Abstract

Fairness is a desirable and crucial property of many protocols that handle, for instance, exchanges of message. It states that if at least one agent engaging in the protocol is honest, then either the protocol will unfold correctly and fulfill its intended goal for all participants, or it will fail for everyone. In this work, we present a game-based framework for the study of fairness protocols, that does not define a priori an attacker model. It is based on the notion of strong secure equilibria, and leverages the conceptual and algorithmic toolbox of game theory. In the case of finite games, we provide decision procedures with tight complexity bounds for determining whether a protocol is immune to nefarious attacks from a coalition of participants, and whether such a protocol could exist based on the underlying graph structure and objectives., Comment: 53 pages, 14 figures

Abstract

We report on the melting dynamics of frozen olive oil in quiescent water for Rayleigh numbers up to $10^9$. The density difference results in an upward buoyancy-driven flow of liquid oil forming a thin film around the frozen oil. We experimentally investigate flat, cylindrical, and spherical shapes and we derive theoretical expressions for the local film thickness, velocity, and the local melt rate for these three canonical geometries. 8. The Perspectives of Agents Working in Positions of Authority within Health Sector Organisations Regarding the Involvement of Experts by Experience with an Intellectual Disability: An Exploratory Study

Abstract

Background: In this study, agents working in positions of authority within health sector organisations shared their perspectives concerning both the involvement of experts by experience with intellectual disabilities within their own organisations and their recommendations as to how best to involve experts by experience in policy and practice. Method: Using purposive sampling, eight agents were selected for semi-structured interviews. The interviews were analysed using thematic analysis. Results: The results indicated the importance of involving experts by experience. However, they also highlighted several barriers such as a lack of emphasis on genuine attention, insufficient visibility and recognition of expertise, and a lack of support for sustainable inclusion. Conclusions: Agents are willing to involve experts by experience in health sector organisations, but practical challenges hindered them from actually doing so. This study emphasises the importance to find solutions for better inclusion in policies.

Abstract

We study the Reynolds-number effects on the outer region of moderate adverse-pressure-gradient (APG) turbulent boundary layers (TBLs) and find that their small scale energy reduces with increasing friction Reynolds-number ($Re_{\tau}$). The trend is based on analyzing APG TBL data across 600 $\lesssim$ $Re_{\tau}$ $\lesssim$ 7000, and contrasts with the negligible variation in small scale energy noted for canonical wall flows. The datasets considered include those from a well-resolved numerical simulation (Pozuelo et al. 2022), which provides access to an APG TBL maintained at near-equilibrium conditions across 1000 $\lesssim$ $Re_{\tau}$ $\lesssim$ 2000, with a well-defined flow history, and a new high-$Re_{\tau}$ ($\sim$ 7000) experimental study from the large Melbourne wind tunnel, with its long test section modified to permit development of an APG TBL from a 'canonical' upstream condition. The decrease in small scale energy with $Re_{\tau}$ is revealed via decomposing the streamwise normal stresses into small and large scale contributions, based on a sharp spectral cut-off. The origin for this trend is traced back to the production of turbulent kinetic energy in an APG TBL, the small scale contribution to which is also found to decrease with $Re_{\tau}$ in the outer region. The conclusion is reaffirmed by investigating attenuation of streamwise normal stresses due to changing spatial-resolutions of the numerical grid/hotwire sensors, which reduces with increasing $Re_{\tau}$ and is found to be negligible at $Re_{\tau}$ $\sim$ 7000 in this study. Abstract Nailfold capillary density is lower in patients with pulmonary arterial hypertension (PAH). It is unclear whether this observation signifies a unique systemic manifestation of PAH, or reflects microcirculatory dysfunction secondary to pulmonary hypertension (PH). Capillary density and loop dimensions were measured by nailfold-capillaroscopy (NC) in 30 PAH (23 idiopathic, or iPAH, 7 hereditary, or hPAH), 17 chronic thromboembolic PH (CTEPH) patients and 48 controls. NC-Measurements were repeated after pulmonary endarterectomy (PEA) or balloon pulmonary angioplasty (BPA) in CTEPH patients. Abstract

