., Comment: accepted for publication in Astronomy & Astrophysics

5. The filament of ionized gas in the outskirt of M87

Abstract

We report on the filament of ionized gas found at the NE periphery of M87, the second brightest elliptical galaxy in the Virgo cluster. The object lies at 3.16 arcmin (15.7 kpc) projected distance of the nucleus of M87, and it coincides in position with the Eastern radio lobe of M87. Long slit spectroscopy confirmed the gaseus nature of this object, showing Hbeta, [OIII], [OI], [NII], Halpha and [SII] lines, without any underlying continuum. The redshift of the object, V = 1170+-25 km/s, agrees with the velocity of M87 itself (V = 1280 km/s), and with the bulk of the ionized material found near the center of M87. The Halpha flux of this filament corresponds to 1 % of the ionized material in M87. 6. Optical spectroscopy of galaxies in the direction of the Virgo cluster

Abstract

Optical spectroscopy of 76 galaxies, 48 of which are projected in the direction of the Virgo cluster and 28 onto the Coma-A1367 supercluster, is reported. Adding these new measurements to those found in the literature, the redshift completeness in the Virgo region becomes 92% at B_T<=16.0 and 68% at B_T<=18.0. The one of CGCG galaxies in the direction of the Coma-A1367 supercluster becomes 98%. The Virgo cluster membership estimates obtained on morphological grounds by Binggeli et al. (1985) are confirmed in all cases. However, several "possible members" classified as BCD (if in the cluster) are found instead to be giant emission-line galaxies in the background of the Virgo cluster, Comment: LaTeX, 8 pages, 5 figures, accepted for pubblication in A&A

7. DESIGNING THE METRIC SURVEY FOR BUILT HERITAGE DOCUMENTATION USING 360° IMAGES AND AN ONLINE CLOUD-BASED PLATFORM

Abstract

The documentation process of Built Heritage could be really challenging, and managing the different phases of this process is not always straightforward. The metric survey design is still one of the most complex and delicate tasks in the overall process: it drives the activities of data acquisition, processing, validation, interpretation, and final product delivery. It encompasses several aspects: stakeholders involvement, choice of instruments and techniques, available resources (not only economical but also in terms of human resources) timelines, etc. Moreover, a wrong or inaccurate metric survey design can lead to significant mistakes during the data acquisition phase that can result in the collection of redundant data or, worst-case scenario, a lack of data. After a brief state-of-the-art in the European and Italian framework, the research presented in this work will focus on the different aspects of the documentation process and, more specifically, on new digital tools that can assist this step of the Built Heritage documentation. More specifically, the contribution will focus on 360° cameras and the related cloud-based platforms for using and sharing these types of data. This market sector has been rapidly growing in the last years, and we faced a lowering of the purchase costs for these systems together with a wider availability of different sensors. Finally, the resolution has reached exciting levels with sensors that can record 360° data up to 6K/8K.

Abstract

In 2002 we published an article describing a population of vessel-associated progenitors that we termed mesoangioblasts (MABs). During the past decade evidence had accumulated that during muscle development and regeneration things may be more complex than a simple sequence of binary choices (e.g., dorsal vs. ventral somite). LacZ expressing fibroblasts could fuse with unlabelled myoblasts but not among themselves or with other cell types. Bone marrow derived, circulating progenitors were able to participate in muscle regeneration, though in very small percentage. Searching for the embryonic origin of these progenitors, we identified them as originating at least in part from the embryonic aorta and, at later stages, from the microvasculature of skeletal muscle. While continuing to investigate origin and fate of MABs, the fact that they could be expanded in vitro (also from human muscle) and cross the vessel wall, suggested a protocol for the cell therapy of muscular dystrophies. We tested this protocol in mice and dogs before proceeding to the first clinical trial on Duchenne Muscular Dystrophy patients that showed safety but minimal efficacy. In the last years, we have worked to overcome the problem of low engraftment and tried to understand their role as auxiliary myogenic progenitors during development and regeneration.

Abstract

Background The aim of this study was to investigate the impact of anti-HBc (HBcAb) positivity on the progression of liver fibrosis (Fibrosis-4 score >3.25) in the Italian cohort of HIV-infected individuals naïve to antiretroviral treatment (ICONA). Methods All patients with FIB-4 Results Patients who were HBcAb+/HCV-/HBs antigen (HBsAg)- and HCV+/HBcAb+/HBsAg- or HBsAg+/HBcAb+/HCV- had CD4+ cell counts below the nadir and significantly higher prevalence of AIDS diagnosis at baseline than the other groups (P < .0001). A Cox regression model adjusted for age, HIV transmission mode, country of birth, and alcohol consumption showed a higher relative risk (HR) of progression to FIB-4 >3.25 in HCV+/HBcAb+/HBsAg- patients (HR, 7.2; 95% CI, 3 8–13.64). Conclusions HBcAb+ contributes to liver damage in HIV+/HCV+/HBcAb+/HBsAg- subjects. A careful monitoring for signs of previous HBV infection is needed in this kind of patients.

