Regulation of gene expression in trypanosomatid parasitic protozoa is mainly achieved posttranscriptionally. RNA-binding proteins (RBPs) associate to 3′ untranslated regions in mRNAs through dedicated domains such as the RNA recognition motif (RRM). Trypanosoma cruziUBP1 (TcUBP1) is an RRM-type RBP involved in stabilization/degradation of mRNAs. TcUBP1 uses its RRM to associate with cytoplasmic mRNA and to mRNA granules under starvation stress. Here, we show that under starvation stress, TcUBP1 is tightly associated with condensed cytoplasmic mRNA granules. Conversely, under high nutrient/low density-growing conditions, TcUBP1 ribonucleoprotein (RNP) complexes are lax and permeable to mRNA degradation and disassembly. After dissociating from mRNA, TcUBP1 can be phosphorylated only in unstressed parasites. We have identified TcP22, the ortholog of mammalian P32/C1QBP, as an interactor of TcUBP1 RRM. Overexpression of TcP22 decreased the number of TcUBP1 granules in starved parasites in vivo. Endogenous TcUBP1 RNP complexes could be dissociated in vitro by addition of recombinant TcP22, a condition stimulating TcUBP1 phosphorylation. Biochemical and in silico analysis revealed that TcP22 interacts with the RNA-binding surface of TcUBP1 RRM. We propose a model for the decondensation of TcUBP1 RNP complexes in T.cruzi through direct interaction with TcP22 and phosphorylation. Fil: Cassola, Alejandro Carlos. Background: Adipose tissue is the main source of the cytokines and adipokines that are increased in the context of obesity. The production of reactive oxygen species (ROS) and cytokines by circulating immune cells can be regulated by these pro-inflammatory factors even before infiltration into adipose tissue. Objective: To investigate the alterations that can occur in circulating monocytes and lymphocytes in paediatric obesity. Methods: In this study, 54 paediatric obese patients and 30 age-matched metabolically healthy individuals were enrolled. Intracellular cytokines were analyzed after phorbol myristate acetate (PMA) or leptin plus PMA stimulation of lymphocytes and monocytes by flow cytometry. ROS generation was measured using dichlorofluorescein-diacetate. Both a 'stimulation index' and a 'fold of increase' were calculated for statistical purposes. Results: Both interferon gamma (IFN-γ) production by circulating CD4+ and CD8+ lymphocytes and ROS production by monocytes following PMA stimulation were increased in obese patients. Leptin induced an increased production of IFN-γ in both subsets of T cells and tumour necrosis factor alpha in monocytes, and linoleic acid induced a higher ROS production in monocytes. Conclusions: The distinct functional responses of circulating cells suggest that alterations in both innate and adaptive immune cells are involved in the maintenance of low-grade inflammation in paediatric obesity. 8. An instrument to measure adherence to weight loss programs: The Compliance Praxis Survey-Diet (COMPASS-Diet)

- Abstract
Adherence to behavioral weight loss strategies is important for weight loss success. We aimed to examine the reliability and validity of a newly developed compliance praxis-diet (COMPASS-diet) survey with participants in a 10-week dietary intervention program. During the third of five sessions, participants of the “slim-without-diet” weight loss program (n = 253) completed the COMPASS-diet survey and provided data on demographic and clinical characteristics, and general self-efficacy. Group facilitators completed the COMPASS-diet-other scale estimating participants’ likely adherence from their perspective. We calculated internal consistency, convergent validity, and predictive value for objectively measured weight loss. Mean COMPASS-diet-self score was 82.4 (SD 14.2) and COMPASS-diet-other score 80.9 (SD 13.6) (possible range 12–108), with lowest scores in the normative behavior subscale. Cronbach alpha scores of the COMPASS-diet-self and -other scale were good (0.82 and 0.78, respectively). COMPASS-diet-self scores (r = 0.31) correlated more highly with general self-efficacy compared to COMPASS-diet-other scores (r = 0.04) providing evidence for validity. In multivariable analysis adjusted for age and gender, both the COMPASS-diet-self (F = 10.8, p < 0.001, r2 = 0.23) and other (F = 5.5, p < 0.001, r2 = 0.19) scales were significantly associated with weight loss achieved at program conclusion. COMPASS-diet surveys will allow group facilitators or trainers to identify patients who need additional support for optimal weight loss.