We study two natural problems about rational behaviors in multiplayer non-zero-sum sequential infinite duration games played on graphs: checking problems, that consist in deciding whether a strategy profile, defined by a Mealy machine, is rational; and rational verification, that consists in deciding whether all the rational answers to a given strategy satisfy some specification. We give the complexities of those problems for two major concepts of rationality: Nash equilibria and subgame-perfect equilibria, and for five major classes of payoff functions: parity, mean-payoff, quantitative reachability, energy, and discounted-sum.

Abstract

Stable isotope analysis of plant remains recovered from archaeological sites is becoming more routine. There remains a lack of consensus, however, on how to appropriately select archaeological plant remains for isotopic analysis, how to account for differences in preservation and the effect of potential contamination, and how to interpret the measured isotope values in terms of the conditions in which the plants grew. In this paper, we outline the main issues to be considered when planning and conducting an isotopic study of archaeobotanical remains. These include: (1) setting out the research question(s) that will be answerable using available analytical approaches, (2) considering the archaeological context from which plant remains derive, (3) determining appropriate sample size through consideration of estimate precision, (4) establishing the conditions in which plant remains have been preserved and potential effects on their isotope values, and (5) accounting for possible contamination during deposition. With these issues in mind, we propose some recommendations for researchers to consider when planning and conducting an isotopic study of archaeobotanical remains.

Abstract

BackgroundDrug-drug interactions (DDIs) may influence the effectiveness and safety of medication treatment, which may require additional monitoring, dose adjustment or avoidance of certain drugs. DDIs involving P-glycoprotein (P-gp) affect many drugs, but current official product information is often insufficient to guide the management of these DDIs in clinical practice. The aim of this paper is to describe a protocol to assess DDIs involving P-gp and to develop and implement practice recommendations for clinically relevant P-gp-mediated DDIs that affect clinical outcomes through changes in systemic drug exposure.MethodsA combined literature review and expert opinion approach will be used according to the following seven steps: set up an expert panel (step 1), establish core concepts and definitions (step 2), select potential P-gp-modulators (i.e., P-gp-inducers and -inhibitors) and P-gp-substrates to be evaluated (step 3), select and extract evidence-based data, and present findings in standardized assessment reports (step 4), discuss and adopt classifications and practice recommendations with the expert panel (step 5), publish and integrate information and alerts in clinical decision support systems (CDSS) (step 6), (re)assessments of DDIs and potential new DDIs when new information is available or when initiated by healthcare providers (step 7).Anticipated resultsThe expert panel will classify potential P-gp-modulators and -substrates as clinically relevant P-gp-inducer, -inhibitor and/or -substrate and draw conclusions about which combinations of classified modulators and substrates will lead to clinically relevant DDIs. This may include the extrapolation of conclusions for DDIs where limited or no data are available, based on the pharmacological characteristics of these drugs. For (potential) DDIs that are considered to be clinically relevant, practice recommendations will be developed.DiscussionThis protocol describes a standardized, evidence- and expert opinion-based assessment of P-gp-mediated DDIs that affect clinical outcomes. This approach will generate alerts with practice recommendations for clinically relevant DDIs and transparent rationales for DDIs that are considered to be irrelevant. These recommendations will improve individual patient care by supporting healthcare professionals to make consistent decisions on how to manage P-gp mediated DDIs.

