Abstract

Identification of the mechanical performance of pharmaceuticals in the drug discovery process can determine the tabletability of a target molecule. Determination of the active slip systems and their ranking in molecular crystals is challenging because molecules offer a set of configurational variables absent from metallic or simple ionic materials, such as bond rotations, molecular rotations, and the relative orientation of molecules. This paper uses two computational methods, the rigid-block and tensor-based shearing methods, to calculate the slip barriers and gain insights regarding the slip deformation of simple molecular crystalline materials, using diatomic solid oxygen and anthracene as examples. Both methods use constrained quasi-static energy minimisation to simulate the materials' displacement and homogeneous shearing. These shearing methods rank the slip systems in oxygen and anthracene in agreement with experiment, including those reported herein where two previously unknown active slip systems in the basal plane of anthracene were identified independently from the computations. Internal degrees of freedom, in the form of shear-induced molecular rotations, critically influence the slip barriers and deformation mechanism. Our results uncover rotational twinning, which is linked to crystallographic symmetry rather than partial dislocations, while homogeneous shear of anthracene leads to a series of polymorphic transitions. The results also provide alternative interpretations of slip-observed morphologies.

14. A three-dimensional anisotropic point process characterization for pharmaceutical coatings

Abstract

Spatial characterization and modeling of the structure of a material may provide valuable knowledge on its properties and function. Especially, for a drug formulation coated with a polymer film, understanding the relationship between pore structure and drug release properties is important to optimize the coating film design. Here, we use methods from image analysis and spatial statistics to characterize and model the pore structure in pharmaceutical coatings. More precisely, we use and develop point process theory to characterize the branching structure of a polymer blended film with data from confocal laser scanning microscopy. Point patterns, extracted by identifying branching points of pore channels, are both inhomogeneous and anisotropic. Therefore, we introduce a directional version of the inhomogeneous K-function to study the anisotropy and then suggest two alternative ways to model the anisotropic three-dimensional structure. First, we apply a linear transformation to the data such that it appears isotropic and subsequently fit isotropic inhomogeneous Strauss or Lennard-Jones models to the transformed pattern. Second, we include the anisotropy directly in a Lennard-Jones and a more flexible step-function model with anisotropic pair-potential functions. The methods presented will be useful for anisotropic inhomogeneous point patterns in general and for characterizing porous material in particular.

Abstract

International audience; Background: In the context of a methicillin-susceptible Staphylococcus aureus (MSSA) outbreak, we aimed to improve our knowledge of S. aureus (SA) epidemiology in the neonatal care center (NCC) of a tertiary care teaching hospital.Methods: We performed a complete one-year review of SA carrier, colonized or infected patients. Monthly prevalence and incidence of SA intestinal carriage, colonization and infection were calculated and the types of infection analysed. During the MSSA outbreak, strains were studied for antimicrobial resistance, content of virulence genes and comparative fingerprint in Pulsed-Field Gel Electrophoresis. Hand hygiene and catheter-related practices were assessed by direct observational audits. Environmental investigation was performed in search of a SA reservoir.Results: Epidemiological analyses showed 2 or 3 prevalence peaks on a background of SA endemicity. In the NCC, during 2009, overall MSSA prevalence did not decrease below 5.5%, while mean MRSA prevalence was about 1.53%. Analysis of infection cases revealed that the outbreak corresponded to the emergence of catheter-related infections and was probably related to the relaxation in infection control practices in a context of high colonization pressure. Health care workers’ white coats appeared as a potential environmental reservoir that could perpetuate SA circulation in the ward.Conclusion: This report emphasizes the importance of integrating MSSA along with methicillin-resistant SA in a program of epidemiological surveillance in the NCC.

36. Spatial scales of mesoscale variability in the North-Atlantic as deduced from geosat data

Abstract

Two years of Geosat data were used to systematically investigate the mesoscale variability and spatial scales of the North Atlantic. We first calculated sea surface height variability and eddy kinetic energy. These compare well with the eddy kinetic energy obtained with surface drifters. Then the spatial scales of mesoscale variability were characterized in 10° (latitude) × 10° (longitude) areas by their wave number spectra and autocorrelation functions. Comparison with existing in situ data shows good agreement, and it is believed that the total error budget is globally below that of the oceanic signal. The wave number spectra show significant differences as a function of latitude and longitude, and these can be related to different types of forcing (instability of a mean current or fluctuating wind). Scales typically decrease from west to east and south to north. Simple proportionality with respect to the first internal Rossby radii does not apply everywhere.

48. Water-Based Latex Dispersions. 2. Adsorption and Dynamics of Nonionic Surfactants on Colloidal Particles with Different Interfacial Properties

Abstract

Studies with the proton nuclear magnetic resonance relaxation technique were carried out on the adsorption on colloidal silica and polystyrene latex dispersions of the nonionic surfactants NP10 and NP100 (poly(ethylene glycol) nonylphenol ether with 10 and 84 oxyethylene groups in mean, respectively) and of poly(ethylene glycol)s (PEGs) of molecular weights 196 and 4120 g/mol. It was found that the larger PEG on silica displayed similar molecular dynamics as compared to the surfactant with 84 oxyethylene groups (NP100) on silica. This suggested that the same relative amounts of trains and loops were present on the polar surface. On the other hand, the PEG4120 on polystyrene showed a more restricted dynamics than the NP100 surfactant adsorbed on the hydrophobic polystyrene surface. The NP10 experienced a high restriction of its dynamics on polystyrene seen as removal of segmental motions, while the PEG196 had a faster dynamic on this surface. The mobility of PEG196 on silica was even higher. Conclusively, the predominant adsorption mechanism on a hydrophobic surface changed from the thermodynamically more favorable situation with contact between the surfactant nonylphenol group and the particle surface to a less favorable contact when oxyethylene segments of the PEG molecules interacted with surface ionic sites.

Abstract

Properties of the surface-active substance polyoxyethylene(100) nonylphenol ether (NF100), in water and in deuterated water at room temperature, were investigated by static and dynamic surface tension measurements, using the fluorescence probe technique with pyrene and prodan and by NMR self-diffusion. It was found that at concentrations below 0.01% w/w, the substance could be described as a random-coil polymer at Θ conditions. The surface tension and fluorescence experiments indicated changes in surface adsorption and domain formations in the bulk at concentrations of between 0.01 and 0.06% w/w. At this higher concentration, no further adsorption took place at the air/water interface. However, the addition of more substance resulted in a bulk process of increased complexity involving aggregate formation and network formation, as was also revealed by the NMR self-diffusion experiments at even higher concentrations.
