8. A GTPase-independent Mechanism of p21-activated Kinase Activation REGULATION BY SPHINGOSINE AND OTHER BIOLOGICALLY ACTIVE LIPIDS*

Abstract

p21-activated kinases (PAKs) are serine/threonine kinases that have been identified as targets for the small GTPases Rac and Cdc42. PAKs have been implicated in cytoskeletal regulation, stimulation of mitogen-activated protein kinase cascades, and in control of the phagocyte NADPH oxidase. Membrane targeting of PAK1 induced increased kinase activity in a GTPase-independent manner, suggesting that other mechanisms for PAK regulation exist. We observed concentration- and time-dependent activation of PAK1 by sphingosine and several related long chain sphingoid bases but not by ceramides or a variety of other lipids. Although phospholipids were generally ineffective, phosphatidic acid and phosphatidylinositol also had stimulatory effects on PAK1. Lipid stimulation induced a similar level of PAK1 activity as did stimulation by GTPases, and the patterns of PAK1 autophosphorylation determined after partial tryptic digestion and two-dimensional peptide analysis were similar with each class of activator. Lipid stimulation of PAK1 activity was dependent upon intact PAK kinase activity, as indicated by studies with a kinase-dead PAK1 mutant. Treatment of COS-7 cells expressing wild type PAK1 with sphingosine, fumonisin B, or sphingomyelinase, all of which are able to elevate the levels of free sphingosine, induced increased activity of PAK1 as determined using a p47(phox) peptide substrate. Studies using PAK1 mutants suggest that lipids act at a site overlapping or identical to the GTPase-binding domain on PAK. The inactive sphingosine derivative N,N-dimethylsphingosine was an effective inhibitor of PAK1 activation in response to either sphingosine or Cdc42. Our results demonstrate a novel GTPase-independent mechanism of PAK activation and, additionally, suggest that PAK(s) may be important mediators of the biological effects of sphingolipids.

16. GEF-H1 is involved in agonist-induced human pulmonary endothelial barrier dysfunction

Abstract

Endothelial cell (EC) permeability is precisely controlled by cytoskeletal elements [actin filaments, rnicrotubules (MT), intermediate filaments] and cell contact protein complexes (focal adhesions, adherens junctions, tight junctions). We have recently shown that the edemagenic agonist thrombin caused partial MT disassembly, which was linked to activation of small GTPase Rho, Rho-mediated actin remodeling, cell contraction, and dysfunction of lung EC barrier. GEF-H1 is an MT-associated Rhospecific guanosine nucleotide (GDP/GTP) exchange factor, which in MT-unbound state stimulates Rho activity. In this study we tested hypothesis that GEF-H1 may be a key molecule involved in Rho activation, myosin light chain phosphorylation, actin remodeling, and EC barrier dysfunction associated with partial MT disassembly. Our results show that depletion of GEF-H1 or expression of dominant negative GEF-H1 mutant significantly attenuated permeability increase, actin stress fiber formation, and increased MLC and MYPT1 phosphorylation induced by thrombin or MT-depolymerizing agent nocodazole. In contrast, expression of wild-type or activated GEF-HI mutants dramatically enhanced thrombin and nocodazole effects on stress fiber formation and cell retraction. These results show a critical role for the GEF-H1 in the Rho activation caused by MT disassembly and suggest GEF-H1 as a key molecule involved in cross talk between MT and actin cytoskeleton in agonist-induced Rho-dependent EC barrier regulation. thrombin; permeability; cytoskeleton; microtubules; pulmonary endothelium; guanine nucleotide exchange factor

17. Control of neutrophil pseudopods by fluid shear: role of Rho family GTPases

Abstract

Blood vessels and blood cells are under continuous fluid shear. Studies on vascular endothelium and smooth muscle cells have shown the importance of this mechanical stress in cell signal transduction, gene expression, vascular remodeling, and cell survival. However, in circulating leukocytes, shear-induced signal transduction has not been investigated. Here we examine in vivo and in vitro the control of pseudopods in leukocytes under the influence of fluid shear stress and the role of the Rho family small GTPases. We used a combination of HL-60 cells differentiated into neutrophils (1.4% dimethyl sulfoxide for 5 days) and fresh leukocytes from Rac knockout mice. The cells responded to shear stress (5 dyn/[cm.sup.2]) with retraction of pseudopods and reduction of their projected cell area. The Rac1 and Rac2 activities were decreased by fluid shear in a time- and magnitude-dependent manner, whereas the Cdc42 activity remained unchanged (up to 5 dyn/[cm.sup.2]). The Rho activity was transiently increased and recovered to static levels after 10 min of shear exposure (5 dyn/[cm.sup.2]). Inhibition of either Racl or Rac2 slightly but significantly diminished the fluid shear response. Transfection with Rac1-positive mutant enhanced the pseudopod formation during shear. Leukocytes from Rac1-null and Rac2-null mice had an ability to form pseudopods in response to platelet-activating factor but did not respond to fluid shear in vitro. Leukocytes in wild-type mice retracted pseudopods after physiological shear exposure, whereas cells in Rac1-null mice showed no retraction during equal shear. On leukocytes from Rac2-null mice, however, fluid shear exerted a biphasic effect. Leukocytes with extended pseudopods slightly decreased in length, whereas initially round cells increased in length after shear application. The disruption of Rac activity made leukocytes nonresponsive to fluid shear, induced cell adhesion and microvascular stasis, and decreased microvascular density. Abstract