Abstract Objectives PD‐1/PD‐L1 immune checkpoint blockade can be an effective treatment for advanced breast cancer patients. However, patients with oestrogen receptor positive (ER+) tumors often display only low lymphocyte infiltration, while a large part of triple negative (TN) breast tumors does not generate an effective immunotherapy response. Therefore, new treatment strategies have to be developed. Here, we investigate Siglec‐7 and Siglec‐9 as novel ITIM‐bearing inhibitory immune checkpoint receptors similar to PD‐1, but expressed on a broader range of immune cells. Methods We assessed Siglec‐7 and Siglec‐9 (ligand) expression in TN and ER+ breast cancer tumors and their breast cancer cell line‐induced signalling. Results We report that Siglec‐7 and Siglec‐9 are highly expressed in TN tumors, and to a low extent in ER+ tumors. Siglec‐7 was observed on myeloid cells, T cells, and NK cells and Siglec‐9 preferentially on myeloid cells. Expression of sialoglycans, including Siglec‐7 and Siglec‐9 ligands, was observed in both TN and ER+ breast cancer tissue sections. Expression levels of Siglec‐7 and Siglec‐9 ligands were higher on in vitro cultured TN cell lines than ER+ cell lines. Importantly, by applying chimeric Siglec‐7 reporter cells, we showed the induction of Siglec‐7 signalling by multiple TN cell lines, but only by one ER+ cell line. Moreover, Siglec‐7 signalling is directly related to Siglec‐7 ligand expression levels of breast cancer cell lines. Conclusion These data imply that immunotherapy targeting Siglec receptors may be particularly interesting for TN breast cancer patients not responding to current treatment strategies with tumors displaying high immune cell infiltration.

25. GRIN3A: A biomarker associated with a cribriform pattern and poor prognosis in prostate cancer

Abstract

Prostate cancer with a cribriform pattern, including invasive cribriform carcinoma (ICC) and/or intraductal carcinoma (IDC) is associated with a poor prognosis, and the underlying mechanisms are unclear. Therefore, we aimed to identify biomarkers for this feature. Using a radical prostatectomy cohort, we performed within-patient differential expression analyses with RNA sequencing data to compare samples with a cribriform pattern to those with non-cribriform Gleason pattern 4 (NcGP4; n=13). ACSM1, GRIN3A, PCDHB2, and REG4 were identified as differentially expressed, and validation was performed using real-time reverse transcription polymerase chain reaction (n=99; 321 RNA samples) and RNA in situ hybridization on tissue microarrays (n=479; 2047 tissue cores). GRIN3A was significantly higher expressed in cribriform pattern vs. NcGP4, when assessed within the same patient (n=27; p=0.005) and between different patients (n=83; p=0.001). Tissue cores with IDC more often expressed GRIN3A compared to ICC, NcGP4, and benign tissue (52 % vs. ≤ 32 %). When IDC and NcGP4 was compared within the same patient (173 pairs of tissue cores; 54 patients), 38 (22 %) of the tissue microarray core pairs had GRIN3A expression in only IDC, 33 (19 %) had expression in both IDC and NcGP4, 14 (8 %) in only NcGP4 and 88 (51 %) were negative in both entities (p=0.001). GRIN3A was as well associated with biochemical recurrence (log-rank, p=0.002). In conclusion, ectopic GRIN3A expression is an RNA-based biomarker for the presence of cribriform prostate cancer, particularly for IDC.

Abstract

Abstract Cardiac fibrosis contributes to the development of heart failure, and is the response of cardiac fibroblasts (CFs) to pressure or volume overload. Limiting factors in CFs research are the poor availability of human cells and the tendency of CFs to transdifferentiate into myofibroblasts when cultured in vitro. The possibility to generate CFs from induced pluripotent stem cells (iPSC), providing a nearly unlimited cell source, opens new possibilities. However, the behaviour of iPSC-CFs under mechanical stimulation has not been studied yet. Our study aimed to assess the behaviour of iPSC-CFs under mechanical stretch and pro-fibrotic conditions. First, we confirm that iPSC-CFs are comparable to primary CFs at gene, protein and functional level. Furthermore, iPSC-derived CFs adopt a pro-fibrotic response to transforming growth factor beta (TGF-β). In addition, mechanical stretch inhibits TGF-β-induced fibroblast activation in iPSC-CFs. Thus, the responsiveness to cytokines and mechanical stimulation of iPSC-CFs demonstrates they possess key characteristics of primary CFs and may be useful for disease modelling.