Abstract

Background There are no Italian data regarding the strategies for preventing neonatal group B streptococcal (GBS) infection. We conducted a national survey in order to explore obstetrical, neonatal and microbiological practices for the GBS prevention. Methods Three distinct questionnaires were sent to obstetricians, neonatologists and microbiologists. Questionnaires included data on prenatal GBS screening, maternal risk factors, intrapartum antibiotic prophylaxis, microbiological information concerning specimen processing and GBS antimicrobial susceptibility. Results All respondent obstetrical units used the culture-based screening approach to identify women who should receive intrapartum antibiotic prophylaxis, and more than half of the microbiological laboratories (58%) reported using specimen processing consistent with CDC guidelines. Most neonatal units (89 out of 107, 82%) reported using protocols for preventing GBS early-onset sepsis consistent with CDC guidelines. Conclusions The screening-based strategy is largely prevalent in Italy, and most protocols for preventing GBS early-onset sepsis are consistent with CDC guidelines. However, we found discrepancies in practices among centers that may reflect the lack of Italian guidelines issued by public health organizations.

Abstract

A comparison of the abilities of rotary versus reciprocating files to eliminate viable Enterococcus faecalis populations from the long oval root canals of extracted human teeth. Fifty teeth were contaminated and randomly distributed into two groups (n = 25 each): BT-RaCe group and WaveOne group. Abstract

Background: The treatment of splanchnic vein thrombosis (SVT) is challenging, due to the increased risk of bleeding and potentially life-threatening complications. Current recommendations are based on evidence from the treatment of venous thrombosis in usual sites, but small observational studies in SVT population suggest that the bleeding risk may offset the benefit of anticoagulant treatment in this setting. The aim of this study was to evaluate the safety of vitamin K antagonists (VKAs) in SVT patients. Methods: We retrospectively included SVT patients treated with VKAs followed by 37 Italian anticoagulation clinics, until June 2013. The primary outcome was the incidence of major bleeding (MB), according to the ISTH definition, during VKA treatment. Vascular events, including both arterial and venous thrombosis, and mortality were also documented. Results: Three hundred and seventy-five patients were included (median age 53 years; 54.7% males). During a median VKA treatment duration of 1.98 years, 15 MB events occurred, corresponding to an incidence rate of 1.24 (95% confidence interval [CI], 0.75-2.06) per 100 patient-years. Gastrointestinal bleeding represented 40% of all MB events. At multivariate analysis, the presence of esophageal varices emerged as independent predictor of MB (hazard ratio 5.4; 95% CI, 1.4-21.1). The incidence rate of vascular events on treatment was 1.37 (95% CI, 0.84-2.23) per 100 patient-years and the mortality rate was 0.83 (95% CI, 0.44-1.54) per 100 patient-years. Conclusions: Selected SVT patients followed by anticoagulation clinics for the management of VKA treatment show a low rate of major bleeding and vascular events.

37. Comparative analysis of SAF, Protaper Next and BT-Race in eliminating Enterococcus faecalis from long oval canals: An ex vivo study

Abstract

Comparison of the ability of newly designed rotary files to eliminate viable Enterococcus faecalis populations from long oval root canals of extracted human teeth to that of the self-adjusting file (SAF). One hundred caries-free, single-rooted, long oval teeth were contaminated with E. faecalis. The teeth were randomly distributed into four groups (n = 25) as follows: G.1, manual; G.2, SAF; G.3, ProTaper Next; and G.4, BT-Race. Two microbial samples were obtained from each tooth with sterile paper points, (s1) before and (s2) after instrumentation. The relative reduction in colony-forming units (CFUs) from s1 to s2 measurements was calculated and compared among the groups using parametric Kruskal–Wallis one-way anova on ranks and Dunn's method (a = 0.05). The results indicated a descending order of the groups with regard to efficacy as follows: BT-Race, Next, SAF and manual. The statistical analysis showed that the relative percentage reduction (RR) of CFUs was lower in the manual group than in the other groups, while the SAF group showed a significantly lower RR than the BT-Race group (P < 0.05). The efficacy in reduction of the microbiological load of viable E. faecalis from long oval root canals was different between the tested endodontic systems. © 2016 Australian Society of Endodontology Inc

