The Coronavirus disease (COVID-19) caused by the novel SARS-CoV-2 coronavirus has spread from China and quickly transmitted to most other countries around the world. The World Health Organization announced COVID-19 as a pandemic that is spreading steadily and soon in most states. Coronavirus genomic characterization showed that it most closely resembled another bat-origin beta-coronavirus. Coronavirus has the largest genome of viruses that have RNA. Spike (S) glycoprotein is present in the virus and is responsible for virus entry into the host cell. COVID-19 can spread through the droplet, direct contact, and aerosol transmission in humans. It can remain in the environment and exists on plastic and steel for the longest time, making it a dangerous and contagious disease that can kill other individuals. The virus has an incubation time of 2 to 14 days. Confirmed cases of COVID-19 have evolved exponentially in the world. Possible preventive steps for disease control include more mask use, hand sanitization, and social distancing. There is no antiviral therapy and only symptomatic care. Many inhibitors of HIV protease and other antimalarial drugs have tested. There is currently no vaccine available for COVID-19 prevention, though others are available in clinical trials. Scientists often use spike proteins for vaccine production. Research is needed to develop a new innovative vaccine and targeted medicine, which will meet people's demands. [ABSTRACT FROM AUTHOR]

Context: About 30% of patients with active acromegaly experience paradoxically increased growth hormone (GH) secretion during the diagnostic oral glucose tolerance test (OGTT). Endogenous glucose-dependent insulinotropic polypeptide (GIP) is implicated in this paradoxical secretion., Objective: We used the GIP receptor (GIPR) antagonist GIP(3-30)NH2 to test the hypothesis that GIP mediates this paradoxical response when GIPR is abundantly expressed in somatotropinomas., Design, Patients, Setting, Interventions: 25 treatment-naïve patients with acromegaly were enrolled. Each patient underwent one OGTT during simultaneous placebo infusion and one OGTT during a GIP(3-30)NH2 infusion. Blood samples were drawn at baseline and regularly after infusions to measure GH. We assessed pituitary adenoma size by magnetic resonance imaging and GIPR expression by immunohistochemistry on resected somatotropinomas. For mechanistic confirmation, we applied in vitro and ex vivo approaches., Main Outcome Measure: The effect of GIP(3-30)NH2 on paradoxical GH secretion during OGTT as a measure of GIP involvement., Results: In four of seven patients with paradoxical GH secretion, GIP(3-30)NH2 infusion completely abolished the paradoxical response (P = 0.0003). Somatotrophs were available from three of four of these patients, all showing abundant GIPR expression. Adenoma size did not differ between patients with and without paradoxical GH secretion., Conclusions: Of 25 patients with acromegaly, seven had paradoxical GH secretion during OGTT, and pharmaceutical GIPR blockade abolished this secretion in four. Corresponding somatotroph adenomas abundantly expressed GIPR, suggesting a therapeutic target in this subpopulation of patients. Cerebral ischemic stroke is a leading cause of morbidity and mortality globally. However, the mechanisms underlying ischemic stroke injury remain poorly understood. Here, it is found that deficiency of the ubiquitin-specific protease USP25 significantly aggravate ischemic stroke injury in mice. USP25 has no impact on neuronal death under hypoxic conditions, but reduced ischemic stroke-induced neuronal loss and neurological deficits by inhibiting microglia-mediated neuroinflammation. Mechanistically, USP25 restricts the activation of NF-κB and MAPK signaling by regulating TAB2. As a deubiquitinating enzyme, USP25 removeds K63-specific polyubiquitin chains from TAB2. AAV9-mediated TAB2 knockdown ameliorates ischemic stroke injury and abolishes the effect of USP25 deletion. In both mouse and human brains, USP25 is markedly upregulated in microglia in the ischemic penumbra, implying a clinical relevance of USP25 in ischemic stroke. Collectively, USP25 is identified as a critical inhibitor of ischemic stroke injury and this data suggest USP25 may serve as a therapeutic target for ischemic stroke., (© 2023 The Authors. The technology known asPROTACs (PROteolysisTArgeting Chimeras) is a method of protein degradation. Utilising bifunctional small molecules, the ubiquitin-proteosome system (UPS) is used to induce the ubiquitination and degradation of target proteins. In addition to being novel chemical knockdown agents for biological studies that are catalytic, reversible, and rapid, PROTACs used in the treatment for disorders like cancer, immunological disorders, viral diseases, and neurological disorders. The protein degradation field has advanced quickly over the last two years, with a significant rise in research articles on the subject as well as a quick rise in smallmolecule degraders that are currently in or will soon enter the clinical stage. Other new degrading technologies, in addition to PROTAC and molecular glue technology, are also emerging rapidly.