Abstract

Abstract Introduction Administration of oxygen therapy is common, yet there is a lack of knowledge on its ability to prevent cellular hypoxia as well as on its potential toxicity. Consequently, the optimal oxygenation targets in clinical practice remain unresolved. The novel PpIX technique measures the mitochondrial oxygen tension in the skin (mitoPO2) which allows for non-invasive investigation on the effect of hypoxemia and hyperoxemia on cellular oxygen availability. Results During hypoxemia, SpO2 was 80 (77–83)% and PaO2 45(38–50) mmHg for 15 min. MitoPO2 decreased from 42(35–51) at baseline to 6(4.3–9)mmHg (p 200 mmHg were both associated with a reduction in mitoPO2. Conclusions Hypoxemia decreases mitoPO2 profoundly, despite complete compensation of global oxygen delivery. In addition, hyperoxemia also decreases mitoPO2, accompanied by a reduction in microcirculatory perfusion. These results suggest that mitoPO2 can be used to titrate oxygen support.

32. Parent Influences on the Dietary Habits of Young Adults

Abstract

To better understand the processes through which eating habits during childhood are carried over into adulthood, I asked the question: under what circumstances and through which patterns do the feeding behaviors of parents become replicated and emerge as habitual in their children during young adulthood? I aim to investigate how parenting style and parent behaviors surrounding food and diet influence children’s dietary habits in the long-term. Previous research indicates that income is a major factor determining parents’ feeding behavior, so I set out to combine income level and parent feeding style in one study. I hypothesized that young adults who recalled their parents engaging in behaviors associated with the authoritative parenting style would be most likely to replicate those eating behaviors as habit from childhood into young adulthood. To better understand the process through which these variables ultimately cultivated the dietary behaviors of the young adults I interviewed, I selected three dietary habits—presence of breakfast, moderation of added sugar intake, sufficient vegetable consumption—that have been associated with positive health outcomes. II observed several distinct patterns: (i) young adults who continue to eat breakfast had previously experienced a high level of engagement from their parents surrounding the meal during childhood; (ii) young adults whose parents engaged with their children’s added sugar intake were more likely to carry similar habits and attitudes related to sugar into young adulthood; (iii) many young adults experienced external factors which also initiated a significant transition towards healthier eating habits independent of their parents’ influence.

Abstract

Abstract Chronic obstructive pulmonary disease (COPD) is a persistent and progressive disorder characterized by airway or alveolar abnormalities, commonly leading to pulmonary hypertension (PH). This clinical observational study investigates the therapeutic mechanisms of Bufei Huoxue capsules (BHC) in treating PH in patients with COPD‐linked PH (COPD‐PH) using network pharmacology and molecular docking methods, and assesses the therapeutic efficacy and safety of BHCs. The active compounds and their target proteins in BHCs were sourced from the Traditional Chinese Medicine Systems Pharmacology database, with additional target proteins derived from the GeneCards and OMIM databases. An active network was constructed using Cytoscape 3.7.1, and interaction networks were established. Intersecting targets underwent Gene Ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis using the Metascape database. Network pharmacology and molecular docking studies demonstrated favorable binding affinities of BHC active ingredients, such as quercetin, bavachalcone, and isobavachin, for key targets including PTGS1, ESR1, and PTGS2. Gene Ontology enrichment analysis highlighted the involvement of these targets in processes such as the positive regulation of locomotion, the transmembrane receptor protein tyrosine kinase signaling pathway, and peptidyl‐tyrosine phosphorylation. KEGG pathway analysis indicated their roles in pathways related to cancer, AGE‐RAGE signaling in diabetic complications, and prostate cancer. BHCs exhibit therapeutic effects on COPD‐PH through multi‐component, multi‐target, and multi‐pathway interactions. This clinical observational study confirms the efficacy and safety of BHCs in improving cardiac and pulmonary functions, enhancing exercise tolerance, and elevating the quality of life in patients with COPD‐PH.