43. Implementation and effectiveness of a physical education intervention in primary school on physical fitness qualities

Abstract

Aim: The aim of this study was to evaluate the effectiveness of 20 physical education lessons taught by qualified physical education teaching assistants in improving physical fitness qualities. A second aim was to estimate the feasibility of a validated [1, 2], functional, and easily administered test battery for measuring children’s physical fitness. Methods: 40,360 children, 20,758 boys and 19,602 girls (Boys: age, 8.7 ± 1.5 years; height, 131.6 ± 10.3 cm; weight, 30.5 ± 8.4 kg; Girls: age, 8.7 ± 1.5 years; height, 130.4 ± 10.8 cm; weight, 29.6 ± 8.1 kg), from 5 to 12 years old, were involved in the study. 20 9 60 min physical education lessons, to be delivered once a week between November 2015 and May 2016, were conducted by a qualified physical education teaching assistant. Anthropometric measures (BMI z-score), cardiorespiratory fitness (6 min walk test_6MWT), musculoskeletal fitness (standing broad jump_SBJ) and motor fitness (4x10 m shuttle run test_SRT) were evaluated at baseline and after 6 months. Results: Overall, the children in each age group were able to perform all of the test items, indicating the suitability of the test battery for children as young as 5 years of age. After 6 months girls slightly decreased BMI z-score (D = -0.0034, p = 0.1054), while boys significantly increased BMI z-score (D = 0.367, p\0.0001). Mean values indicate that all parameters were significantly modified in both groups (6MWT, D = 24 m, p\0.0001; SBJ, D = 5 cm, p\0.0001; SRT, D = -0.6 s, p\0.0001) with respect to baseline data, boys achieving higher improvement in the SRT (p = 0.0141) when compared to their counterparts. Conclusions: The proposed school-based program showed effectiveness and feasibility. Our results suggest that childhood is a possible specific time window for physical fitness enhancement. Abstract

Drift-scan mode (3600-6800 Angstrom) spectra with 500 < R < 1000 resolution are presented for 333 galaxies members of nearby clusters, covering the whole Hubble sequence. The majority (225) were obtained for galaxies in the Virgo cluster where a completeness of 36%, if all Hubble types are considered, and of 51%, restricting to late-types, was reached at m(p) less than or equal to 16. Our data can be therefore considered representative of the integrated spectral properties of giant and dwarf galaxies in this cluster. Intensities and equivalent widths (EWs) are derived for the principal lines, both in emission and in absorption. Deblending of the underlying absorption from emission was achieved in most cases

Abstract

We study the star formation rate and dust extinction properties of a sample of nearby star-forming galaxies as derived from Halpha and UV ( similar to 2000 Angstrom) observations and we compare them to those of a sample of starburst galaxies. The dust extinction in Halpha is estimated from the Balmer decrement and the extinction in UV using the FIR to UV flux ratio or the attenuation law for starburst galaxies of Calzetti et al. (2000). The Halpha and UV emissions are strongly correlated with a very low scatter for the star-forming objects and with a much higher scatter for the starburst galaxies. The Halpha to UV flux ratio is found to be larger by a factor similar to 2 for the starburst galaxies. We compare both samples with a purely UV selected sample of galaxies and we conclude that the mean Halpha and UV properties of nearby star-forming galaxies are more representative of UV-selected galaxies than starburst galaxies. We emphasize that the Halpha to UV flux ratio is strongly dependent on the dust extinction: the positive correlation found between F-Halpha/F-UV and F-FIR./F-UV vanishes when the Halpha and UV flux are corrected for dust extinction. The Halpha to UV flux ratios converted into star formation rate and combined with the Balmer decrement measurements are tentatively used to estimate the dust extinction in UV.

50. The filament of ionized gas in the outskirt of M87

Abstract

We report on the filament of ionized gas found at the NE periphery of M87, the second brightest elliptical galaxy in the Virgo cluster. The object lies at 3.16 arcmin (15.7 kpc) projected distance of the nucleus of M87, and it coincides in position with the Eastern radio lobe of M87. Long slit spectroscopy confirmed the gaseous nature of this object, showing H-beta, [OIII], [OI], [NII], H-alpha and [SII] lines, without any underlying continuum. The redshift of the object, V = 1170 +/- 25 km s(-1), agrees with the velocity of M87 itself (V = 1280 km s(-1)), and with the bulk of the ionized material found near the center of M87. The H-alpha flux of this filament corresponds to 1% of the ionized material in M87. The radio-optical coincidence together with the spectral characteristics suggest that the filament is ionized by a shock