In addition to their role in bacterial killing by leukocytes, reactive oxygen species (ROS) have been increasingly recognized as important components of signaling and host defense in other cell types. The formation of ROS in both phagocytic- and non-phagocytic cells involves membrane-localized NADPH oxidases (Noxs). Nox proteins show structural homology to the cytochrome [b.sub.558] of leukocytes but, until recently, their regulation has been poorly understood. Here, we describe our current understanding of Nox function, and discuss emerging paradigms for regulation of Nox activity by Rac GTPase and/or other cytosolic components.

Abstract

The mechanisms by which Rho family GTPases (Rho, Rac and Cdc42) regulate coordinated changes to the actin cytoskeleton are being elucidated. This review will focus on the current evidence that the p21-activated kinases (PAKs) are involved in regulating some of the diverse cytoskeletal changes induced by Rac and Cdc42. PAKs have been shown to be required for processes including neurite formation and axonal guidance, development of cell polarity and motile responses. Signaling molecules interacting with PAKs that might contribute to the regulation of such processes have recently been identified.

Abstract

Leukocytes are not only important mediators of innate immunity, but they also induce and perpetuate inflammatory responses that are harmful to the host. Although inflammatory mediators activate leukocytes through a common heterotrimeric G protein ([G.sub.i]) signalling intermediate, many downstream inflammatory functions are regulated by distinct small GTPases, which suggests that pharmacological modulation of small GTPase activity would be useful in developing specific anti-inflammatory therapies. The recent identification of multiple small GTPase effectors, the recognition of the role of GTPase regulatory proteins in directing downstream signalling from small GTPases, and detailed structural information on the GTPases themselves suggests new possibilities for the development of effective and selective anti-inflammatory drugs.

Abstract

The guanine nucleotide exchange by the exchange protein, smgGDS, is dependent on the presence of an intact switch 1 and switch 2 region in Rac2, a GTP-binding protein. The reduced binding affinity of the Rac2 D38A and Q61L mutations for smgGDS indicates that the switch 1 at position 38 and switch 2 at position 61 are crucial for the ability to respond to smgGDS. The presence of an intact glutamine at position is necessary for the catalytic activity of smgGDS. The activating mutations: G12V and Q61L, slowly hydrolyze bound GTP and are unresponsive to p190 Rac GTPase-activating protein.

Abstract

D3, D4 polyphosphoinositides and the thrombin receptor-activating peptide(TRAP), GTP and GTPgammaS uncap actin filaments (F-actin) in resting permeabilized platelets. On the other hand GDPbetaS inhibits TRAP-induced F-actin uncapping, and PI(4,5)p2 suppresses this inhibition.

Abstract

A study of the role of Bcr protein in normal and leukemic cells using bcr-null mutant mice reveals that gram-negative endotoxin exposure results in septic shock and enhances tissue injury due to neutrophils. Reactive oxygen metabolite synthesis following the stimulation of neutrophils is enhanced in bcr (-/-) mice. P21(rac2) membrane translocation is 3 times higher in stimulated (-/-) neutrophils than in (+/+) neutrophils, indicating that in vivo regulation of Rac involves Bcr at the time of neutrophil priming and activation, and the regulation is related to the cell types involved in Ph-positive leukemia.

Abstract

A study of the involvement of the small GTP-binding protein Rho in modulating 4,5-bisphosphate (4,5-PIP2) synthesis through the treatment of cells with lovastatin, a GTP-binding protein modification inhibitor, reveals a reduction in PIP2 levels and a decrease in calcium mobilization in response to PDGE and thrombin. Recombinant Rho bound to GTP enhances PIP 5-kinase activity, while GDP-Rho is less effective. Microinjection of botulinum C3 exoenzyme minimizes calcium mobilization in response to PDGF or thrombin, but the effect is reversed by microinjection of activated Rho, indicating that Rho regulates the synthesis and hydrolysis of 4, 5-PIP2.

Abstract

Analyses of the process that ends the production of second messengers stimulated by chemotactic factor-receptor complexes reveal that a large portion of the alpha subunits of the Gn protein (Gnalpha) in inactivated neutrophils was linked to a cytoskeletal portion, and that activation with the chemotactic element fMet-Leu-Phe caused the release of these subunits. fMet-Leu-Phe-stimulated activation of neutrophils includes a particular dissociation of Gn alpha from the cytoskeleton. This release is not because of the influence of fMet-Leu-Phe on the cytoskeleton of neutrophils.