34. An EMDR-based intervention for the ‘Golfers’ Yips’: A case series

Abstract

Abstract: Introduction: The yips in golfers is a task-specific movement disorder. Anxiety might play a role in its emergence and persistence. The objective was to report on the effect of an Eye Movement Desensitization and Reprocessing (EMDR) based intervention on the yips by reducing anxiety. Methods: We present a case-series with seven golfers with observed right-handed yips. The participants were treated with EMDR in one or two sessions and were followed for one year. A bespoke Yips Rating Scale (YRS) was determined and golfing skills were assessed at baseline and after one year. Results: All participants completed treatment without complications. Follow-up data from six golfers after the intervention showed a positive effect of the EMDR based intervention on their yips symptoms. YRS scores improved after the intervention in 6 golfers (p=0.04) and golfers’ performance appeared to improve by an average of 20%. Conclusion: Anxiety might play a crucial role in the emergence and cultivation of the yips in golfers. The reduction of accompanying and anticipatory anxiety by means of EMDR in golfers seems safe and could be beneficial in diminishing the yips. Larger and better studies are needed to further evaluate the effect of EMDR in the golfer’s yips and other sport-related task-specific movement disorders.

Abstract

Summary: Human genetic studies of critical COVID-19 pneumonia have revealed the essential role of type I interferon-dependent innate immunity to SARS-CoV-2 infection. Conversely, an association between the HLA-B∗15:01 allele and asymptomatic SARS-CoV-2 infection in unvaccinated individuals was recently reported, suggesting a contribution of pre-existing T cell-dependent adaptive immunity. We report a lack of association of classical HLA alleles, including HLA-B∗15:01, with pre-omicron asymptomatic SARS-CoV-2 infection in unvaccinated participants in a prospective population-based study in the United States (191 asymptomatic vs. 945 symptomatic COVID-19 cases). Moreover, we found no such association in the international COVID Human Genetic Effort cohort (206 asymptomatic vs. 574 mild or moderate COVID-19 cases and 1,625 severe or critical COVID-19 cases). Finally, in the Human Challenge Characterisation study, the three HLA-B∗15:01 individuals infected with SARS-CoV-2 developed symptoms. As with other acute primary infections studied, no classical HLA alleles favoring an asymptomatic course of SARS-CoV-2 infection were identified.

Abstract

We consider Markov decision processes with synchronizing objectives, which require that a probability mass of $1-\epsilon$ accumulates in a designated set of target states, either once, always, infinitely often, or always from some point on, where $\epsilon = 0$ for sure synchronizing, and $\epsilon \to 0$ for almost-sure and limit-sure synchronizing. We introduce two new qualitative modes of synchronizing, where the probability mass should be either positive, or bounded away from $0$. They can be viewed as dual synchronizing objectives. We present algorithms and tight complexity results for the problem of deciding if a Markov decision process is positive, or bounded synchronizing, and we provide explicit bounds on $\epsilon$ in all synchronizing modes. In particular, we show that deciding positive and bounded synchronizing always from some point on, is coNP-complete.

In this paper, we provide an effective characterization of all the subgame-perfect equilibria in infinite duration games played on finite graphs with mean-payoff objectives. To this end, we introduce the notion of requirement, and the notion of negotiation function. We establish that the plays that are supported by SPEs are exactly those that are consistent with a fixed point of the negotiation function. 38. The CSU Engineering Topic Tree: The First Four Years

Abstract

The Charles Sturt University (CSU) Engineering programme is a new course (degree programmme) established in 2016 by a university that had not previously taught engineering. This start from scratch occasion was taken as an opportunity to build an all-new programme structure and philosophy. Students at CSU Engineering complete a sequence of three semester-long Project-Based Learning (PBL) style challenges across their time face-to-face at the university; after this point, they commence four yearlong paid industry-based work placements and continue studies in an online mode during evenings, weekends, and scheduled study days. The underlying technical curriculum for the engineering programme at Charles Sturt University is delivered mostly on-line via the RealizeIT platform and is based on a philosophy of just-in-time, self-directed learning. Students have freedom in deciding when, how and, to a large extent, which elements of the curriculum they engage within the online environment. This freedom, along with the PBL-style challenges, is enabled by the structure of the technical curriculum which is broken down into fine-grained learning activities called 'topics'. In this paper, we summarise our experiences during the first four years, and the insights gained into student behaviours when offered an opportunity to engage in self-directed learning.

Abstract

Slow-moving landslides move downslope at velocities that range from mm year−1 to m year−1. Such deformations can be measured using satellite-based synthetic aperture radar interferometry (InSAR). We developed a new method to systematically detect and quantify accelerations and decelerations of slowly deforming areas using InSAR displacement time series. The displacement time series are filtered using an outlier detector and subsequently piecewise linear functions are fitted to identify changes in the displacement rate (i.e., accelerations or decelerations). Grouped accelerations and decelerations are inventoried as indicators of potential unstable areas. We tested and refined our new method using a high-quality dataset from the Mud Creek landslide, CA, USA. Our method detects accelerations and decelerations that coincide with those previously detected by manual examination. Second, we tested our method in the region around the Mazar dam and reservoir in Southeast Ecuador, where the time series data were of considerably lower quality. We detected accelerations and decelerations occurring during the entire study period near and upslope of the reservoir. Application of our method results in a wealth of information on the dynamics of the surface displacement of hillslopes and provides an objective way to identify changes in displacement rates. The displacement rates, their spatial variation, and the timing of accelerations and decelerations can be used to study the physical behavior of a slow-moving slope or for regional hazard assessment by linking the timing of changes in displacement rates to landslide causal and triggering factors.

Abstract

Objective: Brain-body interfaces (BBIs) have emerged as a very promising solution for restoring voluntary hand control in people with upper-limb paralysis. The BBI module decoding motor commands from brain signals should provide the user with intuitive, accurate, and stable control. Here, we present a preliminary investigation in a monkey of a brain decoding strategy based on the direct coupling between the activity of intrinsic neural ensembles and output variables, aiming at achieving ease of learning and long-term robustness. Results: We identified an intrinsic low-dimensional space (called manifold) capturing the co-variation patterns of the monkey's neural activity associated to reach-to-grasp movements. We then tested the animal's ability to directly control a computer cursor using cortical activation along the manifold axes. By daily recalibrating only scaling factors, we achieved rapid learning and stable high performance in simple, incremental 2D tasks over more than 12 weeks of experiments. Finally, we showed that this brain decoding strategy can be effectively coupled to peripheral nerve stimulation to trigger voluntary hand movements. Conclusions: These results represent a proof of concept of manifold-based direct control for BBI applications.

Abstract

Abstract The pathobiology of respiratory failure in COVID-19 consists of a complex interplay between viral cytopathic effects and a dysregulated host immune response. In critically ill patients, imatinib treatment demonstrated potential for reducing invasive ventilation duration and mortality. Here, we perform longitudinal profiling of 6385 plasma proteins in 318 hospitalised patients to investigate the biological processes involved in critical COVID-19, and assess the effects of imatinib treatment. Nine proteins measured at hospital admission accurately predict critical illness development. Next to dysregulation of inflammation, critical illness is characterised by pathways involving cellular adhesion, extracellular matrix turnover and tissue remodelling. Imatinib treatment attenuates protein perturbations associated with inflammation and extracellular matrix turnover. These proteomic alterations are contextualised using external pulmonary RNA-sequencing data of deceased COVID-19 patients and imatinib-treated Syrian hamsters. Together, we show that alveolar capillary barrier disruption in critical COVID-19 is reflected in the plasma proteome, and is attenuated with imatinib treatment. This study comprises a secondary analysis of both clinical data and plasma samples derived from a clinical trial that was registered with the EU Clinical Trials Register (EudraCT 2020–001236–10, https://www.clinicaltrialsregister.eu/ctr-search/trial/2020-001236-10/NL ) and Netherlands Trial Register (NL8491, https://www.trialregister.nl/trial/8491 ).
